Wound healing is a critical process essential for the body’s recovery from injuries, often complicated by bacterial infections. Silver nanoparticles (AgNPs) have gained attention due to their antibacterial and tissue-regenerative properties. However, conventional chemical synthesis methods for AgNPs pose environmental risks. This study utilizes Ziziphus spina-christi (ZSC) extract for the eco-friendly synthesis of AgNPs, evaluating their antibacterial and wound-healing capabilities. The AgNPs-ZSC showed an absorption maximum at λ max of 460 nm, a particle size of 111.2 ± 1.09 nm, a polydispersity index of 0.38 ± 0.006, and a zeta potential of −27.0 ± 0.231 mV. The synthesized AgNPs-ZSC were spherical, non-aggregated, and exhibited potent antibacterial activity superior to chloramphenicol. Furthermore, the AgNPs-ZSC cream significantly promoted wound closure, epithelial tissue proliferation, and granulation tissue formation in rats, showing no signs of toxicity or adverse reactions. In conclusion, AgNPs-ZSC cream demonstrates excellent antibacterial and wound-healing properties, presenting a sustainable alternative to conventional chemical methods for AgNP synthesis.

Sorafenib (SFB) is an anticancer drug with sparingly water solubility and reduced bioavailability. Nanoformulation of SFB can increase its dissolution rate and solubility. The current study aimed to formulate SFB in nanoparticles to improve their solubility. The sorafenib nanoparticles (SFB-PNs) were synthesized using the solvent evaporation method, then evaluated for their particle size, polydispersity index (PDI), zeta-potential, morphological structure, and entrapment efficiency (EE%). Further, the anticancer efficacy in A549 and Michigan Cancer Foundation-7 (MCF-7) cancer cell lines was evaluated. The SFB-NPs were uniform in size, which have 389.7 ± 16.49 nm, PDI of 0.703 ± 0.12, and zeta-potential of −13.5 ± 12.1 mV, whereas transmission electron microscopy showed a well-identified spherical particle. The EE% was found to be 73.7 ± 0.8%. SFB-NPs inhibited the cell growth by 50% after 48 h incubation, with IC50 of 2.26 and 1.28 µg/mL in A549 and MCF-7, respectively. Additionally, SFB-NPs showed a significant decrease (P < 0.05) in p21, and stathmin-1 gene expression levels in both cell lines. Moreover, SFB-NPs showed a significant increase in DNA damage of 25.50 and 26.75% in A549 and MCF-7, respectively. The results indicate that SFB-NPs are a potential candidate with an effective anticancer agent compared with free drugs.

Drug uptake and distribution through cell–receptor interactions are of prime interest in reducing the adverse effects and increasing the therapeutic effectiveness of delivered formulations. This study aimed to formulate silver nanoparticles (AgNPs) conjugated to somatostatin analogs for specific delivery through somatostatin receptors (SSTRs) expressed on cells and by nebulizing the prepared AgNPs formulations into lung cells for in vivo application. AgNPs were prepared using the citrate reduction method, yielding AgNPs–CTT, which was further chemically conjugated to octreotide (OCT) to form AgNPs–OCT through an amide linkage. The AgNPs–OCT formulation was coated using alginate to yield a carrier, AgNPs–OCT–Alg, feasible for drug delivery through nebulization. AgNPs were uniform in size with an acceptable range of zeta potential. Furthermore, the concentrations of AgNP formulations were found safe for the model cell lines used, and cell proliferation was significantly reduced in a dose-dependent manner (p < 0.05). In the healthy lung tissues, AgNPs–OCT–Alg accumulated at a concentration of 0.416 ± 5.7 mg/kgtissue, as determined via inductively coupled plasma optical emission spectrometry. This study established the accumulation of AgNPs, specifically the AgNPs–OCT–Alg, in lung tissues, and substantiated the active, specific, and selective targeting of SSTRs at pulmonary sites. The anticancer efficacy of the formulations was in vitro tested and confirmed in the MCF-7 cell lines. Owing to the delivery suitability and cytotoxic effects of the AgNPs–OCT–Alg formulation, it is a potential drug delivery formulation for lung cancer therapy in the future.

