Apricot breeding programs could be strongly improved by the availability of molecular markers linked to the main fruit quality traits. Fruit acidity is one of the key factors in consumer acceptance, but despite its importance, the molecular bases of this trait are still poorly understood. In order to increase the genetic knowledge on the fruit acidity, an F1 apricot population (‘Lito’ ‘BO81604311’) has been phenotyped for titratable acidity and juice pH for the three following years. In addition, the contents of the main organic acids of the juice (malate, citrate, and quinate) were also evaluated. A Gaussian distribution was observed for most of the traits in this progeny, confirming their quantitative inheritance. An available simple sequence repeat (SSR)- based molecular map, implemented with new markers in specific genomic regions, was used to perform a quantitative trait loci (QTL) analysis. The molecular map was also anchored to the recently published apricot genome sequence of ‘Stella.’ Several major QTLs linked to fruit acidity-related traits have been identified both in the ‘Lito’ (no. 21) and ‘BO81604311’ (no. 13), distributed in five linkage groups (LG 4, 5, 6, 7, and 8). Some of these QTLs show good stability between years and their linked markers were used to identify candidate genes in specific QTLs genomic regions.

Background AKI commonly occurs in patients with coronavirus disease 2019 (COVID-19). Its pathogenesis is poorly understood. The urokinase receptor system is a key regulator of the intersection between inflammation, immunity, and coagulation, and soluble urokinase plasminogen activator receptor (suPAR) has been identified as an immunologic risk factor for AKI. Whether suPAR is associated with COVID-19-related AKI is unknown. Methods In a multinational observational study of adult patients hospitalized for COVID-19, we measured suPAR levels in plasma samples from 352 adult patients that had been collected within 48 hours of admission. We examined the association between suPAR levels and incident in-hospital AKI. Results Of the 352 patients (57.4% were male, 13.9% were black, and mean age was 61 years), 91 (25.9%) developed AKI during their hospitalization, of whom 25 (27.4%) required dialysis. The median suPAR level was 5.61 ng/ml. AKI incidence rose with increasing suPAR tertiles, from a 6.0% incidence in patients with suPAR,4.60 ng/ml (first tertile) to a 45.8% incidence of AKI in patients with suPAR levels.6.86 ng/ml (third tertile). None of the patients with suPAR,4.60 ng/ml required dialysis during their hospitalization. In multivariable analysis, the highest suPAR tertile was associated with a 9.15-fold increase in the odds of AKI (95% confidence interval [95% CI], 3.64 to 22.93) and a 22.86-fold increase in the odds of requiring dialysis (95% CI, 2.77 to 188.75). The association was independent of inflammatory markers and persisted across subgroups. Conclusions Admission suPAR levels in patients hospitalized for COVID-19 are predictive of in-hospital AKI and the need for dialysis. Brazil contains two-thirds of remaining Amazonian rainforests and is responsible for the most Amazon forest loss. Primary forest loss in the Brazilian Amazon has declined considerably since 2004 but secondary forest loss has never been quantified. We use a recently developed high-resolution land use/land cover dataset to track secondary forests in the Brazilian Amazon over 14 yr, providing the first estimates of secondary forest loss for the region. We find that secondary forest loss increased by (187 ± 48)% from 2008 to 2014. Moreover, the proportion of total forest loss accounted for by secondary forests rose from (37 ± 3)% in 2000 to (72 ± 5)% in 2014. The recent acceleration in secondary forests loss occurred across the entire region and was not driven simply by increasing secondary forest area but probably a conscious preferential shift towards clearance of a little-protected forest ecosystem (secondary forests). Our results suggest that secondary forests loss has eased deforestation pressure on primary forests. However, this has been at the expense of a lost carbon sequestration opportunity of 2.59–2.66 Pg C over our study period.

