1. JWST observations of the Horsehead photon-dominated region I. First results from multi-band near- and mid-infrared imaging

Abstract

The JWST has captured the sharpest IR images ever taken of the Horsehead nebula, a prototypical moderately irradiated PDR that is fully representative of most of the UV-illuminated molecular gas in the Milky Way and star-forming galaxies. We investigate the impact of FUV radiation of a molecular cloud and constrain the structure of the edge of the PDR and its illumination conditions. We used NIRCam and MIRI to obtain 17 broadband and 6 narrowband maps from 0.7 to 28 $\mu$m. We mapped the dust emission, scattered light, and several gas phase lines. We also used HST-WFC3 maps at 1.1 and 1. 6 $\mu$m, along with HST-STIS spectroscopic observations of the H$\alpha$ line. We probed the structure of the edge of the Horsehead and resolved its spatial complexity. We detected a network of faint striated features extending perpendicularly to the PDR front into the H\,II region in filters sensitive to nano-grain emission and light scattered by larger grains. This may indeed figure as the first detection of the entrainment of dust particles in the evaporative flow. The map of the 1-0 S(1) line of H$_2$ presents sharp sub-structures on scales as small as 1.5 arcsec. The ionization and dissociation fronts appear at distances 1-2 arcsec behind the edge of the PDR and seem to spatially coincide, indicating a thickness of the neutral atomic layer below 100 au. All broadband maps present strong color variations which can be explained by dust attenuation. Deviations of the emissions in the H$\alpha$, Pa$\alpha,$ and Br$\alpha$ lines also indicate dust attenuation. With a very simple model, we derive the main features of the extinction curve. A small excess of extinction at 3 $\mu$m may be attributed to icy H$_2$O mantles onto grains. In all lines of sight crossing the inner regions of the Horsehead, it appears that dust attenuation is non-negligible over the entire spectral range., Comment: 25 pages, 17 figures

Abstract

We present experimental single and multimode sustained 1H NMR masers in solution on thermally polarized spins at room temperature and 9.4 T achieved through the electronic control of radiation feedback (radiation damping). Our observations illustrate the breakdown of the usual three-dimensional Maxwell-Bloch equations for radiation feedback and a simple toy model of few coupled classical moments is used to interpret these experiments.

Abstract

Mid-infrared emission features probe the properties of ionized gas, and hot or warm molecular gas. The Orion Bar is a frequently studied photodissociation region (PDR) containing large amounts of gas under these conditions, and was observed with the MIRI IFU aboard JWST as part of the "PDRs4All" program. The resulting IR spectroscopic images of high angular resolution (0.2") reveal a rich observational inventory of mid-IR emission lines, and spatially resolve the substructure of the PDR, with a mosaic cutting perpendicularly across the ionization front and three dissociation fronts. We extracted five spectra that represent the ionized, atomic, and molecular gas layers, and measured the most prominent gas emission lines. An initial analysis summarizes the physical conditions of the gas and the potential of these data. We identified around 100 lines, report an additional 18 lines that remain unidentified, and measured the line intensities and central wavelengths. The H I recombination lines originating from the ionized gas layer bordering the PDR, have intensity ratios that are well matched by emissivity coefficients from H recombination theory, but deviate up to 10% due contamination by He I lines. We report the observed emission lines of various ionization stages of Ne, P, S, Cl, Ar, Fe, and Ni, and show how certain line ratios vary between the five regions. We observe the pure-rotational H$_2$ lines in the vibrational ground state from 0-0 S(1) to 0-0 S(8), and in the first vibrationally excited state from 1-1 S(5) to 1-1 S(9). We derive H$_2$ excitation diagrams, and approximate the excitation with one thermal (~700 K) component representative of an average gas temperature, and one non-thermal component (~2700 K) probing the effect of UV pumping. We compare these results to an existing model for the Orion Bar PDR and highlight the differences with the observations., Comment: 26 pages, 12 figures, 3 tables. Abstract

The MRS mode of the JWST-MIRI instrument has been shown to be a powerful tool to characterise the molecular gas emission of the inner region of planet-forming disks. Here, we analyse the spectrum of the compact T-Tauri disk DR Tau, which is complemented by high spectral resolution (R~60000-90000) CO ro-vibrational observations. Various molecular species, including CO, CO$_2$, HCN, and C$_2$H$_2$ are detected in the JWST-MIRI spectrum, for which excitation temperatures of T~325-900 K are retrieved using LTE slab models. The high-resolution CO observations allow for a full treatment of the line profiles, which show evidence for two components of the main isotopologue, $^{12}$CO: a broad component tracing the Keplerian disk and a narrow component tracing a slow disk wind. Rotational diagrams yield excitation temperatures of T>725 K for CO, with consistently lower temperatures found for the narrow components, suggesting that the disk wind is launched from a larger distance. The inferred excitation temperatures for all molecules suggest that CO originates from the highest atmospheric layers close to the host star, followed by HCN and C$_2$H$_2$, which emit, together with $^{13}$CO, from slightly deeper layers, whereas the CO$_2$ originates from even deeper inside or further out in the disk. Additional analysis of the $^{12}$CO line wings hint at a misalignment between the inner (i~20 degrees) and outer disk (i~5 degrees). Abstract

