Background: The adverse effects of cytotoxic medications like methotrexate, particularly reproductive damage reported in the numerous experimental studies, limit their effectiveness as anticancer therapies. The current study's objectives were to identify potential histological and immune-histochemical unfavorable changes in the testicles due to methotrexate medication and to assess any possible protective effects of erythropoietin (EPO). Methods and Results: The study included 60 mature male albino rats weighing 200-250g. The animals were divided equally into three groups, each with 20 rats. In Group 1, the control group, the animals received intraperitoneal injections of normal saline twice a week for nine weeks at a dose of 0.5 mg/kg. For nine weeks, animals in Group 2 received intraperitoneal injections of methotrexate hydrate at 0.5 mg/kg twice a week. Animals in Group 3 received subcutaneous injections of 100 IU/kg recombinant human EPO once a week for nine weeks and intraperitoneal injections of methotrexate hydrate at a dosage of 0.5 mg/kg twice a week for nine weeks. Each rat was weighed every week throughout the examinations. An ELISA technique was used to measure the levels of testosterone, malondialdehyde (MDA), total antioxidant capacity (TAC), and ROS in blood serum. Morphological and histopathological changes in testicular tissue were assessed. The body weight in the rats treated with methotrexate was considerably lower than in control group rats. EPO showed clear androgenic and antioxidant activities and reduced the adverse effects of methotrexate on testicular histology. Our results suggest further research into the use of EPO as a drug to protect patients from the adverse effects of methotrexate.

Background: Nursing and midwifery professionals’ well-being may be affected by various factors, including the bullying of new nurses. Purpose: To analyze the relationship between bullying behaviors, perceived stress, and acute stress among undergraduate nursing and midwifery students in Saudi Arabia, as well as the moderating role of gender and academic majors in this relationship. Method: A cross-sectional correlation design was used, and data were collected from 322 undergraduate nursing and midwifery students enrolled in four major nursing universities in Saudi Arabia. Results: Educational level, environment, and personal attacks had a significant negative impact on perceived and acute stress (social and non-social), while hostility toward academic achievement and direct negative behaviors had significant positive impacts on perceived and acute stress (social and non-social). Female nursing students reported a stronger impact of bullying behavior on their perceived stress, while male students reported a greater impact on social and non-social stress. Nursing students reported a stronger impact of bullying on perceived stress, while midwifery students reported a greater impact on social and non-social stress. Discussion: Nursing educators should raise awareness about the harmful effects of bullying and emphasize the importance of creating a safe, supportive learning environment.

Abstract Obesity among adolescents is a global health apprehension which requires early prevention. The aim of this study was to determine the association between lifestyle habits including physical activity, sedentary behaviors and eating habits with obesity indices of body mass index (BMI) and waist-to-height ratio (WHtR) among male adolescents in Riyadh, Saudi Arabia. We randomly selected 471 secondary school male adolescents aged 14–18 years. A pre-validated self-reported questionnaire was used to record the data on physical activity level, sedentary behaviors, sleep duration and eating habits. The International Obesity Task Force (IOTF) cutoff values for adolescents under 18 years of age were used to define overweight and obesity. Total energy expenditure was calculated using metabolic equivalent-minutes per week. Anthropometry including weight, height, BMI, waist circumference, waist/height ratio (WHtR), were assessed. 53.7% and 48.4% of the adolescents were overweight/obese and had abdominal obesity; respectively. Those with overweight and obesity or above 50% of WHtR were much less active in terms of METs-min/week from vigorous-intensity sports, sum of all METs-min/week from all vigorous-intensity physical activity, total METs-min/week from all physical activity compared with non-obese adolescents and below 50% of WHtR. The present study identified the lifestyle habits that were associated with obesity and may represent valid targets for the prevention and management of obesity among Saudi adolescents. Knowledge of the factors that contribute to obesity could be used in preventive programs for the control of obesity among adolescents.

