Abstract The tumor suppressor gene adenomatous polyposis coli (APC) is the initiating mutation in approximately 80% of all colorectal cancers (CRC), underscoring the importance of aberrant regulation of intracellular WNT signaling in CRC development. Recent studies have found that early-onset CRC exhibits an increased proportion of tumors lacking an APC mutation. We set out to identify mechanisms underlying APC mutation-negative (APC mut– ) CRCs. We analyzed data from The Cancer Genome Atlas to compare clinical phenotypes, somatic mutations, copy number variations, gene fusions, RNA expression, and DNA methylation profiles between APC mut– and APC mutation-positive (APC mut+) microsatellite stable CRCs. Transcriptionally, APC mut– CRCs clustered into two approximately equal groups. Cluster One was associated with enhanced mitochondrial activation. Cluster Two was strikingly associated with genetic inactivation or decreased RNA expression of the WNT antagonist RNF43, increased expression of the WNT agonist RSPO3, activating mutation of BRAF, or increased methylation and decreased expression of AXIN2. APC mut– CRCs exhibited evidence of increased immune cell infiltration, with significant correlation between M2 macrophages and RSPO3. APC mut– CRCs comprise two groups of tumors characterized by enhanced mitochondrial activation or increased sensitivity to extracellular WNT, suggesting that they could be respectively susceptible to inhibition of these pathways.

5. A multiple optical tracking based approach for enhancing hand-based interaction in virtual reality simulations

Research exploring natural virtual reality interaction has seen significant success in optical tracker-based approaches, enabling users to freely interact using their hands. Optical based trackers can provide users with real-time, high-fidelity virtual hand representations for natural interaction and an immersive experience. However, work in this area has identified four issues: occlusion, field-of-view, stability and accuracy. To overcome the four key issues, researchers have investigated approaches such as using multiple sensors. Research has shown multi-sensor-based approaches to be effective in improving recognition accuracy. However, such approaches typically use statically positioned sensors, which introduce body occlusion issues that make tracking hands challenging. Machine learning approaches have also been explored to improve gesture recognition. However, such approaches typically require a pre-set gesture vocabulary limiting user actions with larger vocabularies hindering real-time performance. This thesis presents an optical hand-based interaction system that comprises two Leap Motion sensors mounted onto a VR headset at different orientations. Novel approaches to the aggregation and validation of sensor data are presented. A machine learning sub-system is developed to validate hand data received by the sensors. Occlusion detection, stability detection, inferred hands and a hand interpolation sub-system are also developed to ensure that valid hand representations are always shown to the user. In addition, a mesh conformation sub-system ensures 3D objects are appropriately held in a user’s virtual hand. The presented system addresses the four key issues of optical sessions to provide a smooth and consistent user experience. The MOT system is evaluated against traditional interaction approaches; gloves, motion controllers and a single front-facing sensor configuration. The comparative sensor evaluation analysed the validity and availability of tracking data, along with each sensors effect on the MOT system. The results show the MOT provides a more stable experience than the front-facing configuration and produces significantly more valid tracking data. The results also demonstrated the effectiveness of a 45-degree sensor configuration in comparison to a front-facing. Furthermore, the results demonstrated the effectiveness of the MOT systems solutions at handling the four key issues with optical trackers.

Arbitration is often hampered by obstacles to the taking of evidence, either because one party fails to produce relevant documents when requested or the documents are held by a third party outside the tribunal’s powers. Parties engaged in arbitration seated in Mainland China are constrained by the Chinese state court’s limited powers to assist in evidence taking. This article considers the wider scope of options for the taking of evidence in arbitrations seated in Mainland China. The first port of call may be to seek an order from the arbitral tribunal to impose sanctions within the arbitration, such as adverse inferences or adverse cost orders. If the arbitral tribunal cannot compel the recalcitrant party or a third party to produce documents or other evidence, the party may seek assistance from the court at the arbitral seat or a foreign court connected to the arbitration. This article compares the options for state court assistance in evidence taking available in the state courts of Mainland China, England and Wales, Hong Kong, and the United States. Practitioners should be aware that the powers of state courts to assist in evidence taking in international arbitration varies widely between these jurisdictions, from allowing only orders for preservation of key evidence in Mainland China to wide-ranging discovery from third parties by way of Section 1782 applications in the Unitead States.

