One of the major contaminants in aquatic ecosystems is bisphenol A (BPA), a synthetic industrial chemical widely used in the production of epoxy resins and plastics. BPA has negative effects on fish health. Proanthocyanidin, a natural immune stimulant extracted from grape seeds (GR), possesses various biological and antioxidant properties. This study aimed to mitigate the harmful effects of BPA by supplementing the fish diet with GR. Healthy Nile tilapia (120 fish, 14.95±0.10g) were divided into four groups: Group 1 served as the control; group 2 was fed 400mg GR per kg of feed; group 3 was exposed to 3mg of BPA per liter of water without GR supplementation, and group 4 was exposed to 3mg of BPA per liter of water and fed a GR-supplemented diet (400mg GR per kg). After 28 days, blood and tissue samples were collected to assess serum biochemical parameters, cytokines, catalase (CAT) activity, malondialdehyde (MDA) levels, and the histological features of the liver, spleen, and kidney. Co-treatment with GR improved serum protein levels as well as urea and creatinine levels that were previously altered by BPA exposure. BPA exposure elevated serum interleukin (IL)-1β and hepatic MDA levels, which were reduced by GR administration. Additionally, CAT activity decreased in the BPA group but increased in the GR+BPA group. BPA also significantly upregulated the mRNA transcripts of IL-6 and tumor necrosis factor (TNF-α), which may trigger inflammation, while GR significantly downregulated these genes in the BPA-treated group. Histological analysis showed that GR alleviated tissue damage caused by BPA. Simple Summary: Donkey welfare and pain relief are of particular importance in the veterinary field. Therefore, pain management during surgical operations in field settings is mandatory. The current study sheds light on the antinociceptive effect of a combination of dexmedetomidine and nalbuphine premedication prior to total intravenous anesthesia using ketamine-propofol in castration procedures in field conditions. Shortly after castration, the proposed combination of dexmedetomidine and nalbuphine resulted in decreased postoperative pain. In addition, the cardiac function was improved compared to using dexmedetomidine alone during anesthesia. This was demonstrated using established clinicophysiological assessments, serum biochemical markers, and behavioral pain scores for six hours post-recovery. Dexmedetomidine and nalbuphine premedication prior to total intravenous anesthesia using ketamine-propofol showed potential benefits compared to dexmedetomidine alone in providing more analgesia and managing postoperative pain in jacks undergoing castration under field conditions. This study evaluated the antinociceptive effect of dexmedetomidine-nalbuphine vs. dexmedetomidine alone in jacks undergoing field castration under total intravenous anesthesia. Jacks were premedicated with intravenous (IV) dexmedetomidine (5 µg/kg), either alone (Group D, n = 6) or in combination with 0.3 mg/kg nalbuphine (Group DN, n = 6). IV ketamine (1.5 mg/kg) and propofol (0.5 mg/kg) were used to induce general anesthesia, which was maintained by a continuous propofol (0.2 mg/kg/min) IV infusion. The quality of anesthesia, analgesia, and recovery were evaluated. A simple descriptive scale (SDS) was used to measure pain from the recovery time to 6 h later. The DN group exhibited improvements in analgesic and recovery quality and SDS of pain at 1-, 2-, and 3-h post-recovery. There was an apparent improvement in cardiac status, as evidenced by the enhanced heart rate and electrocardiogram findings compared to group D during surgery and recovery time. The DN group had a lower level of inflammatory cytokines, both during the surgery and shortly after recovery. Therefore, the dexmedetomidine-nalbuphine combination prior to IV anesthesia of ketamine and propofol in jacks undergoing field castration resulted in a stable surgical plane of anesthesia, improved antinociception, less pain postoperatively, and better cardiac stability. The modulatory role of primrose oil (PO) supplementation enriched with γ-linolenic acid and D/L-alpha tocopherol acetate against a carbon tetrachloride (CCl4)-induced