Mycobacterium ulcerans ecovar Liflandii (Mu Liflandii) was identified as the causative agent of mycobacteriosis in a research colony of Zaire dwarf clawed frogs (Hymenochirus boettgeri) at the University of Michigan. Clinical presentation included lethargy, generalized septicemia, cutaneous granulomas, coelomic effusion, and acute mortality. Identification of the mycobacterial species was based on molecular, microbiological, and histopathologic characteristics. These findings indicate that Mu Liflandii is a primary cause of morbidity and mortality in Zaire dwarf clawed frogs and should be considered in the differential diagnosis of sepsis and coelomic effusion in amphibians. Mycobacterial speciation is important given the variability in pathogenesis within the family Mycobacteriaceae and the implications for both animal and human health as potential zoonoses. The Zaire dwarf clawed frog is a species common in the pet trade, and these findings provide consideration for this pathogen as a potentially important public health concern. This is the first report of Mu Liflandii infection in the genus Hymenochirus and illustrates the diagnostic challenges of differentiating among both mycolactone-producing mycobacteria and Mycobacterium marinum. Furthermore, we demonstrate the utility of environmental sampling for this pathogen within the tank system, suggesting this mode of sampling could replace the need for direct frog surveillance. Bovine tuberculosis (bTB) is a well-known zoonotic disease that affects cattle all over the world and results in significant economic loss, particularly in impoverished nations. The present communication describes the pathology, isolation, and molecular detection of Mycobacterium in an organized farm in Assam which has previous records on animals with confirmed M. bovis infection. During the period 2020-2021, a total of 40 animals (4 males and 36 females) of one year and above were included in the present study for screening of bovine tuberculosis by single intradermal comparative tuberculin test (SICCT). The milk, nasal swabs were collected from only tuberculin positive cattle and the tissue samples from necropsied animal and then processed for bacteriology, histopathology, and molecular detection from direct samples. Out of 40, four cows showed positive reactors by SICCT, and out of these four, one animal died. At necropsy, there was the presence of circumscribed yellowish-white lesions of various sizes and numbers. The smear prepared from granulomatous tissue samples showed the presence of acid-fast bacilli by Ziehl–Neelsen stain. Mycobacterium could be isolated from tissue samples. The DNA extracted from the samples could amplify Mycobacteria genus specific hsp65 gene and MTBC specific a 123-bp segment of the insertion sequence IS6110.

An approximately 25-year-old, female, yellow-naped Amazon parrot (Amazona auropalliata) was evaluated for a chronic, raised, ulcerative mass on the lateral aspect of the left thigh. Histopathology of an excisional biopsy revealed severe, chronic, multifocal-to-coalescing, ulcerated dermal and subcutaneous granulomas. No infectious organisms were observed on Ziehl-Neelsen or Gomori methenamine silver stains. The parrot was treated with oral sulfamethoxazoletrimethoprim and meloxicam. When reexamined 2 weeks later, the biopsy site had healed. Surgical biopsies were resubmitted 14 months after the original presentation due to recurrence of similar ulcerative lesions on the right leg. Histopathology revealed a similar inflammatory pattern, and hematoxylin-eosin, Ziehl-Neelsen, and silver stains on the biopsy samples were all negative. A Fite-Faraco stain revealed rare acid-fast bacilli throughout the lesion. Tissue polymerase chain reaction test was negative for Mycobacterium avium and Mycobacterium genavense. Mycobacterial culture and subsequent genotyping revealed Mycobacterium kansasii. Mycobacterium kansasii is a significant cause of mycobacteriosis in humans and, therefore, should be considered a potential zoonotic organism. Background Histoplasmosis (HP) is diagnosed by visualizing intracellular microorganisms in biopsy and/or culture. Periodic-acid Schiff (PAS) and Gomori methenamine silver (GMS) staining methods are routinely used for identification. The acid-fast property of Histoplasma was identified decades ago, but acid-fast staining has not been practiced in current surgical pathology. Awareness of the acid-fast property of Histoplasma, which is due to