Recent studies have found that early-onset CRC exhibits an increased proportion of tumors lacking an APC mutation. We set out to identify regulatory mechanisms underlying APC mutation-negative (APCmut-) CRCs. Methods: We analyzed data from The Cancer Genome Atlas to compare clinical phenotypes, somatic mutations, copy number variations, gene fusions, RNA expression, and DNA methylation profiles between APCmut- and APC mutation-positive (APCmut+) microsatellite stable CRCs. We also constructed and compared regulatory networks between APCmut- and APCmut+ CRCs by integrating transcription factor (TF) binding motifs, protein-protein interactions, and gene co-expression using the algorithms PANDA and LIONESS. Single sample networks were used to identify each tumor’s contribution to the differences in transcriptional regulation observed between the APCmut- and APCmut+ groups. Results: APCmut- CRC expression profiles clustered into two approximately equal groups. Cluster One was associated with enhanced mitochondrial activation. Cluster Two was strikingly associated with genetic inactivation or decreased RNA expression of the WNT antagonist RNF43, increased expression of the WNT agonist RSPO3, activating mutation of BRAF, or increased methylation and decreased expression of AXIN2. APCmut- CRCs exhibited evidence of increased immune cell infiltration, with significant correlation between M2 macrophages and RSPO3. Consistent with this, APCmut-CRC regulatory networks showed strong activation of immune pathways relative to APCmut+ CRCs. The second most prominently targeted pathway was RAS signaling, with the TFs TCF15 and RUNX2 regulating key pathway members including RAF1, GEFs, and GAPs. Single sample networks revealed that tumors with the strongest transcriptional regulation of RAS genes tend to lack a KRAS mutation and exhibit a slight skew towards earlier onset. Conclusions: APCmut- CRCs comprise two groups of tumors characterized by enhanced mitochondrial activation or increased sensitivity to extracellular WNT, suggesting that they could be respectively susceptible to inhibition of these pathways. APCmut- tumors exhibit increased regulation of RAS pathway genes compared to APCmut+ tumors. The samples contributing most to this regulatory signature tend to lack a KRAS mutation, suggesting that APCmut- tumors have developed ways to modulate RAS pathway activity through non-mutational mechanisms. Focal emphasis on the RAF1 gene suggests these tumors may be more vulnerable to inhibition of the downstream MAPK pathway. Network analysis implicates TCF15 and RUNX2 as regulators of RAS pathway genes, suggesting a possible WNT-RAS crosstalk occurring in APCmut- CRC that warrants further investigation. In the summer of 1998 the Yangtze River in China suffered its worst floods in 50 years. In response to this flooding the Chinese government devised the National Forest Protection Plan, a key component of this plan is the Tuigeng huanlin (Sloping Land Conversion) policy. Under this scheme, farmers are offered grain subsidies and small cash payments in compensation for voluntarily returning marginal farmland to forest. This article looks at the initial demonstration stage of Tuigeng huanlin in the southwest and northwest of China and focuses on rural households: • their participation in the policy, • the choices made in terms of tree species (economic or ecological), and • the effect of participation on incomes and livelihoods. It also examines the stability of the policy and the impact these aspects will have on the potential of the Sloping Land Conversion Policy (SLCP) to attain its objectives of environmental protection and increased forest cover in China. Employees make decisions every day about whether to contribute to others--and their willingness to help is crucial to group and organizational effectiveness. But in a competitive, often zero-sum, world of work, generosity can be a dangerous path. How can leaders foster it without cutting into productivity, undermining fairness, and allowing employees to become doormats? The key, explains Wharton's Adam Grant, is to help givers reach a more nuanced understanding of what generosity is and is not. They'll be better positioned for sustainable giving when they can distinguish generosity from three attributes that often travel with it: timidity, availability, and empathy. Givers can overcome timidity, Grant says, by learning to act as agents--using "relational accounts" to advocate for others while negotiating for themselves. They can set boundaries on when, how, and whom to help. And they can strive to be perspective takers, not just empathizers, gathering knowledge about others that can lead to more-productive allocations of time that will benefit the organization as a whole.
