INTRODUCTION: A three-pronged approach to acne treatment combining an antibiotic, antimicrobial, and retinoid may be more efficacious than single/double treatments while potentially reducing antibiotic resistance. This study evaluated the efficacy and safety of the first fixed-dose, triple-combination topical acne product, clindamycin 1.2%/adapalene 0.15%/benzoyl peroxide (BPO) 3.1% gel (CAB) using pooled phase 3 data. METHODS: In two identical phase 3 (N = 183; N = 180), double-blind, 12-week studies, participants aged ≥ 9 years with moderate-to-severe acne were randomized 2:1 to receive once-daily CAB or vehicle gel. Endpoints included ≥ 2-grade reduction from baseline in Evaluators Global Severity Score and clear/almost clear skin (treatment success) and least-squares mean percent change from baseline in acne lesion counts. Treatment-emergent adverse events (TEAEs) and cutaneous safety/tolerability were evaluated. RESULTS: At week 12, 50.0% of participants achieved treatment success with CAB versus 22.6% with vehicle gel (P  70% reductions in inflammatory and noninflammatory lesions at week 12 (77.9% and 73.0%, respectively), which were significantly greater than vehicle (57.9% and 48.2%; P

Weaned dairy heifers are a relatively understudied production group. Bovine respiratory disease (BRD) is the most common cause of antimicrobial drug (AMD) use, morbidity, and mortality in this production group. The study of antimicrobial resistance (AMR) is complicated because many variables that may affect AMR are related. This study generates hypotheses regarding the farm- and animal-level variables (e.g., vaccination, lane cleaning, and AMD use practices) that may be associated with AMR in respiratory isolates from weaned dairy heifers. A cross-sectional study was performed using survey data and respiratory isolates (Pasteurella multocida, Mannheimia haemolytica, and Histophilus somni) collected from 341 weaned dairy heifers on six farms in California. Logistic regression and Bayesian network analyses were used to evaluate the associations between farm- and animal-level variables with minimum inhibitory concentration (MIC) classification of respiratory isolates against 11 AMDs. Farm-level variables associated with MIC classification of respiratory isolates included the number of source farms of a calf-rearing facility, whether the farm practiced onsite milking, the use of lagoon water for flush lane cleaning, and respiratory and pinkeye vaccination practices. Animal-level variables associated with a MIC classification included whether the calf was BRD-score-positive and time since the last phenicol treatment.

Low soil phosphorus (P) availability is a limiting factor for crop production. Application of livestock manure as organic fertilizer can increase soil P availability, but may cause soil contamination with antibiotics such as sulfadiazine (SD), thus adversely affecting plant growth and nutrient uptake. The effects of P (0 and 50 mg kg−1) and SD (0, 1, 10, and 100 mg kg−1) levels on plant growth, root development, and nitrogen (N) and P uptake of maize and soybean were examined in a pot experiment using a loess soil as the test soil. High levels of SD considerably inhibited plant growth of both crops. Both P fertilization and SD negatively affected root development of both crops, especially at higher SD levels. For both crops, the average root diameter and the proportion of thick roots increased with increasing soil SD level. When soil SD level was ≤10 mg kg−1, the effects of P fertilization and SD on plant N and P concentrations, and N:P, N- and P-utilization efficiency were not significant in most cases. The effects of SD on plant N and P nutrition could not be of a great concern when soil SD level was ≤10 mg kg−1. Overall, soil SD pollution could reduce plant biomass and inhibit root development of soybean and maize, with a stronger effect on root development than on plant biomass at a high soil SD level (≥10 mg kg−1). Kasugamycin (KSM), an aminoglycoside antibiotic, has been widely used as a natural fungicide to control plant diseases, particularly for managing rice blast. However, its uptake mechanism and transport in rice remain to be explored. In this article, rice seedlings were treated by the foliar spraying method, and the content of KSM in rice leaves, stems and roots under different treatments was detected by high‐performance liquid chromatography tandem mass spectrometry (HPLC‐MS/MS), respectively. Results showed that leaf‐applied KSM could be transported to the phloem and migrate to roots and stems after uptake by leaves. Concentration, temperature and pH had significant effects on the uptake of KSM. Compared with the control, the competitive inhibitors d‐glucose and phlorizin both inhibited the uptake of KSM, demonstrating that sugar transporter proteins were involved in the uptake process. The energy inhibitors dinitrophenol (DNP) and carbonyl cyanide chlorophenylhydrazone (CCCP) also significantly inhibited the uptake of KSM, indicating that the uptake of KSM required energy consumption. Thus, the uptake of KSM by rice was an active process involving sugar transporter proteins, and it could migrate downward through the phloem. This study contributes to the promotion of the scientific application of antibiotics and the biological control of crop diseases. The antibiotic administration in dairy livestock is a current widespread practice. The purpose of the study was to evaluate whether milk from antibiotic‐treated animals, following the application of the withdrawal time and fully complying with European requirements in relation to residue content, affected the development of lactic acid bacteria. Forty‐five raw milk samples were collected after the withdrawal period and analysed to verify their compliance with the European maximum limits using four commercial screening tests. Lactobacillus delbrueckii subsp. bulgaricus growth and acidifying activity were delayed from 5 to 8 h in milk from cow treated with β‐lactams and sulfonamides present in low concentration (cephalosporins < 15 μg/kg; penicillins < 8 μg/kg; sulfonamides < 10 μg/kg) and their effect persisted over time. No influences were detected in Streptococcus thermophilus and Lb. helveticus development. A novel zinc‐based metal–organic framework (Zn‐MOF), designated as [Zn3(L)(bpyp)2(HCOO)3·2DMF·H2O] (1), was synthesized using 2,5‐bis(pyrid‐4‐yl)pyridine (bpyp), 1,3,5‐benzenetricarboxylic acid (H3L), and Zn(CH3COO)2·2H2O in a DMF solution. Single‐crystal X‐ray diffraction analysis revealed that MOF 1 crystallizes in the monoclinic system with a C2/c space group, featuring a (3,5)‐connected binodal network. The structural integrity and characteristics of MOF 1 were confirmed by Fourier transform infrared (FT‐IR), thermal gravimetric analysis (TGA), powder X‐ray diffraction (PXRD), and Brunauer–Emmett–Teller (BET) analyses, showing type IV isotherms indicative of microporous structures. The photocatalytic efficiency of MOF 1 was evaluated for the degradation of various antibiotics under UV light, with nitrofurantoin (NFT) demonstrating the highest degradation rate of 83.9% at an optimal concentration of 40 ppm and catalyst loading of 5 mg. The degradation process followed a pseudo‐first‐order kinetic model with a rate constant of 0.02492 min−1. Radical trapping experiments identified superoxide radicals (O2˙−) and holes (h+) as the predominant reactive species, while hydroxyl radicals (˙OH) played a negligible role. Reusability tests over four cycles confirmed the stability and durability of MOF 1, with consistent photocatalytic performance and unchanged structural morphology as evidenced by PXRD and SEM analyses. The proposed mechanism involves photoinduced electron–hole pair generation, with subsequent formation of reactive oxygen species (ROS) facilitating NFT degradation. This study highlights MOF 1 as a promising photocatalyst for environmental remediation, specifically for the efficient degradation of pharmaceutical contaminants in aquatic systems, providing insights into its structural properties, photocatalytic activity, and underlying degradation mechanisms. Introduction: Surgical site infections (SSI) are one of the most common complications after extensive sarcoma resections and represent a daily challenge. SSI occur in up to 50% of cases particularly in the peripelvic area. One possible approach to reduce infection rate is perioperative antibiotic prophylaxis. The aim of this study therefore was to investigate the influence of perioperative antibiotic prophylaxis on the infection rate and the possible influence of location-specific antibiotic prophylaxis with ampicillin/sulbactam. Methods: This monocentric retrospective study included 366 patients who underwent sarcoma resections in the groin, proximal thigh, or gluteal region. All patients were operated on by 2 surgeons after neoadjuvant pretreatment if necessary. 