mycolic acid in the cell wall, is important in distinguishing Histoplasma from other infective microorganisms. Here, we examined acid-fastness in previously diagnosed cases of Histoplasma using the Ziehl-Neelsen (ZN) stain and correlated those findings with other known fungal stains. Methods All cases diagnosed as HP were retrieved and reviewed along with ZN staining and other fungal stains. We also stained cases diagnosed with Cryptococcus and Leishmania as controls for comparison. Results A total of 54 patients ranging in age from 11 to 69 years were examined. The most common sites of infection were the skin, adrenal tissue, and respiratory tract. Of the total 43 tissue samples, 20 (46.5%) stained positive with the ZN stain. In viable cases, a significant proportion of microorganisms were positive while necrotic cases showed only rare ZN-positive yeasts. In comparison to PAS and GMS stains, there was a low burden of ZN-positive yeasts. Cryptococcus showed characteristic ZN staining and all cases of Leishmania were negative. Conclusions Although the morphology of fungal organisms is the foundation of identification, surgical pathologists should be aware of the acid-fast property of fungi, particularly when there is the potential for confusion with other infective organisms.

Objective: This case report describes a clinical case of Johne's disease (JD) in a Friesian calf aging one and half years. Materials and methods: Physical examination of the calf was carried out, history of the farm as well as samples for laboratory examinations were obtained. The laboratory examinations included hematological examinations by direct wet mount, hematocrit centrifugation technique and Giemsa stain, biochemical evaluation of serum, bacterial culture and isolation from feces and aspirate from the swollen jaw. Results: Clinical history showed that the farm had a history of JD, and routine health screening revealed that one of the calves had clinical signs suggestive of Johne's disease. Physical examination of the calf revealed a lumpy jaw, enlarged bilateral pre-scapular and pre-femoral lymph nodes, while hematological and biochemical findings showed a normocytic normochromic anemia, severe leukocytosis with neutropilic left shift and lymphocytosis with hyperproteinemia characterized by hyperglobulinemia. Fecal bacterial tests showed the presence of acid fast bacilli. Based on the history and laboratory findings, the cow was diagnosed with JD. In order to ensure effective control measures, the calf was culled from the heard. Postmortem examination revealed inflamed pre-scapular and pre-femoral lymph nodes with the presence of paramphistomes in the rumen mucosa. Conclusion: Since JD is a chronic disease that lingers in livestock farms, there is need for early identification and culling of infected animal in order to limit its devastation on the farm. Background Tuberculous effusions are common. Classically, they are described as bacteria poor and lymphocyte rich. Our experience, however, has been more varied. We compiled this rare group of bacteria-positive tuberculous fluids to document their cytologic spectrum and to look for possible predictors of bacillary positivity. Methods Fifty-one cases of bacillary positive fluids were identified and their clinicopathological details were noted. Per case, the smear background was assigned as either clear, caseous, suppurative, granular proteinaceous or frankly hemorrhagic. Fine, punched-out vacuoles in the smear background were also noted. The bacillary load in each case was classified from scanty to 3+. Eventually, the clinicopathologic variables were tabulated for frequency and studied for any association with bacillary presence. Results Only 19 of the 51 patients had a history of tuberculosis. Retropositive patients comprised a small proportion (9.8%) and did not always indicate strong (3+) bacillary positivity. The granular proteinaceous background was the most frequent (35%) pattern. Only a suppurative background was associated with strong bacillary positivity. Fine vacuoles were seen almost always with caseous and granular proteinaceous backgrounds but without statistical significance. Conclusion Tuberculous effusions can have diverse smear backgrounds, not necessarily one rich in caseous material. When tuberculosis is known or clinically suspected, non-classical findings such as abundant neutrophils or suppurative background should not dissuade one from requisitioning mycobacterial stains.