liver damage model was assessed in this study. Twenty male Albino rats were divided into four groups. The control group received corn oil orally. The PO group received 10 mg/kg P O orally. The CCl4 group received 2 mL/kg CCl4 orally and PO/CCl4 group; received PO and 2 mL/kg CCl4 orally. The relative liver weight was recorded. Serum liver enzymes, hepatic malondialdehyde (MDA), hepatic reduced glutathione (GSH) and the expression of hepatic tumor necrosis factor-alpha (TNF-α), interleukin 1 beta (IL-1β), and interleukin 6 (IL-6) were assessed. The binding affinities of γ-linolenic acid and D/L-alpha tocopherol constituents with IL-1β, IL-6 and TNF-α were investigated using molecular docking simulations. Histopathological and electron microscopic examinations of the liver were performed. The results indicated that CCl4 elevated serum liver enzyme and hepatic MDA levels, whereas GSH levels were diminished. The upregulation of IL-1β, IL-6, and TNF-α gene expressions were induced by CCl4 treatment. The PO/CCl4-treated group showed amelioration of hepatic injury biomarkers and oxidative stress. Restoration of histopathological and ultrastructural alterations while downregulations the gene expressions of TNF-α, IL1-β and IL-6 were observed. Obesity's dangers are becoming more widely recognized around the world. Obese people are more likely to develop a variety of serious diseases, including fatty liver. The purpose of this study was to investigate the effect of Origanum majorana on light microscopic changes in the liver, as well as hematology and some liver enzymes in experimentally obese albino rats. Four groups of twenty-four male albino rats were formed. For three months, the control group was fed a standard laboratory diet and received saline orally by gavage tube. Obese rats were given a high-fat diet. The marj group received 20 mg/kg of aqueous extract of marjoram via gavage tube. The obese/marj group received the same treatment as the obese group and received 20 mg/kg of aqueous extract of marjoram daily via gavage tube. The liver and visceral fat weights of animals were recorded, blood samples for hematology and liver functions were estimated. Liver samples were fixed in 10% formol saline then stained by Hematoxylin and Eosin for histological examination. Obese animals had significantly higher liver, visceral fat weights, and liver function than the control group. Histologically, there was an increase of fatty degeneration in the obese group. However, the obese group that received the aqueous leaf extract of Origanum majorana was lower in all the measured and examined liver parameters than that of the control group. Obesity is a global public health issue. It is associated with a spectrum of liver abnormalities, like nonalcoholic fatty liver disease (NAFLD). The purpose of this study was to investigate the effect of Vitamin D3 on light microscopic changes in the liver, as well as hematology and some liver enzymes in experimentally overweight albino rats. Five groups of thirty-five male albino rats were formed. For three months, Control (C) group received a standard laboratory diet. Corn oil (CO) group received normal balanced diet and given orally 20 mg/ kg corn oil daily. Vitamin D (D) group received normal balanced diet and orally treated daily with 5000 IU/kg of vitamin D3. Overweight (OW) group rats received high caloric diet. Overweight/ vitamin D (OWD) group received high caloric diet and treated with oral vitamin D3 5000 IU/kg daily. Liver weights of animals were recorded, blood samples for hematology, some liver functions, catalase (CAT) and total antioxidant capacity (TAC) were collected. Liver samples were fixed in 10% formal saline then stained by H&E for histological examination. OW group had significantly higher liver weights, and liver function than CO group. Histologically, there was an increase of fatty degeneration in the OW group. However, OWD group was lower in all the measured and examined liver parameters than that of CO group in this study vitamin D3 supplement could ameliorate the abnormal changes in the hepatocytes induced by obesity. Adrenaline is widely used drug to combat several conditions such as allergy and anaphylaxis. The