Effector secretion is crucial for root endophytes to establish and protect their ecological niche. We used time-resolved transcriptomics to monitor effector gene expression dynamics in two closely related Sebacinales, Serendipita indica and Serendipita vermifera, during symbiosis with three plant species, competition with the phytopathogenic fungus Bipolaris sorokiniana, and cooperation with root-associated bacteria. We observed increased effector gene expression in response to biotic interactions, particularly with plants, indicating their importance in host colonization. Some effectors responded to both plants and microbes, suggesting dual roles in intermicrobial competition and plant-microbe interactions. A subset of putative antimicrobial effectors, including a GH18-CBM5 chitinase, was induced exclusively by microbes. Functional analyses of this chitinase revealed its antimicrobial and plant-protective properties. We conclude that dynamic effector gene expression underpins the ability of Sebacinales to thrive in diverse ecological niches with a single fungal chitinase contributing substantially to niche defense.

Although public–private collaboration abounds in addressing societal grand challenges, little is known about its performance relative to private-sector collaboration. Drawing from the innovation collaboration literature, and qualitative insights from interviews and contracts, we argue that collaboration risk impairs task performance of public–private versus private–private collaboration. We attribute this to two collaboration risk mechanisms: cooperation problems arising from misaligned social versus economic incentives, and coordination problems due to differences in organizational governance and processes. We test our hypotheses using the development history of 2,496 antimicrobial drugs from 1995–2019. After accounting for endogenous selection into collaboration, we find that public–private collaboration is less effective than private–private collaboration. However, this performance gap reduces for innovation tasks under high technological uncertainty (i.e., in the discovery stage and for developing new drug action mechanisms). To probe the distinctive cooperation and coordination mechanisms, we also examine innovation task efficiency. Findings reveal a wider gap in task effectiveness but not efficiency between public–private and private–private collaboration, suggesting more significant cooperation than coordination problems. Our theoretical and practical insights pertain to whether, when, and how public–private and private–private collaboration offer viable pathways for socially beneficial innovation tasks. Purpose: To analyze the effectiveness of coating of abutments with antimicrobial agents and their influence on the physicochemical and biologic properties of the coated materials. Materials and Methods: This work was registered in Open Science Framework (osf.io/6tkcp) and followed the PRISMA protocols. A search of two independent reviewers of articles published up to October 29, 2021, was performed in the Embase, PubMed, Science Direct, and Scopus databases. Results: The databases found a total of 1,474 references. After excluding the duplicates, 1,050 remained. After reading the titles and abstracts and applying the inclusion criteria, 13 articles remained and were read in full. A total of 8 articles were included in this systematic review. Different antimicrobial agents have been used to coat abutments, including graphene oxide, polydopamine, titanium and zirconium nitride, lactoferrin, tetracycline, silver, and doxycycline with varied release times. Titanium-coated silver showed a better antimicrobial agent release time of up to 28 days. Chemical analysis confirmed the presence of antimicrobials on the surface after coating. Different pathogenic microorganisms, such as Streptococcus sanguinis, Streptococcus oralis, and Staphylococcus aureus, were inhibited when in contact with the coated surface. Conclusions: This review showed that there is still no consensus on which is the better antimicrobial agent and which coated materials have the better performance. However, the association of surface coating of abutments with antimicrobials is feasible and can benefit many patients, which can support their clinical use to favor the healing process and prevent infections that can lead to treatment failure with dental implants. Purpose: To investigate the possible antimicrobial activity of glycine air polishing by comparing peri-implant microbiota before and after treatment. Materials and Methods: A total of 15 patients who received implant-supported full-arch fixed rehabilitations were included. After prosthesis removal (T0), Plaque Index (PI), probing depth (PD), and bleeding on probing (BOP) were recorded. In each hemiarch, the implant with the highest PD score was selected for microbiologic sample collection from the peri-implant sulcus (T0). All patients received two different hygienic protocols (randomly administered, one per each hemiarch): glycine air-polishing (G) and cleansing with cotton pellets soaked in saline (C). At 7 days (T1) and 3 months (T2) after the intervention, PI and BOP were recorded, and new microbiologic samples were taken. Traditional polymerase