3 groups of patients were defined. Group 1: In 60.4% of all cases, antibiotic prophylaxis was administered with cephalosporins (also clindamycin in case of penicillin allergy). Group2: In 9.8% of cases, ampicillin/sulbactam was used. Group 3: 29.8% of patients did not receive any antibiotic prophylaxis. Results: In 31.1% of treated cases, antibiotic therapy was prolonged due to extended tumor resections. Postoperative infections occurred in 23.2% (85 cases), in 77 cases within the first 90 days (on average after 20 days). The median operating time, blood loss was higher, and tumor size were significantly larger in cases with infections, compared to patients without infection. In group 1 and 2 with perioperative single-shot prophylaxis, infection occurred in 24.1% of cases, compared to 13.5% of cases without prophylaxis (group 3) (p= 0.032). In the patients with prolonged antibiotic therapy, infection occurred in 31.6% of cases, compared to 16.3% of cases without prolongation (p< 0.001). In the group 2, infection occurred in 19.4% of cases compared to 24.9% of cases in the group 1 (p= 0.479). In the multivariate analysis, surgery time longer 80 min, blood substitution, neoadjuvant radio- and chemotherapy proved to be a risk factor for SSI. Discussion: Region adapted perioperative antibiotic prophylaxis may reduce the risk of infection after extended sarcoma resection in the peripelvic area. Objective: Bacterial vaginosis is a disease caused by vaginal microecology disorder, which seriously affects female health. At present, there are many drugs to treat BV, and this study aims to compare the efficacy and safety of multiple drugs for BV through a network meta-analysis (NMA). Methods: All studies were sourced from PubMed and Embase databases from the establishment date to April 13, 2023. We evaluated the clinical cure success rate and adverse effects (abnormal increase in vaginal discharge, external genital irritation, and vulvar itching) and performed subgroup analyses of the clinical cure success rate for different modes of administration. All statistical analyses were performed using R and STATA 14.0 software for network meta-analysis. Results: Weincluded42studies thatmetthecriteria, involving a total of8382patients. Network meta-analysis results showed that metronidazole and secnidazole had a higher rate of adverse reactions than placebo (RR 7.06; 95%-CI 2.61-19.10, RR 4.03; 95%-CI1.63-9.98), theadverse reaction rateofprobioticsgroupwaslowerthanthat of metronidazole group (RR 0.44; 95%-CI 0.21-0.93). The clinical cure rate of oral ornidazole was better than clindamycin (RR 16.08; 95%-CI 1.72-150.47), Secnidazole (RR 8.17; 95%-CI 1.66-40.25) and probiotics. Directmeta-analysis results showed that ornidazole had a better clinical cure rate than Secnidazole (RR1.22; 95%-CI 1.10-1.34), oral ornidazole had a better clinical cure rate than Secnidazole (RR 1.23; 95%-CI 1.11-1.36). The clinical cure rate of vaginal application of sucrose was better than metronidazole (RR 1.12; 95%-CI 1.03-1.21) and metronidazole had a lower clinical cure rate than probiotics (RR 0.68; 95%-CI 0.52-0.88). Conclusions: The results of this systematic review and network meta-analysis suggest that ornidazole may be an effective alternative for the treatment of BV, and that sucrose and probiotics are potential BV treatments that need to be validated by more high-quality clinical studies in the future. Introduction: Oritavancin is a semi-synthetic lipoglycopeptide antibiotic primarily used to treat serious infections caused by Gram-positive bacteria. The aim of this study was to elucidate possible molecular targets of oritavancin in human and microbes in relevance to its mechanism of action and model its pharmacokinetics for optimal dose selection in clinical practice. Methods: Computational methods were used in this study which include target prediction, molecular docking, molecular dynamics simulation, pharmacokinetics prediction, and physiological-based pharmacokinetics (PBPK) modeling. Results: Oritavancin was moderately soluble in water and did not permeate the blood-brain barrier. Seven molecular targets were identified in humans. Molecular docking results showed highest binding affinity of oritavancin with PI3-kinase p110-gamma subunit (−10.34 kcal/mol), followed by Acyl-CoA desaturase (−10.07 kcal/mol) and Cytochrome P450 2C19 (−8.384 kcal/mol). Oritavancin PBPK modelling in adult human showed that infusion has lower peak concentrations (Cmax) compared to bolus administration, with 1200 mg dose yielded Cmax of 16.559 mg/L, 800 mg dose yielded Cmax of 11.258 mg/L, and 200 mg over 3 days dose yielded Cmax of 7.526 mg/L. Notably, infusion gave extended half-life (t1/2) for all doses and slightly higher clearance rates compared to bolus, particularly for the 1200 mg and 800 mg doses. The results corroborated existing clinical pharmacokinetic data, and confirmed the model's accuracy and predictive capability. Conclusion: This comprehensive computational study has provided invaluable insights into the pharmacological profile of Oritavancin, aiding its further development and optimization for clinical use. Nigella sativa (N. sativa; Ranunculaceae), commonly referred to as black cumin, is one of the most widely used medicinal plants worldwide, with its seeds having numerous applications in the pharmaceutical and food industries. With the emergence of antibiotic resistance in pathogens as an important health challenge, the need for alternative microbe-inhibitory agents is on the rise, whereby black cumin has gained considerable attention from researchers for its strong antimicrobial characteristics owing to its high content in a wide range of bioactive compounds, including thymoquinone, nigellimine, nigellidine, quercetin, and O-cymene. Particularly, thymoquinone increases the levels of antioxidant enzymes that counter oxidative stress in the liver. Additionally, the essential oil in N. sativa seeds effectively inhibits intestinal parasites and shows moderate activity against some bacteria, including Bacillus subtilis and Staphylococcus aureus. Thymoquinone exhibits minimum inhibitory concentrations (MICs) of 8–16 μg/mL against methicillin-resistant Staphylococcus aureus (MRSA) and exhibits MIC 0.25 µg/mL against drug-resistant mycobacteria. Similarly, quercetin shows a MIC of 2 mg/mL against oral pathogens, such as Streptococcus mutans and Lactobacillus acidophilus. Furthermore, endophytic fungi isolated from N. sativa have demonstrated antibacterial activity. Therefore, N. sativa is a valuable medicinal plant with potential for medicinal and food-related applications. Background: Approximately, 10% of people report a penicillin allergy; however, more than 90% can safely undergo delabeling after a detailed history, oral challenge, or other investigations such as penicillin skin testing (PST). Although PST is the gold standard, the results can be heterogeneous, and awaiting specialist assessment may take an inordinate amount of time. Therefore, oral provocation challenge has become acceptable for individuals with low-risk penicillin allergy histories. There also appears to be an association with increased prevalence of adverse drug reaction reporting in female individuals, which may translate to penicillin allergy prevalence; however, the evidence has not been assessed through a sex and gender lens. This systematic review will identify and synthesize the findings from studies that report measures of effectiveness and safety of interventions aimed at delabeling penicillin allergies in low-risk individuals. Information related to sex and gender will be extracted, where available, to understand potential differences in allergy reporting and patient outcomes. Methods: The Cochrane Handbook for Systematic Reviews of Interventions and the Centre for Review and Dissemination's Guidance for Undertaking Reviews in Health Care will be used as frameworks for conducting this systematic review. The literature search will be conducted by a medical librarian (B. M. M.) and will consist of a search strategy to identify and retrieve published studies that meet our inclusion criteria. Studies that require penicillin skin testing (PST) as a step prior to other interventions will be excluded. Integrated knowledge translation involving co-design was carried out for this systematic review protocol creation. Data extraction will be conducted at four levels: (1) study level, (2) patient level, (3) intervention level, and (4) outcome level. A narrative descriptive synthesis of results and risk of bias of all included studies will be provided, and, if relevant, a meta-analysis will be performed. Discussion: The dissemination of findings from this knowledge synthesis to various stakeholders is intended to inform on options for evidence-based interventions to aid in delabeling penicillin allergies in individuals with a low risk of experiencing a hypersensitivity reaction. Detailed reporting on the characteristics of delabeling