Chronic stable angina (CSA) remains prevalent, and the goal of treatment is control of symptoms and reduction in cardiovascular events. Ranolazine is a selective inhibitor of the late sodium current in myocytes with anti-ischemic and metabolic properties. It was approved by the US Food and Drug Administration in 2006 for use in patients with CSA. Multiple, randomized, placebo-controlled trials have shown that ranolazine improves functional capacity and decreases anginal episodes in CSA patients, despite a lack of a significant hemodynamic effect. Ranolazine did not improve cardiovascular mortality or affect incidence of myocardial infarction in the MERLIN (Metabolic Efficiency with Ranolazine for Less Ischemia in Non-ST-Elevation Acute Coronary Syndrome)-TIMI (Thrombolysis In Myocardial Infarction) 36 trial, but significantly decreased the incidence of recurrent angina. More recently, ranolazine has been shown to have beneficial and potent antiarrhythmic effects, both on supraventricular and ventricular tachyarrhythmias, largely due to its inhibition of the late sodium current. Randomized controlled trials testing these effects are underway. Lastly, ranolazine appears to be cost-effective due to its ability to decrease angina-related hospitalizations and improve quality of life. The article discusses the study which describes the areas of bank erosion that causes heavy loss of land by Brahmaputra river at the Kaziranga National Park in Assam,India. It uses geographic informations systems (GIS to analyze sequential changes in banklines of the river and geomorphology to characterize the irver, while changes in time over spaces are measured using Sol topographic maps. It shows that the main cause of erosion at the river is the loose non-cohesive sediments of the bank.

Abstract Background Viral diversity and abundance are defining properties of human immunodeficiency virus (HIV)-1's biology and pathogenicity. Despite the increasing availability of antiretroviral therapy, HIV-associated dementia (HAD) continues to be a devastating consequence of HIV-1 infection of the brain although the underlying disease mechanisms remain uncertain. Herein, molecular diversity within the HIV-1 non-structural gene, Vpr, was examined in RNA sequences derived from brain and blood of HIV/AIDS patients with or without HIV-associated dementia (HAD) together with the ensuing pathobiological effects. Results Cloned brain- and blood-derived full length vpr alleles revealed that amino acid residue 77 within the brain-derived alleles distinguished HAD (77Q) from non-demented (ND) HIV/AIDS patients (77R) (p < 0.05) although vpr transcripts were more frequently detected in HAD brains (p < 0.05). Full length HIV-1 clones encoding the 77R-ND residue induced higher IFN-α, MX1 and BST-2 transcript levels in human glia relative to the 77Q-HAD encoding virus (p < 0.05) but both viruses exhibited similar levels of gene expression and replication. Myeloid cells transfected with 77Q-(pVpr77Q-HAD), 77R (pVpr77R-ND) or Vpr null (pVpr(-))-containing vectors showed that the pVpr77R-ND vector induced higher levels of immune gene expression (p < 0.05) and increased neurotoxicity (p < 0.05). Vpr peptides (amino acids 70-96) containing the 77Q-HAD or 77R-ND motifs induced similar levels of cytosolic calcium activation when exposed to human neurons. Human glia exposed to the 77R-ND peptide activated higher transcript levels of IFN-α, MX1, PRKRA and BST-2 relative to 77Q-HAD peptide (p < 0.05). The Vpr 77R-ND peptide was also more neurotoxic in a concentration-dependent manner when exposed to human neurons (p < 0.05). Stereotaxic implantation of full length Vpr, 77Q-HAD or 77R-ND peptides into the basal ganglia of mice revealed that full length Vpr and the 77R-ND peptide caused greater neurobehavioral deficits and neuronal injury compared with 77Q-HAD peptide-implanted animals (p < 0.05). Conclusions These observations underscored the potent neuropathogenic properties of Vpr but also indicated viral diversity modulates innate neuroimmunity and neurodegeneration.

