Até recentemente, o tratamento da hemofilia A consistia em repor fator VIII ou agentes de by-pass (se inibidor positivo) para tratar (demanda) ou evitar (profilaxia) sangramentos, baseado em infusões intravenosos várias vezes por semana. O emicizumabe surgiu como uma alternativa na profilaxia, mostrando superioridade frente aos produtos anteriores para evitar sangramentos em pessoas com hemofilia A (PcHA) sem e com inibidores. A administração é subcutânea com doses semanais a mensais. Essa mudança na efetividade e na posologia tem sido acompanhada de uma readaptação das orientações às PcHA acerca da doença e do manejo terapêutico. O objetivo deste estudo foi compreender a percepção de enfermeiros, psicólogos e assistentes sociais, aqui descritos como equipe de suporte biopsicossocial (esBPS), que atendem PcHA em diversos centros de tratamento de hemofilia do Brasil, a respeito das mudanças de mentalidade e comportamento relacionadas à introdução do emicizumabe. Este estudo exploratório e quantitativo começou em julho/2021. A partir da discussão da literatura sobre o tema, uma esBPS (n = 5) enumerou os principais pontos do suporte biopsicossocial prestado à PcHA que poderiam ser impactados com a introdução da profilaxia com emicizumabe. Em janeiro/2024, após maior experiência com a terapia, esses pontos foram mantidos e transformados em afirmativas validadas por profissionais não envolvidos com o projeto. Entre 5 e 30/07/2024, profissionais das esBPS foram convidados para participar e, mediante aceite, receberam um formulário envolvendo 26 afirmativas para serem avaliadas quanto à concordância em uma escala de Likert: discordo totalmente e discordo parcialmente (agrupados como discordo), não discordo nem concordo (neutro), e concordo parcialmente e concordo totalmente (agrupados como concordo). Uma pergunta alternativa solicitou que selecionassem o desfecho mais adequado para ser avaliado na profilaxia com emicizumabe.Dos 44 profissionais convidados, 17 (39%) completaram a enquete, com representatividade semelhante entre enfermeiros, psicólogos e assistentes sociais. A mediana de tempo de atendimento às PcHA foi 10 anos (amplitude 2-42), com média mediana de 15 atendimentos/semana (amplitude 1-57). Todos concordaram que é importante que a PcHA em uso de emicizumabe tenha autonomia e liberdade para participar da decisão da melhor terapia a ser instituída. A maior parte (94%) concordou que a frequência de consultas (rotina e urgência) foi reduzida e que um modelo alternativo (por exemplo, consultas online ) junto com o convencional seria importante para garantir a autonomia e a liberdade que o tratamento proporciona. A maioria também concordou que a equipe interdisciplinar deva elaborar novos métodos de garantir o treinamento e a compreensão pela PcHA sobre reconhecimentos de sangramentos (88%), mantendo-se o treinamento de obtenção de acesso venoso para tratar sangramentos (76%). O indicador mais importante foi qualidade de vida (65%). Finalmente, todos concordaram que a PcHA deva ser instruída quanto ao risco da associação do emicizumabe com medicamentos (por exemplo, complexo protrombínico parcialmente ativado). Em conclusão, existe uma necessidade de adaptação de modelo assistencial atual à PcHA após a introdução da profilaxia com emicizumabe.

Introdução: A legislação que normatiza a hemoterapia no Brasil estabelece que o serviço de hemoterapia deve possuir equipe profissional suficiente e adequada à necessidade e complexidade do serviço. Para as equipes de enfermagem as normativas apresentam os parâmetros mínimos e características a serem consideradas para dimensionar o quantitativo de profissionais das diferentes categorias de enfermagem. Nos serviços de hemoterapia, além dos parâmetros mínimos estabelecidos na legislação, especificidades da natureza da atividade também devem ser consideradas. O conhecimento da capacidade produtiva e da força de trabalho necessária, possibilitam o planejamento de ações e a organização do atendimento aos doadores. Objetivo: Descrever o processo para o dimensionamento de pessoal de enfermagem e capacidade de produção, por esses profissionais, nas unidades da Fundação Hemominas. Materiais e métodos: Trata-se de um estudo descritivo do tipo relato de experiência do trabalho de dimensionamento de pessoal de enfermagem e identificação da capacidade de produção das unidades da Fundação Hemominas. Resultados: No período de janeiro de 2020 a março de 2021 realizou-se o dimensionamento da força de trabalho e a análise da capacidade produtiva nos procedimentos de triagem hematológica e coleta de sangue total. Para essas análises aliou-se metodologia baseada nos mapas de atribuição por produto (MAP), proposta pela Secretaria de Planejamento e Gestão do Estado de Minas Gerais, as recomendações do Conselho Federal de Enfermagem e as características das atividades. Procedeu-se a mensuração do tempo de execução para cada atividade desenvolvida para a realização dos procedimentos de triagem hematológica e coleta de sangue total, incluindo ações como a organização do setor, a lavagem de mãos e descarte de resíduos. Obteve-se uma capacidade de atendimento de até cinco doadores por hora para o procedimento de coleta de sangue total e de até 20 triagens hematológicas por hora. Para o dimensionamento de pessoal de enfermagem considerou-se a capacidade laboral individual em uma hora, o total de horas improdutivas (absenteísmo, feriados), a jornada de trabalho de cada categoria profissional da enfermagem; padrões de desempenho dos profissionais; índice de segurança técnica e a proporção de profissionais de enfermagem de nível superior e de nível médio. Discussão: A metodologia baseada nos MAP possibilitou diagnosticar as horas produtivas de trabalho ou a capacidade laboral de cada servidor, aliada aos parâmetros estabelecidos na legislação foi possível determinar a capacidade de produção e determinar o quantitativo de pessoal de enfermagem para cada unidade da Fundação Hemominas, o que permitiu ações de planejamento, metas, organização dos serviços e de pessoal nas unidades e na rede Hemominas. Conclusão: A metodologia utilizada mostrou-se adequada para a determinação da capacidade de produção por servidor/ por hora e, juntamente com os parâmetros utilizados viabilizou o dimensionamento da equipe de enfermagem para as unidades da Fundação Hemominas, contribuindo para diferentes processos de organização da instituição.

