Multifunctional probes play an increasing role even beyond applications in biomedicine. Multifunctionality introduced by the dual types of complementary probes is always attractive because, in this case, functionalized objects inherit the function of both materials. Porous calcium carbonate microparticles are becoming popular carriers of biomolecules and biosensors, as well as imaging enhancers. We demonstrate here a dual function of these carriers by incorporating both magnetic and silver nanoparticles. Magnetic nanoparticles enable movements and displacements by a magnetic field, while silver nanoparticles provide surface-enhanced Raman signal amplification necessary for the detection of biomolecules. Application of such dual-functional carriers is foreseen beyond the applications of biomedicine and theranostics. [ABSTRACT FROM AUTHOR]

The status of sentinel lymph nodes (SLNs) has a substantial prognostic value because these nodes are the first place where cancer cells accumulate along their spreading route. Routine SLN biopsy ("gold standard") involves peritumoral injections of radiopharmaceuticals, such as technetium‐99m, which has obvious disadvantages. This review examines the methods used as "gold standard" analogs to diagnose SLNs. Nonradioactive preoperative and intraoperative methods of SLN detection are analyzed. Promising photonic tools for SLNs detection are reviewed, including NIR‐I/NIR‐II fluorescence imaging, photoswitching dyes for SLN detection, in vivo photoacoustic detection, imaging and biopsy of SLNs. Also are discussed methods of SLN detection by magnetic resonance imaging, ultrasonic imaging systems including as combined with photoacoustic imaging, and methods based on the magnetometer‐aided detection of superparamagnetic nanoparticles. The advantages and disadvantages of nonradioactive SLN‐detection methods are shown. The review concludes with prospects for the use of conservative diagnostic methods in combination with photonic tools. [ABSTRACT FROM AUTHOR]

In recent years, luminescent materials doped with Ln3+ ions have attracted much attention for their application as optical thermometers based on both downshifting and upconversion processes. This study presents research done on the development of highly sensitive optical thermometers in the physiological temperature range based on poly(methyl methacrylate) (PMMA) films doped with two series of visible Ln3+ complexes (Ln3+=Tb3+, Eu3+, and Sm3+) and SiO2 nanoparticles (NPs) coated with these PMMA films. The best performing PMMA film doped with Tb3+ and Eu3+ complexes was the PMMA[TbEuL1tppo]1 film (L1=4,4,4‐trifluoro‐1‐phenyl‐1,3‐butadionate; tppo=triphenylphosphine oxide), which showed good temperature sensing of Sr=4.21 % K−1 at 313 K, whereas for the PMMA films doped with Tb3+ and Sm3+ complexes the best performing was the PMMA[TbSmL2tppo]3 film (L2=4,4,4‐trifluoro‐1‐(4‐chlorophenyl)‐1,3‐butadionate), with Sr=3.64 % K−1 at 313 K. Additionally, SiO2 NPs coated with the best performing films from each of the series of PMMA films (Tb–Eu and Tb–Sm) and their temperature‐sensing properties were studied in water, showing excellent performance in the physiological temperature range (PMMA[TbEuL1tppo]1@SiO2: Sr=3.84 % °C at 20 °C; PMMA[TbSmL2tppo]3@SiO2: Sr=3.27 % °C at 20 °C) and the toxicity of these nanoparticles on human cells was studied, showing that they were nontoxic. Lanthanide‐doped luminescent nanoparticles are an appealing system for nanothermometry with biomedical applications due to their sensitivity, reliability, and minimal invasive thermal sensing properties. Here, four unique hybrid organic–inorganic materials prepared by combining β‐NaGdF4 and PMOs (periodic mesoporous organosilica) or mSiO2 (mesoporous silica) are proposed. PMO/mSiO2 materials are excellent candidates for biological/biomedical applications as they show high biocompatibility with the human body. On the other hand, the β‐NaGdF4 matrix is an excellent host for doping lanthanide ions, even at very low concentrations with yet very efficient luminescence properties. A new type of Er3+–Yb3+ upconversion luminescence nanothermometers operating both in the visible and near infrared regime is proposed. Both spectral ranges permit promising thermometry performance even in aqueous environment. It is additionally confirmed that these hybrid materials are non‐toxic to cells, which makes them very promising candidates for real biomedical thermometry applications. In several of these materials, the presence of additional voids leaves space for future theranostic or combined thermometry and drug delivery applications in the hybrid nanostructures. In the present work, we have successfully prepared and characterized novel nanocomposite material exhibiting temperature-dependent surface wettability changes, based on grafted brush coatings of non-fouling poly(di(ethylene glycol)methyl ether methacrylate) (POEGMA) with the embedded CaCO3 nanoparticles. Grafted polymer brushes attached to the glass surface were prepared in a three-step process using atom transfer radical polymerization (ATRP). Subsequently, uniform CaCO3 nanoparticles (NPs) embedded in POEGMA-grafted brush coatings were synthesized using biomineralized precipitation from solutions of CaCl2 and Na2CO3. An impact of the low concentration of the embedded CaCO3 NPs on cell adhesion and growth depends strongly on the type of studied cell line: keratinocytes (HaCaT), melanoma (WM35) and osteoblastic (MC3T3-e1). Based on the temperature-responsive properties of grafted brush coatings and CaCO3 NPs acting as biologically active substrate, we hope that our research will lead to a new platform for tissue engineering with modified growth of the cells due to the release of biologically active substances from CaCO3 NPs and the ability to detach the cells in a controlled manner using temperature-induced changes of the brush. In advancing tissue engineering, we introduce a particle system combining the strength of calcium carbonate with the flexibility of hydrogels enhanced with alkaline phosphatase (ALP) for improved bone regeneration. Our innovation lies in creating large hybrid macrospheroids by bonding mineral nanostructured microparticles loaded with ALP through hydrogel polymerization. These composite macrospheroids address critical challenges in cell seeding, growth, and handling within three-dimensional (3D) environments. We conducted extensive characterization of these particles using scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), mechanical property assessment, and fluorescence microscopy. The results demonstrate that the hybrid macrospheroids significantly enhance cell manipulation and growth in three-dimensional structures. Specifically, ALP-loaded macrospheroids showed a marked improvement in osteogenic activity, promoting effective bone tissue regeneration. This study not only showcases a unique approach to overcoming the limitations of traditional hydrogels in tissue engineering but also opens pathways for bone tissue regeneration. Our findings offer a promising tool for cell seeding and growth in 3D structures, potentially revolutionizing practices in tissue engineering and regenerative medicine.

