2. Characterization of a cryptic, pyrroloquinoline quinone-dependent dehydrogenase of Gluconobacter sp. strain CHM43

ABSTRACT: We characterized the pyrroloquinoline quinone (PQQ)-dependent dehydrogenase 9 (PQQ-DH9) of Gluconobacter sp. strain CHM43, which is a homolog of PQQ-dependent glycerol dehydrogenase (GLDH). We used a plasmid construct to express PQQ-DH9. The expression host was a derivative strain of CHM43, which lacked the genes for GLDH and the membrane-bound alcohol dehydrogenase and consequently had minimal ability to oxidize primary and secondary alcohols. The membranes of the transformant exhibited considerable d-arabitol dehydrogenase activity, whereas the reference strain did not, even if it had PQQ-DH9-encoding genes in the chromosome and harbored the empty vector. This suggests that PQQ-DH9 is not expressed in the genome. The activities of the membranes containing PQQ-DH9 and GLDH suggested that similar to GLDH, PQQ-DH9 oxidized a wide variety of secondary alcohols but had higher Michaelis constants than GLDH with regard to linear substrates such as glycerol. Cyclic substrates such as cis-1,2-cyclohexanediol were readily oxidized by PQQ-DH9.

Abstract: Gluconobacter frateurii: CHM 43 have D-mannitol dehydrogenase (quinoprotein glycerol dehydrogenase) and flavoprotein D-fructose dehydrogenase in the membranes. When the two enzymes are functional, D-mannitol is converted to 5-keto-D-fructose with 65% yield when cultivated on D-mannitol. 5-Keto-D-fructose production with almost 100% yield was realized with the resting cells. The method proposed here should give a smart strategy for 5-keto-D-fructose production.

Upstream of the gene for flavin adenine dinucleotide (FAD)-dependent D-sorbitol dehydrogenase (SLDH), sldSLC, a putative transcriptional regulator was found in Gluconobacter frateurii THD32 (NBRC 101656). In this study, the whole sboR gene and the adjacent gene, sboA, were cloned and analyzed, sboR mutation did not affect FAD-SLDH activity in the membrane fractions. The SboA enzyme expressed and purified from an Escherichia coli transformant showed NADPH-dependent L-sorbose reductase (NADPH-SR) activity, and the enzyme was different from the NADPH-SR previously reported for Gluconobacter suboxydans IFO 3291 in molecular size and amino acid sequence. A mutant defective in sboA showed significantly reduced growth on L-sorbose, indicating that the SboA enzyme is required for efficient growth on L-sorbose. The sboR mutant grew on L-sorbose even better than the wild-type strain did, and higher NADPH-SR activity was detected in cytoplasm fractions. Reverse transcription-PCR experiments indicated that sboRA comprises an operon. These data suggest that sboR is involved in the repression of sboA, but not in the induction of sldSLC, on o-sorbitol and that another activator is required for the induction of these genes by D-sorbitol or L-sorbose.

35. Characterization and spontaneous mutation of a novel gene, polE, involved in pellicle formation in Acetobacter tropicalis SKU 1100

Acetobacter tropicalis SKU 1100 produces a pellicle polysaccharide, consisting of galactose, glucose and rhamnose, which attaches to the cell surface. This strain forms two types of colony on agar plates: a rough-surfaced colony (R strain) and a mucoid smooth-surfaced colony (S strain). The R strain forms a pellicle, allowing it to float on the medium surface in static culture, while the S strain does not. The pellicle is an assemblage of cells which are tightly associated with capsular polysaccharides (CPS) on the cell surface. In this study, a gene required for pellicle formation by the R strain was investigated by transposon mutagenesis using Tn 10. The resulting mutant, designated [Pel.sup.-] , has a smooth-surfaced colony and a defect in pellicle formation, as for the S strain. The mutant produced polysaccharide which was instead secreted into the culture medium as extracellular polysaccharide (EPS). An ORF was identified at the Tn 10 insertion site, designated polE, upstream of which polABCD genes were also found. The deduced amino acid sequences of polABCD showed a high level of homology to those of rfbBACD which are involved in dTDP-rhamnose synthesis, whereas polE had a relatively low level of homology to glycosyltransferase. In this study a polB (rfbA) disruptant was also prepared, which lacked both CPS and EPS production. A plasmid harbouring the polE or polB genes could restore pellicle formation in the [Pel.sup.-] mutant and S strains, and in the [DELTA]polB mutant, respectively. Thus both polE and polB are evidently involved in pellicle formation, most likely by anchoring polysaccharide to the cell surface and through the production of dTDP-rhamnose, respectively. The [Pel.sup.-] and [DELTA]polB mutants were unable to grow in static culture and became more sensitive to acetic acid due to the loss of pellicle formation. Additionally, this study identified the mutation sites of several S strains which were spontaneously isolated from the original culture and found them to be concentrated in a sequence of 7 C residues in the coding sequence of polE, with the deletion or addition of a single C nucleotide.