ABSTRACT This study was conducted to estimate the dig. valine to lysine (Val:Lys) ideal ratio for meat-type quails from 15 to 35 d of age. A total of 385, 15-day-old not sexed quails, were randomly assigned to five treatments, each one seven times replicated with eleven quails per experimental unit. Experimental treatments consisted of five dig. Val:Lys ratios (0.65; 0.70; 0.75; 0.80 and 0.85). Ideal dig. Val:Lys ratio was estimated using polynomial regression and linear broken-line regression model. Significant effect was considered when p≤0.05. Feed conversion ratio was not affected by dig. Val:Lys ratios in any of the assessed phases. From 15 to 21 d of age Val:Lys ratios increased linearly feed intake, weight gain and body weight. From 15 to 28 d of age, weight gain and body weight were both optimized by the Val:Lys ratio of 0.77. A linear effect of dig. Val:Lys ratios was observed on weight gain and final body weight from 15 to 35 d of age. From 15 to 35 d of age, according to linear broken-line regression model, the dig. Val:Lys ideal ratios for optimum final body weight and weight gain were 0.77 and 0.76, respectively. Based on the results the estimated dig. Val:Lys ratio for optimum meat-type quail performance from 15 to 35 d of age is 0.77

Introduction: This study evaluates the efficacy of a group-based career intervention designed to promote employability resources among unemployed individuals conducted in face-to-face and online modalities., Methods: Employing a longitudinal quasi-experimental design, with two intervention groups and a control group, data was collected pre-, post, and two months post-intervention., Results: Conditional Latent Growth Curve analysis revealed a significant decline in employability resources over time, mitigated by both intervention groups. Human capital and professional development were key at baseline and follow-up, while social capital and networking were crucial post-intervention. Career identity self-career management, and environmental monitoring resources significantly contributed to employability post-intervention., Discussion: These findings highlight the intervention's effectiveness in counteracting the negative trend among the general population, underscoring the critical role of ongoing participation in career development activities for unemployed individuals. The forward design of biosensors that implement Boolean logic to improve detection precision primarily relies on programming genetic components to control transcriptional responses. However, cell- and gene-free nanomaterials programmed with logical functions may present lower barriers for clinical translation. Here we report the design of activity-based nanosensors that implement AND-gate logic without genetic parts via bi-labile cyclic peptides. These actuate by releasing a reporter if and only if cleaved by a specific pair of proteases. AND-gated nanosensors that detect the concomitant activity of the granzyme B protease secreted by CD8 T cells and matrix metalloproteinases overexpressed by cancer cells identify the unique condition of cytotoxic T cell killing of tumour cells. Innovation in the drying process during the roasting of cashew nut almonds has the potential to significantly improve product quality. This study aimed to investigate the drying kinetics of the almond, comparing the experimental data with the mathematical models of Fick, Page, Cavalcanti Mata, and Henderson and Pabis. The research was conducted at the Laboratory of Physical Measurements and Drying of the Academic Unit of Food Engineering at the Federal University of Campina Grande. For drying, equipment with high air velocity and precise temperature control was used, operating at 120, 140, 160, and 180 °C, with an air velocity of 3 m s⁻ 1 . Based on the drying kinetics parameters, the activation energy for each mathematical model was determined, which indicates the energy required to break the bonds that maintain water in the product. The thermodynamic properties of the process were also determined, which are essential to help understand the drying behavior of the fruit, optimize energy efficiency and the quality of the final product. The Fick model, using six terms in the series and considering the cylindrical shape of the almond, along with the other models analyzed, showed a good fit for the experimental data, as evidenced by the R 2 values obtained. Deep-UV microscopy enables high-resolution, label-free molecular imaging by leveraging biomolecular absorption properties in the UV spectrum. Recent advances in UV-imaging hardware have renewed interest in this technique for quantitative live cell imaging applications. However, UV-induced photodamage remains a concern for longitudinal dynamic imaging studies. Here, we quantify UV phototoxicity with several cell types at notable UV wavelengths. We find that the fluence required for cell death via UV phototoxicity with continuous UV exposure varies with cell type and wavelength from ∼0.5µJ/µm 2 to 2µJ/µm 2 , but is independent of typical illumination power/radiant flux of UV microscopy (e.g., 0.1-20 nW/µm 2 ). We also show results from fractionation studies that reveal cell repair following UV exposure, which increases the tolerance to UV radiation by a factor of 2 or more, depending on the fractionation paradigm. Results further show that UV tolerance exceeds ANSI guidelines for maximum permissible exposure. Finally, we calculate imaging limits for a typical application of UV microscopy, such as hematology analysis. Together, this work provides UV fluence thresholds that can serve as guidelines for nondestructive, longitudinal, and dynamic deep-UV microscopy experiments., Competing Interests: Francisco E. Sensorimotor synchronization (SMS) is the mainly specifically human ability to move in sync with a periodic external stimulus, as in keeping pace with music. The most common experimental paradigm to study its largely unknown underlying mechanism is the paced finger-tapping task, where a participant taps to a periodic sequence of brief stimuli. Contrary to reaction time, this task involves temporal prediction because the participant needs to trigger the motor action in advance for the tap and the stimulus to occur simultaneously, then an error-correction mechanism takes past performance as input to adjust the following prediction. In a different, simpler task, it has been shown that exposure to a distribution of individual temporal intervals creates a "temporal context" that can bias the estimation/production of a single target interval. As temporal estimation and production are also involved in SMS, we asked whether a paced finger-tapping task with period perturbations would show any time-related context effect. In this work we show that a perturbation context can indeed be generated by exposure to period perturbations during paced finger tapping, affecting the shape and size of the resynchronization curve. Response asymmetry is also affected, thus evidencing an interplay between context and intrinsic nonlinearities of the correction mechanism. The ST6GAL1 sialyltransferase is overexpressed in multiple cancers, including pancreatic ductal adenocarcinoma (PDAC). ST6GAL1 adds an α2-6-linked sialic acid to N-glycosylated membrane receptors, which consequently modulates receptor structure and function. While many studies have investigated the effects of ST6GAL1 on cell phenotype, there is a dearth of knowledge regarding mechanisms that regulate ST6GAL1 expression. In the current study, we evaluated the regulation of ST6GAL1 by two pro-inflammatory cytokines, IL-1β and IL-6, which are abundant within the PDAC tumor microenvironment. Cytokine activity was monitored using the Suit-2 PDAC cell line and two Suit-2-derived metastatic subclones, S2-013 and S2-LM7AA. For all three cell models, treatment with IL-1β or IL-6 increased the expression of ST6GAL1 protein and mRNA. Specifically, IL-1β and IL-6 induced expression of the ST6GAL1 YZ mRNA isoform, which is driven by the P3 promoter. The ST6GAL1 H and X isoforms were not detected. Promoter reporter assays confirmed that IL-1β and IL-6 activated transcription from the P3 promoter. We then examined downstream signaling mechanisms. IL-1β is known to signal through the NFκB transcription factor, whereas IL-6 signals through the STAT3 transcription factor. CUT&RUN experiments revealed that IL-1β promoted the binding of NFκB to the ST6GAL1 P3 promoter, and IL-6 induced the binding of STAT3 to the P3 promoter. Finally, we determined that inhibitors of NFκB and STAT3 blocked the upregulation of ST6GAL1 stimulated by IL-1β and IL-6, respectively. Obesity is a multifactorial disease that is difficult to control worldwide. Although nuts are recognized health foods, the application of food in obesity management is unclear. We systematically reviewed the literature and performed a meta-analysis to evaluate if nut consumption favors people on energy restriction (ER) dietary interventions. Four databases were used to search for eligible articles in May 2024. This review was conducted according to the PRISMA guide, and the bias risk of papers was evaluated. For the meta-analysis, we extracted the endpoint values of the group's variables and estimated the effect sizes by the random-effects model. Sixteen and ten articles were included in the systematic review and meta-analysis, respectively. Almonds were evaluated in the majority of studies ( n = 6). The consumption of nuts (28 to 84 g/d, 4 to 72 months) included in ER (-250 to 1000 kcal/d) did not differently affect anthropometry (weight loss, BMI, waist and hip circumferences), body composition (fat mass, fat-free mass, or lean mass), markers of glucose (glycemia and insulinemia), lipid metabolism (total cholesterol, HDL-c, LDL-c, LDL-c/HDL-c, or triglycerides), and systolic and diastolic blood pressure. In most analyses, stratifying studies by type of nut or intervention time did not present different results in the meta-analysis. As there are few studies, in addition to great methodological variability, more high-quality trials are needed to confirm these results. The PROSPERO registration number is CRD42023444878.

