The angelshark, Squatina squatina, is listed as Critically Endangered on the IUCN Red List of Threatened Species, and remaining populations are highly fragmented throughout its historical distribution. The Canary Islands archipelago in the North East Atlantic has been identified as a uniquely large stronghold for the species. In the present study, we compared the population genetic structure of S. squatina across different islands of the Canary Island archipelago using both microsatellite and single nucleotide polymorphism (SNP) markers. Both markers revealed significant differentiation of angelsharks between islands in the archipelago, with three main genetic units at: (1) Tenerife, (2) Gran Canaria and (3) the island group consisting of La Graciosa, Lanzarote and Fuerteventura. Our results imply a connectivity barrier between some adjacent islands, most likely driven by abyssal depths, and varying geological history and formation of each island and oceanographic patterns (i.e. seasonal coastal upwellings off the African coast). Therefore, we suggest that in the Canary Islands, S. squatina populations should be managed locally, with conservation and research priorities designed and implemented specifically for each of the three genetic units.

Establishing how threatened wildlife are distributed spatially and temporally is essential for effective conservation and management planning. While many shark species are threatened globally, knowledge on sex-specific differences in behaviour and fine-scale habitat use remains limited, hindering the implementation of appropriate conservation actions. Here, acoustic telemetry was used to investigate sex-specific space use in the Critically Endangered angelshark Squatina squatina in the Canary Islands (Atlantic Ocean), a key stronghold for the species. We collected data on space use, residency and movement of adult males (n = 32) and females (n = 72) in Spain’s largest marine reserve (La Graciosa Marine Reserve) over 4 yr (2018-2022). Presence in offshore sites (>80 m depth) indicated long-term utilisation of deep-water habitat, not previously observed in this species. Males were more likely to be detected in deep water and displayed significantly greater levels of activity and roaming behaviour than females. Patterns of behaviour in shallow waters indicated both social and habitat sexual segregation. Diel variability was also recorded, with greater activity occurring nocturnally. This study demonstrates the importance of considering the spatiotemporal distribution of both sexes, horizontally and vertically, when designing effective conservation measures.

This paper proposes a new methodology to solve the bone remodeling problem using one-dimensional (1D) elements to discretize trabecular structures more efficiently for 2D and 3D domains. An Euler integration scheme is coupled with the momentum equations to obtain the evolution of material density at each step. For the simulations, the equations were solved by using the finite element method, and two benchmark tests were solved varying mesh parameters. Proximal femur and calcaneus bone were selected as study cases given the vast research available on the topology of these bones, and compared with the anatomical features of trabecular bone reported in the literature. The presented methodology has proven to be efficient in optimizing topologies of lattice structures; It can predict the trend of formation patterns of the main trabecular groups from two different cancellous bones (femur and calcaneus) using domains set up by discrete elements as a starting point. Preliminary results confirm that the proposed approach is suitable and useful in bone remodeling problems leading to a considerable computational cost reduction. There is an unmet need to improve the response to immune checkpoint blockade in patients with head and neck squamous cell carcinoma (HNSCC). Here the authors show that aberrant HER3 activation sustains the proliferation of PIK3CA wild type HNSCC cells and that HER3 inhibition increases response to PD-1 blockade in HNSCC preclinical models.Immune checkpoint blockade (ICB) therapy has revolutionized head and neck squamous cell carcinoma (HNSCC) treatment, but <20% of patients achieve durable responses. Persistent activation of the PI3K/AKT/mTOR signaling circuitry represents a key oncogenic driver in HNSCC; however, the potential immunosuppressive effects of PI3K/AKT/mTOR inhibitors may limit the benefit of their combination with ICB. Here we employ an unbiased kinome-wide siRNA screen to reveal that HER3, is essential for the proliferation of most HNSCC cells that do not harbor PIK3CA mutations. Indeed, we find that persistent tyrosine phosphorylation of HER3 and PI3K recruitment underlies aberrant PI3K/AKT/mTOR signaling in PIK3CA wild type HNSCCs. Remarkably, antibody-mediated HER3 blockade exerts a potent anti-tumor effect by suppressing HER3-PI3K-AKT-mTOR oncogenic signaling and concomitantly reversing the immune suppressive tumor microenvironment. Ultimately, we show that HER3 inhibition and PD-1 blockade may provide a multimodal precision immunotherapeutic approach for PIK3CA wild type HNSCC, aimed at achieving durable cancer remission.

This work is the result of an international research effort to determine the main impacts of the COVID-19 pandemic on marine recreational fishing. Changes were assessed on (1) access to fishing, derived from lockdowns and other mobility restrictions; (2) ecosystems, because of alterations in fishing intensity and human presence; (3) the blue economy, derived from alterations in the investments and expenses of the fishers; and (4) society, in relation to variations in fishers’ health and well-being. For this, a consultation with experts from 16 countries was carried out, as well as an international online survey aimed at recreational fishers, that included specific questions designed to capture fishers’ heterogeneity in relation to behavior, skills and know-how, and vital involvement. Fishers’ participation in the online survey (5,998 recreational fishers in 15 countries) was promoted through a marketing campaign. The sensitivity of the fishers’ clustering procedure, based on the captured heterogeneity, was evaluated by SIMPER analysis and by generalized linear models. Results from the expert consultation highlighted a worldwide reduction in marine recreational fishing activity. Lower human-driven pressures are expected to generate some benefits for marine ecosystems. However, experts also identified high negative impacts on the blue economy, as well as on fisher health and well-being because of the loss of recreational fishing opportunities. Most (98%) of the fishers who participated in the online survey were identified as advanced, showing a much higher degree of commitment to recreational fishing than basic fishers (2%). Advanced fishers were, in general, more pessimistic about the impacts of COVID-19, reporting higher reductions in physical activity and fish consumption, as well as poorer quality of night rest, foul mood, and raised more concerns about their health status. Herein we present an innovative route for model biomass compounds upgrading via “H2-free” hydrodeoxygenation (HDO) reactions. The underlaying idea is to implement a multifunctional catalyst able to activate water and subsequently use in-situ generated hydrogen for the HDO process. In this sense we have developed a series of effective Ni and Pt based catalysts supported on N-promoted graphene decorated with ceria. The catalyst reached commendable conversion levels and selectivity to mono-oxygenated compounds considering the very challenging reaction conditions. Pt outperforms Ni when the samples are tested as-prepared. However, Ni performance is remarkably boosted upon applying a pre-conditioning reductive treatment. Indeed, our NiCeO2/GOr- N present the best activity/selectivity balance and it is deemed as a promising catalyst to conduct the H2-free HDO reaction. Overall, this “proof-concept” showcases an economically appealing route for bio-compounds upgrading evidencing the key role of advanced catalysts for a low carbon future.
