This research article gives out the Mathematical design and system for solving a time cost trade-off problems by using fuzzy linear programming problem and Multiple Attribute Group Decision Making (MAGDM) problems. A linear numerical template for project time cost trade-off problem is evolved through this work which gives the optimum solution. The activities presented in the network take the form of decision matrices which are solved by using aggregation operators available in the literature. Several aggregation operators are employed in the decision process and the best alternative which comprises the normal cost, normal duration, crash cost and crash duration is selected for each activity and then the optimal solution of the network is obtained. Chondroitin sulfate (CS) is a type of sulfated glycosaminoglycan that is manufactured by extraction from animal tissues for the treatment of osteoarthritis and in drug delivery applications. Here, the authors report the development of single microbial cell factories capable of compete, one-step biosynthesis of animal-free CS production in E. coli.

Abstract N-glycolyl chondroitin (Gc-CN) is a metabolite of N -glycolylneuraminic acid (Neu5Gc), a sialic acid that is commonly found in mammals, but not humans. Humans can incorporate exogenous Neu5Gc into their tissues from eating red meat. Neu5Gc cannot be biosynthesized by humans due to an evolutionary mutation and has been implicated in causing inflammation causing human diseases, such as cancer. The study Neu5Gc is important in evolutionary biology and the development of potential cancer biomarkers. Unfortunately, there are several limitations to detecting Neu5Gc. The elimination of Neu5Gc involves a degradative pathway leading to the incorporation of N-glycolyl groups into glycosaminoglycans (GAGs), such as Gc-CN. Gc-CN has been found in humans and in animals including mice, lamb and chimpanzees. Here, we present the biosynthesis of Gc-CN in bacteria by feeding chemically synthesized N-glycolylglucosamine to Escherichia coli. A metabolically engineered strain of E. coli K4, fed with glucose supplemented with GlcNGc, converted it to N-glycolylgalactosamine (GalNGc) that could then be utilized as a substrate in the chondroitin biosynthetic pathway. The final product, Gc-CN was converted to disaccharides using chondroitin lyase ABC and analyzed by liquid chromatography–tandem mass spectrometry with multiple reaction monitoring detection. This analysis showed the incorporation of GalNGc into the backbone of the chondroitin oligosaccharide.

Background: Recent reports have unveiled the potential of flavonoids to enhance bone formation and assuage bone resorption due to their involvement in cell signaling pathways. They also act as an effective alternative to circumvent the disadvantages associated with existing treatment methods, which has increased their scope in orthopedic research. Valproic acid (VA, 2-propylpentanoic acid) is one such flavonoid, obtained from an herbaceous plant, used in the treatment of epilepsy and various types of seizures., Objective: In this study, the role of VA in osteogenesis and the molecular mechanisms underpinning its action in mouse mesenchymal stem cells (mMSCs) were determined., Methods: Results: Cytotoxic studies validated VA's amiable nature in mMSCs. Alizarin red and von Kossa staining results showed an increased deposition of calcium phosphate in VA-treated mMSCs, which confirmed the occurrence of osteoblast differentiation and mineralization at a cellular level. At the molecular level, there were increased levels of expression of Runx2, a vital bone transcription factor, and other major osteoblast differentiation marker genes in the VA-treated mMSCs. Further, VA-treatment in mMSCs upregulated mir-21 and activated the mitogen-activated protein kinase/extracellular signal-regulated kinase signaling pathway, which might be essential for the expression/activity of Runx2., Conclusion: Thus, the current study confirmed the osteoinductive nature of VA at the cellular and molecular levels, opening the possibility for its application in bone therapeutics with mir-21., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.net.)

Traditional methods of bone defect repair include autografts, allografts, surgical reconstruction, and metal implants that have several disadvantages such as donor site morbidity, rejection, risk of disease transmission, and repetitive surgery. Biomaterial-based bone reconstructions can, therefore, be an efficient alternative due to the inherent properties of the materials. Chitosan (CS), the deacetylated form of chitin, is a biopolymer having a wide array of applicability in regenerative tissue applications owing to its biocompatible, in vitro degradative and bioresorbable nature. Extensive studies are being carried out using CS to augment the properties of the already existing methods and to also improve the applicability of CS-based biocomposites in bone tissue repair. In this review, the suitability of CS as a surface modifier has been discussed in detail for the already existing implants, surface modifications of CS-based natural biocomposites for bone tissue regeneration, and the wide range of techniques that can introduce these modifications. CS, being a natural polymer, possesses advantageous properties including surface modifier that makes it a suitable candidate for bone regeneration, and further research to investigate its osteogenic potential in vivo along with the molecular and signaling mechanisms involved in bone regeneration can aid in expanding its applicability in clinical trials., (© 2019 WILEY-VCH Verlag GmbH & Co. Flooding is one of the most disastrous global hazards, which has been occurring more frequently in recent times. It is observed that climate change is likely to increase the intensity and the frequency of floods and river basins have become more vulnerable to fluvial flooding. In this study, the impact of climate change on fluvial flooding was analyzed over the Adyar sub-basin. This study applied statistically downscaled Global Climate Model (GCM) data in a CMIP5 dataset of IPCC Assessment Report 5 (AR5). Based on the performance to simulate the observed climate, four GCMs, namely, cesm1-cam5, mpi-esm-mr, ncar-ccsm4, and bnu-esm, for RCP 4.5 were selected for projections of the future scenario. The Intensity-Duration-Frequency (IDF) curves for the past and future scenarios were derived from the IMD-observed and GCM-projected rainfall data. Integrated flood modeling was performed with hydrologic (HEC-HMS) and hydraulic (HEC-RAS) models. Finally, in order to visualize the inundation areas according to the future climate projection, flood inundation maps were prepared geospatially using the ArcGIS software. For the 100-year return period, the results predict that the peak discharge for the future climate scenario would increase by 34.3%-91.9% as compared to the present climate scenario. Similarly, the future projections show an increase in the flooded area ranging from 12.6% to 26.4% based on GCMs. This simulation helps in understanding the flood risk over the Adyar sub-basin under the changing climate and the requirement for the regulation of developmental activities over the flood-prone areas., Competing Interests: The authors have declared that no competing interests exist.

Intercellular communication in bacteria (quorum sensing, QS) is an important phenomenon in disease dissemination and pathogenesis, which controls biofilm formation also. This study reports the anti-QS and anti-biofilm efficacy of seaweed Gracilaria gracilis associated Vibrio alginolyticus G16 against Serratia marcescens. Purification and mass spectrometric analysis revealed the active principle as phenol, 2,4-bis(1,1-dimethylethyl) [PD]. PD affected the QS regulated virulence factor production in S. marcescens and resulted in a significant (p < 0.05) reduction in biofilm (85%), protease (41.9%), haemolysin (69.9%), lipase (84.3%), prodigiosin (84.5%) and extracellular polysaccharide (84.62%) secretion without hampering growth, as evidenced by XTT [2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide] assay. qPCR analysis confirmed the down-regulation of the fimA, fimC, flhD and bsmA genes involved in biofilm formation. Apart from biofilm inhibition and disruption, PD increased the susceptibility of S. marcescens to gentamicin when administered synergistically, which opens another avenue for combinatorial therapy where PD can be used to enhance the efficacy of conventional antibiotics.
