Abstract

We investigate the mean-field dynamics of stochastic McKean differential equations with heterogeneous particle interactions described by large network structures. To express a wide range of graphs, from dense to sparse structures, we incorporate the recently developed graph limit theory of graphops into the limiting McKean--Vlasov equations. Global stability of the splay steady state is proven via a generalized entropy method, leading to explicit graph-structure dependent decay rates. We highlight the robustness of the entropy approach by extending the results to the closely related Sakaguchi-Kuramoto model with intrinsic frequency distribution. We also present central examples of random graphs, such as power law graphs and the spherical graphop, and analyze the limitations of the applied methodology.

3. A minimax optimal control approach for robust neural ODEs

Abstract

In this paper, we address the adversarial training of neural ODEs from a robust control perspective. This is an alternative to the classical training via empirical risk minimization, and it is widely used to enforce reliable outcomes for input perturbations. Neural ODEs allow the interpretation of deep neural networks as discretizations of control systems, unlocking powerful tools from control theory for the development and the understanding of machine learning. In this specific case, we formulate the adversarial training with perturbed data as a minimax optimal control problem, for which we derive first order optimality conditions in the form of Pontryagin's Maximum Principle. We provide a novel interpretation of robust training leading to an alternative weighted technique, which we test on a low-dimensional classification task., Comment: 7 pages, 2 figures and 1 table. Abstract

The goal of this work is to find the sharp rate of convergence to equilibrium under the quadratic Fisher information functional for solutions to Fokker-Planck equations governed by a constant drift term and a constant, yet possibly degenerate, diffusion matrix. A key ingredient in our investigation is a recent work of Arnold, Signorello, and Schmeiser, where the $L^2$-propagator norm of such Fokker-Planck equations was shown to be identical to the propagator norm of a finite dimensional ODE which is determined by matrices that are intimately connected to those appearing in the associated Fokker-Planck equations.

Abstract

The goal of this work is to introduce and investigate a generalised Fisher Information in the setting of linear Fokker-Planck equations. This functional, which depends on two functions instead of one, exhibits the same decay behaviour as the standard Fisher information, and allows us to investigate different parts of the Fokker-Planck solution via an appropriate decomposition. Focusing almost exclusively on Fokker-Planck equations with constant drift and diffusion matrices, we will use a modification of the well established Bakry-Emery method with this newly defined functional to conclude the sharp long time behaviour of relative entropies of solutions to such equations when the diffusion matrix is positive definite and the drift matrix is defective. This novel approach is different to previous techniques, such as those presented in the authors' previous work, which relied heavily on spectral theory.

Abstract

This paper is dealing with two $L^2$ hypocoercivity methods based on Fourier decomposition and mode-by-mode estimates, with applications to rates of convergence or decay in kinetic equations on the torus and on the whole Euclidean space. The main idea is to perturb the standard $L^2$ norm by a twist obtained either by a nonlocal perturbation build upon diffusive macroscopic dynamics, or by a change of the scalar product based on Lyapunov matrix inequalities. We explore various estimates for equations involving a Fokker-Planck and a linear relaxation operator. We review existing results in simple cases and focus on the accuracy of the estimates of the rates. The two methods are compared in the case of the Goldstein-Taylor model in one-dimension.

Abstract

Blockchain has evolved into a platform for decentralized applications, with beneficial properties like high integrity, transparency, and resilience against censorship and tampering. However, blockchains are closed-world systems which do not have access to external state. To overcome this limitation, oracles have been introduced in various forms and for different purposes. However so far common oracle best practices have not been dissected, classified, and studied in their fundamental aspects. In this paper, we address this gap by studying foundational blockchain oracle patterns in two foundational dimensions characterising the oracles: (i) the data flow direction, i.e., inbound and outbound data flow, from the viewpoint of the blockchain; and (ii) the initiator of the data flow, i.e., whether it is push or pull-based communication. We provide a structured description of the four patterns in detail, and discuss an implementation of these patterns based on use cases. On this basis we conduct a quantitative analysis, which results in the insight that the four different patterns are characterized by distinct performance and costs profiles.