Acetic acid bacteria are obligate aerobes able to oxidize ethanol, sugar alcohols, and sugars into their corresponding acids. Among them, Acetobacter and Gluconacetobacter species have very high ethanol oxidation capacity, leading to accumulation of vast amounts of acetic acid outside the cell. Since these bacteria are able to grow in media with high concentrations of acetic acid, they must possess a specific mechanism such as an efflux pump by which they can resist the toxic effects of acetic acid. In this study, the efflux pump of Acetobacter aceti IFO 3283 was examined using intact cells and membrane vesicles. The accumulation of acetic acid/acetate in intact cells was increased by the addition of a proton uncoupler and/or cyanide, suggesting the presence of an energy-dependent efflux system. To confirm this, right-side-out and inside-out membrane vesicles were prepared from A. aceti IFO 3283, and the accumulation of acetic acid/acetate in the vesicles was examined. Upon the addition of a respiratory substrate, the accumulation of acetic acid/acetate in the right-side-out vesicles was largely decreased, while its accumulation was very much increased in the inside-out vesicles. These respiration-dependent phenomena observed in both types of membrane vesicles were all sensitive to a proton uncoupler. Acetic acid/acetate uptake in the inside-out membrane vesicles was dependent not on ATP but on the proton motive force. Furthermore, uptake was shown to be rather specific for acetic acid and to be pH dependent, because higher uptake was observed at lower pH. Thus, A. aceti IFO 3283 possesses a proton motive force-dependent efflux pump for acetic acid.

Carbon monoxide (CO) complexes generated for copper/topa quinone (TPQ)-containing amine oxidases have been studied using spectroscopy. Carbon monoxide that is able to bind to the Cu(I) centers in a variety of copper proteins was used. For the first time it has been shown that CO does bind to the copper amine oxidases for Arthrobactor globiformix and Aspergillus niger. Evidence indicates that vibrational properties of CO bound to the Cu(I) ion are sensitively affected by the coordination structure of the Cu atom, which may be altered by the chemical/redox states of the TPQ cofactor.

Research was conducted to examine the molecular properties of azurin, a blue protein copper found in Pseudomonas putida HK5 and purified from the soluble fraction of the organism grown on n-butanol. Its interaction with type II alcohol dehydrogenase (ADH) was also investigated. Results strongly indicate that azurin acts as an electron-transfer mediator in a pyrroloquinoline quinone-dependent alcohol oxidase respiratory chain in P. putida HK5.

Con el objetivo de diseñar un biocatalizador para la reducción del contenido en L-Fenilalanina con aplicación a la producción de fórmulas para pacientes con Fenilcetonuria, PAL fue obtenida a partir de Rhodosporidium toruloides NBRC 0559. Luego, PAL fue recuperada, purificada, inmovilizada por encapsulación en membranas semipermeables y empleada en: a) reactor de columna con recirculación y b) reactor tipo tanque agitado. La bioconversión se monitoreó espectrofotométricamente (DO290) siguiendo la producción de ácido t-cinámico. La enzima obtenida evidenció una elevada actividad enzimática, gran afinidad por el sustrato y una temperatura óptima de 43C. En cuanto al empleo de PAL inmovilizada, la utilización de un reactor tipo tanque agitado resultó óptima para la reducción de Lfenilalanina dado el efecto de la agitación en la reducción del espesor de la película estanca adyacente a la membrana. El factor más influyente en la velocidad de reacción fue el espesor de membrana, alcanzando de este modo la eliminación completa de una solución 0,1 mM de L-Fenilalanina en 6 h. En conclusión, PAL inmovilizada por encapsulación en membranas celulósicas es un método viable para la reducción del contenido de L-Fenilalanina en soluciones diluidas. Este método podría extrapolarse a sustratos más complejos como lo son hidrolizados proteicos. For the design of a biocatalyst capable of reducing the L-Phenylalanine content to produce formulas for Phenylketonuria patients, PAL was obtained from Rhodosporidium toruloides NBRC 0559. PAL was produced, recovered from the culture medium, purified, immobilized by encapsulation in semipermeable membranes and used in: a) column reactor with recirculation and b) stirring tank reactor. The bioconversion was spectrophotometrically monitored (OD290) by t-cinnamic acid production. PAL demonstrated to have high activity and affinity for L-Phenylalanine, with an optimal temperature of 43C. In relation to PAL utilization, the stirring tank reactor was the most appropriate system for bioconversion because of agitation effect in the diffusion of substrate and products to the membrane. In addition, membrane wall thickness was a key factor in bioconversion rate, reaching the complete removal of 0,1 mM L-Phenylalanine in 6 h. These results indicate that purified PAL immobilized by encapsulation in semipermeable membranes can be applied for the reduction of L-Phenylalanine content in diluted solutions. Spectroscopic investigation of the structure of the binuclear sites of two terminal ubiquinol oxidases (cytochromes alpha1 and 0) of Acetobacter aceti suggests that conformational flexibility of the binuclear center in the reduced state is a common feature of the heme-copper oxidase superfamily. Raman data indicate that the formyl C=O group may make a hydrogen bond network with water molecules and conserved distal amino acid residues. This network may be important to the high affinity in cyanide binding.

The structure and behavior of inactive alcohol dehydrogenase (ADH) and the process of physiologically activating inactive ADH in the growing and resting cells of Gluconobacter suboxydans was studied. Active ADH has 10 times more enzyme reactions than inactive ADH. Interconversion between the two forms of ADH was obtained either by changing the pH of the culture medium or by incubating the resting cells in different levels of pH while exposing them to an energy source.

Pseudomonas putida can produce three different types of alcohol dehydrogenase (ADH) depending on the nature of the alcohol used as its major carbon source. The bacterium produces ADH I when grown on ethanol, ADH IIB when grown on 1-butanol and ADH IIG when grown on 1,2-propanediol. Antibodies against the enzymes could distinguish one from another.