The study of protoplanetary disks has become increasingly important with the Kepler satellite finding that exoplanets are ubiquitous around stars in our galaxy and the discovery of enormous diversity in planetary system architectures and planet properties. High-resolution near-IR and ALMA images show strong evidence for ongoing planet formation in young disks. The JWST MIRI mid-INfrared Disk Survey (MINDS) aims to (1) investigate the chemical inventory in the terrestrial planet-forming zone across stellar spectral type, (2) follow the gas evolution into the disk dispersal stage, and (3) study the structure of protoplanetary and debris disks in the thermal mid-IR. The MINDS survey will thus build a bridge between the chemical inventory of disks and the properties of exoplanets. The survey comprises 52 targets (Herbig Ae stars, T Tauri stars, very low-mass stars and young debris disks). We primarily obtain MIRI/MRS spectra with high S/N (~100-500) covering the complete wavelength range from 4.9 to 27.9 {\mu}m. For a handful of selected targets we also obtain NIRSpec IFU high resolution spectroscopy (2.87-5.27 {\mu}m). We will search for signposts of planet formation in thermal emission of micron-sized dust - information complementary to near-IR scattered light emission from small dust grains and emission from large dust in the submillimeter wavelength domain. We will also study the spatial structure of disks in three key systems that have shown signposts for planet formation, TW Hya and HD 169142 using the MIRI coronagraph at 15.5 {\mu}m and 10.65 {\mu}m respectively and PDS70 using NIRCam imaging in the 1.87 {\mu}m narrow and the 4.8 {\mu}m medium band filter. ..., Comment: accepted for publication in PASP

Abstract

Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photo-dissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, impacting planet formation within the disks. We report JWST and Atacama Large Millimetere Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modelling their kinematics and excitation allows us to constrain the physical conditions within the gas. We quantify the mass-loss rate induced by the FUV irradiation, finding it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.

Abstract

We study the emission of dust grains within the Orion Bar - a well-known, highly far-UV (FUV)-irradiated PDR. The Orion Bar because of its edge-on geometry provides an exceptional benchmark for characterizing dust evolution and the associated driving processes under varying physical conditions. Our goal is to constrain the local properties of dust by comparing its emission to models. Taking advantage of the recent JWST PDRs4All data, we follow the dust emission as traced by JWST NIRCam (at 3.35 and 4.8 micron) and MIRI (at 7.7, 11.3, 15.0, and 25.5 micron), along with NIRSpec and MRS spectroscopic observations. First, we constrain the minimum size and hydrogen content of carbon nano-grains from a comparison between the observed dust emission spectra and the predictions of the THEMIS dust model coupled to the numerical code DustEM. Using this dust model, we then perform 3D radiative transfer simulations of dust emission with the SOC code and compare to data obtained along well chosen profiles across the Orion Bar. The JWST data allows us, for the first time, to spatially resolve the steep variation of dust emission at the illuminated edge of the Orion Bar PDR. By considering a dust model with carbonaceous nano-grains and submicronic coated silicate grains, we derive unprecedented constraints on the properties of across the Orion Bar. To explain the observed emission profiles with our simulations, we find that the nano-grains must be strongly depleted with an abundance (relative to the gas) 15 times less than in the diffuse ISM. The NIRSpec and MRS spectroscopic observations reveal variations in the hydrogenation of the carbon nano-grains. The lowest hydrogenation levels are found in the vicinity of the illuminating stars suggesting photo-processing while more hydrogenated nano-grains are found in the cold and dense molecular region, potentially indicative of larger grains., Comment: 18 pages, 11 figures, accepted for publication in A&A

Abstract

Water is a key ingredient for the emergence of life as we know it. Yet, its destruction and reformation in space remains unprobed in warm gas. Here, we detect the hydroxyl radical (OH) emission from a planet-forming disk exposed to external far-ultraviolet (FUV) radiation with the James Webb Space Telescope. The observations are confronted with the results of quantum dynamical calculations. The highly excited OH infrared rotational lines are the tell-tale signs of H2O destruction by FUV. The OH infrared ro-vibrational lines are attributed to chemical excitation via the key reaction O+H=OH+H which seeds the formation of water in the gas-phase. We infer that the equivalent of the Earth ocean's worth of water is destroyed per month and replenished. These results show that under warm and irradiated conditions water is destroyed and efficiently reformed via gas-phase reactions. Abstract

