1. The role of ADAMTS6 and ADAMTS17 polymorphisms in susceptibility to lumbar disc herniation in Chinese Han population.

Abstract

Purpose: LDH caused by lumbar disc degeneration is associated with genetic factors. However, the role of ADAMTS6 and ADAMTS17 genes in LDH risk is still unknown. Methods: To investigate the interaction between ADAMTS6 and ADAMTS17 variants in the susceptibility of LDH, five SNPs were genotyped in 509 patients and 510 healthy individuals. The experiment used logistic regression to calculate odds ratio (OR) and 95% confidence interval (CI). Multi-factor dimensionality reduction (MDR) was chosen to evaluate impact of interaction of SNP-SNP on susceptibility to LDH. Results: ADAMTS17-rs4533267 is significantly associated with reducing risk of LDH (OR = 0.72, 95% CI = 0.57–0.90, p = 0.005). Stratified analysis indicates that ADAMTS17-rs4533267 is significantly associated with the reducing risk of LDH among participants aged ≤ 48 years old. In addition, we observed that ADAMTS6-rs2307121 was associated with increasing risk of LDH in females. MDR analysis shows that single-locus model composed by ADAMTS17-rs4533267 can be chosen as the best model for predicting susceptibility to LDH (CVC = 10/10, test accuracy = 0.543). Conclusion: ADAMTS6-rs2307121 and ADAMTS17-rs4533267 are potentially associated with LDH susceptibility. In particular, ADAMTS17-rs4533267 has a strong association with reducing risk of LDH. Abstract

BackgroundGenome-wide association studies conducted on QRS duration, an electrocardiographic measurement associated with heart failure and sudden cardiac death, have led to novel biological insights into cardiac function. However, the variants identified fall predominantly in non-coding regions and their underlying mechanisms remain unclear.ResultsHere, we identify putative functional coding variation associated with changes in the QRS interval duration by combining Illumina HumanExome BeadChip genotype data from 77,898 participants of European ancestry and 7695 of African descent in our discovery cohort, followed by replication in 111,874 individuals of European ancestry from the UK Biobank and deCODE cohorts. We identify ten novel loci, seven within coding regions, including ADAMTS6, significantly associated with QRS duration in gene-based analyses. ADAMTS6 encodes a secreted metalloprotease of currently unknown function. In vitro validation analysis shows that the QRS-associated variants lead to impaired ADAMTS6 secretion and loss-of function analysis in mice demonstrates a previously unappreciated role for ADAMTS6 in connexin 43 gap junction expression, which is essential for myocardial conduction.ConclusionsOur approach identifies novel coding and non-coding variants underlying ventricular depolarization and provides a possible mechanism for the ADAMTS6-associated conduction changes.

Abstract

ADAMTS family members control mammalian development and disease, primarily through their function as proteases, by regulation of extracellular matrix composition. Until recently, ADAMTS6 was known as one of the orphan proteinases of the nineteen-member family with a relatively unknown expression pattern and function. Emerging focus on this enzyme has started to uncover these unknowns and revealed a vast importance and requirement of ADAMTS6 in cardiovascular and musculoskeletal development. In addition, ADAMTS6 has been linked to numerous disease settings including several types of cancer. This review summarizes the necessity of ADAMTS6 during development, its role in disease and requirement for essential prospective studies to fully realize its biological implications and potential for therapeutic intervention.

5. 慢病毒载体沉默 ADAMTS6 人非小细胞肺癌稳转株的构建.

