A cardiovascular disease is a group of diseases of the heart and blood vessels. The most common cause of death in humans is associated precisely with cardiovascular diseases, and α2-adrenergic receptors (α2-AR) are known to be involved. α2-AR reduce the central sympathetic output and peripheral release of catecholamines and therefore, can prevent sympathetic hyperactivity and hypertension. Therefore, various agonists and antagonists of adrenergic receptors are widely used in the treatment of cardiovascular diseases in medicine, among them clonidine and ST-91. The objective was to compare the effects of clonidine and ST-91, selective agonists of α2-AR, on the cardiac activity in rats. Stimulation of α2-AR by ST-91 (0.01 mg/kg) causes a short-term negative chronotropic effect, while stimulation of α2-adrenergic receptors with clonidine (0.01 mg/kg) led to a negative chronotropic effect throughout the experiment. We hypothesize that the short-term effect of ST-91 on heart rate is associated with the activation of one of the three subtypes of α2-adrenergic receptors. Identification of the specific subtype due to which this effect was observed is part of future research. [ABSTRACT FROM AUTHOR]

[EN] Adverse environmental conditions, such as rising temperatures and increased droughts, put crop productivity at risk. Under stress conditions, plants produce the hormone abscisic acid (ABA), which is involved in the response and adaptation to adverse conditions such as drought. In the plant cell, ABA is sensed through its binding to soluble PYR/PYL receptors. ABA signalling is orchestrated through a module that includes the PYR/PYL family of ABA receptors, PP2C family phosphatases, SnRK2 family kinases and ABA-responsive transcription factors. The exogenous application of this phytohormone is able to improve crop yields, however, it has the disadvantage of having a high production cost, as well as poor stability and photosensitivity. In order to find solutions to water stress, and thanks to the development of chemical genetics, synthetic molecules capable of intervening in water saving by closing the stomata are being studied. In this work, 70 ABA receptor agonist candidates have been characterised, both in vitro and in vivo. We found that the synthetic ligand, C3, shows potent in planta activity and strong activation of dimeric subfamily III receptors in both Arabidopsis and maize. Our results indicate that dimeric receptors play a key role in the activation of certain responses to ABA, such as stomatal closure, root growth inhibition and germination. The compound C3 is an ABA receptor agonist with an activity very similar to that of the best compound described to date and shows great potential for field use.

Background: Behavioral traits of pigs have been shown to be partly under genetic control, which raises the possibility that behavior might be altered by genetic selection, resulting in pigs with better growth performance. Objective: To evaluate the behavior and growth of finishing pigs and investigate pigs selected for high or low social breeding value (SBV) in relation to social behavior and group growth. Methods: Thirty-five females and 35 boars from five positive and five negative SBV groups of finishing pigs were grown from 30 to 90 kg and housed in 10 test pens (3.0 × 3.3 m, 7 pigs/pen). Pigs were recorded with video technology for nine consecutive hours on days 1, 15, and 30 after mixing. Pigs were weighed at approximately 90 kg body weight and the number of days to reach 90 kg was then calculated. Results: The frequency and duration of behaviors were present in the positive and negative SBV groups after mixing. On day 1 after mixing, agonistic behavior was significantly higher (p=0.027) for the -SBV group compared with the +SBV group. Feeding and feeding-together behaviors were significantly higher (p<0.003) in the +SBV group on days 1 and 30 after mixing. Moreover, growth performance to reach 90 kg body weight was significantly faster (p<0.002) in the +SBV group than in the -SBV group. Conclusion: Social interactions, such as feeding-together behavior, among pen mates might affect their growth rate and feed intake. Selection for SBV could be used as an indirect technique for improving growth performance of pigs. [ABSTRACT FROM AUTHOR]

Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology & Toxicology Student: Miriama Hutníková Supervisor: Prof. PharmDr. Petr Pávek, PhD. Title of diploma thesis: Screening of selected alkaloids of Fumariaceae and Amaryllidaceae families on the farnesoid X receptor and the G protein coupled receptor 1 Farnesoid X receptor (FXR) and bile acid receptor associated with G protein 1 (TGR5) significantly affect metabolic processes in the human body. The role of FXR in neuronal apoptosis in Alzheimer's disease (AD) has also been discovered. The possible structural similarity of the small lipophilic molecules binding to these receptors and the alkaloids found in the plants Corydalis cava and Narcissus pseudonarcissus, as well as the richoften use of these plants in traditional medicine, represent a potential therapeutic intervention for these molecules. In our screening methods, we performed tests using a luciferase gene reporter assay to determine the ability of the alkaloids to interact with FXR and TGR5 in the HepG2 cell line. Many derivatives have shown a strong ability to antagonize FXR and TGR5 activated by obethicholic (OCA) or litocholic (LCA) acids in this assay. Some of the compounds also demonstrated the ability to potentiate the effects of OCA or LCA. Cytotoxicity...