(Abridged) We investigate the impact of radiative feedback from massive stars on their natal cloud and focus on the transition from the HII region to the atomic PDR (crossing the ionisation front (IF)), and the subsequent transition to the molecular PDR (crossing the dissociation front (DF)). We use high-resolution near-IR integral field spectroscopic data from NIRSpec on JWST to observe the Orion Bar PDR as part of the PDRs4All JWST Early Release Science Program. The NIRSpec data reveal a forest of lines including, but not limited to, HeI, HI, and CI recombination lines, ionic lines, OI and NI fluorescence lines, Aromatic Infrared Bands (AIBs including aromatic CH, aliphatic CH, and their CD counterparts), CO2 ice, pure rotational and ro-vibrational lines from H2, and ro-vibrational lines HD, CO, and CH+, most of them detected for the first time towards a PDR. Their spatial distribution resolves the H and He ionisation structure in the Huygens region, gives insight into the geometry of the Bar, and confirms the large-scale stratification of PDRs. We observe numerous smaller scale structures whose typical size decreases with distance from Ori C and IR lines from CI, if solely arising from radiative recombination and cascade, reveal very high gas temperatures consistent with the hot irradiated surface of small-scale dense clumps deep inside the PDR. The H2 lines reveal multiple, prominent filaments which exhibit different characteristics. This leaves the impression of a "terraced" transition from the predominantly atomic surface region to the CO-rich molecular zone deeper in. Abstract

The JWST has captured the most detailed and sharpest infrared images ever taken of the inner region of the Orion Nebula, the nearest massive star formation region, and a prototypical highly irradiated dense photo-dissociation region (PDR). We investigate the fundamental interaction of far-ultraviolet photons with molecular clouds. The transitions across the ionization front (IF), dissociation front (DF), and the molecular cloud are studied at high-angular resolution. These transitions are relevant to understanding the effects of radiative feedback from massive stars and the dominant physical and chemical processes that lead to the IR emission that JWST will detect in many Galactic and extragalactic environments. Due to the proximity of the Orion Nebula and the unprecedented angular resolution of JWST, these data reveal that the molecular cloud borders are hyper structured at small angular scales of 0.1-1" (0.0002-0.002 pc or 40-400 au at 414 pc). A diverse set of features are observed such as ridges, waves, globules and photoevaporated protoplanetary disks. At the PDR atomic to molecular transition, several bright features are detected that are associated with the highly irradiated surroundings of the dense molecular condensations and embedded young star. Toward the Orion Bar PDR, a highly sculpted interface is detected with sharp edges and density increases near the IF and DF. This was predicted by previous modeling studies, but the fronts were unresolved in most tracers. A complex, structured, and folded DF surface was traced by the H2 lines. This dataset was used to revisit the commonly adopted 2D PDR structure of the Orion Bar. JWST provides us with a complete view of the PDR, all the way from the PDR edge to the substructured dense region, and this allowed us to determine, in detail, where the emission of the atomic and molecular lines, aromatic bands, and dust originate.

Abstract

(Abridged) Mid-infrared observations of photodissociation regions (PDRs) are dominated by strong emission features called aromatic infrared bands (AIBs). The most prominent AIBs are found at 3.3, 6.2, 7.7, 8.6, and 11.2 $\mu$m. The most sensitive, highest-resolution infrared spectral imaging data ever taken of the prototypical PDR, the Orion Bar, have been captured by JWST. We provide an inventory of the AIBs found in the Orion Bar, along with mid-IR template spectra from five distinct regions in the Bar: the molecular PDR, the atomic PDR, and the HII region. We use JWST NIRSpec IFU and MIRI MRS observations of the Orion Bar from the JWST Early Release Science Program, PDRs4All (ID: 1288). We extract five template spectra to represent the morphology and environment of the Orion Bar PDR. The superb sensitivity and the spectral and spatial resolution of these JWST observations reveal many details of the AIB emission and enable an improved characterization of their detailed profile shapes and sub-components. While the spectra are dominated by the well-known AIBs at 3.3, 6.2, 7.7, 8.6, 11.2, and 12.7 $\mu$m, a wealth of weaker features and sub-components are present. We report trends in the widths and relative strengths of AIBs across the five template spectra. These trends yield valuable insight into the photochemical evolution of PAHs, such as the evolution responsible for the shift of 11.2 $\mu$m AIB emission from class B$_{11.2}$ in the molecular PDR to class A$_{11.2}$ in the PDR surface layers. This photochemical evolution is driven by the increased importance of FUV processing in the PDR surface layers, resulting in a "weeding out" of the weakest links of the PAH family in these layers. Abstract

