2. Effects of inulin supplementation on selected faecal characteristics and health of neonatal Saanen kids sucking milk from their dams

Fifty newborn Saanen kids were used to study the effects of inulin supplementation on faecal score, faecal pH, selected faecal bacterial population, BW, body temperature, haematological traits, selected health parameters and the incidence of diarrhoea. Kids were sorted by parity of their dams and multiple birth (twin or triplet) and assigned to one of the two groups (control: CG, and experimental: EG) at birth. Each group consisted of 25 kids. The groups were similar with regard to sex and birth weight. All kids were fed colostrum for the first 3 days after birth, and then the kids in EG were adapted to inulin supplementation by an increased dosage from day 4 to 7. Each kid in EG was supplemented with 0.2 g, 0.3 g, 0.4 g, 0.5 g and 0.6 g inulin on day 4, 5, 6, 7 and from day 8 to 28, respectively, whereas the kids in CG did not receive inulin. Faecal score and faecal bacterial population were not affected by inulin supplementation (P > 0.05). There were differences in faecal pH on day 14 (P = 0.01) and 28 (P 0.05) was detected between groups. No differences (P > 0.05) in BW and haematological traits were found between groups. Body temperature did not differ on day 14 and 21 (P > 0.05), whereas there was a difference in body temperature on day 28 (P = 0.01) between groups. The numbers of kids with pneumonia and kids treated for pneumonia and diarrhoea were similar for CG and EG. Kid losses during the study were the same for CG and EG. The incidence of diarrhoea was not affected by inulin supplementation (P > 0.05). Inulin supplemented to kids did not adversely affect faecal score. The effect of inulin on faecal pH was not consistent. The results of our study suggested that daily dose (0.6 g) of inulin might not be enough to observe effects of it. Our data will be useful to determine the dose and timing of inulin supplementation in future studies investigating the effects of inulin on the parameters associated with performance and health status in kids and other young ruminants.

- Abstract
Vitamin D deficiency is an important public health problem as it is a major risk factor for many diseases. Fortification of foods with vitamin D to increase serum vitamin D levels is an important approach in addressing the current problem and milk is the most commonly used food source for fortification. Over the years, however, milk consumption has decreased, particularly due to lactose intolerance. Yogurt, considered as a functional food, is a significant nutrient due to its high nutritional value, immune system booster and disease risk reducing effects as well as humans' tolerance to even high levels of consumption without any side effects. For this reason, fortification of vitamin D with yogurt may be expected to effectively reduce or prevent diseases related to vitamin D deficiency. In this review, the effects of vitamin D on health, stability of vitamin D during shelf life of vitamin D fortified yogurt, and the metabolic effects of regular consumption have been described in the context of the literature. Bu çalışma, Bursa Uludağ Üniversitesi Sağlık Bilimleri Enstitüsünde Artun Yıbar'ın danışmanlığında Seva Can Küçük tarafından yazılan "50 yaş ve üzeri kadınlarda D vitamini ve probiyotik takviye edilmiş yoğurdun günlük tüketiminin yaşam kalitesi, depresyon, antropometrik ölçümler ve bazı kan parametreleri üzerine etkisi" adlı yüksek lisans tezine dayanılarak hazırlanmıştır. Bu araştırmada, postmenopozal kadınların probiyotik bilgi düzeylerini ve probiyotik tüketim durumlarını belirlemek amaçlanmıştır. Bu kesitsel araştırma, Mart 2019 tarihinde Bursa’da Milli Eğitim Bakanlığı’na bağlı bir hobi kursuna giden 50 yaş ve üzeri 150 kadın ile gerçekleştirilmiştir. Araştırma verileri yüz yüze görüşülerek anket yöntemiyle toplanmıştır. Kadınların yaşları 50-70 arasında değişmekte olup, yaş ortalaması 55,26+5,11 yıldır. Kadınların %66,7’si probiyotik terimini bilmekte olup %26’sı probiyotik katkılı gıda tüketmektedir. Probiyotik katkılı gıda tüketen kadınların %86,6’sı, sağlık yararı gördüğü için tüketmektedir. Tüketmeyen kadınların ise %36,4’ü pahalı bulduğu, %30,3’ü ihtiyaç duymadığı, %27,3’ü doğal bulmadığı ve %6,1’i ise bu gıdaların neler olduğunu bilmediği için tüketmemektedir. Elde edilen verilere göre, kadınlar arasında probiyotik terimi yanlış ve eksik bilinmektedir. Ayrıca, probiyotik ve fermente gıdalar ile ilgili bilgi karmaşası da söz konusudur. This study aimed to determine the probiotic knowledge levels and probiotic consumption characteristics of postmenopausal women. This cross-sectional study was conducted with 150 women aged 50 and over who attended a hobby course affiliated with the Ministry of National Education in Bursa in March 2019. Research data were collected by face-to-face interview method. Ages of women varied between 50-70 years, with an average age of 55.26+5.11 years. A total of 66.7% of the women were aware of the term “probiotic” and 26% consumed probiotic-added food. A total of 86.6% of the women were consuming food with probiotic additives because they believed they obtained health benefits. On the other hand, 36.4% of the women stated that they did not prefer probiotic food because they found it expensive, 30.3% believed they did not need it, 27.3% did not find it natural, and 6.1% did not know what these foods were. According to the data obtained, the term probiotic is known incorrectly and incompletely among women. There is also a confusion of information regarding probiotics and fermented foods.

