Avian coccidiosis, a common disease caused by Eimeria species, results in significant losses in global poultry production. Mycotoxins are low-molecularweight natural products (i.e., small molecules) produced as secondary metabolites by filamentous fungi and they have the potential to economically and significantly affect global poultry production. Little is known about the relationship between mycotoxins and avian coccidiosis, although they often co-occur in the field. This comprehensive review examines the intricate relationship between mycotoxins and avian coccidiosis, in particular how mycotoxins, including aflatoxins, ochratoxins, trichothecenes as well as Fusarium mycotoxins, compromise the health of the poultry flock and open the door to Eimeria parasites in the gut. In addition, this review sheds light on the immunosuppressive effects of mycotoxins, their disruption of cellular signaling pathways, and the consequent exacerbation of coccidiosis infections. The mechanisms of mycotoxin toxicity are also reviewed, emphasizing direct damage to intestinal epithelial cells, impaired nutrient absorption, inflammation, oxidative stress, and changes in the gut microbiota. Finally, the consequences for the prevention and treatment of coccidiosis when mycotoxins are present in the feed are discussed. This review emphasizes the need for effective management strategies to mitigate the combined risks of mycotoxins and coccidiosis and highlights the complexity of diagnosing and controlling these interrelated problems in poultry. The review advocates a holistic approach that includes strict feed management, disease prevention measures and regular monitoring to maintain the health and productivity of poultry against these significant challenges. The codon usage bias (CUB) of genes encoded by different species' genomes varies greatly. The analysis of codon usage patterns enriches our comprehension of genetic and evolutionary characteristics across diverse species. In this study, we performed a genome-wide analysis of CUB and its influencing factors in six sequenced Eimeria species that cause coccidiosis in poultry: Eimeria acervulina, Eimeria necatrix, Eimeria brunetti, Eimeria tenella, Eimeria praecox, and Eimeria maxima. The GC content of protein-coding genes varies between 52.67% and 58.24% among the six Eimeria species. The distribution trend of GC content at different codon positions follows GC1 > GC3 > GC2. Most high-frequency codons tend to end with C/G, except in E. maxima. Additionally, there is a positive correlation between GC3 content and GC3s/C3s, but a significantly negative correlation with A3s. Analysis of the ENC-Plot, neutrality plot, and PR2-bias plot suggests that selection pressure has a stronger influence than mutational pressure on CUB in the six Eimeria genomes. Finally, we identified from 11 to 15 optimal codons, with GCA, CAG, and AGC being the most commonly used optimal codons across these species. This study offers a thorough exploration of the relationships between CUB and selection pressures within the protein-coding genes of Eimeria species. Genetic evolution in these species appears to be influenced by mutations and selection pressures. This study determines the effects of probiotics - Saccharomyces cerevisiae on oocysts shedding, haematology and serum proteins of broilers infected with mixed Eimeria oocysts. 120-day-old broiler chicks were randomly assigned to four groups (Groups A - D) of 30 birds each. Group A - was fed a plain diet and not infected, Group B - was fed a supplemented diet and infected, Group C - was fed a plain diet and infected, and Group D - was fed a supplemented diet and not infected. S. cerevisiae was supplemented at 0.1 %/kg of feed. At week 3, Groups B and C were infected with sporulated mixed Eimeria oocysts orally, followed by a daily faecal examination of all groups. The rate of oocyst shedding, packed cell volume (PCV), haemoglobin concentration (Hb), red blood cell (RBC) count, total white blood cell (WBC) count, differential WBC count and serum levels of total protein, albumin and globulin were determined at five days' intervals. The data collected were analyzed using a one-way analysis of variance at a 95% confidence interval (p<0.05). Group C had the significantly higher (p<0.05) number of oocysts in faeces, total WBC count and globulin levels, with the lowest PCV, Hb, RBC count, total protein and albumin levels. Group D had significantly higher (p<0.05) heterophil, eosinophil and monocyte counts. In conclusion, dietary supplementation with S. cerevisiae (1 g/kg) ameliorated the effect of Eimeria infection in broilers, by reducing the rate of oocyst shedding, anaemia and hypoproteinaemia due to the activity of the parasite. Introduction: Avian coccidiosis presents a significant challenge to the poultry industry in Egypt, highlighting the urgent need for validating new drug targets offering promising prospects for the development of advanced anticoccidials. Although numerous reports highlight the activity of lactoferrin (LF) against various microorganisms, its potential against Eimeria has not been explored. The present study evaluated the potential anticoccidial effect of LF and diclazuril in broiler chickens experimentally infected with Eimeria tenella. Methods: A total of 100 one-day-old broiler chicks were divided into five equal groups (20 each) as follows: Group 1 (G1) served as the normal healthy control group, Group 2 (G2) consisted of chickens infected with 1 × 105 sporulated E. tenella oocysts at 14 days of age, Group 3 (G3) comprised infected chickens treated with diclazuril (0.5 mL/L in drinking water) for 3 days successively, Group 4 (G4) included infected chickens treated with LF (at a dose of 250 mg/kg of diet) from one day of age until the end of the study, and Group 5 (G5) comprised infected chickens treated with both LF and diclazuril. Results: The positive control group (G2) experienced significant reductions in body weight (BW), BW gain, serum glucose, lipase, amylase, total antioxidant capacity, several hematological indices, and total proteins, along with alterations in various antioxidant enzymes. Conversely, serum levels of aspartate aminotransferase (AST), Alanine aminotransferase (ALT), Alkaline phosphatases (ALP), urea, creatinine, nitric oxide, mean corpuscular volume (MCV), White blood cells (WBCs), heterophils, alpha 2, beta 1, and liver contents of malondialdehyde were elevated in this group. Moreover, higher oocyst counts and lesion scores, along with histopathological alterations, were observed in G2. Remarkably, treatment with diclazuril and/or LF demonstrated potent antioxidant and anticoccidial effects, resulting in reduced shedding of oocysts, lesion scores, and lymphocytic infiltrates in the cecum. Additionally, these treatments improved the antioxidant and immune systems in chickens and restored all histopathological changes reported in the infected non-treated group (G2). Conclusion: This study offers novel perspectives on the potential anticoccidial effects of the combination of LF and diclazuril in broiler chickens infected with E. tenella, highlighting the potential synergistic actions of LF in treating poultry coccidiosis. Avian coccidiosis, a parasitic disease prevalent in poultry, is caused by Eimeria species and leads to significant economic losses. The use of attenuated live oocyst vaccines has been adopted as an alternative to the use of anticoccidial drugs. However, the accurate detection and differentiation of vaccine strains from virulent ones remain challenging. Therefore, this study presents a novel TaqMan polymerase chain reaction (PCR) detection method that offers enhanced sensitivity, specificity, and reproducibility compared with traditional PCR techniques. Through whole-genome resequencing and bioinformatics analysis, we identified a molecular marker gene, Em_marker6, with a unique 21-base pair deletion specific to the Eimeria maxima attenuated vaccine strain. Optimized primers and probes targeting this marker enabled rapid quantification cycle value achievement and high fluorescence intensity. The standard curve's slope of -3.540 and correlation coefficient of 0.9971 confirmed precise quantification capabilities. The TaqMan PCR method detected as few as 30 plasmid DNA copies and 50 oocysts per reaction, outperforming traditional PCR techniques by an order of magnitude. No cross-reactivity was observed with other E. maxima wide-type strains or common intestinal pathogens, ensuring the exclusive detection of the E. maxima EMPY vaccine strain. Weekly testing over 3 weeks demonstrated minimal variability, indicating robust consistency in the method's application. Testing on 61 clinical samples revealed a 57.38% positivity rate for E. maxima species and 13.11% for the vaccine strain. The Em_marker6 gene exhibited genetic stability across multiple generations, confirming the detection method's robust stability for the attenuated E. maxima vaccine strain. This study significantly advances the field of avian coccidiosis research and control by providing a valuable tool for monitoring vaccine purity and preventing inadvertent infections in vaccinated flocks, aligning with global efforts to curb antibiotic use in animal feed. Although anticoccidial drugs have been used to treat avian coccidiosis for nearly a century, resistance, bird harm, and food residues have caused health concerns. Thus, Nannochloropsis oculata was investigated as a possible coccidiosis treatment for broilers. A total of 150 1-day-old male Cobb broiler chicks were treated as follows: G1-Ng: fed a basal diet; G2-Ps: challenged with Eimeria spp. oocysts and fed basal diet; G3-Clo: challenged and fed basal diet with clopidol; G4-NOa: challenged and fed 0.1% N. oculata in diet, and G5-NOb: challenged and fed 0.2% N. oculata. Compared to G2-Ps, N. oculata in the diet significantly (P < 0.05) decreased dropping scores, lesion scores, and oocyst shedding. Without affecting breast meat colour metrics, N. oculata improved meat quality characters. At 28 days of age, birds received 0.2% N. oculata had significantly (P < 0.05) higher serum levels of MDA, T-SOD, HDL, and LDL cholesterol compared to G2-Ps. Serum AST, ALT, and urea levels were all decreased when N. oculata (0.2%) was used as opposed to G2-Ps. Histopathological alterations and the number of developmental and degenerative stages of Eimeria spp. in the intestinal epithelium were dramatically reduced by 0.2% N. oculata compared to G2-Ps. Molecular docking revealed a higher binding affinity of N. oculata for E. tenella aldolase, EtAMA1, and EtMIC3, which hindered glucose metabolism, host cell adhesion, and invasion of