This study aimed to evaluate the Mangaba Genebank of Embrapa Tabuleiros Costeiros, Brazil, since its implementation in 2006 and characterize the accessions in the reproductive stage. We used 21 descriptors (botanical, morphological, growth, biometric, and physicochemical) to assess mangaba germplasm, as well as the data of enrichment and survival since the genebank implementation. There was a success in the enrichment, maintenance, and diversity of the germplasm evaluated. The results showed a significant difference between the accessions for most descriptors. The fruits presented good quality characteristics both for fresh consumption and processing. Moreover, these results supported the publication of the minimum descriptors for this species by Bioversity International and Embrapa. The knowledge of mangaba variability will subsidize conservation strategies and should encourage future researches on breeding.

We used 30 descriptors in 54 plants from 10 accesses in the fruiting stage (CA, AB, PT, PR, TC, PA, LG, BI, IP and AD) from the Active Germplasm Bank of Embrapa Coastal Tablelands located in the Experimental Field of Itaporanga d'Ajuda, Sergipe, Brazil, faithful trustee of the species. The diversity was verified between and within the accesses, however, without direct relation with its origins. The mean vitamin C content was considered high (394.45 mg/100g) and there was no difference between the accesses. There was a significant difference for most descriptors related to the growth of the plants and the physicochemical characteristics of the fruits. The results are important for the conservation actions of the species and individuals selection for future improvement programs. Lethal yellowing is a disease that causes severe damages to coconut palms and it has devastated the coconut canopy of several Caribbean islands, and areas of southern Florida. The Fiji dwarf coconut accession has demonstrated tolerance to this disease. Pollen storage and germination provides useful information for conservation strategy and genetic improvement programs. The objective of this study was to evaluate pollen viability of Fiji dwarf coconut by in vitro germination under different nutritional and abiotic conditions. For the first experiment, two liquid culture media were evaluated for in vitro germination of Fiji dwarf coconut pollen; the Lora culture medium and the Armendariz culture medium combined with four temperature levels. In the second experiment, different sucrose concentrations in the Lora culture medium (4, 8, 12 and 16%) were evaluated combined with two incubation times (6 and 24 h) in the presence or absence of light. The highest in vitro germination of pollen grain was obtained in Lora culture medium by 24 h incubation period at 28 ºC. The light condition does not influence the germination process. In vitro germination percentage improved as the sucrose concentration increased according to a linear regression. The ex situ conservation of seeds in cryobanks demands previous studies of dehydration procedures for the adjustment of the adequate moisture that makes long-term conservation possible. The objective of this work was to evaluate the effect of dehydration time on the regenerative capacity of embryonic axes of two genipap accessions (Umbaúba and Núcleo Bandeirante). Seeds were submitted to desiccation in policarbonate box containing silica gel for 0, 12, 16 and 20 h at room temperature and after each period, seed moisture content was determined. Seeds were inoculated in germination medium and samples were inserted into cryotubes and immersed in liquid nitrogen (-196ºC). Desiccation in silica gel was efficient in reducing the moisture content in both accessions. Umbaúba accession presented 100% germination at all desiccation treatments before cryopreservation and Núcleo Bandeirante was more sensitive to desiccation with 10% germination in 20 h. Umbaúba accession showed a reduction of growth variables with increased desiccation time. The treatments tested did not affect the growth variables of Núcleo Bandeirante accession. The desiccation, although changes observed in the ultra-structures of embryonic axis, did not influence the regeneration percentage of cryopreserved accessions.