13. Evaluation of the relationship between sleep quality, diet quality and appetite in adult individuals

Amaç: Bu çalışmada yetişkin bireylerde uyku kalitesi, diyet kalitesi ve iştah arasındaki ilişkinin araştırılması amaçlandı. Gereç ve Yöntem: Bu çalışma, Bursa ilinde özel bir psikolojik danışmanlık merkezine başvuran, yan etkisi veya etkisi uyku arttırıcı veya düzenleyici herhangi bir ilaç, takviye kullanmayan, 18 yaş ve üzeri 50 katılımcı ile yürütüldü. Araştırmaya katılanlara kişisel bilgi formu, besin tüketim sıklığı anketi, 24 Saatlik Besin Tüketim Kaydı, Üç Faktörlü Beslenme Anketi, Duygusal İştah Anketi (DİA) ve Pittsburgh Uyku Kalitesi İndeksi (PUKİ) uygulandı ve vücut kompozisyon analizi standartlara uygun bir şekilde yapıldı. Diyet kalite puanı Sağlıklı Yeme İndeksi(2015) kullanılarak hesaplandı. Bulgular: Katılımcıların %52'sinin uyku kalitesi iyi iken, % 8'inin uyku kalitesinin kötü olduğu tespit edildi. Toplam PUKİ puanı ortalaması 6,16± ,11 olarak bulundu. Uyku kalitesi iyi olan grup ile kötü olan grup arasında diyet kalite puanı arasında anlamlı bir fark görülmedi (p>0,05). Diyet kalite faktörlerinden sadece tam tahıl puanı PUKİ toplam uyku kalitesi puanı ile negatif ilişkiliydi. Tam tahıl puanı aynı zamanda DİA pozitif durum ve toplam puanları ile pozitif ilişkiydi. Toplam PUKİ puanı ile tüketilen kafein miktarları arasında anlamlı pozitif yönde ilişki varken (p0,05). Uyku kalitesi grupları ve toplam uyku kalite puanı ile duygusal iştah arasında anlamlı bir ilişki bulunamadı (p>0,05). Sonuç: Diyet kalitesi ve uyku kalitesinin belirlenmesi ve değerlendirilmesi uyku ve beslenme arasındaki karmaşık ilişkiyi çözmeye yardımcı olur. Beslenme davranışları ve iştah hem dolaylı hem de doğrudan uyku kalitesi ve diyet kalitesini etkiler bu nedenle bu alanlarda da daha fazla araştırmaya ihtiyaç vardır. Aim: In this study, it was aimed to investigate the relationship between sleep quality, diet quality and emotional appetite in adults. Materials and Methods: This study was conducted with 50 participants over the age of 18 who applied to a private psychological counseling center in Bursa, did not use any sleep-enhancing or regulating drugs and supplements. Sleep quality was assessed through Pittsburgh Sleep Quality Index (PSQI), dietary quality through Healthy Eating Index 2015 (HEI 2015), emotional appetite through Emotional Appetite Questionnaire (EMAQ), and dietary habits through Three Factor Eating Questionnaire (TFEQ). Body composition and anthropometric measurements were performed in accordance with predetermined standards. Results: While 52% of the participants had good sleep quality, 48% of them had poor sleep quality. The mean total PUKI score was 6.16± .11. There was no significant difference in diet quality score between the groups with good sleep quality and the group with poor sleep quality (p>0.05). Only the whole grain score, which is one of the diet quality factors, was negatively associated with the PSQI total sleep quality score. Whole grain score was also positively correlated with EMAQ positive status and total scores. While there was a significant positive correlation between the total PSQI score and the amount of caffeine consumed (p0.05). There was no significant relationship between sleep quality groups and total sleep quality score and emotional appetite (p>0.05). Conclusion: Determination and evaluation of diet quality and sleep quality helps to unravel the complex relationship between sleep and nutrition. Nutritional behaviors and appetite both indirectly and directly affect sleep quality and diet quality, so more research is needed in these areas.