Eimeria. Salinomycin is a coccidiostat approved as a feed additive for the prevention of coccidiosis in poultry. Official control of its residues is set by the Commission Delegated Regulation (EU) 2022/1644. The aim of our study was to assess the suitability of three microbial inhibition tests (MITs), Premi®Test, Explorer 2.0, and the Screening Test for Antibiotic Residues (STAR) linked to the enzyme-linked immunosorbent assay (ELISA), for the screening of salinomycin residues in the tissues of broiler chickens (breast and thigh muscle, heart, liver, gizzard, kidneys, lungs, spleen, skin, and fat) fed commercially produced feed containing 70 mg.kg−1 of salinomycin in the complete feed. The first residue screening (Sampling A) was performed on the last day of administration of the salinomycin-medicated feed (day 30), and the second screening (Sampling B) was performed on the day of slaughter (day 37) after the expiry of the withdrawal period with the feeding of non-medicated feed. Based on the quantitative confirmation of salinomycin residues in the examined chicken tissues by the ELISA method (Sampling A from 0.025 to 0.241 mg.kg−1; Sampling B from 0.003 to 0.076 mg.kg−1), all the MITs with the preference of the bacterial strain Bacillus stearothermophilus var. calidolactis ATCC 10149 demonstrated the ability to detect the residues of salinomycin in the examined tissues of broiler chickens at the level of the maximum residue limits set from 0.015 to 0.150 mg.kg−1 by Commission Implementing Regulation (EU) 2017/1914 and confirmed the relevance of their sensitivity to the coccidiostat salinomycin. Coccidia of the genus Eimeria are important pathogens that cause coccidiosis in livestock and poultry. Due to the expansion of intensive farming, coccidiosis has become more difficult to control. In addition, the continued use of anti-coccidiosis drugs has led to drug resistance and residue. Some herbs used in traditional Chinese medicine (TCM) have been shown to alleviate the clinical symptoms of coccidiosis, while enhancing immunity and growth performance (GP) of livestock and poultry. Previous in vitro and in vivo studies have reported that the TCM herb Portulaca oleracea exhibited anti-parasitic activities. In total, 36 female Hu lambs were equally divided into six treatment groups: PL (low-dose P. oleracea), PH (high-dose P. oleracea), PW (P. oleracea water extract), PE (P. oleracea ethanol extract), DIC (diclazuril), and CON (control). The treatment period was 14 days. The McMaster counting method was used to evaluate the anti-coccidiosis effects of the different treatments. Untargeted metabolomics and 16S rRNA gene sequencing were used to investigate the effects of treatment on the gut microbiota (GM) and GP. The results showed that P. oleracea ameliorated coccidiosis, improved GP, increased the abundances of beneficial bacteria, and maintained the composition of the GM, but failed to completely clear coccidian oocysts. The Firmicutes to Bacteroides ratio was significantly increased in the PH group. P. oleracea increased metabolism of tryptophan as well as some vitamins and cofactors in the GM and decreased the relative content of arginine, tryptophan, niacin, and other nutrients, thereby promoting intestinal health and enhancing GP. As an alternative to the anti-coccidiosis drug DIC, P. oleracea effectively inhibited growth of coccidia, maintained the composition of the GM, promoted intestinal health, and increased nutrient digestibility. Coccidiosis is an intestinal protozoan disease that affects the poultry industry worldwide. The severity of this disease varies depending on the identity of the infectious agents. Therefore, this study was carried out to identify the Eimeria species that affect broiler chickens, Gallus gallus domesticus, through morphological and molecular phylogenetic analyses. Twenty‐five faecal samples were collected from the broiler chickens in a commercial poultry farm in Riyadh (Saudi Arabia). Using the floatation technique, faeces were examined microscopically for the Eimeria occurrence. Identification of Eimeria species was performed based on morphological criteria and molecular tools (DNA amplification for the partial small subunit ribosomal RNA (18S rRNA), internal transcribed spacer (ITS)‐1, and mitochondrial cytochrome c oxidase I (COI) genes. In this study, 32% (8 out of 25) of collected samples were found to be positive for coccidiosis. After sporulation in potassium dichromate (K2Cr2O7), the sporulated oocysts were observed as ovoid and measured 18.37–23.19 µm (19.87) long and 15.07–18.67 µm (16.46) wide, with the anterior location of a polar granule and absence of micropyle. These Eimeria oocysts were assumed to size and shape characteristics of Eimeria acervulina. Molecular analysis was conducted on the sequences of the polymerase chain reaction products from the three genes studied (18S rRNA, ITS‐1, and COI). At the three genes, results showed that the resultant sequences clustered with E. acervulina from different regions confirming morphological description. This study highlighted the importance of molecular techniques to detect avian Eimeria species more than the traditional morphology‐based tool to optimise the appropriate anticoccidial strategies for long‐term control in the studied area. Eimeria are the absolute intestinal parasites belonging to phylum Apicomplexa, causing the intestinal disease known as coccidiosis. It is a lethal disease that affects a large variety of animals and birds including cattle, camel, goats, sheep, poultry, etc. causing severe devastation and mortalities. Controlling poultry coccidiosis by the means of preventive and prophylactic use of chemical drugs is currently being practiced on a commercial scale. Regardless of a history of successful use of these drugs, resistance and public health concerns demand their alternatives. Several alternatives including vaccination, organic acids and botanicals, are being suggested. Several preparations of botanicals have been experimented and found useful, but exact compound identification is a question of interest for scientists. Phenolic compounds are among the most abundant phytochemicals found in diverse structural forms comprising mainly phenolic acids, tannins, flavonoids, and others. Research states that they have the potential to control coccidiosis because of their direct and indirect anticoccidial activities. Phenolics have been reported to control coccidiosis by targeting Eimeria species directly and reducing the damages by antioxidant, immunomodulatory, and anti-inflammatory properties. Phenolics kill Eimeria by engulfing schizonts, rupturing the cell membrane of sporozoites, and inhibiting growth by stopping ATP synthesis mechanisms and other pathways depending on their chemical nature and the subgroup of phenolic compounds. Because of the high potential of being oxidized; they capture the reactive species produced because of cell rupture during rapid schizogony and immune invasion. Phenolics give negative feedback to several inflammatory mediators and reduce the inflammatory destruction of the gut, hence minimizing the loss. Regardless of these activities, phenolics have some drawbacks i.e., they reduce nutrient uptake, disturb carbohydrate disturbance, have cytotoxic effects, and undefined long-term response. Extensive research is needed to investigate the proper anticoccidial compound and reduce the side effects of phenolics to manage coccidiosis in an accurate, rapid, and safe way. Two local isolates of Eimeria tenella gametocytes against coccidiosis were immunologically evaluated in chickens. Cell mediated immune response was detected by modified splenic cell migration inhibition assay (MSCMIA) and data were expressed in terms of per cent migration index. No significant difference in per cent migration index was detected for the chickens immunized either with Vaccine-I (local isolate-I) or with Vaccine-II (local isolate-II); however per cent migration index was comparatively lower in chickens immunized with Vaccine-II as compared to Vaccine-I; indicating a somewhat higher cell mediated immune (CMI) response. Humoral immune response was monitored by ELISA in vaccinated and control chickens. Significantly elevated (P<0.05) antibody titer (IgG) in chickens immunized with Vaccine-II as compared to Vaccine-I was detected. Significantly higher protection (67%) in chickens immunized with Vaccine-II followed by vaccine-I (49%) was recorded. Further, oocyst count was significantly lower (P<0.05) in chickens immunized with Vaccine-II as compared to those immunized with Vaccine-I. It was concluded that vaccinal strain (Vaccine-II) contained additional protein of high molecular weight (49.23 kDa) in its gametocytes provided cross protection and can be used to prepare commercial vaccine against coccidiosis in poultry. For years, there has been an increasing interest in natural alternatives to the conventional coccidiostats applied as feed additives, which have been used for decades to prevent coccidiosis in poultry and fattening rabbits. This study aimed to compare the possible anticoccidial effects of oregano oil to the established substance diclazuril in growing rabbits. The control group (CG) received a non-supplemented basal compound feed, to which either diclazuril (1 mg/kg; DG) or oregano oil (75 mg/kg; OG) was added. In each of the three trials, subgroups of 50 rabbits each were assigned to one of the three experimental groups (CG, DG and OG). Natural Eimeria infection was monitored weekly by fecal oocyst counts and Eimeria species identification following sporulation. Additionally, the performance parameters were determined at the middle and the end of the trials, and the deceased rabbits were subjected to necropsy. Neither oocyst excretion nor the performance parameters differed significantly between the three experimental groups. Eimeria media, Eimeria magna, Eimeria perforans and Eimeria exigua were identified as the occurring species. The highest animal losses (16.0%) occurred in the OG, while the losses were 12.7% in the DG and 12.0% in the CG. However, these differences were not statistically significant. Overall, neither diclazuril nor oregano oil was superior to the non-supplemented feed. This underlines the importance of diagnostics, as this study's results indicate that in the absence of the highly pathogenic Eimeria species, economic rabbit rearing and fattening is achievable without the use of coccidiostats. The objective of the current investigation was to evaluate several Eimeria challenges and the resulting alterations in intestinal permeability, intestinal morphology, and intestinal lesion scores in broiler chickens. This