current work was to investigate the renal and hepatic complications following adrenaline injection, in addition, the impact of Cleome (SAMWA) methanolic extract on adrenaline- induced renal and hepatic alterations. Twenty- four male Wister rats were divided equally into four groups; Normal control group received oral distilled water for 30 consecutive days and administered subcutaneous saline on the 31st and 32nd days. The Cleome extract (200 mg/kg) group received Cleome methanolic extract (200 mg/kg, P.O) for 30 consecutive days and administered subcutaneous saline on the 31st and 32nd days. The adrenaline group received distilled water orally for 30 consecutive days and administered subcutaneous adrenaline (2 mg/kg, s.c.) divided into two doses (1 mg/kg, s.c) each on the 31st and 32nd days. Cleome extract (200 mg/kg)/ adrenaline received Cleome methanolic extract (200 mg/kg, P. O) for 30 consecutive days and administered subcutaneous adrenaline (2 mg/kg, s.c.) divided into two doses (1 mg/kg, s.c) each on the 31st and 32nd days. Liver and kidney function biomarkers in addition to histopathological analyses were evaluated. Adrenaline caused alteration in liver and kidney function biomarkers without affecting the histological structure of the liver and the kidney. SAMWA methanolic extract pretreatment significantly decreased serum urea, uric acid, aspartate aminotransferase (AST), alanine transaminase (ALT) and alkaline phosphatase (ALP). SAMWA ameliorated serum albumin reduction and total protein induced by adrenaline injection. SAMWA methanolic extract could protect liver and kidney of rats exposed to adrenaline. A high-fat diet (HFD) is characterized by an excessive intake of dietary fats, while often being deficient in essential nutrients and fiber. Studies in animal models have consistently demonstrated that high-fat diets can lead to significant elevation of weight gain, body organs and biochemical parameters. The present research aimed to clarify the adverse physiological consequences of HFD on female rats. The present study was performed on 12 female rats that were divided into equal groups. Group I fed normal basal diet and group II fed HFD for 12 months. The body weight and gain, relative organs weight as well as abdominal fat mass were recorded. Hematological parameters, lipid profile, liver and kidney functions and heart fatty acid binding protein (H-FABP) were determined. Also, histopathology for liver, kidney and heart were performed. Electrocardiography (ECG) was performed. The body weight, weight gain, relative organs weight and abdominal fat mass were significantly increased in group II than group I. Hematological parameters, lipid profile, liver and kidney functions, H-FABP and ECG as well as histopathology were deteriorated in group II than group I. The findings revealed that prolonged consumption of a high-fat diet resulted in significant physiological perturbations. Understanding the intricate interactions between high-fat diets, body weight, body organs and biochemical parameters is essential for devising effective prevention and treatment strategies for obesity and related metabolic disorders. For poultry farmers and quails' producers', one of the biggest challenges is dealing with natural diet contaminants like mycotoxins. Worldwide, mycotoxins are present in all feed sources, primarily in corn, and they significantly reduce the health, immune function, and performance of birds. For this purpose, the effect of citric acid (CA) supplementation on diet contaminated with Aflatoxins (AFL) was investigated in male Japanese quail (Coturnix coturnix) through studying hepatic biochemical, histological, and ultrastructural changes. Influences of experimental diets were assessed in 3 replications of 6 birds each (n = 18 per treatment). Quails two weeks old were assigned into 4 equal groups. The control quails fed only basal diet, AFL group quails were given basal diet contaminated with 2.5 mg AFL/kg diet, citric group quails fed basal diet with 10 g citric acid/Kg, and AFL/citric group quails fed basal diet contaminated with 2.5 mg AFL /Kg and augmented with 10 g citric acid/Kg. After four weeks, feeding AFL to quails induced