Abstract

Liver fibrosis is a key determinant of the progression of metabolic dysfunction-associated steatotic liver disease (MASLD). Its increasing prevalence and a lack of effective treatments make it a major health problem worldwide, particularly in people living with HIV, among whom the prevalence of advanced fibrosis is higher. We have published preclinical data showing that Rilpivirine (RPV), a widely used anti-HIV drug, selectively triggers hepatic stellate cell (HSC) inactivation and apoptosis through signal transducer and activator of transcription (STAT)1-mediated pathways, effects that clearly attenuate liver fibrosis and promote regeneration. We performed a retrospective, cross-sectional study of RPV-induced effects on steatosis, inflammation, and fibrosis in liver biopsies from well-controlled HIV-infected subjects diagnosed with MASLD. Patients on RPV exhibited similar levels of HIV-related parameters to those not receiving this drug, while showing a tendency toward improved liver function and lipid profile, as well as an enhanced activation of STAT1 in hepatic non-parenchymal cells in those with identified liver injury. This protective effect, promoting STAT1-dependent HSC inactivation, was observed at different stages of MASLD. Our results suggest that RPV-based therapy is especially indicated in HIV-infected patients with MASLD-derived liver injury and highlight the potential of RPV as a new therapeutic strategy for liver diseases. Abstract

The synthesis of crystalline one-dimensional polymers provides a fundamental understanding about the structure??? property relationship in polymeric materials and allows the preparation of materials with enhanced thermal, mechanical, and conducting properties. However, the synthesis of crystalline one-dimensional polymers remains a challenge because polymers tend to adopt amorphous or semicrystalline phases. Herein, we report the synthesis of a crystalline one-dimensional polymer in solution by dynamic covalent chemistry. The structure of the polymer has been unambiguously confirmed by microcrystal electron diffraction that together with charge transport studies and theoretical calculations show how the ir-stacked chains of the polymer generate optimal channels for charge transport.

Abstract

Herein, we show that twisted molecular nanoribbons with as many as 322 atoms in the aromatic core are efficient gelators capable of self-assembling into ordered pi-gels with morphologies and sol-gel transitions that vary with the length of the nanoribbon. In addition, the nanoribbon gels show a red fluorescence and also pseudoconductivity values in the same range as current state-of-the-art pi-gels.

Three-dimensional covalent organic frameworks (3D COFs) with a pcu topology have been obtained from distorted polycyclic aromatic hydrocarbons acting as triangular antiprismatic (D3d) nodes. Such 3D COFs are six-fold interpenetrated as the result of interframework π-stacking, which enable charge transport properties that are not expected for 3D COFs.

Abstract

A high degree of crystallinity is an essentialaspect in two-dimensional covalent organic frameworks, asmany properties depend strongly on the structural arrangement of the different layers and their constituents. Weintroduce herein a new design strategy based on core-twistedpolycyclic aromatic hydrocarbon as rigid nodes that give riseto a two-dimensional covalent organic framework with awavy honeycomb (chairlike) lattice. The concave−convex self- complementarity of the wavy two-dimensional lattice guides thestacking of framework layers into a highly stable and ordered covalent organic framework that allows a full 3D analysis by transmission electron microscopy revealing its chairlike honeycomb facets and aligned mesoporous channels. Remarkably, the waviness of the framework does not disrupt the interlayerπ−πstacking that shows charge transporting properties similar to those of planar covalent organic frameworks. The implementation of core-twisted aromatics as building blocks for covalent organic frameworks brings new possibilities in the design of highly ordered organic materials.

Abstract

The synthesis and optical, electrochemical, thermal, and electrical character- ization of a new and unexpected 1-n-octyloxyperopyrene is reported. The structure of 1-n- octyloxyperopyrene has been unambiguously established by single-crystal X-ray diffraction. The solubility of this polycyclic aromatic hydrocarbon, endowed by the alkoxy substituent, allows the fabrication of thin-filmfield-effect transistors by liquid deposition methods. These devices show hole mobilities up to 1.61×10−3cm2V−1s−1.

Abstract

The integrated analytical approach developed in this study offers a powerful methodology for the structural characterisation of complex molecular nanomaterials. Structures of a covalent organic framework based on boronate esters (COF-5) and a conjugated microporous polymer (Aza-CMP) have been investigated by a combination of several electron microscopy techniques elucidating the three-dimensional topology of the complex polycrystalline (COF) and non-crystalline (CMP) materials. Unexpected, aperiodic mesoporous channels of 20-50 nm in diameter were found to be penetrating the COF and CMP particles, which cannot be detected by X-ray diffraction techniques. The mesopores appear to be stable under a range of different conditions and accessible to gas molecules, exhibiting a particular bonding capability with CO2 in the case of the CMP. The mesoporosity is unrelated to the intrinsic chemical structures of the COF or CMP but rather it reflects the mechanisms of polymer particle formation in a polycondensation reaction. The mesopores may be templated by clusters of solvent molecules during the COF or CMP synthesis, leaving cavities within the polymer particles. The unexpected mesoporosity discovered in COF and CMP materials begs for re-assessment of the nature of framework materials and may open new opportunities for applications of these molecular materials in gas sorption or catalysis.

