Major depressive disorder (MDD) is a recurrent episodic mood disorder that represents the third leading cause of disability worldwide. In MDD, several factors can simultaneously contribute to its development, which complicates its diagnosis. According to practical guidelines, antidepressants are the first-line treatment for moderate to severemajor depressive episodes. Traditional treatment strategies often follow a one-size-fits-all approach, resulting in suboptimal outcomes for many patients who fail to experience a response or recovery and develop the so-called "therapy-resistant depression". The high biological and clinical inter-variability within patients and the lack of robust biomarkers hinder the finding of specific therapeutic targets, contributing to the high treatment failure rates. In this frame, precision medicine, a paradigm that tailors medical interventions to individual characteristics, would help allocate themost adequate and effective treatment for each patientwhile minimizing its side effects. In particular, multi-omic studies may unveil the intricate interplays between genetic predispositions and exposure to environmental factors through the study of epigenomics, transcriptomics, proteomics, metabolomics, gut microbiomics, and immunomics. The integration of the flow of multi-omic information into molecular pathways may produce better outcomes than the current psychopharmacological approach, which targets singular molecular factors mainly related to the monoamine systems, disregarding the complex network of our organism. The concept of systembiomedicine involves the integration and analysis of enormous datasets generated with different technologies, creating a "patient fingerprint", which defines the underlying biological mechanisms of every patient. This review, centered on precision medicine, explores the integration of multi-omic approaches as clinical tools for prediction in MDD at a single-patient level. It investigates how combining the existing technologies used for diagnostic, stratification, prognostic, and treatment-response biomarkers discovery with artificial intelligence can improve the assessment and treatment of MDD. BIOACTIVE glasses , *T cells , *BONE substitutes , *COPPER , *IMMUNE response , *EOSINOPHILS , *FLOW cytometry - Abstract
Biomaterials are extensively used as replacements for damaged tissue with bioactive glasses standing out as bone substitutes for their intrinsic osteogenic properties. However, biomaterial implantation has the following risks: the development of implant-associated infections and adverse immune responses. Thus, incorporating metallic ions with known antimicrobial properties can prevent infection, but should also modulate the immune response. Therefore, we selected silver, copper and tellurium as doping for bioactive glasses and evaluated the immunophenotype and cytokine profile of human T-cells cultured on top of these discs. Results showed that silver significantly decreased cell viability, copper increased the T helper (Th)-1 cell percentage while decreasing that of Th17, while tellurium did not affect either cell viability or immune response, as evaluated via multiparametric flow cytometry. Multiplex cytokines assay showed that IL-5 levels were decreased in the copper-doped discs, compared with its undoped control, while IL-10 tended to be lower in the doped glass, compared with the control (plastic) while undoped condition showed lower expression of IL-13 and increased MCP-1 and MIP-1β secretion. Overall, we hypothesized that the Th1/Th17 shift, and specific cytokine expression indicated that T-cells might cross-activate other cell types, potentially macrophages and eosinophils, in response to the scaffolds. Proteomic analysis of extracellular vesicles presents several challenges due to the unique nature of these small membrane-bound structures. Alternative analyses could reveal outcomes hidden from standard statistics to explore and develop potential new biological hypotheses that may have been overlooked during the initial evaluation of the data. An analysis sequence focusing on deviating protein expressions from donors' primary cells was performed, leveraging machine-learning techniques to analyze small datasets, and it has been applied to evaluate extracellular vesicles' protein content gathered from mesenchymal stem cells cultured on bioactive glass discs doped or not with metal ions. The goal was to provide additional opportunities for detecting details between experimental conditions that are not entirely revealed with classic statistical inference, offering further insights regarding the experimental design and assisting the researchers in interpreting the outcomes. The methodology extracted a set of EV-related proteins whose differences between conditions could be partially explainable with statistics, suggesting the presence of other factors involved in the bioactive glasses' interactions with tissues. Outlier identification of extracellular vesicles' protein expression levels related to biomaterial preparation was instrumental in improving the interpretation of the experimental outcomes. Load Frequency Control (LFC) is an issue of top importance to ensure microgrids (MGs) safe and reliable operation in AC MGs. Primary frequency control of each energy source can be guaranteed in AC MGs in order to integrate other energy sources. This paper proposes a micro-hydro frequency control scheme, combining the control of a reduced dump load and the nozzle flow control of Pelton turbines operating in isolated from electricity grid. Some researchs have reported the integration of dump load and flow control methods, but they did not reduce the dump load value and adjust the nozzle flow linearly to the power value demanded by users, causing the inefficient use of water. Simulation results were obtained in Matlab®/Simulink® using models obtained from previous research and proven by means of experimental studies. The simulation of the proposed scheme shows that instantaneous frequency variation is about 2 % even for perturbations magnitude up to 12 %. The validation result shows a 60 % reduction in overshoot and settling time of frequency temporal behaviour operating the micro-hydro autonomously.

