Abstract Intracellular electrophysiology is essential in neuroscience, cardiology, and pharmacology for studying cells’ electrical properties. Traditional methods like patch-clamp are precise but low-throughput and invasive. Nanoelectrode Arrays (NEAs) offer a promising alternative by enabling simultaneous intracellular and extracellular action potential (iAP and eAP) recordings with high throughput. However, accessing intracellular potentials with NEAs remains challenging. This study presents an AI-supported technique that leverages thousands of synchronous eAP and iAP pairs from stem-cell-derived cardiomyocytes on NEAs. Our analysis revealed strong correlations between specific eAP and iAP features, such as amplitude and spiking velocity, indicating that extracellular signals could be reliable indicators of intracellular activity. We developed a physics-informed deep learning model to reconstruct iAP waveforms from extracellular recordings recorded from NEAs and Microelectrode arrays (MEAs), demonstrating its potential for non-invasive, long-term, high-throughput drug cardiotoxicity assessments. This AI-based model paves the way for future electrophysiology research across various cell types and drug interactions.

Understanding how to manage the hydrophilicity and hydrophobicity of a surface has been the focus of a lot of research in recent years. The surface's energy often controls its hydrophobic state. There are numerous techniques to realize a change in the surface energy. Smart nano-based materials are being used to create super hydrophobic coatings that will serve as layers of defence against mechanical abrasion, corrosion, and fouling agents on the surface of metallic components. These coatings, which have recently gained popularity, have shown to be excellent choices for protecting steel pipelines. The recently created super hydrophobic coatings for glass surfaces, papers, cotton, steel pipes, etc., are examined in-depth and critically in this review study, emphasizing their use in different industries. It explains how to create super hydrophobic coatings on glass substrates using various techniques and the most recent research results on various coating production techniques. An in-depth discussion is also given to the recent applications of these created super hydrophobic coatings for treatments, including anti-bio fouling, dicing, and corrosion prevention over the past five years. According to the literature, spraying is the most adaptable and popular technique for creating super hydrophobic coatings for any substrate.

Background: Little is known regarding the effect of the coronavirus pandemic on the characteristics of dermatology admissions, particularly in the Indian context. Objectives: To conduct a retrospective analysis of the discharge records of all patients admitted to a tertiary care centre in Western India from 1 January 2019 to 31 December 2022. Methods: Patient records were reviewed and compiled. Diagnoses were grouped to achieve consistency with international studies. Finally, the effect of COVID on admissions was determined. Data samples were assessed using descriptive statistics. Continuous variables were expressed as median or mean. The student’s t-test and chi-square test were utilised to investigate wherever appropriate. Results: Inpatient care was required for 1,817 patients. Males (1000, 55.04%) were significantly more likely to be admitted than females (817, 44.96%). The leading causes of hospitalisation were bacterial infections (26.42%), vesiculobullous disorders (17.45%), and psoriasiform disorders (11.34%). The greatest number of admissions occurred in 2019, followed by a substantial decline in 2020. Admission rates for vesiculobullous disorders, psoriasiform disorders, and malignancies were significantly higher during the pandemic period. Conclusions: Our study offers an analysis of the impact of the coronavirus pandemic on the patient profiles of patients admitted to a dermatology ward.

Pathogenic bacteria in wounds impede successful skin grafting. However, their detection relies on culture methods, which delay confirmation by several days. Real-time fluorescence imaging detects bacteria, allowing for rapid assessment and documentation. We herein report a post modified radical mastectomy, surgical site infection with multidrug-resistant Pseudomonas spp. that underwent repeated antibiotic therapy and debridement and eventually grafting. In this case, a real-time fluorescence imaging device helped prevent graft rejection.

Abstract Numerous speculations have continually emerged, trying to explore the association between COVID-19 infection and a varied range of demographic and clinical factors. Frontline healthcare workers have been the primary group exposed to this infection, and there have been limited global research that examine this cohort. However, while there are a few large studies conducted on Indian healthcare professionals to investigate their potential risk and predisposing factors to COVID-19 infection, to our knowledge there are no studies evaluating the development of long COVID in this population. This cross-sectional study systematically utilized the demographic and clinical data of 3329 healthcare workers (HCW) from a tertiary hospital in India to gain significant insights into the associations between disease prevalence, severity of SARS-Cov-2 infection and long COVID. Most of the study population was found to be vaccinated (2,615, 78.5%), while 654 (19.65%) HCWs were found to be SARS-CoV-2 positive at least once. Of the infected HCWs, 75.1% (491) did not require hospitalization, whereas the rest were hospitalized for an average duration of 9 days. A total of 206 (6.19%) individuals were found to be suffering from long COVID. Persistent weakness/tiredness was the most experienced long-COVID symptom, while females (1.79, 1.25–2.57), individuals who consumed alcohol (1.85, 1.3–2.64) or had blood group B (1.9, 1.33–2.7) were at a significantly higher risk for developing long COVID.