Abstract

Objective: To predict the expression of ADAMTS6 in non-small cell lung cancer(NSCLC) tissue and the prognosis of NSCLC patients via database, constract the ADAMTS6 shRNA vector and establish ADMATS6 stable knockdown cell line. Methods:The different expression of ADAMTS6 in NSCLC tissue and normal lung tissue was analyzed through the Oncomine database. The relationship between the expression level of ADAMTS6 and the prognosis of patients with NSCLC was analyzed by Kaplan-Meier Plotter database. Subsequently, the shRNA interference sequences of ADAMTS6 were designed and synthesized, and then was cloned into the pGLV3-GFP vector linearized by digestion with the restriction enzymes. The positive transformed bacteria were picked and sent for DNA sequencing. The lentiviral vector was subjected to virus packaging and then infected the NSCLC cell line NCI-H358. The stable knockdown cell line was selected through puromycin. The density of lentivirus-infected cells were observed by fluorescence microscope,and the knockdown effect of m RNA and protein levels of ADAMTS6 were detected by qRT-PCR and Western blot analysis,respectively. Results: The m RNA levels of ADAMTS6 were significantly higher in NSCLC tissue than in normal lung tissue according to the Oncomine database(P <0.001). Kaplan-Meier Plotter database analysis showed that NSCLC patients with high expression of ADAMTS6 had a poorer prognosis than patients with low expression(P<0.05). The shRNA annealing template was successfully cloned into the restriction digestion and linearized pGLV3-GFP vector. The DNA sequencing result was correct. Fluorescence observation showed that the density of lentivirus-infected cells was about 95%. The m RNA and protein levels of ADAMTS6 were successfully knocked down detected by qRT-PCR and Western blot analysis. Conclusions: The high expression of ADAMTS6 might be related to the poor clinical prognosis of NSCLC patients. We constructed the ADAMTS6 lentiviral vector and successfully established the stable knockdown cell line to lay the foundation for the future study of ADAMTS6 in NSCLC. [ABSTRACT FROM AUTHOR]

Abstract

Here, we report on a male patient with developmental delay, speech impairment, mild dysmorphic features, and borderline intellectual disability, bearing a de novo balanced t(5;6)(q11;q25.3). By combining FISH and long distance inverse PCR, we identified two genes, ADAMTS6 and ARID1B, which were disrupted at the translocation breakpoints. Due to the opposing transcriptional directions of the two genes, no fusion transcripts could be formed. ADAMTS6 on chromosome 5 encodes a zinc metalloprotease. To date, there has been no information about the substrates and the exact role of this enzyme protein. ARID1B on chromosome 6 is involved in chromatin remodeling and transcriptional activation and is known to play a role in neural development. To our knowledge, this is the fourth translocation involving ARID1B reported in association with intellectual disability. ARID1B haploinsufficiency has already been described in patients with intellectual disabilities with or without corpus callosum abnormalities, Coffin-Siris syndrome and autism (OMIM 614562 and OMIM 614556). A review of patients with ARID1B mutations reveals their broad phenotypic variability. The phenotype of the present patient is of the mildest described to date and further underscores this observation. We conclude that the most prominent and consistent clinical findings in patients with ARID1B haploinsufficiency are developmental delay, speech impairment and intellectual disability and propose that patients with unresolved genetic background and these clinical findings should be considered for ARID1B mutation screening. © 2014 Wiley Periodicals, Inc. Abstract

Objective: A Disintegrin-like Metalloproteinase with Thrombospondin Motifs (ADAMTS) proteins are kind of matrix metalloproteinase enzymes that primarily founds in the extracellular matrix (ECM). Insulin is an important anabolic hormone, which acts on many tissues. The aim of this study is to evaluate the time-dependent effects of insulin on the two functionally unknown enzyme expressions (ADAMTS6 & 19) in OUMS-27 human chondrosarcoma cell line. Methods: OUMS-27 cells were cultured in Dulbecco's modified Eagle' medium (DMEM) alone and DMEM containing 10 μg/mL insulin. The medium was changed every other day up to 11th day. Cells were harvested at 1, 3, 7, and 11th days and RNA isolation was performed at appropriate times according to study setup. The levels of RNA expression of ADAMTS6 and 19 were estimated by qRT-PCR using appropriate primers. Results: According to qRT-PCR analysis, ADAMTS6 mRNA expression was found to be decreased as early as one day after insulin application and continued up to day 11, the last day of insulin induction (p=0.008). The ratio of ADAMTS6 in insulin-applied groups was changed between 1/2 and 1/4 of control values. The changes in ADAMTS19 mRNA levels in insulin-applied cells were not statistically significant compared to control cell group. Conclusion: Our results demonstrated that insulin has a potential effect on alleviation of loss of extracellular matrix compounds by diminishing ADAMTS6 levels. To test this hypothesis and finding, more investigations are needed to recognize the real functions of orphan ADAMTS proteins. [ABSTRACT FROM AUTHOR]

