Plants release a wealth of metabolites into the rhizosphere that can shape the composition and activity of microbial communities in response to environmental stress. The connection between rhizodeposition and rhizosphere microbiome succession has been suggested, particularly under environmental stress conditions, yet definitive evidence is scarce. In this study, we investigated the relationship between rhizosphere chemistry, microbiome dynamics, and abiotic stress in the bioenergy crop switchgrass grown in a marginal soil under nutrient-limited, moisture-limited, and nitrogen (N)-replete, phosphorus (P)-replete, and NP-replete conditions. We combined 16S rRNA amplicon sequencing and LC-MS/MS-based metabolomics to link rhizosphere microbial communities and metabolites. We identified significant changes in rhizosphere metabolite profiles in response to abiotic stress and linked them to changes in microbial communities using network analysis. N-limitation amplified the abundance of aromatic acids, pentoses, and their derivatives in the rhizosphere, and their enhanced availability was linked to the abundance of bacterial lineages from Acidobacteria, Verrucomicrobia, Planctomycetes, and Alphaproteobacteria. Conversely, N-amended conditions increased the availability of N-rich rhizosphere compounds, which coincided with proliferation of Actinobacteria. Treatments with contrasting N availability differed greatly in the abundance of potential keystone metabolites; serotonin and ectoine were particularly abundant in N-replete soils, while chlorogenic, cinnamic, and glucuronic acids were enriched in N-limited soils. Serotonin, the keystone metabolite we identified with the largest number of links to microbial taxa, significantly affected root architecture and growth of rhizosphere microorganisms, highlighting its potential to shape microbial community and mediate rhizosphere plant-microbe interactions.

Tall wheatgrass is cultivated for pasture in marginal areas around the world due to its tolerance to abiotic stress including seasonal droughts and floods, as well as saline and alkaline soils. However, there is concern about its biomass productivity, which often does not exceed that of natural pastures. In this work we hypothesize that tall wheatgrass is a plant more sensitive to low P availability than commonly assumed. A glasshouse sand culture experiment was conducted to analyze morpho-physiological responses of tall wheatgrass to different P availability levels: 1 µmol L−1 phosphorus (severe deficiency, P1), 10 µmol L−1 phosphorus (representative value of soil solution in the region, P10) and 500 µmol L−1 phosphorus (full P supply, P500). Our results show that both limiting P conditions strongly decreased plant development and biomass accumulation, and altered above- and below-ground morphology, in comparison with P500 plants. The latter, in turn, had relative growth rate values comparable to those of other cultivated grasses under non-limiting conditions. In both P-deficient treatments, plant development was severely delayed starting from leaf number 3, even when P concentration in the pseudostem at early stages was similar or even higher than that of P500 plants. Our results suggest that developmental inhibition of tall wheatgrass at low P supply may be the consequence of a specific plant response to P concentration in soil solution, rather than to direct P starvation in plant tissues, and underlines the need of improvement of this species to enhance productivity in this environment. Background: Squamosa promoter-binding protein-like (SPL) is a plant-specific transcription factor that is widely involved in the regulation of plant growth and development, including flower and grain development, stress responses, and secondary metabolite synthesis. However, this gene family has not been comprehensively evaluated in barley, the most adaptable cereal crop with a high nutritional value. Results: In this study, a total of 15 HvSPL genes were identified based on the Hordeum vulgare genome. These genes were named HvSPL1 to HvSPL15 based on the chromosomal distribution of the HvSPL genes and were divided into seven groups (I, II, III, V, VI, VII, and VIII) based on the phylogenetic tree analysis. Chromosomal localization revealed one pair of tandem duplicated genes and one pair of segmental duplicated genes. The HvSPL genes exhibited the highest collinearity with the monocotyledonous plant, Zea mays (27 pairs), followed by Oryza sativa (18 pairs), Sorghum bicolor (16 pairs), and Arabidopsis thaliana (3 pairs), and the fewest homologous genes with Solanum lycopersicum (1 pair). The distribution of the HvSPL genes in the evolutionary tree was relatively scattered, and HvSPL proteins tended to cluster with SPL proteins from Z. mays and O. sativa, indicating a close relationship between HvSPL and SPL proteins from monocotyledonous plants. Finally, the spatial and temporal expression patterns of the 14 HvSPL genes from different subfamilies were determined using quantitative real-time polymerase chain reaction (qRT-PCR). Based on the results, the HvSPL gene family exhibited tissue-specific expression and played a regulatory role in grain development and abiotic stress. HvSPL genes are highly expressed in various tissues during seed development. The expression levels of HvSPL genes under the six abiotic stress conditions indicated that many genes responded to stress, especially HvSPL8, which exhibited high expression under multiple stress conditions, thereby warranting further attention. Conclusion: In this study, 15 SPL gene family members were identified in the genome of Hordeum vulgare, and the phylogenetic relationships, gene structure, replication events, gene expression, and potential roles of these genes in millet development were studied. The frequencies and lengths of drought periods are increasing in subtropical and temperate regions worldwide. Epigenetic responses to water stress could be key for plant resilience to these largely unpredictable challenges. Experimental DNA demethylation, together with application of a stress factor is an appropriate strategy to reveal the contribution of epigenetics to plant responses to stress. We analysed leaf cytosine methylation changes in adult plants of the annual Mediterranean herb, Erodium cicutarium, in a greenhouse, after seed demethylation with 5‐Azacytidine and/or recurrent water stress. We used bisulfite RADseq (BsRADseq) and a newly reported reference genome for E. cicutarium to characterize methylation changes in a 2 × 2 factorial design, controlling for plant relatedness. In the long term, 5‐Azacytidine treatment alone caused both hypo‐ and hyper‐methylation at individual cytosines, with substantial hypomethylation in CG contexts. In control conditions, drought resulted in a decrease in methylation in all but CHH contexts. In contrast, the genome of plants that experienced recurrent water stress and had been treated with 5‐Azacytidine increased DNA methylation level by ca. 5%. Seed demethylation and recurrent drought produced a highly significant interaction in terms of global and context‐specific cytosine methylation. Most methylation changes occurred around genic regions and within Transposable Elements. The annotation of these Differentially Methylated Regions associated with genes included several with a potential role in stress responses (e.g., PAL, CDKC, and ABCF), confirming an epigenetic contribution in response to stress at the molecular level. Background: Invertases (INVs) are key enzymes in sugar metabolism, cleaving sucrose into glucose and fructose and playing an important role in plant development and the stress response, however, the INV gene family in passion fruit has not been systematically reported. Results: In this study, a total of 16 PeINV genes were identified from the passion fruit genome and named according to their subcellular location and chromosome position. These include six cell wall invertase (CWINV) genes, two vacuolar invertase (VINV) genes, and eight neutral/alkaline invertase (N/AINV) genes. The gene structures, phylogenetic tree, and cis-acting elements of PeINV gene family were predicted using bioinformatics methods. Results showed that the upstream promoter region of the PeINV genes contained various response elements; particularly, PeVINV2, PeN/AINV3, PeN/AINV5, PeN/AINV6, PeN/AINV7, and PeN/AINV8 had more response elements. Additionally, the expression profiles of PeINV genes under different abiotic stresses (drought, salt, cold temperature, and high temperature) indicated that PeCWINV5, PeCWINV6, PeVINV1, PeVINV2, PeN/AINV2, PeN/AINV3, PeN/AINV6, and PeN/AINV7 responded significantly to these abiotic stresses, which was consistent with cis-acting element prediction results. Sucrose, glucose, and fructose are main soluble components in passion fruit pulp. The contents of total soluble sugar, hexoses, and sweetness index increased significantly at early stages during fruit ripening. Transcriptome data showed that with an increase in fruit development and maturity, the expression levels of PeCWINV2, PeCWINV5, and PeN/AINV3 exhibited an up-regulated trend, especially for PeCWINV5 which showed highest abundance, this correlated with the accumulation of soluble sugar and sweetness index. Transient overexpression results demonstrated that the contents of fructose, glucose and sucrose increased in the pulp of PeCWINV5 overexpressing fruit. It is speculated that this cell wall invertase gene, PeCWINV5, may play an important role in sucrose unloading and hexose accumulation. Conclusion: In this study, we systematically identified INV genes in passion fruit for the first time and further investigated