Amaç: Bu çalışmada, D vitamini eksikliği/yetersizliği olan 50 yaş ve üzeri kadınlarda D vitamini ve probiyotik takviye edilmiş yoğurdun günlük tüketiminin yaşam kalitesi, depresyon, antropometrik ölçümler ve bazı kan parametreleri üzerine etkisinin araştırılması amaçlandı.Gereç ve Yöntem: Bu çalışmada, Mart- Mayıs 2019 tarihleri arasında Bursa'da bir hobi kursuna giden 50 yaş ve üzeri kadınlar üç ayrı durumda ziyaret edildi. İlk ziyarette kan örnekleri alındı ve serum 25(OH)D düzeyi 30 ng/mL'den düşük olan 150 kadın ile çalışmaya devam edildi. Ardından, genel özellikler, yaşam kalitesi ve depresyon ile ilgili anketler tamamlandı. Antropometrik ölçümler ise standartlara uygun şekilde ölçüldü. Daha sonra; kontrol grubu (Grup 1), yoğurt tüketen grup (Grup 2), yoğurt ile birlikte 5x109 kob/g probiyotik takviyesi alan grup (Grup 3), yoğurt ile birlikte D vitamini alan grup (Grup 4), yoğurt ile birlikte D vitamini ve probiyotik takviyesi alan grup olmak üzere beş farklı çalışma grubu oluşturuldu. Çalışmanın 12. haftasındaki üçüncü ziyarette tekrar kan örnekleri alındı ve birinci ziyarette uygulanan anketler ve ölçümler tekrar gerçekleştirildi. Bulgular: Kadınların %21,3'ünde D vitamini yetersizliği, %78,7'sinde ise D vitamini eksikliği vardı. Tüm gruplar arasındaki başlangıç antropometrik ölçümler, kan bulguları, yaşam kalitesi ve depresyon skorları açısından anlamlı bir farklılık saptanmadı (p>0,05). Grup 1, Grup 2, Grup 3 ve Grup 4'teki kadınlara kıyasla Grup 5'teki kadınların vücut ağırlıkları ortalamalarının daha fazla azaldığı, serum 25(OH)D seviyelerinin daha fazla yükseldiği ve depresyon skorlarının daha fazla iyileştiği tespit edildi (p0,05). The main objectives of our study were to measure the major adipokines adiponectin, leptin and resistin in goat milk, to assess their interrelationships and to assess their relationships with the plasma and serum concentrations of total protein, cholesterol, total lipids, plasma C-reactive protein (CRP), milk somatic cell count (SCC), milk total aerobic colony and lactobacillus count, and milk components in lactating Saanen goats. The study was performed on eighteen lactating Saanen goats. Milk and blood samples were collected on days 20, 35, 50, 65 and 80 of lactation postpartum. The milk and plasma adiponectin levels on days 50, 65 and 80 postpartum were significantly higher than those on day 20. The milk and plasma leptin levels were lower on day 20 than on days 35, 50, 65 and 80. The milk concentrations of these major adipokines were positively intercorrelated. The milk and plasma concentrations of these three adipokines were also positively correlated. The plasma CRP concentrations correlated positively with milk leptin and resistin concentrations and inversely with milk adiponectin concentration. Milk adiponectin concentration was inversely related with its SCC. These data confirm that adiponectin, leptin and resistin are present in goat milk. The milk concentrations of these three adipokines were interrelated and interacted with the general inflammatory marker, CRP. The inverse relationship between milk adiponectin concentrations and its SCC suggests that variations in milk adiponectin might be involved in the udder health of lactating goats, but clinical trials are needed to support this hypothesis.