Targeted alpha therapy (TAT) pairs the specificity of antigen targeting with the lethality of alpha particles to eradicate cancerous cells. Actinium-225 [225Ac; t1/2 = 9.920(3) days] is an alpha-emitting radioisotope driving the next generation of TAT radiopharmaceuticals. Despite promising clinical results, a fundamental understanding of Ac coordination chemistry lags behind the rest of the Periodic Table due to its limited availability, lack of stable isotopes, and inadequate systems poised to probe the chemical behavior of this radionuclide. In this work, we demonstrate a platform that combines an 8-coordinate synthetic ligand and a mammalian protein to characterize the solution and solid-state behavior of the longest-lived Ac isotope, 227Ac [t1/2 = 21.772(3) years]. We expect these results to direct renewed efforts for 225Ac-TAT development, aid in understanding Ac coordination behavior relative to other +3 lanthanides and actinides, and more broadly inform this element's position on the Periodic Table.

Abstract

Yersinia pestis, the causative agent of plague and a biological threat agent, presents an urgent need for novel medical countermeasures due to documented cases of naturally acquired antibiotic resistance and potential person-to-person spread during a pneumonic infection. Immunotherapy has been proposed as a way to circumvent current and future antibiotic resistance. Here, we describe the development and characterization of two affinity matured human antibodies (αF1Ig AM2 and αF1Ig AM8) that promote survival of mice after exposure to aerosolized Y. pestis. We share details of the error prone PCR and yeast display technology-based affinity maturation process that we used. The resultant matured antibodies have nanomolar affinity for Y. pestis F1 antigen, are produced in high yield, and are resilient to 37°C stress for up to 6 months. Importantly, in vitro assays using a murine macrophage cell line demonstrated that αF1Ig AM2 and αF1Ig AM8 are opsonic. Even more importantly, in vivo studies using pneumonic plague mouse models showed that 100% of the mice receiving 500 μg of IgGs αF1Ig AM2 and αF1Ig AM8 survived lethal challenge with aerosolized Y. pestis CO92. Combined, these results provide evidence of the quality and robustness of αF1Ig AM2 and αF1Ig AM8 and support their development as potential medical countermeasures against plague.

Abstract Hyperparasitism is a common pattern in nature that is not limited to cellular organisms. Giant viruses infecting protists can be hyperparasitized by smaller ones named virophages. In addition, both may carry episomal DNA molecules known as transpovirons in their particles. They all share transcriptional regulatory elements that dictate the expression of their genes within viral factories built by giant viruses in the host cytoplasm. This suggests the existence of interactions between their respective transcriptional networks. Here we investigated Acanthamoeba castellanii cells infected by a giant virus (megavirus chilensis), and coinfected with a virophage (zamilon vitis) and/or a transpoviron (megavirus vitis transpoviron). Infectious cycles were monitored through time-course RNA sequencing to decipher the transcriptional program of each partner and its impact on the gene expression of the others. We found highly diverse transcriptional responses. While the giant virus drastically reshaped the host cell transcriptome, the transpoviron had no effect on the gene expression of any of the players. In contrast, the virophage strongly modified the giant virus gene expression, albeit transiently, without altering the protein composition of mature viral particles. The virophage also induced the overexpression of transpoviron genes, likely through the indirect upregulation of giant virus-encoded transcription factors. Together, these analyses document the intricated transcriptionally regulated networks taking place in the infected cell.

Abstract

The understanding of planet formation has changed recently, embracing the new idea of pebble accretion. This means that the influx of pebbles from the outer regions of planet-forming disks to their inner zones could determine the composition of planets and their atmospheres. The solid and molecular components delivered to the planet-forming region can be best characterized by mid-infrared spectroscopy. With Spitzer low-resolution (R=100, 600) spectroscopy, this approach was limited to the detection of abundant molecules such as H2O, C2H2, HCN and CO2. This contribution will present first results of the MINDS (MIRI mid-IR Disk Survey, PI: Th. Henning) project. Due do the sensitivity and spectral resolution (R~1500-3500) provided by JWST we now have a unique tool to obtain the full inventory of chemistry in the inner disks of solar-types stars and brown dwarfs, including also less abundant hydrocarbons and isotopologues. The Integral Field Unit (IFU) capabilities enable at the same time spatial studies of the continuum and line emission in extended sources such as debris disks, the flying saucer and also the search for mid-IR signatures of forming planets in systems such as PDS70. Abstract

