Objective: A study was planned to investigate the effect of exposure to cigarette smoke and the effects of calcium channel blockers in experimentally induced sciatic nerve injuries. Meteryal-Metod: The sciatic nerve was cut on unilaterally in all the groups, and repair was performed at hour 0. After repair, single dose of 1 ml of 0.9% saline was administered to group 1, cigarette smoke to group 2, 1mg/kg calcium channel blocker to group 3, cigarette smoke and 1mg/kg Calcium channel blocker was given to group 4. Results: At the end of the 12th week, gait analysis at and sciatic function index (SFI), in 30 minutes for 90 days the number of myelinated axons, axon areas and myelin diameters were assessed. Ethanol (ETH) delays myelination in the sciatic nerve of rat following fetal exposure. It has been suggested that melatonin (MLT), a naturally occurring indole, may be a potential protective agent for ETH toxicity, possibly due to its protective effects against free radical damage in experimental models. However, the precise mechanism of MLT protection in the development of the rat sciatic nerves and their myelination is unclear. In this study, the effect of ETH, MLT and ETH+MLT on the myelin thickness, axon number and axonal area of rat sciatic nerve were investigated. Pregnant rats were injected with either 25% ETH (dosage 2g/kg body weight), 25% ETH plus MLT (dosage 2g/kg and 10 mg/kg, respectively), MLT (dosage 10 mg/kg) or physiological saline (dosage 1 ml/kg) at 7, 15 and 20 days post-conception (first, second and third trimesters, respectively). At postnatal day 14, sciatic nerves of offspring were dissected out and processed for stereological analyses. Our results demonstrated that the myelin thickness was increased by exogenous MLT, and decreased by acute ETH, during first and second trimesters. It was found that MLT was protective to developing myelin thickness following fetal exposure during the first trimester, whereas it was toxic to myelin thickness during the third trimester. The presented study is the frst stereological study indicating that exogenous MLT has a toxic efect on the myelin thickness of rat sciatic nerves during the third trimester. Based on our findings, we suggested that exogenous MLT, given during the third trimester as an antioxidant agent, as well as acute ETH, given in the first and second trimesters, may alter the normal development of sciatic nerve myelination. © 2009 OMU All rights reserved.

Çalışmada, koyunlarda in vitro ortamda şekillenen morula ve blastosist verimlerinin geliştirilmesi amacı ile farklı gelişim evrelerinde uygulanan farklı miktarlarda (10 ng/ml, 50 ng/ml ve 100 ng/ml) aktivin A proteini içeren medyumlarda oosit ve embriyoların gelişimleri değerlendirildi. Ayrıca blastosistlerde büyüme ve apoptoz faktörlerinin bağıl miktarları incelenerek Bmp4, Smad4, Bax, Blc-2 ve Caspase-3 gen transkriptlerinin değişim oranları karşılaştırılmıştır. Bu amaçla mezbahadan alınan koyun ovaryumlarından 1237 adet oosit kullanılarak metafaz I (MI), metafaz II (MII), bölünen, morula öncesi, morula sonrası, blastosist ve dejenere hücre sayıları değerlendirilmiştir. Farklı zaman aralıklarında aktivin A'nın etkilerinin incelenmesi amacıyla iki deney planı oluşturulmuştur. Deney planı A'da 10, 50 ve 100 ng/ml aktivin A konsantrasyonları maturasyon aşamasında (IVM) kullanılarak; deney planı B'de ise aynı konsantrasyonlar IVF sonrası medyumlara eklenerek aktivin A'nın etkileri incelenmiştir. Aktivin A'nın bölünme oranları üzerine olan en etkili sonucu IVF sonrası ortama 50 ng/ml konsantrasyonunda eklenmesi ile %92,7 verim elde edilmiştir. Maturasyon ortamına eklendiğinde morula gelişiminde sadece 100 ng/ml konsantrasyonda bir artışa neden olduğu (%81,1) ancak IVF sonrası bütün konsantrasyonlarda artışa neden olduğu ve maksimum düzeye (%89,3 morula) 50 ng/ml konsantrasyonu ile ulaştığı görülmüştür. Mezbahadan alınan örneklerden testis ve epididimisin 3 farklı bölgesinden gelişim aşamasındaki spermatozoonlar elde edilmiştir. Koç spermatozoonlarının gelişim ve maturasyonunda geçirdiği morfolojik değişimler, kromatin kondensasyonu aşamalarının