Abstract

8-Chrysoeriol, a bioactive flavanoid, was firstly identified to bind directly to BCL-2 as BH3 mimetics by structure-based virtual ligand screening. And 3D docking model revealed the molecular basis of 8-Chrysoeriol targeting to BCL-2. The interaction between 8-Chrysoeriol and BCL-2 was further confirmed using Microscale Thermophoresis (MST) technique. Meanwhile, high expression level of antiapoptotic protein BCL-2 was detected in SW1990 pancreatic cancer cells and 8-Chrysoeriol showed obvious proapoptosis effect against SW1990 in vitro. Collectively, the results showed that 8-Chrysoeriol as a natural dietary product potentially targeting to BCL-2 could serve as a lead compound for SW1990 pancreatic cancer therapy.

11. [Association of KCNE1 and KCNE4 gene polymorphisms with atrial fibrillation among Uygur and Han Chinese populations in Xinjiang]

Abstract

To assess the association of KCNE1 (rs1805127) and KCNE4 (rs12621643) polymorphisms with atrial fibrillation (AF) among ethnic Uygur and Han Chinese in Xinjiang.A case-control study was carried out. The patients and controls were selected based on ethnicity, gender and age with an 1:1 ratio. DNA was extracted from peripheral blood samples. Genotypes of KCNE1 (rs1805127) and KCNE4 (rs12621643) were determined with a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay.Multivariate Logistic regression analysis showed KCNE1 (rs1805127) to be an independent risk factor for AF among Uygurs, while KCNE4 (rs12621643) was a risk factor for both Uygur and Han patients with AF (P0.05). The population attributable risk percentage (PARc%) of obstructive sleep apnea hpoventilation syndrome, obesity, hypertension, cholesterol, Hcy, hs-CRP, IL-6, KCNE1 (rs1805127) and KCNE4 (rs12621643) were 9.68%, 12.06%, 15.76%, 6.91%, 11.37%, 17.78%, 9.31%, 11.27% and 6.46% among the Uygurs, respectively. The PARc% of drinking, hypertension, cholesterol, Hcy, hs-CRP, IL-6, and KCNE4 (rs12621643) were 12.94%, 14.48%, 7.24%, 8.49%, 17.29%, 9.49% and 7.41% among Hans.The KCNE1 (rs1805127) appears to an independent risk factor for AF in the Uygur population. And the KCNE4 (rs12621643) was an independent risk factor for AF among both Uygurs and Hans. Management of the risk factors of AF based on testing of "risk genes" may have an impact on the prevention and treatment of AF.

12. Neuroprotective effects of Fomes officinalis Ames polysaccharides on Aβ25–35-induced cytotoxicity in PC12 cells through suppression of mitochondria-mediated apoptotic pathway.

Abstract

Aggregation of Aβ is a pathological hallmark of Alzheimer's disease (AD). The purpose of this study was to identify the protective roles of different polysaccharide components in Fomes officinalis Ames polysaccharides (FOAPs) against Aβ25–35-induced neurotoxicity in PC12 cells. Different doses of FOAPs components (i.e. FOAPs-a and FOAPs-b) were added to PC12 cells about 2 h before β-amyloid protein fragment 25–35 (Aβ25–35) exposure. The AD cellular model of PC12 cells was established using Aβ25–35. Then the PC12 cells were divided into 9 groups including: control group, Donepezil hydrochloride (DHCL) group, model group treated using 40 μM Aβ25–35, followed by FOAPs-a and FOAPs-b interference (50, 100 and 200 μg/mL). The mitochondrial reactive oxygen species (ROS), ATP, superoxide dismutase (SOD), malondialdehyde (MDA), lactate dehydrogenase (LDH) and mitochondrial membrane potential (MMP) were determined by commercial kits. The Cytochrome C, Bcl-2 and Bax expressions in the mitochondria and cytosol was determined by using Western blot analysis. FOAPs-a and FOAPs-b could significantly inhibit the LDH release, MDA level and the over accumulation of ROS induced by Aβ25–35 in PC12 cells in a dose-dependent manner. They could also effectively prevent Aβ25–35-stimulated cytotoxicity, which involved in attenuating cell apoptosis, increasing the ratio of Bcl-2/Bax and inhibiting Cytochrome C release from mitochondria to cytosol in PC12 cells. Moreover, FOAPs-a and FOAPs-b significantly alleviated mitochondrial dysfunction by regulating the MMP, as well as promoting the mitochondrial ATP synthesis. FOAPs-a and FOAPs-b played neuroprotective roles against Aβ25–35-induced cytotoxicity in PC12 cells through suppressing the mitochondria-mediated apoptotic pathway. [ABSTRACT FROM AUTHOR]

Abstract

To investigate prevalence of atrial fibrillation (AF) in Uygur and Han elderly populations in Xinjiang Uygur autonomous region (Xinjiang).Epidemiological survey was conducted among the residents selected through stratified random cluster sampling in the southern, northern and eastern Xinjiang.The overall AF prevalence among Uygur and Han elderly people was 3.56%. The crude prevalence of AF was 2.91% among Uygur elderly people and 4.13% among Han elderly people. The sex specific prevalence of AF were 3.19% and 2.61% among Uygur males and females respectively, and 5.01% and 3.31% among Han males and females respectively. The prevalence of valvular AF among Uygur ethnic group was higher than that in Han ethnic group; the prevalence of non-valvular and isolated AF in Han ethnic group were higher than those in Uygur ethnic group. The compliance of aspirin and β-blocker medication among Han ethnic group was better than that in Uygur ethnic group. The compliance of warfarin medication was poor in both Uygur ethnic group and Han ethnic group. The prevalence of ischemic stroke were 8.82% and 0.98% in Uygur elderly people with or without AF. The prevalence of ischemic stroke were 6.08% and 0.70% in Han elderly people with or without AF.The prevalence of AF in elderly people in Xinjiang is similar to the results from other domestic studies, the prevalence of AF in Han elderly people was higher than that in Uygur elderly peoples.