their physicochemical properties, evolution, and expression patterns. Furthermore, we screened out a key candidate gene involved in hexose accumulation. Background: Alfalfa (Medicago sativa L.) is an essential leguminous forage with high nutrition and strong adaptability. The TIFY family is a plant-specific transcription factor identified in many plants. However, few reports have been reported on the phylogenetic analysis and gene expression profiling of TIFY family genes in alfalfa. Result: A total of 84 TIFY genes belonging to 4 categories were identified in alfalfa, including 58 MsJAZs, 18 MsZMLs, 4 MsTIFYs and 4 MsPPDs, respectively. qRT-PCR data from 8 genes in different tissues revealed that most MsTIFY genes were highly expressed in roots. The expression of MsTIFY14 was up-regulated after different times in both thrips-resistant and susceptible alfalfa after thrips feeding, and the expression of the remaining MsTIFYs had a strong correlation with the time of thrips feeding. Different abiotic stresses, including drought, salt, and cold, could induce or inhibit the expression of MsTIFY genes to varying degrees. In addition, the eight genes were all significantly up-regulated by JA and/or SA. Interestingly, MsTIFY77 was induced considerably by all the biotic, abiotic, or plant hormones (JA or SA) except ABA. Conclusion: Our study identified members of the TIFY gene family in alfalfa and analyzed their structures and possible functions. Summary Arsenic poses a global threat to living organisms, compromising crop security and yield. Limited understanding of the transcriptional network integrating arsenic‐tolerance mechanisms with plant developmental responses hinders the development of strategies against this toxic metalloid. Here, we conducted a high‐throughput yeast one‐hybrid assay using as baits the promoter region from the arsenic‐inducible genes ARQ1 and ASK18 from Arabidopsis thaliana, coupled with a transcriptomic analysis, to uncover novel transcriptional regulators of the arsenic response. We identified the GLABRA2 (GL2) transcription factor as a novel regulator of arsenic tolerance, revealing a wider regulatory role beyond its established function as a repressor of root hair formation. Furthermore, we found that ANTHOCYANINLESS2 (ANL2), a GL2 subfamily member, acts redundantly with this transcription factor in the regulation of arsenic signaling. Both transcription factors act as repressors of arsenic response. gl2 and anl2 mutants exhibit enhanced tolerance and reduced arsenic accumulation. Transcriptional analysis in the gl2 mutant unveils potential regulators of arsenic tolerance. These findings highlight GL2 and ANL2 as novel integrators of the arsenic response with developmental outcomes, offering insights for developing safer crops with reduced arsenic content and increased tolerance to this hazardous metalloid. Pearl millet is a nutri-cereal that is mostly grown in harsh environments, making it an ideal crop to study heat tolerance mechanisms at the molecular level. Despite having a better-inbuilt tolerance to high temperatures than other crops, heat stress negatively affects the crop, posing a threat to productivity gain. Hence, to understand the heat-responsive genes, the leaf and root samples of two contrasting pearl millet inbreds, EGTB 1034 (heat tolerant) and EGTB 1091 (heat sensitive), were subjected to heat-treated conditions and generated genome-wide transcriptomes. We discovered 13,464 differentially expressed genes (DEGs), of which 6932 were down-regulated and 6532 up-regulated in leaf and root tissues. The pairwise analysis of the tissue-based transcriptome data of the two genotypes demonstrated distinctive genotype and tissue-specific expression of genes. The root exhibited a higher number of DEGs compared to the leaf, emphasizing different adaptive strategies of pearl millet. A large number of genes encoding ROS scavenging enzymes, WRKY, NAC, enzymes involved in nutrient uptake, protein kinases, photosynthetic enzymes, and heat shock proteins (HSPs) and several transcription factors (TFs) involved in cross-talking of temperature stress responsive mechanisms were activated in the stress conditions. Ribosomal proteins emerged as pivotal hub genes, highly interactive with key genes expressed and involved in heat stress response. The synthesis of secondary metabolites and metabolic pathways of pearl millet were significantly enriched under heat stress. Comparative synteny analysis of HSPs and TFs in the foxtail millet genome demonstrated greater collinearity with pearl millet compared to proso millet, rice, sorghum, and maize. In this study, 1906 unannotated DEGs were identified, providing insight into novel participants in the molecular response to heat stress. The identified genes hold promise for expediting varietal development for heat tolerance in pearl millet and similar crops, fostering resilience and enhancing grain yield in heat-prone environments. Background: Jackfruit (Artocarpus heterophyllus Lam.) is the world's largest and heaviest fruit and adapts to hot, humid tropical climates. Low-temperature injury in winter is a primary abiotic stress, which affects jackfruit growth and development. Therefore, breeding cold-resistant varieties and identifying the vital genes in the process of cold resistance are essential. The dehydration-responsive element binding (DREB) gene family is among the subfamily of the APETALA2/ethylene response factor transcription factor family and is significant in plant abiotic stress responses. Methods: In this study, a comparative analysis of the cold resistance property of 'GuangXi' ('GX') and 'Thailand' ('THA') jackfruit strains with different cold resistance characteristics was performed through chlorophyll fluorescence and transcriptome sequencing. Results: We found that differentially expressed genes (DEGs) are significantly enriched in the metabolic processes. Here, 93 DREB genes were identified in the jackfruit genome, and phylogenetic analysis was used to classify them into seven groups. Gene structure, conserved motifs, chromosomal location, and homologous relationships were used to analyze the structural characteristics of the DREB family. Transcriptomics indicated that most of the AhDREB genes exhibited down-regulated expression in 'THA.' The DEGs AhDREB12, AhDREB21, AhDREB29, and AhDREB34 were selected for quantitative real-time PCR, and the results showed that these genes also had down-regulated expression in 'THA.' Background: Phytochrome-interacting factors (PIFs) plays an important role in plants as hubs for intracellular signaling regulation. The PIF gene family has been identified and characterized in many plants, but alfalfa (Medicago sativa L.), an important perennial high-quality legume forage, has not been reported on the PIF gene family. Results: In this study, we presented the identification and characterization of five MsPIF genes in alfalfa (Medicago sativa L.). Phylogenetic analysis indicated that PIFs from alfalfa and other four plant species could be divided into three groups supported by similar motif analysis. The collinearity analysis of the MsPIF gene family showed the presence of two gene pairs, and the collinearity analysis with AtPIFs showed three gene pairs, indicating that the evolutionary process of this family is relatively conservative. Analysis of cis-acting elements in promoter regions of MsPIF genes indicated that various elements were related to light, abiotic stress, and plant hormone responsiveness. Gene expression analyses demonstrated that MsPIFs were primarily expressed in the leaves and were induced by various abiotic stresses. Conclusion: This study conducted genome-wide identification, evolution, synteny analysis, and expression analysis of the PIFs in alfalfa. Our study lays a foundation for the study of the biological functions of the PIF gene family and provides a useful reference for improving abiotic stress resistance in alfalfa. Background: Sulphotransferase (SOT) enzyme (encoded by a conserved family of SOT genes) is involved in sulphonation of a variety of compounds, through transfer of a sulphuryl moiety from 3'phosphoadenosine- 5'phosphosulphate (PAPS) to a variety of secondary metabolites. The PAPS itself is derived from 3'adenosine-5'phosphosulphate (APS) that is formed after uptake of sulphate ions from the soil. The process provides tolerance against abiotic stresses like drought and heat in plants. Therefore, a knowledge of SOT genes in any crop may help in designing molecular breeding methods for improvement of tolerance for drought and heat. Methods: Sequences of rice SOT genes and SOT domain (PF00685) of corresponding proteins were both used for identification of SOT genes in wheat and six related species (T. urartu, Ae. tauschii, T. turgidum, Z. mays, B. distachyon and Hordeum vulgare), although detailed analysis was conducted only in wheat. The wheat genes were mapped on individual chromosomes and also subjected to synteny and collinearity analysis. The proteins encoded by these genes were examined for the presence of a complete SOT domain using 'Conserved Domain Database' (CDD) search tool at NCBI. Results: In wheat, 107 TaSOT genes, ranging in length from 969 bp to 7636 bp, were identified and mapped onto individual chromosomes. SSRs (simple sequence repeats), microRNAs, long non-coding RNAs (lncRNAs) and their target sites were also identified in wheat SOT genes. SOT proteins were also studied in detail. An expression assay of TaSOT genes via wheat RNA-seq data suggested engagement of these genes in growth, development and responses to various hormones and biotic/abiotic stresses. Lentil (Lens culinaris Medik.) is a