Terrestrial and sub-Neptune planets are expected to form in the inner ($<10~$AU) regions of protoplanetary disks. Water plays a key role in their formation, although it is yet unclear whether water molecules are formed in-situ or transported from the outer disk. So far Spitzer Space Telescope observations have only provided water luminosity upper limits for dust-depleted inner disks, similar to PDS 70, the first system with direct confirmation of protoplanet presence. Here we report JWST observations of PDS 70, a benchmark target to search for water in a disk hosting a large ($\sim54~$AU) planet-carved gap separating an inner and outer disk. Our findings show water in the inner disk of PDS 70. This implies that potential terrestrial planets forming therein have access to a water reservoir. The column densities of water vapour suggest in-situ formation via a reaction sequence involving O, H$_2$, and/or OH, and survival through water self-shielding. This is also supported by the presence of CO$_2$ emission, another molecule sensitive to UV photodissociation. Dust shielding, and replenishment of both gas and small dust from the outer disk, may also play a role in sustaining the water reservoir. Abstract

(Abridged) Massive stars can determine the evolution of molecular clouds with their strong ultraviolet (UV) radiation fields. Moreover, UV radiation is relevant in setting the thermal gas pressure in star-forming clouds, whose influence can extend from the rims of molecular clouds to entire star-forming galaxies. Probing the fundamental structure of nearby molecular clouds is therefore crucial to understand how massive stars shape their surrounding medium and how fast molecular clouds are destroyed, specifically at their UV-illuminated edges, where models predict an intermediate zone of neutral atomic gas between the molecular cloud and the surrounding ionized gas whose size is directly related to the exposed physical conditions. We present the highest angular resolution (~$0.5$", corresponding to $207$ au) and velocity-resolved images of the molecular gas emission in the Horsehead nebula, using CO J=3-2 and HCO$^+$ J=4-3 observations with ALMA. We find that CO and HCO$^+$ are present at the edge of the cloud, very close to the ionization (H$^+$/H) and dissociation fronts (H/H$_2$), suggesting a very thin layer of neutral atomic gas (<$650$ au) and a small amount of CO-dark gas ($A_V=0.006-0.26$ mag) for stellar UV illumination conditions typical of molecular clouds in the Milky Way. The new ALMA observations reveal a web of molecular gas filaments with an estimated thermal gas pressure of $P_{\mathrm{th}} = (2.3 - 4.0) \times 10^6$ K cm$^{-3}$, and the presence of a steep density gradient at the cloud edge that can be well explained by stationary isobaric PDR models with pressures consistent with our estimations. However, in the HII region and PDR interface, we find $P_{\mathrm{th,PDR}} > P_{\mathrm{th,HII}}$, suggesting the gas is slightly compressed. Abstract

MIRI/MRS on board the JWST allows us to probe the inner regions of protoplanetary disks. Here we examine the disk around the classical T Tauri star Sz 98, which has an unusually large dust disk in the millimetre with a compact core. We focus on the H$_2$O emission through both its ro-vibrational and pure rotational emission. Furthermore, we compare our chemical findings with those obtained for the outer disk from Atacama Large Millimeter/submillimeter Array (ALMA) observations. In order to model the molecular features in the spectrum, the continuum was subtracted and LTE slab models were fitted. The spectrum was divided into different wavelength regions corresponding to H$_2$O lines of different excitation conditions, and the slab model fits were performed individually per region. We confidently detect CO, H$_2$O, OH, CO$_2$, and HCN in the emitting layers. The isotopologue H$^{18}_2$O is not detected. Additionally, no other organics, including C$_2$H$_2$, are detected. This indicates that the C/O ratio could be substantially below unity, in contrast with the outer disk. The H$_2$O emission traces a large radial disk surface region, as evidenced by the gradually changing excitation temperatures and emitting radii. The OH and CO$_2$ emission are relatively weak. It is likely that H$_2$O is not significantly photodissociated; either due to self-shielding against the stellar irradiation, or UV-shielding from small dust particles. The relative emitting strength of the different identified molecular features point towards UV-shielding of H$_2$O in the inner disk of Sz 98, with a thin layer of OH on top. The majority of the organic molecules are either hidden below the dust continuum, or not present. In general, the inferred composition points to a sub-solar C/O ratio (<0.5) in the inner disk, in contrast with the larger than unity C/O ratio in the gas in the outer disk found with ALMA., Comment: Updated to accepted version. Abstract

The mammalian protein siderocalin binds bacterial siderophores and their iron complexes through cation-π and electrostatic interactions, but also displays high affinity for hydroxypyridinone complexes of trivalent lanthanides and actinides. In order to circumvent synthetic challenges, the use of siderocalin-antibody fusion proteins is explored herein as an alternative targeting approach for precision delivery of trivalent radiometals. We demonstrate the viability of this approach in vivo, using the theranostic pair 90Y (β-, t1/2 = 64 h)/86Y (β+, t1/2 = 14.7 h) in a SKOV-3 xenograft mouse model. Ligand radiolabeling with octadentate hydroxypyridinonate 3,4,3-LI(1,2-HOPO) and subsequent protein binding were achieved at room temperature. The results reported here suggest that the rapid non-covalent binding interaction between siderocalin fusion proteins and the negatively charged Y(iii)-3,4,3-LI(1,2-HOPO) complexes could enable purification-free, cold-kit labeling strategies for the application of therapeutically relevant radiometals in the clinic.