study included four groups with ten replicate cages per treatment, in which each group received a different treatment dosage of Eimeria, characterizing high, medium-high, and medium-low challenges. Five days after the challenge, intestinal lesions and permeability were assessed. The results showed that the increase in Eimeria challenge led to a considerable decrease in the height of intestinal villosities, in the ratio between villosity size and crypt depth, and in goblet cells. Moreover, after the challenge, there was a considerable increase in intestinal permeability. In conclusion, the medium-low, medium-high, and high-challenge models can be utilized for experimental infection. In the context of clinical studies, it has been observed that the administration of medium-high and high-challenge doses has proven to be adequate. However, it is advisable to utilize a medium-low challenge level to develop a subclinical challenge model for forthcoming investigations that aim to evaluate nutritional recommendations. Purpose: Coccidiosis of domestic chicken is an important disease caused by any of seven species of Eimeria which, by developing within the epithelial cells of the intestine, cause lesions therein. We carried out a study on poultry farms located in various regions of Iran to determine the incidence and spread of Eimeria species by employing a single PCR test. Methods: A total of 64 fully confirmed clinically intestinal tracts were collected from different parts of Iran. From these 64 intestinal tracts, 82 samples were prepared from the different sites involved in the digestive tract. In morphological assessment, 23 samples could not be isolated and its information was not evaluated. Results: Using morphological methods, the following seven species of Eimeria were identified: E. acervulina (15/59; 25.42%), E. tenella (30/59; 50.84%), E. maxima (12/59; 20.33%), E. praecox (1/59; 1.69%), E. necatrix (2/59; 3.38%), E. mitis (5/59; 8.47%), and E. mivati (2/59; 3.38%). Mixed infections were found in eight (13.55%) samples. In molecular assessment, 31 samples could not be isolated and its information was not evaluated. Totally, the following five species were identified using molecular methods: E. acervulina (35/51; 68.62%), E. tenella (33/51; 64.70%), E. maxima (6/51; 11.76%), E. brunetti (5/51; 9.80%), and E. necatrix (2/51; 3.92%). Mixed infections were found in 23 (45.09%) samples. Conclusions: The present study is an update on the situation of poultry coccidiosis in Iran and provides the first data on the molecular detection, identification, and characterization of Eimeria spp. in the poultry population of this country and confirmed the presence of different species of this parasite in this area. According to the results, E. acervulina and E. tenella, as the main disease-causing species, should be considered in control programs such as treatment and vaccination strategies. Purpose: The in vivo efficacy of ultrasonicated Rosmarinus officinalis ethanolic extract (UROEE) and its chitosan-loaded nanoparticles (UROEE-CsNPs) was investigated as a dietary prophylactic agent and as a therapeutic treatment against Eimeria tenella infected broiler chickens. Methods: Chickens were infected with 4 × 104E. tenella oocysts at 21 days old for primary infection and with 8 × 104 oocysts at 35 days old for secondary infection. Eleven experimental groups were conducted. Dietary addition of 100 mg/kg UROEE and 20 mg/kg for CsNPs as well as UROEE-CsNPs were included for prophylactic groups from day 1 to 42. The same doses were used for therapeutic treatment groups for 5 constitutive days. Oocyst output in feces was counted. Histopathological and immunohistochemical studies were conducted. Gene expression of pro-inflammatory cytokines as IFN-γ, IL-1β and IL-6 as well as anti-inflammatory cytokines as IL-10 and TGF-β4 was analyzed using semi-quantitative reverse transcriptase-PCR. Results: The results showed an efficacy of UROEE, CsNPs and UROEE-CsNPs in reduction of oocyst excretion and improving the cecal tissue architecture. CD4+ and CD8+ T lymphocytes protein expression were reduced. E. tenella infection lead to upregulation of pro-inflammatory cytokines as IFN-γ, IL-1β, IL-6 and anti-inflammatory cytokines as TGF-β4 following primary infection, while their expression was downregulated following secondary infection. Conclusion: The dietary prophylactic additives and therapeutic treatments with UROEE, CsNPs and UROEE-CsNPs could decrease the inflammatory response to E. tenella as indicated by oocyst output reduction, histopathological improvements, CD4+ and CD8+ T cells protein expression reduction as well as reducing mRNA expression levels of the tested cytokines following primary and secondary infections. Consequently, these results will help to develop better-combating strategies for the control and prevention of coccidiosis on poultry farms as a dietary prophylactic agent or as a therapeutic treatment. Aim: The epidemiological survey was carried out to determine the prevalence of eimeriosis in broiler chickens slaughtered depending the season, to determine the different Eimeria species causing the coccidiosis in poultry farms; and to assess the impact of Eimeria parasite on histomorphological structure and oxidative stress parameters of the intestine. Materials and Methods: The study was conducted from December 2018 to December 2019 in the province of Bejaia, Algeria. The intestines chickens (n = 366) were obtained immediately after slaughter, each cut into different segments (duodenum, jejunum, ileum, and caecum). Microscopic and parasitological examinations were performed according to standard procedures. Histomorphometric measurements of intestine were obtained using Image J software. Oxidative stress parameters were carried out from intestine tissue. Results: Eimeria spp. were detected in 73.77% (95% CI 71.20–76.34) of broiler gut samples. The prevalence varied significantly according to the season, with the lowest rates in winter (42.81%, 95% CI 40.35–45.27) and the highest in autumn (97.92%, 95% CI 97.6–99.4). All seven Eimeria species were identified, most commonly E. necatrix (27.70%), E. brunetti (26.47%), and E. tenella (20.96%). The mean lesion score ranged from 1.51 ± 0.05 to 1.79 ± 0.04. Significant differences in VH/CD ratio of intestinal epithelium (P < 0.001) were observed in different intestinal portions of infested broiler chickens compared to non-infested. The mean MDA concentration of intestinal segments was significantly higher in Eimeria species infested broilers compared to non-infested (P < 0.05). The results show at least one difference in CAT, SOD, and ABTS−+ concentrations (P < 0.05) in both chicken's groups. Conclusion: Our results revealed that coccidiosis is extremely prevalent in slaughtered broilers, with an abundance of pathogenic Eimeria species. Purpose: Avian coccidiosis is an important and widely distributed disease that affects global agricultural economies through losses. In Algeria, there is limited epidemiological and ecological knowledge about this disease and this hinders implementation of control strategies. A recent study, in Algeria, demonstrated a high prevalence and diversity of Eimeria species in broiler chickens. However, very little is known about the Eimeria species that exist on chicken farms raised on the floor and older than broiler chickens (for example, future laying hens and breeding hens) in Algeria. Methods: Samples were collected from 32 poultry farms located in 6 northeastern Algerian provinces (Algiers, Batna, Bejaia, Bordj Bou Arréridj, Jijel, Mila). These included 22 pre-laying pullet farms, with hens aged between 11 and 17 weeks, and 10 breeding hen farms with older hens (over 20 weeks). FTA cards were used to capture DNA and internal transcribed Spacer 1 PCR (ITS1-PCR) was used to determine the prevalence and composition of Eimeria species in the chickens. Results: This showed the presence of six species of Eimeria with a diverse prevalence range. Eimeria necatrix (63%) was the most common species, followed by E. maxima (53%), E. tenella (31%), E. brunetti (19%), E. acervulina and E. mitis (both 0.3%). Eimeria praecox was absent. Eimeria infection affected all farms studied where co-infections by different Eimeria species (63%) were more frequent than single infections (38%). The number of oocyts, per ml of enriched oocyst suspension was higher in breeding hen farms compared to pre-laying pullet farms. Chicken coccidiosis costs the poultry industry over GBP 10 billion per year. The main method of preventing and controlling coccidiosis in chickens continues to be the use of drugs. Unfortunately, the prevalence of drug resistance in the field reduces or even eliminates the effectiveness of drugs, and drug residues in the food supply chain can also can be harmful to humans. Therefore, safe and effective anticoccidial drugs are urgently needed. Natural products have many advantages such as being safe, effective and inexpensive and are a sustainable way to control coccidiosis. In this study, the anticoccidial effects of six natural compounds were tested by Eimeria tenella infection. Oocyst production, cecum lesion, body weight gain, feed conversion ratio, and intestinal microbiota were measured. The results showed that nerolidol had a moderate effect on maintaining both body weight gain and feed conversion ratio. Silymarin and dihydroartemisinin showed significant anticoccidial effects by reducing total oocyst output. Dihydroartemisinin also significantly reduced the cecum lesion caused by Eimeria infection, but this compound may be toxic to the host at such informed doses because it decreases growth and survival rates. In addition, both silymarin and dihydroartemisinin partly restored the microbiota after challenge. This indicates that silymarin, dihydroartemisinin, and nerolidol are effective in the control of chicken coccidiosis. Poultry coccidiosis is an intestinal infection caused by an intracellular parasitic protozoan of the genus Eimeria. Coccidia-induced gastrointestinal inflammation results in large economic losses, hence finding methods to decrease its prevalence is critical for industry participants and academic researchers. It has been demonstrated that coccidiosis can be effectively controlled and managed by employing anticoccidial chemical compounds. However, as a result of their extensive use, anticoccidial drug resistance in Eimeria species has raised concerns. Phytochemical/herbal medicines (Artemisia annua, Bidens pilosa, and garlic) seem to be a promising strategy for preventing coccidiosis, in accordance with the "anticoccidial chemical-free" standards. The impact of herbal