Bacillus cereus is a type of bacteria that can cause severe food poisoning. The aim of this study was to determine the incidence of B. cereus in various full-fat milk and cheese samples and to assess the HBL (haemolysin BL) and NHE (nonhaemolytic enterotoxin) production and the resistance to several antimicrobial agents of the isolates. A total of 259 samples of full-fat milk (raw, pasteurized and UHT) and cheese obtained from different retail markets in Bursa province between July and December 2013 were analysed. Isolation of B. cereus was performed using Bacara agar according to the method suggested by FDA. Twenty six (10.04%) out of 259 samples were found to be contaminated with presumptive B. cereus based on their colony morphology and microscopic appearance, by counts that ranged from 1x10(1) to 1.1x10(3) CFU/mL in raw and pasteurized milk and from 4x10(1) to 3.8x10(5) CFU/g in cheese. Thirteen isolates of B. cereus were identified by API system. However, further analysis using MALDI-TOF-MS confirmed 19 isolates as B. cereus. Thirteen out of 19 (68.4%) isolates showed evidence of only NHE toxin production while six out of 19 (31.6%) isolates were positive for both NHE and HBL production. All isolates were resistant to penicillin G, although they were susceptible to oleondamycin, erythromycin and streptomycin. There were seven different patterns of multiple antibiotic resistance in this study. In our study, 84.2% (n = 16) of B. cereus isolates exhibited multiple antibiotic resistance. B.cereus izolasyonu FDA tarafından önerilen metoda göre Bacara agar kullanılarak yapıldı. Analize alınan çiğ ve pastörize süt örneklerinde 1x101 ile 1.1x103 kob/mL ve peynirde 4x101 ile 3.8x105 kob/g arasında değişen sayılarda olmak üzere, toplamda 259 örneğin 26 adetinin (%10.04) koloni morfolojileri ve mikroskobik görünüşlerine de dayalı olarak B. cereus ile kontamine olduğu tespit edilmiştir. Örneklerden elde edilen 13 adet B. cereus izolatı API sistemi ile identifiye edilmiştir. Bununla birlikte, MALDI-TOF-MS kullanılarak yapılan analizde, 19 adet izolat B. cereus olarak identifiye edilmiştir. Bu 19 izolatın 13’ünün (%68.4) sadece NHE toksini bakımından, altı izolatın da (%31.6) NHE ve HBL toksinleri bakımından pozitif olduğu gözlenmiştir. Tüm izolatlar oleondamisin, eritromisin ve streptomisine duyarlı olmalarına rağmen penisilin G’ye dirençlidir. Bu çalışmada, çoklu antibiyotik direnci gösteren yedi farklı model bulunmuştur. Çalışmamızda, B. cereus izolatlarının %84.2’si (n = 16) çoklu antibiyotik direnci göstermiştir.

Son birkaç yıl içinde hayvan refahı birçok ülkede ve Avrupa Birliği’nde önemli hale gelmiştir. Tüm etçi hayvanlar kesim öncesinde bazı düzeylerde stres yaşayabilmektedirler ve bu da et kalitesi üzerinde zararlı etkiler oluşturabilmektedir. Kesim öncesi koşullar ve taşıma, mezbahada bekleme, taşımada kullanılan araçlar, diğer müdahale işlemleri ve olumsuz mevsimsel koşullar gibi süregelen çevresel stres faktörleri hayvanlarda stres, çeşitli davranışsal değişiklikler ve bağışıklık sistemlerinin olumsuz yönde etkilenmesi gibi değişikliklere sebep olabilmektedir. Uygun olmayan kesim öncesi koşulların bir sonucu olarak kas glikojen depoları tükenmekte, bunun sonucunda karkas ve et kalite özellikleri (yüksek et Ph’sı, daha fazla su tutma kapasitesi, koyu et rengi ve sert et oluşumu) etkilenmektedir ve bu yüzden ekonomik kayıplar oluşmaktadır. Ekonomik ve kalite kayıplarını önlemek için kesim öncesi hayvan refahı uygulamalarına dikkat edilmelidir. In the last few years animal welfare has become an important issue in many countries and the European Union. All meat animals will experience some level of stress prior to slaughter and this in turn, may have detrimental effects to meat quality. Pre-slaughter conditions and continued environmental stressors such as duration of transportation, waiting time in slaughterhouse pens, the models of vehicles used for transportation, extra handling procedures, adverse seasonal conditions can potentially stress animals and consequently, alter behavioural or immune responses. As a consequence of the improper pre-slaughter conditions, muscle glycogen reserves are depleted before slaughter, which may result in carcass and meat quality characteristics (e.g., higher ultimate meat pH, greater waterholding capacity, darker meat colour and tougher meat) and therefore may cause economic loss. Animal welfare practices should be performed carefully in pre-slaughter period to avoid the quality problems and production losses