A novel hybrid bio-composite, based on Mg-alloy (AZ31) and reinforced with Titanium dioxide (TiO2) & Hydroxyapatite (HAp) particles, has been successfully created using the powder metallurgy technique. The efficacy of this approach in synthesizing hybrid composites is remarkable. Insightful microstructural analysis of the bio-composite specimens unveiled a uniform dispersion of reinforcement particles across the matrix. By conducting rigorous compressive and microhardness assessments, the influence of integrating TiO2-HAp into AZ31 on its mechanical attributes was thoroughly explored. Notably, the peak ultimate compressive strength reached an impressive 430 MPa, while the microhardness peaked at 58 HV. Moreover, a detailed investigation into the tribological performance of the hybrid composite was also undertaken. Simultaneously, the assessment of cell viability through the MTT assay yielded valuable insights. The fundamental mechanisms reinforcing the hybrid bio-composite, attributed to the incorporation of TiO2-HAp, are primarily linked to the elevation of microhardness, compressive strength, and a concurrent reduction in wear loss. Notably, a noteworthy enhancement in cytocompatibility was observed, with cell viability ranging from 78 to 83 percent for hybrid composites following a nine-day incubation period.

Generative autoencoders create full conformational ensembles of intrinsically disordered proteins from short molecular dynamics simulations.

Abstract Background Autoinflammatory disorders are the group of inherited inflammatory disorders caused due to the genetic defect in the genes that regulates innate immune systems. These have been clinically characterized based on the duration and occurrence of unprovoked fever, skin rash, and patient’s ancestry. There are several autoinflammatory disorders that are found to be prevalent in a specific population and whose disease genetic epidemiology within the population has been well understood. However, India has a limited number of genetic studies reported for autoinflammatory disorders till date. The whole genome sequencing and analysis of 1029 Indian individuals performed under the IndiGen project persuaded us to perform the genetic epidemiology of the autoinflammatory disorders in India. Results We have systematically annotated the genetic variants of 56 genes implicated in autoinflammatory disorder. These genetic variants were reclassified into five categories (i.e., pathogenic, likely pathogenic, benign, likely benign, and variant of uncertain significance (VUS)) according to the American College of Medical Genetics and Association of Molecular pathology (ACMG-AMP) guidelines. Our analysis revealed 20 pathogenic and likely pathogenic variants with significant differences in the allele frequency compared with the global population. We also found six causal founder variants in the IndiGen dataset belonging to different ancestry. We have performed haplotype prediction analysis for founder mutations haplotype that reveals the admixture of the South Asian population with other populations. The cumulative carrier frequency of the autoinflammatory disorder in India was found to be 3.5% which is much higher than reported. Conclusion With such frequency in the Indian population, there is a great need for awareness among clinicians as well as the general public regarding the autoinflammatory disorder. To the best of our knowledge, this is the first and most comprehensive population scale genetic epidemiological study being reported from India.

Abstract Background Hepatocellular carcinoma (HCC) is the most prevalent primary liver cancer, accounting for 80–90% of cases. Mutations are commonly found in the signaling regulating the PI3K/Akt pathway, leading to oncogenic cell proliferation and survival. Key transcription factors that are negatively regulated downstream of PI3K/Akt are members of the forkhead box O family (FOXO). FOXOs were initially considered as tumor suppressors by inducing cell cycle arrest and apoptosis. However, there is increasing evidence showing that FOXOs, especially FOXO3, can support tumorigenesis. Methods To understand the roles of FOXO3 in liver tumorigenesis and hepatocarcinogenesis, we analyzed HCC patient specimens and also established a doxycycline-regulated transgenic mouse model with hepatocyte-specific FOXO3 expression in a constitutively active form. Results We found that FOXO3 protein is significantly overexpressed and activated in livers of HCC patients. Hepatic activation of FOXO3 induced extensive hepatic damage and elevated gene expression of several HCC-associated factors. Furthermore, FOXO3 expression enhanced hepatotoxicin-induced tumorigenesis. Mechanistically, FOXO3 activation caused oxidative stress and DNA damage and triggered positive feedback-loop for Akt activation as well as mTORC2 activation. Interestingly, FOXO3 activated not only reactive oxygen species (ROS)-promoting pathways, but also ROS-eliminating systems, which can be associated with the activation of the pentose phosphate pathway. Conclusions FOXO3 is a master regulator of ROS in a ‘carrot and stick’ manner; on one side avoiding cellular crisis while also supporting hepatocellular carcinogenesis. Clinically, we suggest analyzing FOXO3 activation status in patients with liver diseases, in addition to PI3K/Akt signaling. Personalized therapy of FOXO3 inhibition may be a reasonable, depending on the activation status of FOXO3.