chain reaction (PCR) and quantitative PCR real-time were employed for microbiologic analysis to investigate how the presence of different bacterial species varied according to the hygienic treatment performed. Results: Treatment G provided a significantly higher PI score reduction around implants compared to treatment C (P = .015). No statistical difference was found in the microbial population around G and C implant sites, with Tannerella forsythia being the most commonly detected bacterial species in both G and C groups. No statistical differences were found between the antimicrobial activity of treatments C and G. Conclusions: Glycine powder air polishing is a valid method for professional hygienic care of implants and was more effective in PI reduction compared to the control treatment. BackgroundOcular infections caused by antibiotic-resistant pathogens can result in partial or complete vision loss. The development of pan-resistant microbial strains poses a significant challenge for clinicians as there are limited antimicrobial options available. Synthetic peptoids, which are sequence-specific oligo-N-substituted glycines, offer potential as alternative antimicrobial agents to target multidrug-resistant bacteria.MethodsThe antimicrobial activity of synthesised peptoids against multidrug-resistant (MDR) ocular pathogens was evaluated using the microbroth dilution method. Hemolytic propensity was assessed using mammalian erythrocytes. Peptoids were also incubated with proteolytic enzymes, after which their minimum inhibitory activity against bacteria was re-evaluated.ResultsSeveral alkylated and brominated peptoids showed good inhibitory activity against multidrug-resistant Pseudomonas aeruginosa strains at concentrations of ≤15 μg mL-1 (≤12 µM). Similarly, most brominated compounds inhibited the growth of methicillin-resistant Staphylococcus aureus at 1.9 to 15 μg mL-1 (12 µM). The N-terminally alkylated peptoids caused less toxicity to erythrocytes. The peptoid denoted as TM5 had a high therapeutic index, being non-toxic to either erythrocytes or corneal epithelial cells, even at 15 to 22 times its MIC. Additionally, the peptoids were resistant to protease activity.ConclusionsPeptoids studied here demonstrated potent activity against various multidrug-resistant ocular pathogens. Their properties make them promising candidates for controlling vision-related morbidity associated with eye infections by antibiotic-resistant strains.

Host-directed therapy (HDT) is an emerging approach to overcome antimicrobial resistance in pathogenic microorganisms. Specifically, HDT targets host-encoded factors required for pathogen replication and survival without interfering with microbial growth or metabolism, thereby eliminating the risk of resistance development. By applying HDT and a drug repurposing approach, we demonstrate that (R)-DI-87, a clinical-stage anticancer drug and potent inhibitor of mammalian deoxycytidine kinase (dCK), mitigates Staphylococcus aureus abscess formation in organ tissues upon invasive bloodstream infection. Mechanistically, (R)-DI-87 shields phagocytes from staphylococcal death-effector deoxyribonucleosides that target dCK and the mammalian purine salvage pathway-apoptosis axis. In this manner, (R)-DI-87-mediated protection of immune cells amplifies macrophage infiltration into deep-seated abscesses, a phenomenon coupled with enhanced pathogen control, ameliorated immunopathology, and reduced disease severity. Thus, pharmaceutical blockade of dCK represents an advanced anti-infective intervention strategy against which staphylococci cannot develop resistance and may help to fight fatal infectious diseases in hospitalized patients.

The growing concerns regarding foodborne illnesses related to fresh produce accentuate the necessity for innovative material solutions, particularly on surfaces that come into close contact with foods. This study introduces a sustainable, efficient, and removable antimicrobial and antifouling coating ideally suited for hydrophobic food-contact surfaces such as low-density polyethylene (LDPE). Developed through a crosslinking reaction involving tannic acid, gelatin, and soy protein hydrolysate, these coatings exhibit proper stability in aqueous washing solutions and effectively combat bacterial contamination and prevent biofilm formation. The unique surface architecture promotes the formation of halamine structures, enhancing antimicrobial efficacy with a rapid contact killing effect and reducing microbial contamination by up to 5 log10 cfu·cm-2 against both Escherichia coli (Gram-negative) and Listeria innocua (Gram-positive). Notably, the coatings are designed for at least five recharging cycles under mild conditions (pH6, 20 ppm free active chlorine) and can be easily removed with hot water or steam to refresh the depositions. This removal process not only conveniently aligns with existing sanitation protocols in the fresh produce industry but also facilitates the complete eradication of potential developed biofilms, outperforming uncoated LDPE coupons. Overall, these coatings represent sustainable, cost-effective, and practical advancements in food safety and are promising candidates for widespread adoption in food processing environments.