- Abstract
Soon after the discovery of the first active component of Cannabis sativa - -A9-THC, it has been shown that its action is mediated mainly through the activation of specific membrane protein - cannabinoid receptor (CB). So far two types of this protein were isolated: CB1 and CB2. It is already known that they constitute the intracellular signaling system, called Endocannabinoid System (ECS), which also includes their ligands and enzymes involved in the synthesis and degradation of this compounds. ECS is abundant in the brain regions associated with feeding control and also in peripheral organs regulating food intake and glucose and lipids metabolism, suggesting the crucial role of this system in pathogenesis of obesity. Much evidence has proven potent orexigenic action of endocannabinoids induced via the CB1 receptors activation in CNS. They also stimulate lipogenesis, leading to the fat accumulation in such organs like the liver. Moreover, the CB1 receptor agonist-based treatment induced insulin resistance and development of hyperglycaemia - characteristic metabolic parameters of obesity. These results has led to the development of CBl-blocking drugs with potential efficacy in obesity treatment. Preclinical and clinical studies with the use of antagonist/inverse agonist - rimonabant have shown significant decrease in body weight and improve in cardiometabolic risk factors. However, serious psychiatric effects of this substance contributed to its withdrawal from the market. The main goal is to design a drug which might prevent obesity without these side effects. Therapies based on the CB1 receptor's neutral antagonists or selective, inverse agonists might be a promising strategy in obesity treatment. [ABSTRACT FROM AUTHOR]

This article discusses emerging methodologies in Moana Nui a Kiwa (MNak) (Pasific Peoples) performance art practices in Aotearoa New Zealand (NZ). It explores the ways in which artistic research guides research questions and final research outputs within the context of a practice led PhD degree. John Vea’s (Tonga/NZ) underlying research methods reference Timote Vaioleti’s work on ‘talanoa’ as a MNak notion about respectfulness in personal encounters with people. Vea’s performance practice engages with MNak minority groups — exploring tropes of migration and subsequent interaction with hegemony — where ‘co-operations’ with collectives and small groups challenge some traditional research models of leadership and authorship. These indigenous approaches encourage a different reading of theorists such as Chantel Mouffe and her ideas of navigating artistic activism and agonistic spaces of shared cooperation. The differential action of the novel agonist JN403 at neuronalα7 and muscle nicotinic receptors (AChRs)was explored by using a combination of functional and structural approaches. Single-channel recordings reveal that JN403 is a potent agonist of α7 but a very low-efficacy agonist of muscle AChRs. JN403 elicits detectable openings of α7 and muscle AChRs at concentrations∼1000-fold lower and∼20-fold higher, respectively, than that for ACh. Single-channel activity elicited by JN403 is very similar to that elicited by ACh in α7 but profoundly different in muscle AChRs, where openings are brief and infrequent and do not appear in clusters at any concentration. JN403 elicits single-channel activity of muscle AChRs lacking theεsubunit, with opening events being more frequent and prolonged than those of wild-type AChRs. This finding is in line with the molecular docking studies predicting that JN403 may form a hydrogen bond required for potent activation at the α−δ but not at the α−ε binding site. JN403 does not elicit detectable Ca2+ influx in muscle AChRs but inhibits (±)-epibatidine-elicited influx mainly by a noncompetitive mechanism. Such inhibition is compatible with single-channel recordings revealing that JN403 produces open-channel blockade and early termination of ACh-elicited clusters, and it is therefore also a potent desensitizing enhancer of muscle AChRs. The latter mechanism is supported by the JN403-induced increase in the level of binding of [3H]cytisine and [3H]TCP to resting AChRs. Elucidation of the differences in activity of JN403 between neuronal α7 and muscle AChRs provides further insights into mechanisms underlying selectivity for α7 AChRs. Fil: Arias, Hugo Ruben. The purpose of this study was to compare the effect of concentric and eccentric training in developing maximal strength of quadriceps and hamstrings. In this sense, and before applying the training program, it was important to examine whether the protocols for evaluating the strength or the reliability of equipment we would use. So from the five studies that we did, the first two were devoted to this issue. The first empirical study showed a high reproducibility in concentric, isometric and eccentric peak torque for knee extensor and flexors muscles indicating that the REV9000 (Technogym) isokinetic dynamometer can be used in rehabilitation, sports medicine and for future sports performance studies. This study also showed that dynamic control ratio (Hecc:Qconc) can be a more valid indicator for imbalance reciprocal parameters. In the second study we also wanted to assess the reliability, but now of surface electromyography of the average full wave rectified signal (AvgEMG) and peak torque (PT) of quadriceps. The results of this study showed a high reproducibility of eccentric, concentric and isometric isokinetic strength for knee extensor muscles assessed with the REV9000 showing that this isokinetic device can be used for legitimate research or patient evaluation. With respect of EMG parameters we have to be more carefull, because if we don`t remove the electrodes the ICC is high, but if we remove them then reproducibility becames lower, probably due to factors affecting new electrode placement. Furthermore, the RF muscle seems to be less reliable that VL and VM muscles. In the third study the purposes were to examine the effect of 8 - week concentric versus eccentric isokinetic training program on the isometric and dynamic strength of the knee extensors and flexors. Additionally, the experimental program was followed by a 4-week detraining period that was also inspected. In conclusion, this study showed that strength gains after eccentric training were greater and more specific comparing with concentric training. In addition, the isokinetic strength bilateral ratios increased more in eccentric than concentric group and Hconc:Qconc ratio did not suffer significant changes. Greater declines in muscle strength were observed in eccentric training but still retained at a higher level than concentric training probably due to the higher gains achieved during the training period. Neural adaptations have been one of the factors that contributed to the increase of isometric and dynamic peak torque after isokinetic concentric and eccentric training. However, these neural adaptations seem to be higher in the eccentric training, which may be related to higher intensity developed during eccentric actions. This was the purpose of fourth study, to examine the effect of 8 - week concentric versus eccentric isokinetic training program on the electromyogram signal amplitude (EMG) and isometric and dynamic maximum strength on the knee extensors. In conclusion, this study showed that concentric training improve peak torque and electrical activation in all kind of contractions (concentric, isometric and eccentric), but just with statistically significant differences in peak torque and Vastus Medialis of isometric contraction. The eccentric training also improve the electrical activation in all muscles with statistically significant differences on Vastus Lateralis in all contractions and Vastus Medialis in concentric contraction, regarding to peak torque also improve in all contractions with statistically significant differences in eccentric contraction. Available research is contradictory when explaining the relationship between isokinetic knee tests and vertical jump tests. The aim of fifth study was to to identify relations among various bilateral closed kinetic chain tests and traditional isokinetic dynamometry tests. A low to moderate correlation between isokinetic strength and vertical jump tests was found supporting the view that in maximal efforts, the maximal power potential of lower limbs may be independent from the kind of exercise or tests. Further research needs to be conducted to determine a simple yet valid test to evaluate maximal power according to specific task (open or closed kinetic chain) usefull for strength and conditioning specialist to analyse the performance or for clinicians to analyse possible asymmetries. Overall, the results of these studies suggest that eccentric training achieve, for the same period of training, higher gains compared to concentric training. However, we believe it is important to prepare the athlete for a more intense workout, such as eccentric training or assess the athletes before the implementation of the training program to see if they have the capacity to perform this type of training to develop muscular strength, safely without any risk of injury.