Abstract A facile carbothermal route was adopted to obtain niobium carbide nanoparticles (NPs) embedded in carbon network from Nb2O5 to study photocatalytic behavior. Optimization of synthesis parameters to obtain single phase NbC NPs has been successfully done. The phase identification, morphology and nature of carbon were determined with the help of X-ray diffraction, transmission electron microscopy (TEM) and Raman spectroscopy. X-ray photoelectron spectroscopy (XPS) suggested the presence of multiple oxidation states of Nb associated to NbC and NbCxOy centers on the surface of NPs. Due to the presence of NbCxOy on the surface of NPs, absorption under visible region of EM spectrum has been observed by UV-visible spectroscopy. Different organic dyes (RhB, MB and MO) were used to study the effect of holding time on the photocatalytic performance of as-synthesized samples. RhB dye was found to be the most sensitive organic molecule among all the considered dyes and degraded 78% in 120 min.

Background: The relationship between impaired quality of life (QoL) due to melasma and its clinical severity remains equivocal despite several studies. Aim: The aim was to study the correlation, if any, between the clinical severity and the impairment in QOL due to melasma. Methods: This cross-sectional, questionnaire-based study was conducted on a cohort of 141 patients of melasma attending the outpatient department of our referral hospital. A physician measured the severity of melasma using the melasma area and severity index (MASI), while melasma-related QoL (MELASQOL) score was calculated utilizing the validated Hindi version of the MELASQOL questionnaire filled by the patients. Correlations of these two scores with each other and with components of the demographic data were attempted using the Statistical Package for the Social Sciences, version 20. Results: Significantly greater impairment in QoL was found in patients with a history of prior use of triple combination therapy and in patients with hirsutism and/or polycystic ovarian disease. The severity of melasma was found to be significantly higher in patients with a history of recurrence and tobacco chewing. Limitations: The sample size could have been larger. Ultrasonography could have been carried out in all cases of hirsutism. Conclusion: The severity of melasma does not correlate with the impairment in QoL.

Background: Chronic alcohol intake impacts skin directly, through organ dysfunction or by modifying preexisting dermatoses. However, dermatoses afflicting chronic alcoholics figure in a few studies only. Aim: This study aims to correlate the spectrum of dermatoses in chronic alcoholics with the quantum/duration of alcohol intake and raised liver transaminases. Materials and Methods: Adult males, totaling 196, ascertained to fulfill the Royal College of Psychiatry criteria for chronic alcoholism by the de-addiction center and referred for dermatological consult were enrolled as cases, and similar number of age-/sex-matched teetotallers, as controls. Data emanating from detailed history, clinical examination, and routine liver functions tests were summarized and subsequently analyzed, including statistically using the Chi-square, independent “t” and Spearman's rank correlation tests, and compared with data from previous studies. Results: Majority (104) drank 41–50 units of alcohol/week since 3–40 (mean: 20.01 ± 9.322) years. Generalized pruritus (odds ratio [OR]: 31.15, P < 0.001), xerosis (OR: 3.62, P = 0.008), and seborrheic dermatitis (OR: 12.26, P < 0.001) were significantly more common in cases than controls. Infections (73; 37.2%), eczemas (45; 22.9%), and generalized hyperpigmentation (28; 14.2%) were the major presenting complaints. Spider nevi, gynecomastia, and pellagroid dermatitis were present in 34 (17.3%), 19 (9.7%), and 8 (4.1%) respectively exclusively in cases only. Commonly seen systemic abnormalities were an alcoholic liver disease (45; 22.9%), diabetes mellitus (23; 11.7%), and peripheral neuropathy (19; 9.7%). Conclusion: Knowledge of cutaneous manifestations of chronic alcoholism could prompt in-depth history taking of alcohol intake, lead to specialist referral and thereby enable timely de-addiction, hopefully before serious adversities in the chronic alcoholics.