Although analysis of retrospective studies has documented survival benefit from the addition of a macrolide to the treatment regimen for community-acquired pneumonia (CAP), no data are available to determine if there is differential efficacy between members of the macrolide family. In order to investigate this, an analysis was undertaken of data from 1174 patients with CAP who met the new Sepsis-3 definitions and were enrolled prospectively in the data registry of the Hellenic Sepsis Study Group. Four well-matched treatment groups were identified with 130 patients per group: clarithromycin and β-lactam; azithromycin and β-lactam; respiratory fluoroquinolone and β-lactam monotherapy. The primary endpoint was comparison of the effects of clarithromycin with β-lactam monotherapy on 28-day mortality. The secondary endpoint was resolution of CAP. Mortality rates for the clarithromycin, azithromycin, respiratory fluoroquinolone and β-lactam groups were 20.8%, 33.8% (P=0.026 vs clarithromycin), 32.3% (P=0.049 vs clarithromycin) and 36.2% (P=0.009 vs clarithromycin), respectively. After stepwise Cox regression analysis among all groups, clarithromycin was the only treatment modality associated with a favourable outcome (hazard ratio 0.61; P=0.021). CAP resolved in 73.1%, 65.9% (P=0.226 vs clarithromycin), 58.5% (P=0.009 vs clarithromycin) and 61.5% (P=0.046 vs clarithromycin) of patients, respectively. It is concluded that the addition of clarithromycin to the treatment regimen of patients with severe CAP leads to better survival rates. © 2019 Elsevier B.V. and International Society of Chemotherapy

Interim analysis of the phase III ACTIVATE trial to evaluate protection against infection in elderly patients reveals that BCG vaccination is safe, increases the time to first infection, and shows protection against viral respiratory infections. © 2020 Elsevier Inc. BCG vaccination in children protects against heterologous infections and improves survival independently of tuberculosis prevention. The phase III ACTIVATE trial assessed whether BCG has similar effects in the elderly. In this double-blind, randomized trial, elderly patients (n = 198) received BCG or placebo vaccine at hospital discharge and were followed for 12 months for new infections. At interim analysis, BCG vaccination significantly increased the time to first infection (median 16 weeks compared to 11 weeks after placebo). The incidence of new infections was 42.3% (95% CIs 31.9%–53.4%) after placebo vaccination and 25.0% (95% CIs 16.4%–36.1%) after BCG vaccination; most of the protection was against respiratory tract infections of probable viral origin (hazard ratio 0.21, p = 0.013). No difference in the frequency of adverse effects was found. Data show that BCG vaccination is safe and can protect the elderly against infections. Larger studies are needed to assess protection against respiratory infections, including COVID-19 (ClinicalTrials.gov NCT03296423). © 2020 Elsevier Inc.

The sudden clinical deterioration 7–8 days after initial symptom onset suggests that severe respiratory failure (SRF) in COVID-19 is driven by a unique pattern of immune dysfunction. We studied immune responses of 54 COVID-19 patients, 28 of whom had SRF. All patients with SRF displayed either macrophage activation syndrome (MAS) or very low human leukocyte antigen D related (HLA-DR) expression accompanied by profound depletion of CD4 lymphocytes, CD19 lymphocytes, and natural killer (NK) cells. Tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) production by circulating monocytes was sustained, a pattern distinct from bacterial sepsis or influenza. SARS-CoV-2 patient plasma inhibited HLA-DR expression, and this was partially restored by the IL-6 blocker Tocilizumab; off-label Tocilizumab treatment of patients was accompanied by increase in circulating lymphocytes. Thus, the unique pattern of immune dysregulation in severe COVID-19 is characterized by IL-6-mediated low HLA-DR expression and lymphopenia, associated with sustained cytokine production and hyper-inflammation. © 2020 Elsevier Inc.