Human Pose Estimation (HPE) is a crucial step towards understanding people in images and videos. HPE provides geometric and motion information of the human body, which has been applied to a wide range of applications (e.g., human-computer interaction, motion analysis, augmented reality, virtual reality, healthcare, etc.). An extremely useful task of this kind is the 2D pose estimation of bedridden patients from infrared (IR) images. Here, the IR imaging modality is preferred due to privacy concerns and the need for monitoring both uncovered and covered patients at different levels of illumination. The major drawback of this research problem is the unavailability of covered examples, which are very costly to collect and time-consuming to label. In this work, a deep learning-based framework was developed for human sleeping pose estimation on covered images using only the uncovered training images. In the training scheme, two different image augmentation techniques, a statistical approach as well as a GAN-based approach, were explored for domain adaptation, where the statistical approach performed better. The accuracy of the model trained on the statistically augmented dataset was improved by 124 % as compared with the model trained on non-augmented images. To handle the scarcity of training infrared images, a transfer learning strategy was used by pre-training the model on an RGB pose estimation dataset, resulting in a further increment in accuracy of 4 %. Semi-supervised learning techniques, with a novel pose discriminator model in the loop, were adopted to utilize the unannotated training data, resulting in a further 3 % increase in accuracy. The recent advances in pigeon pea genomics, including high-quality whole genome and chloroplast genome sequence information helped develop improved varieties. However, a comprehensive Cajanus proteome, including the organelle proteome, is yet to be fully mapped. The spatial delineation of pigeon pea proteins at sub-cellular levels and inter-organelle communication could offer valuable insights into its defense mechanism against various stresses. However, the major bottleneck in the proteomic study is the lack of a suitable method of protein extraction and sample preparation compatible with two-dimensional gel electrophoresis (2D-PAGE), liquid chromatography-mass spectrometry (LCMS), or matrix-assisted laser desorption ionization-time of flight (MALDi-ToF). Our study introduces two efficient methods, one for isolating total proteins and another for organelle (chloroplast) proteins from various Cajanus spp. For total protein extraction, we have optimized a protocol using phenol in combination with a reducing agent (DTT) and protease inhibitor cocktail, also washing (6-7 times) with ice-cold acetone after overnight protein precipitation of total proteins. Our modified extraction method using phenol for total leaf protein yielded approximately 2-fold more proteins than the previously reported protocols from C. cajan (3.18 ± 0.11 mg/gm) and C. scarabaeoides (2.06 ± 0.08 mg/gm). We have also optimized a protocol for plastid protein extraction, which yielded 1.33 ± 0.25 mg/10 gm plastid proteins from C. cajan and 0.88 ± 0.19 mg/10 gm plastid proteins from C. scarabaeoides. The 2D-PAGE analysis revealed 678 ± 08 reproducible total protein spots from C. cajan and 597 ± 22 protein spots from C. scarabaeoides. Similarly, we found 566 ± 10 and 486 ± 14 reproducible chloroplast protein spots in C. cajan and C. scarabaeoides, respectively. We confirmed the plastid protein fractions through immunoblot analysis using antibodies against LHCb1/LHCⅡ type Ⅰ protein. We found both methods suitable for 2D-PAGE and mass spectrometry (MS). We developed a bio-cheminformatics method, exploring disease inhibition mechanisms using machine learning-enhanced quantitative structure-activity relationship (ML-QSAR) models and knowledge-driven neural networks. ML-QSAR models were developed using molecular fingerprint descriptors and the Random Forest algorithm to explore the chemical spaces of Chalcones inhibitors against diverse disease properties, including antifungal, anti-inflammatory, anticancer, antimicrobial, and antiviral effects. We generated and validated robust machine learning-based bioactivity prediction models (https://github.com/RatulChemoinformatics/QSAR) for the top genes. These models underwent ROC and applicability domain analysis, followed by molecular docking studies to elucidate the molecular mechanisms of the molecules. Through comprehensive neural network analysis, crucial genes such as AKT1, HSP90AA1, SRC, and STAT3 were identified. The PubChem fingerprint-based model revealed key descriptors: PubchemFP521 for AKT1 , PubchemFP180 for SRC , PubchemFP633 for HSP90AA1, and PubchemFP145 and PubchemFP338 for STAT3 , consistently contributing to bioactivity across targets. Notably, chalcone