Background: Obesity is associated with low-grade inflammation and increased intestinal permeability (IP). The Brazil nut (BN) (Bertholletia excelsa H.B.K.) appears to be a promising dietary intervention to control inflammation by enhancing antioxidant defenses., Objectives: We aimed to assess the effect of daily BN consumption on inflammatory biomarkers and IP in the context of an energy-restricted intervention. Furthermore, we evaluated the correlation between the changes in these inflammatory markers and the changes in serum selenium and IP., Methods: In this 8-wk nonrandomized controlled trial, 56 women with overweight or obesity were allocated into 2 groups, both following an energy-restricted diet (-500 kcal/d). The control group (CO) consumed a nut-free diet, while the BN group consumed 8 g BN/d, providing 347.2 μg selenium (Se). Inflammatory cytokines were analyzed in plasma and Se in serum. IP was assessed using the lactulose/mannitol test (LM ratio)., Results: Forty-six women completed the intervention. Both groups achieved similar energy restriction (CO Δ= -253.7 ± 169.4 kcal/d; BN Δ= -265.8 ± 141.8 kcal/d) and weight loss (CO Δ= -2.5 ± 0.5 kg; BN Δ= -3.5 ± 0.5 kg). The BN group showed lower values of C-reactive protein, tumor necrosis factor, interleukin (IL)1-β, IL-8, percentage lactulose excretion, and LM ratio than the CO group. Additionally, changes in serum Se concentration were predictive of changes in IL-8 concentration (β: -0.054; adjusted R 2 : 0.100; 95% confidence interval [CI]: -0.100; -0.007; P = 0.025), and changes in IL-8 were predictive of changes in the LM ratio (β: 0.006; adjusted R 2 : 0.101; 95% CI: 0.001, 0.011; P = 0.024)., Conclusions: Regular intake of BNs can be a promising complementary dietary strategy for controlling low-grade inflammation and improving IP in women with overweight/obesity undergoing energy-restricted treatment. However, the effects of BNs seem to be Se status-dependent. Aim: To evaluate the urinary biomarkers related to sepsis in preterm newborns (NBs) and to investigate the predictive capacity of these biomarkers for a longer hospital stay. Methods: Serum and urine were collected from 27 healthy NBs, 24 NBs with neonatal infection without sepsis and 11 NBs with sepsis for the measurement of sindecan-1, lipocalin associated with urinary neutrophil gelatinase (uNGAL), urinary cystatin-C (uCysC) and urinary kidney injury molecule-1. Results: Levels of uNGAL and urinary cystatin-C were elevated in NBs with sepsis and neonatal infection, and uNGAL was significant predictor of hospital stay longer than 30 days (odds ratio: 1.052; 95% CI: 1.012-1.093; p  = 0.01). Conclusion: uNGAL was associated with sepsis in preterm NBs and was useful to predict extended hospital stay.