Purpose: Clubfoot is among the most common musculoskeletal congenital anomalies. Poor understanding of clubfoot can cause lack of awareness that leads to complications in treating this condition because of late medical intervention. This is considered as a significant public health problem, especially in communities where the burden of clubfoot deformity remains unrecognized. We assessed the level of awareness and knowledge on clubfoot among residents of rural areas and determined the knowledge and attitudes of the public toward the risk factors and general consequences of clubfoot. Methods: This cross-sectional study, conducted from January to July 2021, included the general population of small city and rural area residents. The participants completed a self-administered survey on the web. The questionnaire was pre-tested in a pilot study to ensure comprehension and ease of administration. Results: Altogether, 41.6% of the participants recognized the scientific name of clubfoot in their native language, whereas 38.9% identified the disorder after they were shown a photograph of it. The most recognized risk factors of clubfoot according to the participants were family history (52.2%), complicated pregnancy (46.5%), and medications (43.4%). Only 8.8% of the participants considered casting the initial treatment of clubfoot. Conclusion: An individual's residential area plays a role in their level of awareness concerning clubfoot. Our results suggest that many educational interventions for clubfoot must be provided in rural areas, particularly regarding therapeutic options and plans of care.

In this study, calcium-rich resource minerals such as brushite, tricalcium phosphate (TCP), and hydroxyapatite were tested as catalysts for the methanolysis of alkaline solutions of NaBH4 to generate hydrogen H2. The synthesis of calcium phosphate compounds was characterized by means of X-ray powder diffraction (XRD) and scanning electron microscopy (SEM). The hydrogen generation rate with the TCP catalyst (15,214 mL min−1 g−1) was higher than with the hydroxyapatite catalyst (12,437 mL min−1 g−1) and brushite catalyst (6210 mL min−1 g−1) for the methanolysis of 250 mg NaBH4 at 298 K using 25 mg of catalyst. The impact of TCP weight on hydrogen generation was studied. The methanolysis reaction led to a higher hydrogen volume generation over time with an increase in the weight of the TCP catalyst at a temperature of 308 K. The calculated activation energy for NaBH4 hydrolysis with the TCP catalyst was 23.944 kJ mol−1, suggesting the high catalytic activity of TCP. The values of enthalpy (ΔH) and entropy (ΔS) were calculated, and the results showed that ΔH was 21.28 kJ mol−1 and ΔS was −93.096 J·mol−1. ΔH was positive, meaning that the reaction was endothermic, and the negative ΔS meant a decrease in the disorder of the methanolysis reaction. The stability of the catalysis was tested in successive methanolysis tests. The catalyst's efficiency decreased to 89% after four cycles. [ABSTRACT FROM AUTHOR]

Cyclin-dependent kinase 5 (CDK5) plays a crucial role in various biological processes, including immune response, insulin secretion regulation, apoptosis, DNA (deoxyribonucleic acid) damage response, epithelial−mesenchymal transition (EMT), cell migration and invasion, angiogenesis, and myogenesis. Overactivation of CDK5 is associated with the initiation and progression of cancer. Inhibiting CDK5 has shown potential in suppressing cancer development. Despite advancements in CDK5-targeted inhibitor research, the range of compounds available for clinical and preclinical trials remains limited. The marine environment has emerged as a prolific source of diverse natural products with noteworthy biological activities, including anti-cancer properties. In this study, we screened a library of 47,450 marine natural compounds from the comprehensive marine natural product database (CMNPD) to assess their binding affinity with CDK5. Marine compounds demonstrating superior binding affinity compared to a reference compound were identified through high-throughput virtual screening, standard precision and extra-precision Glide docking modes. Refinement of the selected molecules involved evaluating molecular mechanics–generalized born surface area (MM/GBSA) free binding energy. The three most promising compounds, (excoecariphenol B, excoecariphenol A, and zyzzyanone B), along with the reference, exhibiting favorable binding characteristics were chosen for molecular dynamics (MD) simulations for 200 nanoseconds. These compounds demonstrated interaction stability with the target during MD simulations. The marine compounds identified in this study hold potential as effective CDK5 inhibitors and warrant subsequent experimental validation.