Abstract

This study aimed to identify significantly altered long non-coding RNAs (lncRNAs), microRNAs (miRNAs), mRNAs, pathways in preeclampsia (PE), and to investigate their targeted relationships and biological functions. GSE96985 from Gene Expression Omnibus database was extracted, involving 3 PE and 4 normal tissues. After the differential expression analysis of miRNAs, lncRNAs, and mRNAs using the limma package, protein-protein interaction (PPI) network and module analyses were performed for differentially expressed mRNAs (dif-mRNAs). Combined with the miRanda and miRWalk tools, a regulatory relationship between dif-miRNAs and dif-mRNAs/lncRNAs (dif-mRNAs/dif-lncRNAs) was predicted. Finally, mRNA-miRNA-lncRNA regulatory network construction was performed using Cytoscape software. A total of 511 dif-mRNAs were screened in PE. The top 5 nodes in the PPI networks included up-regulated complement component 3 (C3), C-X-C motif chemokine ligand 8 (CXCL8), and fibronectin 1 (FN1). Three significant network modules were identified for dif-mRNAs. C3 and CXCL8 were identified in module A, and FN1 was identified in module C. A disintegrin and metalloproteinase with thrombospondin motifs 6 (ADAMTS6) was down-regulated by the miR-210-3p. Therefore, lnc-CTD-2383M3.1 functions as a competing endogenous RNA in ADAMTS6 expression regulation by competitively binding to miR-210-3p during the regulation process of PE. C3, CXCL8, FN1, and ADAMTS6 might be involved in the development of PE. The lnc-CTD-2383M3.1-miR-210-3p-ADAMTS6 axis might be a potential regulatory mechanism in PE.

10. Molecular mechanism of CD163 + tumor-associated macrophage (TAM)-derived exosome-induced cisplatin resistance in ovarian cancer ascites.

Abstract

Background: Studies have found that tumor-associated macrophages (TAMs) in the malignant ascites of patients with serous ovarian cancer exhibit a mixed polarization phenotype and highly variable expression of the surface marker CD163. The exosomes secreted by mesenchymal cells can be taken up by tumor cells, affecting the malignant biological behavior of them., Methods: Using reverse transcription-polymerase chain reaction (RT-PCR) to detect the genes expression of drug resistance related factors cancer stem cells (CSCs)/multidrug resistance (MDR)/epithelial-mesenchymal transition (EMT) after CD163 + TAMs exosomes in ovarian cancer ascites co-cultured with A2780 and A2780/cis-diamminedichloroplatinum (DDP). Differences of the level of miR-221-3p expression between CD163 + TAMs cells (M2) and peripheral blood mononuclear cells M0 detect by microarray screening, and bioinformatics analysis predicts that its target gene is ADAMTS6 . Western blot (WB) and immunohistochemical detection of ADAMTS6 expression level in epithelial ovarian cancer (EOC) clinical sample tissues, and analysis of the ADAMTS6 expression in ovarian cancer tissues and its correlation with clinicopathological factors. WB was used to detect the expression of AKT pathway and downstream EMT-related molecules [ SNAIL1 , ZEB1 , Vimentin ( VIM )] after co-culture of CD163 + TAMs exosomes with ovarian cancer cell lines A2780, A2780/DDP. WB detects the expression of EGFR and TGF-β1 upstream molecules of the AKT signaling pathway ( AKT - pAKT ) and downstream EMT molecules ( SNAIL1 , ZEB1 , VIM ) after overexpression of ADAMTS6 in A2780 and A2780/DDP., Results: We demonstrated that after CD163 + TAM exosomes were taken up by EOC cells, the highly expressed miR-221-3p could downregulate the level of ADAMTS6 , further activate the AKT signaling pathway, and increase the expression of EMT transcription factors SNAIL1 and ZEB1 and the mesothelial marker VIM . Decreased expression of the epithelial marker E-cadherin induced EMT, triggering a switch to a CSC-like phenotype and MDR, thereby promoting EOC cell proliferation, adhesion, migration, and resistance. Compared with benign ovarian tumors, ADAMTS6 expression was low in EOC tissues and was closely related to the clinical stage, age, and survival curve of ovarian cancer patients., Conclusions: Overexpression of ADAMTS6 reduced the IC50 of cisplatin in ovarian cancer cells. Noncoding RNAs have drawn significant attention in carcinogenesis. In this study, we identified a novel gene named nickel-related gene1 (NRG1) associated with nickel-induced cancer. By using rapid amplification of cDNA end PCR, we obtained the full length of the cDNA. The sequence was analyzed by using related bioinformatics software and comparative genomics methods. The results showed that NRG1 was located on chromosome 2q12, within intron2 of ADAMTS6, a disintegrin and metalloproteinase with thrombospondin motifs. And, NRG1 had a high level of homology (76 %) to rat LINE1 sequence RL1.3 (long interspersed middle repetitive DNA). What's more, there was no continuous open reading frame present in NRG1 sequence. Taken together, these data demonstrate that NRG1 is a novel noncoding RNA, and we predicted it may be a transposon-like gene. However, how ADAMTS6 influences cancer remains unknown. We investigated the biological function and clinical implications of ADAMTs6 in breast cancer (BC). Its functional significance in BC cell lines was confirmed by ADAMTs6 overexpression or downregulation both in vitro and in vivo studies. Enhanced ADAMTS6 expression suppressed cell migration, invasion, and tumorigenesis, whereas knockdown promoted these characteristics. The extracellular signal-regulated kinase (ERK) pathway was partially involved in ADAMTS6-mediated inhibition of BC development, and miR-221-3p was identified as a predicted target for ADAMTS6. Results from the luciferase assay confirmed that miR-221-3p directly inhibited ADAMTS6 expression by binding its 3′-untranslated region. In addition, immunohistochemistry data from specimens from 182 BC patients showed that high ADAMTS6 expression was significantly correlated with favorable disease-free survival (DFS, p = 0.045). Subgroup analysis of patients with ER positive, PR positive or HER-2 negative tumors revealed that high ADAMTS6 expression more strongly extended DFS compared to low expression ( p = 0.004, p = 0.009, p = 0.017). Multivariate analyses confirmed that ADAMTS6 expression was an independent risk factor for DFS ( p = 0.011). Together, these data demonstrate that ADAMTS6 inhibits tumor development by regulating the ERK pathway via binding of miR-221-3p. Thus, its expression may be a potential prognostic biomarker for BC.