Nano-based systems can be used to transport active medicinal products to specific parts of the body. Most challenges with drug delivery, such as low water solubility and poor bioavailability, can be solved using nanotechnology. In addition, nanoparticles can overcome various physiological obstacles to increase load distribution to desired sites. Nanoparticles can carry a load of medication or therapeutic agent, such as a DNA-related substance, to enhance distribution time and deliver the drug to the target site in either a nonspecific (through enhanced permeability and retention (EPR)) or specific (through binding specific target receptors) manner. Moreover, nanoparticle drug delivery systems have been employed in the clinic since the early 1990s. Since then, the field of nanomedicine has developed with growing technical needs to improve the delivery of various medications. Over these past decades, newer generations of nanoparticles have emerged that are capable of conducting new delivery activities that could enable therapy via innovative therapeutic modalities. This review highlights different types of approved and currently marketed nanoparticles, such as nanocrystals, liposomes, lipid nanoparticles, PEGylated polymeric nanoparticles, protein-based nanoparticles, and metal-based nanoparticles. Furthermore, it explores the use of vaccine-loaded nanoparticles for COVID-19 prophylaxis.

This review focuses on nano-structured delivery devices prepared from biodegradable and biocompatible natural and synthetic polymers, organic raw materials, metals, metal oxides, and their other compounds that culminated in the preparation of various nano-entities depending on the preparative techniques, and starting raw materials’ utilizations. Many nanoparticles (NPs) made of polymeric, metallic, magnetic, and non-magnetic origins, liposomes, hydrogels, dendrimers, and other carbon-based nano-entities have been produced. Developments in nanomaterial substrate and end products’ design, structural specifications, preparative strategies, chemo-biological interfacing to involve the biosystems interactions, surface functionalization, and on-site biomolecular and physiology-mediated target-specific delivery concepts, examples, and applications are outlined. The inherent toxicity, and safety of the design concepts in nanomaterial preparation, and their applications in biomedical fields, especially to the organs, cellular and sub-cellular deliveries are deliberated. Bioapplications, the therapeutic delivery modules’ pharmacokinetics and medicinal values, nanopharmaceutical designs, and their contributions as nano-entities in the healthcare biotechnology of drug delivery domains have also been discussed. The importance of site-specific triggers in nano-scale deliveries, the inherent and induced structural specifications of numerous nanomaterial entities belonging to NPs, nano-scale composites, nano-conjugates, and other nano-devices of organic and inorganic origins, near biological systems are detailed. Modifications that provide nano-deliveries of their intrinsic therapeutic actions, through structural and physicochemical characteristics modifications, and the proven success of various nano-delivery devices and currently available commercial nanomedicinal and nanopharmaceutical products are also provided.

Herein, we report on the development of chitosan-capped silver nanoparticles (AgNPs-CHI) with an intrinsic activity against breast cancer cells. Following chemical synthesis via a simple, one-pot reaction, the chitosan coating of AgNPs was verified using Fourier-transform infrared and ultraviolet–visible spectroscopies. The physicochemical properties and morphology were characterized using dynamic light scattering, scanning electron microscopy, and transmission electron microscopy. The shelf stability of the optimized platform was tracked for 3 months upon storage at either room temperature or 4°C. Then, the anticancer activities of AgNPs-CHI on human breast cancer cells, MCF-7, versus normal human cells, human skin fibroblasts (HSF), were evaluated via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cytotoxicity assay and tumor-associated biomarkers determination by enzyme-linked immunosorbent assay, in comparison with plain silver nitrate (AgNO3) solution. AgNPs were successfully coated with chitosan and demonstrated acceptable physicochemical properties, with a spherical morphology and high stability upon long-term storage. Although AgNPs-CHI and AgNO3 demonstrated comparable cytotoxicity to MCF-7 cells, AgNPs-CHI resulted in 10-fold lower toxicity to HSF cells, suggesting a higher selectivity. In addition, AgNPs-CHI lowered IL-6 and tumor necrosis factor-alpha levels in MCF-7 cells by 90 and 30%, respectively, compared to 60 and 10% in the case of plain AgNO3. The interesting therapeutic modality presented in this study is promising for potential clinical applications.

