Abstract

Breast cancer is a chronic health problem that causes 690,000 deaths worldwide. The development of secondary metabolite compounds from natural preparations through an in silico approach is needed as a predictive tool to prevent breast cancer, one of them is cladosporol from Cladosporium spp. This study aims to utilize an in silico approach to predict the potential of cladosporol against alpha-estrogen receptors. The alpha-estrogen receptor with code 6CBZ was selected based on group function as pharmacophore in ligand-receptor interaction. The methods used in this study are by using an in silico approach with Molegro Virtual Docker (MVD) Ver 5.5 for the docking process and CABS-flex 2.0 for identifying the stability of the complexes. ADMET properties analysis was also performed to know the pharmacokinetics attributes of cladosporol. Based on research conducted, stated that cladosporol octanoate has the lowest rerank score with a -84.3593 value and the RMSD value is 1.195 Å so it's valid for molecular docking. Exploration of cladosporol for anti-breast cancer from Cladosporium spp fungi can be a novelty for the development of future pharmaceutical research. Thus, the development of anticancer drugs for early prevention can be carried out to reduce the number of breast cancer cases worldwide. [ABSTRACT FROM AUTHOR]

Abstract

The xenicane-type diterpene 13-epi-9-deacetylxenicin (1) was isolated from polar extract of the soft coral Asterospicularia laurae. The structure of 13-epi-9-deacetylxenicin was determined by X-ray diffraction analysis, and the absoulte configuration was determined using the modified Mosher′s method. Eleven acylation products of 13-epi-9-deacetylxenicin were prepared: 13-epi-9-O-p-anisoylxenicin (2), 13-epi-9-O-2-furoylxenicin (3), 13-epi-9-O-2-thiophenecarbonylxenicin (4), 13-epi-9-O-2-benzoylxenicin (5), 13-epi-9-O-p-toluoylxenicin (6), 13-epi-9-O-4-chlorobenzoylxenicin (7), 13-epi-9-O-4-fluorobenzoylxenicin (8), 13-epi-9-O-4-propylbenzoylxenicin (9), 13-epi-9-O-4-nitrobenzoylxenicin (10), and 13-epi-9-O-cinnamoylxenicin (11) are new compounds, and one known compound, 13-epixenicin (12). The structures of the new metabolites were determined on the basis of extensive spectroscopic analysis, particularly mass spectroscopy and 2D NMR (1H–1H COSY, HMQC, HMBC, and NOESY) spectroscopy. Methods have been developed for the synthesis of acyl and diacyl derivatives of 1′H-spiro[cyclo-heptane-1,2′-naphthalenes] and 2′-substituted spiro[cycloheptane-1,5′-naphtho[1,2-d][1,3]oxazin]-4′(6′H)-ones starting from ethyl 4′-amino-1′H-spiro[cycloheptane-1,2′-naphthalene]-3′-carboxylate (1) through variation of the amount of reactants, solvent nature, and reaction conditions. The reaction of amino ester 1 with hydrazine hydrate gave benzo[g]indazole derivative. 3-Amino-2-phenyl-3H-spiro[benzo[h]quinazolin-5,1′-cycloheptan]-4(6H)-one (8) and 3-amino-1H-spiro[benzo[h]quinazoline-5,1′-cycloheptane]-2,4(3H,6H)-dione (13) were syn-thesized and subjected to acylation with acetyl and benzoyl chlorides to obtain the corresponding amides. Abstract

