INTRODUCTION: Vascular contributions to cognitive impairment and dementia (VCID) represent a major factor in cognitive decline in older adults. The present study examined the relationship between cerebrovascular reactivity (CVR) measured by magnetic resonance imaging (MRI) and cognitive function in a multi-site study, using a predefined hypothesis. METHODS: We conducted the study in a total of three analysis sites and 263 subjects. Each site performed an identical CVR MRI procedure using 5% carbon dioxide inhalation. A global cognitive measure of Montreal Cognitive Assessment (MoCA) and an executive function measure of item response theory (IRT) score were used as outcomes. RESULTS: CVR and MoCA were positively associated, and this relationship was reproduced at all analysis sites. CVR was found to be positively associated with executive function. DISCUSSION: The predefined hypothesis on the association between CVR and a global cognitive score was validated in three independent analysis sites, providing support for CVR as a biomarker in VCID. HIGHLIGHTS: This study measured a novel functional index of small arteries referred to as cerebrovascular reactivity (CVR). CVR was positively associated with global cognition in older adults. This finding was observed in three independent cohorts at three sites. Our statistical analysis plan was predefined before beginning data collection.

Peroxiredoxin 6 (Prdx6) repairs peroxidized membranes by reducing oxidized phospholipids, and by replacing oxidized sn-2 fatty acyl groups through hydrolysis/reacylation by its phospholipase A2 (aiPLA2) and lysophosphatidylcholine acyltransferase activities. Prdx6 is highly expressed in the lung, and intact lungs and cells null for Prdx6 or with single-point mutations that inactivate either Prdx6-peroxidase or aiPLA2 activity alone exhibit decreased viability, increased lipid peroxidation, and incomplete repair when exposed to paraquat, hyperoxia, or organic peroxides. Ferroptosis is form of cell death driven by the accumulation of phospholipid hydroperoxides. We studied the role of Prdx6 as a ferroptosis suppressor in the lung. We first compared the expression Prdx6 and glutathione peroxidase 4 (GPx4) and visualized Prdx6 and GPx4 within the lung. Lung Prdx6 mRNA levels were five times higher than GPx4 levels. Both Prdx6 and GPx4 localized to epithelial and endothelial cells. Prdx6 knockout or knockdown sensitized lung endothelial cells to erastin-induced ferroptosis. Cells with genetic inactivation of either aiPLA2 or Prdx6-peroxidase were more sensitive to ferroptosis than WT cells, but less sensitive than KO cells. We then conducted RNA-seq analyses in Prdx6-depleted cells to further explore how the loss of Prdx6 sensitizes lung endothelial cells to ferroptosis. Prdx6 KD upregulated transcriptional signatures associated with selenoamino acid metabolism and mitochondrial function. Accordingly, Prdx6 deficiency blunted mitochondrial function and increased GPx4 abundance whereas GPx4 KD had the opposite effect on Prdx6. Moreover, we detected Prdx6 and GPx4 interactions in intact cells, suggesting that both enzymes cooperate to suppress lipid peroxidation. Notably, Prdx6-depleted cells remained sensitive to erastin-induced ferroptosis despite the compensatory increase in GPx4. These results show that Prdx6 suppresses ferroptosis in lung endothelial cells and that both aiPLA2 and Prdx6-peroxidase contribute to this effect. These results also show that Prdx6 supports mitochondrial function and modulates several coordinated cytoprotective pathways in the pulmonary endothelium.