Abstract

The Goldstein-Taylor equations can be thought of as a simplified version of a BGK system, where the velocity variable is constricted to a discrete set of values. It is intimately related to turbulent fluid motion and the telegrapher's equation. A detailed understanding of the large time behaviour of the solutions to these equations has been mostly achieved in the case where the relaxation function, measuring the intensity of the relaxation towards equally distributed velocity densities, is constant. The goal of the presented work is to provide a general method to tackle the question of convergence to equilibrium when the relaxation function is not constant, and to do so as quantitatively as possible. In contrast to the usual modal decomposition of the equations, which is natural when the relaxation function is constant, we define a new Lyapunov functional of pseudodifferential nature, one that is motivated by the modal analysis in the constant case, that is able to deal with full spatial dependency of the relaxation function. The approach we develop is robust enough that one can apply it to multi-velocity Goldstein-Taylor models, and achieve explicit rates of convergence. Abstract

Recent Alzheimer's disease (AD) patient studies have focused on retinal analysis, as the retina is the only part of the central nervous system which can be imaged non-invasively by optical methods. However as this is a relatively new approach, the occurrence and role of pathological features such as retinal layer thinning, extracellular amyloid beta (A$\beta$) accumulation and vascular changes is still debated. Animal models of AD are therefore often used in attempts to understand the disease. In this work, both eyes of 24 APP/PS1 transgenic mice (age: 45-104 weeks) and 15 age-matched wildtype littermates were imaged by a custom-built multi-contrast optical coherence tomography (OCT) system. The system provided a combination of standard reflectivity data, polarization-sensitive data and OCT angiograms. This tri-fold contrast provided qualitative and quantitative information on retinal layer thickness and structure, presence of hyper-reflective foci, phase retardation abnormalities and retinal vasculature. While abnormal structural properties and phase retardation signals were observed in the retinas, the observations were very similar in transgenic and control mice. At the end of the experiment, retinas and brains were harvested from a subset of the mice (14 transgenic, 7 age-matched control) in order to compare the in vivo results to histological analysis, and to quantify the cortical A$\beta$ plaque load.

Abstract

Rapid technological progress in computer sciences finds solutions and at the same time creates ever more complex requirements. Due to an evolving complexity todays programming languages provide powerful frameworks which offer standard solutions for recurring tasks to assist the programmer and to avoid the re-invention of the wheel with so-called out-of-the-box-features. In this paper, we propose a way of comparing different programming paradigms on a theoretical, technical and practical level. Furthermore, the paper presents the results of an initial comparison of two representative programming approaches, both in the closed SAP environment.

Abstract

The popularity of cloud based Infrastructure-as-a- Service (IaaS) solutions is becoming increasingly popular. However, since IaaS providers and customers interact in a flexible and scalable environment, security remains a serious concern. To handle such security issues, defining a set of security parameters in the service level agreements (SLA) between both, IaaS provider and customer, is of utmost importance. In this paper, the European Network and Information Security Agency (ENISA) guidelines are evaluated to extract a set of security parameters for IaaS. Furthermore, the level of applicability and implementation of this set is used to assess popular industrial and open-source IaaS cloud platforms, respectively VMware and OpenStack. Both platforms provide private clouds, used as backend infrastructures in Industry 4.0 application scenarios. The results serve as initial work to identify a security baseline and research needs for creating secure cloud environments for Industry 4.0.

13. Sharp Decay Estimates in Local Sensitivity Analysis for Evolution Equations with Uncertainties: from ODEs to Linear Kinetic Equations

Abstract

We review the Lyapunov functional method for linear ODEs and give an explicit construction of such functionals that yields sharp decay estimates, including an extension to defective ODE systems. As an application, we consider three evolution equations, namely the linear convection-diffusion equation, the two velocity BGK model and the Fokker-Planck equation. Adding an uncertainty parameter to the equations and analyzing its linear sensitivity leads to defective ODE systems. By applying the Lyapunov functional construction, we prove sharp long time behavior of order $(1+t^{M})e^{-\mu t}$, where $M$ is the defect and $\mu$ is the spectral gap of the system. The appearance of the uncertainty parameter in the three applications makes it important to have decay estimates that are uniform in the non-defective limit., Comment: Minor corrections for clarity