Lipophilic marine toxins (LMTs) are made up of multiple groups of toxic analogues, which are characterised by different levels of cellular and toxic action. The most prevalent groups in the southern Pacific zone are: a) okadaic acid group (OA-group) which consists of okadaic acid (OA) and dinophysistoxin-1 (DTX-1); and, b) pectenotoxin-2 (PTX2) group which consists of pectenotoxin-2 (PTX-2). The main objective of our study was to examine in vitro biotransformation of OA-group and PTX-group in the tissues of two endemic species of bivalves from southern Chile; blue mussels (Mytilus chilensis) and clams (Ameghinomya antiqua). The biotransformation processes of both groups were only detected in the digestive glands of both species using LC-MS/MS. The most frequently detected analogues were acyl derivatives (≈2.0 ± 0.1 μg ml−1) for OA-group and PTX-2SA (≈1.4 ± 0.1 μg ml−1) for PTX-group, with a higher percentage of biotransformation for OA-group (p <.001). In addition, simultaneous incubations of the different analogues (OA/PTX-2; DTX-1/PTX-2 and OA/DTX-1/PTX-2) did not show any interaction between the biotransformation processes. These results show that the toxicological variability of endemic species leads to biotransformation of the profile of toxins, so that these new analogues may affect people's health. Antimicrobial peptides are essential components of innate defense mechanisms and make promising candidates for novel anti-infective agents. The advantages of these peptides in clinical applications include their potential for broad-spectrum and rapid bactericidal activities, and low propensity for resistance development, whereas possible disadvantages include their high cost, limited stability, and unknown toxicology and pharmacokinetics. Granulysin (Gr) is a cytolytic and proinflammatory molecule expressed by activated human cytotoxic T lymphocytes and natural killer (NK) cells. This paper aims to study bacteriostatic and bactericidal activity against Mycobacterium tuberculosis by synthetic analogues of human Gr between 12 and 26 amino acids (AA) and their acyl derivatives. Considering results of previous studies, five new peptides were designed: a cyclic of 20 AA (Gr-SL1); one of 21 AA (linear) (Gr-SL2), another of 12 AA (cyclic) (Gr-SL3) and two lipopeptides (Gr-SL3-lauric and Gr-SL3-palmitic). Peptides were manually synthesized as C-terminal carboxamides by the solid-phase method following Fmoc chemistry. Gr synthetic analogues were purified by reverse phase HPLC and analyzed by analytical C18RP-HPLC and Maldi Tof. The antimycobacterial activity of synthesized Gr analogues was assessed using a microdilution susceptibility test as described previously. Although peptides studied here had neither higher antimycobacterial activity nor lower toxicity than analogs of human Gr previously evaluated, fresh knowledge concerning the influence of acylation and structural aspects analyzed will optimize the design of novel peptides combining the most favorable aspects for the maintenance of antimycobacterial activity with minimum toxicity. Monoacylated derivatives O-p-anisoyl- d-tartaric acid and its N,N-dimethylammonium salt are synthesized by the partial hydrolysis of O,O′-di- p-anisoyl- d-tartaric acid. Crystal and molecular structures of both compounds have been determined and analyzed. In both of them, some strong and moderate strength [O-H...O] hydrogen bonds exist between the carboxylic units. Abstract

On the basis of studies of the electronic absorption spectra and quantum-chemical calculations of the energies and bond orders of a series of N-acetyl-N-aryl substituted 2-amino-4,5, 6, 7-tetrahydro-1,3- thiazepines and N-aryl substituted 2-iminohexahydro-1,3-thiazepines it has been concluded that the reason for the weak basicity of the ring nitrogen atom of the former is the acceptor properties of the amide carbonyl which obstructs the interaction of the unshared pair (USP) of this atom with the phenyl ring; in the case of the derivatives of hexahydroazepine the strengthening of the basic properties of the exocyclic nitrogen atom is associated with the conjugation of its USP with the 71-electrons of the benzene ring, which is strengthening by delocalisation of the USP of the sp³ -hybridized ring nitrogen atom into the azomethine bond. Some methods of functionalization of closo -borate anions on the example of the interaction with carboxylic acids are considered. Oxonium derivatives of closo -decaborate anion [2-B 10 H 9 OC(OR)CH 3 ] − (R = H, Et) were prepared and transformed upon subsequent heating into 2,6-disubstituted derivative [2,6-B 10 H 8 (O 2 CCH 3 )] − . The obtained compounds were characterized by IR and NMR ( 1 H, 11 B, 13 C) spectroscopies. The crystal structure of (Bu 4 N)[2,6-B 10 H 8 (O 2 CCH 3 )] was determined by X-ray diffraction. Protonation of amides most often occurs on oxygen, unless the amide bond is twisted out of planarity allowing the more favored nitrogen protonation to occur. Protonation of oxygen is believed to take place over that of nitrogen due to increased resonance stabilization of the O-protonated species. Rapid progress in the synthetic application of benzotriazole derivatives in the last 20 years has resulted in over 1000 scientific papers on the subject. This fact is reflected in Section 5.01.7, which involves almost a half of the volume of this chapter. The section is arranged according to hybridization of the C-α atom and atomic numbers of the atoms in positions β and γ to allow an easy access to the material of interest. Recent discovery of copper catalysis in [3+2] cycloadditions of azides to acetylenes, the so-called ‘click chemistry’, which boosted application of the 1,2,3-triazole derivatives, especially in medicinal chemistry, is presented in Section 5.01.9. From the point of view of practical applications, Section 5.01.11 is organized according to the number, position, and combination of the substituents at the aromatic rings. Another novel feature that has no precedence in the previous editions of Comprehensive Heterocyclic Chemistry is an addition of triazole and benzotriazole complexes with various transitions metals to Section 5.01.4.