BACKGROUND: Elephant seals exhibit extreme hypoxemic tolerance derived from repetitive hypoxia/reoxygenation episodes they experience during diving bouts. Real-time assessment of the molecular changes underlying protection against hypoxic injury in seals remains restricted by their at-sea inaccessibility. Hence, we developed a proliferative arterial endothelial cell culture model from elephant seals and used RNA-seq, functional assays, and confocal microscopy to assess the molecular response to prolonged hypoxia. RESULTS: Seal and human endothelial cells exposed to 1% O2 for up to 6 h respond differently to acute and prolonged hypoxia. Seal cells decouple stabilization of the hypoxia-sensitive transcriptional regulator HIF-1α from angiogenic signaling. Rapid upregulation of genes involved in glutathione (GSH) metabolism supports the maintenance of GSH pools, and intracellular succinate increases in seal but not human cells. High maximal and spare respiratory capacity in seal cells after hypoxia exposure occurs in concert with increasing mitochondrial branch length and independent from major changes in extracellular acidification rate, suggesting that seal cells recover oxidative metabolism without significant glycolytic dependency after hypoxia exposure. CONCLUSIONS: We found that the glutathione antioxidant system is upregulated in seal endothelial cells during hypoxia, while this system remains static in comparable human cells. Furthermore, we found that in contrast to human cells, hypoxia exposure rapidly activates HIF-1 in seal cells, but this response is decoupled from the canonical angiogenesis pathway. These results highlight the unique mechanisms that confer extraordinary tolerance to limited oxygen availability in a champion diving mammal.

Introduction: The objective of the present study was to determine if emotional intelligence and family stress are predictors of satisfaction with studies in university students. Method: Cross-sectional predictive study with a quantitative approach. The population was made up of 414 university students of both sexes who applied the TMMS-24 Emotional Intelligence Scale by Salovey and Mayer Family Stress Scale, the original version of Olson, and the Brief Satisfaction with Studies EBSE scale. Results: When performing statistical processing, it was evident that family stress has a standardized beta coefficient [beta] = -0.116; p < 0.05 indicates a low, statistically significant, indirect prediction. In other words, the greater the family stress, the lower the satisfaction with studies. In the same way, the standardized beta coefficient that represents the prediction of emotional intelligence on satisfaction with studies, which was [beta] = 0.470; p < 0.05, showed a direct, moderate, and statistically significant predictive value. Discussion and Conclusions: The results establish that emotional intelligence and family stress predict satisfaction with studies. The areas that are part of emotional intelligence, such as perception, understanding, and regulation, are related to family stress levels, and these influence the degree of satisfaction with studies. The greater the emotional intelligence, the greater the satisfaction with studies and different life stages.

Antecedentes: El grupo Progetto Nazionale Emorragia Digestiva (PNED) de Italia publicó su propuesta de escala predictora de riesgo de mortalidad en pacientes con hemorragia digestiva alta no variceal (HTDANV), que desde su publicación no ha sido validada en Latinoamérica. Objetivo: Comparar el sistema PNED contra la escala de Rockall en la predicción de mortalidad de pacientes hospitalizados por HTDANV. Material y métodos: Estudio multicéntrico prospectivo, transversal, analítico, en el cual se reclutó a pacientes con diagnóstico de hemorragia de tubo digestivo alto no variceal del 2011 al 2015. Se contó con la colaboración de 6 centros hospitalarios mexicanos. Se calculó la puntuación por escala Rockall y por sistema PNED, clasificando a los pacientes en enfermedad leve, moderada o severa. Finalmente, se determinó la asociación entre mortalidad y riesgo mediante aplicación de la χ2 y con cálculo de riesgo relativo (RR), con una p significativa al presentar valor menor de 0.05; Resultados: Se recolectó información de 198 pacientes. Solo 8 pacientes (4%) fallecieron por causas directamente asociadas con la hemorragia. Al aplicar escala Rockall, 46 pacientes alcanzaron puntuación de enfermedad severa (23.2%), de los cuales 5 fallecieron, con un RR de 5.5 (IC 1.35-22.02, p=0.006). En cuanto a PNED, solo 8 pacientes alcanzaron puntuación de enfermedad severa (4%), de los cuales 5 fallecieron, con un RR de 38.7 (IC 11.4-137.3, p=0.001). Conclusiones: El sistema PNED fue más selectivo para calificar un caso grave, pero tuvo mayor capacidad de predicción de mortalidad comparado con la escala Rockall.