interventions as well as the effectiveness of similar interventions will benefit policy makers considering the implementation of a penicillin allergy delabeling protocol. Additionally, findings from this systematic review will report on the current evidence regarding the role of sex and gender in both the prevalence and outcomes associated with the presence of penicillin allergies. The kinetic tendencies in the destruction of cephalosporin antibiotics (ceftriaxone (CEF)) in photoinitiated oxidative systems were studied using a xenon lamp as a source of quasisolar radiation (UV–Vis). It was established that the oxidative systems under study can be arranged in the following series according to the efficiency and rate of antibiotic destruction: {UV–Vis/Fe2+/S2O } > {Fe2+/S2O } {UV–Vis/S2O } > {UV–Vis}. The optimum conditions for the oxidative destruction of CEF in the {UV–Vis/Fe2+/S2O } system are reached at [S2O ] : [CEF] = 30 : 1 and [S2O ] : [Fe2+] = 1 : 0.1. As the temperature increases to 40°C, the initial CEF oxidation rate and efficiency increase. The apparent activation energy of the CEF oxidation in the {UV–Vis/Fe2+/S2O } system was 45 kJ mol–1, which is comparable to the values obtained for cephalosporin antibiotics. It was proved, using inhibitors of radical reactions, that the oxidative destruction of CEF in combined {UV–Vis/Fe2+/S2O } proceeds by a multiradical mechanism involving reactive oxygen species (ROS): hydroxyl radicals and sulfate and superoxide radical anions. The obtained tendencies are in good agreement with the results of open-air studies with natural solar radiation; in the {Solar/Fe2+/S2O } system, the oxidation of CEF is significantly intensified due to combined activation of persulfate by the iron ions, the UV-C (<300 nm) component of natural solar radiation, and the thermal exposure. The results indicate that using the combined {Solar/Fe2+/S2O } oxidation system is promising for the destruction of antibiotics in order to reduce their release into the environment. 400 test chicken (1-day-old AA broilers) were randomly selected and divided into 4 groups. The test chicken in the blank control group was only fed with the basal diet, the test chicken in the test groups Ⅰ, Ⅱ and Ⅲ were fed with the basal diet + 0. 1% gentamycin, the basal diet + 0. 2% compounded Chinese herbal medicine preparation, the basal diet + 0. 2% fermented compounded Chinese herbal medicine preparation respectively. The effects of the fermented compound medicinal plant agent on the growth performance, blood biochemical indices and antioxidant properties of broilers were investigated. The results showed that at 1- 25 day-age, compared with control group, the daily weight gain of broilers in test groups Ⅰ, Ⅱ and Ⅲ increased by 2. 24%, 2. 24% and 4. 50% respectively (P<0. 05), feed-to-weight ratio all decreased by 2. 21% (P<0. 05). At 26-49 day-age, the daily weight gain of the broilers in test groups Ⅰ, Ⅱ and Ⅲ increased by 2. 31%, 4. 52% and 4. 99% respectively (P<0. 05), the daily weight gain of broilers in test group Ⅲ increased by 0. 45% and 2. 62% respectively compared with that in test groups Ⅱ,Ⅰ. Compared with the control group, the crude protein utilization rate of the broilers in test groups Ⅰ, Ⅱ and Ⅲ increased by 1. 86%, 1. 77% and 3. 46% respectively (P<0. 05), and the crude protein utilization rate of the broilers in test group Ⅲ increased by 1. 66% and 1. 57% respectively (P<0. 05) compared than that in test group. The crude fat utilization rate of the broilers in test groups Ⅰ, Ⅱ and Ⅲ increased by 0. 98%, 0. 97%, 1. 76% respectively (P<0. 05) compared with that in the control group. The excessive presence of antibiotics such as Oxytetracycline (OTC) in the wastewater has increased health problems due to their toxic impact on the aquatic ecosystem. Therefore, their removal has become an important topic. This study aims to produce high surface area-activated carbon derived from low-cost and environmentally friendly barley lignocellulosic wastes to remove OTC from aqueous solutions. The synthesized barley wastes-activated carbon (BW-AC) was characterized using Fourier-Transform Infrared spectroscopy, Thermal Gravimetric Analysis, X-ray diffraction analysis, N2 adsorption/desorption isotherms, and Scanning Electron Microscopy. A Central Composite Design under the Response Surface Methodology (CCD-RSM) was applied to optimize the operational parameters (adsorbent dosage, pH, OTC initial concentration, and contact time) affecting the adsorption capacity as the response factor. The optimum condition of OTC adsorption by BW-AC was the adsorbent dosage of 16.25 mg, pH of 8.25, initial concentration of 62.50 mg/L, and contact time of 23.46 min. An analysis of variance (ANOVA) was performed to investigate the significance of the designed quadratic model and evaluate the parameters interactions. The linear regression coefficient (R2) of 0.975 shows a good correlation between predicted and actual results. The adsorption isotherms were used to determine the contaminant distribution over the adsorbent surface, and the equilibrium data was best described by the Freundlich isotherm due to the R2 value of 0.99 compared to other isotherms and β parameter of 0.23 in Redlich-Peterson equation. Moreover, the n value of 1.25 in Freundlich equation and E value of 0.31 in Dubinin–Radushkevich equation indicates a physical nature of adsorption process. According to the equations results, the maximum adsorption capacity of BW-AC for OTC removal was 500 mg/g, based on the Langmuir isotherm equation. In addition, the thermodynamic studies indicated an endothermic process based on the 0.31 value of ΔH° and spontaneous nature due to the negative amount of ΔG° within the temperature range of 288–318 K. Consequently, the prepared BW-AC can be deemed as a highly effective adsorbent with a large surface area, resulting in significant capacity for removing OTC. This synthesized BW-AC can serve as an environmentally friendly adsorbent for affordable wastewater treatment and is poised to make valuable contributions to future research in this field. A review of medical records in dogs that underwent medical dissolution of upper urinary tract uroliths utilizing diet, administration of antibiotics, and double‐pigtail ureteral stent(s) placement, when indicated, was performed. Medical management was generally continued for 4 weeks beyond urolith dissolution. Information on biochemical, microbiological, imaging, and clinical outcomes before and after dissolution were recorded.Six dogs (9 kidneys) were included with bilateral (3) or unilateral (3) nephrolithiasis, ureterolithiasis, or a combination. A ureteral stent(s) was placed endoscopically in 5/6 dogs (6/9 kidneys) for obstructive ureterolithiasis (n = 5) or a nonobstructive massive nephrolith (n = 1). All dogs had a positive urine culture of Staphylococcus pseudintermedius with a median urine pH of 7.25 (range, 6.5‐8) and 4/5 had pyonephrosis. All dogs had initial evidence of urolith dissolution at a median of 1.1 months (range, 0.42‐5.9), with complete dissolution of ureteroliths at a median of 3.9 months (range, 1.5‐7.6), nephroliths at 5.3 months (range, 1.5‐7.6), and lower urinary tract uroliths at 0.87 months (range, 0.42‐5.9). Stents were removed in 3/6 once dissolution was documented. The median follow‐up time was 519 days (range, 177‐2492 days).Medical dissolution and decompression of upper urinary tract struvite uroliths should be considered a minimally invasive treatment for dogs before more invasive options. Due to a combination of increased urbanization, industrialization, and population growth, many pharmaceutical pollutants are currently being discharged into the environment. A possible strategy is critical for eliminating antibiotic pollutants from the environment, and photocatalysis has been generally recognized as an excellent method for successfully degrading antibiotics at a faster pace. In this work, we employed a hydrothermal synthesis approach to create a novel C3N5/NiCo2S4 Z-scheme-based heterojunction with better interfacial charge transfer and used it as a catalyst for the degradation of norfloxacin antibiotic. The optimized 1:1 C3N5/NiCo2S4 (50CN/NCS) shows the highest photocatalytic efficiency of 86.5% in 120 min towards the degradation of norfloxacin (NOR). Such an effective performance can be attributed to the high responsive nature of photocatalyst in the visible region and superior transfer of interfacial charges via Z-scheme transfer in heterojunction. The high charge transfer efficiency and reduced recombination of charge carriers in heterojunction was confirmed by EIS and PL results. The influence of some key factors such as pollutant concentration, catalyst dosage, pH, and coexisting ions on the photocatalytic activity is also investigated in this work. Low oral bioavailability is a common feature in most drugs, including antibiotics, due to low solubility in physiological