Abstract

Switchable luminescent organic materials with unusual high light‐sensitivity in the bulk at room temperature are described. The compounds consist of two promesogenic photoactive cyanostilbene units linked by a flexible spacer with an odd number of carbon atoms, analogous to the structural design of a liquid crystal dimer. Experimental and computational studies support the formation of excimers in solution due to the characteristic geometry of the dimers. In their condensed phases they show a high tendency to pack in lamellar phases, exhibiting bent‐core lamellar liquid crystal phases and crystalline polymorphs with different photoluminescence and high quantum yields. The compounds are highly photoreactive, and their photoactivity in terms of photoisomerization and/or photocycloaddition reactions in solution and in the bulk is different. By irradiation of the compounds in the condensed phases at room temperature a fast photochemical process takes place, identified as a photocycloaddition reaction favored by the lamellar packing. Due to this photoreaction a luminescence modulation with green/blue color switching and an increase in quantum yield takes place.

40. Organizaciones supramoleculares luminiscentes de moléculas de tipo 'bent-core'

Abstract

El trabajo desarrollado en esta Tesis Doctoral se ha centrado en la preparación y el estudio de organizaciones supramoleculares de moléculas de tipo bent-core luminiscentes. Para la consecución de estos objetivos se han seleccionado dos motivos estructurales con desigual trayectoria en el campo de los materiales luminiscentes: el sistema cianoestilbeno de gran atractivo y actualidad, y el pireno, más clásico pero de vigente interés. En el estado sólido, ambos son sensibles al entorno que les rodea, presentando drásticos cambios en la luminiscencia frente a distintos estímulos o ambientes. Sin embargo, hasta este trabajo no habían sido incorporados y estudiados en moléculas mesógenas de tipo bent-core, ofreciendo un campo virgen en lo que a los estudios estructura química - estructura supramolecular - propiedades, se refiere. Dependiendo del sistema luminiscente seleccionado, el diseño molecular propuesto ha variado. Así, se han incorporado unidades cianoestilbeno como estructuras laterales de moléculas de tipo bent-core para la consecución de fases cristal líquido de tipo bent-core luminiscentes, propiedades ópticas no lineales y propiedades fotoquímicas. Por otro lado, también se ha incorporado la unidad pireno en las cadenas terminales de moléculas de tipo bent-core con el fin de conseguir tres objetivos diferentes: obtener organizaciones cristal líquido de tipo bent-core luminiscentes, conseguir geles luminiscentes y acceder a monocapas vía interacciones pireno grafeno. Se ha desarrollado la síntesis de diferentes compuestos orgánicos mediante rutas sintéticas multietapa, así como la caracterización de los diferentes materiales mediante una amplia variedad de técnicas experimentales: microscopias (MOP, TEM, SEM), calorimetría, difracción de rayos X, y mediante espectroscopia UV-vis, de fluorescencia y dieléctrica, etc., permitiendo la adecuada caracterización estructural y física de los mismos. Como resultado de esta investigación se ha podido concluir que la combinación de estructuras bent-core con estas dos unidades funcionales fotoactivas ha permitido obtener materiales luminiscentes novedosos: fases cristal líquido de tipo bent core, materiales con elevados rendimientos cuánticos de fluorescencia, agregados en disolventes, geles luminiscentes con organizaciones características de fases bent core, materiales mecano- y termocrómicos y con respuesta a la luz, y también sistemas basados en la interacción grafeno-pireno, abriendo nuevos caminos a explorar en el desarrollo de dispositivos fotoactivos y en el marco general de los materiales supramoleculares funcionales.

41. A Wavy Two-Dimensional Covalent Organic Framework from Core-Twisted Polycyclic Aromatic Hydrocarbons

Abstract

A high degree of crystallinity is an essential aspect in two-dimensional covalent organic frameworks, as many properties depend strongly on the structural arrangement of the different layers and their constituents. We introduce herein a new design strategy based on core-twisted polycyclic aromatic hydrocarbon as rigid nodes that give rise to a two-dimensional covalent organic framework with a wavy honeycomb (chairlike) lattice. The concave–convex self-complementarity of the wavy two-dimensional lattice guides the stacking of framework layers into a highly stable and ordered covalent organic framework that allows a full 3D analysis by transmission electron microscopy revealing its chairlike honeycomb facets and aligned mesoporous channels. Remarkably, the waviness of the framework does not disrupt the interlayer π–π stacking that shows charge transporting properties similar to those of planar covalent organic frameworks. The implementation of core-twisted aromatics as building blocks for covalent organic frameworks brings new possibilities in the design of highly ordered organic materials.