cool season legume crop that plays vital roles in food and nutritional security, mostly in the least developed countries. Lentil is often cultivated in dry and semi-dry regions, where the primary abiotic factor is drought, which negatively impacts lentil growth and development, resulting in a reduction of yield. To withstand drought-induced multiple negative effects, lentil plants evolved a variety of adaptation strategies that can be classified within three broad categories of drought tolerance mechanisms (i.e., escape, avoidance, and tolerance). Lentil adapts to drought by the modulation of various traits in the root system, leaf architecture, canopy structure, branching, anatomical features, and flowering process. Furthermore, the activation of certain defensive biochemical pathways as well as the regulation of gene functions contributes to lentil drought tolerance. Plant breeders typically employ conventional and mutational breeding approaches to develop lentil varieties that can withstand drought effects; however, little progress has been made in developing drought-tolerant lentil varieties using genomics-assisted technologies. This review highlights the current understanding of morpho-physiological, biochemical, and molecular mechanisms of lentil adaptation to drought stress. Introduction: Crop Wild Relatives (CWR) have great socioeconomic importance for humans harbouring a broad spectrum of diversity and being important elements of different habitats. Beta vulgaris subsp. maritima ((L.) Arcangeli), also known as sea beet, is an important CWR of cultivated beets (GP-1). The high adaptability of this taxon to different environmental conditions, as well as its tolerance/resistance to different biotic and abiotic stresses, makes it a vital source for sugar beet improvement. Aim of this work was to analyse the in situ and ex situ status of sea beet population conservation in Italy, so as to guide protection activities and new collecting missions. Methods: Geographical distribution data of populations were that occur in the wild and conserved in genebanks were assembled from different databases and submitted to data quality control. Distribution, habitat characterization, land cover and use of the involved sites were then evaluated to provide insight into the current condition of areas hosting this CWR diversity. The presence of populations within vs. outside Italian protected areas was also evaluated. A density analysis of the records was performed and the adequateness of sea beet ex situ conservation, in terms of number of conserved accessions, was finally estimated. Results: A collection of 138 B. vulgaris subsp. maritima high quality georeferenced records were obtained, mainly distributed into Mediterranean Biogeographic Region. About 22% of the considered populations occurs in protected sites of the Natura 2000 Network, while about 15% in areas recorded in the Official list of protected natural areas (EUAP). Occurrences within protected areas are founded primarily in natural environments, whereas those outside are mainly located in urban and cultivated areas. The comparison of distribution and density analysis results revealed the presence of several gaps between sites hosting in situ populations and sites where ex situ conserved accessions were originally collected. Discussion: Here presented data indicates that the protection status for sea beet in Italy can be considered only partially adequate; more proactive protection measures should be foreseen to increase the role of protected areas in safeguarding in situ conservation. Global climate change has significantly reduced the yield of many crops due to various abiotic stressors. These stressors include water-related issues such as drought and flooding, thermal changes like extremely low and high temperatures, salinity, and adverse soil pH conditions including alkalinity and acidity. Biostimulants have emerged as promising and effective tools for mitigating the damage caused by these abiotic stressors in plants, ultimately enhancing both the quantity and quality of crops. Biostimulants are naturally derived substances that include humic acid, protein hydrolysates, nitrogenous compounds, seaweed extracts, beneficial bacteria, and molds. Even at low concentrations, biostimulants play a critical role in activating important plant enzymes, inducing antioxidant defenses, improving water relations and photosynthetic activity, stimulating hormone-like activities (particularly auxins, gibberellins, and cytokinins), and modulating root system development. Purple-leafed plants not only have a higher resistance to biotic and abiotic stresses, but also have higher ornamental value. Anthocyanins are vital for leaf color formation, growth and development of purple leaves. However, the molecular mechanism underlying purple leaf formation in Lagerstroemia indica remains unclear. Metabolomic and transcriptomic analysis of purple-leafed cultivar 'Ebony Embers' and greenleafed cultivar 'Arapahoe' showed that the high expression of anthocyanin structure genes induced hyperaccumulation of cyanidin and pelargonidin derivatives, making the leaves purple. LfiHY5, LfiMYB75 and LfibHLH1 were identified using correlation analysis and weighted gene co-expression network analysis. In 'Arapahoe' x 'Ebony Embers' population, LfiHY5 and LfiMYB75 showed significant positive correlation with leaf anthocyanin content. Transient expression of LfiMYB75 and LfiHY5 in tobacco and purple-leafed crape myrtle indicated that the two genes activated anthocyanin synthesis. Yeast two-hybrid analysis showed that LfiMYB75 and LfibHLH1 could form a complex that enhanced anthocyanin synthesis. Yeast monohybrid and dual-luciferase assays confirmed that LfiHY5 activated the expression of LfiMYB75, to activate the transcription of anthocyanin structural genes LfiCHS and LfiANS. Moreover, there were three alleles of LfiHY5 in crape myrtle, and the different sequences had different activation effects on LfiMYB75. In conclusion, the results showed that LfiHY5 led to upregulate the transcription of LfiMYB75, and LfiMYB75 formed a complex with LfibHLH1, which increased the transcription level of LfiCHS and LfiANS to affect anthocyanin synthesis in crape myrtle. Sexual reproduction, growth, and survival are crucial demographic strategies for plant population viability. Here, we propose a conceptual model predicting demographic responses of species based on their ecological strategy and the heterogeneity of environmental conditions within a biogeographical unit and then applied it to a case study from a 5˚ latitudinal gradient in the Patagonian steppes. We also aim to disentangle genetic from environmental effects on demographic responses. We performed in situ and common garden experiments with two species from six local populations of the Occidental Phytogeographical District of the Patagonian steppes. Species differ in key ecological traits, and thus fit into Grime's model for evolutionary strategies in plants: one as competitive species and the other as stress‐tolerant species. We calculated population growth rate (λ) and performed elasticity analyses to compare the contribution of each demographic strategy to population fitness between species and among local populations distributed along 600 km latitudinal gradient with differences in mean annual precipitation (MAP). We highlight four results. First, the competitive species change from sexual reproduction to growth as MAP increases. Second, the stress‐tolerant species relied on growth and survival along the MAP gradient. Third, interannual variation in resource availability modulated demographic responses for both strategies. Fourth, based on the comparison of the in situ and common garden experiments, we submit that demographic responses were genetically driven. Our study shows that demographic responses can be roughly predicted by the ecological strategy across environmental gradients. We show that differences arise not only between species, but also were genetically driven differences within species among local populations. Scaling up plant‐level responses to population‐level dynamics allows for a process‐based understanding of current and future biogeographical species organization. The impact of climate change on plant growth and productivity is profound, resulting in significant stresses such as biotic and abiotic factors. Among the abiotic stresses, temperature extremes, nutrient imbalances, heavy metals, drought, and salinity stand out as prominent contributors to substantial crop yield losses globally, exacerbating food insecurity concerns. Various methods are currently being explored to enhance plants' resilience to these stresses. We undertook an extensive literature review to assess the current landscape of knowledge in Nano biotechnology and identify areas where existing research is deficient or underexplored. This examination focused on identifying gaps related to methodology, scope, applications, and specific challenges within nanobiotechnology. We also evaluated recent developments and emerging trends in the field, identifying areas where research interest is expanding but remains relatively uncharted. By narrowing our research scope, we aimed to pinpoint specific aspects of Nano biotechnology requiring further investigation or that have not been sufficiently addressed. One promising approach involves the use of nanoparticles, which have shown to positively influence plant performance under stressful conditions. Nanoparticles facilitate nutrient uptake by plants, combat plant diseases and pathogens, and enable the detection and monitoring of trace components in soil through signal absorption. Understanding