The transmission of antimicrobial resistant bacteria in the urban environment is poorly understood. We utilized genomic sequencing and phylogenetics to characterize the transmission dynamics of antimicrobial resistant Escherichia coli (AMR-Ec) cultured from putative canine (caninep) and human feces present on urban sidewalks in San Francisco, California. We isolated a total of fifty-six AMR-Ec isolates from human (n = 20) and caninep (n = 36) fecal samples from the Tenderloin and South of Market (SoMa) neighborhoods of San Francisco. We then analyzed phenotypic and genotypic antimicrobial resistance (AMR) of the isolates, as well as clonal relationships based on cgMLST and single nucleotide polymorphisms (SNPs) of the core genomes. Using Bayesian inference, we reconstructed the transmission dynamics between humans and caninesp from multiple local outbreak clusters using the marginal structured coalescent approximation (MASCOT). Our results provide evidence for multiple sharing events of AMR-Ec between humans and caninesp. In particular, we found one instance of likely transmission from caninesp to humans as well as an additional local outbreak cluster consisting of one caninep and one human sample. Based on this analysis, it appears that non-human feces act as an important reservoir of clinically relevant AMR-Ec within the urban environment for this study population. This work showcases the utility of genomic epidemiology to reconstruct potential pathways by which antimicrobial resistance spreads.

Purpose: To verify the parameters and characteristics evaluated in 3D-printed complete denture (CD) bases and how they influence CD properties. Materials and Methods: This work was registered in Open Science Framework (osf.io/4um6v) and followed the PRISMA Extension for Scoping Reviews. A search of peer-reviewed articles published up to April 9, 2020, was performed on the PubMed, LILACS, Cochrane Library, and Science Direct databases. The review question based on the PCC (population, concept, and context) was: What printing parameters and characteristics of CD bases can influence their properties? Results: The database search resulted in 1,945 articles, and the initial screening was carried out in 1,390 articles, 17 of which were selected for inclusion in this review. The present authors verified that cleaning the prosthesis with alcohol after printing followed by a postcuring cycle influences biocompatibility and residual monomers. The printing angle influences physicomechanical properties, microbial adhesion, and tissue adaptation. Accuracy is influenced by layer thickness and manufacturing technique, which also influences retention and tissue adaptation. The incorporation of antimicrobial agents influences physicomechanical properties and antimicrobial activity. The method of union between the denture base and teeth influences mechanical strength. Conclusions: Printed denture bases showed good adaptation to tissues and accuracy with 100-µm layer thickness, but the base must be cleaned in ethanol followed by postcuring for better biocompatibility. Purpose: To evaluate the applica.on of chitosan as a cleanser in the control of biofilm forma.on on cobalt-chromium (Co-Cr) alloy and acrylic resin surfaces. Materials and Methods: In total, 172 Co-Cr discs and 172 acrylic resin discs (14 mm x 3 mm) were contaminated with Streptococcus mutans, Staphylococcus aureus, Candida albicans, or Candida glabrata and incubated for 48 hours. Then, specimens were randomly divided into groups and immersed in the following solu.ons for 15 minutes: solu.on without chitosan (WC/control); chitosan solu.on (CH: 5 mg/mL); chitosan nanopar.cle solu.on (CN: 3.8 mg/mL); and effervescent tablet (ET). Biofilm recovery rates (n = 9) were evaluated by coun.ng the colony-forming units (CFU/mL). Biofilm morphology was evaluated using scanning electron microscopy (SEM). Data were compared using the Kruskal-Wallis or ANOVA tests followed by the Tukey post hoc test. Results: For acrylic resin, ET showed the lowest number of CFU for S aureus and S mutans (P < .001). CH exhibited intermediate values for S mutans, S aureus, and