Diffusible iodine-based contrast-enhanced computed tomography (diceCT) is an increasingly used digital complement, supplement, or alternative to traditional dissection-based anatomical research. The diceCT protocol, which has evolved and expanded over the past decade, employs passive diffusion of Lugol's iodine (KI3) to increase soft tissue radiodensity and improve structure contrast in the CT or microCT imaging of specimens. The development and application of diceCT has focused largely on specimens under 1 kg, and the varying reporting of methods on studies of both small and large specimens has initiated, but not yet established, an effective diceCT protocol for larger specimens based on monitored experiments of several fundamental variables (e.g., Lugol's iodine concentration, duration, and impacts of Lugol's iodine on tissues). In this study, we have experimentally assessed the efficacy of diceCT protocols for imaging whole-body specimens of the 1–4.5 kg Australian brushtail possum (Trichosurus vulpecula) using sequential CT imaging assessment across experimental conditions. We assessed the impact of varying Lugol's iodine concentration, the presence/absence of skin, solution volume and agitation on tissue radiodensity changes through weekly CT-based monitoring of tissue radiodensities over an 8-week experimental period. We have also quantified tissue volumetric changes across our experiment to assess the impact of diceCT applications on subsequent analyses of imaging datasets. Our results indicate that substantial changes in both soft-tissue radiodensity and soft-tissue volume occur within the first 28 days of Lugol's iodine treatment, followed by a slower rate of progressive soft-tissue radiodensity and volume changes across the experiment duration. Our results demonstrate the negligible benefit of skinning larger specimens to improve solution diffusion, and document significant soft-tissue volumetric changes with high concentration solutions (e.g., 10%) and long-duration exposure (e.g., beyond 5 weeks) that should guide individual diceCT protocol design and/or quantification and analysis for mammal specimens above 1 kg. The practical aspect of human developmental biology education is often limited to the observation and use of animal models to illustrate developmental anatomy. This is due in part to the difficulty of accessing human embryonic and fetal specimens, and the sensitivity inherent to presenting these specimens as teaching materials. This report presents a new approach using three-dimensional (3D) printed replicas of actual human materials in practical classes, thus allowing for the inclusion of accurate examples of human developmental anatomy in the educational context. A series of 3D prints have been produced from digital data collected by computed tomography (CT) imaging of an archived series of preserved human embryonic and fetal specimens. The final versions of 3D prints have been generated in a range of single or multiple materials to illustrate the progression of human development, including the development of internal anatomy. Furthermore, multiple copies of each replica have been printed for large group teaching. In addition to the educational benefit of examining accurate 3D replicas, this approach lessens the potential for adverse student reaction (due to cultural background or personal experience) to observing actual human embryonic/fetal anatomical specimens, and reduces the potential of damage or loss of original specimens. This approach, in combination with ongoing improvements in the management and analysis of digital data and advances in scanning technology, has enormous potential to allow embryology students access to both local and international collections of human gestational material.

Reconstructing the detailed dietary behaviour of extinct hominins is challenging.sup.1--particularly for a species such as Australopithecus africanus, which has a highly variable dental morphology that suggests a broad diet.sup.2,3. The dietary responses of extinct hominins to seasonal fluctuations in food availability are poorly understood, and nursing behaviours even less so; most of the direct information currently available has been obtained from high-resolution trace-element geochemical analysis of Homo sapiens (both modern and fossil), Homo neanderthalensis.sup.4 and living apes.sup.5. Here we apply high-resolution trace-element analysis to two A. africanus specimens from Sterkfontein Member 4 (South Africa), dated to 2.6-2.1 million years ago. Elemental signals indicate that A. africanus infants predominantly consumed breast milk for the first year after birth. A cyclical elemental pattern observed following the nursing sequence--comparable to the seasonal dietary signal that is seen in contemporary wild primates and other mammals--indicates irregular food availability. These results are supported by isotopic evidence for a geographical range that was dominated by nutritionally depauperate areas. Cyclical accumulation of lithium in A. africanus teeth also corroborates the idea that their range was characterized by fluctuating resources, and that they possessed physiological adaptations to this instability. This study provides insights into the dietary cycles and ecological behaviours of A. africanus in response to food availability, including the potential cyclical resurgence of milk intake during times of nutritional challenge (as observed in modern wild orangutans.sup.5). The geochemical findings for these teeth reinforce the unique place of A. africanus in the fossil record, and indicate dietary stress in specimens that date to shortly before the extinction of Australopithecus in South Africa about two million years ago. Establishing relationships between soft tissues and bones in living species can inform our understanding of functional adaptations in their extinct kin in the absence of direct data on habitual behaviours. The koala and the wombats are the only surviving species of the vombatiform suborder, and represent the extant phylogenetic bracket for dozens of fossil marsupial species across nine families. Here we present the first quantitative architectural data on the forelimb myology of the koala and common wombat, alongside redescriptions based on physical and digital dissections. Despite obvious contrasts in how their forelimbs are used in arboreal (koala) and fossorial (wombat) functional contexts, overall they showed only minor differences in qualitative and quantitative measures of myology. When the architectural properties of antagonistic pairs of muscle groups crossing each joint were compared, the greatest contrasts were seen in muscles crossing the elbow, where the wombat exhibited greater relative emphasis on forceful elbow extension than the koala, indicative of adaptations for digging. Our findings reinforce the importance of the elbow as a useful system to explore when seeking discrimination between habitual patterns of forelimb use in extinct vombatiform species. Most of the diversity of extant southern true seals (Phocidae: Monachinae) is present in the Southern Ocean, but a poor fossil record means that the origin of this fauna remains unknown. Australia represents a large gap in the record bordering the Southern Ocean that could possibly inform on the origins of the extant Antarctic monachines, with most known fossils remaining undescribed. Here we describe the oldest Australian fossil pinniped assemblage, from the Late Miocene to the Early Pliocene of Beaumaris. Two fossils are referrable to Pinnipedia, five (possibly six) to Phocidae and a humerus is referrable to Monachinae. The humerus is not referrable to any extant tribe, potentially representing an archaic monachine. The description of this assemblage is consistent with the Neogene pinniped fauna of Australia being exclusively monachine before the arrival of otariids (fur seals and sea lions). The Beaumaris humerus, along with other Neogene phocids from the Southern Ocean margins, were smaller than their extant Antarctic relatives, possibly driven by longer food chains with less energy efficiency between trophic levels. This suggests that small archaic phocids potentially used the Southern Ocean as a means of dispersal before the arrival of extant Antarctic monachines. [ABSTRACT FROM AUTHOR]

The teaching of medical pathology has undergone significant change in the last 30–40 years, especially in the context of employing bottled specimens or 'pots' in classroom settings. The reduction in post-mortem based teaching in medical training programs has resulted in less focus being placed on the ability of students to describe the gross anatomical pathology of specimens. Financial considerations involved in employing staff to maintain bottled specimens, space constraints and concerns with health and safety of staff and student laboratories have meant that many institutions have decommissioned their pathology collections. This report details how full-colour surface scanning coupled with CT scanning and 3 D printing allows the digital archiving of gross pathological specimens and the production of reproductions or replicas of preserved human anatomical pathology specimens that obviates many of the above issues. With modern UV curable resin printing technology, it is possible to achieve photographic quality accurate replicas comparable to the original specimens in many aspects except haptic quality. Accurate 3 D reproductions of human pathology specimens offer many advantages over traditional bottled specimens including the capacity to generate multiple copies and their use in any educational setting giving access to a broader range of potential learners and users.