the mechanisms by which nanoparticles assist plants in mitigating abiotic challenges holds great promise for developing enduring solutions. While reactive oxygen species (ROS) have been identified as key redox signaling agents contributing to aging process, which and how specific oxidants trigger healthy longevity remain unclear. This paper aimed to explore the precise role and signaling mechanism of superoxide (O 2 •−) in health and longevity. A tool for precise regulation of O 2 •− levels in vivo was developed based on the inhibition of superoxide dismutase 1 (SOD1) by tetrathiomolybdate (TM) in Caenorhabditis elegans (C. elegans). Then, we examined the effects of TM on lifespan, reproduction, lipofuscin accumulation, mobility, and stress resistance. Finally, the signaling mechanism for longevity induced by TM-O 2 •− was screened by transcriptome analysis and tested in sod-1 and argk-1 RNAi strains, sod-2 , sod-3 , and daf-16 mutants. TM promoted longevity in C. elegans with a concomitant extension of healthy lifespan as indicated by increasing fertility and mobility and reducing lipofuscin accumulation, as well as enhanced resistance to different abiotic stresses. Mechanically, TM could precisely regulate O 2 •− levels in nematodes via modulating SOD1 activity. An O 2 •− scavenger Mn(III)TBAP abolished TM-induced lifespan extension, while an O 2 •− generator paraquat at low concentration mimicked the life prolongation effects. The longevity in TM-treated worms was abolished by sod-1 RNAi but was not affected in sod-2 or sod-3 mutants. Further transcriptome analysis revealed arginine kinase ARGK-1 and its downstream insulin/insulin-like growth factor 1 signaling (IIS) as potential effectors for TM-O 2 •‾-induced longevity, and argk-1 RNAi or daf-16 mutant nullified the longevity. Climate change has intensified drought stress in plants, altering the chemical composition of medicinal plant seeds. Addressing the impact of drought stress is crucial, particularly for species like Cumin (Cuminum cyminum L.), a valuable aromatic medicinal plant. Developing drought-tolerant varieties and comparing the quantity and quality with parent genotypes is essential. This study aimed to investigate the effects of drought stress on the quantitative and qualitative traits of cumin seeds, including the content and amount of oil and essence, essence chemical composition, fatty acids, and oil body number in a synthetic cumin variety compared to selected parent genotypes over two cropping seasons. Drought stress significantly decreased seed yield, essence yield, oil yield, and oil body number, while increasing essence and oil percentages and oil body size. The synthetic variety outperformed parent genotypes across all studied traits under both normal irrigation and drought stress conditions. The improvement in seed yield of the synthetic cultivar, compared to the average yield of the parental genotypes under normal irrigation conditions and drought stress, was estimated to be 27.84 % and 84.04 %, respectively. Linoleic (53.08–60.03 %) and oleic (28.67–31.52 %) acids were identified as primary fatty acids in cumin. Under drought stress, the synthetic variety showed increased Polyunsaturated fatty acids (31.77 %), while parent genotypes exhibited a decrease. In drought stress condition, the synthetic variety consistently displayed higher levels of unsaturated fatty acids, and superior fatty acid indices (AI = 0.06, TI = 0.35, h/ H = 26.57 and HPI= 12.63), indicating enhanced oil quality compared to parent genotypes. The objectives encompassed gene identification, domain confirmation, physicochemical characterization, subcellular localization prediction, genomic organization analysis, gene duplication events, conserved motif identification, gene architecture analysis, phylogenetic connections, promoter regions' cis-acting regulatory components, and RNA sequencing-based expression profiling. The results revealed 51 B3 genes in bitter gourd, classified into ARF, REM, LAV, and RAV classes. Drought, as a primary environmental factor, imposes significant constraints on developmental processes and productivity of plants. PHDs were identified as stress‐responsive genes in a wide range of eukaryotes. However, the regulatory mechanisms governing PHD genes in maize under abiotic stress conditions are still largely unknown and require further investigation. Here, we identified a mutant, zmvil2, in the EMS mutant library with a C to T mutation in the exon of the Zm00001d053875 (VIN3‐like protein 2, ZmVIL2), resulting in premature termination of protein coding. ZmVIL2 belongs to PHD protein family. Compared to WT, zmvil2 mutant exhibited increased sensitivity to drought stress. Consistently, overexpression of ZmVIL2 enhances drought resistance in maize. Y2H, BiFC, and Co‐IP experiments revealed that ZmVIL2 directly interacts with ZmFIP37 (FKBP12‐interacting protein of 37). zmfip37 knockout mutants also exhibit decreased drought tolerance. Interestingly, we demonstrated that ZmABF4 directly binds to the ZmVIL2 promoter to enhance its activity in yeast one hybrid (Y1H), electrophoretic mobility shift assay (EMSA) and dual luciferase reporter assays. Therefore, we uncovered a novel model ZmABF4‐ZmVIL2/ZmFIP37 that promotes drought tolerance in maize. Overall, these findings have enriched the knowledge of the functions of PHD genes in maize and provides genetic resources for breeding stress‐tolerant maize varieties. As the global climate continues to change, plants will increasingly experience abiotic stress(es). Stomata on leaf surfaces are the gatekeepers to plant interiors, regulating gaseous exchanges that are crucial for both photosynthesis and outward water release. To optimise future crop productivity, accurate modelling of how stomata govern plant–environment interactions will be crucial. Here, we synergise optical and thermal imaging data to improve modelled transpiration estimates during water and/or nutrient stress (where leaf N is reduced). By utilising hyperspectral data and partial least squares regression analysis of six plant traits and fluxes in wheat (Triticum aestivum), we develop a new spectral vegetation index; the Combined Nitrogen and Drought Index (CNDI), which can be used to detect both water stress and/or nitrogen deficiency. Upon full stomatal closure during drought, CNDI shows a strong relationship with leaf water content (r2 = 0.70), with confounding changes in leaf biochemistry. By incorporating CNDI transformed with a sigmoid function into thermal‐based transpiration modelling, we have increased the accuracy of modelling water fluxes during abiotic stress. These findings demonstrate the potential of using combined optical and thermal remote sensing‐based modelling approaches to dynamically model water fluxes to improve both agricultural water usage and yields. The vermiremediation potential of Eudrilus eugeniae was assessed in heavy metals (HMs) contaminated soil in a microcosm experiment. In detail, efficacy of E. eugeniae was investigated in terms of reduction in HMs content, enhancement in the soil enzyme activities, synergistic correlation between the HMs and enzyme activities and bioaccumulation of HMs in the earthworm's biomass. Moreover, a seed germination assay was also conducted to assess the HMs toxicity in soil after remediation. The results showed that Eudrilus eugeniae was capable of decreasing the Cu, Zn, Mn, Cr, and Ni contents in the soil by 17.56–26.30% after 90 days of experimental trial. Introduction of E. eugeniae increased cellulase, amylase, polyphenol oxidase, peroxidase, urease, dehydrogenase, and catalase activities by 30.3–80.95% compared to controls. Earthworm addition led to a 12.98% increase in G-Mean and 2.89% increase in T-QSI values, indicating significant soil health improvement. PCA revealed the negative impact of HMs on synergistic enzyme activities during vermiremediation. HMs content in earthworm biomass increased 13-fold. Seed germination assay confirmed HMs reduction by the end of the experiment. Summary: MicroRNA169 (miR169) has been implicated in multi‐stress regulation in annual species such as Arabidopsis, maize and rice. However, there is a lack of experimental functional and mechanistic studies of miR169 in plants, especially in perennial species, and its impact on plant growth and development remains unexplored. Creeping bentgrass (Agrostis stolonifera L.) is a C3 cool‐season perennial turfgrass of significant environmental and economic importance. In this study, we generated both miR169 overexpression and knockdown transgenic creeping bentgrass lines. We found that miR169 acts as a positive regulator in abiotic stress responses but is negatively associated with plant growth and development, playing multiple critical roles in the growth and environmental adaptation of creeping bentgrass. These roles include differentiated spatial hormone accumulation patterns associated with growth and stress accommodation, elevated antioxidant activity that alleviates oxidative damage induced by stress, ion‐channelling membrane components for maintaining homeostasis under saline conditions, and potential cross‐talks with stress‐regulating transcription factors such as AsHsfA and AsWRKYs. Our results unravel the role of miR169 in modulating plant development and stress responses in perennial grass species. SUMMARY: Mangroves grow in tropical/subtropical intertidal habitats with extremely high salt tolerance. Trehalose and trehalose‐6‐phosphate (T6P) have an alleviating function against abiotic stress. However, the roles of trehalose in the salt tolerance of salt‐secreting mangrove Avicennia marina is not documented. Here, we found that