Although the long-term use of immunosuppressives – supplemented with more aggressive treatments such as immunoadsorption, intravenous immunoglobulins, or plasmapheresis in recalcitrant cases has dramatically improved the prognosis of pemphigus vulgaris, opportunistic infections secondary to immunosuppression continue to cause significant mortality. We report four cases– three old ones, who had accumulated significant morbidities over their disease duration ranging from 5 to 10 years, and the fourth, a teenage female intolerant to corticosteroids and idiosyncratic to methotrexate– who achieved complete remission on administration of rituximab by the lymphoma protocol. One of the old cases who had recalcitrant mucositis experienced its complete subsidence without any adjuvant whatsoever. All continue to remain asymptomatic for 11–20 months. None had infusion reactions or any delayed side effects.

Integrins play an important role in tumor progression, invasion and metastasis. Therefore we aimed to evaluate a preclinical imaging approach applying ανβ3 integrin targeted hybrid Fluorescence Molecular Tomography/X-ray Computed Tomography (FMT-XCT) for monitoring tumor progression as well as early therapy response in a syngeneic murine Non-Small Cell Lung Cancer (NSCLC) model. Lewis Lung Carcinomas were grown orthotopically in C57BL/6 J mice and imaged in-vivo using a ανβ3 targeted near-infrared fluorescence (NIRF) probe. ανβ3-targeted FMT-XCT was able to track tumor progression. Cilengitide was able to substantially block the binding of the NIRF probe and suppress the imaging signal. Additionally mice were treated with an established chemotherapy regimen of Cisplatin and Bevacizumab or with a novel MEK inhibitor (Refametinib) for 2 weeks. While μCT revealed only a moderate slowdown of tumor growth, ανβ3 dependent signal decreased significantly compared to non-treated mice already at one week post treatment. ανβ3 targeted imaging might therefore become a promising tool for assessment of early therapy response in the future.

Metabolic syndrome, a constellation of interrelated risk factors of metabolic origin namely, abdominal obesity, insulin resistance, dyslipidemia and hypertension, confers greater risk of cardiovascular disease on its patients than the sum of the individual components. It is increasingly being associated with inflammatory dermatoses, especially psoriasis. Determination of the diagnostic criteria of this syndrome is conditioned by the changing views regarding its pathogenesis. Approximately, a quarter of the world′s population harbors this syndrome, whose reported prevalence in India (5-30%) has escalated with an increase in urbanization and socioeconomic status. Due to, up to 3 times, the risk of cardiovascular mortality and up to 24 times risk of diabetes mellitus, the epidemiological significance of metabolic syndrome ideally necessitates formulation of preventive guidelines by public health authorities. Chronic inflammation, involving several cytokines and adipokines, forms the bridge between this syndrome and psoriasis and underlies the formation of atherosclerotic plaque, the primary lesion of coronary artery disease, in whose pathogenesis oxidative stress and genetic factors also play a role. Up to 4-fold increase in the prevalence of metabolic syndrome and 3-fold increased risk of cardiovascular mortality is reported globally in psoriatics. Increasing index of suspicion of this syndrome by the dermatologists, prevention of cardiovascular damage by lifestyle modifications, smoking cessation and redressal of the inherent depression in these patients is as imperative in management as is the specific therapy of the skin lesions of this systemic, rather than "just a skin," disease as well as the lipid-lowering, antihypertensive and antidiabetic agents.

Background: Acne vulgaris is known to impair many aspects of the quality of life (QoL) of its patients. Aim: To translate the Cardiff Acne Disability Index (CADI) from English into Hindi and to assess its validity and reliability in Hindi speaking patients with acne from India. Methods: Hindi version of CADI, translated and linguistically validated as per published international guidelines, along with a previously translated Hindi version of dermatology life quality index (DLQI) and a demographic questionnaire were administered to acne patients. The internal consistency reliability of the Hindi version of CADI and its concurrent validity were assessed by Cronbach′s alpha co-efficient and Spearman′s correlation co-efficient respectively. Construct validity was examined by factor analysis. Statistical analysis was carried out using the Statistical Package for the Social Sciences (SPSS) version 20 (SPSS Inc., Chicago, IL, USA) for Windows. Results: One hundred Hindi speaking patients with various grades of acne participated in the study. Hindi version of CADI showed high internal consistency reliability (Cronbach′s alpha co-efficient = 0.722). Mean item-to-total correlation co-efficient ranged from 0.502 to 0.760. Concurrent validity of the scale was supported by a significant correlation with the Hindi DLQI. Factor analysis revealed the presence of two dimensions underlying the factor structure of the scale. Conclusion: Hindi CADI is equivalent to the original English version and constitutes a reliable and valid tool for clinical assessment of the impact of acne on QoL.