Abstract

Carbon is an essential element for life but how much can be delivered to young planets is still an open question. The chemical characterization of planet-forming disks is a crucial step in our understanding of the diversity and habitability of exoplanets. Very low-mass stars ($<0.2~M_{\odot}$) are interesting targets because they host a rich population of terrestrial planets. Here we present the JWST detection of abundant hydrocarbons in the disk of a very low-mass star obtained as part of the MIRI mid-INfrared Disk Survey (MINDS). In addition to very strong and broad emission from C$_2$H$_2$ and its $^{13}$C$^{12}$CH$_2$ isotopologue, C$_4$H$_2$, benzene, and possibly CH$_4$ are identified, but water, PAH and silicate features are weak or absent. The lack of small silicate grains implies that we can look deep down into this disk. These detections testify to an active warm hydrocarbon chemistry with a high C/O ratio in the inner 0.1 au of this disk, perhaps due to destruction of carbonaceous grains. The exceptionally high C$_2$H$_2$/CO$_2$ and C$_2$H$_2$/H$_2$O column density ratios suggest that oxygen is locked up in icy pebbles and planetesimals outside the water iceline. Abstract

Synthetic meat or the production of edible animal tissues in vitro, addresses major concerns such as climate change, animal ethics and human health. This article explores its epistemological specificity and its impact on our conception of human and non-human life. Cellular meat, derived from cellular agriculture, questions living materiality and its appropriation via technoscientific means. We analyze the ontological status of cellular meat through the technical recomposition of life, characterized by the process of biofabricating “meat,” thus revealing the material production of new forms of edible biological matter. A critical analysis of neo-materialism illustrates the limits of this materiality and its social implications, specifically on the relationship with animals and rural communities. By examining prospective discourses and semantic shifts surrounding cellular agriculture, we question the promises and imaginaries associated with cellular meat.

Abstract

Abstract Targeted alpha therapy (TAT) pairs the specificity of antigen targeting with the lethality of alpha particles to eradicate cancerous cells. Actinium-225 [225Ac; t1/2 = 9.920(3) days] is an alpha-emitting radioisotope driving the next generation of TAT radiopharmaceuticals. Despite promising clinical results, a fundamental understanding of Ac coordination chemistry lags behind the rest of the Periodic Table due to its limited availability, lack of stable isotopes, and inadequate systems poised to probe the chemical behavior of this radionuclide. In this work, we demonstrate a platform that combines an 8-coordinate synthetic ligand and a mammalian protein to characterize the solution and solid-state behavior of the longest-lived Ac isotope, 227Ac [t1/2 = 21.772(3) years]. We expect these results to direct renewed efforts for 225Ac-TAT development, aid in understanding Ac coordination behavior relative to other +3 lanthanides and actinides, and more broadly inform this element’s position on the Periodic Table.

In this work, we present a modified simultaneous growth and self-aggregation method that produces ceria and silver Janus nanoparticles for the conversion of 4-nitrophenol, a chemical widely used in several industries. The nanoparticles had cerium-to-silver ratios ranging from 0 to 1.35 and well-defined heterodimer morphologies. By controlling the growth conditions, we have manipulated the interface between ceria and silver, maximizing its exposure to the chemical reactants and increasing the reaction rate constants between 2- and 4-fold. Taken together, these results can inform the design rules to achieve better performing hybrid nanocatalysts.

Abstract

We present JWST-MIRI MRS spectra of the protoplanetary disk around the low-mass T Tauri star GW Lup from the MIRI mid-INfrared Disk Survey (MINDS) GTO program. Emission from $^{12}$CO$_{2}$, $^{13}$CO$_{2}$, H$_{2}$O, HCN, C$_{2}$H$_{2}$, and OH is identified with $^{13}$CO$_{2}$ being detected for the first time in a protoplanetary disk. We characterize the chemical and physical conditions in the inner few au of the GW Lup disk using these molecules as probes. The spectral resolution of JWST-MIRI MRS paired with high signal-to-noise data is essential to identify these species and determine their column densities and temperatures. The $Q$-branches of these molecules, including those of hot-bands, are particularly sensitive to temperature and column density. We find that the $^{12}$CO$_{2}$ emission in the GW Lup disk is coming from optically thick emission at a temperature of $\sim$400 K. $^{13}$CO$_{2}$ is optically thinner and based on a lower temperature of $\sim$325 K, may be tracing deeper into the disk and/or a larger emitting radius than $^{12}$CO$_{2}$. The derived $N_{\rm{CO_{2}}}$/$N_{\rm{H_{2}O}}$ ratio is orders of magnitude higher than previously derived for GW Lup and other targets based on \textit{Spitzer}-IRS data. This high column density ratio may be due to an inner cavity with a radius in between the H$_{2}$O and CO$_{2}$ snowlines and/or an overall lower disk temperature. Abstract