Abstract

The evolutionary processes that drive universal therapeutic resistance in adult patients with diffuse glioma remain unclear1,2. Here we analysed temporally separated DNA-sequencing data and matched clinical annotation from 222 adult patients with glioma. By analysing mutations and copy numbers across the three major subtypes of diffuse glioma, we found that driver genes detected at the initial stage of disease were retained at recurrence, whereas there was little evidence of recurrence-specific gene alterations. Treatment with alkylating agents resulted in a hypermutator phenotype at different rates across the glioma subtypes, and hypermutation was not associated with differences in overall survival. Acquired aneuploidy was frequently detected in recurrent gliomas and was characterized by IDH mutation but without co-deletion of chromosome arms 1p/19q, and further converged with acquired alterations in the cell cycle and poor outcomes. The clonal architecture of each tumour remained similar over time, but the presence of subclonal selection was associated with decreased survival. Finally, there were no differences in the levels of immunoediting between initial and recurrent gliomas. Collectively, our results suggest that the strongest selective pressures occur during early glioma development and that current therapies shape this evolution in a largely stochastic manner.

Abstract

Optical coherence tomography (OCT) is a powerful technology for rapid volumetric imaging in biomedicine. The bright field imaging approach of conventional OCT systems is based on the detection of directly backscattered light, thereby waiving the wealth of information contained in the angular scattering distribution. Here we demonstrate that the unique features of few-mode fibers (FMF) enable simultaneous bright and dark field (BRAD) imaging for OCT. As backscattered light is picked up by the different modes of a FMF depending upon the angular scattering pattern, we obtain access to the directional scattering signatures of different tissues by decoupling illumination and detection paths. We exploit the distinct modal propagation properties of the FMF in concert with the long coherence lengths provided by modern wavelength-swept lasers to achieve multiplexing of the different modal responses into a combined OCT tomogram. We demonstrate BRAD sensing for distinguishing differently sized microparticles and showcase the performance of BRAD-OCT imaging with enhanced contrast for ex vivo tumorous tissue in glioblastoma and neuritic plaques in Alzheimer's disease.

Abstract

We establish sharp long time asymptotic behaviour for a family of entropies to defective Fokker-Planck equations and show that, much like defective finite dimensional ODEs, their decay rate is an exponential multiplied by a polynomial in time. The novelty of our study lies in the amalgamation of spectral theory and a quantitative non-symmetric hypercontractivity result, as opposed to the usual approach of the entropy method.

Abstract

IntroductionAmino-acid positron emission tomography (PET) is a validated metabolic imaging approach for the diagnostic work-up of gliomas. This study aimed to evaluate sex-specific radiomic characteristics of L-[S-methyl-11Cmethionine (MET)-PET images of glioma patients in consideration of the prognostically relevant biomarker isocitrate dehydrogenase (IDH) mutation status.MethodsMET-PET of 35 astrocytic gliomas (13 females, mean age 41 ± 13 yrs. and 22 males, mean age 46 ± 17 yrs.) and known IDH mutation status were included. All patients underwent radiomic analysis following imaging biomarker standardization initiative (IBSI)-conform guidelines both from standardized uptake value (SUV) and tumor-to-background ratio (TBR) PET values. Aligned Monte Carlo (MC) 100-fold split was utilized for SUV and TBR dataset pairs for both sex and IDH-specific analysis. Borderline and outlier scores were calculated for both sex and IDH-specific MC folds. Feature ranking was performed by R-squared ranking and Mann-Whitney U-test together with Bonferroni correction. Correlation of SUV and TBR radiomics in relation to IDH mutational status in male and female patients were also investigated.ResultsThere were no significant features in either SUV or TBR radiomics to distinguish female and male patients. In contrast, intensity histogram coefficient of variation (ih.cov) and intensity skewness (stat.skew) were identified as significant to predict IDH +/-. In addition, IDH+ females had significant ih.cov deviation (0.031) and mean stat.skew (-0.327) differences compared to IDH+ male patients (0.068 and -0.123, respectively) with two-times higher standard deviations of the normal brain background MET uptake as well.DiscussionWe demonstrated that female and male glioma patients have significantly different radiomic profiles in MET PET imaging data. Future IDH prediction models shall not be built on mixed female-male cohorts, but shall rely on sex-specific cohorts and radiomic imaging biomarkers.