supplements on poultry coccidiosis is based on the reduction of oocyst output by preventing the proliferation and growth of Eimeria species in chicken gastrointestinal tissues and lowering intestinal permeability via increased epithelial turnover. Background: The prevalence of avian coccidiosis in the poultry industry has grown, resulting in substantial financial losses from high mortality, stunted growth, reduced productivity, and expensive medical expenses. Aim: The purpose of the current study was to assess the immunological effects of neem leaf extract and toltrazuril on broilers that had contracted coccidiosis. Methods: In this investigation, 100 one-day-old Cobb broiler chicks without sexes were employed. The chicks were divided into five equal groups, with 20 birds in each. On the 14th day of life, the birds in groups 2, 3, 4, and 5 received an oral inoculation with 1 × 105 sporulated oocysts of Eimeria tenella (E. tenella) (field isolate). The first group (Gp), which consists of 20 healthy broilers, served as a negative control. Gp (2) contains experimentally infected broilers and nontreated (served as a positive control). Gp (3) contains experimentally infected broilers treated with toltrazuril (1 ml/l drinking water) for two consecutive days. Gp (4) contains experimentally infected broilers treated with neem leaf extract 4% (50 ml/l drinking water) for 5 successive days, and Gp (5) contains experimentally infected broilers treated with toltrazuril (1 ml/l drinking water) and a half dose of neem leaves extract 4% (25 ml/l drinking water) for 5 successive days. For the purpose of estimating body weight growth and feed conversion ratio, each broiler was weighed separately at the start of the trial and again on the 1st and 10th day after treatment. In addition to obtaining intestinal samples for immunohistochemistry, blood samples were also obtained for immunological examination. Results: As compared to the negative control group, the experimentally infested broilers with E. tenella showed significant decreases in serum nitric oxide, lysosome, phagocytic percent, and phagocytic index, along with significant increases in white blood cells (WBCs), lymphocyte, heterophilis, eosinophilis, basophilis, monocyte, serum total protein, γ globulin, fibrinogen, and haptoglobin. When compared to the control positive group, experimentally infested broilers treated with either neem or toltrazuril alone or in combination demonstrated significant increases in serum total protein, nitric oxide, lysozyme, phagocytic percent, and phagocytic index, but significant decreases in WBCs, lymphocytes, heterophile, eosinophile, basophile, and monocyte. The intestinal peroxidase stain of broilers infected with E. tenella exhibited a significant positive expression for CD4, but the infected broilers treated with toltrazuril and half a dosage of neem displayed a negative expression for CD4, identical to the negative control. Conclusion: The broiler chickens infested with E. tenella may have a variety of negative impacts on their immune systems and immunohistopathological findings. This study assessed the impact of thyme (Thymus vulgaris), oregano (Origanum vulgare), and garlic (Allium sativum) essential oils, as well as their bioactive compounds, on the initial phase of schizogony in Madin–Darby bovine kidney cells, comparing their effectiveness to two commercially used anticoccidial drugs. Using image analysis and quantitative PCR, the study confirmed the efficacy of commercial anticoccidials in reducing invasion and schizont formation, and it found that essential oils were equally effective. The poultry sector is facing multiple challenges among which infectious diseases are major hindrances. Coccidiosis, especially cecal coccidiosis is the most prominent disease because of its acute nature and high mortality. Controlling through synthetic chemicals and ionophores is becoming less effective, botanicals especially essential oils are being considered for the control of coccidiosis. In this study, broiler birds at day 10 of age were divided into 6 groups. Three concentrations of flaxseed (Linum usitatissimum) essential oil 1, 2 and 3% (v/w) were administered orally to estimate their anticoccidial effects compared to toltrazuril medicated, infected non-medicated, and non-infected non-medicated controls. Oocysts per gram of feces, oocyst score, fecal score, lesion score, feed conversion ratio, weight gain, complete blood profile, and liver and renal function-related serum parameters were evaluated. Analysis of variance and Tukey's post hoc test were used to compare the means and analyze statistically. The results showed that flaxseed essential oil had significant (p<0.05) anticoccidial activity in reducing oocysts per gram, oocyst score, fecal score, and lesion score. There was a significant decrease in mortalities, while weight gain and FCR were also improved by therapy of flaxseed essential oil at a concentration of 3%. Flaxseed essential oil also showed significant positive effects on red blood cell counts, packed cell volume, mean corpuscular hemoglobin, and hemoglobin concentration. Total proteins and albumins were significantly (p<0.05) increased by the flaxseed essential oil while other serum-related parameters remained in the normal range. Avian coccidiosis compromises welfare and production performance, increases susceptibility to bacterial infections and leads to substantial economic and production losses upon the poultry industry. The objective of this study was to determine the impact of a mixed coccidiosis (Eimeria acervulina, Eimeria maxima, Eimeria tenella and Eimeria necatrix) challenge on oxidative stress biomarkers, performance indices, carcass and meat quality of broilers. Broilers challenged with coccidiosis had higher concentrations of catalase, glutathione-S-transferase and malondialdehyde and lower concentration of superoxide dismutases at all sampling days. The same group of broilers had a lower feed intake on day 21 and 35, and a lower daily weight gain and body weight on day 35. Healthy broilers had higher hot and cold carcass weights and breast weight, but a lower back and pelvis weight. Also, healthy broilers produced better meat quality in terms of higher water-holding capacity (lower drip and thawing loss in breast and thigh muscle) and less yellow colour (lower b* value in breast and thigh muscle). Chemical composition of healthy broiler meat was better in terms of higher content of dry matter, ash and protein in breast muscle. Avian coccidiosis arises from co-infection involving multiple Eimeria species, which could give rise to substantial economic losses in the global poultry industry. As a result, multivalent anticoccidial vaccines containing common Eimeria antigens offer considerable promise for controlling co-infection in clinical practice. In our previous study, Elongation factor 2 (EF2) was deemed as an immunogenic common antigen across various Eimeria species. This current investigation aimed to further assess the immunogenicity and protective efficacy of EF2 in recombinant subunit vaccine format against three Eimeria species. The EF2 gene cloned from Eimeria maxima (E. maxima) cDNA was designated as EF2 of E. maxima (EmEF2). The immunogenicity of the recombinant protein EmEF2 (rEmEF2) was assessed through Western blot analysis. The evaluation of the vaccine-induced immune response encompassed the determination of T lymphocyte subset proportions, cytokine mRNA transcription levels, and specific IgY concentrations in rEmEF2-vaccinated chickens using flow cytometry, quantitative real-time PCR (qPCR), and indirect enzyme-linked immunosorbent assay (ELISA). Subsequently, the protective efficacy of rEmEF2 was evaluated through vaccination and challenge experiments. The findings demonstrated that rEmEF2 was effectively recognized by the His-tag monoclonal antibody and E. maxima chicken antiserum. Vaccination with rEmEF2 increased the proportions of CD4+ and CD8+ T lymphocytes, elevated IL-4 and IFN-γ mRNA transcription levels, and enhanced IgY antibody levels compared to the control groups. Moreover, compared to the control groups, vaccination with rEmEF2 led to decreased weight loss, reduced oocyst outputs, and alleviated enteric lesions. Furthermore, in the rEmEF2-immunized groups, challenges with E. maxima and E. acervulina resulted in anticoccidial index (ACI) scores of 166.35 and 185.08, showing moderate-to-excellent protective efficacy. Nevertheless, challenges with E. tenella and mixed Eimeria resulted in ACI scores of 144.01 and 127.94, showing low protective efficacy. In conclusion, EmEF2, a common antigen across Eimeria species, demonstrated the capacity to induce a significant cellular and humoral immune response, as well as partial protection against E. maxima, E. acervulina, and E. tenella. Clinical avian coccidiosis is typically caused by coinfection with several Eimeria species. Recombinant protein and DNA vaccines have shown promise in controlling coccidiosis. On this basis, DNA vaccines that encode multiple epitopes from different Eimeria species may provide broad protection against coinfections. In this study, we designed a fusion gene fragment, 14EGT, that contained concentrated T-cell epitopes from four common antigens of Eimeria species (14-3-3, elongation factor 2, glyceraldehyde-3-phosphate dehydrogenase, and transhydrogenase). The multiepitope DNA vaccine pVAX1-14EGT and recombinant protein vaccine pET-32a-14EGT (r14EGT) were then created based on the 14EGT fragment. Subsequently, cellular and humoral immune responses were measured in vaccinated chickens. Vaccination-challenge trials were also conducted, where the birds were vaccinated with the 14EGT preparations and later exposed to single or multiple Eimeria species to evaluate the protective efficacy of the vaccines. According to the results, vaccination with 14EGT preparations effectively increased the proportions of CD4+ and CD8+ T cells and the levels of Th1 and Th2 hallmark cytokines. The levels of serum IgG antibodies were also significantly increased. Animal vaccination trials revealed alleviated enteric lesions, weight loss, and oocyst output compared to those of the control groups. The preparations were found to be moderately effective against single Eimeria species, with the anticoccidial index (ACI) ranging from 160 to 180. However, after challenge with multiple Eimeria species, the protection provided by the 14EGT preparations was not satisfactory, with ACI values of 142.18 and 146.41. Collectively, the results suggest that a multiepitope vaccine that encodes the T-cell epitopes of common antigens derived from Eimeria parasites could be a potential and