trehalose was significantly accumulated in A. marina under salt treatment. Furthermore, exogenous trehalose can enhance salt tolerance by promoting the Na+ efflux from leaf salt gland and root to reduce the Na+ content in root and leaf. Subsequently, eighteen trehalose‐6‐phosphate synthase (AmTPS) and 11 trehalose‐6‐phosphate phosphatase (AmTPP) genes were identified from A. marina genome. Abscisic acid (ABA) responsive elements were predicted in AmTPS and AmTPP promoters by cis‐acting elements analysis. We further identified AmTPS9A, as an important positive regulator, that increased the salt tolerance of AmTPS9A‐overexpressing Arabidopsis thaliana by altering the expressions of ion transport genes and mediating Na+ efflux from the roots of transgenic A. thaliana under NaCl treatments. In addition, we also found that ABA could promote the accumulation of trehalose, and the application of exogenous trehalose significantly promoted the biosynthesis of ABA in both roots and leaves of A. marina. Ultimately, we confirmed that AmABF2 directly binds to the AmTPS9A promoter in vitro and in vivo. Taken together, we speculated that there was a positive feedback loop between trehalose and ABA in regulating the salt tolerance of A. marina. These findings provide new understanding to the salt tolerance of A. marina in adapting to high saline environment at trehalose and ABA aspects. Tomato (Solanum lycopersicum L.) belongs to the Solanaceae family and is considered an important vegetable worldwide. Its production has been challenged due to soil salinization and water shortages. Local tomato cultivars could be better adapted to salt stress. Based on this hypothesis, the present study was carried out to determine the variation in fourteen different tomato cultivars for salt tolerance. Seed germination, mean germination time, shoot length, and root length were examined under salinity stress (1.5 g L-1 NaCl). Plant growth and seed germination were severely affected by saline conditions. The results of this in vitro experiment showed that the seed germination of Yellow Milk was significantly increased (73.33%) however, Pink Jade and Red Jade cultivars were significantly decreased under the NaCl treatment. Similarly, among the fourteen tomato cultivars, the mean germination time of only Yellow Pearl and Black Current significantly increased. Moreover, the shoot length of eight tomato cultivars decreased compared with the control, while the highest shoot length (12.5 cm) was recorded in the case of Saint cultivar. The root length of Taiwan Red Saint, Purple Beauty, Yellow Milk, Red Pearl, Yellow Pearl, Pink Cooperative 906, Qinzu Shanghai 903, and Scarlet cultivars significantly increased, while the other five cultivars significantly decreased under NaCl treatment compared to control. In plants, the pathogenesis-related (PR) proteins have been identified as important regulators of biotic and abiotic stresses. PR proteins branch out into 19 different classes (PR1–PR19). Basically, all PR proteins display a well-established method of action, with the notable exception of PR1, which is a member of a large superfamily of proteins with a common CAP domain. We have previously isolated and characterized the first PR1 from durum wheat, called TdPR-1.2. In the current research work, TdPR1.2 gene was used to highlight its functional activities under various abiotic (sodium chloride (100 mM NaCl) and oxidative stresses (3 mM H2O2), hormonal salicylic acid (SA), abscisic acid (ABA) and jasmonic acid (JA), and abiotic stresses (Botrytis cinerea and Alternaria solani). Enhancement survival index was detected in Arabidopsis transgenic plants expressing TdPR1.2 gene. Moreover, quantitative real-time reverse transcription PCR (qRT-PCR) analysis demonstrated induction of antioxidant enzymes such as catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD). It equally revealed a decrease of malondialdehyde (MDA) as well as hydrogen peroxide (H2O2) levels in transgenic Arabidopsis plants compared to control lines, confirming the role of TdPR1.2 in terms of alleviating biotic and abiotic stresses in transgenic Arabidopsis plants. Eventually, RT-qPCR results showed a higher expression of biotic stress-related genes (PR1 and PDF1.2) in addition to a downregulation of the wound-related gene (LOX3 and VSP2) in transgenic lines treated with jasmonic acid (JA). Notably, these findings provide evidence for the outstanding functions of PR1.2 from durum wheat which can be further invested to boost tolerance in crop plants to abiotic and biotic stresses. Rich nutrients, climate resilience, and the economic importance of millets are believed to ensure food security for future generations. Millets have the habit of growing against abiotic stresses (particularly drought stress). Millets show much better climate resilience and nutrient supplementation properties compared to other major cereals. Understanding the molecular mechanisms of genes that respond to stresses and nutrient transport will help understand the tolerance mechanism and improve against both stresses. Genome sequences are currently available for two major (sorghum and pearl millet) and eight minor millets (foxtail millet, finger millet, kodo millet, barnyard millet, proso millet, job's tear, fonio millet, and tef). Five minor millets (little millet, kodo millet, brown top millet, guinea millet, and raishan) do not have genome sequences to date. Transcriptome studies help to identify differentially expressed genes (DEGs), mine genes induced in a particular stress and develop several molecular markers for all plants, including millets. Some millets have reports on transcriptome datasets for plants exposed to various biotic and abiotic stresses and for nutritional traits. Unfortunately, the transcriptome datasets of millets have not been adequately leveraged to explore genes associated with traits such as climate resilience, nutrient enrichment, and crop improvement. This underutilization stems from a lack of high-resolution studies and limited exploration within the field. As a result, the potential insights and genetic understanding offered by these datasets remain largely untapped. Through this review, we plan to elucidate the current status of transcriptome resources on millets and draw future insights on the utilization of these resources. This review summarizes recent progress in our current understanding of the mechanisms underlying the cell death pathways in bryophytes, focusing on conserved pathways and particularities in comparison to angiosperms. Regulated cell death (RCD) plays key roles during essential processes along the plant life cycle. It is part of specific developmental programmes and maintains homeostasis of the organism in response to unfavourable environments. Bryophytes could provide valuable models to study developmental RCD processes as well as those triggered by biotic and abiotic stresses. Some pathways analogous to those present in angiosperms occur in the gametophytic haploid generation of bryophytes, allowing direct genetic studies. The growth of bivoltine silk production in eastern and north-eastern india lags behind drastically in comparison to southern states due to non-availability of suitable bivoltine hybrids. The major constraints in bivoltine popularization are adverse climatic conditions. to overcome this limitation and to continuously rear bivoltine hybrids in these harsh climatic conditions throughout the year, the present proposal aims to identify robust and abiotic stress tolerant bivoltine silkworm breeds through integrating conventional breeding (directional selection) for improved productivity and advanced genome technologies such as MAS (marker assisted selection), gene specific markers for high humidity and thermo-tolerant DNA markers (Pyx3, Pyx4, S0803 and S0816) for tolerance to high temperature and high humidity and other abiotic factors. These robust hybrids could be reared throughout the year and would provide opportunities for further popularizing and improving bivoltine raw silk production in the eastern and north-eastern region. the present investigation identified wb1hh bivoltine breed to be having highest survival rate and better shell ratio of 18.81% in abiotic stress condition. MAS selection identified DNA markers in all breeds and among them WB1HH showed all four markers, confirming the genetic ability of the breed for tolerance to high temperature and humidity. Transcriptional regulation of the high-affinity K+ (HAK) transporter gene is an important mechanism of plant response to low potassium stress. Cloning and analysis of the promoter of potassium transporter gene is not only helpful to its expression pattern and regulation mechanism but also important to improve the potassium absorption efficiency in sugarcane. The potassium transporter gene ScHAK10 is highly expressed and induced by low potassium stress in sugarcane, but the functions of its promoter is still unclear. In the present article, the 1918 bp promoter region of the ScHAK10 gene (pScHAK10) was cloned by genomic walking technique. Computational analysis affirmed the existence of abiotic stress-responsive cis and core cis-elements, such as TATA box, CAAT box, phytohormone responsive, stress response and light response motifs. GUS histochemical staining of transgenic Arabidopsis thaliana seedlings showed that the leaf, corolla, and root tip were deeply stained. The 5′-terminal deletion of the promoter was cloned, and the lengths of 1918 bp (full), 1623 bp (Q1), 1332 bp (Q2), 957 bp (Q3), 576 bp (Q4), and 357 bp (Q5) were cloned into the GUS reporter vector for A. thaliana transient transformation. The transgenic plants generated