The ionotropic glutamate receptors activated by N-Methyl-D-Aspartate (iGluR-NMDA) are of great importance in pharmacology since they are involved in neurodegenerative and neuropsychiatric disorders; they even participate in processes such as synaptic plasticity that are essential for memory formation. Subunit NR1 iGluRs-NMDA is of paramount importance for the appropriate activation of this type of receptors; in fact, many of the pharmaceutical products studied for the abovementioned disorders are targeted specifically to the NR1 subunit. Previous studies have shown that the lowest energy unoccupied molecular orbital (LUMO) can be used as a parameter to estimate the agonist and antagonist activity of the NR1subunit. Objective. Evaluate the semiemprical method CNDO for the rapid calculation of the LUMO energy with the aim of preparing a simple model for the in silico design of new pharmacological substances. Materials and methods. 168 molecules with agonist and antagonist activity in the NR1 subunit were selected. Energy of each structure was optimized and then we calculated the energy of the frontier orbital, the LogP, total energy, capacity of forming hydrogen bonds, binding energy, and dipolar moment. Results. We demonstrate that LUMO energy is enough for discriminating agonist and antagonist molecules of the NR1 subunit and that the CNDO method evaluates these properties in a rapid and efficient way. Conclusions. The CNDO method facilitates a rapid calculation, enabling a future development of effective procedures for the characterization of potential pharmacological substances acting on this particular site. Classically it was assumed that the compounds with therapeutic effect exert their action interacting with a single receptor. Nowadays it is widely recognized that the pharmacological effect of most drugs is more complex and involves a set of receptors, some associated to their positive effects and some others to the side effects and toxicity. Antipsychotic drugs are an example of effective compounds characterized by a complex pharmacological profile binding to several receptors (mainly G protein-coupled-receptors, GPCR). In this work we will present a detailed study of known antipsychotic drugs and the receptors potentially involved in their binding profile, in order to understand the molecular mechanisms of the antipsychotic pharmacologic effects. The study started with obtaining homology models for all the receptors putatively involved in the antipsychotic drugs receptorome, suitable for building consistent drug-receptor complexes. These complexes were structurally analyzed and compared using multivariate statistical methods, which in turn allowed the identification of the relationship between the pharmacological properties of the antipsychotic drugs and the structural differences in the receptor targets. The results can be exploited for the design of safer and more effective antipsychotic drugs with an optimum binding profile., Tradicionalmente se asumía que los fármacos terapéuticamente efectivos actuaban interaccionando con un único receptor.