With the constant expansion of computational systems, the amount of data that requires durability increases exponentially. All data persistence must be replicated in order to provide high-availability and fault tolerance according to the surrogate application or use-case. Currently, there are numerous approaches and replication protocols developed supporting different use-cases. There are two prominent variations of replication protocols, generic protocols, and database specific ones. The two main techniques associated with generic replication protocols are the active and passive replication. Although generic replication techniques are fully matured and widely used, there are inherent problems associated with those protocols, namely: performance issues of the primary replica of passive replication and the determinism required by the active replication. Some of those disadvantages are mitigated by specific database replication protocols (e.g., using multi-master) but, those protocols do not allow a separation between logic and data and they can not be decoupled from the database engine. Moreover, recent strategies consider highly-scalable and fault tolerant distributed logging mechanisms, allowing for newer designs based purely on logs to power replication. To mitigate the shortcomings found in both active and passive replication mechanisms, but also in partial variations of these methods, this dissertation presents a hybrid replication middleware, SQLware. The cornerstone of the approach lies in the decoupling between the logical replication layer and the data store, together with the use of a highly scalable distributed log that provides fault-tolerance and high-availability. We validated the prototype by conducting a benchmarking campaign to evaluate the overall system’s performance under two distinct infrastructures, namely a private medium class server, and a private high performance computing cluster. Across the evaluation campaign, we considered the TPCC benchmark, a widely used benchmark in the evaluation of Online transaction processing (OLTP) database systems. Results show that SQLware was able to achieve 150 times more throughput when compared with the native replication mechanism of the underlying data store considered as baseline, PostgreSQL., This work was partially funded by FCT - Fundação para a Ciência e a Tecnologia, I.P., (Portuguese Foundation for Science and Technology) within project UID/EEA/50014/2019

ABSTRACT: The pressure of the oceans and rivers on the coastal territories presents itself as one of the greatest challenges to face during the 21st century. Climate change promotes the rise in the mean sea level and therefore increase erosion of the coast, imposes new paradigms in the approach to spatial planning, different treatments of coastal ecosystems and new architectural and urbanistic solutions that move towards a sense of adaptation and mitigation of a phenomenon that is becoming more common, so as to consecrate the safeguarding of people and goods that have been or will be established in these biomes and enhancing the activities and qualities inherent in these territories. Along Portugal, the coastal area between Cova do Vapor and Fonte da Telha, in Almada is the most vulnerable and it is also here that some of the most vulnerable communities lie, vulnerable not only regarding the natural dynamics but also in economic and social terms. Illegal genesis villages on the Portuguese coast are often linked to fishing communities which in certain areas have established themselves and expanded over the years, becoming considerable clusters but which have been increasingly more vulnerable over the last decades to the dynamic conditions of the natural environments in which they were established. This work intends to explore new approaches in architectonic and urban planning in an area of illegal coastal genesis (with all the legal and social issues associated with it), a common phenomenon along the Portuguese coast, in order to improve the living conditions and living conditions of its population and to improve their coexistence with the surrounding, fast changing natural environment. In the context of the CloudDBAppliance (CDBA) project, fault tolerance and high-availability are provided in layers: within each appliance, within a data centre and between datacentres. This paper presents the recovery mechanisms in place to fulfill the provision of high-availability within a datacentre. The recovery mechanism takes advantage of CDBA's in-middleware replication mechanism to bring failed replicas up-to-date. Along with the description of different variants of the recovery mechanism, this paper provides their comparative evaluation, focusing on the time it takes to recover a failed replica and how the recovery process impacts throughput., The research leading to these results has received funding from the European Union's Horizon 2020 - The EU Framework Programme for Research and Innovation 2014-2020, under grant agreement No. 732051 and by the ERDF - European Regional Development Fund through the Operational Programme for Competitiveness and Internationalization - COMPETE 2020.