derivatives demonstrated significant bioactivity against target genes, with compound RA1 displaying a predictive pIC 50 value of 5.76 against HSP90AA1 and strong binding affinities across other targets. Compounds RA5 to RA7 also exhibited high binding affinity scores comparable to or exceeding existing drugs. These findings emphasize the importance of knowledge-based neural network-based research for developing effective drugs against diverse disease properties. These interactions warrant further in vitro and in vivo investigations to elucidate their potential in rational drug design. The presented models provide valuable insights for inhibitor design and hold promise for drug development. In 1998, Fields medallist Stephen Smale [Smale (1998) [1]] proposed his famous eighteen problems to the mathematicians of this century. The statement of his eighteenth problem is simple but very important. The statement of his problem is, "What are the limits of intelligence, both artificial and human?". To answer the limit of human intelligence, in this paper, we introduce cognitive-consequence space and cognitive-consequence topology, and mainly prove that deductive and non-deductive parts of a human mind will never be empty. It proves a human being will continue to think and solve problems using both deductive and non-deductive inferences as long as they are alive. Hence, we conclude that human intelligence is limitless. We also introduce cognitive closure, cognitive similarity distance, cognitive limit point, cognitive-continuous function, consequence ideal, consequence filter, Gödel's incompleteness black hole, and study related properties. We also provide suitable justifications to show that cognitive consequence topological space is not similar to that of any existing topological space because it connects cognitive space and consequence operator in one frame to find the limit of human intelligence. Moreover, we also provide justifications to state that artificial intelligence has limitations. Dunbar's number is the cognitive limit of human beings to maintain stable relationships with other individuals in their social networks, and it is found to be 150. It is based on the neocortex size of humans. Usually, Dunbar's number and related phenomena are studied from the perspective of an individual. Dunbar's number also plays a crucial role in evolutionary psychology and allied areas. However, no study done so far has considered a couple who are in a stable relationship as a system from the perspective of Dunbar's number and its hierarchy layers. In this paper, we study the impact of Dunbar's number and Dunbar's hierarchy from the perspective of a couple by studying mathematically the conjoint Dunbar graphs for a couple. The cost of romance is the loss of almost two people from one's support network when a human being enters into a new relationship. Thus, we obtain mathematically that there is no significant change in one's friendship if human beings spend negligible time with their partners. Also, along with marriage and friendship development, we attempt to assess how a person's social network structure holds up over the course of a romantic relationship. In ecology, foraging requires animals to expend energy in order to obtain resources. The cost of foraging can be reduced through kleptoparasitism, the theft of a resource that another individual has expended effort to acquire. Thus, kleptoparasitism is one of the most significant feeding techniques in ecology. The phenomenon of kleptoparasitism has garnered significant attention from scholars due to its substantial impact on the food chain. However, the proportionate amount of mathematical modelling to facilitate the analysis has made limited progress in the literature. This circumstance motivated us to develop mathematical models that could explain the population dynamics of the prey-predator food chain. This study explores a scenario with two predators and one prey, where one predator is a kleptoparasite and the other is a host. The energy depletion caused by the predator's counterattack subsequent to kleptoparasitism, notwithstanding the nonlethal nature of this antagonism, is an additional component incorporated into this model. It has been suggested that biologically viable equilibria must meet certain parametric conditions in order to exist and to be stable both locally and globally. This article delves deeply into the occurrences of various one-parametric bifurcations, such as saddle-node bifurcation, transcritical bifurcation, and Hopf bifurcation, as well as two-parametric bifurcations, such as Bautin bifurcation. A subcritical Hopf bifurcation happens when the growth rate of the first predator is relatively low, while a supercritical Hopf bifurcation occurs when the growth rate of the first predator is quite large, allowing for the coexistence