Abstract

Background: Genome-wide association studies conducted on QRS duration, an electrocardiographic measurement associated with heart failure and sudden cardiac death, have led to novel biological insights into cardiac function. However, the variants identified fall predominantly in non-coding regions and their underlying mechanisms remain unclear. Results: Here, we identify putative functional coding variation associated with changes in the QRS interval duration by combining Illumina HumanExome BeadChip genotype data from 77,898 participants of European ancestry and 7695 of African descent in our discovery cohort, followed by replication in 111,874 individuals of European ancestry from the UK Biobank and deCODE cohorts. We identify ten novel loci, seven within coding regions, including ADAMTS6, significantly associated with QRS duration in gene-based analyses. ADAMTS6 encodes a secreted metalloprotease of currently unknown function. In vitro validation analysis shows that the QRS-associated variants lead to impaired ADAMTS6 secretion and loss-of function analysis in mice demonstrates a previously unappreciated role for ADAMTS6 in connexin 43 gap junction expression, which is essential for myocardial conduction. Conclusions: Our approach identifies novel coding and non-coding variants underlying ventricular depolarization and provides a possible mechanism for the ADAMTS6-associated conduction changes.

Abstract

Background/aim: The resistance to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs), such as gefitinib or erlotinib, is considered a major challenge in the treatment of patients with non-small cell lung cancer (NSCLC). Herein, we identified the critical roles of anterior gradient 2 (AGR2) in gefitinib (Gef) resistance of mutant NSCLC cells., Materials and Methods: Using datasets from a pair of NSCLC-sensitive and NSCLC-resistant cells, immunoblotting, immunofluorescence and immunohistochemistry, and cell viability assays were applied to identify the effects of AGR2., Results: AGR2 was found to be significantly over-expressed in Gef-resistant cells and was highly associated with drug resistance, proliferation, migration, and invasion of cancer cells. Moreover, AGR2 and ADAMTS6 formed a negative feedback loop in drug-resistant cells., Conclusion: Modulation of overexpression of AGR2 in mutant NSCLC cells may be an attractive therapeutic strategy for the treatment of EGFR-TKI-resistant NSCLC., (Copyright© 2020, International Institute of Anticancer Research (Dr. George J. Abstract