Immune checkpoint inhibitors (ICIs) represent new therapeutic candidates against glioblastoma multiforme (GBM); however, their efficacy is clinically limited due to both local and systemic immunosuppressive environments. Hence, therapeutic approaches that stimulate local and systemic immune environments can improve the efficacy of ICIs. Here, we report an adoptive cell therapy employing neutrophils (NE) that are activated via surface attachment of drug-free disk-shaped backpacks, termed Cyto-Adhesive Micro-Patches (CAMPs) for treating GBM. CAMP-adhered neutrophils (NE/CAMPs) significantly improved the efficacy of an anti-PD1 antibody (aPD-1) in a subcutaneous murine GBM model (GL261). A combination of NE/CAMPs and aPD-1 completely regressed subcutaneous GL261 tumors in mice. The efficacy of NE/CAMPs against GBM was also tested in an orthotopic GL261 model. Neutrophil's ability to migrate into the brain was not affected by CAMP attachment, and intracerebral NE/CAMP accumulation was observed in mice-bearing orthotopic GBM. The combination treatment of NE/CAMPs and aPD-1 activated systemic immune responses mediated by T cells and showed improved therapeutic responses compared with aPD-1 alone in the orthotopic GBM model. Toxoplasma gondii is a food-borne zoonotic parasite widespread in a variety of hosts, including humans. With a majority of infections in Europe estimated to be meat-borne, pork, as one of the most consumed meats worldwide, represents a potential risk for consumers. Therefore, we aimed to investigate the progress of T. gondii infection and tissue tropism in experimentally infected pigs, using different T. gondii isolates and infectious stages, i.e. tissue cysts or oocysts. Twenty-four pigs were allocated to treatment in four groups of six, with each group inoculated orally with an estimated low dose of either 400 oocysts or 10 tissue cysts of two European T. gondii isolates, a type II and a type III isolate. The majority of pigs seroconverted two weeks post-inoculation. Pigs infected with the type III isolate had significantly higher levels of anti-T. gondii antibodies compared to those infected with the type II isolate. Histopathological exams revealed reactive hyperplasia of the lymphatic tissue of all pigs. Additionally, a selected set of nine tissues was collected during necropsy at 50 dpi from each of the remaining 22 pigs for T. gondii DNA detection by quantitative real-time PCR. A positive result was obtained in 29.8 % (59/139) of tested tissues. The brain was identified as the most frequently positive tissue in 63.6 % (14/22) of the animals. In contrast, liver samples tested negative in all animals. The highest mean parasite load, calculated by interpolating the average Cq values on the standard curve made of ten-fold serial dilutions of the genomic DNA, corresponding to 10 0 to 10 4 tachyzoites/µL, was observed in shoulder musculature with an estimated concentration of 84.4 [0.0-442.5] parasites per gram of tissue. The study highlights the variability in clinical signs and tissue distribution of T. gondii in pigs based on the combination of parasite stages and strains, with type III isolates, particularly oocysts, causing a stronger antibody response and higher tissue parasite burden. Background: Exploring the value of baseline and early 18 F-FDG PET/CT evaluations in prediction PFS in ER+/HER2- metastatic breast cancer patients treated with a cyclin-dependent kinase inhibitor in combination with an endocrine therapy., Methods: Sixty-six consecutive breast cancer patients who underwent a pre-therapeutic 18 F-FDG PET/CT and a second PET/CT within the first 6 months of treatment were retrospectively included. Metabolic tumour volume (MTV) and total lesion glycolysis (TLG) and D max , which represents tumour dissemination and is defined as the distance between the two most distant lesions, were computed. The variation in these parameters between baseline and early evaluation PET as well as therapeutic evaluation using PERCIST were assessed as prognosticators of PFS at 18 months., Results: The median follow-up was equal to 22.5 months. Thirty progressions occurred (45.4%). The average time to event was 17.8 ± 10.4 months. At baseline, D max was the only predictive metabolic parameter. Patients with a baseline D max ≤ 18.10 cm had a significantly better 18 m-PFS survival than the others: 69.2% (7.7%) versus 36.7% (8.8%), p = 0.017. There was no association between PERCIST evaluation and 18 m-PFS status (p = 0.149) and there was no difference in 18 m-PFS status between patients classified as complete, partial metabolic responders or having stable metabolic disease., Conclusion: Disease spread at baseline PET, as assessed by D max , is predictive of an event occurring within 18 months. Objective: To assess maternal mortality (MM) in Brazilian Black, Pardo, and White women., Methods: We evaluated the maternal mortality rate (MMR) using data from the Brazilian Ministry of Health public databases from 2017 to 2022. We compared MMR among Black, Pardo, and White women according to the region of the country, age, and cause. For statistical analysis, the Q2 test prevalence ratio (PR) and confidence interval (CI) were calculated., Results: From 2017 to 2022, the general MMR was 68.0/100,000 live births (LB). The MMR was almost twice as high among Black women compared to White (125.81 vs 64.15, PR = 1.96, 95%CI:1.84-2.08) and Pardo women (125.8 vs 64.0, PR = 1.96, 95%CI: 1.85-2.09). MMR was higher among Black women in all geographical regions, and the Southeast region reached the highest difference among Black and White women (115.5 versus 60.8, PR = 2.48, 95%CI: 2.03-3.03). During the covid-19 pandemic, MMR increased in all groups of women (Black 144.1, Pardo 74.8 and White 80.5/100.000 LB), and the differences between Black and White (PR = 1.79, 95%CI: 1.64-1.95) and Black and Pardo (PR = 1.92, 95%CI: 1.77-2.09) remained. MMR was significantly higher among Black women than among White or Pardo women in all age ranges and for all causes., Conclusion: Black women presented higher MMR in all years, in all geographic regions, age groups, and causes. In Brazil, Black skin color is a key MM determinant. Reducing MM requires reducing racial disparities.

Aim: This systematic review and meta-analysis evaluated theobromine's (Theobroma cacao) potential in remineralizing white spot lesions in dental enamel. Methods: This study is reported according to the PRISMA checklist and was registered in PROSPERO (CRD42023414371). In vitro tests that evaluated the remineralizing potential of theobromine compared to fluoride ion after demineralization for the formation of white spot lesions on enamel were selected, with no limitation on the year of publication. Electronic searches were performed in PubMed/MEDLINE, Scopus, and Web of Science by two independent researchers. Thirty articles were received of which six were selected according to the inclusion criteria., Results: The selected studies evaluated the Enamel Surface Microhardness (SMH), Vickers or Knoop, before and after treatment with theobromine and fluoride solutions. For the SMH Vickers, there were no differences between groups at baseline (p=1.00; mean difference: -0.00; CI: -11.36 to 11.36) and after treatment (p=0.51; mean difference: 4.12; CI: -8.16 to 16.41). The results of SMH Knoop showed differences between groups at baseline, favoring the experimental group (p=0.002; mean difference: 9.84; CI: 3.65 to 16.02) and after treatment favoring the control group (p=0.00001; mean difference: -5.45; CI: -7.62 to -3.27)., Conclusion: The use of theobromine increases the microhardness of dental enamel subjected to a demineralization process, thus being effective in the remineralization of this tissue with success equivalent to that obtained with the use of fluoride., (©Operative Dentistry, 2024.)