Two isomeric cyanostilbene photoswitchable bent-core mesogens with polar liquid crystal phases in which macroscopic polarization and luminescence can be light-modulated are introduced. Z/E isomerization or [2+2] cycloaddition photochemical processes occur depending on the chemical structure, which make the compounds very innovative multifunctional advanced materials.

Abstract

Multiresponsive luminescent materials with dicyanodistyrylbenzene chromophores incorporated into bent-core and rod-like molecules have been prepared and characterized. Rod-like compounds exhibit nematic or smectic liquid crystal phases, whereas bent-core compounds melt directly to the isotropic liquid at high temperatures. Fluorescence properties are very sensitive to external stimuli such as heat, light, pressure and solvent vapors. All compounds undergo Z/E photoisomerization in solution under UV irradiation. In addition, photochemical activity in bulk has been investigated. Whereas the crystal phases are stable under UV irradiation, the liquid crystal phases are unstable giving rise to Z/E photoisomerization and [2+2]-photocycloaddition products. As a consequence, photoinduced phase changes are observed. This study shows the versatilty of dicyanodistyrylbenzene-based liquid crystals as novel types of photoreactive materials.

45. Organizaciones supramoleculares luminiscentes de moléculas de tipo 'bent-core'

Abstract

El trabajo desarrollado en esta Tesis Doctoral se ha centrado en la preparación y el estudio de organizaciones supramoleculares de moléculas de tipo bent-core luminiscentes. Para la consecución de estos objetivos se han seleccionado dos motivos estructurales con desigual trayectoria en el campo de los materiales luminiscentes: el sistema cianoestilbeno de gran atractivo y actualidad, y el pireno, más clásico pero de vigente interés. En el estado sólido, ambos son sensibles al entorno que les rodea, presentando drásticos cambios en la luminiscencia frente a distintos estímulos o ambientes. Sin embargo, hasta este trabajo no habían sido incorporados y estudiados en moléculas mesógenas de tipo bent-core, ofreciendo un campo virgen en lo que a los estudios estructura química - estructura supramolecular - propiedades, se refiere. Dependiendo del sistema luminiscente seleccionado, el diseño molecular propuesto ha variado. Así, se han incorporado unidades cianoestilbeno como estructuras laterales de moléculas de tipo bent-core para la consecución de fases cristal líquido de tipo bent-core luminiscentes, propiedades ópticas no lineales y propiedades fotoquímicas. Por otro lado, también se ha incorporado la unidad pireno en las cadenas terminales de moléculas de tipo bent-core con el fin de conseguir tres objetivos diferentes: obtener organizaciones cristal líquido de tipo bent-core luminiscentes, conseguir geles luminiscentes y acceder a monocapas vía interacciones pireno grafeno. Se ha desarrollado la síntesis de diferentes compuestos orgánicos mediante rutas sintéticas multietapa, así como la caracterización de los diferentes materiales mediante una amplia variedad de técnicas experimentales: microscopias (MOP, TEM, SEM), calorimetría, difracción de rayos X, y mediante espectroscopia UV-vis, de fluorescencia y dieléctrica, etc., permitiendo la adecuada caracterización estructural y física de los mismos. 49. Bent-core liquid crystal phases promoted by azo-containing molecules: from monomers to side-chain polymers

Abstract

New bent-core monomethacrylates and their side-chain polymers containing the 3,4′-biphenylene moiety as a central core and both all-ester and azo-ester lateral structures have been prepared and characterized. Attractive results concerning the scarcely studied side-chain homo- and copolymers, synthesized either by in situ polymerization in the mesophase or ATRP in solution, are reported. The research is focused on the synthesis of these materials and the effect that structural changes, such as the number of aromatic rings (5 or 6), the presence of -NN- versus -COO- links on the lateral structures, and the relative positions of the reactive groups, have on the liquid crystal properties of these new bent-core based-materials. The mesomorphic behavior of these low and high-molecular compounds, and also of the synthetic intermediates with bulky terminal bromo-substituents, have been characterized by POM, XRD, DSC and electrooptic studies, showing that the majority of these new molecules have the ability to organize forming non classical lamellar or columnar mesophases. Significant stabilization of the lamellar SmCP mesophase ranges is achieved upon polymerization in comparison to the monomers and interestingly the vitrification of these supramolecular organizations occurs at room temperature. © 2014 the Partner Organisations.