C albicans; CN exhibited intermediate values for S mutans and S aureus. For C glabrata, there was no sta.s.cal difference between the solu.ons (P = .264). For Co-Cr, ET showed the highest level of an.microbial ac.on against all microorganisms (P < .001), and CH showed an intermediate level of ac.on against S mutans and S aureus. Against C albicans and C glabrata, there was no significant difference among CH, CN, and WC. This study aimed to investigate how the presence of co-morbid conditions influenced antimicrobial usage as presumptive prophylaxis for suspected bacteremia in dogs and cats undergoing dental treatments at primary care veterinary clinics in the United States. In 2020, data was collected from 1076 veterinary clinics across 44 US states. A total of 681,541 general anesthesia dental procedures were conducted on 592,472 dogs and 89,069 cats. This revealed that systemic antimicrobials were administered in 8.8% of dog procedures and 7.8% of cat procedures in the absence of concurrent periodontal disease or extractions. Cefpodoxime, clindamycin, and amoxicillin-clavulanate were the most frequently used antimicrobials in dogs, while cefovecin, amoxicillin-clavulanate, and clindamycin topped the list for cats. Dogs with cardiovascular, hepato-renal, and endocrine co-morbidities, as well as those undergoing concurrent removal of cutaneous or subcutaneous neoplasia, displayed higher antimicrobial use. Similarly, cats with endocrine or hepato-renal disease, retroviral infection (i.e., feline leukemia virus (FeLV), feline immunodeficiency virus (FIV)), and concurrent removal of cutaneous or subcutaneous neoplasia exhibited increased antimicrobial use. Dogs with hepato-renal abnormalities had longer treatment durations compared to those without (10.1 vs. 9.6 days). Conversely, cats with concurrent removal of cutaneous or subcutaneous neoplasia had shorter durations of treatment as compared to those that did not have this procedure performed (8.4 vs 9.2 days). The findings of this study underscore the necessity for further research and collaboration within the veterinary community to develop evidence-based guidelines, promoting responsible antimicrobial use, and advancing the field of veterinary dentistry for enhanced patient outcomes.

The objective of this study was to evaluate the impact of dietary zinc supplementation in pre-weaned dairy calves on the phenotypic antimicrobial resistance (AMR) of fecal commensal bacteria. A repository of fecal specimens from a random sample of calves block-randomized into placebo (n = 39) and zinc sulfate (n = 28) groups collected over a zinc supplementation clinical trial at the onset of calf diarrhea, calf diarrheal cure, and the last day of 14 cumulative days of zinc or placebo treatment were analyzed. Antimicrobial susceptibility testing was conducted for Enterococcus spp. (n = 167) and E. coli (n = 44), with one representative isolate of each commensal bacteria tested per sample. Parametric survival interval regression models were constructed to evaluate the association between zinc treatment and phenotypic AMR, with exponentiated accelerated failure time (AFT) coefficients adapted for MIC instead of time representing the degree of change in AMR (MIC Ratio, MR). Findings from our study indicated that zinc supplementation did not significantly alter the MIC in Enterococcus spp. for 13 drugs: gentamicin, vancomycin, ciprofloxacin, erythromycin, penicillin, nitrofurantoin, linezolid, quinupristin/dalfopristin, tylosin tartrate, streptomycin, daptomycin, chloramphenicol, and tigecycline (MR = 0.96-2.94, p > 0.05). In E. coli, zinc supplementation was not associated with resistance to azithromycin (MR = 0.80, p > 0.05) and ceftriaxone (MR = 0.95, p > 0.05). However, a significant reduction in E. coli MIC values was observed for ciprofloxacin (MR = 0.17, 95% CI 0.03-0.97) and nalidixic acid (MR = 0.28, 95% CI 0.15-0.53) for zinc-treated compared to placebo-treated calves. Alongside predictions of MIC values generated from these 17 AFT models, findings from this study corroborate the influence of age and antimicrobial exposure on phenotypic AMR.