effective strategy to control avian coccidiosis. Coccidiosis is acclaimed as the most prevalent enteric parasitic ailment of poultry. It is caused by an apicomplexan protozoon of the genus Eimeria, which resides in chicken intestinal epithelium leading to intestinal damage. As a result, bloody droppings are there, feed efficiency is reduced, the growth rate is impaired, and egg production is temporarily decreased. Treatment and prevention of coccidiosis are primarily accomplished by inoculating live vaccines and administering anticoccidial drugs. Due to anticoccidials' continuous and excessive use, the mounting issue is drug resistant Eimeria strains. The poultry industry has managed resistance-related issues by suggesting shuttle and rotation schemes. Furthermore, new drugs have also been developed and introduced, but it takes a long time and causes cost inflation in the poultry industry. Moreover, government disallows growth promoters and drugs at sub-therapeutic doses in poultry due to increased concerns about the drug residues in poultry products. These constraints have motivated scientists to work on alternative ways to control coccidiosis effectively, safely, and sustainably. Background: Coccidiosis, a prominent intestinal protozoan disease, carries significant economic implications for the poultry industry. The aim of this study was to evaluate the effects of Fengqiang Shengtai (BLES), a probiotics product, and coccidiosis vaccine in modulating the intestinal microbiome and providing insight into mitigating the occurrence and management of avian coccidiosis. Methods: Broilers included in the study were divided into four pre-treatment groups: the Pre-Con group (commercial diet), Pre-BLES group (BLES supplement), Pre-Vac group (coccidiosis vaccination) and Pre-Vac-BLES group (combined vaccination and BLES). Body weight gain, feed consumption and feed conversion ratio were monitored from age 25 to 55 days. Cecum contents were collected at 8 and 15 days of age for comparative analysis of intestinal microbiomes. In the Pre-BLES and Pre-Vac-BLES groups, probiotics were administered at a dose of 0.01 g per chicken between ages 3 to 6 days and 10–13 days. At 3 days of age, chickens in the Pre-Vac and Pre-Vac-BLES groups were vaccinated with 1700 sporulated oocysts of the live coccidiosis vaccine per chicken. At the age of 25 days, Eimeria spp. challenge experiments were performed based on the aforementioned immunization strategy, and the oocysts per gram (OPG) in the feces, intestinal lesion score and intestinal pathological characteristics were evaluated. Specifically, 30 chickens were randomly selected from each group and orally administered 34,000 sporulated oocysts of Eimeria spp. per chicken, re-defined as Eimeria group, BLES-Eimeria group, Vac-Eimeria group and Vac-BLES-Eimeria group, respectively. Additionally, 30 chickens were randomly selected from the Pre-Con group and included as negative control without Eimeria spp. challenge. Intestinal microbiota was sequenced and analyzed when the broilers were 32 days old. Results: A significant improvement was observed in body weight gain of the broilers in the Pre-BLES and Pre-Vac-BLES group at 45 days of age. Analysis of the intestinal microbiota revealed a positive correlation between the experimental groups receiving BLES and coccidiosis vaccines at 8 and 15 days of age with the Enterococcus genus and Lachnospiraceae NK4A136 group, respectively. In addition to the reduced lesion score and OPG values, the combination of coccidiosis vaccine and BLES also reduced the intestinal epithelial abscission induced by coccidiosis vaccines. The results of intestinal microbial function prediction demonstrated that N-glycan biosynthesis and ferroptosis were the prominent signal pathways in the Vac-BLES-Eimeria group. Conclusions: Taken together, the results of the present study suggest that supplementation of BLES with coccidiosis vaccine represents a promising strategy for improving growth performance, alleviating clinical manifestations and inducing favorable alterations to the intestinal microbiota in broiler chickens affected by coccidiosis. The objective of this study was to evaluate the effectiveness of live attenuated commercial vaccine LIVACOX® T against avian coccidiosis upon parasite exposure through its correlation with productive and economic performance, clinical observation, and oocyst excretion of broiler chickens. For this purpose, 420 1-day-old Cobb chicks were divided into five groups of 84 birds: (G1) unvaccinated and unchallenged control; (G2) vaccinated on day 0; (G3) challenged on day 1; (G4) vaccinated on day 0 and challenged on day 14; and (G5) challenged on day 14. For 28 days, the clinical signs of infection, weight and feed conversion of the birds, and excretion of oocysts in the feces were evaluated. Macroscopic analysis of intestinal lesions in birds was also performed. After vaccination in G2, G3, and G4 as well as after challenge in G3, G4, and G5, there was an increase in oocyst excretion. In the analysis of weight gain, the difference in final weight between groups G3 and G4 is − 105.74 g per bird. Therefore, if we multiply this value by the average number of birds slaughtered per day in a medium/large slaughterhouse (250,000), we have 26,435 kg of chicken meat per day of slaughter, representing 581,570 kg of monthly losses (22 days of slaughter/month), or approximately R$3,489,420.00 (US$872,355.00), considering the commercial value at R$6.00/kg (US$ 1.5/kg). Thus, the productive and economic impact of coccidiosis in broiler chickens is evident, and the importance of vaccination to prevent the occurrence of the disease and reduce subsequent loss is highlighted. The present study was done to know the incidence of coccidiosis in naturally affected broiler birds in 10 different farms located in Ayodhya and Sultanpur districts of Uttar Pradesh for six months (October, 2021 to March, 2022). These farms were visited during study period to screen the cases of coccidiosis on the basis of clinical signs and symptoms like marked depression and weakness, decreased feed intake, feed conversion and diarrhoea. Out of total 22600 birds under observation at the ten farms, 7300 were clinically found to be naturally affected with coccidiosis. The overall incidence of the coccidiosis was 30.96%. Out of 5 farms in Ayodhya district, total 11500 birds were under observation, 3781 birds were clinically affected. The overall incidence in Ayodhya district was 31.49%. Out of 5 farms in Sultanpur district, total of 11100 birds were under observation, 3519 birds were clinically affected. The overall incidence in Sultanpur district was 30.43%. In the present study, the overall mortality was 26.74%. Both in vitro and in vivo studies were conducted to evaluate the beneficial effects of green tea extract (GT), cinnamon oil (CO), and pomegranate extract (PO) on avian coccidiosis. In experiment (EXP) 1, an in vitro culture system was used to investigate the individual effects of GT, CO, and PO on the proinflammatory cytokine response and integrity of tight junction (TJ) in chicken intestinal epithelial cells (IEC), on the differentiation of quail muscle cells and primary chicken embryonic muscle cells, and anticoccidial and antibacterial activities against Eimeria tenella sporozoites and Clostridium perfringens bacteria, respectively. In EXP 2 and 3, in vivo trials were carried out to study the dose-dependent effect of blended phytochemicals (GT, CO, PO) on coccidiosis in broiler chickens infected with E. maxima. For EXP 2, one hundred male broiler chickens (0-day-old) were allocated into the following five treatment groups: Control group for non-infected chickens (NC), Basal diet group for E. maxima-infected chickens (PC), PC group supplemented with phytochemicals at 50 (Phy 50), 100 (Phy 100), and 200 (Phy 200) mg/kg feed diets for E. maxima-infected chickens. For EXP 3, one hundred twenty male broiler chickens (0-day-old) were allocated into the following six treatment groups: NC, PC, PC supplemented with phytochemicals at 10 (Phy 10), 20 (Phy 20), 30 (Phy 30), and 100 (Phy 100) mg/kg feed for E. maxima-infected chickens. Body weights (BW) were measured on days 0, 7, 14, 20, and 22, and jejunum samples were used to measure cytokine, TJ protein, and antioxidant enzyme responses at 8 days post-infection (dpi). Fecal samples for oocyst enumeration were collected from 6 to 8 dpi. In vitro, CO and PO reduced LPS-induced IL-1b and IL-8 in IEC, respectively, and GT enhanced the gene expression of occludin in IEC. PO at 1.0 and 5.0 mg/mL exerted antimicrobial effect against E. tenella sporozoites and C. perfringens bacteria, respectively. In vivo, chickens fed a diet supplemented with phytochemicals showed enhanced BW, reduced oocyst shedding, and decreased proinflammatory cytokines following E. maxima challenge. In conclusion, the combination of GT, CO, and PO in the diet of broiler chickens infected with E. maxima induced enhanced host disease resistance including innate immunity and gut health, which contributed to improved growth and reduced disease responses. Coccidiosis causes huge economic losses worldwide. Current study evaluated the effect of biosynthesized Zinc oxide nanoparticles (ZnONPs) using Nigella sativa, on Eimeria tenella infected broilers. Scanning electron microscopy showed spherical ZnONPs with 50–100 nm diameter, Fourier transforms infrared spectroscopy revealed the functional groups involved in the reduction of zinc acetate dihydrate to ZnONPs, UV–vis spectroscopy showed a peak at 354 nm, and Zeta potential exhibited stability at − 30 mV. A total of 150, a day-old broiler chicks were divided into 5 equal groups. Control negative: uninfected and untreated; Control positive: Infected and untreated; 3rd, 4th and 5th group were infected orally with 5 × 104 sporulated oocysts of Eimeria tenella and treated with 60 mg/kg ZnONPs, 1% Nigella sativa seeds and amprolium 125 ppm, respectively. ZnONPs significantly (p < 0.05) improved the growth performance in the infected birds and decreased the oocyst shedding and anti-coccidial index. A significant (p < 0.05) decrease in the level of aspartate transferase and alanine transferase, whereas, a significantly higher amount of antioxidants like catalase and superoxide dismutase in ZnONPs treated group was observed. Pro-inflammatory cytokines like IL-2 and TNF-α were significantly decreased by ZnONPs (p < 0.05). Background and Aim: Eimeria tenella is a causative agent of avian cecal coccidiosis resulting in bleeding, diarrhea, weight loss, high morbidity, and mortality in chickens. Zinc supplementation increases body weight gain, reduces mortality, and improves