Background & Aims: In France, bulevirtide (BLV) became available in September 2019 through an early access program to treat patients with HDV. The aim of this analysis was to evaluate the efficacy and safety of BLV in patients with HIV and HDV coinfection. Methods: Patients received BLV 2 mg ± pegylated interferon-α (pegIFNα) according to the physician’s decision. The primary endpoint (per-protocol analysis) was the virological response rate at Week 48, defined as the proportion of patients with undetectable serum HDV RNA or a HDV RNA decline >2 log10 IU/ml from baseline. Results: The characteristics of the 38 patients were as follows: 28 male, mean age 47.7 years, and mean baseline HDV RNA viral load 5.7 ± 1.2 log10 IU/ml. Median HIV viral load and mean CD4 count were 32 (30–65) copies/ml and 566 ± 307/mm3, respectively. Eight patients stopped treatment before Week 48. At Week 48, 10 of 19 patients (52.6%) in the 2 mg BLV group and five of seven patients (71.4%) in the 2 mg BLV + pegIFNɑ group had reached virological response (no HDV RNA available in four patients). At Week 48, seven of 19 patients in the 2 mg BLV group and three of six patients in the 2 mg BLV + pegIFNɑ group had a combined response (virological response and normal alanine aminotransferase level). Conclusions: Adults living with HIV coinfected with HDV can be treated by BLV with a virological response in more than 50% of patients. The combination of BLV and pegIFNɑ showed a strong virological response. Impact and implications: Bulevirtide is the only EMA-approved drug for HDV treatment, and we showed that it can be used in adults living with HIV, with an overall good tolerability. Bulevirtide induces a virological response in more than 50% of patients, suggesting that bulevirtide should be considered as a first-line therapy in this specific population. Bulevirtide in combination with pegIFNα could be used in patients without pegIFNα contraindication. No specific drug–drug interaction is reported. Bulevirtide is the only EMA-approved drug for HDV treatment, and we showed that it can be used in adults living with HIV, with an overall good tolerability. Bulevirtide induces a virological response in more than 50% of patients, suggesting that bulevirtide should be considered as a first-line therapy in this specific population. Bulevirtide in combination with pegIFNα could be used in patients without pegIFNα contraindication. No specific drug–drug interaction is reported. Abstract

In view of the mounting requirements for courses in nuclear chemistry and radiochemistry, hands-on experience in the separations of radioisotopes is imperative. Three separation techniques commonly used were introduced, coprecipitation, liquid-liquid extraction, and ion-exchange chromatography. Their principles and the associated fundamental thermodynamic concepts are detailed. Their respective experimental applications to the separations of radioisotopes of one rare earth element (europium) and two transition metals (cobalt and nickel) are discussed. In-classroom experimental settings are proposed, and experiment outcomes are presented, which can be beneficial for future implementation within nuclear chemistry and radiochemistry programs in higher education.

Abstract

Context. Carbonaceous nano-grains play a fundamental role in the physico-chemistry of the interstellar medium (ISM) and especially of photon-dominated regions (PDRs). Their properties vary with the local physical conditions and affect the local chemistry and dynamics. Aims. We aim to highlight the evolution of carbonaceous nano-grains in three different PDRs and propose a scenario of dust evolution as a response to the physical conditions. Methods. We used Spitzer/IRAC (3.6, 4.5, 5.8, and 8 $\mu$m) and Spitzer/MIPS (24 $\mu$m) together with Herschel/PACS (70 $\mu$m) to map dust emission in IC63 and the Orion Bar. To assess the dust properties, we modelled the dust emission in these regions using the radiative transfer code SOC together with the THEMIS dust model. Results. Regardless of the PDR, we find that nano-grains are depleted and that their minimum size is larger than in the diffuse ISM (DISM), which suggests that the mechanisms that lead nano-grains to be photo-destroyed are very efficient below a given critical size limit. The evolution of the nano-grain dust-to-gas mass ratio with both G0 and the effective temperature of the illuminating star indicates a competition between the nano-grain formation through the fragmentation of larger grains and nano-grain photo-destruction. We modelled dust collisions driven by radiative pressure with a classical 1D approach to show that this is a viable scenario for explaining nano-grain formation through fragmentation and, thus, the variations observed in nano-grain dust-to-gas mass ratios from one PDR to another. Conclusions. We find a broad variation in the nano-grain dust properties from one PDR to another, along with a general trend of nano-grain depletion in these regions. We propose a viable scenario of nano-grain formation through fragmentation of large grains due to radiative pressure-induced collisions.