Essential oils (EOs) from plants have attracted the attention of scientists due to their multiple biological activities. Antimicrobial activity of essential oils is the main reason for their widespread use in industries, which is determined through minimum inhibitory concentration (MIC) by using in vitro models. Therefore, the MIC is a crucial parameter to evaluate the antimicrobial potential of EOs. 492 papers were screened through the critical review of the related literatures in the past 50 years using four databases and read deeply for an outline. This review first listed common MIC determination methods of EOs, including agar diffusion method, agar dilution method, broth dilution method, E-Test, thin-layer chromatography-bioautography, etc. and their merits and demerits were introduced in detail. Agar and broth dilution methods are the gold standards in a more exact approach for MIC determination. Then, their functions and factors influencing the accuracy were listed and analyzed deeply. The application of the MIC determination in food and pharmaceuticals was finally discussed. Lactic acid bacteria (LAB) are known to exhibit various beneficial roles in fermentation, serving as probiotics, and producing a plethora of valuable compounds including antimicrobial activity such as bacteriocin-like inhibitory substance (BLIS) that can be used as biopreservative to improve food safety and quality. However, the yield of BLIS is often limited, which poses a challenge to be commercially competitive with the current preservation practice. Therefore, the present work aimed to establish an optimised two-plasmid CRISPR/Cas9 system to redirect the carbon flux away from lactate towards compounds with antimicrobial activity by disrupting lactate dehydrogenase gene (ldh) on various strains of LAB. The lactic acid-deficient (ldhΔ) strains caused a metabolic shift resulting in increased inhibitory activity against selected foodborne pathogens up to 78 % than the wild-type (WT) strain. The most significant effect was depicted by Enterococcus faecalis-ldh∆ which displayed prominent bactericidal effects against all foodborne pathogens as compared to the WT that showed no antimicrobial activity. The present work provided a framework model for economically important LAB and other beneficial bacteria to synthesise and increase the yield of valuable food and industrial compounds. Two novel phosphonate-substituted cucurbiturils have been synthesized and characterized. The inherent solubilities of the phosphonate hosts were proved to be higher than those of sulfonate analogues. Benefiting from the interaction between phosphonate and guanidium, the phosphonate hosts exhibited a high affinity towards guests like phenformin, and chlorhexidine. In addition, the phosphonate hosts displayed circular dichroism signals in response to the chiral levofloxacin. Bioceramics composed of calcium and magnesium silicates have garnered increasing attention for the development of porous scaffolds in bone tissue engineering (BTE). This heightened interest is primarily attributed to their remarkable bioactivity and their capacity to form strong bonds with hard tissue. Fabricating nanocomposite scaffolds is a recognized approach for improving the characteristics of scaffolds used in BTE. This research investigates the mechanical and biological properties, antibacterial activity, and drug-release characteristics of scaffolds composed of akermanite (AKT), monticellite (MON), and monticellite-akermanite (MON-AKT). These scaffolds were fabricated utilizing the space holder process. Additionally, the in vitro drug release profile and antimicrobial activity of Azithromycin (AZT)-loaded MON-AKT composite scaffolds were investigated. The findings showed that the Mon-15 wt% AKT nanocomposite scaffold had the highest density, the smallest grain and micropore sizes, and the lowest porosity. In contrast, integrating AKT into MON-based composite scaffolds resulted in materials characterized by high mechanical strength and stability within physiological environments. The MON-AKT nanocomposite scaffolds exhibited cytocompatibility and demonstrated a high level of alkaline phosphatase (ALP) activity in osteogenic studies. Furthermore, antimicrobial activity assessments revealed that the AZT-encapsulated MON-AKT composite scaffolds effectively inhibited the growth of both S. aureus and E. coli bacteria. The outcomes showed that the antibacterial efficacy of the scaffold depends on both the amount of AZT and the type of bacteria. Overall, MON-AKT/3AZT scaffolds exhibited significantly superior bacterial inhibition compared to other scaffolds, making it a promising option for treating bone tissue defects. The existing and emerging demand for materials for life and health has contributed to the cultivation and development of respective markets. Nevertheless, the