Abstract

To study the chemical constituents from Punica granatum.The chemical constituents were isolated by column chromatography on silica gel and MCI-gel CHP 20P, Sephadex LH-20. Their structures were elucidated on the basis of spectral analysis and physical constants.Ten compounds were isolated and elucidated as: gallic acid (1), methyl gallate (2), ellagic acid (3), (+) catechin (4), isoquerecitrin (5), D-mannitol (6), ursolic acid (7), Oleanolic acid (8), beta-Sitosterol (9) and Daucosterol (10).Compounds 1-10 are isolated from this plant for the first time.

19. Antitumor activity of flavonoids from Alpinia officinarumhance on gastric cancer

Abstract

Introduction: To investigate antitumor activity and mechanism of flavonoids from Alpinia officinarumHance against gastric cancer. Methods: Transplanted mouse fore-stomach carcinoma (MFC) tumor mice were divided into six groups: control group, model group, low dose (20 mg/kg), middle dose (40 mg/kg), and high dose (80 mg/kg) groups of TFAO and 5-Fu group. Mice were treated with TFAO or 5-Fu for 14 days, except those of control and model group. Tumor inhibitory rate, spleen, and thymus index were calculated. Contents of proliferating cell nuclear antigen, MMP-9, vascular endothelial growth factor, IL-1β, IL-6, and IL-17 in serum were detected. Effect of galangin on BGC-823 cell growth was detected. Cell apoptosis and cell cycle distribution were measured. Enzyme activity of Caspase-3, Caspase-8, and Caspase-9 was detected. Western blot was used to detect STAT3, Bcl-2, Bax, Caspase-3, Caspase-8, Caspase-9, CyclinB1, and CyclinD1 protein expression in BGC-823 cell. Results: Compared with model group, tumor weight of mice decreased significantly (p< .01) in 5-Fu group, low dose, middle dose, and high dose group of TFAO; thymus index of mice decreased significantly (p< .05) in 5-Fu group; and spleen index decreased significantly (p< .05) in low dose and middle dose groups of TFAO. Compared with model group, levels of PCNA, MMP-9, IL-1β, and IL-6 in serum of mice decreased obviously (p< .01) in all administration groups; levels of VEGF in serum of mice decreased obviously (p< .01) in low dose and high dose group of TFAO and 5-Fu group; and levels of IL-17 in serum of mice decreased significantly (p< .01) in low-dose and middle-dose groups of TFAO and 5-Fu group. Galangin could inhibit BGC-823 cell growth; accelerate apoptosis; block cell cycle; increase cell Caspase-3, Caspase-8, and Caspase-9 enzyme activity; upregulate expression of Caspase-3, Caspase-8, Caspase-9, and Bax; and downregulate expression of STAT3, CyclinB1, CyclinD1, and Bcl-2 protein. Conclusion: Flavonoids from A. officinarumshowed antitumor activity in gastric cancer. Mechanisms may be associated with inhibition of tumor angiogenesis, tumor cell proliferation, and cancer-associated inflammation.

Abstract

A neutral polysaccharide Gi-A1 was isolated from the roots of Glycyrrhiza inflata Bat. It had a molecular mass of over 2000 kDa and showed [α] + 81.4° (c 1.05, H2O). Acid hydrolysis and methylation analysis indicated that Gi-A1 was mainly composed of α-D-glucose, α-L-arabinose, and α-D-galactose with a molar ratio of 8.0:1.8:1.0. Abstract

Introduction: Septic acute kidney injury (AKI) is a leading cause of morbidity and mortality with no accepted method of therapy. Growth differentiation factor 15 (GDF15) is induced in many diseases, however its role in Lipopolysaccharide (LPS)-induced AKI is unknown. We aim to determine the role of GDF15 in LPS-induced AKI to assess its therapeutic potential. As GDF15 is known to have anti-inflammatory and anti-apoptotic effects, we hypothesize that GDF15 has a protective effect in LPS-induced AKI in the mouse model. Methods: GDF15 knockout (KO), transgenic (TG) and wild-type (WT) mice received 4mg/kg injections of LPS. Renal function was determined using serum BUN and creatinine levels. Kidney tissues were stained with H&E and scored based on area of tubular damage. Apoptosis and neutrophil infiltration were also detected by the TUNEL and MPO assays, respectively. The expression of inflammatory mediators was measured by quantitative RT-PCR. Results: LPS treatment induced AKI as evidenced by the increased levels of BUN and serum creatinine. Our results show GDF15 KO mice treated with LPS had the highest levels of BUN and creatinine. In contrast, GDF15 TG mice had significantly lower BUN and creatinine levels compared to KO, indicating that GDF15 has a protective effect in LPS-induced renal injury. GDF15 KO mice generally had greater levels of tubular damage, as indicated by swollen epithelia and luminal precipitate in histological sections, compared to GDF15 TG mice. We also found that GDF15 deficiency in KO mice augmented the expression of TNF-α and MCP-1 compared to WT mice. Conclusion: Our results show that GDF15 may play a protective role in LPS-induced acute kidney injury, potentially through inhibiting the NFκB pathway.