media and inadequate cell permeability, which may limit their efficacy or restrict their administration in a clinical setting. Cefuroxime is usually administered in its prodrug form, cefuroxime axetil. However, its preparation requires further reaction steps and additional metabolic pathways to be converted into its active form. The combination of Active Pharmaceutical Ingredients (APIs) with biocompatible organic molecules as salts is a viable and documented method to improve the solubility and permeability of a drug. Herein, the preparations of five organic salts of cefuroxime as an anion with enhanced physicochemical characteristics have been reported. These were prepared via buffer-assisted neutralization methodology with pyridinium and imidazolium cations in quantitative yields and presented as solids at room temperature. Cell viability studies on 3T3 cells showed that only the cefuroxime salts combined with longer alkyl chain cations possess higher cytotoxicity than the original drug, and while most salts lost in vitro antibacterial activity against E. coli, P. aeruginosa and B. subtilis, one compound, [PyC10Py][CFX]2, retained the activity. Cefuroxime organic salts have a water solubility 8-to-200-times greater than the original drug at 37 °C. The most soluble compounds have a very low octanol-water partition, similar to cefuroxime, while more lipophilic salts partition predominantly to the organic phase. [ABSTRACT FROM AUTHOR]

Objectives: This study aimed to investigate the in vitro and in vivo antibacterial and cytoprotective activities of marine fungal tripeptide derivatives with cinnamic acid moiety asterripeptides A–C (1–3). Methods: The antimicrobial and antibiofilm activities of asterripeptides A–C were tested using the Staphylococcus aureus ATCC 21027 strain. Human HaCaT keratinocytes infected with S. aureus were used for the in vitro investigation of the various aspects of the influence of asterripeptides A–C by lumino- and fluorospectrometry, ELISA, flow cytometry, Western blotting, and microscopy techniques. In the in vivo experiments, mice with burns and scalped S. aureus-infected wounds were used according to ethical committee resolution. Results: Asterripeptides A–C (10 µM) inhibited S. aureus growth and biofilm formation. Asterripeptides A–C increased the viability, proliferation, and migration of S. aureus-infected HaCaT cells and reduced the release of reactive oxygen species (ROS), NO, TNF-α, and IL-18. Asterripeptides A–C protected HaCaT cells against TNF-α-induced inflammation, decreased the transcriptional level of NF-κB in JB6 Cl41 cells, and increased the protein levels of Nrf2 and glutathione synthetase in HaCaT cells. More active asterripeptide C was tested in in vivo burn wounds and S. aureus-infected incised wounds. Asterripeptide C significantly enhanced wound healing, normalized cytokine levels and profiles of peripheral blood samples, and decreased S. aureus contamination of wounds and blood in mice with infected incised wounds. Conclusions: Taken together, these results confirm the dual antibacterial and Nrf2-dependent anti-inflammatory activities of asterripeptides A-C in in vitro and in vivo assays. The aim of conducting a systematic review of the available literature on antibiotic self-medication was to obtain comprehensive data on prevalence, reasons, common antibiotics, self-treated diseases, and drug and information sources of antibiotics used for self-medication worldwide. PubMed, ScienceDirect, SpringerLink, and Taylor & Francis Online were used to perform a comprehensive search on articles published between 2011 and 2020 in the English language. Studies focusing on the prevalence of antibiotic self-medication and its associated factors were included in this review. AXIS Appraisal tool for Cross-sectional studies (AXIS tool) was used for quality assessment of the included articles. A total of 76 studies from diverse geographical locations, including 62668 individuals, were included in this systematic review. The prevalence of SMA in the included studies ranges from 2.6% to 86.5%. Major reasons for non-prescribed use of antibiotics include healthcare costs and time constraints. Respiratory tract infections, fever, and prophylaxis of various symptoms appeared to be prominent self-treated health issues. Pharmacy or superstore, leftovers, and relatives or friends were the most common sources of acquiring antibiotics for self-medication. Similarly, the majority tend to self-medicate with antibiotics based on information obtained from a pharmacy, previous prescription, or personal experience. Self-medication with antibiotics was considered a pervasive issue, with penicillin being the most commonly self-medicated antibiotic. Lack of public awareness and little attention of researchers to not only self-medication but also other related issues such as antibiotic resistance, necessitates the immediate development and execution of public health policies and multifaceted interventions to ensure rational use of antibiotics eventually reducing antibiotic resistance. Photocatalysis is an effective technique to remove antibiotic residues from aquatic environments. Typical metal sulfides like Zn3In2S6 have been applied to a wide range of photocatalytic applications. However, there are currently no readily accessible methods to increase its antibiotic‐degrading activity. Here, a facile hydrothermal approach is developed for the preparation of flower‐like Zn3In2S6 with tunable sulfur lattice defects. Photogenerated carriers can be separated and transferred more easily when there is an adequate amount of lattice defects. Moreover, lattice defect‐induced electronic modulation enhances light utilization and adsorption properties. The modified Zn3In2S6 demonstrates outstanding photocatalytic degradation activity for levofloxacin, ofloxacin, and tetracycline. Bacterial keratitis (BK) is an infection that causes inflammation of the cornea and, if severe, can result in blindness. Topical fluoroquinolones combined with corticosteroids have been shown to be useful in the treatment of BK. A rapid, selective, and sensitive bioanalytical method for simultaneous quantification of Gatifloxacin (GAT) and Dexamethasone (DEX) has been developed and validated using tandem mass spectrometry (LC–MS/MS). Optimal separation was accomplished in under 5 min using an Agilent Zorbax C18 column (100 mm × 4.6 mm, 3.5 μm). The mobile phase was composed of a blend of 0.2% formic acid in triple distilled water and methanol with a flow rate of 0.65 mL/min in isocratic mode. GAT and DEX were detected in positive electrospray ionization multiple reaction monitoring mode (MRM), and the retention time was found to be at 1.64 and 2.93 min, respectively. The linearity of GAT and DEX was found to be in the range of 1.56–400 ng mL−1 with good precision and accuracy. The method was validated according to USFDA regulatory guidelines. The validated method was effectively utilized for preclinical pharmacokinetic analysis of GAT and DEX in rabbit tear fluid following the topical application of a commercial formulation. Among the natural tetramic acids with a decalinoyl part, signermycin B is unique because it contains a cis‐decalin. In this paper, we demonstrate that the cis‐decalin section of signermycin B can be accessed by an anionic oxy‐Cope rearrangement. The substrate, a tricyclic dienol was prepared by an intramolecular Diels‐Alder reaction of a masked ortho‐benzoquinone, generated by oxidation of an α‐methoxyphenol in presence of cis‐2‐hexenol. After a superfluous bromine on the cycloadduct was removed, reaction of the tricyclic ketone with isopropenylmagnesium bromide led to the tricyclic trienol that underwent the oxy‐Cope rearrangement to a cis‐decalinone. Research concerning coordination polymers has been intense due to their significant variability and structural stability. With this in mind, an ionic neodymium coordination polymer was synthesized, composed of an anionic one‐dimensional polymer interconnected to a cationic three‐dimensional porous polymer, poly[dodecaaquabis(μ‐pyridine‐4‐carbohydrazide‐κ2N:O)bis(μ2‐4‐sulfobenzoato‐κ2O:O′)bis(μ3‐4‐sulfobenzoato‐κ3O:O′:O′′)trineodymium(III)] catena‐poly[aquabis(μ‐pyridine‐4‐carbohydrazide‐κ2N:O)bis(μ2‐4‐sulfobenzoato‐κ2O:O′)neodymium(III)] 4.33‐hydrate, {[Nd3(C7H4O5S)4(C6H7N3O)2(H2O)12][Nd(C7H4O5S)2(C6H7N3O)2(H2O)]·4.33H2O}n. The ligands used were 4‐sulfobenzoate (PSB) and pyridine‐4‐carbohydrazide, popularly known as isoniazid (INH), an antibiotic drug. The compound crystallizes in the monoclinic space group C2/c, with Z = 4. Solid‐state calculations suggest that the crystal structure is mainly stabilized by hydrogen bonds, i.e. O—H...O and N—H...O interactions among the polymers, and by van der Waals interactions involving the organic side chains. Objectives: This study aimed to determine the feasibility of capturing antimicrobial usage data from prisons for inclusion in the Antimicrobial