current generation of biomedical materials has yet to fully satisfy the clinical requirements of the market, which is still in its relative infancy. Research and development in this area must be prioritized in light of the pivotal role of new life and health materials in the biological field. Among many life and health materials, GQDs, an emerging nanomaterial, exhibit considerable promise in the biomedical field, primarily due to their exceptional properties. Furthermore, the direct preparation and functionalization of GQDs have facilitated the development of specific functional composites based on GQDs. The biological applications of GQDs are undergoing rapid growth, which makes it necessary to publish a review article presenting the latest advances in this field. This review provides an overview of the significant advances in synthesizing GQDs, the techniques employed for structural characterizations, and the properties that have been elucidated. Furthermore, it presents recent findings on applying GQDs in antimicrobial, anticancer, biosensing, drug delivery, and bioimaging applications. Two new libraries of diethyl and dimethyl dihydropyridines‐based pyrazole and 1,2,3‐triazole hybrids (7a–g and 7h–n) were synthesized via a series of reactions involving click chemistry, pyrazole synthesis, and multicomponent reaction. All the new compounds were screened for their in vitro anticancer activity and antimicrobial activities. The 4‐chloro substituted analogue (7i) in dimethyl ester series showed an excellent activity against all three, namely, human breast adenocarcinoma (MCF‐7), human cervical cancer (HeLa), and human breast cancer triple‐positive (BT‐474) cell lines with IC50 values of 0.119 ± 0.58 µM, 0.135 ± 0.13 µM, and 0.163 ± 0.31 µM, respectively compared to abemaciclib. Molecular docking studies performed on the best active compound 7i against crystal structure of CDK6 revealed its best binding score and interactions. Additionally, these libraries were screened for their antimicrobial activities and determined their MIC. Compounds with ─Cl (7b), ─OMe (7d), ─Me (7a) substitution, and without substitution (7e) significantly showed better MIC values in comparison to ampicillin and nystatin. [ABSTRACT FROM AUTHOR]

Discovery of novel antimicrobial drugs is one of the hottest topics in the field of medicinal chemistry. An efficient and catalyst free synthesis of novel fluorinated 5‐substituted 1,2,4‐oxadiazole‐3‐yl benzoxazole derivatives is described. A series of novel compounds have been synthesized and characterized by different spectroscopic techniques such as FTIR, 1H NMR, 13C NMR, and LCMS. When tested for antibacterial activity, the synthesized benzoxazole derivatives exhibited outstanding to good activities in comparison to the conventional medications. Moreover, the theoretical forecasts grounded in molecular docking against microbial DNA gyrase may offer a glimpse into the conceivable mode of operation and establish a connection between the detected antimicrobial efficacy and the binding affinity, illuminating particular thermodynamic (bonded and nonbonded) interactions that regulate the activity. The produced compounds also exhibited promising qualities to develop into viable oral medication candidates when their absorption, distribution, metabolism, and excretion were examined. In summary, the research proposes that these novel compounds have emerged as interesting contender for the development of next generation antimicrobial drugs. In this study, we present a green synthesis approach for synthesis of silver nanoparticles (Ag NPs) using Grewia Asiatica L. berries through a phytosynthesis method. In this synthesis procedure the Grewia Asiatica L. berry extract served as a natural reducing agent in the phytosynthesis of GA‐Ag NPs. The synthesized Ag NPs were characterized using various physico‐chemical techniques, revealing their spherical morphology of average size of ∼20 nm. These Ag NPs exhibited significant antibacterial activity against both Gram‐negative and Gram‐positive bacterial strains. Additionally, the GA‐Ag NPs demonstrated anticancer properties, with an IC50 value of approximately 5 µM. The study highlights the potential of Grewia Asiatica L. berry extract as a novel, nontoxic, eco‐friendly, and cost‐effective method for synthesizing silver nanoparticles with promising antibacterial and anticancer activities. ABSTRACT Pomegranate is one of the most popular fruits worldwide, and it is important to maintain the overall quality and bioaccessibility of freshly squeezed pomegranate juices (PJS). The adverse effects of heat treatment on sensory properties and phytochemicals encourage the use of non‐thermal processes in the juice industry. Hereby, the effects of high‐pressure homogenization (HPH) (50, 100, and 150 MPa) on the physicochemical properties, antimicrobial activity, in