Background: Considering that beliefs may be assessed and changed, inventories measuring employability and career beliefs may be of utmost importance for career interventions., Objective: This study introduces the psychometric properties of a brief version of the Employability and Career Beliefs Inventory (ECBI) in a sample of unemployed persons., Methods and Results: Altogether, 2023 unemployed persons aged from 18 to 66 years old and living in Southern Portugal participated in an online survey. The ECBI's original internal structure was tested and did not fit the data. Exploratory and confirmatory factor analyses were implemented, and a three-factor solution was retained. The three factors discriminate three types of beliefs named growth, pessimism, and flexibility. Measurement invariance models identified scalar equivalence across gender and educational degree, and metric invariance across age. All items fit the graded response model's parameters. The growth and flexibility subscales were less effective in the assessment of low latent trait levels, whereas the opposite was observed with the pessimism subscale. Internal consistency is good yet discrimination between factors is questionable. Correlations to career decision-making self-efficacy evidence validity based on the relations to other constructs., Conclusion: Despite the limitations, the brief version of the ECBI proposed in this study is ready for further use and development among unemployed persons., (© 2024. This article aims to map the global scientific production on social or cultural representations and breast cancer in Public Health and discuss how it is presented in the literature. We conducted a scoping review guided by the question: "How are cultural or social representations in the context of breast cancer described in the global scientific Public Health production?". We searched for works in five scientific literature sources and included 45 studies. The analytical process followed the content analysis technique in the thematic modality. The analyzed collection can be thematized into the following categories: (1) Compromised body image and interactions, (2) Spirituality, (3) Loss of control over life, (4) Going on with life, and (5) Association with ethnic-racial issues. Despite advances in biomedicine, we observed that representations of breast cancer still have metaphors associated with cancer in the last century. We conclude that, among other aspects, care for women with breast cancer cannot be guided only by biomedical and epidemiological approaches since this disease is traversed by knowledge that competes with these approaches.

Objective. Traditional quantification of fluorescence signals, such asΔF/F, relies on ratiometric measures that necessitate a baseline for comparison, limiting their applicability in dynamic analyses. Our goal here is to develop a baseline-independent method for analyzing fluorescence data that fully exploits temporal dynamics to introduce a novel approach for dynamical super-resolution analysis, including in subcellular resolution. Approach. We introduce ARES (Autoregressive RESiduals), a novel method that leverages the temporal aspect of fluorescence signals. By focusing on the quantification of residuals following linear autoregression, ARES obviates the need for a predefined baseline, enabling a more nuanced analysis of signal dynamics. Main result. We delineate the foundational attributes of ARES, illustrating its capability to enhance both spatial and temporal resolution of calcium fluorescence activity beyond the conventional ratiometric measure (ΔF/F). Additionally, we demonstrate ARES's utility in elucidating intracellular calcium dynamics through the detailed observation of calcium wave propagation within a dendrite. Significance. ARES stands out as a robust and precise tool for the quantification of fluorescence signals, adept at analyzing both spontaneous and evoked calcium dynamics. The aim of this article is to present the state of the art, in the field of public health, on cis homoparental reproduction, from 28 studies addressing barriers to reproduction by homoparental couples for legal, ethical, technical or economic reasons, in addition to prejudice and discrimination. Six studies addressed facilitators, such as receptiveness in services, availability of conception and contraceptive methods and training of health professionals. The results show that the discussion has focused more on the barriers than on the facilitating factors. This may indicate a continuing need to problematise the hegemonic model of a heterosexual, nuclear family.

Imaging flow cytometry (IFC) combines flow cytometry and fluorescence microscopy to enable high-throughput, multiparametric single-cell analysis with rich spatial details. However, current IFC techniques remain limited in their ability to reveal subcellular information with a high 3D resolution, throughput, sensitivity, and instrumental simplicity. In this study, we introduce a light-field flow cytometer (LFC), an IFC system capable of high-content, single-shot, and multi-color acquisition of up to 5,750 cells per second with a near-diffraction-limited resolution of 400-600 nm in all three dimensions. The LFC system integrates optical, microfluidic, and computational strategies to facilitate the volumetric visualization of various 3D subcellular characteristics through convenient access to commonly used epi-fluorescence platforms. We demonstrate the effectiveness of LFC in assaying, analyzing, and enumerating intricate subcellular morphology, function, and heterogeneity using various phantoms and biological specimens. Aim: To synthesize HER2 aptamer-conjugated iron oxide nanoparticles with a coating of poly(2-(dimethylamino) ethyl methacrylate)-poly(2-methacryloyloxyethylphosphorylcholine) block copolymer (IONPPPs). Methods: Characterization covered molecular structure, chemical composition, thermal stability, magnetic characteristics, aptamer interaction, crystalline nature and microscopic features. Subsequent investigations focused on IONPPPs for in vitro cancer cell identification. Results: Results demonstrated high biocompatibility of the diblock copolymer with no significant toxicity up to 150 μg/ml. The facile coating process yielded the IONPP complex, featuring a 13.27 nm metal core and a 3.10 nm polymer coating. Functionalized with a HER2-targeting DNA aptamer, IONPPP enhanced recognition in HER2-amplified SKBR3 cells via magnetization separation. Conclusion: These findings underscore IONPPP's potential in cancer research and clinical applications, showcasing diagnostic efficacy and HER2 protein targeting in a proof-of-concept approach.