through a single event exhibited a promising expression of the GUS reporter protein, which was treated with salt, low potassium, IAA and cold stress conditions. The results showed that the promoter activity correlates with the promoter fragment's length, and the long promoter fragment exhibits higher training. The Q5 was the least active and could not drive GUS expression. Under abiotic stress, the expression of GUS enzyme activity varies among different promoter fragments. Under low potassium and high salt stress, Q3 and Q4 showed the highest promoter activity. The Q1 and Q4 led the highest promoter activity during IAA and cold stress. Environmental stress affects food productivity by producing reactive oxygen species (ROS) that damage cell organelles and biomolecules, leading to apoptosis. Plants have developed a variety of defense responses over time to counteract the detrimental effects of biotic and abiotic stresses. However, in order to combat climate change and the deterioration of ecosystems, it will be necessary to promote the use of beneficial bacteria including plant growth-promoting bacteria (PGPR) in agricultural ecosystems. PGPR potentially improves nutrient absorption, modulates hormone levels, and/or improves tolerance to abiotic stress factors and plant pathogens. PGPR offers a sustainable replacement for conventional agrochemicals and traditional farming practices by improving plant growth and stress tolerance. In this context, studying the fundamental evolutionary and ecological relationships among plants and their microbiomes is necessary to build vital approaches to strengthen traditional agricultural techniques. Stress-tolerant PGPR can release bioactive substances such as gibberellins and indole acetic acid which participate in mitigating stresses. PGPR could minimize the adverse effect of salinity, drought, heavy metals, floods, and other stresses on plants by inducing antioxidant enzymes, like catalase, peroxidase, and superoxide dismutase. Elicitation, the process of stimulating plants to enhance the production of valuable secondary metabolites, has emerged as a significant avenue in the field of plant biotechnology. This review paper provides a comprehensive examination of elicitation, encompassing its mechanisms, the imperative for its utilization, and the diverse array of elicitors employed, including abiotic, biotic, and nanoparticle-based agents. By exploring the intricate signaling pathways and molecular responses triggered by external stimuli in this review, we gain insights into how plants tailor their metabolite production in reaction to their environment. Intricately intertwined with the mechanisms, the types of elicitors utilized are extensively discussed. Abiotic elicitors, encompassing physical factors and chemicals, biotic elicitors involving microbe-derived molecules, plant hormones and symbiotic microorganisms, and the emerging application of nanoparticles as elicitation agents are explored. The review further examines the application of these elicitors in both in-vitro and in-vivo cultures, showcasing their practical utility in controlled laboratory settings and real-world environments. By assessing the efficacy of diverse elicitors, this paper provides valuable insights into tailoring elicitation strategies to enhance secondary metabolite yields for various applications. Addressing the limitations and leveraging emerging technologies will undoubtedly steer elicitation research towards innovative breakthroughs, further enhancing our capacity to harness plants for sustainable and efficient secondary metabolite production. Phytoremediation is gaining momentum among bioremediation strategies for addressing high levels of metal(loid)s and organic pollutants in the environment, which threaten plants, wildlife, and human safety due to their cytotoxic, mutagenic, and carcinogenic effects. An impediment to this bioremediation method is the limitation in the innate abilities of phytoremediation species to efficiently cope with pollutant-mediated stress, which often restricts growth, development, and efficient pollutant removal. Phlorotannins, a class of polyphenols derived from marine brown algae, possess a number of bioactivities that may be beneficial for boosting phytoremediation efficiency. This review provides a concise overview of phlorotannins, their chemical nature and structural classes, and the few (indicating a paucity of research data) bioactivities of phlorotannins that have been reported in plants. In addition, included are synopses on different phytoremediation strategies and highlights of major future research perspectives on harnessing phlorotannin bioactivities to ameliorate growth, development, and stress tolerance in phytoremediation species for the benefit of phytoremediation efforts. Azadirachta indica or neem is recognized for its unique properties, both against insects and in improving human health. However, proteomics studies of this plant's response to abiotic stresses have yet to be reported. Application of this technique highly depends on efficient and optimized sample preparation, in deriving highly reproducible proteomics data. The present study highlights the comparison of protein extraction method which effectively recovers proteins from recalcitrant phenolic-rich tissues of neem callus. Four protocols based on TCA-acetone or/and phenol extraction methods were compared. The TCA-acetone complemented with PVPP method was the most effective approach and generated the greatest total protein yields, spot number, good resolution, and spot intensities. Subsequently, the differential proteomics performed revealed that proteins associated with oxidoreduction, energy, transcriptional, stress response, respiration, and cell division were differentially regulated in salinity stress treatment. Meanwhile, the expression of proteins classified in the group of energy, photosynthesis, protein synthesis, oxidoreduction, and post-transcriptional regulation was altered in the JA-stressed callus. The accumulation of AZA was also noticeably reduced in salinity- and JA-treated callus, suggesting these tested stress parameters were not effective in improving AZA production in the neem callus. Overall, this analysis provides valuable information on the application of plant tissue culture as an alternative for sustainable production of major bioactive constituents in A. indica. Main conclusion: In this review, we have discussed the untapped potential of orchid endophytic bacteria as a valuable reservoir of bioactive metabolites, offering significant contributions to plant growth promotion and disease protection in the context of sustainable agriculture. Orchidaceae is one of the broadest and most diverse flowering plant families on Earth. Although the relationship between orchids and fungi is well documented, bacterial endophytes have recently gained attention for their roles in host development, vigor, and as sources of novel bioactive compounds. These endophytes establish mutualistic relationships with orchids, influencing plant growth, mineral solubilization, nitrogen fixation, and protection from environmental stress and phytopathogens. Current research on orchid-associated bacterial endophytes is limited, presenting significant opportunities to discover new species or genetic variants that improve host fitness and stress tolerance. The potential for extracting bioactive compounds from these bacteria is considerable, and optimization strategies for their sustainable production could significantly enhance their commercial utility. This review discusses the methods used in isolating and identifying endophytic bacteria from orchids, their diversity and significance in promoting orchid growth, and the production of bioactive compounds, with an emphasis on their potential applications in sustainable agriculture and other sectors. Oryza sativa ssp. indica is not amenable to genetic modifications owing to its recalcitrant nature, rendering it difficult to transform and recover improved varieties to combat the persisting biotic and abiotic stress conditions. Therefore, in the present study, we have attempted to enhance transformation efficiency by applying polyamines and vacuum infiltration in Agrobacterium tumefaciens-mediated genetic transformation of Indica rice cv. IR64 seed-derived embryogenic calli. The highest number of regenerating calli, with maximum number of shoots per callus, was achieved in regeneration medium supplemented with 0.2 mM Spermidine, and the regenerated shoots displayed improved rooting when cultured on MS medium supplemented with 0.1 mM Putrescine. Further, vacuum infiltration of calli for 4 min resulted in the highest transformation efficiency of 8.1% as evident by GUS positive calli. The transformed calli were screened on the selection medium supplemented with hygromycin, and the integration and expression of the T-DNA in the rice genome were confirmed by GUS histochemical assay, PCR and Southern hybridization. Abiotic stresses such as salinity, submergence, drought and extreme temperatures greatly affect rice production worldwide. Marker-assisted selection was effectively used to introgress QTLs for salinity tolerance (Saltol) and submergence tolerance (Sub1) into the background of elite rice variety Jyothi. Saltol introgressed BC1F2 lines of Jyothi with 79.3% of recipient parent genome was crossed with Sub1 introgressed BC2F2 lines of Jyothi showing 95.9% of parent genome recovery. F1 progenies were screened with Saltol and Sub1 linked foreground markers and recombinant markers. F1 heterozygous plants were selfed to produce F2 generation. F2 progenies homozygous for both the loci were selected. Phenotypic