Background: Obesity has become a prevalent issue worldwide, leading to various complications such as hyperlipidemia, diabetes, and cardiovascular problems. Statins, as FDA approved anti-hyperlipidemic drugs, still pose some concerns upon their administration. Recently, researchers have looked for natural products as an alternative to manage hyperlipidemia and obesity. Aim: This work aimed to study the hypolipidemic effect of Lepidium sativum garden cress (GC) from different preparations; orally administered seeds, and hydrogel, in comparison to atorvastatin. Methods: GC hydrogel was prepared from the GC aqueous extract and pharmaceutically evaluated for its pH, spreadability, seeds content, homogeneity, rheology, and in vitro release. The rat's body weight, blood glucose levels, total lipid profile, and liver biomarkers were evaluated on obese rats for one month. In addition, the histopathology study was also performed. Results: GC hydrogel had acceptable pharmaceutical properties and showed a sustained release performance over 24 h. Oral and topical GC significantly reduced the lipid profiles, blood sugar and ALT, AST levels more than the negative control group and comparable to atorvastatin. It was found that oral GC showed a significant effect on the percentage decrease in the rat's body weight than the applied hydrogel. Histopathology study revealed a better outcome in the histological structure of pancreas and liver compared with rats feed on high fat diet post-treatment for one month. Conclusion: GC orally administered, or topically applied hydrogel could be a promising, safe alternative formulation to atorvastatin in managing hyperlipidemia and normalizing body weight of obese rats. Background: A fracture is considered a medical emergency leading to considerable complications. Aim: This study aimed to describe the accelerating action of Ag-NPs-FG on fracture healing in rabbits. Methods: Silver NPs (AgNPs) were reduced with fenugreek (FG), loaded into a starch gel base, and investigated for their morphology, size, and charge. Four equal groups were randomly formed of 40 adult male rabbits. A 3.5 mm diameter bone defect was created at the proximal metaphysis of the right tibia in each rabbit. Groups 1-4 were injected with placebo saline, AgNPs-FG, plain gel, and FG-gel at the bone defect zone, respectively. The healing was assessed for 8 weeks postoperatively based on the radiographic, bone turnover markers, and histopathological examinations. Results: The AgNPs-FG was obtained as a faint reddish color, spherical in shape, with an absorbance of 423 nm, a size of 118.0 ± 1.7 nm, and a surface charge of -7.8 ± 0.518 mV. The prepared AgNPs-FG hydrogel was clear, translucent, and homogenous. The pH values were 6.55-6.5 ± 0.2, the viscosity of 4,000 and 1,875 cPs, and spreadability of 1.6 ± 0.14 and 2.0 ± 0.15 for both FG and AgNPs-FG hydrogel, respectively. The radiographic union scale was significantly (p < 0.05) improved in group 2 with a significant (p < 0.05) increase in bone turnover markers was found in comparison to other treated groups. Histopathological examination revealed the formation of mature bone on the 28th postoperative day in groups 2 and 4. Conclusion: Colloidal nano-formulation of AgNPs-FG loaded hydrogel could be a promising formulation to accelerate rabbits' tibial bone healing process. [ABSTRACT FROM AUTHOR]

Lepidium sativum, Garden Cress (GC), seeds have a lot of natural molecules with a pronounced activity against different disorders. It was reported that GC seeds have the ability to lower the blood glucose level. The aim of this work was to formulate GC seeds into oral tablets containing a fixed dose of the grounded seeds. Furthermore, the anti-diabetic performance of the prepared tablets was studied in the streptozotocin rats' model in comparison with positive control metformin. Micrometrics of GC grounded seeds with different excipients were investigated. Then, GC tablets were prepared via direct compression technique. GC tablets were characterized for their uniformity of dosage unit, friability, hardness, disintegration time, and in vitro release. The antidiabetic effect was studied in rats for a period of 28 days. Glycosylated hemoglobin, liver performance, and lipid levels include total cholesterol (TC), triglycerides (TGs), high-density lipoprotein (HDL), and low-density lipoprotein (LDL) were also estimated. In addition, histopathological study of liver and pancreas was also performed. Prosolv®EasyTab produced tablets with higher hardness, lower disintegration time, and fast release. GC tablets significantly lower the elevated blood glucose level. In addition, they have antihyperlipidemic activity, hepatocellular protective role and restore the histology of the liver and pancreas. Bacterial infections caused by different strains of bacteria still