Objective: A Disintegrin-like Metalloproteinase with Thrombospondin Motifs (ADAMTS) proteins are kind of matrix metalloproteinase enzymes that primarily founds in the extracellular matrix (ECM). Insulin is an important anabolic hormone, which acts on many tissues. The aim of this study is to evaluate the time-dependent effects of insulin on the two functionally unknown enzyme expressions (ADAMTS6 & 19) in OUMS-27 human chondrosarcoma cell line. Methods: OUMS-27 cells were cultured in Dulbecco\'s modified Eagle\' medium (DMEM) alone and DMEM containing 10 μg/mL insulin. The medium was changed every other day up to 11th day. Cells were harvested at 1, 3, 7, and 11th days and RNA isolation was performed at appropriate times according to study setup. The levels of RNA expression of ADAMTS6 and 19 were estimated by qRT-PCR using appropriate primers. Results: According to qRT-PCR analysis, ADAMTS6 mRNA expression was found to be decreased as early as one day after insulin application and continued up to day 11, the last day of insulin induction (p=0.008). The ratio of ADAMTS6 in insulin-applied groups was changed between 1/2 and 1/4 of control values. The changes in ADAMTS19 mRNA levels in insulin-applied cells were not statistically significant compared to control cell group. Conclusion: Our results demonstrated that insulin has a potential effect on alleviation of loss of extracellular matrix compounds by diminishing ADAMTS6 levels. To test this hypothesis and finding, more investigations are needed to recognize the real functions of orphan ADAMTS proteins., Amaç: A Disintegrin-like Metalloproteinase with Thrombospondin Motifs (ADAMTS) proteinleri ekstraselüler matrikste (ECM) bulunan bir çeşit matriks metalloproteinaz enzimidir. İnsulin birçok dokuda etki gösteren önemli bir anabolik hormondur. Bu çalışmanın amacı OUMS-27 insan kondrosarkom hücre kültüründe insülinin, fonksiyonları tam olarak bilinmeyen ADAMTS enzimlerinden ADAMTS6 ve 19 ekspresyonu üzerine zamana bağımlı olarak etkilerini değerlendirmek ve insülinin anabolik etkilerinden dolayı ADAMTS ekspresyonunu azaltma hipotezini test etmektir. Yöntemler: OUMS-27 hücrelerinin, 10 μg/mL insulin içeren ve içermeyen Dulbecco\'s modified Eagle besiyerinde (DMEM) kültürü yapıldı. 11. güne kadar iki günde bir besiyeri ile birlikte insülin takviyesi yapıldı. Hücreler 1, 3, 7 ve 11. günlerde harvest edilerek uygun zamanlarda RNA izolasyonu yapıldı. ADAMTS6 ve 19 RNA ekspresyonları uygun primerler kullanılarak qRT-PCR yöntemi ile ölçüldü. Bulgular: qRT-PCR cihazının verilerine göre, insülin uygulanmasından 1 gün sonra başlamak üzere deneyin son günü olan 11. güne kadar ADAMTS6\'nın mRNA düzeyinde ciddi bir azalma meydana geldi (p=0.008). İnsülin uygulanan gruplarda ADAMTS6 oranı kontrol grubuna kıyasla 1/2 ve 1/4 aralığında değişti. İnsülin verilen hücre grubunda ADAMTS19 mRNA düzeyindeki değişiklikler ise kontrol grubuna kıyasla istatistiksel olarak anlamsızdı. Sonuç: Sonuçlarımız, insülinin ADAMTS6 düzeylerini azaltarak ekstrasellüler matriks bileşenlerinin kaybının telafi edilmesinde potansiyel bir etkisinin olabileceğini gösterdi. Bu hipotez ve bulgunun anlaşılması için yetim ADAMTS proteinlerinin gerçek fonksiyonlarının inceleneceği çok sayıda araştırma yapılmasına ihtiyaç vardır.

Abstract

A disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) family is involved in tumor development. However, how ADAMTS6 influences cancer remains unknown. We investigated the biological function and clinical implications of ADAMTs6 in breast cancer (BC). Its functional significance in BC cell lines was confirmed by ADAMTs6 overexpression or downregulation both in vitro and in vivo studies. Enhanced ADAMTS6 expression suppressed cell migration, invasion, and tumorigenesis, whereas knockdown promoted these characteristics. The extracellular signal-regulated kinase (ERK) pathway was partially involved in ADAMTS6-mediated inhibition of BC development, and miR-221-3p was identified as a predicted target for ADAMTS6. Results from the luciferase assay confirmed that miR-221-3p directly inhibited ADAMTS6 expression by binding its 3'-untranslated region. In addition, immunohistochemistry data from specimens from 182 BC patients showed that high ADAMTS6 expression was significantly correlated with favorable disease-free survival (DFS, p = 0.045). Subgroup analysis of patients with ER positive, PR positive or HER-2 negative tumors revealed that high ADAMTS6 expression more strongly extended DFS compared to low expression (p = 0.004, p = 0.009, p = 0.017). Multivariate analyses confirmed that ADAMTS6 expression was an independent risk factor for DFS (p = 0.011). Together, these data demonstrate that ADAMTS6 inhibits tumor development by regulating the ERK pathway via binding of miR-221-3p.
