1. Predictive impact of rare genomic copy number variations in siblings of individuals with autism spectrum disorders.

Abstract

Identification of genetic biomarkers associated with autism spectrum disorders (ASDs) could improve recurrence prediction for families with a child with ASD. Here, we describe clinical microarray findings for 253 longitudinally phenotyped ASD families from the Baby Siblings Research Consortium (BSRC), encompassing 288 infant siblings. By age 3, 103 siblings (35.8%) were diagnosed with ASD and 54 (18.8%) were developing atypically. Thirteen siblings have copy number variants (CNVs) involving ASD-relevant genes: 6 with ASD, 5 atypically developing, and 2 typically developing. Within these families, an ASD-related CNV in a sibling has a positive predictive value (PPV) for ASD or atypical development of 0.83; the Simons Simplex Collection of ASD families shows similar PPVs. Polygenic risk analyses suggest that common genetic variants may also contribute to ASD. CNV findings would have been pre-symptomatically predictive of ASD or atypical development in 11 (7%) of the 157 BSRC siblings who were eventually diagnosed clinically.

Abstract

We report the analysis of 1 year of data from the first cohort of 15 patients enrolled in an open-label, first-in-human, dose-escalation phase I study (ClinicalTrials.gov: NCT03282760, EudraCT2015-004855-37) to determine the feasibility, safety, and tolerability of the transplantation of allogeneic human neural stem/progenitor cells (hNSCs) for the treatment of secondary progressive multiple sclerosis. Participants were treated with hNSCs delivered via intracerebroventricular injection in combination with an immunosuppressive regimen. No treatment-related deaths nor serious adverse events (AEs) were observed. All participants displayed stability of clinical and laboratory outcomes, as well as lesion load and brain activity (MRI), compared with the study entry. Longitudinal metabolomics and lipidomics of biological fluids identified time- and dose-dependent responses with increased levels of acyl-carnitines and fatty acids in the cerebrospinal fluid (CSF). The absence of AEs and the stability of functional and structural outcomes are reassuring and represent a milestone for the safe translation of stem cells into regenerative medicines.

Abstract

The degradation of PK99 was studied by heating the polymer in both isothermal and dynamic conditions under nitrogen flow and in a static air atmosphere. In both environments this polymer's thermal stability exceeded that of PEEK. The dynamic experiments showed that in both nitrogen and air atmospheres the polymer degraded in two stages with formation of the same residue, which was quite stable under nitrogen, while almost completely burned in air. Good agreement was observed with the isothermal experiments. Apparent activation energy values were evaluated. Abstract

OBJECTIVE: An increased cardiovascular mortality and morbidity has been widely reported in patients with atrial fibrillation (AF). In this study, a subanalysis of the AntiThrombotic Agents Atrial Fibrillation (ATA‐AF) is performed with the aim to evaluate estimated glomerular filtration rate (eGFR) as an independent prognostic marker of cardiovascular mortality and morbidity in patients with AF. METHODS AND RESULTS: The ATA‐AF study enrolled 7148 patients with AF, in 360 Italian centers. The eGFR was calculated from data reported in patient notes or hospital database. This post‐hoc analysis included 1097 AF patients with eGFR data available and 1‐year clinical follow‐up. The endpoint was assessed as cardiovascular mortality and/or hospital admission for cardiovascular causes at follow‐up. Patients were also divided in two groups according to the eGFR (

Abstract

El visón (Mustela vison) es el hospedador que brinda mas especificidad al nematodo Dioctophyma renale (Goeze, 1782) en él, el verme manifiesta totalmente su tropismo y alcanza su localización definitiva,en tejido renal, logra así su cometido, desde allí podrá continuar su ciclo biológico. Son casuales las localizaciones ectópicas, encontradas en Mustela vison. Sin embargo, en los caninos, hospedadores menos específicos, se observan mas frecuentemente las localizaciones aberrantes. Algunas formas extrarrenales son: las subcutáneas, abdominales, torácicas,vesicales, hepáticas y bronquiales. La Ciudad de Berisso, situada en la costa ribereña del Río de La Plata, presenta características geográficas, que propician, determinadas parasitosis de ciclo biológico acuático, entre ellas la Dioctofimosis., Facultad de Ciencias Veterinarias

Abstract

We have developed a technique and a prototype for [Beta]-ray quantitative imaging of biological samples which have been radioactively labeled. The prototype is based on a solid state pixel detector, bump-bonded to a low threshold, single particle counting chip: the detector we used is 200-[micro]m thick GaAs, with pixel size 170 [micro]m. In this paper we describe an experiment that shows the capability of monitoring a dynamic process. Specifically, [[sup.14]C]L-Leucine uptake by eggs of mollusk cephalopod Octopus vulgaris and the consequent migration of marker into the ooplasm was followed.

Contains fulltext : 171920.pdf (publisher's version ) (Open Access), Anopheles gambiae s.s. mosquitoes are efficient vectors for Plasmodium falciparum, although variation exists in their susceptibility to infection. This variation depends partly on the thioester-containing protein 1 (TEP1) and TEP depletion results in significantly elevated numbers of oocysts in susceptible and resistant mosquitoes. Polymorphism in the Plasmodium gene coding for the surface protein Pfs47 modulates resistance of some parasite laboratory strains to TEP1-mediated killing. Here, we examined resistance of P. falciparum isolates of African origin (NF54, NF165 and NF166) to TEP1-mediated killing in a susceptible Ngousso and a refractory L3-5 strain of A. gambiae. All parasite clones successfully developed in susceptible mosquitoes with limited evidence for an impact of TEP1 on transmission efficiency. In contrast, NF166 and NF165 oocyst densities were strongly reduced in refractory mosquitoes and TEP1 silencing significantly increased oocyst densities. Our results reveal differences between African P. falciparum strains in their capacity to evade TEP1-mediated killing in resistant mosquitoes. There was no significant correlation between Pfs47 genotype and resistance of a given P. falciparum isolate for TEP1 killing. These data suggest that polymorphisms in this locus are not the sole mediators of immune evasion of African malaria parasites.

The juggernaut of technological development has and will continue to dramatically alter how we as health professionals in the neurosciences pursue our efforts in research, training and treatment.