The USDA-ARS National Plant Germplasm System (NPGS) maintains 946 accessions of sugarcane (Saccharum spp.) in the field at the Subtropical Horticult sure Research Station in Miami, Florida. These accessions are particularly vulnerable to hurricanes, diseases and other threats. We sought to identify a method whereby clonally propagated sugarcane accessions could be successfully introduced into tissue culture, multiplied, and then cryopreserved as shoot tips for long-term preservation at he National Laboratory for Genetic Resources Preservation in For Collins, Colorado. For many accessions, 70% isopropyl alcohol and 20% commercial bleach treatments, followed by three rinses of sterile water weresufficient to remove microbial contaminants during the introduction process. However, in some cases, cefotaxime was particularly effective for removing bacterial contamination. We found that antioxidant treatments of glutathione, glycine betaine, and ascorbic acid did not improve regrowth after liquid nitrogen exposure usine either PVS2 or PVS3 as cryoprotectants in droplet vitrification cryopreservation methods. Exposure durations of PVS2 and PVS3 were optimized, with and without exposure to liquid nitrogen (LN), and shoot tip regrowth levels ranged from 0 to 37% after LN exposure. Published methods for encapsulation dehydration and V-plate vitrification cryopreservation procedures were tested to determine if acceptable results could be obtained. Using these methods, shoot tip regrowth levels ranged from 0 to 50% after LN exposure. Information about the establishment phase of Miscanthus sinensis Andersson as sterile procedure to avoid the contamination were few reported. The aim of this study was to evaluate different immersion sterilization procedures with and without the biocide, Plant Preservative Mixture (PPM?) on in vitro contamination of two Miscanthus genotypes. The plant material were canes from adult plants of two accessions PI 668371 and PI 668375 cultivated in the germplasm bank of USDA-ARS, Griffin, Georgia. Apical meristems were submitted to immersion sterilization procedures (70% isopropyl alcohol and 1.5, 5.0 or 8.25% NaOCl for different time periods, followed by rinsing three times with autoclaved distilled water. After that, the explants were inoculated on MS basal medium in the presence or absence of 1 mL L-1 PPM?. The experimental design was fully randomized in a 2 x 5 x 2 factorial scheme (genotypes x sterilization process x PPM?) with five replicates per treatment and five apical meristems per experimental unit. Aseptic treatments showed no differences for percentage of bacterial and fungal contaminations and percentage of explant survival. In the presence of PPM ? there was less bacterial contamination in the accessions (28%) than in the absence (100%). For the percentage of fungal contamination, PPM ? had no significant effect on PI 668371 accession. However, on PI 668375 accession there was a lower percentage of fungal contamination (16%). The biocide PPM ? may be an efficient agent to prevent bacterial contamination on in vitro cultures of Miscanthus spp and fungal contamination in PI 668375 accession. A mandioca é a única espécie do gênero Manihot comercialmente produzida. A produção de mudas micropropagadas possui limitações, e entre estas está o alto custo com energia utilizadas nas salas de crescimento, e na taxa de sobrevivência das plantas na fase de aclimatização, porém a cultura de tecidos é uma ferramenta de multiplicação rápida e obtenção de plantas sadias (cultura de meristema), pois o método de propagação vegetativa acarreta problemas fitossanitários como disseminação de patógenos. Uma alternativa para diminuir gastos dessa técnica seria a redução de sacarose ao meio e indução de enraizamento sob luz natural em casa de vegetação. Sendo assim, o objetivo desse trabalho foi avaliar a influência da luz e sacarose na sobrevivência e desenvolvimento de mudas de mandioca micropropagada. As variedades BRS Formosa, Lagoão e BRS Verdinha, cultivadas em meio MS acrescido de diferentes concentrações de sacarose (10; 20 e 30 g.L-1), e após 30 dias em sala de crescimento, os frascos foram incubados em sala de crescimento e casa de vegetação por 15 dias. Em cultivos envolvendo diferentes tipos de luz, BRS Formosa e Lagoão, alcançaram melhores médias sob luz natural e altas concentrações de sacarose, enquanto que BRS Verdinha obteve melhor desenvolvimento em luz natural