Superficial fungal infections are of serious concern worldwide due to their morbidity and increasing distribution across the globe in this era of growing antimicrobial resistance. The delivery of antifungals to the target regions of the skin and sustaining the effective drug concentration are essential for successful treatment of such mycoses. Topical formulations get extra benefits here if they penetrate into the hair follicles since fungal hyphae can proliferate and produce spores in such reservoirs. We designed a novel particulate system for the encapsulation and intrafollicular delivery of griseofulvin (Gf) antifungal drug, which is water-insoluble and currently commercially available in oral dosage forms. Micron-sized calcium carbonate (vaterite) carriers containing 25 ± 3% (w/w) of Gf were prepared via the wet chemical method. The successful in vivo transportation of the carriers into the hair follicles of rats was demonstrated using scanning electron and confocal laser scanning microscopy. In addition, we introduced an approach toward Gf release prolongation for the proposed system. The stabilizing coatings were formed on the surface of the obtained particles via the layer-by-layer technique. The formulations displayed sufficient biocompatibility and good cellular uptake in contact with fibroblast cells in vitro . Four different coatings were tested for their preserving ability in the course of continued carrier incubation in the model media. The best release prolonging formulation liberated 38% of the loaded Gf during 5 days, while the uncoated carriers demonstrated more than 50% drug release within the first 24 h in water. To assess the in vivo release properties, free Gf drug and Gf-loaded carriers (uncovered and covered with the stabilizing shell) were administered topically in rats and the drug excretion profiles were further studied. By comparing the daily Gf levels in urine, we verified the sustained effect (longer than a week) of the stabilizing shell formed on the carrier surface. Conversely, the application of the free drug did not provide reliable Gf detection for this period. These findings open new prospects for the efficiency enhancement of topical therapeutics. Importantly, the elaborated system could be adapted for the dermal delivery of various water-insoluble drugs beyond the scope of antifungal therapy.