Use and Resistance in Australia (AURA) surveillance system and to analyse 2021 and 2022 South Australian (SA) usage data for notable trends. Methods: Monthly antimicrobial supply data for eight SA prisons were collected. Antimicrobial volume was converted into the World Health Organization metric, defined daily doses (DDD). Usage rates were calculated relative to prison occupied bed days (OBD). Results: Annual usage of systemic antimicrobials across eight SA prisons totalled 26,448 DDD and 23,526 DDD in 2021 and 2022 respectively. Antibacterials accounted for 80.6% of all antimicrobials dispensed during the study period. The average antibacterial usage rate in female prisons was higher on average than in male prisons. The state-wide systemic antibacterial usage rate in SA prisons declined by 11.3% from 23.8 DDDs/1000 OBD in 2021 to 21.1 DDDs/1000 OBD. Doxycycline, amoxicillin, flucloxacillin, amoxicillin-clavulanic acid, and cefalexin accounted for 72% of the total systemic antibacterial usage rate. Variation in the oral and topical antifungal agents used and the rate of use was observed between prisons. Conclusions: This SA pilot study demonstrates the feasibility of including prisons in routine national antimicrobial surveillance using similar methodology to hospital surveillance. The contributing facilities comprised 6.1% of all Australian prison beds, and extrapolation of the results suggests that the identified gap in surveillance may equate to over 400,000 DDD per annum in prisons nationwide, equating to approximately 5% of hospital inpatient antimicrobial usage. Objective: Medical students are demanders and future suppliers of antibiotic use. Understanding their knowledge, attitude and practice is important for appropriately using antibiotics and controlling antibiotic resistance. The objective of this study was to assess the measurement properties of existed instruments and summarize measurement items and results. Methods: Five English and Chinese databases were searched to comprehensively identify related studies between January 2000 and May 2023. Included instruments were assessed using consensus‐based standards for the selection of health measurement instruments checklist. Descriptive tables and narrative texts were applied to summary the data. Results: Of 3524 studies identified, 25 were finally included, from which 22 measurement instruments were included. Of these, 14 instruments were found with adequate content validity, only one study reported structural validation process and two studies reported reliability test results. Similar items were divided into different dimensions in different studies. Gaps and misconceptions in knowledge and attitude were indicated in antibiotic use principles, including antibiotic use indications and selection. Conclusions: Limitations existed in the current measurement instrument including lacking validation, inconsistent classification of item and lacking item regarding the perspective of suppliers. Heavy metal ions and antibiotic contamination have become a major environmental concern worldwide. The development of efficient recognition strategies of these pollutants at ultra‐low concentrations in aqueous solutions as well as the elucidation of the intrinsic sensing mechanism are challenging tasks. In this work, unique luminescent Ln‐MOF materials (NIIC‐3‐Ln) were assembled by rational ligand design. Among them, NIIC‐3‐Tb demonstrated highly selective luminescence quenching response toward Hg2+ and sulfadiazine (SDI) at subnanomolar concentrations in less than 7 s. In addition, a Hg2+ sensing mechanism through chelation was proposed on the basis of single‐crystal X‐ray diffraction analysis and Hg2+ adsorption study. The interaction mechanism of NIIC‐3‐Tb with SDI was revealed using a newly developed approach involving a (TD‐)DFT based quantification of the charge transfer of a MOF‐analyte supramolecular complex model in the ground and excited states. Effect of ultrasonic treatment on the surface morphology important for MOF sensing performance was revealed by gas adsorption experiments. The presented results indicate that NIIC‐3‐Ln is not only an advanced sensing material for the efficient detection of Hg2+ and SDI at ultra‐low concentrations, but also opens up a new approach to study the sensing mechanism at the molecular level at ultra‐low concentrations. Our objectives were to compare the efficacy of ketoprofen or ceftiofur for treatment of metritis in dairy cows considering subsequent health, production, and reproduction. Cows from 2 commercial dairy farms in Ontario, Canada were examined with a Metricheck device 3 times per week from 2 to 14 DIM. Cows with metritis (fetid vaginal discharge; n = 193) were blocked by parity and fever (rectal temperature ≥39.5°C or <39.5°C) and within each block per farm, randomly assigned to receive 3 mg/kg BW of ketoprofen (KET) or 2.2 mg/kg of ceftiofur hydrochloride (CEF), once a day for 3 d. Day of enrollment was considered study d 0. Rectal temperature and attitude were evaluated in cows with metritis on study d 0, 3, 4, 7, 10, and 13, and vaginal discharge was evaluated on study d 4, 7, 10, and 13. Body condition was scored at enrollment and at 35 DIM, and serum concentration of haptoglobin was measured at d 0, 2, 4, and 7. Cows with rectal temperature ≥39.5°C or a depressed attitude on d 3 were classified as clinical failure and received treatment with ceftiofur for 3 d (KET), or 2 additional days (CEF), to a maximum of 5 d of treatment with ceftiofur. At 35 ± 3 DIM cows were examined for uterine involution via transrectal palpation, purulent vaginal discharge (PVD) via Metricheck, and endometritis via endometrial cytology. Time to onset of cyclicity was assessed by serum progesterone (P4) measurements at 28, 42, and 56 DIM. Contemporary cows from the same farms without metritis (NOMET; n = 1,043) were used for comparison. Data were analyzed with mixed linear or logistic regression or Cox's proportional hazard models, including herd as a random effect. The proportion of clinical resolution of metritis on d 3 (96% vs. 92%), of cows with fever (from d 3 to d 13 after enrollment) or fetid discharge (from d 4 to d 13 after enrollment), and the number of medical treatments (3.1 vs. 3.3) were not different between CEF and KET, respectively. Cows in KET received fewer antibiotic treatments than cows in CEF (0.3 vs. 3.1). Uterine involution, prevalence of PVD (50% vs. 47%) and subclinical endometritis (6.6% vs. 4.3%), and proportion of cyclic cows (82% vs. 86%) did not differ between CEF and KET. Cows in KET had greater serum haptoglobin concentration from d 2 to 7 after enrollment. The incidence of mastitis, lameness, or displaced abomasum to 60 DIM and subclinical ketosis to 21 DIM did not differ among CEF, KET, and NOMET. There were no differences in median time to first AI (CEF = 68 d, 95% CI: 65–70; KET = 69 d, 95% CI: 68–72; NOMET = 69 d, 95% CI: 68–70), median time to pregnancy (CEF = 118 d, 95% CI: 92–145; KET = 113 d, 95% CI: 90–135; NOMET = 105 d, 95% CI: 101–109), pregnancy at first AI at 33 d after insemination (CEF = 42%; KET = 41%; NOMET = 41%), pregnancy loss after first AI (CEF = 8%; KET = 11%; NOMET = 8%), hazard of pregnancy, or hazard of culling up to 300 DIM. Milk yield was not different between CEF and KET during the first 10 wk, but was lesser in KET at wk 2 and 4 and CEF at wk 2, 4, and 6 than in NOMET. In this pilot-scale study, given early detection, we did not detect differences in subsequent health, milk yield, or reproductive performance in cows with metritis initially treated for 3 d with CEF or KET. Patients are sometimes mislabeled as having an immune-mediated antibiotic allergy in their medical records. Therefore, the aim of this study was to investigate the prevalence of subjects with non-immune mediated reactions to antibiotics using a standardized questionnaire. Subjects aged 18 years and older with a documented antibiotic allergy were identified and recruited from 2 outpatient clinics in the greater Chicago area. Subjects completed a standardized questionnaire during a single visit regarding their previous adverse reaction to an antibiotic. For subjects with multiple documented antibiotic allergies, 1 questionnaire was filled out for each antibiotic allergy. Investigators subsequently evaluated the questionnaire responses to determine whether the adverse reaction was a true immune-mediated allergic reaction or an adverse drug reaction. A total of 98 subjects were recruited with completion of 159 questionnaires. Eighteen subjects (18.37%, 95% CI: 10.7%, 26.3%) had antibiotic allergy labels with no corresponding immune-mediated reaction history. There were 35 allergy labels (22.0%, 95% CI: 14.7%, 29.4%) that were unlikely to be immune-mediated. Antibiotics with the highest percentage of clinical histories that were unlikely to be immune-mediated were macrolides (8 of 11 subjects), nitrofurantoin (1 of 2 subjects), and