e baixa concentração de sacarose. Drought is the most limiting environmental factor to crop productivity and presents a great variability in the degree of tolerance among and within species, among varieties. The aim of this study was to characterize sugarcane accessions regarding tolerance to water stress during in vitro cultivation based on changes in biometric, physiological and biochemical characteristics, within species and among species, to support future breeding programs. Adventitious shoots of five sugarcane accessions: Saccharum robustum, Saccharum spontaneum and Saccharum officinarum species, cultivated in Murashige and Skoog medium supplemented with 2% sucrose and 4 g/l Phytagel were used in five water potentials, 0, -0.3, -0.6, -0.9, -1.2 MPa, induced by mannitol. Survival, length of shoots and roots, number of shoots and roots, biomass, proline content in leaves and activity of antioxidant enzymes were analyzed. There is difference among species, and also, within the same sugarcane species when submitted to in vitro drought stress, and S. officinarum was shown to be the most tolerant. Proline can be used as a biochemical indicator of response to drought in sugarcane accessions and its accumulation was intensified in S. robustum and S. spontaneum accessions. The aim of this study was to evaluate the in vitro morphogenic potential of genipap (Genipa americana L.) zygotic embryos. Seeds obtained from ripe fruits had their zygotic embryos excised and inoculated in MS medium with 4.44µM of 6-benzylaminopurine (BAP) and supplemented with 0.0; 1.07; 2.14 and 3.21µM of naphthalene acetic acid (NAA). The potential of explants regeneration and the shoot length and number of leaves in plantlets were evaluated. The in vitro regeneration of genipap is possible from the conversion of zygotic embryos in a MS medium with 4.44µM BAP supplemented with 3.21µM NAA. RESUMO: O objetivo do trabalho foi avaliar o potencial morfogênico in vitro de embriões zigóticos de jenipapeiro (Genipa americana L.). Sementes obtidas de frutos tiveram seus embriões zigóticos excisados e inoculados em meio MS com 4,44µM de 6-benzilaminopurina (BAP) suplementado com 0,0; 1,07; 2,14 e 3,21µM de ácido naftaleno acético (ANA). O potencial de regeneração dos explantes e o comprimento da parte aérea e o número de folhas nas plântulas formadas foi avaliado. Observou-se que é possível a regeneração in vitro de jenipapeiro a partir da conversão de embriões zigóticos em meio MS com 4,44µM de BAP, suplementado com 3,21µM de ANA.

this work describes the establishment of procedures to induce in vitro callogenesis from Poincianellapyramidalis (Tul.) L. P. Queiroz, Fabaceae, explants. Nodal, internodal and leaf segments were iso-lated from in vitro germinated seedlings and cultured in MS medium with 0, 2.5, 5 and 10 mg l−1of2,4-dichlorophenoxyacetic acid. After 30 days, the explants with induced callus showed a quadraticresponse for the segments nodal, internodal and leaf, with increasing the callus formation in 2,4-dichlorophenoxyacetic acid concentrations of 6.28, 6.49 and 4.91 mg l−1, respectively. In 30 days there wasa linear oxidation rise with the increase to the 2,4-dichlorophenoxyacetic acid. After 60 days, oxidationvalues were minimum, at 2,4-dichlorophenoxyacetic acid concentrations of 5.13 mg l−1(internodal) and3.98 mg l−1(leaf). The highest callus production was observed after 30 days in the presence of 6.09 mg l−1,5.82 mg l−1and 4.91 mg l−1of 2,4-dichlorophenoxyacetic acid in nodal, internodal and leaf segments,respectively. After 60 days these segments showed peaks of production at 7.0 mg l−1(nodal), 6.15 mg l−1(internodal) and 5.08 mg l−1(leaf) of 2,4-dichlorophenoxyacetic acid. For callus induction the intake of2,4-dichlorophenoxyacetic acid was essential. The greater intensity in callus formation was observed in4.91 mg l−1in leaf segments after 30 days. O objetivo do presente trabalho foi estudar as respostas morfogenéticas de explantes de jenipapeiro em diferentes condições de cultura in vitro para subsidiar futuros protocolos de multiplicação e produção in vitro de metabólitos secundários. Após 90 dias de cultivo, as plântulas de jenipapeiro foram segmentadas (segmentos nodal e foliar) e transferidas para meio MS suplementado com 30 g L-1 de sacarose e diferentes concentrações de 2,4 - Diclorofenoxiacético - 2,4-D (0; 4 e 8 mg L-1) combinadas com quatro concentrações de benzilaminopurina-BAP (0; 1; 2 e 