hepatotoxicity as evidenced by significant decline in body weight, serum albumin and total protein while it significantly increased serum ALT, and AST activities. AFL also induces liver oxidative stress by the elevation of lipid peroxidation and reducing GPx, ADH, SOD and catalase activities. Descriptive hepatic histological and ultrastructural alteration were also noted in the AFL group. Treatment with CA induced an increase in total protein, albumin, SOD, GPx, ADH and significantly decreased ALT and AST activities and MDA level. Moreover, it also improved the histological and ultrastructure alternations induced in the liver of AFL group. Myocardial infarction (MI) is a life-threatening ischemic disease and is one of the leading causes of morbidity and mortality worldwide. Serotonin (5-HT) release during myocardial ischemia plays an important role in the progression of myocardial cellular injury. This study was conducted to investigate the possible cardioprotective effect of flibanserin (FLP) against isoproterenol (ISO)-induced MI in rats. Rats were randomly divided into five groups and were treated orally (p.o.) with FLP (15, 30, and 45 mg/kg) for 28 days. ISO was administered subcutaneously (S.C.) (85 mg/kg) on the 27th and 28th days to induce MI. ISO-induced myocardial infarcted rats exhibited a significant increase in cardiac markers, oxidative stress markers, cardiac and serum 5-HT levels, and total cardiac calcium (Ca2+) concentration. ISO-induced myocardial infarcted rats also revealed a remarkable alteration of electrocardiogram (ECG) pattern and significantly upregulated expression of the 5-Hydroxytryptamine 2A (5-HT2A) receptors gene. Moreover, ISO-induced myocardial infarcted rats showed significant histopathological findings of MI and hypertrophic signs. However, pretreatment with FLP significantly attenuated the ISO-induced MI in a dose-dependent manner, as the effect of FLP (45 mg/kg) was more pronounced than that of the other two doses, FLP (15 and 30 mg/kg). This study provided a preliminary framework for the effects of Ropadiar (Origanum vulgare L.) on Nile tilapia (Oreochromis niloticus) on some immune parameters over a 60-day feeding trial and after bacterial challenge with Aeromonas hydrophila. A total of 60 O. niloticus 14.5-15.3 g were divided into two groups (10 fish/aquarium/3 replicate). The first group was fed a basal diet, which served as control, the second group was fed 2% Oreganum oil added to the diet for 60 days, then they were inoculated with Aeromonas hydrophila. Hematological parameters and cytokines levels, phagocytic index and nitric oxide level (NO) as well as serum lysozyme activity were determined. Histopathology for the spleen was done. Results revealed significant improvement in hemoglobin (HB), red blood cells (RBCs) and white blood cells (WBCs) count, lysozyme, phagocytic activity, nitric oxide levels and cytokines up-regulation in 2% oreganum oil supplemented group than the control. Histopathology for the spleen was significantly improved in 2% of the oreganum oil-supplemented group compared to the control. The incorporation of Ropadiar 2% in the diet of Nile tilapia is recommended to improve hematological, histopathological and immune parameters in Nile tilapia. [ABSTRACT FROM AUTHOR]

Background: Isoflavones are estrogenic compounds that exist in soy, clover, and peanuts. They are selective estrogen receptor modulators. Aim: The study was planned to explain the interactions of isoflavones with estrogen receptors alpha (ERα), beta (ERβ), and vascular endothelial growth factor (VEGF) expressions in ovarian and uterine tissues during different stages of the estrous cycle of regular cyclic female Wistar rats. Methods: Thirty-two regular cyclic females were divided equally into control group: fed casein-based diet and isoflavones group: fed casein-based diet and gavaged 50 mg/kg/day soy isoflavones extract 40%. The regularity of estrus cycles was monitored. Final body weight (FBW), weight gain (BWG), and ovarian and uterine weights were estimated. Histopathology and immunohistochemistry for ERα, Erβ, and VEGF in ovarian and uterine tissues were performed. Results: All females (100%, n = 16) in control group