amoxicillin/clavulanate (2 of 8 subjects). The most common antibiotic allergy labels were penicillin (43 of 159 subjects), sulfonamides (25 of 159 subjects), and fluoroquinolones (21 of 159 subjects). Identification of adverse reactions to antibiotics that are unlikely to be immune-mediated can be accomplished using a standardized questionnaire in the outpatient setting. Florfenicol (FF) is a new antibiotic commonly used in sericulture. This study aimed to examine the effects of low (LC: 0.06 g/L), medium (MC: 0.12 g/L), and high (HC: 1.2 g/L) concentrations of FF on the midgut physiological functions of the silkworm, Bombyx mori (L.) (Lepidoptera: Bombycidae). The results showed that the body weight and the whole cocoon weight of silkworms decreased and the development duration of the fifth instar was prolonged in the HC group. The structure of intestinal microbiota of silkworm larvae was changed by high FF exposure. Specifically, the abundance of conditional pathogens (Curtobacterium, Sanguibater, etc.) was significantly increased, whereas the abundance of Pseudomonas and Pedobacter was decreased significantly. Additionally, the intestinal reactive oxygen species level at 72 h was significantly elevated, and the muscle layer had become loose, together with the appearance of gut goblet cell atrophy. We observed that the increase in Lactobacillus abundance led to a reduction in intestinal fluid pH, resulting in decreased α‐amylase and protease activities, whereas lipase activity exhibited an elevation. The results demonstrated that the accumulation of peroxides induced by exposure to 1.2 g/L FF caused intestinal damage, and the decrease in pH resulting from alterations in microbial composition and structure affected digestive enzyme activity, collectively leading to physiological impairment of the silkworm midgut. Transrectal prostate biopsy (TRPB) is a common procedure used to obtain a prostate biopsy. Although generally safe, complications may occur including infection. Preprocedural antimicrobial prophylaxis is recommended to minimize risk of subsequent infection. This study is a retrospective chart review via the computerized patient record system from January 1, 2018 to February 28, 2022. The study included patients who underwent a TRPB at the Western New York, Syracuse, or Albany Stratton Veterans Affairs Healthcare Systems. This study included a total of 932 patients who underwent TRPB. Postoperative infection occurred in 3.2% (n = 30) of patients within 14 days of the TRPB. Of the 30 patients who developed an infection, 30% (n = 9) resulted in bacteremia. For the 932 patients evaluated, 24 different antibiotic regimens were used, none of which followed guideline recommendations. None of the regimens were found to have an impact on rates of subsequent infection. The results of this study suggest a need for guideline adherence. There was no benefit to using the guideline-discordant regimens as they were not associated with a decreased risk of infection, and in many cases exposed patients to unnecessarily broad and prolonged antibiotic regimens. • Antibiotics are overutilized in prophylaxis for transrectal prostate biopsies. • Antibiotic regimens vary greatly and are discordant with AUA recommendations. • Antibiotics were broader and of prolonged duration compared with current guidelines. • There was no difference in rates of infection with antibiotic duration > 1 day. Background: Antibiotics are frequently prescribed to critically ill patients admitted to intensive care units (ICUs). About 30-60% of antibiotics prescribed in ICUs are unnecessary, inappropriate, or suboptimal and contribute substantially to the development of antimicrobial resistance. Aims and Objectives: The present study was undertaken to assess the use of antibiotics based on the World Health Organization (WHO) access, watch, and reserve (AWaRe) index in a multidisciplinary ICU of a medical college and hospital in Eastern India. Materials and Methods: The descriptive past record-based study was carried out among patients admitted in the multidisciplinary ICU of a medical college and hospital over 6 months. Information from the ICU bed head tickets (BHTs) was captured in a data record form and analyzed. Results: A total of 97 BHTs were reviewed, and 257 antibiotics were prescribed to 90 patients. The prevalence of antibiotic use was 92.78%. Average number of antibiotics per patient was 2.85. In most instances (88.89%, 80), either two (28.88%), three (27.77%), or even four (20%) antibiotics were used. Frequency of use of AWaRe antibiotics was 21.43% (51/238), 67.23% (160/238), and 11.34% (27/238), respectively. Meropenem (15.12%, 36), piperacillin-tazobactam (36, 15.12%), and ceftriaxone (35, 14.70%) were the three most frequently prescribed watch group antibiotics. Most antibiotics were used empirically (60%), and 40% (36/90) use was targeted. Conclusion: Empirical treatment with antibiotics is common in ICUs. The WHO AWaRe tool may check the injudicious and unnecessary use of antibiotics and thus promotes the rational use of antibiotics and prevent the development of antibiotic resistance in ICUs. Ngari is a non-salted fermented fish of Manipur and was made by the indigenous people of Manipur North east India. Traditional Knowledge (TK) possessed by different communities is often closely linked to their way of life. The present study tried to document the control of microbial contaminants isolated from Ngari (A fermented fish product) by natural plants products. Two fungi were isolated from the "Ngari" stored at room temperature (150C-200C) for 45 day. Antimicrobial activity of plant extract was done by agar well diffusion method. Of 8 plants extracts, cinnamon ethanolic extract showed antifungal activity with zone of inhibition 55mm and 15mm against green coloured and black coloured fungal isolates, respectively. Mango ethanolic extract showed antifungal activity against black coloured fungal isolate with zone of inhibition 10mm. Mulberry, neen, guava, curry and stem of ginger and termaric extracts did not show any activity against the both fungal isolates. Antibiotic susceptibility pattern of microbial isolates was done by agar disc diffusion method. Ketaconazole showed the best activity against the both fungal i.e (green and black) with zone of inhibition (18mm) followed by Clotrimazole (CC) (12 and 20mm) and Itraconazole (IT) (12 and 17mm), respectively. Nystatin (NS) showed the activity only against the black fungal 13mm. Both fungal isolates were resistant to Amphotericin and Fluconazole. The Cinnamon ethanolic extract exhibited the MIC of 10% w/v against both fungal isolates. The photochemical analysis of cinnamon ethanolic extract was done and showed the presence of alkaloid, tannins and phenols and absence of flavonoid and saponins. It may be suggested from the findings that cinnamon ethanolic extract can be used as a potential source of natural antimicrobial compound to be used as food preservative. Further research is needed for the to identify fungal isolates from Ngari and also to isolate, purify and identify the active antifungal from the cinnamon ethanolic extract. [ABSTRACT FROM AUTHOR]

Background: The human gut microbiota is an extensive population of microorganisms, and it shows significant variations between periods of optimal health and periods of illness. Vancomycin-resistant Enterococcus (VRE) and carbapenem-resistant Klebsiella pneumoniae (CRKP) are both pathogenic agents (BPAs) that can colonize in the gut after dysbiosis of microbiotal composition following antibiotic treatment. Methods: This study aimed to investigate the impact of antibiotics on the microbiotal composition of the gut. For this purpose, the first pass meconiums of 20 patients and the first rectal swabs containing BPAs of the same patients after antibiotic treatment were studied using next-generation sequencing-based 16S rRNA gene analysis. The V1–V9 region of 16S rRNA was sequenced with Oxford Nanopore. Results: Twenty-five phyla were detected in the meconiums, and 12 of them were absent after antibiotic treatment. The four most prevalent phyla in meconiums were Bacillota, Pseudomonadota, Bacteroidota, and Actinomycetota. Only the relative abundance of Pseudomonadota was increased, while a significant decrease was observed in the other three phyla (p < 0.05). A significant decrease was observed in alpha-diversity in rectal swabs containing BPAs versus meconiums (p = 0.00408), whereas an increased variance was observed in beta-diversity in all samples (p < 0.05). As a result of a LEfSe analysis, Pseudomonadota was found to have a higher relative abundance in rectal swabs, and Bacillota was significantly higher in the meconiums of the twins. Conclusions: Our study strongly verified the relationship between the administration of antibiotics, dysbiosis, and colonization of BPAs in the infants' gut microbiota. The current study aimed to determine the molecular mechanisms