spermatozoon gelişim aşamaları ile olan ilişkisi ile birlikte ejakulat ve dondurulup çözdürülmüş koç spermatozoonlarında in vitro kapasitasyon/akrozom reaksiyonu geçirmeden ve geçirdikten sonraki yapısal özellikleri ışık mikroskopisi çalışmayla ortaya konması amaçlanmıştır. Testis ve epididimisten alınan spermatozoon örneklerinin morfolojik özellikleri elektron mikroskobik çalışma ile incelenmiştir. Aynı gruplarda immunofloresan yöntem ile Fertilin-β, Calmegin, İzumo-1, P34H, ACE ve Fibronektin'in varlıkları ve konumlarının tespit edilmesi aynı zamanda bu gruplarda western blot tekniği ile kantitatif olarak aynı proteinlerin ekspresyon analizleri yapılmıştır. Kromatin kondensasyonu çalışmasında numuneler arasında istatistiksel olarak anlamlı bir farklılık tespit edilmemiştir (p>0,05). İndirekt immunofloresan yöntemi ile Fertilin-β, Calmegin, P34H proteinlerinde kaput, korpus, kauda ve matur spermatozoonlarda bölgeye göre farklı yoğunluk ve dağılımda işaretlenme görülmiştür. Dondurulup-çözdürülmüş örneklerde ise hem ejakulat örneklerine hem de kauda örneklerine göre çok daha az yoğun bir işaretlenme tespit edilmiştir. İn vitro kapasitasyon/akrozom reaksiyonu geçiren örneklerde ise İzumo-1 proteini dışında diğer incelenen proteinlerde bir işaretlenme görülmemiştir. İn vitro kapasitasyondan sonra İzumo-1'lerin akrozomun üzerinde ekvatoral bölgeye doğru artan bir bant şeklinde işaretlenmesi görülürken in vitro akrozom reaksiyonu sonrası sadece ekvatoral bölgede bant şeklinde işaretlenme görülmüştür. Fibronektin antikoru ile numunelerde beklenenin tersine spermatozoonun baş bölgesinde değil de bölgeye göre farklı konumlarda non spesifik olarak işaretlenmiştir. ACE antikoru ile numunelerin hiç birinde işaretlenme görülmemiştir. Western blot yöntemi ile incelenen proteinlerin kDa düzeylerinde numunelerin alındığı bölgelere özgü farklılıklar olduğu ve koç spermatozoonlarına özgü olası izoformlar ya da benzer epitoplara sahip proteinler işaretlenmiştir. Ultrastrüktür çalışmasında da, testis ve epididimisin kaput, korpus ve kauda bölgelerindeki spermatozoonların yapısal özelliklerinin birbirlerinden farklı olmadıkları gözlenmiştir. Determination of morphological changes of ram spermatozoa during development and maturation have been aimed and relation ship between chromatin condensation stages and spermatozoa developmental stages with ejaculate and structural features of freezed-thawed ram spermatozoas with and without in vitro capacitation/acrosome reaction also been evaluated by using light microscopy. Morphology of spermatozoas obtained from testis ande epididymis were evaluated by electron microscopy techniques. In the same groups, determination of existence and localisation of Fertilin-β, Calmegin, Izumo-1, P34H, ACE and Fibronectin were analyzed quantatively via their protein expression profiles by western blotting technique. In chromatin condensation experiments, statistically meaning full results could not be obtained between samples (p>0.05). Fertilin-β, Calmegin, P34H proteins in caput, corpus, cauda and mature spermatozoas showed marking in different density and distrubition with indirect immunofluorescence technique. Freezed-thawed samples had lower density and marking than both ejaculate and cauda samples. Marking was not obtained except Izumo-1 protein from the samples undergo in vitro capatitation/acrosome reaction. Marking of Izumo-1 protein was seen as increasing band formation through equatorial region on acrosome, after in vitro capacitation, however after acrosome reaction, the band formation was only equatorial region. In contrast to expected marking on spermatozoa head, non specific marking was obtained on different localization changing with the region in fibronectin antibody and samples. ACE antibody did not mark the samples. Region specific differences of proteins at kDa level were obtained with western blotting and possible isoforms specific to ram spermatozoa or proteins with similar epitops were marked. In ultrastructure study, spermatozoa structure did not show any differences in testis, caput, corpus and cauda region of epididyms. 149