showed regularity in estrous cycle compared to 62.5% (n = 10) in isoflavones group. Estrus and diestrus phases revealed prolongation and shortening in isoflavones rats than control, respectively. Nonsignificant variation was noted in the duration of the whole cycle of both groups. FBW and BWG significantly decreased however, ovarian and uterine weights increased significantly in all estrous phases of isoflavones group than control. Histopathology demonstrated an increase in number of follicles/ovaries besides, hyperplasia and proliferation of luminal epithelium with hydropic degeneration in the isoflavones group. Also, uterine connective tissue stroma showed edema in the isoflavones group during all estrous phases. Immunostaining percentages of ERα, Erβ, and VEGF protein expression were significantly elevated in the isoflavones group during all estrous phases. Conclusion: Isoflavones induced irregularity of the estrous cycle that was encountered by increased and altered ERα, Erβ, and VEGF expressions in ovarian and uterine tissues. [ABSTRACT FROM AUTHOR]

The present study investigated the possible ameliorative effect of Beta-MOS® on lead-induced toxicity in Nile tilapia. A total of ninety O. niloticus (34.43 ± 0.31 g and 7 weeks age) were used in this study. Fish of mixed sex were divided into 4 groups. The first group which served as control received a basal diet (0% Beta-MOS®). The second group received a diet supplemented with 0.3% Beta-MOS®. The third group was exposed to 10 mg Pb acetate L-1 in water and received a basal diet. The fourth group was exposed to the same dose of Pb acetate (10 mg L-1 ) in water and given 0.3% Beta-MOS®. Initial and final weight as well as weight gain were recorded. Red blood cell count and hemoglobin concentration were determined. Creatinine plasma level was estimated as well as histopathological changes in the kidney. The lead acetate induced a significant reduction in final body weight, weight gain, red blood cell count and hemoglobin concentration than control. Moreover, creatinine was significantly increased in the lead group than control, with retrogressive changes in histopathological sections. Background: Sodium nitrate has been used extensively as a meat additive to keep meat products fresh. Several strategies have been implemented in counteracting such toxicity. Objective: The goal of the current investigation was to determine if ascorbic acid and crude honey extract can protect male rats from the harmful biochemical and histopathological effects of sodium nitrate. Materials and methods: 36 male Albino rats were alienated into 6 groups; control was given saline, honey group was given (2.5g/kg/day honey, ascorbic group was given (200mg/kg/day) L-ascorbic acid, NaNO3 group was given 200mg/kg/day, honey/NaNO3 group rats given honey and nitrate with the former doses, and Ascorbic/NaNO3 group was given L-ascorbic acid and NaNO3 with the former doses. Organs' relative weight, liver and kidney blood biochemical determinants, serum tumor necrosis factor alpha (TNF-a) and interleukin 6 (IL-6), histology and histochemistry were determined. Results: The sodium nitrate had significantly increased liver, kidney and spleen relative weights while decreased thymus weight. Higher levels of AST, ALT, urea, creatinine, IL-6 and TNF-a were observed in NaNO3 treated group. Liver, kidney, and spleen of rats treated with NaNO3 revealed histological and histochemical alterations. The treatment with L-ascorbic acid and crude honeybee improved the later dysfunctions with more palliative effect for L-ascorbic acid. Conclusions: We can conclude that sodium nitrate has to be limited in use. L-ascorbic acid was therefore recommended to be used in reducing the harmful metabolic alterations induced by sodium nitrate better than the honeybee. [ABSTRACT FROM AUTHOR]