38. Multi-temporal speckle reduction with self-supervised deep neural networks

Abstract

Speckle filtering is generally a prerequisite to the analysis of synthetic aperture radar (SAR) images. Tremendous progress has been achieved in the domain of single-image despeckling. Latest techniques rely on deep neural networks to restore the various structures and textures peculiar to SAR images. The availability of time series of SAR images offers the possibility of improving speckle filtering by combining different speckle realizations over the same area. The supervised training of deep neural networks requires ground-truth speckle-free images. Such images can only be obtained indirectly through some form of averaging, by spatial or temporal integration, and are imperfect. Given the potential of very high quality restoration reachable by multi-temporal speckle filtering, the limitations of ground-truth images need to be circumvented. We extend a recent self-supervised training strategy for single-look complex SAR images, called MERLIN, to the case of multi-temporal filtering. This requires modeling the sources of statistical dependencies in the spatial and temporal dimensions as well as between the real and imaginary components of the complex amplitudes. Quantitative analysis on datasets with simulated speckle indicates a clear improvement of speckle reduction when additional SAR images are included. Our method is then applied to stacks of TerraSAR-X images and shown to outperform competing multi-temporal speckle filtering approaches. Abstract

Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Abstract

Lanthanides are a series of elements essential to a wide range of applications, from clean energy production to healthcare. Despite their presence in multiple products and technologies, their toxicological characteristics have been only partly studied. Recently, our group has employed a genomic approach to extensively characterize the toxicity mechanisms of lanthanides. Even though we identified substantially different behaviors for mid and late lanthanides, the toxicological profiles of early lanthanides remained elusive. Here, we overcome this gap by describing a multidimensional genome-wide toxicogenomic study for two early lanthanides, namely, lanthanum and praseodymium. We used Saccharomyces cerevisiae as a model system since its genome shares many biological pathways with humans. By performing functional analysis and protein-protein interaction network analysis, we identified the main genes and proteins that participate in the yeast response to counter metal harmful effects. Moreover, our analysis also highlighted key enzymes that are dysregulated by early lanthanides, inducing cytotoxicity. Several of these genes and proteins have human orthologues, indicating that they may also participate in the human response against the metals. By highlighting the key genes and proteins in lanthanide-induced toxicity, this work may contribute to the development of new prophylactic and therapeutic strategies against lanthanide harmful exposures.

Abstract

Targeted alpha therapy offers unique opportunities for the treatment of tumours and infections. Here, we report the development of a new radioimmunoconjugate construct that targets SARS-CoV-2 infected cells, which act as viral reservoirs and promote virus replication and infection spread. The chosen antibody selectively binds to the ACE2-receptor binding domain of the spike protein, and prevents the protein binding to the receptor. Furthermore, the antibody has been radiolabelled with 225Ac, and the therapeutic performance of the resulting radioimmunoconjugate has been demonstrated in vitro against cells mimicking SARS-CoV-2 infection.

Abstract

Massive stars disrupt their natal molecular cloud material through radiative and mechanical feedback processes. These processes have profound effects on the evolution of interstellar matter in our Galaxy and throughout the Universe, from the era of vigorous star formation at redshifts of 1-3 to the present day. The dominant feedback processes can be probed by observations of the Photo-Dissociation Regions (PDRs) where the far-ultraviolet photons of massive stars create warm regions of gas and dust in the neutral atomic and molecular gas. PDR emission provides a unique tool to study in detail the physical and chemical processes that are relevant for most of the mass in inter- and circumstellar media including diffuse clouds, proto-planetary disks and molecular cloud surfaces, globules, planetary nebulae, and star-forming regions. PDR emission dominates the infrared (IR) spectra of star-forming galaxies. Most of the Galactic and extragalactic observations obtained with the James Webb Space Telescope (JWST) will therefore arise in PDR emission. In this paper we present an Early Release Science program using the MIRI, NIRSpec, and NIRCam instruments dedicated to the observations of an emblematic and nearby PDR: the Orion Bar. These early JWST observations will provide template datasets designed to identify key PDR characteristics in JWST observations. These data will serve to benchmark PDR models and extend them into the JWST era. We also present the Science-Enabling products that we will provide to the community. These template datasets and Science-Enabling products will guide the preparation of future proposals on star-forming regions in our Galaxy and beyond and will facilitate data analysis and interpretation of forthcoming JWST observations., Comment: Submitted to PASP