Abstract

PurposeModern techniques for improved tumor visualization have the aim to maximize the extent of resection during brain tumor surgery and thus improve patient prognosis. Optical imaging of autofluorescence is a powerful and non-invasive tool to monitor metabolic changes and transformation in brain tumors. Cellular redox ratios can be retrieved from fluorescence emitted by the coenzymes reduced nicotinamide adenine dinucleotide (phosphate) (NAD(P)H) and flavin adenine dinucleotide (FAD). Recent studies point out that the influence of flavin mononucleotide (FMN) has been underestimated.Experimental designFluorescence lifetime imaging and fluorescence spectroscopy were performed through a modified surgical microscope. We acquired 361 flavin fluorescence lifetime (500-580 nm) and fluorescence spectra (430-740 nm) data points on freshly excised different brain tumors: low-grade gliomas (N=17), high-grade gliomas (N=42), meningiomas (N=23), metastases (N=26) and specimens from the non-tumorous brain (N=3).ResultsProtein-bound FMN fluorescence in brain tumors did increase with a shift toward a more glycolytic metabolism (R=-0.87). This increased the average flavin fluorescence lifetime in tumor entities with respect to the non-tumorous brain. Further, these metrics were characteristic for the different tumor entities and showed promise for machine learning based brain tumor classification.ConclusionsOur results shed light on FMN fluorescence in metabolic imaging and outline the potential for supporting the neurosurgeon in visualizing and classifying brain tumor tissue during surgery.

Abstract

Adult diffuse gliomas are a diverse group of brain neoplasms that inflict a high emotional toll on patients and their families. The Cancer Genome Atlas and similar projects have provided a comprehensive understanding of the somatic alterations and molecular subtypes of glioma at diagnosis. However, gliomas undergo significant cellular and molecular evolution during disease progression. We review the current knowledge on the genomic and epigenetic abnormalities in primary tumors and after disease recurrence, highlight the gaps in the literature, and elaborate on the need for a new multi-institutional effort to bridge these knowledge gaps and how the Glioma Longitudinal Analysis Consortium (GLASS) aims to systemically catalog the longitudinal changes in gliomas. The GLASS initiative will provide essential insights into the evolution of glioma toward a lethal phenotype, with the potential to reveal targetable vulnerabilities and, ultimately, improved outcomes for a patient population in need.

Abstract

BackgroundRare multicentric lower-grade gliomas (LGGs) represent a unique opportunity to study the heterogeneity among distinct tumor foci in a single patient and to infer their origins and parallel patterns of evolution.MethodsIn this study, we integrate clinical features, histology, and immunohistochemistry for 4 patients with multicentric LGG, arising both synchronously and metachronously. For 3 patients we analyze the phylogeny of the lesions using exome sequencing, including one case with a total of 8 samples from the 2 lesions.ResultsOne patient was diagnosed with multicentric isocitrate dehydrogenase 1 (IDH1) mutated diffuse astrocytomas harboring distinct IDH1 mutations, R132H and R132C; the latter mutation has been associated with Li-Fraumeni syndrome, which was subsequently confirmed in the patient's germline DNA and shown in additional cases with The Cancer Genome Atlas data. In another patient, phylogenetic analysis of synchronously arising grade II and grade III diffuse astrocytomas demonstrated a single shared mutation, IDH1 R132H, and revealed convergent evolution via non-overlapping mutations in ATRX and TP53. In 2 cases, there was divergent evolution of IDH1-mutated and 1p/19q-codeleted oligodendroglioma and IDH1-mutated and 1p/19q-intact diffuse astrocytoma, occurring synchronously in one case and metachronously in a second.ConclusionsEach tumor in multicentric LGG cases may arise independently or may diverge very early in their development, presenting as genetically and histologically distinct tumors. Comprehensive sampling of these lesions can therefore significantly alter diagnosis and management. Additionally, somatic IDH1 R132C mutation in either multicentric or solitary LGG identifies unsuspected germline TP53 mutation, validating the limited number of published cases.

The major aim of this survey is to identify the strengths and weaknesses of a representative set of Data-Mining and Integration (DMI) query languages. We describe a set of properties of DMI-related languages that we use for a systematic evaluation of these languages. In addition, we introduce a scoring system that we use to quantify our opinion on how well a DMI-related language supports a property. The languages surveyed in this paper include: DMQL, MineSQL, MSQL, M2MQL, dmFSQL, OLEDB for DM, MINE RULE, and Oracle Data Mining. Abstract