screening for salinity and submergence tolerance was performed to validate the introgressed genes. The plants able to tolerate both salinity and submergence stresses were selected for selfing to raise F3 progenies. Background: Nuclear factor Y (NF-Y) plays a vital role in numerous biological processes as well as responses to biotic and abiotic stresses. However, its function in ginger (Zingiber officinale Roscoe), a significant medicinal and dietary vegetable, remains largely unexplored. Although the NF-Y family has been thoroughly identified in many plant species, and the function of individual NF-Y TFs has been characterized, there is a paucity of knowledge concerning this family in ginger. Methods: We identified the largest number of NF-Y genes in the ginger genome using two BLASTP methods as part of our ginger genome research project. The conserved motifs of NF-Y proteins were analyzed through this process. To examine gene duplication events, we employed the Multiple Collinearity Scan toolkit (MCScanX). Syntenic relationships of NF-Y genes were mapped using the Dual Synteny Plotter software. Multiple sequence alignments were performed with MUSCLE under default parameters, and the resulting alignments were used to generate a maximum likelihood (ML) phylogenetic tree with the MEGA X program. RNA-seq analysis was conducted on collected samples, and statistical analyses were performed using Sigma Plot v14.0 (SYSTAT Software, USA). Results: In this study, the ginger genome was utilized to identify 36 NF-Y genes (10 ZoNF-YAs, 16 ZoNF-YBs, and 10 ZoNF-YCs), which were renamed based on their chromosomal distribution. Ten distinct motifs were identified within the ZoNF-Y genes, with certain unique motifs being vital for gene function. By analyzing their chromosomal location, gene structure, conserved protein motifs, and gene duplication events, we gained a deeper understanding of the evolutionary characteristics of these ZoNF-Y genes. Detailed analysis of ZoNF-Y gene expression patterns across various tissues, performed through RNA-seq and qRT-PCR, revealed their significant role in regulating ginger rhizome and flower growth and development. Additionally, we identified the ZoNF-Y family genes that responded to abiotic stresses. Conclusion: This study represents the first identification of the ZoNF-Y family in ginger. Our findings contribute to research on evolutionary characteristics and provide a better understanding of the molecular basis for development and abiotic stress response. Drought stress is a common abiotic challenge that profoundly impacts plant growth and development. As sessile organisms, plants rely on various physiological and morphological adaptations to cope with drought conditions. The CIPK (calcineurin B‐like protein‐interacting protein kinase) family proteins play a pivotal role in mediating plant responses to abiotic stress through modulation of cellular membrane events via the CBL‐CIPK complex. However, reports documenting the CIPKs’ regulation of non‐membrane events are scant. In this study, we discovered a novel subcellular localisation pattern of the AtCIPK20 protein of Arabidopsis, specifically to cortical microtubules (cMT), which is distinct from previously reported localisation patterns of plant CIPKs. AtCIPK20 regulates ABA‐induced loss of cMT organisation in guard cells, thereby facilitating stomatal closure, mitigating leaf water loss, and protecting plants from drought stress in Arabidopsis. The C‐terminal regulatory domain of AtCIPK20 governs its cMT targeting, whereas the interaction of AtCIPK20 with its CBL partners disrupts this localisation. Notably, the cMT targeting characteristic of AtCIPK20 is not exclusive, as several other CIPK members in Arabidopsis, maize, and rice exhibit similar localisation patterns. These findings broaden our current understanding of the role of plant CIPK members in abiotic stress resistance and suggest that future exploration of CIPK molecular functions should adopt a more comprehensive perspective. The availability of high‐throughput sequencing technologies increased our understanding of different genomes. However, the genomes of all living organisms still have many unidentified coding sequences. The increased number of missing small open reading frames (sORFs) is due to the length threshold used in most gene identification tools, which is true in the genic and, more importantly and surprisingly, in the intergenic regions. Scanning the cucumber genome intergenic regions revealed 420 723 sORF. We excluded 3850 sORF with similarities to annotated cucumber proteins. To propose the functionality of the remaining 416 873 sORF, we calculated their codon adaptation index (CAI). We found 398 937 novel sORF (nsORF) with CAI ≥ 0.7 that were further used for downstream analysis. Searching against the Rfam database revealed 109 nsORFs similar to multiple RNA families. Using SignalP‐5.0 and NLS, identified 11 592 signal peptides. Five predicted proteins interacting with Meloidogyne incognita and Powdery mildew proteins were selected using published transcriptome data of host‐pathogen interactions. Gene ontology enrichment interpreted the function of those proteins, illustrating that nsORFs' expression could contribute to the cucumber's response to biotic and abiotic stresses. miRNAs function as negative regulators that significantly influence plant growth and stress responses. Within rice and other monocotyledonous plants, miR1432 plays a conserved role in seed development and disease resistance. However, its involvement in the response to abiotic stresses remains unclear. Our study aimed to elucidate this mechanism by predicting the targeting of the rice P‐type IIB Ca2+ ATPase gene OsACAs by miR1432 and identifying its cleavage sites via 5′RACE. We observed induced expression of miR1432 and its target gene, OsACA6, under abiotic stresses. Overexpression (OX) of miR1432 and suppression of OsACA6 resulted in reduced cold, salt, and drought tolerance, while OsACA6 suppression/knockout and OX had opposite effects on cold tolerance. Additionally, miR1432 may target other OsACA6 homologs. RNA‐sequencing data highlighted the differential expression of stress‐related genes in miR1432‐overexpressing rice. Furthermore, miR1432‐overexpressing rice exhibited weakened vigor, dwarfism, yellowing leaves and reduced fertility. Collectively, our results strongly suggest that miR1432 not only negatively modulates abiotic stress tolerance by suppressing Ca2+ ATPase gene(s) but also influences plant growth and development. Background: The DELLA proteins, a class of GA signaling repressors, belong to the GRAS family of plant-specific nuclear proteins. Members of DELLA gene family encode transcriptional regulators with diverse functions in plant development and abiotic stress responses. To date, DELLAs have been identified in various plant species, such as Arabidopsis thaliana, Malus domestica, Populus trichocarpa, and other land plants. Most information of DELLA family genes was obtained from A. thaliana, whereas little is known about the DELLA gene family in blueberry. Results: In this study, we identified three DELLA genes in blueberry (Vaccinium darrowii, VdDELLA) and provided a complete overview of VdDELLA gene family, describing chromosome localization, protein properties, conserved domain, motif organization, and phylogenetic analysis. Three VdDELLA members, containing two highly conserved DELLA domain and GRAS domain, were distributed across three chromosomes. Additionally, cis-acting elements analysis indicated that VdDELLA genes might play a critical role in blueberry developmental processes, hormone, and stress responses. Expression analysis using quantitative real-time PCR (qRT-PCR) revealed that all of three VdDELLA genes were differentially expressed across various tissues. VdDELLA2 was the most highly expressed VdDELLA in all denoted tissues, with a highest expression in mature fruits. In addition, all of the three VdDELLA genes actively responded to diverse abiotic stresses. Based on qRT-PCR analysis, VdDELLA2 might act as a key regulator in V. darrowii in response to salt stress, whereas VdDELLA1 and VdDELLA2 might play an essential role in cold stress response. Under drought stress, all of three VdDELLA genes were involved in mediating drought response. Furthermore, their transiently co-localization with nuclear markers in A. thaliana protoplasts demonstrated their transcriptional regulator roles. Conclusions: In this study, three VdDELLA genes were identified in V. darrowii genome. Three VdDELLA genes were closely related to the C. moschata DELLA genes, S. lycopersicum DELLA genes, and M. domestica DELLA genes, respectively, indicating their similar biological functions. Expression analysis indicated that VdDELLA genes were highly efficient in blueberry fruit development. Expression patterns under different stress conditions revealed the differentially expressed VdDELLA genes responding to salt, drought, and cold stress. Overall, these results enrich our understanding of evolutionary relationship and potential functions of VdDELLA genes, which provide valuable information for further studies on genetic improvement of the plant yield and plant resistance. Atrazine, a commonly employed herbicide for corn production, can leave residues in soil, resulting in photosynthetic toxicity and impeding growth in subsequent alfalfa (Medicago sativa L.) crops within alfalfa-corn rotation systems. The molecular regulatory mechanisms by which atrazine affects alfalfa growth and development, particularly its impact on the microbial communities of the alfalfa rhizosphere, are not well understood. This study carried out field