vitro bioaccessibility, and antioxidant capacity of freshly‐squeezed PJS from different genotypes were investigated. Instrumental color values (L*, a*, and b*) of PJS generally decreased, while pH and aw values increased with HPH processing (p < 0.05). The antimicrobial activity of PJS was not significantly affected by HPH processing (p > 0.05). Sarıcakaya‐1 had higher bioactive constituent content and antioxidant capacity (DPPH‐ARA, ABTS‐ARA, and FRAP) than other genotypes (p < 0.05). Total phenolic content (TPC) and antioxidant capacity of PJS were increased by HPH treatment, whereas total anthocyanin content (TAC) and ascorbic acid content (AAC) were decreased (p < 0.05). Accordingly, HPH treatment enhanced the release of phenolics and antioxidant capacity in PJS, while largely maintaining the presence of heat‐sensitive compounds. Regardless of genotype and HPH treatment, in vitro gastrointestinal digestion adversely affected TPC, TAC, AAC, and antioxidant capacity of all PJS. However, the 150 MPa HPH treatment of Saricakaya‐1 had the highest values for TPC bioaccessibility and recovered ABTS‐ARA after in vitro gastrointestinal digestion (p < 0.05). Considering the overall data, 150 MPa HPH treatment can be recommended as an innovative approach to obtain minimally processed PJS to meet new trends in consumer demand. [ABSTRACT FROM AUTHOR]

Rifampicin (RF) is a potent antibiotic, but its clinical effectiveness is limited by poor water solubility and stability. To overcome these limitations, RF is encapsulated in solid‐lipid nanoparticles (SLNs) using a one‐pot microwave microemulsion method. This study aimed to synthesize and characterize RF‐loaded SLNs, evaluating their physicochemical characteristics and antimicrobial efficacy. The synthesized SLNs exhibit particle sizes ranging from 150 to 200 nm, with a high encapsulation efficiency of ≈55% and high loading capacity. The particles are imaged by cryogenic‐transmission electron microscopy and found to be circular and elongated, with RF homogenously distributed throughout the particles. The RF‐loaded SLNs demonstrate potent antimicrobial activity, as evidenced by disc diffusion assays. The SLNs exhibits a sustained drug release profile, highlighting their potential as an effective drug delivery system for antimicrobial treatment. Recent considerations of natural sources as potential anticancer agents have arisen due to the origins of numerous drugs commonly used in chemotherapy. Plant‐based drugs, in particular, have attracted attention for offering the advantage of low adverse effects. Among these, the black mulberry plant (Morus nigra L.) stands out as a natural source of polyphenols, widely used to treat metabolic dysfunctions and confer benefits on human health. This study explores the potential of this plant as an anticancer agent, examining its effectiveness based on the type of application of the plant extracts or isolated substances, extraction methods, and its potential biological effects on cancer cells. Consequently, this study contributes to a better understanding of the distribution of phytochemicals in M. nigra and their applications in the context of cancer field. Among the compounds found in black mulberry are flavonoids, chlorogenic acid, cryptochlorogenic acid, and protocatechuic acid, along with cyanidin‐3‐O‐glucoside as the main anthocyanin on the fruit. The phytochemicals derived from M. nigra exhibit antinociceptive and antimicrobial activities, while also showing protective effects, such as antioxidant properties that underline their potential as anticancer agents. The black mulberry's roots, stem bark, pulp, and leaves are particularly rich sources of anti‐inflammatory compounds. Ethanol and methanol extraction methods appear to be the most effective in cancer management, offering compounds that facilitate the integration of apoptosis induction, cell growth inhibition, and cytotoxicity modulation. These results collectively represent the salient biological attributes that positioned black mulberry as a promising anticancer agent. Therefore, these findings highlight the multifaceted potential of M. nigra as an anticancer agent, making a compelling case for further research to advance prospects in the medical field. A series of new pyrazolo[1,5‐a]pyrimidine 4–15 derivatives were synthesized starting from 3,5‐diaminopyrazole derivatives 3 a,b as precursor using a simple, efficient procedure, and screened for their in‐vitro antimicrobial activities. All the synthesized compounds were fully characterized by IR, 1HNMR, 13CNMR, MS spectral data, and elemental analysis. Among the investigated against the antimicrobial activity, compounds 4 b, 4 d, 4 e, 8 b, 9 and 12 showed the best activity. Metallo‐β‐lactamases (MBLs), enzymes of class B, employ zinc ions to degrade β‐lactam antibiotics such as penicillins, cephalosporins, carbapenems, and