some immune response parameters of broilers infected with E. tenella. This study aimed to investigate the effects of zinc hydroxychloride (ZnOHCl) supplementation and ZnOHCl combined with an anticoccidial drug on E. tenella infection in broiler chickens. Materials and Methods: Forty one-day-old broilers were randomly divided into five groups, the study was replicated twice and had four chickens per replicate. Group 1 was an uninfected and unmedicated control group, and Group 2 was an infected but unmedicated control group. Group 3 was infected and treated with 120 mg/kg ZnOHCl, Group 4 was infected and medicated with 7 mg/kg toltrazuril (TOL), and Group 5 was infected and treated with 120 mg/kg ZnOHCl and 7 mg/kg TOL. Body weight gain, feed intake, and feed conversion ratio were monitored on days 15, 21, and 28. Oocyst shedding, hematological data, and lesion scores were analyzed on 7 days post-infection. Results: The average weight gain, feed intake, and packed cell volume of chickens treated with ZnOHCl and TOL were significantly higher than those of the infected and unmedicated controls (p = 0.05). Lesion scores, oocyst output, and lymphocytes of the chickens treated with ZnOHCl and TOL were significantly lower than those of the infected and unmedicated controls (p = 0.05). Conclusion: This study demonstrated that zinc supplementation alone reduced only oocyst output. However, growth performance, lesion scores, and oocyst output were affected by the combination of ZnOHCl and TOL supplementation. Currently, coccidiostats are the primary and standard method of combating coccidiosis in poultry farms. However, consumer organizations still point to the need to phase out all chemotherapeutic substances from animal nutrition to protect human health. The research material consisted of 720 cocks of the Cobb 500 hybrid, which were divided into three groups (C – coccidiostat/control, V – vaccine, and H – herbals). The following parameters were analyzed: body weight (BW), feed conversion ratio (FCR), mortality, foot pad dermatitis (FPD), European Production Efficiency Factor (EPEF), and the number of oocysts per 1g of feces (OPG). On day 42, the BW of the C group was higher compared with the V and H groups. The C group was also characterized by the best FCR and the highest EPEF. In the V group, oocysts were noted in feces from the 14th day of rearing, and the highest oocysts content was observed on the 21st day of life. In the C and H groups, the highest number of oocysts was recorded in the feces on the 28th day. The investigated alternative methods to coccidiostats showed good antiparasitic potential. Coccidiosis caused by protozoan parasite of Eimeria species is one of the most important poultry diseases prevalent in different parts of globe including India. The parasite invades the intestinal cells and the disease is characterised by enteritis and diarrhoea leading to weight loss, altered feed conversion ratio, poor growth and eventually death of affected birds. A survey on commercial poultry farms was conducted in 10 districts of Haryana state, India, using a questionnaire to assess the status of coccidiosis, associated risk factors and management practices followed. On analysis of data, it was found that overall, 15 (28.8%) farms had history of coccidiosis. One-fourth (25%) of the farmers reported the use of anticoccidial drugs for chemoprophylaxis or therapeutic purposes at their farms. Further, on statistical analysis using Chi-square, several factors were found to be statistically significant (p<0.05) with occurrence of coccidiosis at these farms including type of birds, capacity of birds, visitor's entry, litter removal frequency, number of antibiotics used for treatment and foggers use in summer at farms under study. These factors were identified as predisposing and/ or risk factors associated with occurrence of coccidiosis at the farms. The odds of occurrence of coccidiosis were significantly higher (p<0.001) at the farms with presence of risk factors identified in the study as compared to the farms where risk factors were absent. The study revealed history of coccidiosis at several poultry farms leading to undue economic burden to farmers. For effective management of coccidiosis at the farms, the use of foggers may be discouraged. Essential oils are potent medicinal substances that can control multiple diseases through their bioactivities. In the current research, the anticoccidial potential of the Amomum subulatum essential oil was estimated in broiler chicks. A day-old, 270 broiler chicks were divided equally into 6 groups, each having 3 replicates. All the groups, except the infected non-medicated control group, were given sporulated oocysts of mixed Eimeria spp. The first three groups received essential oil of A. subulatum @ 1%, 2%, and 3% (v/w) in the feed, and their activities were evaluated in comparison to the Symocox® (Toltrazuril) treated groups and infected non-medicated groups. Feed conversion ratio (FCR), feed intake, weight gain, rates of mortality, organ weight ratios, fecal scores, lesion scores, and oocyst per gram of feces were taken as the parameters of the study. Blood parameters e.g. red blood cell, white blood cells, packed cell volume, mean corpuscular hemoglobin, and serum metabolites like alanine aminotransferase, aspartate transferase, alkaline phosphatases, lactate dehydrogenase, urea, proteins, and albumins were also evaluated. A. subulatum essential oil @ 3% concentration had the best FCR, survival rates, and feed intakes. A. subulatum essential oils also significantly reduced lesions lowered oocyst counts and improved fecal scores. Hematological and serum profiles were improved by the essential oil of A. subulatum in the feed and the best results were achieved by the 3% concentration of the oil. This research concluded that the A. subulatum essential oil @ 3% concentration can be used for the treatment of avian coccidiosis however formulation of the dose requires further studies on broiler chicks. Background: Avian coccidiosis is thought to be one of the most expensive infectious diseases in the poultry industry. Objectives: Safe and alternative anti‐coccidial drugs are herbal extracts because they do not result in tissue residue and drug resistance. The objective of the present study was to evaluate the anti‐coccidial effect of the herbal mixture, a complex of two plants (Echinacea purpurea, Glycyrrhiza glabra) in broiler chickens in comparison with toltrazuril. Methods: One hundred twenty broiler chickens were used in this experiment and divided into 4 equally numbered groups. All the groups, except Group D, were experimentally infected with mixed Eimeria spp. (E. Tenella, E. maxima, E. necatrix and E. brunetti) on day 14. Group A treated with an herbal mixture [Glycyrrhiza glabra Extract 5% (standardised to 5.4% glycyrrhizic acid) and Echinacea purpurea Extract 2% (standardised to 4% total phenolic content based on chlorogenic acid); Coxinin‐EC®; Shamim Teb Sepid Giti]. Group B treated with toltrazuril. Group C was experimentally infected with mixed Eimeria spp. but they did not have any treatment, this group was our positive control. Performance indices, faecal oocyst excretion, and intestinal lesion score were determined during the experiment. Results: Positive control group had the poorest results and more mortality than other groups. Group D was not infected and was healthy all the experiment period. Treatment with herbal complex significantly reduced the negative performance and pathogenic effects associated with Eimeria spp. at a level that was comparable with toltrazuril. Conclusions: In summary, the anti‐coccidial activity of the studied herbal complex suggests its use as an alternative anti‐coccidial agent to chemotherapeutic drugs for controlling coccidiosis in poultry. Avian coccidiosis causes huge economic losses to the poultry industry worldwide and currently lacks effective live vector vaccines. Achieving efficient antigen delivery to mucosa-associated lymphoid tissue (MALT) is critical for improving the effectiveness of vaccines. Here, chicken claudin-3 (CLDN3), a tight junction protein expressed in MALT, was identified as a target, and the C-terminal region of Clostridium perfringens enterotoxin (C-CPE) was proven to bind to chicken CLDN3. Then, a CLDN3-targeting Lactobacillus plantarum NC8-expressing C-CPE surface display system (NC8/GFP-C-CPE) was constructed to successfully express the heterologous protein on the surface of L. plantarum. The colonization level of NC8/GFP-C-CPE was significantly increased compared to the non-targeting strain and could persist in the intestine for at least 72 h. An oral vaccine strain expressing five EGF domains of Eimeria tenella microneme protein 8 (EtMIC8-EGF) (NC8/EtMIC8-EGF-C-CPE) was constructed to evaluate the protective efficacy against E. tenella infection. The results revealed that CLDN3-targeting L. plantarum induced stronger mucosal immunity in gut-associated lymphoid tissues (GALT) as well as humoral responses and conferred better protection in terms of parasite replication and pathology than the non-targeting strain. Overall, we successfully constructed a CLDN3-targeting L. plantarum NC8 surface display system characterized by MALT-targeting, which is an efficient antigen delivery system to confer enhanced protective efficacy in chickens against E. tenella infection. For many years, ionophores have been used to control coccidiosis in poultry. However, misuse of ionophores can cause toxicity with significant clinical symptoms. The most critical factors influencing ionophores' toxicity are administration dose, species, and animal age. Although clinical signs of ionophore intoxication are well studied, the toxicity mechanisms of the ionophores at the molecular level still are not fully elucidated. This review summarizes the studies focused on polyether ionophores toxicity mechanisms in animals at the clinical and molecular levels. Studies show that ionophore toxicity mainly affects myocardial and skeletal muscle cells. The molecular mechanism of the toxication could be explained by the inhibition of oxidative phosphorylation via dysregulation of ion concentration. Tiamulin-ionophore interaction and the synergetic effect of tiamulin in ionophore biotransformation are discussed. Furthermore, in recent years ionophores were candidates for reprofiling as antibacterial and anti-cancer drugs. Avian coccidiosis is the primary parasitic disease that afflicts poultry and is responsible for significant economic losses. Vaccination has emerged as a viable alternative to chemical treatments for controlling broiler coccidiosis and has shown considerable