experiments to explore the influence of atrazine stress on the biomass, chlorophyll content, antioxidant system, and rhizosphere microbial communities of the atrazine-sensitive alfalfa variety WL- 363 and the atrazine-resistant variety JN5010. The results revealed that atrazine significantly reduced WL-363 growth, decreasing plant height by 8.58cm and root length by 5.42cm (p <0.05). Conversely, JN5010 showed minimal reductions, with decreases of 1.96cm in height and 1.26cm in root length. Chlorophyll content in WL-363 decreased by 35% under atrazine stress, while in JN5010, it was reduced by only 10%. Reactive oxygen species (ROS) accumulation increased by 60% in WL- 363, compared to a 20% increase in JN5010 (p <0.05 for both). Antioxidant enzyme activities, such as superoxide dismutase (SOD) and catalase (CAT), were significantly elevated in JN5010 (p <0.05), suggesting a more robust defense mechanism. Although the predominant bacterial and fungal abundances in rhizosphere soils remained generally unchanged under atrazine stress, specific microbial groups exhibited variable responses. Notably, Promicromonospora abundance declined in WL-363 but increased in JN5010. FAPROTAX functional predictions indicated shifts in the abundance of microorganisms associated with pesticide degradation, resistance, and microbial structure reconstruction under atrazine stress, displaying different patterns between the two varieties. This study provides insights into how atrazine residues affect alfalfa rhizosphere microorganisms and identifies differential microbial responses to atrazine stress, offering valuable reference data for screening and identifying atrazine-degrading bacteria. Introduction: The WRKY transcription factor (TF) family is one of the largest TF families in plants and is widely involved in responses to both biotic and abiotic stresses. Methods: To clarify the function of the WRKY family in blueberries, this study identified the WRKY genes within the blueberry genome and systematically analyzed gene characteristics, phylogenetic evolution, promoter cis-elements, expression patterns, and subcellular localization of the encoded products. Results: In this study, 57 VcWRKY genes were identified, and all encoding products had a complete WRKY heptapeptide structure and zinc-finger motif. The VcWRKY genes were divided into three subgroups (I-III) by phylogenetic analysis. Group II was divided into five subgroups: IIa, IIb, IIc, IId, and IIe. 57 VcWRKY genes were distributed unevenly across 32 chromosomes. The amino acids ranged from 172 to 841, and molecular weights varied from 19.75 to 92.28 kD. Intra-group syntenic analysis identified 12 pairs of duplicate segments. Furthermore, 34 cis-element recognition sites were identified in the promoter regions of VcWRKY genes, primarily comprising phytohormone-responsive and light-responsive elements. Comparative syntenic maps were generated to investigate the evolutionary relationships of VcWRKY genes, revealing the closest homology to dicotyledonous WRKY gene families. VcWRKY genes were predominantly expressed in the fruit flesh and roots of blueberries. Gene expression analysis showed that the responses of VcWRKY genes to stress treatments were more strongly in leaves than in roots. Notably, VcWRKY13 and VcWRKY25 exhibited significant upregulation under salt stress, alkali stress, and saline-alkali stress, and VcWRKY1 and VcWRKY13 showed notable induction under drought stress. Subcellular localization analysis confirmed that VcWRKY13 and VcWRKY25 function within the nucleus. Conclusion: These findings establish a foundation for further investigation into the functions and regulatory mechanisms of VcWRKY genes and provide guidance for selecting stress-tolerant genes in the development of blueberry cultivars. Cd (cadmium) is a highly toxic heavy metal pollutant often present in soil and detrimentally impacting the production and quality of horticultural crops. Cd affects various physiological and biochemical processes in plants, including chlorophyll synthesis, photosynthesis, mineral uptake and accumulation, and hormonal imbalance, leading to cell death. The MYB family of transcription factors plays a significant role in plant response to environmental influences. However, the role of MYB116 in abiotic stress tolerance remains unclear. In this study, we reported that Chinese cabbage transcription factor BrMYB116 enhanced Cd stress tolerance in yeast. The expression level of BrMYB116 was increased by Cd stress in Chinese cabbage. Additionally, yeast cells overexpressing BrMYB116 showed improved Cd stress tolerance and reduced Cd accumulation. Moreover, we found that BrMYB116 interacted with facilitator of iron transport (FIT3) to enhance Cd stress tolerance. ChIP-qPCR results showed that ScFIT3 was activated through specific binding to its promoter. Additionally, the overexpression of ScFIT3 induced Cd stress tolerance and reduced Cd accumulation in yeast and Chinese cabbage. Bell pepper (Capsicum annuum L.); an important spice crop of the region is a rich source of vitamins and antioxidants having many health benefits. Many biotic and abiotic factors contribute towards growth and yield losses of this crop. Arsenic (As) toxicity is a global issue, but it is particularly critical in developing countries. The current study was designed to evaluate the efficacy of selenium (Se) in mitigating the toxic effects of As in two varieties (HSP-181 A and PS09979325) of Capsicum annuum L. Different concentrations of As (0, 50, and 100 µM) and Se (0, 5, and 10 µM) were tested using 14 days old seedlings of C. annuum L. The As stress caused a significant (P ≤ 0.001) reduction in growth, uptake of nutrients, and eco-physiological attributes in both varieties however, the response was specific. While the overproduction of osmo-protectants and antioxidants intensified the symptoms of oxidative stress. The maximum reduction in shoot length (45%), fresh weight (29%), and dry weight (36%) was observed in under 100 µM As stress. The organic acids exudation from the roots of both cultivars were significantly increased with the increase in As toxicity. The Se treatment significantly (p ≤ 0.001) improved growth, nutrient uptake, gas exchange attributes, antioxidant production, while decreased oxidative stress indicators, and As uptake in the roots and shoots of all the subjects under investigation. It is concluded from the results of this study that Se application increased photosynthetic efficiency and antioxidant activity while decreasing As levels, organic acid exudation, and oxidative stress indicators in plants. Background: Plant long non-coding RNAs (lncRNAs) have important regulatory roles in responses to various biotic and abiotic stresses, including light quality. However, no lncRNAs have been specifically linked to the Shade Avoidance Response (SAS). Results: To better understand the involvement of lncRNAs in shade avoidance, we examined RNA-seq libraries for lncRNAs with the potential to function in the neighbor proximity phenomenon in Arabidopsis thaliana (A. thaliana). Using transcriptomes generated from seedlings exposed to high and low red/far-red (R/FR) light conditions, we identified 13 lncRNA genes differentially expressed in cotyledons and 138 in hypocotyls. To infer possible functions for these lncRNAs, we used a 'guilt-by-association' approach to identify genes co-expressed with lncRNAs in a weighted gene co-expression network. Of 34 co-expression modules, 10 showed biological functions related to differential growth. We identified three potential lncRNAs co-regulated with genes related to SAS. T-DNA insertions in two of these lncRNAs were correlated with morphological differences in seedling responses to increased FR light, supporting our strategy for computational identification of lncRNAs involved in SAS. Conclusions: Using a computational approach, we identified multiple lncRNAs in Arabidopsis involved in SAS. T-DNA insertions caused altered phenotypes under low R/FR light, suggesting functional roles in shade avoidance. Phenylalanine ammonia lyase (PAL) is a widely studied enzyme in plant biology due to its role in connecting primary metabolism to secondary phenylpropanoid metabolism, significantly influencing plant growth, development, and stress response. Although PAL genes have been extensively studied in various plant species but their exploration in cucumber has been limited. This study successfully identified 11 CsPAL genes in Cucumis sativus (cucumber). These CsPAL genes were categorized based on their conserved sequences revealing patterns through MEME analysis and multiple sequence alignment. Interestingly, cis-elements related to stress were found in the promoter regions of CsPAL genes, indicating their involvement in responding to abiotic stress. Furthermore, these gene's promoters contained components associated with light, development and hormone responsiveness. This suggests that they may have roles in hormone developmental processes. MicroRNAs were identified as a key regulators for the CsPAL genes, playing a crucial role in modulating their expression. This discovery underscores the complex regulatory network involved in the plant's response to various stress conditions. The influence of these microRNAs further highlights the complicated mechanisms that plants use to manage stress. Gene expression patterns were analyzed using RNA-seq data. The significant upregulation of CsPAL9 during HT3h (heat stress for 3 h) and the heightened upregulation of both CsPAL9 and CsPAL7 under HT6h (heat stress for 6 h) in the transcriptome study suggest a potential role for these genes in cucumber's tolerance to heat stress.