3 mg L-1). Aos 120 dias a porcentagem de explantes com resposta morfoge-nética quanto à formação de calos e/ou organogênese foi avaliada. O 2,4-D nas concentrações de 4 e 8 mg L-1 induz aumento na resposta morfogenética, principalmente na formação de calos, segmentos foliar e nodal de Genipa americana L. acesso SIR. O BAP a 1,77 mg L-1 induz maior formação de calos em segmentos foliares e maior regeneração de brotos nos segmentos foliar e nodal. O trabalho teve como objetivo avaliar alternativas de manejo fitotécnico da bananeira comprida, cultivar D?Angola, visando ao controle da Sigatoka-negra. O experimento foi conduzido no campo experimental da Embrapa Acre no município de Rio Branco-AC com os tratamentos distribuídos em blocos ao acaso, completos, com seis repetições e constituídos por diferentes densidades de plantio, correspondendo: T1 - 3 m x 3 m (1.111 plantas/hectare); T2 - 3 m x 2 m (1.666 plantas/hectare); T3 - 2,5 m x 2 m (2.000 plantas/hectare); T4 - 4 m x 2 m x 1,5 m (1.333 plantas/hectare); T5 - 2 m x 2m (2.500 plantas/hectare) e; T6 - 3 m x 3 m (1.111 plantas/hectare), este último alocado sob o bosque de seringueira. Foram realizadas avaliações durante dois ciclos de produção. As plantas da cv. D'Angola (AAB) cultivadas no espaçamento 3,0 x 3,0m, sombreadas com seringueira, apresentaram maior altura de plantas, maior número de folhas vivas na floração e colheita, maior peso médio do cacho, maior massa das pencas, maior ciclo de produção e menor severidade da Sigatoka-negra. Maiores produtividades foram obtidas quando se adotaram maiores densidades de plantio. O consorciamento de bananeiras com árvores de seringueiras mostrou-se ser uma boa alternativa para viabilizar o cultivo da cv. D'Angola (banana comprida).

O objetivo deste estudo foi avaliar o potencial de armazenamento de grãos de pólen de acessos de coqueiro Anão Verde do Brasil de Jiqui (AVeBrJ), coqueiro Gigante do Brasil Praia do Forte (GBrPF) e coqueiro Anão Vermelho dos Camarões (AVC) em diferentes condições de armazenamento. Plantas matrizes dos acessos foram selecionados no Banco Ativo de Germoplasma de Coco da Embrapa Tabuleiros Costeiros. Os grãos de pólen foram coletados, armazenados em criotubos e mantidos nas seguintes condições de armazenamento: T1: Congelador (-4°C) T2: Freezer (-20°C), T3: Freezer (-80°C) e T4: Nitrogênio líquido (-196°C). A viabilidade grãos de pólen foi determinada por coloração com carmim acético a 1% e por germinação in vitro em meio de cultura Lora aos 30 e 60 dias após o armazenamento. O armazenamento em refrigeração (-4°C), freezer (-80°C) e nitrogênio líquido (-196°C) promoveu uma melhor viabilidade de grãos de pólen para o acesso GBrPF . O armazenamento em freezer (-20°C) e nitrogênio líquido (-196°C) proporcionou melhor viabilidade de grãos de pólen para o acesso AVC. O armazenamento em freezer (-20°C e -80°C) e nitrogênio líquido (-196°C) promoveu uma melhor viabilidade de grãos de pólen do acesso AVeBrJ. A viabilidade de grãos de pólen do acesso GBrPF é estável após 30 e 60 dias de armazenamento.

The objective of this work was to determine the mannitol concentration for in vitro germination and minimum growth of coconut (Cocos nucifera). The following accessions were used: Brazilian Red Dwarf Gramame (BRDG), Malaysian Yellow Dwarf (MYD), Brazilian Tall (BRA), Polynesian Tall (PYT), and Rennell Island Tall (RIT). Embryos were inoculated in an Y3 culture medium at different mannitol concentrations (0, 0.1, 0.2, 0.3, and 0.4 mol L?1). Inhibition of shoot development was verified at 0.1, 0.2, and 0.3 mol L?1, for MYD and BRDG, and at 0.1 and 0.2 mol L?1, for PYT, BRA, and RIT. This research had as objective to characterize genetically individuals of physic nut cultivated in experimental areas in Sergipe, Brazil by means of RAPD molecular markers. Leaves of 40 individuals were collected and DNA was isolated using CTAB 2% method. Were used 30 primers RAPD for DNA amplification, and this data was used to estimate the genetic similarity among the pairs of individuals, using Jaccard coefficient, and group them out for the UPGMA method. Also, the genetic structure and diversity of the populations were assessed using AMOVA. Of the 100 fragments generated, 29 of were polymorphic. A similarity average of 0.54 among the individuals was found and the amplitude similarities varied from 0.18 to 1.00. One of them (U5) was unit clusters and formed by the most divergent individuals.