potential for mitigating its impact. This study aimed to evaluate the effectiveness of Fortegra® vaccination in controlling coccidiosis in broiler chickens. A total of 245 unsexed one-day-old broiler chickens obtained from Masr poultry were divided into seven groups, with 35 birds in each group. Four groups (GA1, GA2, GA3, and GA4) were vaccinated with Fortegra® while three groups (GB1, GB2, and GB3) served as non-vaccinated controls. At 6 days of age, all chicks in the vaccinated group were administered Fortegra®, while all groups except GA3 and GB3 were infected with coccidia at day 22. The results showed that vaccination did not prevent infection but resulted in lower lesion scores compared to the conventional and control groups. Body weight in the vaccinated group (GA) was lower than that of the non-vaccinated (GB) groups. Microscopic examination of fecal samples collected at different ages confirmed coccidia infection in all experimental groups. Broiler intestinal mucosa scrapings revealed typical forms of Eimeria species, leading to bloody diarrhea and petechial hemorrhage. After a 72-hour period at 30°C with forced aeration, sporulation occurred, and oocysts were stored in a potassium dichromate solution. Microscopic examination confirmed the presence and shape of oocysts, and experimental samples displayed a 500 bp amplicon size specific to Eimeria spp. While Fortegra® vaccination did not prevent infection, it resulted in lower lesion scores compared to the conventional and control groups. Thus, vaccination may serve as an effective approach for controlling coccidiosis in broiler chickens, but further research is necessary to optimize vaccination protocols and evaluate its long-term effects on broiler production. Coccidiosis poses a significant challenge in poultry production and is typically managed with ionophores and chemical anticoccidials. However, the emergence of drug resistance and limitations on their use have encouraged the exploration of alternative solutions, including botanical compounds and improvements in in vitro screening methods. Prior research focused only on the impact of these alternatives on Eimeria invasion, with intracellular development in cell cultures receiving limited attention. This study assessed the impact of thyme (Thymus vulgaris), oregano (Origanum vulgare), and garlic (Allium sativum) essential oils, as well as their bioactive compounds, on the initial phase of schizogony in Madin–Darby bovine kidney cells, comparing their effectiveness to two commercially used anticoccidial drugs. Using image analysis and quantitative PCR, the study confirmed the efficacy of commercial anticoccidials in reducing invasion and schizont formation, and it found that essential oils were equally effective. Notably, thymol and carvacrol exhibited mild inhibition of intracellular replication of the parasite but significantly reduced schizont numbers, implying a potential reduction in pathogenicity. In conclusion, this research highlights the promise of essential oils and their bioactive components as viable alternatives to traditional anticoccidial drugs for mitigating coccidiosis in poultry, particularly by disrupting the intracellular development of the parasites.

Avian coccidiosis arises from co-infection involving multiple Eimeria species, which could give rise to substantial economic losses in the global poultry industry. As a result, multivalent anticoccidial vaccines containing common Eimeria antigens offer considerable promise for controlling co-infection in clinical practice. In our previous study, Elongation factor 2 (EF2) was deemed as an immunogenic common antigen across various Eimeria species. This current investigation aimed to further assess the immunogenicity and protective efficacy of EF2 in recombinant subunit vaccine format against three Eimeria species. The EF2 gene cloned from Eimeria maxima (E. maxima) cDNA was designated as EF2 of E. maxima (EmEF2). The immunogenicity of the recombinant protein EmEF2 (rEmEF2) was assessed through Western blot analysis. The evaluation of the vaccine-induced immune response encompassed the determination of T lymphocyte subset proportions, cytokine mRNA transcription levels, and specific IgY concentrations in rEmEF2-vaccinated chickens using flow cytometry, quantitative real-time PCR (qPCR), and indirect enzyme-linked immunosorbent assay (ELISA). Subsequently, the protective efficacy of rEmEF2 was evaluated through vaccination and challenge experiments. The findings demonstrated that rEmEF2 was effectively recognized by the His-tag monoclonal antibody and E. maxima chicken antiserum. Vaccination with rEmEF2 increased the proportions of CD4+ and CD8+ T lymphocytes, elevated IL-4 and IFN-γ mRNA transcription levels, and enhanced IgY antibody levels compared to the control groups. Moreover, compared to the control groups, vaccination with rEmEF2 led to decreased weight loss, reduced oocyst outputs, and alleviated enteric lesions. Furthermore, in the rEmEF2-immunized groups, challenges with E. maxima and E. acervulina resulted in anticoccidial index (ACI) scores of 166.35 and 185.08, showing moderate-to-excellent protective efficacy. Nevertheless, challenges with E. tenella and mixed Eimeria resulted in ACI scores of 144.01 and 127.94, showing low protective efficacy. In conclusion, EmEF2, a common antigen across Eimeria species, demonstrated the capacity to induce a significant cellular and humoral immune response, as well as partial protection against E. maxima, E. acervulina, and E. tenella. These results highlight EmEF2 as a promising candidate antigen for the development of multivalent vaccines targeting mixed infections by Eimeria species.

Rumex nervosus leaves (RNL) and Cinnamomum verum bark (CNB), phytogenic herbs, have received much attention in recent years for their antimicrobial properties; however, there is limited knowledge about their potential anticoccidial functions. The prophylactic effects of RNL and CNB were compared with salinomycin, a standard synthetic anticoccidial agent, in broilers experimentally infected with Eimeria tenella (E. tenella). One-day-old broiler chicks (n = 225) were randomly divided into nine groups. Birds were either fed a basal diet containing 1, 3, or 5 g RNL or 2, 4, or 6 g CNB/kg feed, or treated with salinomycin within the basal diet, or the infected (IUT) or non-infected (UUT) groups were fed a basal diet only. Birds infected (n = 25 bird/group) with 40,000 sporulated E. tenella oocysts/bird at d 21 except UUT. Bodweight gain (BWG) and feed conversion ratio (FCR) were significantly (p <.05) lower in IUT compared to UUT. On day 7 post-infection (DPI), birds treated with RNL, CNB, or salinomycin had fewer lesions in the caeca and a lower oocyst value, and a higher oocyst reduction rate in the faeces than birds in the IUT. Although RNL was not able to reduce weight loss caused by coccidiosis, CNB at 6 g improved FCR and production efficiency index (PEI) at 7 DPI compared to the infected groups. In conclusion, RNL at 5 g and CNB at 6 g have moderate anti-coccidial activity and could be used to treat poultry coccidiosis in the field. However, more research is needed to identify active ingredients that make it effective compared to commercially available drugs. Phytogenic feed additives prevented weight loss and caecum pathology in broiler chickens at risk of coccidiosis. Rumex nervosus leaves and Cinnamomum verum bark had similar effects to the coccidiostat salinomycin at high doses. Simple Summary: Avian coccidiosis is one of the major diseases threatening the poultry industry in Egypt and worldwide. As such, developing a new oocysticidal to control this disease seems crucial. The present study examined the in vitro effects of allicin and alcoholic garlic extract at different concentrations as natural disinfectant candidates compared with KOH 5% as a chemical disinfectant against Eimeria tenella oocysts and the in vivo infectivity of these treated oocysts in chickens. The morphological changes in the treated oocysts were examined using light microscopy and scanning electron microscopy. The in vivo infectivity was evaluated using a gross and histopathological examination of the cecal tissues to determine the most significant changes. The study showed promising results for using Allicin at a concentration of 180 mg/mL and garlic at a concentration of 360 mg/mL in reducing the number of oocysts, the sporulation percentage, and the number of oocysts with marked deformities. The sporulated oocysts exposed to Allicin and alcoholic garlic extract revealed the lowest infectivity and pathogenicity, resulting in a minor gross lesions score. In addition, restoration of the major reported histopathological changes compared to oocysts sporulated in media containing the KOH 5%. Collectively, this study provides novel information regarding the potential use of natural extracts of allicin and alcoholic garlic as environmentally eco-friendly disinfectants in poultry farms to reduce the spread of coccidiosis. To determine their effect on the viability of Eimeria tenella oocysts in vitro, allicin and alcoholic garlic extract, which are natural, less toxic, and inexpensive products, were compared to KOH 5% (chemical disinfectant) using an in vitro culture system. Three concentrations of allicin (45, 90, and 180 mg/mL) and alcoholic garlic extract (90, 180, and 360 mg/mL, were used. Subsequently, destructive and sporulation-inhibiting effects on Eimeria oocysts were detected using light and electron microscopy. Young chickens were infected with treated sporulated oocysts to determine their effect on infectivity. After 7 days pi, the percentage of excreted oocysts (oocyst shedding) was determined, and the chickens were slaughtered for histopathological examination of the cecal tissues. Under an electron microscope, allicin at a concentration of 180 mg/mL and alcoholic garlic extract at a concentration of 360 mg/mL demonstrate a high oocysticidal activity with severe destruction of the oocyst wall and the appearance of pores. In addition, both concentrations directly affected the infectivity of sporulated oocysts by reducing the shedding of oocysts and the pathological lesions of infected young chickens. Avian coccidiosis is a common enzootic disease caused by infection of Eimeria species parasites. It causes huge economic losses in the global poultry industry. Current control using anticoccidial drugs or vaccination is limited due to drug resistance and the relatively high cost of vaccines. Improving host genetic resistance to Eimeria species is considered an effective strategy for improved control of coccidiosis. Circular RNAs (circRNAs) have been found to