of all three species. Apoptosis-inducing factor 1 (AIF1) overexpression is intimately linked to the sensitivity of yeast cells towards hydrogen peroxide or acetic acid. Therefore, studying the mechanism of AIF1 regulation in the cell would provide a significant understanding of the factors guiding yeast apoptosis. In this report, we show the time-dependent induction of AIF1 under hydrogen peroxide stress. Additionally, we find that AIF1 expression in response to hydrogen peroxide is mediated by two transcription factors, Yap5 (DNA binding) and Cdc73 (non-DNA binding). Furthermore, substituting the H3K36 residue with another amino acid significantly abrogates AIF1 expression. However, substituting the lysine (K) in H3K4 or H3K79 with alanine (A) does not affect AIF1 expression level under hydrogen peroxide stress. The traditional rice genotypes of Assam are considered to have biological value due to the presence of several bioactive compounds like flavonoids, polyphenols, and anthocyanins, which have antioxidant, anti-cancer, anti-diabetic, and anti-aging properties. The pigmented genotypes are considered to have high iron (Fe) content. However, the effect of Fe and Zinc (Zn) accumulation on anthocyanin content is yet to be studied in pigmented rice of Assam. We studied the Fe, Zn, and anthocyanin content in grains of 204 traditional rice of Assam, which are traditionally preferred for their nutraceutical properties. We performed phenotypic and biochemical compositional analyses of 204 genotypes to identify those having high Fe, Zn, and anthocyanin. We also carried out the differential expression of a few selected Fe and Zn transporter genes along with the expression of anthocyanin biosynthesis genes. Interestingly, all pigmented rice genotypes contained a higher amount of phenolic compound than the non-pigmented form of rice. We found the highest (32.73 g) seed yield per plant for genotype Jengoni followed by Kajoli chokuwa and Khau Pakhi 1. We also listed 30 genotypes having high levels of Fe and Zn content. The genotype Jengoni accumulated the highest (186.9 μg g -1 ) Fe, while the highest Zn (119.9 μg g -1 ) content was measured in genotype Bora (Nagaon), The levels of Ferritin 2 gene expression were found to be significantly higher in Bora (Nagaon) (> 2-fold). For Zn accumulation, the genotype DRR Dhan-45, which was released as a high Zn content variety, showed significant up-regulation of the ZIP4 gene at booting (> 7-fold), post-anthesis (7.8-fold) and grain filling (> 5-fold) stages followed by Bora (Nagaon) (> 3-fold) at post-anthesis. Anthocyanidin synthase gene, Flavanone 3-dioxygenase 1-like (FDO1), and Chalcone-flavanone isomerase-like genes were up-regulated in highly pigmented genotype Bora (Nagaon) followed by Jengoni. Based on our data there was no significant correlation between iron and zinc content on the accumulation of anthocyanin. This challenges the present perception of the higher nutritive value in terms of the micronutrient content of the colored rice of Assam. This is the first report on the detailed characterization of traditional rice genotypes inclusive of phenotypic, biochemical, nutritional, and molecular attributes, which would be useful for designing the breeding program to improve Fe, Zn, or anthocyanin content in rice., (© 2024. Plant extracts have been used to treat microbiological diseases for centuries. This study examined plant triterpenoids tormentic acid (TA) and 23-hydroxycorosolic acid (HCA) for their antibiofilm effects on Staphylococcus aureus strains (MTCC-96 and MTCC-7405). Biofilms are bacterial colonies bound by a matrix of polysaccharides, proteins, and DNA, primarily impacting healthcare. As a result, ongoing research is being conducted worldwide to control and prevent biofilm formation. Our research showed that TA and HCA inhibit S. aureus planktonic growth by depolarizing the bacterial membrane. In addition, zone of inhibition studies confirmed their effectiveness, and crystal violet staining and biofilm protein quantification confirmed their ability to prevent biofilm formation. TA and HCA exhibited substantial reductions in biofilm formation for S. aureus (MTCC-96) by 54.85% and 48.6% and for S. aureus (MTCC-7405) by 47.07% and 56.01%, respectively. Exopolysaccharide levels in S. aureus biofilm reduced significantly by TA (25 μg/mL) and HCA (20 μg/mL). Microscopy, bacterial motility, and protease quantification studies revealed their ability to reduce motility and pathogenicity. Furthermore, TA and HCA treatment reduced the mRNA expression of S. aureus virulence genes. In silico analysis depicted a high binding affinity of triterpenoids for biofilm and quorum-sensing associated proteins in S. aureus, with TA having the strongest affinity for TarO (- 7.8 kcal/mol) and HCA for AgrA (- 7.6 kcal/mol). TA and HCA treatment reduced bacterial load in