In the present study, a total of 512 food samples composed of raw milk, dairy products, meat/meat products, chicken meat, seafood and raw vegetables were analyzed for the presence of Listeria monocytogenes. The results of the standard identification methods showed that 20 (3.9%) of the analyzed samples were found to harbor this pathogen. Further, 8.4% (13/155) of chicken meats, 0.9% (1/105) of meat/meat products and 13.6% (6/44) of fresh vegetables were contaminated with L.monocytogenes. Interestingly, only 18 of these isolates gave expected band size when they were subjected to molecular confirmation by polymerase chain reaction (PCR). Multiplex PCR serotyping of the strains revealed that 66.6% (12/18) of which belonged to serotype 1/2a (or 3a), 5.6% (1/18) to serotype 1/2b (or 3b, 7), 5.6% (1/18) to serotype 1/2c (or 3c) and 11.1% (2/18) to serotype 4b (or 4d, 4e). Two strains could not be serotyped by multiplex PCR. The strains were also evaluated by disk diffusion assay for their susceptibility to 15 commonly used antimicrobials. Antimicrobial resistance was most frequently observed for clindamycin (94.4%), followed by streptomycin and kanamycin (88.9%); penicillin (72.2%), tetracycline and gentamicin (66.7%); quinopristin/dalfopristin and erythromycin (61.1). Interestingly, 13 strains were resistant to more than five antibiotics. All strains were susceptible to linezolid, teicoplanin and vancomycin. Practical ApplicationsListeria monocytogenes is one of the most important foodborne pathogens responsible for several outbreaks and cases of listeriosis in human. This study focused on the prevalence of L.monocytogenes in different raw and ready-to-eat foodstuffs, and serotype distribution among the isolates. Antibiotic resistance profiles of the isolates were also reported. Information and relief provided to consumers could help elaborate public health and food safety.

Enzyme Linked Immunosorbent Assay (ELISA) method has been implemented to screen the residues of nitrofuran metabolite AOZ in chicken livers obtained from local markets in Bursa province of Turkey. For this purpose, ELISA kit specific for this metabolite was used. ELISA screening demonstrated the presence of AOZ residues in 11 of 90 chicken liver samples, with a positive rate of 12.2%. The residual level of nitrofuran metabolite AOZ in chicken livers ranged from 103.8 to 1027.8 ng kg(-1), having a mean of 212.2 ng kg(-1). Present results showed that the use of these compounds in broiler has the potential and public awareness should be raised to prevent the usage of AOZ in livestock production.

Analysis of residual levels of tetracyclines (TCs) in chicken meat was performed using a validated liquid chromatography coupled with a tandem mass spectrometry (LC-MS/MS) technique. Overall, the recoveries for TCs ranged from 56.9% to 101.2%, with standard deviations of 4.5-13.2%. Detection limits ranged from 7.9 to 14.6 mu g kg(-1). In four of 60 samples, doxycycline (DXC) was determined in a range from 19.9 to 35.6 mu g kg(-1); and in one sample tetracycline was detected at 17.2 mu g kg(-1). Chlortetracycline (CTC) and oxytetracycline (OTC) were not detected in any of the tested samples. This study indicates that chicken meat sold in Bursa, Turkey, contained some residues of TCs. Therefore, stricter regulations for the use of antibiotics in the poultry industry and the monitoring of drug residues in chicken meat prior to marketing are needed. Finally, this method has been applied successfully for the confirmation of TCs in chicken meat.

Thirty seven strains of Lactococcus laths subsp. locus and 5 strains of L. locus subsp. cremoris isolated from White Pickled and Kashar cheeses were identified according to phenotypic and molecular techniques using subspecies specific primers. Then, acidification, proteolysis and autolysis of strains were assessed in order to select bacteria for starter development. The strains were divided into slow, moderate and rapid acidifying groups according to their acidifications (P < 0.05) in reconstituted skim milk (RSM). Acidification of strains from White Pickled cheese was significantly higher (P < 0.05) compared to those of Kashar. Majority (86%) of the isolates exhibited Prt(+) phenotype. Bacterial autolysis which was tested under starvation conditions evidenced that strains have an autolytic extent at the end of 24 h ranging from 37 to 70%. The results of this study showed a large diversity among the isolates and provide criteria for choosing strains to be tested as starters in cheesemaking. experimentation.