In the present study, four Lactobacillus strains from the cheese were analyzed for its probiotic potential against enteropathogenic bacteria. The probiotic properties of the selected strains were also analyzed and the selected bacterial strains showed high tolerance in bile salts and organic acid. The strain L. plantarum LP049 showed maximum survival rate (92 ± 4.2% and 93.3 ± 2%) after 3 h of treatment at 0.25% (w/v) bile salts and 0.25% (w/v) organic acid concentrations. The ability of the Lactobacillus strains to adhere to human epithelial cells (HT-29 cell lines) was evaluated and L. plantarum LP049 showed maximum adhesion property (19.2 ± 1.1%) than other tested strains. The Lactobacillus strains produced lactic acid at various concentrations. Compared with other strains, maximum level of lactic acid (3.1 g/L), hydrogen peroxide (4.31 mM) and bacteriocin (31 AU/mg) was detected in LB049. The inhibitory activity of culture supernatant against various bacterial pathogens was observed. The zone of inhibition ranged between 6 ± 2 mm and 23 ± 2 mm. The cell free extract showed activity against, Escherichia coli (ATCC 10536), Salmonella enteritidis (ATCC 13076), Shigella flexneri (ATCC 29903), and Enterococcus faecium (ATCC 8459). Consequently, L. plantarum LP049 may be considered as a potential candidate for the production of novel bioactive metabolites for therapeutic and bio-protective applications.

The present study aimed to analyze the nephroprotective property of violacein obtained from the bacterium, Chromobacterium violaceum. The nephrotoxicity in the animal model was induced by gentamicin, potassium dichromate, mercuric chloride, and cadmium chloride-induced nephrotoxicity in the Wistar rats was analyzed by measuring the serum creatinine, uric acid, and urea level. The present investigation revealed the nephroprotective property on convoluted proximal tubule (S1 and S2 segments) and the straight proximal tubule (S3 segment). Also, violacein significantly improved the renal function by the renal protective property on S2 segment of proximal tubule from the nephrotoxicity stimulated by mercuric chloride, potassium dichromate, cadmium chloride and gentamicin in animal models. Animal model studies revealed that violacein at 20 and 40 mg/kg p.o improved the renal function and significantly reduced the increased amount of uric acid, creatinine, and blood urea compared to the control.

Abstract Background Raising the general population knowledge of Alzheimer’s disease is crucial for optimizing care and ultimately enhancing the quality of life of people diagnosed and their caregivers. Aim To assess knowledge among the general population in Makkah Region about symptoms, diagnosis, causes, course, life impact, care giving, and management of Alzheimer disease. Material and methods A descriptive cross-sectional study was conducted during July to August 2018 in three main cities of Makkah Region, i.e., Makkah, Jeddah, and Taif, Saudi Arabia. Two to three mosques and/or malls were selected randomly from a list of all mosques and malls in each city, and then every fifth subject was selected using a systematic random technique, entering the mall or getting out from the mosques. Both genders, aged 18 years or more and able to understand Arabic were included. Valid and reliable Alzheimer’s Disease Knowledge Scale (ADKS) was used to assess knowledge about the diseases, in addition to socio-demographic characteristics of the participants. Results The study included 862 adult participants. Their age ranged between 18 and 65 years with a mean ± standard deviation (SD) of 31.2 ± 10.3 years. History of having a relative with Alzheimer’s disease was mentioned by 20.8% of the participants. Overall, the percentage of total Alzheimer’s disease knowledge score ranged between 26.7 and 100% with a mean of 58.5%, median of 60%, mode of 56.7%, and standard deviation of ± 11.5%. There was a significant positive correlation between participant’s age and their knowledge regarding Alzheimer’s disease (Spearman’s correlation coefficient “r” = 0.099, p = 0.004). The highest known subscale of Alzheimer’s disease was assessment (mean percentage was 62.21%), followed by treatment (62.09), care giving (61.83%) and course of the disease (60.99%) whereas the lowest known subscale was symptoms (53.25%). Participants who reported a history of having a relative with Alzheimer’s disease were more knowledgeable regarding symptoms than those who did not had such history (mean ranks were 470.48 and 421.28, respectively), p = 0.014. Conclusion Knowledge of the general population in Makkah Region is deficient, particularly regarding symptoms, risk factors and life impacts. Therefore, health education campaigns are required for general public in this regards.