1.ÖZETBakteriyel İletişim İnhibitörü Olarak Tasarlanan Açil Homoserin Lakton AnaloglarınınPseudomonas aeruginosa Suşlarında Virülans Faktörlerinin Üretimine Etkilerinin AraştırılmasıAmaç:Çalışmamızda bakteriyel iletişim sistemi inhibitörü olarak sentezlenenN-açil homoserin lakton anoloğu moleküllerinPseudomonas aeruginosa PA01 suşunda virülans faktörlerinin üretimine ve biyofilm oluşumuna etkisinin araştırılması amaçlanmıştır.Gereçler ve Yöntemler: Çalışmamızda mikrodalga sentez reaktörü içerisinde sentezlenen 16 adetN-açil homoserin lakton anoloğu maddenin QSIS(Quorum Sensing Inhibitor Selector)1 biyosensör suşu kullanılarakçevreyi algılama sistemi (quorum sensing) üzerindeki inhibisyon etkisi test edilmiştir. Ayrıca bu maddelerin P. aeruginosa PA01 suşunda Elastin Congo Red testi elastaz, kloroform ekstraksiyonu yöntemi ile piyosiyaninüretimi analiz edilmiştir. Ayrıca mikroplak yöntemi ile sentezlenen maddelerin biyofilm oluşumuna etkileri test edilmiştir.Bulgular:Çalışılan 16N-açil homoserin lakton anoloğunun 11 tanesinin 2 mM derişimde QSIS1 biyosensör suşunda inhibitör etki gösterdiği görülmüştür. İnhibitör etki gösteren bu 11 maddenin P. aeruginosa PA01 suşunda elastaz ve piyosiyanin üretimi ile biyofilm oluşumunu da etkilediği gözlenmiştir. Elastaz aktivitesini % 28.18 (U151) ile % 64.06 (U153) arasında,piyosiyanin üretimini ise %26 (U139) ile %75.56 (U137)arasında değişen oranlarda azalttığı tespit edilmiştir. The structures of these compounds were confirmed by NMR, IR and elemental analysis. Quorum sensing inhibitor selector (QSIS) assay using biomonitor strain These compounds were also assessed for their capacities to prevent production of QS-regulated virulence factors such as elastase, pyocyanin and biofilm formation in P. aeruginosa PA01 strain.Results :some of 11 compunds were found to have anti-QS activities againstbiosensor bacteria (QSIS1) at 2mM concentration. These compounds were also found to significantly affect QS regulated elastase and pyocyanin production and biofilm formation in P. aeruginosa PA01 strain. Elastse activity U140 (%40.63) U141 (%46.88) U142 (%45.33) U153 (%35.54), pyocyanin activity U137 (%24.48) U138 (%37) U148 (%24.48) were found .Conclusion :These results of this study suggest that AHL analogues may provide a starting point for the design and development of new QS inhibitory drugs that restrict virulence of clinically important human pathogensKeywords: quorum sensing, cell-to-cell communication,AHL,Pseudomonas aeruginosa

ÖZET Bu çalışmada 2-hidroksi-l-naftaldehit (163), anilin ve p-metoksi anilinden naftolik Schiff bazları (165 a,b) hazırlanmıştır. Hazırlanan naftolik Schiff bazları NaBH4 ile indirgenmiş ve amin bileşikleri (166 a,b) elde edilmiştir. Amin bileşiklerinin (166 a,b) kloroasetilklorür ile reaksiyonu sonunda acil türevleri sentezlenmek istenmiş ancak acil türevleri yerine diaçil türevleri (167 a,b) elde edilmiştir. 1,4-naftoksazepinlerin (168 a,b) elde edilmesi için acil türevleri NaOH ile muamele edilmiştir. Reaksiyon sonunda halkalaşma ve 1,4- naftoksazepinler (168 a,b) sentezlenmiştir. Sentezlenen bileşiklerin yapıları İR, UV, *H-NMR, 13C-NMR (DEPT) ve kütle spektrumları ile tayin edilmiştir.