cephamycins. Carbapenem‐resistant Acinetobacter baumannii (CRAB) is linked to the existence of carbapenemase enzymes such as oxacillinase and MBL. The most prevalent resistance mechanisms include imipenemases (IMP), verona integron‐encoded MBL, and New Delhi MBL‐1. The effectiveness of current antibiotics against the MBL enzyme is limited due to the presence of metal ions, underscoring the need for new antimicrobial agents. Recent research has demonstrated that natural compounds can effectively inhibit MBL. This study aims to screen natural phytochemicals against IMP‐2 MBL using in silico virtual screening techniques via AutoDock Vina and molecular dynamic simulations with GROMACS for 200 ns, followed by molecular mechanics/Poisson‒Boltzmann surface area analysis. This procedure identified new lead molecules against A. baumannii that produce IMP. A total of 588 natural compounds were screened against IMP, along with the imipenem substrate and known inhibitors of L‐captopril. The top four compounds, N025‐0038 (NC1), N062‐0008 (NC2), eupalitin, and Rosmorinic acid, demonstrated binding affinities of ‒8.5, ‒8.4, ‒7.5, and ‒7.2 kcal/mol, respectively. The structural stability of these complexes was observed to be maintained throughout the simulation in a dynamic environment, as determined by molecular dynamics trajectory analysis, and all these compounds met the SWISS‐ADME (adsorption, distribution, metabolism, and excretion) properties. NC1 and NC2 compounds are considered potential drug molecules against IMP. However, while these selected compounds showed superior binding energy in computational analysis, further in vitro analysis is required to establish an effective drug regimen against A. baumannii that produces IMP. The endolichenic Fusarium solani (EF5), known to show induced metabolite production when exposed to red and green lights, was selected for characterization of their putative light-regulated bioactive compounds. To achieve this, fractionation was first performed for crude extracts from cultures of F. solani (EF5) incubated in green, red, white-fluorescent light and dark conditions. The extract yielded 12 (dark condition) to 15 (exposed to green, red, and white-fluorescent lights) fractions, and each of the fractions was tested for antimicrobial activities. The fraction (fraction 5) that showed the most promising antimicrobial activity was then subjected to high-performance liquid chromatography (HPLC) and liquid chromatography–mass spectrometry (LC-MS) to identify the bioactive compounds. Results revealed detection of two new metabolites from endolichenic F. solani, putatively identified as 8-deoxyjavanicin and fusolanone A, which are known to have antimicrobial properties. This study revealed that red and green lights trigger the production of 8-deoxyjavanicin and fusolanone A, which likely contributed to the antimicrobial properties demonstrated by endolichenic F. solani. The aim of this systematic review was to assess the antimicrobial effectiveness of silver nanoparticles (AgNPs) incorporated to different orthodontic bonding systems. Additionally, the review investigated the impact of AgNPs on the bonding properties of these materials. The hypothesis posed that the addition of AgNPs would enhance the antimicrobial efficacy of orthodontic bonding systems while maintaining their bonding properties. The systematic review employed a PICO-based search strategy, targeting in vitro studies focusing on the integration of nano silver particles into orthodontic bonding systems with potential antimicrobial activity. The intervention involved the use of nano silver in orthodontic bonding systems, with a comparison to systems lacking nano silver. The primary outcomes assessed were antimicrobial activity and shear bond strength (SBS). The search process, conducted without publication date restrictions, yielded 551 potential articles: 34 from PubMed, 360 from PubMed Central, 42 from Embase, 54 from Scopus, and 61 from Web of Science. Ultimately, a qualitative synthesis was conducted on 13 papers. The PRISMA diagram, visually represented the search strategy, screening process, and inclusion criteria. The study protocol was registered in PROSPERO CRD42023487656 to enhance transparency and adherence to systematic review guidelines. Quality assessment of the included studies was performed using the Newcastle-Ottawa Scale, revealing that the 13 articles meeting the inclusion criteria demonstrated a high level of evidence. Seven studies were included in the meta-analysis regarding shear bond strength. In summary, the synthesized findings from these studies strongly underscore the promising potential of orthodontic materials modified with AgNPs. These materials exhibit effective resistance against cariogenic bacteria without compromising bonding properties below clinical acceptability.