Since the role of estrogen in postmenauposal-associated dementia is still debatable, this issue urges the search for other medications. Dimethyl fumarate (DMF) is a drug used for the treatment of multiple sclerosis and has shown a neuroprotective effect against other neurodegenerative diseases. Accordingly, the present study aimed to evaluate the effect of DMF on an experimental model of Alzheimer disease (AD) using D-galactose (D-Gal) administered to ovariectomized (OVX) rats, resembling a postmenopausal dementia paradigm. Adult 18-month old female Wistar rats were allocated into sham-operated and OVX/D-Gal groups that were either left untreated or treated with DMF for 56 days starting three weeks after sham-operation or ovariectomy. DMF succeeded to ameliorate cognitive (learning/short- and long-term memory) deficits and to enhance the dampened overall activity (NOR, Barnes-/Y-maze tests). These behavioral upturns were associated with increased intact neurons (Nissl stain) and a reduction in OVX/D-Gal-mediated hippocampal CA1 neurodegeneration and astrocyte activation assessed as GFAP immunoreactivity. Mechanistically, DMF suppressed the hippocampal contents of AD-surrogate markers; viz., apolipoprotein (APO)-E1, BACE1, Aβ42, and hyperphosphorylated Tau. Additionally, DMF has augmented the neuroprotective parameters p-AKT, its downstream target CREB and BDNF. Besides, it activated AMPK, and enhanced SIRT-1, as well as antioxidant defenses (SOD, GSH). On the other hand, DMF inhibited the transcription factor NF-κB, IL-1β, adiponectin/adiponectin receptor type (AdipoR)1, GSK-3β, and MDA. Accordingly, in this postmenopausal AD model, DMF treatment by pursuing the adiponectin/AdipoR1, AMPK/SIRT-1, AKT/CREB/BDNF, AKT/GSK-3β, and APO-E1 quartet hampered the associated tauo-/amyloidopathy and NF-κB-mediated oxidative/inflammatory responses to advance insights into its anti-amnesic effect., (Copyright © 2021 Elsevier Ltd. This study has investigated the effect of using the Uncaria tomentosa (UT) extract against immunotoxicity that induced by fipronil (FP) in male Wistar rats. Twenty-eight, male Wistar rats were assigned randomly into four groups (7 rats each). Control group received distilled water. FP group received FP 9.7 mg/kg b. wt orally via gastric tube. UT group received 120 mg/kg b. wt. of UT extract orally. FP-UT group received both FP and UT (9.7 and 120 mg/kg b.wt, respectively) for 30 days. Hematological parameters, malondialdehyde (MDA), total antioxidant capacity (TAC), estradiol, histamine and immunoglobulin E (IGE) were assayed. Histopathological and electron microscopical examinations were performed to the lymphoid organs. Hematological parameters, were decreased in the FP group than the control group. There was a rise in MDA of FP group followed by a decrease in TAC content with histological and ultrastructure degenerative changes. UT extract treatment ameliorated the FP-induced perturbations for the former parameters. The results showed that FP treatment exerted an immunotoxic effect through acting as an endocrine disruptor and allergic, pro-inflammatory that was confirmed by histopathological and ultrastructure study of the lymphoid organs. Present research explored the anti-obesity effect of Moringa olifera seed oil extract and lycopene (LYC). Forty eight male Sprauge Dawely rats were divided equally into 6 groups. Group Ι (C) served as control, group ΙΙ (MC) was given Moringa olifera seed oil extract (800 mg/kg b.wt) for 8 weeks, group ΙΙΙ (LC) was given (20 mg/kg b.wt) LYC for 8 weeks, group ΙV (O) received high fat diet (HFD) for 20 weeks, group Ѵ (MO), was given HFD for 20 weeks and received (800 mg/kg b.wt) Moringa olifera seed oil extract for last 8 weeks and group ѴΙ (LO), received HFD for 20 weeks and was given (20 mg/kg b.wt) LYC for last 8 weeks. Hematology, lipid peroxidation and antioxidants, non-esterified fatty acids (NEFA), glucose, lipid profile, serum liver and kidney biomarkers, inflammatory markers, leptin, resistin and heart fatty acid binding protein (HFABP) were determined. Also histopathology for liver, kidney and aorta were performed besides immunohistochemistry (IHC) for aortic inducible nitric oxide synthase (iNOS). Administration of Moringa olifera seed oil extract and LYC significantly ameliorated the HFD induced hematological and metabolic perturbations as well as reduced leptin and resistin. Both treatments exerted these effects through promotion of antioxidant enzymes and reducing lipid peroxidation as well as inflammatory cytokines along with reduced iNOS protein expression.