one of the most important disorders affecting humans worldwide. Polymers nanocomposite systems could be considered as an alternative to conventional antibiotics to eradicate bacterial infections. In an attempt to enhance the antibacterial performance of silver and iron oxide nanoparticles, decrease their aggregation and toxicity, a polymeric hybrid nanocomposite system combining both nanoparticles is produced. Magnetic Ag–Fe3O4@polymer hybrid nanocomposites prepared using different polymers, namely polyethylene glycol 4000, ethyl cellulose, and chitosan were synthesized via wet impregnation and ball-milling techniques. The produced nanocomposites were tested for their physical properties and antibacterial activities. XRD, FT-IR, VSM, and TEM results confirmed the successful preparation of hybrid nanocomposites. Hybrid nanocomposites have average crystallite sizes in the following order Ag–Fe3O4@CS (8.9 nm) < Ag–Fe3O4@EC (9.0 nm) < Ag–Fe3O4@PEG4000 (9.4 nm) and active surface area of this trend Ag–Fe3O4@CS (130.4 m2g−1) > Ag–Fe3O4@EC (128.9 m2g−1) > Ag–Fe3O4@PEG4000 (123.4 m2g−1). In addition, they have a saturation magnetization in this order: Ag–Fe3O4@PEG4000 (44.82 emu/g) > Ag–Fe3O4@EC (40.14 emu/g) > Ag–Fe3O4@CS (22.90 emu/g). Hybrid nanocomposites have a pronounced antibacterial action against Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus intermedius compared to iron oxide nanoparticles and positive antibacterial drug. In addition, both Ag–Fe3O4@EC and Ag–Fe3O4@CS have a lower MIC values compared to Ag–Fe3O4@PEG and positive control. Magnetic Ag–Fe3O4 hybrid nanocomposites could be promising antibacterial nanomaterials and could pave the way for the development of new materials with even more unique properties and applications. Predicting dorsalgia involves a multifaceted approach that encompasses the analysis of demographic, lifestyle, and medical data. Machine learning algorithms and advanced data analytics play a pivotal role in forecasting the risk of developing back pain. Early prediction aids in proactive interventions and personalized healthcare strategies, thereby mitigating the burden of dorsalgia on individuals and healthcare systems. The proposed feature selection is the initial feature set's most educational elements by evolutionary gravitational search-based feature selection (EGSFS). Specifically, the framework is trained and fine-tuned using spinal geometry parameters, enabling precise identification of individuals at risk of developing dorsalgia. This study presents a novel approach for classification tasks using a Genetic Algorithm (GA)-optimized hybrid Convolutional Neural Network (CNN) and Long Short-Term Memory (LSTM) model. The GA optimizes the model's architecture and hyperparameters to enhance its performance. The framework is implemented using Python. In the categorization procedure, the Single Neutrosophic sets aid in capturing ambiguity, which is particularly beneficial when handling dorsalgia disorders that may present with confusing symptoms, thus enhancing the accuracy of classifying various dorsalgia conditions. Experimental results demonstrate that this hybrid approach significantly improves classification accuracy, making it a viable option for several practical applications. Experimental results exhibit remarkable improvements in accuracy and predictive power, underscoring the potential of this innovative approach in advancing preventative and personalized healthcare strategies for back pain management. The experiment was built on the lower back pain symptoms dataset. A comparison is made between the experimental results and previous prediction models like Logistic Regression, Decision Tree Classifier, Random Forest, and Support Vector Machine in terms of accuracy, F1-score, precision, and recall. Driver drowsiness has been identified as a major cause of roadway accidents globally. Efficiently determining the extent of drowsiness can greatly enhance preventive measures. This study proposes a novel approach, combining convolutional neural networks (CNN) and Gated Recurrent Units (GRU) to dynamically evaluate both the presence of drowsiness and its severity based on the Eye Aspect Ratio (EAR). By bridging spatial features extracted by CNNs with temporal sequences through GRU, our model offers a robust and real-time assessment of drowsiness levels, paving the way for enhanced safety measures in vehicular systems. Incorporating Neutrosophic Logic enables a more robust representation of uncertainty and ambiguity in the data and enhances the accuracy of driver drowsiness level detection within the Hybrid CNN-GRU framework. The model's hybrid CNN-GRU structure combines CNN layers to extract spatial information from Human eye Images and GRU units to represent temporal correlations between frames. In-car cameras and sensors must be integrated