The Brain Tumor Epidemiology Consortium (BTEC) is an international consortium that aims to advance the development of multicenter and interdisciplinary collaborations that focus on research related to the etiology, outcomes, and prevention of brain tumors. The 18th annual BTEC meeting was held in Banff, AB, Canada, on June 27 - 29, 2017. The meeting focused on the intersection between epidemiology and precision medicine, that is, the use of molecular indicators of risk, early disease and prognosis or precision epidemiology. While traditional epidemiologic approaches group large numbers of participants for statistical power, precision epidemiology is founded on the uniqueness and biology of individual disease characteristics. With this in mind, plenary speakers described the molecular heterogeneity of adult and pediatric brain tumors and how those characteristics are currently being used to guide therapy and etiologic research. Rare subtypes and novel mechanisms for recruitment of individuals for research on brain tumors were discussed along with concepts and methodology related to biological and etiologic heterogeneity. The incorporation of relevant molecular classifiers into population registries was emphasized for its role in future research endeavors, ensuring the accessibility of such tools for researchers and clinicians seeking to improve the lives of individuals with brain tumors and those at risk. The next BTEC meeting will be held in Copenhagen, Denmark, in June 2018.

Abstract

ObjectiveThe intraoperative visualization of adult-type diffuse gliomas with 5-aminolevulinic acid (5-ALA) induced fluorescence is widely used in the neurosurgical field. While visible 5-ALA induced fluorescence is found in the majority of high-grade gliomas, most low-grade gliomas lack visible fluorescence during surgery. Recently, the heme biosynthesis pathway was identified as crucial influencing factor for presence of visible fluorescence since it metabolizes 5-ALA to fluorescing Protoporphyrin IX (PpIX). However, the exact alterations within the heme biosynthesis pathway resulting in visible 5-ALA induced fluorescence in gliomas are still unclear. The aim of the present study was thus to compare the mRNA and protein expression of promising intramitochondrial heme biosynthesis enzymes/transporters in glioma tissue samples of different fluorescence behavior.MethodsA total of 19 strongly fluorescing and 21 non-fluorescing tissue samples from neurosurgical adult-type diffuse gliomas (WHO grades II-IV) were included in the current analysis. In these samples, we investigated the mRNA expression by quantitative real time PCR and protein expression using immunohistochemistry of the intramitochondrial heme biosynthesis enzymes Coproporphyrinogen Oxidase (CPOX), Protoporphyrinogen Oxidase (PPOX), Ferrochelatase (FECH), and the transporter ATP-binding Cassette Subfamily B Member 2 (ABCG2).ResultsRegarding mRNA expression analysis, we found a significantly decreased ABCG2 expression in fluorescing specimens compared to non-fluorescing samples (p = 0.001), whereas no difference in CPOX, PPOX and FECH was present. With respect to protein expression, significantly higher levels of CPOX (p = 0.005), PPOX (p < 0.01) and FECH (p = 0.003) were detected in fluorescing samples. Similar to mRNA expression analysis, the protein expression of ABCG2 (p = 0.001) was significantly lower in fluorescing samples.ConclusionDistinct alterations of the analyzed heme biosynthesis factors were found primarily on protein level. Our data indicate that heme biosynthesis pathway activity in general is enhanced in fluorescing gliomas with upregulation of PpIX generating enzymes and decreased ABCG2 mediated PpIX efflux outweighing the also increased further metabolization of PpIX to heme. Intramitochondrial heme biosynthesis factors thus constitute promising pharmacological targets to optimize intraoperative 5-ALA fluorescence visualization of usually non-fluorescing tumors such as low-grade gliomas.

Abstract

The Brain Tumor Epidemiology Consortium (BTEC) is an international consortium that aims to advance development of multicenter and interdisciplinary collaborations that focus on research related to the etiology, outcomes, and prevention of brain tumors. The 17th annual BTEC meeting was held in Barcelona, Spain on June 21 - 23, 2016. The meeting focused on immune and viral factors that influence brain tumor development. Fundamentals of innate and adaptive immunity were reviewed, the role of immune checkpoint inhibitors in primary and secondary brain tumors was addressed, vaccination strategies for glioma treatment were presented, and the potential contribution of immune dysfunction and viruses tropic for glial cells in gliomagenesis was discussed. Further contributions addressed the risk of non-ionizing radiation, molecular and birth characteristics on brain tumor induction/outcomes, and patterns of care and effects of different treatments on brain tumor survival in the real world setting. The next BTEC meeting will be held in June 2017 in Banff, Canada, and will focus on brain tumor epidemiology in the era of precision medicine.