S. aureus-infected peritoneal macrophages and RAW264.7 cells. Dunbar's number is the cognitive limit of an individual to maintain stable relationships with others in his network. It is based on the size of the neocortex of the human brain. On the other hand, trust is one of the major issues for one while selecting members for his social network and the evolution of his social network with time. Trust and Dunbar's number are interconnected in the case of one's stable social network. Trust needs time to be built after several social interactions, intimacy, etc. In this paper, we try to provide answers to the following important questions related to social networks: (i) Do trust levels remain the same for individuals from one's perspective in his social network when the network size increases? (ii) What is the relation between the power-law exponent α and the trust cutoff? (iii) Do trust levels help to diffuse information quickly or vice versa to reach Dunbar's number 150 along with hierarchy layers of 5, 15, and 50 individuals in networks of different sizes? We find that there is a requirement for trust levels to increase among the same individuals in one's social network if the size of the network increases. As a relation between the power-law exponent α and the trust cutoff, it is found that α ∝ 1/(trust cutoff). Memristive devices have garnered significant attention in the field of electronics over the past few decades. The reason behind this immense interest lies in the ubiquitous nature of memristive dynamics within nanoscale devices, offering the potential for revolutionary applications. These applications span from energy-efficient memories to the development of physical neural networks and neuromorphic computing platforms. In this research article, the angle toppling technique (ATT) was employed to fabricate titanium dioxide (TiO 2 ) nanoparticles with an estimated size of around 10 nm. The nanoparticles were deposited onto a 50 nm SiO x thin film (TF), which was situated on an n-type Si substrate. Subsequently, the samples underwent annealing processes at temperatures of 550 °C and 950 °C. The structural studies of the sample were done by field emission gun-scanning electron microscope (FEG-SEM) (JEOL, JSM-7600F). The as-fabricated sample exhibited noticeable clusters of nanoparticles, which were less prominent in the samples annealed at 550 °C and 950 °C. The element composition revealed the presence of titanium (Ti), oxygen (O 2 ), and silicon (Si) from the substrate within the samples. X-ray diffraction (XRD) analysis revealed that the as-fabricated sample predominantly consisted of the rutile phase. The comparative studies of charge storage and endurance measurements of as-deposited, 550 °C, and 950 °C annealed devices were carried out, where as-grown device showed promising responses towards brain computing applications. Furthermore, the teaching-learning-based optimization (TLBO) technique was used to conduct further comparisons of results.

Background: Bacillus thuringiensis (Bt) is a gram-positive ubiquitous saprophytic bacterium that produces proteins (Crystal protein, Vegetative insecticidal protein, and Secreted insecticidal protein) toxic to insects during its growth cycle. In the present study, the whole genome of a locally isolated B. thuringiensis strain BA04 was sequenced to explore the genetic makeup and to identify the genes responsible to produce insecticidal proteins including the virulence factors. The strain was isolated from the soil sample of the Kaziranga National Park, Assam, North-Eastern part of India (Latitude: 26°34'39.11''N and Longitude: 93°10'16.04''E)., Results: The whole genome sequencing (WGS) of the BA04 strain revealed that it has a circular genome of size 6,113,005 bp with four numbers of plasmids. A total of 6,111 genes including two novel crystal protein-encoding genes (MH753362.1 and MH753363.1) were identified. The BLASTn analysis of MH753362.1 showed 84% similarities (maximum identity) with Cry1Ia (KJ710646.1) gene, whereas MH753363.1 exhibited 66% identity with Insecticidal Crystal Protein (ICP)-6 gene (KM053257.1). At the protein level, MH753362.1 and MH753363.1 shared 79% identity with Cry1Ia (AIW52613.1) and 40% identity with Insecticidal Crystal Protein (ICP)-6 (AJW76687.1) respectively. Three-dimensional structures of these two novel protein sequences revealed that MH753362.1 have 48% structural similarity with Cry8ea1 protein, whereas MH753363.1 showed only 20% structural similarity with Cry4Aa protein. Apart from these insecticidal genes, the strain was also found to contain virulence and virulence-associated factors including the antibiotic resistance genes and Clustered regularly interspaced short palindromic repeat (CRISPR) sequences., Conclusion: This is the first report on the whole genome sequence of Bt strain BA04 isolated from Assam, a