The objective of this work was to analyze land use dynamics in the Brazilian Cerrado region from 2002 to 2013. This analysis was based on the interpretation of Landsat satellite images carried out by the projects Projeto de Conservação e Utilização Sustentável da Diversidade Biológica Brasileira (Probio) and TerraClass Cerrado 2013, both coordinated by Ministério do Meio Ambiente. In 2002, 38.9% of the Cerrado was covered by some type of anthropic activity. In 2013, this percentage increased to 43.4%. High mobility group box 1 (HMGB1) is released from macrophages as a late biomarker of sepsis. Conditions associated with pre-existing macrophage activation may modify HMGB1 expression. This study aimed to assess the impact of HMGB1 kinetics on 28-day mortality. In a sub-study of a previous randomized clinical trial among patients with systemic inflammatory response syndrome and gram-negative infections, patients were classified in early and late HMGB1 peak groups. Serial measurements of HMGB1, ferritin and interferon-gamma (IFNγ) were performed in all available sera. Two hundred ten patients were included; 118 (46.5%) had at least one inflammatory disease (diabetes, chronic obstructive pulmonary disease, chronic heart failure, or chronic renal disease). Mortality after 28 days was higher among patients with a late peak of HMGB1 (OR 2.640; P=0.026). Co-existence of late peak and inflammatory disease synergistically impacted mortality (odds ratio of logistic regression analysis 3.17; P: 0.027). Late peak was concomitantly associated with higher values of ferritin (P=0.035), and IFNγ (P=0.002) among patients with hyperferritinemia. It is concluded that late HMGB1 peak was associated with worse prognosis, especially in patients with underlying chronic inflammatory conditions. © 2019 by the Shock Society. The unavoidable diet change in emerging countries, projected for the coming years, will significantly increase the global consumption of animal protein. It is expected that Brazilian livestock production, responsible for close to 15% of global production, be prepared to answer to the increasing demand of beef. Consequently, the evaluation of pasture quality at regional scale is important to inform public policies towards a rational land use strategy directed to improve livestock productivity in the country. Our hypothesis is that MODIS images can be used to evaluate the processes of degradation, restoration and renovation of tropical pastures. To test this hypothesis, two field campaigns were performed covering a route of approximately 40,000 km through nine Brazilian states. To characterize the sampled pastures, biophysical parameters were measured and observations about the pastures, the adopted management and the landscape were collected. Each sampled pasture was evaluated using a time series of MODIS EVI2 images from 2000?2012, according to a new protocol based on seven phenological metrics, 14 Boolean criteria and two numerical criteria. The theoretical basis of this protocol was derived from interviews with producers and livestock experts during a third field campaign. The analysis of the MODIS EVI2 time series provided valuable historical information on the type of intervention and on the biological degradation process of the sampled pastures. Of the 782 pastures sampled, 26.6% experienced some type of intervention, 19.1% were under biological degradation, and 54.3% presented neither intervention nor trend of biomass decrease during the period analyzed.

The Brazilian Cerrado is one of the world´s biodiversity hotspot and hosts some of the most intensive agricultural activities for food production in the world. The objective of this study was to produce a land-use and land-cover (LULC) map of the Cerrado based on Landsat-8 Operational Land Imager (OLI) images. A set of 121 scenes from 2013 was processed using the image segmentation technique. The segments were exported in the shapefile format and interpreted visually in a geographical information system software using RGB/564 color composites. The following LULC classes were considered: annual croplands, perennial croplands, cultivated pasturelands, reforestation, mosaic of occupation, urban areas, mining areas, bare soil, forestlands, non-forestlands, water bodies, and non-identified (clouds and burned areas). The overall accuracy was estimated by an independent scientist with large experience in Cerrado´s image interpretation. The results showed that 43.4% of the study area (88.5 million hectares) were already converted into agricultural, urban and mining areas, 54.6% (111 million hectares) were still natural areas, and 1.9% (3.9 million hectares) was classified as non-identified. Cultivated pasturelands were the most representative land-use type (29.5%), followed by annual croplands (8.5%) and perennial croplands (3.1%). The overall accuracy of the final map was 80.2%.