My dissertation focuses on the analysis of Paul Ricoeur’s thinking on the ethical-political conflict. To begin with, it seems important to recognize the essential role of his philosophical anthropology, in which the human being is defined as a combination of subject/body, in contrast to the traditional cogito of the cartesian philosophy. The subject is seen as a dialogical structure between himself and his body, an integral cogito, which allows to understand the importance of the problem of evil. The evil or suffering must identify the individual in his act, as a holder of his freedom. These ideas are crucial to comprehend the ethical-political problem of this author, as well as to demonstrate the intimate relationship between ethics and politics, mediated by the relationship between moral and justice. The ethical-polítical problem facilitates the understanding of the relationship between ethics and moral, due to the fact that the human being wishes to live and relate with others in fair institutions. In fact, the ethical desire has to be submitted to the norms, which must not be set apart from its original ethical aim. Nowadays, in Ricoeur’s opinion, there is a need to separate the concepts of ethics and politics and consider them antagonic; however, they also have to be considered close. Ethics is reflected in politics, as politics is associated with a group of individuals who intend the well-living. The subject realizes the disproportion and the fragility of his act only when confronted with the duality between voluntary and involuntary actions. The novelty of the Ricoeur’s philosophy consists in the identification of the fragile construction of the subject and that the core of this structure is in it disproportion. In addition to that, the subject becomes vulnerable with the recognition of his fallibility. Thus, the human being needs the relationship with the world and the others, because it is the only possible way to reconstruct a new meaning in his identity. A deep analysis of the politics is of utmost importance nowadays, due to the fact that the modern society is aware that the major political systems are insufficient and require urgent changes. The political action and the system it relies on have to be analysed and altered, creating a structure without financial and technocratic assumptions. It is now evident that politics has to suffer rehabilitation, which is the only way to recover the society’s trust in the political action that cannot be subjected to other power. Therefore, it seems essential to me that the human action has to be responsibly re-centered in those who are represented, allowing the Ricoeur’s ethical intention to be realized: live well with others in fair institutions. I am convicted that this work may represent a hopeful future to any modern society.

- Abstract

Purpose: This dissertation was conducted to find out which are the motivations of Sporting CP supporters, and if any of these motivations influence positively their preference for Promoted Youth Players. Methodology used: In order to ascertain if the study variables influence Sporting supporters’ preferences towards Promoted Youth Players, was conducted a Structural Equation Model to measure the regression weights between all study variables and the second-order construct: “Preferences for Promoted Youth Players”. Firstly was conducted a model with the whole sample and also a second attempt only with Sporting members. Findings: This research did not found any significant relation between final study variables and the preferences for Promoted Youth Players: all of the research hypotheses were rejected. The final Structural Equation Model was built with three first-order constructs: “Identification”, “Motivations” and “Value Perception”. All other variables were rejected on the previous stages due to it reliability values. In the end, none of these three variables registered significant Regression weights to be accepted. It was proved that none of chosen research variables influence positively the preference of Sporting supporters towards Promoted Youth Players. Added value: There were not found any studies about the variable Promoted Youth Players. For this reason, this pioneer study can attract the interest of other scholars to research about this topic. The used model can be replicated to future researches about other club’s supporters or improved and used again to research about Sporting supporters’ preferences.

The theoretical and conceptual approach concerning social movements and its surroundings has been given special prominence lately. The economic and social crisis over Europe and some American countries reopened the debate on the protests and demands followed by movements. Characterized primarily by an operational guidance directed to obtain reality changes or maintenance of a particular type of context, social movements contain a mix of individuals with different motivations and objectives, which foresee an opportunity for collective action to try to achieve their intentions. The collective participation of people through these movements, translate into a catalog of repertoires of action, resources and different motivations, culminating with the construction of a modus operandi. To police, it matter, therefore, become wise in relation to social movements which often get involved in political protest, whilst maintaining public order and safety through planning and implementation of safety actions. It is highlighted the importance of police activity inherent to the control of collective action and, using a qualitative approach, we seek to present, specifically, a characterization of the Black Bloc.

EXTRACELLULAR vesicles , *REGENERATION (Biology) , *STROMAL cells , *REGENERATIVE medicine , *MUSCULOSKELETAL system diseases , *VESICLES (Cytology) - Abstract
Tissue regeneration is a hot topic in health sciences, particularly because effective therapies promoting the healing of several cell types are lacking, specifically those of the musculoskeletal system. Mesenchymal Stem/Stromal Cells (MSCs) have been identified as crucial players in bone homeostasis, and are considered a promising therapy for diseases such as osteoarthritis (OA) and Rheumatoid Arthritis (RA). However, some known drawbacks limit their use, particularly ethical issues and immunological rejections. Thus, MSCs byproducts, namely Extracellular Vesicles (EVs), are emerging as potential solutions to overcome some of the issues of the original cells. EVs can be modulated by either cellular preconditioning or vesicle engineering, and thus represent a plastic tool to be implemented in regenerative medicine. Further, the use of biomaterials is important to improve EV delivery and indirectly to modulate their content and secretion. This review aims to connect the dots among MSCs, EVs, and biomaterials, in the context of musculoskeletal diseases. [ABSTRACT FROM AUTHOR]