of carbapenem and colistin resistance among the clinical isolates of Klebsiella pneumoniae from hospitalized patients admitted to a university hospital in Eastern Algeria. In total, 124 non-duplicate isolates of K. pneumoniae were collected from September 2018 to April 2019. Bacterial identification was performed using MALDI-TOF MS. The presence of extended spectrum β-lactamase (ESBL) genes, carbapenemase genes, chromosomal mutation and mcr genes in colistin-resistant K. pneumoniae were evaluated by PCR. ESBLs represented a rate of 49.1% and harbored blaCTX-M, blaTEM and blaSHV genes. Concerning carbapenems, 12 strains (9.6%) were resistant to ertapenem (MIC: 1–32 μg/mL), of which one strain (0.8%) was also resistant to imipenem (MIC: 32 μg/mL). Among these strains, nine (75%) harbored blaOXA-48 gene. Seven strains (5.6%) expressed resistance to colistin (MIC: 2–32 μg/mL), of which two harbored mcr-8 and mgrB genes simultaneously. The existence of a double resistance to colistin in the same strain is new in Algeria, and this could raise concerns about the increase in levels of resistance to this antibiotic (MIC: 32 μg/mL). The mgrB gene alone was observed in five isolates (71.4%), including two strains harboring blaOXA-48. This is the first report revealing the presence of K. pneumoniae strains carrying the blaOXA-48 gene as well as a mutation in the mgrB gene. This work examined the occurrence characteristics and ecological risks of 31 antibiotics across five classes and seven ARGs in the surface waters of Gaoyou Lake. A total of 27 antibiotics, spanning four classes, were detected in the surface waters of Gaoyou Lake, with an overall concentration ranging from 57.5 to 114 ng/L and an average of 78.8 ng/L. Sulfonamide antibiotics exhibited the highest average concentration at 32.7 ng/L. Spatial analysis revealed that antibiotic concentration levels in the western region of the lake were higher than those in other areas. Similarly, ARGs were most abundant in this area, with sulfonamide ARGs demonstrating a notably higher mean abundance than other ARGs. Correlation analysis revealed strong positive associations between sul1 and several antibiotics, including sulfadimidine, sulfamethoxazole, ciprofloxacin, lincomycin, clindamycin, erythromycin, and intl1 (P < 0.05), with intra-group correlations among sulfonamide ARGs exceeding those between different ARG groups. Ecological risk assessment indicated that erythromycin and sulfamethoxazole presented medium risks, whereas roxithromycin, azithromycin, and lincomycin were associated with low risks to aquatic organisms. The ecological risk proportions across monitoring sites were primarily low (10.6%) and moderate (16.7%), with no high-risk areas identified and 72.7% presenting no risk. Spiders produce webs, which are still a largely unexplored source of antibacterial compounds, although the reports of its application in the medical field. Therefore, this study aims to present an integrative review of the antibacterial activity of spider webs. The research was conducted using Google Scholar, Scielo, Web of Science, PubMed, ScienceDirect, Medline EBSCO, LILACS, and Embase. The inclusion criteria were original articles written in English that studied the antibiotic properties of the web or isolated compounds tested. The studies were compared according to the spider species studied, the type of web, treatment of the sample, type of antimicrobial test, and the results obtained. Nine hundred and seventy-three publications were found, and after applying the inclusion and exclusion criteria, sixteen articles were selected. Bacterial inhibition was found in seven studies against various species of bacteria such as Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Salmonella Typhi, Bacillus megaterium, Listeria monocytogenes, Acinetobacter baumannii, Streptococcus pneumoniae, Pasteurella multocida, and Bacillus subtilis. Additionally, there was no apparent relationship between the proximity of the spider species evaluated in the studies and the presence or absence of activity. Methodological problems detected may affected the reproducibility and reliability of the results in some studies, such as the lack of description of the web or microorganism strain, as well as the absence of adequate controls and treatments to sterilize the sample. Seasonal rhythms are gaining attention given their impact on metabolic disorders development such as obesity gut microbiota is emerging as a key factor in mediating this link. However, the underlying mechanisms are still poorly understood. In this regard, corticosterone may play a role as it has been shown to be affected by gut bacteria and seasonal rhythms, and has been linked to obesity. Thus, this study aimed to investigate if seasonal rhythms effects on corticosterone are influenced by gut microbiota in obese rats and whether this may be related to seasonal and clock genes expression in the pituitary gland and colon. Fischer 344 male rats fed with cafeteria diet (CAF) were housed under different photoperiods for 9 weeks and treated with an antibiotic cocktail (ABX) in drinking water during the last 4 weeks. Rats fed with standard chow and CAF-fed rats without ABX were included as controls. ABX altered gut microbiota, corticosterone levels and seasonal clock expression in the pituitary depending on photoperiod conditions. Treating bone infections with common antibiotics is challenging, since pathogens like Staphylococcus aureus can reside inside macrophages. To target these intracellular bacteria, we have proposed nanoparticles (NPs) as drug carriers. This study aims to investigate the efficacy of hydroxyapatite and gelatin NPs, selected in view of their bone mimicry and potential for targeted delivery, as carriers for the antibacterial agents zinc and vancomycin. Therefore, two distinct NPs are fabricated: zinc-doped hydroxyapatite (ZnHA) and vancomycin-loaded gelatin (VGel) NPs. The NPs are characterized based on morphology, size, chemical composition, cellular internalization, and intracellular bactericidal efficacy. Specifically, the intracellular bactericidal efficacy is tested using a validated coculture model of human THP-1 derived macrophages and phagocytosed S. aureus bacteria. Scanning electron microscopy (SEM) and Fourier transform-infrared spectroscopy (FTIR) results show that the spherical NPs are synthesized successfully. These NPs are internalized by THP-1 cells and show >75% colocalization with lysosomes without compromising the viability of the THP-1 cells. Both ZnHA and VGel NPs substantially reduce the intracellular survival of S. aureus compared to the direct addition of dissolved zinc and vancomycin. Introduction: As a widely used traditional Chinese medicine with hot property, aconite can significantly promote energy metabolism. However, it is unclear whether the gut microbiota and bile acids contribute to the energy metabolismpromoting properties of aconite. The aim of this experiment was to verify whether the energy metabolism-promoting effect of aconite aqueous extract (AA) is related to gut microbiota and bile acid (BA) metabolism. Methods: The effect of AA on energy metabolism in rats was detected based on body weight, body temperature, and adipose tissue by HE staining and immunohistochemistry. In addition, 16S rRNA high-throughput sequencing and targeted metabolomics were used to detect changes in gut microbiota and BA concentrations, respectively. Antibiotic treatment and fecal microbiota transplantation (FMT) were also performed to demonstrate the importance of gut microbiota. Results: Rats given AA experienced an increase in body temperature, a decrease in body weight, and an increase in BAT (brown adipose tissue) activity and browning of WAT (white adipose tissue). Sequencing analysis and targeted metabolomics indicated that AA modulated gut microbiota and BA metabolism. The energy metabolism promotion of AA was found to be mediated by gut microbiota, as demonstrated through antibiotic treatment and FMT. Moreover, the energy metabolism-promoting effect of aconite is associated with the bile acid receptor TGR5 (Takeda G-protein-coupled receptor 5)-UCP1 (uncoupling protein 1) signaling pathway. Many prescribers are not currently following these guidelines. To address suboptimal antimicrobial use, the underlying reasons must be understood.To identify factors influencing choices of antimicrobial drug, dose, and duration for companion animals.Twenty‐two veterinarians treating companion animals in Australia. Diversity of participants was deliberately sought.Semistructured interviews were conducted online. Two case studies were discussed, and then a range of broader questions was posed. Transcripts were analyzed thematically, using an inductive approach.Few participants chose guideline‐concordant management for the case studies. Prescribing choices were influenced by a complex array of factors associated with the clinical case, pet