Background/aims: Bladder cancer is considered one of the most aggressive neoplasms due to its recurrence and progression profile, and even with the improvement in diagnosis and treatment methods, the mortality rate has not shown a declining trend in recent decades. From this perspective, the search and development of more effective and safer therapeutic alternatives are necessary. Phytochemicals are excellent sources of active principles with therapeutic potential. [6]-Shogaol is a phenolic compound extracted from the ginger rhizomes that has shown antitumor effects in a wide variety of cancer models. However, there is no record in the literature of studies reporting these effects in models of bladder cancer. Thus, this study aimed to investigate the in vitro cytotoxic and pro-apoptotic potential of [6]-Shogaol against murine bladder cancer urothelial cells (MB49)., Methods: The cytotoxic effects of [6]-Shogaol on cell viability (MTT method), cell morphology (light microscopy), alteration of proliferative processes (clonogenic assay), oxidative stress pathway (levels of reactive oxygen species) and the induction of apoptotic events (flow cytometry and high-resolution epifluorescence imaging) were evaluated in murine urothelial bladder cancer cell lines (MB49), relative to non-tumor murine fibroblasts (L929)., Results: The results showed that [6]-Shogaol was able to induce concentration-dependent cytotoxic effects, which compromised cell viability, exhibiting an inhibitory concentration of 50% of cells (IC50) of 146.8 µM for MB49 tumor cells and 236.0 µM for L929 non-tumor fibroblasts. In addition to inhibiting and altering the proliferative processes if colony formation, it presented pro-apoptotic activity identified through a quantitative analysis and the observation of apoptotic phenotypes, events apparently mediated by the induction of nuclear fragmentation., Conclusion: The data presented suggest that [6]-Shogaol has a higher concentration-dependent cytotoxic and apoptosis-inducing potential in MB49 cells than in L929 fibroblasts. Optimizing a sustained-release drug delivery system for the treatment of cystic fibrosis (CF) is crucial for decreasing the dosing frequency and improving patients' compliance with the treatment regimen. In the current work, we developed an injectable poly(D,L-lactide-co-glycolide) (PLGA) microparticle formulation loaded with ivacaftor, a cystic fibrosis transmembrane conductance regulator (CFTR) potentiator that increases the open probability of the CFTR anion channel, using a single emulsion solvent evaporation technique. We aimed to study the effect of different parameters on the characteristics of the prepared formulations to select an optimized microparticle formulation to be used in an in vivo pharmacokinetic study in mice. First, a suite of ivacaftor-loaded microparticles were prepared using different formulation parameters in order to study the effect of varying these parameters on microparticle size, morphology, drug loading, encapsulation efficiency, and in vitro release profiles. Prepared microparticles were spherical with diameters ranging from 1.91-6.93 µm, percent drug loading (% DL) of 3.91-10.3%, percent encapsulation efficiencies (% EE) of 26.6-100%, and an overall slow cumulative release profile. We selected the formulation that demonstrated optimal combined % DL and % EE values (8.25 and 90.7%, respectively) for further studies. These microparticles had an average particle size of 6.83 µm and a slow tri-phasic in vitro release profile (up to 6 weeks). In vivo pharmacokinetic studies in mice showed that the subcutaneously injected microparticles resulted in steady plasma levels of ivacaftor over a period of 28 days, and a 6-fold increase in AUC 0 - t (71.6 µg/mL*h) compared to the intravenously injected soluble ivacaftor (12.3 µg/mL*h). This study aimed to estimate the prevalence of multimorbidity in Brazilian individuals and its association with sociodemographic and lifestyle factors. This cross-sectional study used data from the National Health Survey conducted in 2019 with 88,531 Brazilian adults Multimorbidity is the presence of two or more non-communicable chronic diseases. Associated factors were assessed by calculating the prevalence ratio (PR) obtained using Poisson regression with robust variance. Multimorbidity was estimated in 29.9% (95%CI: 29.33% to 30.48%) of Brazilian individuals. In the multiple models, the prevalence was high in women (PR: 1.37; 95%CI: 1.32 to 1.42), individuals over 60 years (PR: 4.26; 95%CI: 3.87 to 4.69), non-employed (PR: 1.20; 95%CI: 1.15 to 1.26), individuals with obesity (PR: 1.49; 95%CI: 1.43 to 1.56), and smokers (PR: 1.24; 95%CI: 1.19 to 1.29). This study identified a high prevalence of multimorbidity and its association with sociodemographic and lifestyle factors. Transgender women (TGW) and men who have sex with other men (MSM) often encounter disparities in accessing HIV testing, leading to delayed diagnoses and worse prognoses. We analysed barriers and facilitators for accessing HIV rapid testing by TGW and MSM in Brazil, 2004-2023. Citations were included whether the study population consisted of individuals aged ≥18y old, and studies addressed HIV testing and have been conducted in Brazil. The study protocol was based on Joanna Briggs' recommendations for scoping reviews. We included 11 studies on TGW and 17 on MSM. The belief that one is not at risk of contracting HIV infection, fear expressed in different ways (e.g. lack of confidentiality) and younger age were the main barriers. Feeling at risk for HIV infection, curiosity, and favourable characteristics of the setting where the testing takes place were cited as the main facilitators. Barriers and facilitators specifically for HIV self-testing included, respectively, concerns about conducting the test alone vs. autonomy/flexibility. Brazil is unlikely to achieve the UN' 95-95-95 goal without minimising testing disparities. Combating prejudice against TGW and MSM in testing settings, along with educational campaigns and transparent protocols to ensure confidentiality, can help increase HIV testing among these populations.

In varied environments, microorganisms search for partners or nutritional resources using chemical signals. Microbes are drawn (chemotaxis) or grow directionally (chemotropism) towards the chemical source, enabling them to establish and maintain symbiosis. The hypocrealean fungi Escovopsis enhance their growth towards the basidiomycete fungus Leucoagaricus gongylophorus, which is cultivated by leaf-cutting attine ants for food. Although directional growth is well documented in this symbiosis, it is unclear whether non-volatile or volatile organic compounds participate in the interaction between cultivar and Escovopsis, and which specific chemical compounds might attract and induce chemotropism. In this study, we examined the growth responses of Escovopsis isolates to non-volatile and volatile organic compounds produced by fungal cultivars of higher attine ants. We also isolated and identified molecules released by the ant-cultivar and assessed the chemotropism of Escovopsis towards them. Our results indicate that the growth of Escovopsis is stimulated in the presence of both non-volatile and volatile compounds from fungal cultivars. We also identified three isomeric diketopiperazines molecules from crude extracts of the ant cultivar, suggesting that these might play a role in Escovopsis chemotropism. Objective: To map global scientific production on homoparenting in the field of collective health or public health., Methods: In terms of methodological procedures, a scoping review was carried out, guided by the following question: What are the aspects addressed in global scientific production regarding homoparental families in the field of collective or public health? The searches were carried out in seven sources of scientific literature, including 58 studies, involving scientific articles and dissertations. The analytical treatment given to the studies, most of which were qualitative, followed the content analysis technique in the thematic modality., Results: The results indicate that the perceptions of homosexuals and professionals about the care provided and health services in general was the topic addressed by the largest number of studies (n = 31), followed by heteronormative context of health services (n = 26); disclosure of sexual orientation (n = 20); fertilization (n = 14); educational information and actions (n = 5)., Conclusion: Although the issue of same-sex parenthood has been discussed in some health sectors, there is awareness that it is necessary to rely on a consolidated basis through numerous studies when discussing this issue. It is concluded that, among other aspects, the scope of this review is not sufficiently problematized within the scope of health professionals' training and performance.