to implement the suggested system in real-time and enable continuous driver behavior monitoring. The system alerts early warnings and takes action when drowsiness is detected, lowering the likelihood of accidents caused by weary drivers. The CNN-GRU hybrid architecture accurately detects fatigue during real-time driving. Performance metrics, including accuracy, recall, and F1-score, are provided for comparative research utilizing baseline models. Model behavior may be understood by visualizing tiredness detection and carefully examining false positives and negatives. The proposed CNN-GRU framework outperforms traditional methods such as SVM, KNN, and BPNN by achieving a significantly higher accuracy of 99.5%. It increases the recognition of driver tiredness by proposing a trustworthy and adaptable hybrid CNN-GRU deep learning system. This project is implemented in Python; it offers a practical and versatile solution for real-time driver drowsiness level detection. Chemotherapeutic agents have numerous side effects. There is a major interest in using natural and safe plants as food or drink to prevent from cancer. Origanum marjoram (OMAE) is a medicinal plant that can be used as a tea, food, and additive in traditional medicine. This study aimed to evaluate the potential anticancer effects of OMAE as a soft drink for daily use against a model cancer, prevention and treatment. MCF-7 cells were chosen as model cancer cells. The MTT assay was used to assess the in vitro inhibitory effects of OMAE on cell growth. Moreover, quantitative real‐time PCR (qRT-PCR) was used to detect specific genes associated with cancer, such as ESR1, Bax, Bcl-2, and p53. Furthermore, the DNA damage was evaluated using the comet assay. OMAE has IC50 of 53.1 and IC90 of 97.5 μg/ml dependent inhibition of cell proliferation after 48 h of treatment toward MCF-7. Also, a significant decrease in the expression level of the ESR1 gene in the MCF-7 cell line. Furthermore, there was a significant increase in the comet length and comet-positive cells after treatment with OMAE (88.7%) compared with those in the untreated control cells (9.5%), suggesting a high induction of DNA damage by OMAE. Also, OMAE showed a modification in bcl-2, tumor suppressor gene (p53), and Bax levels and influenced the BAX/BCL-2 ratio via releasing the cytochrome C. The results of the study were promising, suggesting that the reduced apoptotic rate of MCF-7 breast cancer cells in this work was correlated to the potential anticancer effect of OMAE which would be a suitable preventable drink against cancer. The topical delivery of therapeutics is a promising strategy for managing skin conditions. Cyclooxygenase-2 (COX-2) inhibitors showed a possible target for chemoprevention and cancer management. Celecoxib (CXB) is a selective COX-2 inhibitor that impedes cell growth and generates apoptosis in different cell tumors. Herein, an investigation proceeded to explore the usefulness of nano lipid vesicles (transethosomes) (TES) of CXB to permit penetration of considerable quantities of the drug for curing skin cancer. The prepared nanovesicles were distinguished for drug encapsulation efficiency, vesicle size, PDI, surface charge, and morphology. In addition, FT-IR and DSC analyses were also conducted to examine the influence of vesicle components. The optimized formulation was dispersed in various hydrogel bases. Furthermore, in vitro CXB release and ex vivo permeability studies were evaluated. A cytotoxicity study proceeded using A431 and BJ1 cell lines. The expression alteration of the cyclin-dependent kinase inhibitor 2A (CDKN2A) gene and DNA damage and fragmentation using qRT-PCR and comet assays were also investigated. Optimized CXB-TES formulation was spherically shaped and displayed a vesicle size of 75.9 ± 11.4 nm, a surface charge of −44.7 ± 1.52 mV, and an entrapment efficiency of 88.8 ± 7.2%. The formulated TES-based hydrogel displayed a sustained in vitro CXB release pattern for 24 h with an enhanced flux and permeation across rat skin compared with the control (free drug-loaded hydrogel). Interestingly, CXB-TES hydrogel has a lower cytotoxic effect on normal skin cells compared with TES suspension and CXB powder. Moreover, the level of expression of the CDKN2A gene was significantly (p ≤ 0.01, ANOVA/Tukey) decreased in skin tumor cell lines compared with normal skin cell lines, indicating that TES are the suitable carrier for topical delivery of CXB to the cancer cells suppressing their progression. In addition, apoptosis demonstrated by comet and DNA fragmentation assays was evident in skin cancer cells exposed to CXB-loaded TES hydrogel formulation. In conclusion, our results illustrate that CXB-TES-loaded hydrogel could be considered a promising carrier and effective chemotherapeutic agent for the management of skin carcinoma.