In recent times, coronary artery disease (CAD) has become one of the leading causes of morbidity and mortality across the globe. Diagnosing the presence and severity of CAD in individuals is essential for choosing the best course of treatment. Presently, computed tomography (CT) provides high spatial resolution images of the heart and coronary arteries in a short period. On the other hand, there are many challenges in analyzing cardiac CT scans for signs of CAD. Research studies apply machine learning (ML) for high accuracy and consistent performance to overcome the limitations. It allows excellent visualization of the coronary arteries with high spatial resolution. Convolutional neural networks (CNN) are widely applied in medical image processing to identify diseases. However, there is a demand for efficient feature extraction to enhance the performance of ML techniques. The feature extraction process is one of the factors in improving ML techniques’ efficiency. Thus, the study intends to develop a method to detect CAD from CT angiography images. It proposes a feature extraction method and a CNN model for detecting the CAD in minimum time with optimal accuracy. Two datasets are utilized to evaluate the performance of the proposed model. The present work is unique in applying a feature extraction model with CNN for CAD detection. The experimental analysis shows that the proposed method achieves 99.2% and 98.73% prediction accuracy, with F1 scores of 98.95 and 98.82 for benchmark datasets. In addition, the outcome suggests that the proposed CNN model achieves the area under the receiver operating characteristic and precision-recall curve of 0.92 and 0.96, 0.91 and 0.90 for datasets 1 and 2, respectively. The findings highlight that the performance of the proposed feature extraction and CNN model is superior to the existing models.

Introduction: Scrotal orchidopexy for palpable undescended testicle (UDT) has received attention in the last decade due to its lower morbidity. This study was conducted to determine the frequency and factors related to the use of the scrotal approach in the surgical treatment of palpable UDT among surgeons. Methods: An observational cross-sectional study was carried out using an online survey, which was sent to different pediatric urologists, pediatric surgeons, and urologists groups. The survey consisted of questions on demographics as well as surgeons opinions and experience toward scrotal orchidopexy. Results: Of 163 respondents, 57 (35.0%) were pediatric surgeons, 98 (60.1%) were pediatric urologists, and 8 (4.9%) were urologists. There were 86 respondents (52.8%) who used the scrotal orchidopexy approach for UDT at any time in their practice. Pediatric urologists tended to use the scrotal orchidopexy approach for UDT more significantly than others (P < 0.001). There were significantly more scrotal orchidopexies for UDT performed by the pediatric urologists throughout their practice and per year compared to others, respectively (P < 0.001). Fifty-two respondents (31.9%) claimed that scrotal orchidopexy is not a good option for their patients, while seven respondents (4.3%) claimed that the procedure was hard to perform. Discussion: Based on the results of this study, we believe that there is a discrepancy in the reported advantages and success rate of scrotal orchidopexy in the published literature and the utilization of such an approach among surgeons managing palpable UDT in children. Conclusion: Scrotal orchidopexy is an underutilized approach in the management of palpable UDT in children. Only 52.8% of our respondents used it for UDT. One of the main reasons why scrotal orchidopexy is underutilized is due to the surgeons' perception that scrotal orchidopexy is not the procedure of choice for their patients and their unfamiliarity with the procedure.