North-Eastern state of India. The WGS of strain BA04 unveils the presence of two novel types of insecticidal crystal protein-encoding genes which can be used for the development of insect-resistant transgenic crops. Additionally, the strain could be used for the formulations of effective biopesticides. The WGS provides the fastest and cheapest platform for a better understanding of the genetic makeup of a strain and helps to explore the role of virulence genes in pathogenicity against the insect host., (© 2023. The activity of a green tissue-specific promoter of the Rubisco small subunit gene from  Arabidopsis  ( AraSSU ) was studied using transgenic chickpea lines. We generated transgenic chickpea lines expressing an  AraSSU  promoter-driven  cry2Aa  gene through  the Agrobacterium- mediated transformation method. Lines with  AraSSU  expressed the gene in all green tissues at high levels (> 90 ng/mg of fresh weight tissue) compared to lines generated using  CaMV35S  (< 10 ng/mg FW). We used vertical cross sections of various tissues of homozygous progeny using microtome for immunolocalization. The immunolocalization showed the expression of the  cry2Aa  gene in the green mesophyll cells of the leaves of both  AraSSU  and CaMV35 chickpea lines. Moreover, the accumulation of  AraSSU- regulated Cry2Aa protein was also observed in vascular tissues, including enucleate sieve elements and their companion cells. However, no expression was observed in the roots of  AraSSU  lines. In the case of  CaMV35  lines, the transgene expression was observed in all the tissues. Since our data indicated that the  AraSSU  promoter is active in non-green tissues such as vascular bundles. Therefore, we validated this by RT-PCR. We found Cry2Aa RNA transcripts in leaves, stems without epidermis (for vascular tissues), and roots with and without epidermis. Hot chilli ('Bhut Jolokia') ( Capsicum chinense Jacq.) is the hottest chilli widely grown in the North-Eastern region of India for its high pungency. However, little information is available on its physiology, growth and developmental parameters including yield. Therefore, the present research was undertaken to study the physiological responses of Bhut Jolokia under elevated CO 2 ( e CO 2 ) and temperature. Two germplasms from two different agro-climatic zones (Assam and Manipur) within the North-East region of India were collected based on the pungency. The present study explored the interactive effect of e CO 2 [at 380, 550, 750 ppm (parts per million)] and temperature (at ambient, > 2 °C above ambient, and > 4 °C above ambient) on various physiological processes, and expression of some photosynthesis and capsaicin related genes in both the germplasms. Results revealed an increase (> 1-2 fold) in the net photosynthetic rate (P n ), carbohydrate content, and C: N ratio in 'Bhut Jolokia' under e CO 2 and elevated temperature regimes compared to ambient conditions within the germplasms. Gene expression studies revealed an up-regulation of photosynthesis-related genes such as Cs RuBPC2 ( Ribulose biphosphate carboxylase 2 ) and Cs SPS ( Sucrose phosphate synthase ) which, explained the higher P n under e CO 2 and temperature conditions. Both the germplasm showed better performance under CTGT-II (Carbon dioxide Temperature Gradient Tunnel having 550 ppm CO 2 and temperature of 2 °C above ambient) in terms of various physiological parameters and up-regulation of key photosynthesis-related genes. An up-regulation of the Cs  capsaicin synthase gene was also evident in the study, which could be due to the metabolite readjustment in 'Bhut Jolokia'. In addition, the cultivar from Manipur (cv. 1) had less fruit drop compared to the cultivar from Assam (cv. 2) in CTGT II. The data indicated that 550 ppm of e CO 2 and temperature elevation of > 2 °C above the ambient with CTGT-II favored the growth and development of 'Bhut Jolokia'. Specification of migratory cell fate from a stationary population is complex and indispensable both for metazoan development as well for the progression of the pathological condition like tumor metastasis. Though this cell fate transformation is widely prevalent, the molecular understanding of this phenomenon remains largely elusive. We have employed the model of border cells (BC) in Drosophila oogenesis and identified germline activity of an RNA binding protein, Cup that limits acquisition of migratory cell fate from the neighbouring follicle epithelial cells. As activation of JAK-STAT in the follicle cells is critical for BC specification, our data suggest that Cup, non-cell autonomously restricts the domain of JAK-STAT by activating Notch in the follicle cells. Employing genetics and Delta endocytosis assay, we demonstrate that Cup regulates Delta recycling in the nurse cells through Rab11GTPase thus facilitating Notch activation in the adjacent follicle cells.