owner, drug, veterinarian, veterinary colleagues, and external factors. Key factors driving broad‐spectrum antimicrobial use included a sense of safety, habit, ease of administering the drug (especially in cats), pharmaceutical marketing, and the self‐perpetuating dispensary cycle. Many participants were concerned about antimicrobial resistance, but insufficiently informed about how to minimize this risk. Several participants believed that longer duration of treatment and ensuring patients finish a predetermined course would decrease the risk of antimicrobial resistance and improve clinical outcomes.Veterinarians are engaged with the concept of antimicrobial stewardship, but face numerous practical barriers and require more information. The Zn/Cr-MOFs/TiO2 composites were synthesized using the solvothermal method. XRD, FTIR, and SEM techniques were utilized to characterize the Zn/Cr-MOFs/TiO2 composites employed for simulating levofloxacin hydrochloride in wastewater. The impact of the mass of the Zn/Cr-MOFs/TiO2 composite, concentration of levofloxacin hydrochloride, solution pH, and temperature on the adsorption performance was investigated. Experimental findings indicated that at pH 6, the maximum removal efficiency of levofloxacin hydrochloride by the Zn/Cr-MOFs/TiO2 composite was achieved at 88.8%, with an adsorption capacity of 246.3 mg/g. To analyze the experimental data, both pseudo-first-order and pseudo-second-order kinetics models were applied, revealing that the pseudo-second-order model provided a better fit to the data. Additionally, Langmuir and Freundlich isotherm models were used to study equilibrium adsorption behavior and showed good agreement with both kinetic modeling and Langmuir isotherm analysis results. These observations suggest that monolayer adsorption predominates during the removal process of levofloxacin hydrochloride by Zn/Cr-MOFs/TiO2 composites. [ABSTRACT FROM AUTHOR]

Simple Summary: About a 4% decline in fish production was recorded in 2020 in comparison to 2018 in inland aquaculture sectors. In 2014, about 10% of aquatic cultured animals were lost (costing global annual losses > USD 10 billion) due to infectious diseases. Contagious diseases generate several stresses, including oxidative stress in fishes that hampers their reproduction, growth, and production value. Aquaculture management needs to employ preventive measures against various contagious diseases for maintenance. Therefore, vaccination in fish, especially in broodstocks, is one of the most important measures to curb such fish losses. Here, we reviewed the current status of fish vaccines and the modern technology used to produce (alternative) fish vaccines, such as plant-derived vaccines for fish health management. Reliable aquaculture vaccines to increase economic productivity in aquaculture sectors are needed. Emerging contagious diseases in different fish species demand novel vaccines to be produced by adapting biotechnological approaches. Vaccination in broodstocks is important, and the fish vaccines are produced with whole-killed pathogens; subunits of a protein, peptide, or a recombinant protein; DNA vaccines; or live attenuated vaccines via different modes of administration. The use of adjuvants in fish vaccines needs to be researched. Vaccination is an essential method of immunological preventive care required for the health management of all animals, including fish. More particularly, immunization is necessary for in-land aquaculture to manage diseases in fish broodstocks and healthy seed production. According to the latest statistics in 2020, 90.3 million tons of capture fishery production was achieved from the aquaculture sector. Out of the above, 78.8 million tons were from marine water aquaculture sectors, and 11.5 million tons were from inland water aquaculture sectors. About a 4% decline in fish production was achieved in 2020 in comparison to 2018 from inland aquaculture sectors. On the other hand, the digestive protein content, healthy fats, and nutritional values of fish products are comparatively more affordable than in other meat sources. In 2014, about 10% of aquatic cultured animals were lost (costing global annual losses > USD 10 billion) due to infectious diseases. Therefore, vaccination in fish, especially in broodstocks, is one of the essential approaches to stop such losses in the aquaculture sector. Fish vaccines consist of whole-killed pathogens, protein subunits, recombinant proteins, DNA, or live-attenuated vaccines. Challenges persist in the adaption of vaccination in the aquaculture sector, the route of administration, the use of effective adjuvants, and, most importantly, the lack of effective results. The use of autogenous vaccines; vaccination via intramuscular, intraperitoneal, or oral routes; and, most importantly, adding vaccines in feed using top dressing methods or as a constituent in fish feed are now emerging. Background Results Conclusion There are few studies about the differences in the composition of moisture, ash, crude protein, crude fat, crude polysaccharide and ergothioneine in Ganoderma lucidum spore powder (GLSP) from different origins. As for GLSP after oil extraction (OE‐GLSP), there are still lots of bioactive substance in it. It can be seen that OE‐GLSP has certain biological activity. The effect of OE‐GLSP on the improvement of intestinal barrier function has been less studied.The results showed that there were significant differences for GLSP from five different origins (Anhui, Jilin, Jiangxi, Shandong and Zhejiang) in moisture (0.065–0.113%), ash (0.603–0.955%), crude fat (42.444–44.773%), crude polysaccharide (2.977–4.127%), crude protein (14.761–17.639%) and ergothioneine (0.552–1.816 mg g−1) (P < 0.05). The monosaccharides of GLSP polysaccharide mainly consist of glucose, galactose, mannose, rhamnose, etc. Moreover, the effects of OE‐GLSP supplementation on the regulation of organ index, colonic tissue and intestinal microbiota in C57BL/6J mice were investigated. The supplement of OE‐GLSP could restore the organ index and weight loss of antibiotic‐treated mice. Moreover, OE‐GLSP led to the improvement of intestinal dysbiosis by enriching Bacteroidetes, Firmicutes, Lactobacillus and Roseburia, and increasing the Firmicutes/Bacteroidetes ratio. In addition, OE‐GLSP intervention repaired intestinal barrier dysfunction by increasing the expression of tight junction proteins (Occludin, Claudin‐1 and E‐cadherin).Different GLSP from five origins exhibited significant differences in microstructure and contents of crude polysaccharide, crude protein, crude fat, water, ash and ergothioneine. The livestock-associated methicillin-resistant Staphylococcus aureus (LA-MRSA) strains are prevalent in the poultry farming environment and are a common component of the bacterial microbiota on the skin and mucous membranes of healthy animals. The origin and spread of LA-MRSA are attributed to the use of antibiotics in animals, and close contact between people and different animal species increases the risk of animal exposure to humans. The epidemiology of LA-MRSA in poultry significantly changed when ST398 and ST9 were found in food-producing animals. The significance of LA-MRSA and zoonotic risk associated with handling and processing foods of avian origin is highlighted by the LA-MRSA strain's ability to infect chickens. People who work with poultry are more prone to contract LA-MRSA than the general population. There is scientific consensus that individuals who have close contact with chickens can become colonized and subsequently infected with LA-MRSA; these individuals could include breeders, medical professionals, or personnel at chicken slaughterhouses. Background and Objectives: The aim of this study was to investigate the effect of sheep lymph nodes and antibiotics on the bacterium that causes acne and the bacteria that usually exist with it. Materials and Methods: In this laboratory study, 24 patients with acne (18 to 45 years old) were selected to isolate the bacteria. Also, 24 clinical isolates of Staphylococcus aureus and Staphylococcus epidermidis were isolated from the wound samples of patients. After culturing the bacteria, their sensitivity to common antibiotics was evaluated. Data were reported as percentage. Results: The results showed that Cutibacterium acnes and Staphylococcus aureus were 100% sensitive to vancomycin. Also, Cutibacterium acnes showed sensitivity in tetracycline and imipenem groups in 95% of cases and in cotrimoxazole and piperacillin/tazobactam groups in 90% of cases. In addition, the highest sensitivity of Staphylococcus epidermis to piperacillin/tazobactam (62.5%) and the highest drug resistance (91.67%) were related to azithromycin and erythromycin groups. Conclusion: Due to the increase in drug resistance, it is suggested to use antibiotics with the least drug resistance such as vancomycin and imipenem in the treatment of acne. [ABSTRACT FROM AUTHOR]