Metabolic syndrome is a cluster of conditions that results from the interplay of genetic and environmental factors, which increase the comorbidity risk of obesity, hyperglycemia, dyslipidemia, arterial hypertension, stroke, and cardiovascular disease. In this article, we review various high-impact studies which link epigenetics with metabolic syndrome by comparing each study population, methylation effects, and strengths and weaknesses of each research. This study examined the relationship between two variables of the psychological future [future time orientation (FTO) and life project (LP)] and their relationship with career self-efficacy in unemployed individuals. Participants were 216 unemployed adults (151 women, 65 men), aged from 18 to 67 years old ( M  = 42.8, SD  = 10.57), who responded to measures of distance and impact of future time orientation, identification and involvement in life project and career exploration and decision-making self-efficacy. Results of latent mediation analysis and correlational analysis indicated that there is a direct effect between FTO and LP, but also an indirect (i.e., mediating) effect between them through career self-efficacy beliefs. These findings suggest that unemployed individuals with a stronger sense of future time orientation are more likely to identify and engage with their life projects, and that this organization of their life projects is mediated by their levels of career self-efficacy. The role of aberrant glycosylation in pancreatic ductal adenocarcinoma (PDAC) remains an under-investigated area of research. In this study, we determined that ST6 β-galactoside α2,6 sialyltransferase 1 (ST6GAL1), which adds α2,6-linked sialic acids to N-glycosylated proteins, was upregulated in patients with early-stage PDAC and was further increased in advanced disease. A tumor-promoting function for ST6GAL1 was elucidated using tumor xenograft experiments with human PDAC cells. Additionally, we developed a genetically engineered mouse (GEM) model with transgenic expression of ST6GAL1 in the pancreas and found that mice with dual expression of ST6GAL1 and oncogenic KRASG12D had greatly accelerated PDAC progression compared with mice expressing KRASG12D alone. As ST6GAL1 imparts progenitor-like characteristics, we interrogated ST6GAL1's role in acinar to ductal metaplasia (ADM), a process that fosters neoplasia by reprogramming acinar cells into ductal, progenitor-like cells. We verified ST6GAL1 promotes ADM using multiple models including the 266-6 cell line, GEM-derived organoids and tissues, and an in vivo model of inflammation-induced ADM. EGFR is a key driver of ADM and is known to be activated by ST6GAL1-mediated sialylation. Importantly, EGFR activation was dramatically increased in acinar cells and organoids from mice with transgenic ST6GAL1 expression. These collective results highlight a glycosylation-dependent mechanism involved in early stages of pancreatic neoplasia.

The ST6GAL1 sialyltransferase, which adds α2-6-linked sialic acids to N-glycosylated proteins, is upregulated in many malignancies including ovarian cancer. Through its activity in sialylating select surface receptors, ST6GAL1 modulates intracellular signaling to regulate tumor cell phenotype. ST6GAL1 has previously been shown to act as a survival factor that protects cancer cells from cytotoxic stressors such as hypoxia. In the present study, we investigated a role for ST6GAL1 in tumor cell metabolism. ST6GAL1 was overexpressed (OE) in OV4 ovarian cancer cells, which have low endogenous ST6GAL1, or knocked-down (KD) in ID8 ovarian cancer cells, which have high endogenous ST6GAL1. OV4 and ID8 cells with modulated ST6GAL1 expression were grown under normoxic or hypoxic conditions, and metabolism was assessed using Seahorse technology. Results showed that cells with high ST6GAL1 expression maintained a higher rate of oxidative metabolism than control cells following treatment with the hypoxia mimetic, desferrioxamine (DFO). This enrichment was not due to an increase in mitochondrial number. Glycolytic metabolism was also increased in OV4 and ID8 cells with high ST6GAL1 expression, and these cells displayed greater activity of the glycolytic enzymes, hexokinase and phosphofructokinase. Metabolism maps were generated from the combined Seahorse data, which suggested that ST6GAL1 functions to enhance the overall metabolism of tumor cells. Finally, we determined that OV4 and ID8 cells with high ST6GAL1 expression were more invasive under conditions of hypoxia. Apparent diffusion coefficient (ADC) of magnetic resonance imaging (MRI) is an indispensable imaging technique in clinical neuroimaging that quantitatively assesses the diffusivity of water molecules within tissues using diffusion-weighted imaging (DWI). This study focuses on developing a robust machine learning (ML) model to predict the aggressiveness of gliomas according to World Health Organization (WHO) grading by analyzing patients' demographics, higher-order moments, and grey level co-occurrence matrix (GLCM) texture features of ADC. A population of 722 labeled MRI-ADC brain image slices from 88 human subjects was selected, where gliomas are labeled as glioblastoma multiforme (WHO-IV), high-grade glioma (WHO-III), and low-grade glioma (WHO I-II). Images were acquired using 3T-MR systems and a region of interest (ROI) was delineated manually over tumor areas. Skewness, kurtosis, and statistical texture features of GLCM (mean, variance, energy, entropy, contrast, homogeneity, correlation, prominence, and shade) were calculated using ADC values within ROI. The ANOVA f-test was utilized to select the best features to train an ML model. The data set was split into training (70%) and testing (30%) sets. The train set was fed into several ML algorithms and selected most promising ML algorithm using K-fold cross-validation. The hyper-parameters of the selected algorithm were optimized using random grid search technique. Finally, the performance of the developed model was assessed by calculating accuracy, precision, recall, and F1 values reported for the test set. According to the ANOVA f-test, three attributes; patient gender (1.48), GLCM energy (9.48), and correlation (13.86) that performed minimum scores were excluded from the dataset. Among the tested algorithms, the random forest classifier(0.8772 ± 0.0237) performed the highest mean-cross-validation score and selected to build the ML model which was able to predict tumor categories with an accuracy of 88.14% over the test set. The study concludes that the developed ML model using the above features except for patient gender, GLCM energy, and correlation, has high prediction accuracy in glioma grading. Therefore, the outcomes of this study enable to development of advanced tumor classification applications that assist in the decision-making process in a real-time clinical environment., (© 2023. This article is an integral part of the research "Violence in the context of COVID-19: global challenges and vulnerabilities", which proposes a critical reflection on situations of gender-based violence increased by social distancing protocols, required by the COVID-19 pandemic. Based on a 2020 literature survey, we have analyzed recommendations made by researchers and institutions from different countries around the world, with the aim of systematizing and disseminating strategies to deal with this scenario. The material is organized into two thematic areas, namely: gender policies and intersectoral actions; and strategies to face violence against women and children in the health and social work field. The recommendations are focused on the development of actions by States/governments, service networks and society in general. Part of the recommendations suggest increasing or adapting existing surveillance actions and part of them contribute with creative proposals, guiding promotional and preventive actions at an individual and collective level. The adoption of teleassistance, media campaigns raising awareness that violence is unjustifiable and the development of reporting strategies through signs and codes have been reiterated in the literature.