function as biomarkers or diagnoses of various kinds of diseases. The molecular biological functions of circRNAs, miRNAs, and mRNAs related to Sasso chicken have not yet been described during Eimeria species challenge. In this study, RNA-seq was used to profile the expression pattern of circRNAs, miRNAs, and mRNAs in spleens from Eimeria tenella-infected and non-infected commercial dualpurpose Sasso T445 breed chickens. Results showed a total of 40 differentially expressed circRNAs (DEcircRNAs), 31 differentially expressed miRNAs (DEmiRNAs), and 820 differentially expressed genes (DEmRNAs) between infected and non-infected chickens. Regulatory networks were constructed between differentially expressed circRNAs, miRNAs, and mRNAs to offer insights into the interaction mechanisms between chickens and Eimeria spp. Functional validation of a significantly differentially expressed circRNA, circMGAT5, revealed that circMGAT5 could sponge miR-132c-5p to promote the expression of the miR-132c-5p target gene monocyte to macrophage differentiation-associated (MMD) during the infection of E. tenella sporozoites or LPS stimulation. Pathologically, knockdown of circMGAT5 significantly upregulated the expression of macrophage surface markers and the macrophage activation marker, F4/80 and MHC-II, which indicated that circMGAT5 might inhibit the activation of macrophage. miR-132c-5p markedly facilitated the expression of F4/80 and MHC-II while circMGAT5 could attenuate the increase of F4/80 and MHC-II induced by miR-132c-5p, indicating that circMGAT5 exhibited function through the circMGAT5-miR-132c-5p-MMD axis. Together, our results indicate that circRNAs exhibit their resistance or susceptive roles during E. tenella infection. The objective of this study was to evaluate the phytotherapeutic efficacy of Allium sativum L. (garlic) as alternative compounds to antibiotics used by farmers in the treatment of coccidiosis in poultry. In this study, 20 chickens divided into 4 batches received only garlic powder in their feed. The results obtained after a natural infestation revealed a weight loss in all subjects before administration of the different products. Following treatment, a considerable decrease in coccidia eggs in the chickens receiving the highest concentrations of the products. The weight gain was more or less high in the treated batches except in the control batches. In conclusion, this study generated biological results. These data support that garlic could be used as alternatives to antibiotics in poultry and many other livestock operations. This study aimed to investigate the possible anticoccidial efficacy of Moringa oleifera leaves vs. Diclazuril, as well as production performance and antioxidant activities in broilers exposed to experimental coccidiosis. A total of 160 one-day-old broiler chicks (Ross 308) were randomly divided into four groups (n=40) as follows: Group 1: unchallenged, untreated; Group 2: challenged, untreated; Group 3: challenged, treated with diclazuril 1mg/kg ration; and Group 4: challenged, treated with Moringa oleifera leaves (20g/kg ration). The birds of all groups except those of group1 were infected with mixed Eimeria species at the day 14th of the experiment. The mortality rates were 14.8 % in Group 2, 8.8 % in Groups 3 and zero % in group 4. A significant improvement of the final body weight and the feed conversion ratio induced by Moringa oleifera group when compared with that obtained by diclazuril group. The average lesion score was decreased in medicated groups with a better effect in the caecum for Group 3, while Group 4 exhibited a much decrease in lesion score in the upper and the lower intestinal parts and in the caecum. The average oocysts count in Groups 3 and 4 was 7.9x10³ and 14.5x10³, respectively, while Group 2 had 67.1x10³ oocysts. Furthermore, both Moringa oleifera leaves and Diclazuril increased plasma carotenoids levels and growth performance, as well as decreased oxidative stress. Moringa oleifera leaves were sucess to be as efficient as diclazuril against coccidiosis. It could be concluded that, for the prevention of coccidiosis in poultry production, Moringa Oleifera may be a viable alternative product. Background and Objectives: Avian coccidiosis is an intestinal parasitic disease exerting a highly negative impact on the global poultry industry. The aim of the present study is to evaluate the immune protective efficacies against Eimeria tenella infection in chickens orally immunized with combined recombinant probiotics Entercoccus faecalis (E. faecalis) delivering surface-anchored E. tenella proteins. Methods: Four kinds of novel probiotics vaccines that surface-expressing four Eimeria tenella (E. tenella) proteins EtAMA1, EtIMP1, EtMIC2 and Et3-1E were produced, respectively. The expression of four target proteins on the surface of recombinant bacteria was detected by Western blot and indirect immunofluorescence assay (IFA). Then the four kinds of recombinant E. faecalis were combined to immunize chickens via oral route in different combinations. The immunizations were performed three times at two-week intervals, and each for three consecutive days. After immunizations, chickens in each immunized group were orally challenged with E. tenella sporulated oocysts. The immune responses and protective efficacies against homologous infection were evaluated. Results: The results showed that three or four live recombinant E. faecalis induced effective antigen-specific humoral, intestinal mucosal immune responses, stimulated peripheral T lymphocytes proliferation, and displayed partial protections against homologous challenge as measured by cecal lesions, oocyst shedding, and body weight gain (BWG). Notably, higher levels of protective efficacies were observed when the four recombinant E. faecalis delivering target proteins were combined. Conclusion: Chickens orally administrated with three or four, especially the four combined recombinant E. faecalis stimulated specific immune responses, which provided anti-coccidial effects. The study was conducted to consider the anticoccidial and immunogenic effectivities of encapsulated organic acids and anticoccidial drugs in broilers reared on a reused litter infected with Eimeria spp. for simulating in-field exposure to avian coccidiosis. 525 mixed-sex one-day-old broiler chicks (Ross 308) were used in a 2 × 3 factorial experiment as a completely randomized design with seven experimental groups and five replicates of 15 chicks. The seven experimental groups were included: negative (uninfected; T1) and positive (infected; T2) control groups fed a diet without additive, and other infected groups (T3–T7) fed diets supplemented with 0.05% maduramicin, 0.02% diclazuril, 0.1% EOAs, 0.05% maduramicin and 0.1% EOAs, 0.02% diclazuril and 0.1% EOAs. During the experimental period, the evaluated parameters were European production efficiency factor (EPEF; at 22 days of age (d)), oocyst output per gram feces (OPG; at different ages), oocyst reduction rate (ORR; at 22-d), survival rate (SR; at 22-d), caecal lesion score (CLS at 22-d), sporulation percentage (SP; by in vitro anticoccidial tests), bloody diarrhea (BD; by scoring the bloody feces each morning from 13 to 31-d), immunity (humoral test at 28 and 35-d and cell-mediated test at 22-d), goblet cells analysis of the jejunum (GC; at 22-d) and anti-coccidiosis index (ACI; at 22-d). EOAs and anticoccidials, especially their simultaneous feeding improved (P < 0.05) broiler's EPEF, SR, OPG, ORR, SP, CLS, immunity and BD (scored). ACI was improved (P < 0.05) by EOAs more than anticoccidials (marked vs. moderate). The highest ACI was significantly observed in EOAs + diclazuril group. EOAs as a safe alternative had more intensive anticoccidial and immunogenic properties and increased the anticoccidial drugs' effectiveness, especially diclazuril in Eimeria spp-infected broilers. Two local isolates of Eimeria tenella gametocytes against coccidiosis were immunologically evaluated in chickens. Cell mediated immune response was detected by modified splenic cell migration inhibition assay (MSCMIA) and data were expressed in terms of per cent migration index. No significant difference in per cent migration index was detected for the chickens immunized either with Vaccine-I (local isolate-I) or with Vaccine-II (local isolate-II); however per cent migration index was comparatively lower in chickens immunized with Vaccine-II as compared to Vaccine-I; indicating a somewhat higher cell mediated immune (CMI) response. Humoral immune response was monitored by ELISA in vaccinated and control chickens. Significantly elevated (P<0.05) antibody titer (IgG) in chickens immunized with Vaccine-II as compared to Vaccine-I was detected. Significantly higher protection (67%) in chickens immunized with Vaccine-II followed by vaccine-I (49%) was recorded. Further, oocyst count was significantly lower (P<0.05) in chickens immunized with Vaccine-II as compared to those immunized with Vaccine-I. It was concluded that vaccinal strain (Vaccine-II) contained additional protein of high molecular weight (49.23 kDa) in its gametocytes provided cross protection and can be used to prepare commercial vaccine against coccidiosis in poultry. Due to the emergence of drug resistance against synthetic chemicals, effective alternatives like essential oils are required for the control of coccidiosis in poultry. Hence, the present in-vivo experiment was conducted on Citrus (C.) sinensis essential oil for investigation of its anticoccidial efficacy. For this purpose, 72 dayold chicks were procured from local market and divided randomly into six equal groups: A, B, C, D, E and F. On day 14, all the groups except F were administered with 50,000 sporulated oocysts (mixed Eimeria species). On the same day, groups A, B and C were given 1, 2 and 3% of C. sinensis essential oil, respectively, in feed, group D with Toltrazuril® while groups E and F were left unmedicated. The results revealed C. sinensis essential oil to have positive effect on FCR in broilers. Moreover, there was a significant improvement in oocysts per gram of faeces, lesion score, oocyst score and faecal score in oil treated broilers at the maximum inclusion level of 3% comparable to the standard control. However, apart from significant effect on WBCs count and serum levels of ALT and LDH, C. sinensis essential oil had very limited non-significant effect on hematological and serum biochemical parameters, and organs' weight. Thus, the present study provided C. sinensis essential oil to be an alternate anticoccidial agent which can be used in coccidiosis control programs after further validation. [ABSTRACT FROM AUTHOR]