Background and Aims: Nociception is the most prevalent pain mechanism in Parkinson disease (PD). It negatively affects quality of life, and there is currently no evidence-based treatment for its control. Burst spinal cord stimulation has been used to control neuropathic pain and recently has been shown to relieve pain of nociceptive origin. In this study, we hypothesize that burst transspinal magnetic stimulation (bTsMS) reduces nociceptive pain in PD., Materials and Methods: Twenty-six patients were included in a double-blind, sham-controlled, randomized parallel trial design, and the analgesic effect of lower-cervical bTsMS was assessed in patients with nociceptive pain in PD. Five daily induction sessions were followed by maintenance sessions delivered twice a week for seven weeks. The primary outcome was the number of responders (≥ 50% reduction of average pain intensity assessed on a numerical rating scale ranging from 0-10) during the eight weeks of treatment. Mood, quality of life, global impression of change, and adverse events were assessed throughout the study., Results: Twenty-six patients (46.2% women) were included in the study. The number of responders during treatment was significantly higher after active than after sham bTsMS (p = 0.044), mainly owing to the effect of the first week of treatment, when eight patients (61.5%) responded to active and two (15.4%) responded to sham bTsMS (p = 0.006); the number needed to treat was 2.2 at week 1. Depression symptom scores were lower after active (4.0 ± 3.1) than after sham bTsMS (8.7 ± 5.3) (p = 0.011). Patients' global impressions of change were improved after active bTsMS (70.0%) compared with sham bTsMS (18.2%; p = 0.030). Minor adverse events were reported in both arms throughout treatment sessions. One major side effect unrelated to treatment occurred in the active arm (death due to pulmonary embolism). Objective: To estimate the prevalence of exposure to the SARS-CoV-2 virus among individuals living in restricted freedom., Methods: A seroprevalence survey was carried out with the population of the female penitentiary of the Centro de Progressão Penitenciária (CPP) in Butantan (municipality of São Paulo), between June 24 and August 20, 2020. During this period, according to the Secretariat of Penitentiary Administration (SAP), the positivity of rapid tests among inmates ranged from 65% to 78%. The evaluation method used in the study was the "One Step COVID-19" rapid test (chromatography), from the company Wondfo, also using the RT-PCR method in symptomatic participants to confirm the viral condition. The study population consisted of 879 female inmates and 170 employees of the institution., Results: The prevalence of total antibodies (IgG/IgM) against the SARS-CoV-2 virus in the total population of 1049 study participants was 6.1%; among the population of 879 inmates,a prevalence of 5.8% was observed, and among the institution's employees, 7.5%., Conclusions: The prevalence of covid-19 at the Butantan CPP was low, which is due to the implementation of simple prevention measures at the institution, such as the use of masks (with appropriate changes), emphasis on hygiene, hand washing and social distancing, in addition to other strategies, such as suspending inmates' visits from relatives and friends and cutting back on elective medical appointments and outside work.

The transition to the first year of high school constitutes a critical moment because it corresponds to the implementation of a career choice, which can impact students' satisfaction and psychosocial adjustment. The career construction model of adaptation holds potential to explain how students adapt to high school, by suggesting linkages among adaptive readiness, resources, responses, and results. However, research applying the career construction model to school transitions, combining social-emotional, career, and academic variables is still needed. This study explores the roles that social-emotional skills (an indicator of adaptive readiness) and career adaptability (an indicator of adaptability resources) play in explaining first-year high school students' agentic school engagement (an indicator of adapting responses). Measures of social-emotional skills, career adaptability, and school engagement were completed by 136 students (63.2% girls; M age = 15.68). Results from the hierarchical linear regression analysis suggest that social-emotional skills and career adaptability explain 32% of the variance and significantly contribute to explaining agentic school engagement. These findings seem illustrative of the potential of the career construction model of adaptation to deepen knowledge and understanding about the transition to high school and the implementation of career choices. Aligned with the literature, this study supports the calls for integrative psychological practices that acknowledge social-emotional, career, and academic variables when fostering students' psychosocial adjustment.

This study compared the psychological future of unemployed individuals and workers during the Covid-19 pandemic. It used the data from two previous data collections, one with unemployed individuals and another with workers. Participants from the two datasets were paired considering the same gender and similar age and education degree. The analyzed sample consisted of 352 participants, of which 176 were unemployed individuals and 176 were workers. The psychological future was measured by the Future Time Orientation Scale and the Life Project Scale. Both scales fit the sample of unemployed individuals and were invariant at the metric level across occupation status. Partial scalar model met good fit after freeing the intercepts of one item in each scale. In contrast to the hypothesis, compared to workers, unemployed individuals did not have lower rates in the assessed features of their psychological future. Conversely, for some variables, the rates were even higher among unemployed individuals.