Background: The predictive scale for mortality risk in patients with nonvariceal upper gastrointestinal bleeding (NVUGIB) proposed by Italy's PNED (Progetto Nazionale Emorragia Digestiva) group has not been validated in Latin America since its original publication. Aim: To compare the PNED system and the Rockall score as mortality predictors in patients hospitalized for NVUGIB. Material and methods: A multicenter, prospective, cross-sectional, analytic study was conducted that recruited patients diagnosed with nonvariceal upper gastrointestinal bleeding within the time frame of 2011 to 2015. Slaughter is a crucial step in the meat production chain that could induce psychological stress on each animal, resulting in a physiological response that can differ among individuals. The aim of this study was to investigate the relationship between an animal’s emotional state, the subsequent psychological stress at slaughter and the cellular damage as an effect. In all, 36 entire male pigs were reared at an experimental farm and a cognitive bias test was used to classify them into positive bias (PB) or negative bias (NB) groups depending on their decision-making capabilities. Half of the animals, slaughtered in the same batch, were used for a complete study of biomarkers of stress, including brain neurotransmitters and some muscle biomarkers of oxidative stress. After slaughter, specific brain areas were excised and the levels of catecholamines (noradrenaline (NA) and dopamine (DA)) and indoleamines (5-hydroxyindoleacetic acid and serotonin (5HT)) were analyzed. In addition, muscle proteasome activity (20S), antioxidant defence (total antioxidant activity (TAA)), oxidative damage (lipid peroxidation (LPO)) and autophagy biomarkers (Beclin-1, microtubule-associated protein I light chain 3 (LC3-I) and LC3-II) were monitored during early postmortem maturation (0 to 24 h). Black carbon (BC) is a major component of submicron particulate matter (PM), with significant health and climate impacts. Many cities in emerging countries lack comprehensive knowledge about BC emissions and exposure levels. This study investigates BC concentration levels, identifies its emission sources, and characterizes the optical properties of BC at urban background sites of the two largest high-altitude Bolivian cities: La Paz (LP) (3600 m above sea level) and El Alto (EA) (4050 m a.s.l.), where atmospheric oxygen levels and intense radiation may affect BC production. The study relies on concurrent measurements of equivalent black carbon (eBC), elemental carbon (EC), and refractory black carbon (rBC) and their comparison with analogous data collected at the nearby Chacaltaya Global Atmosphere Watch Station (5240 m a.s.l). The performance of two independent source apportionment techniques was compared: a bilinear model and a least-squares multilinear regression (MLR). Maximum eBC concentrations were observed during the local dry season (LP: eBC = 1.5 ± 1.6 µg m−3; EA: 1.9±2.0 µg m−3). While eBC concentrations are lower at the mountain station, daily transport from urban areas is evident. Average mass absorption cross sections of 6.6–8.2 m2 g−1 were found in the urban area at 637 nm. Both source apportionment methods exhibited a reasonable level of agreement in the contribution of biomass burning (BB) to absorption. The MLR method allowed the estimation of the contribution and the source-specific optical properties for multiple sources, including open waste burning.

La Economía Social y Solidaria se refiere a un grupo de organizaciones que a través de su trabajo impulsan el desarrollo socioeconómico, cultural y ambiental en una región, este modelo evoca a las cooperativas, comunidades ejidales, cajas de ahorro y fundaciones. En México, el INAES regula los NODESS, definidas como alianzas entre el sector social de la economía, académico y gubernamental, que unen sus capacidades para incidir positivamente en una región. El objetivo fue analizar la importancia de la Economía Social y Solidaria y la pertinencia de las alianzas NODESS en México, particularmente en Sinaloa, como estrategia de desarrollo regional. La investigación fue cualitativa, con la técnica del estudio por categorías apoyada en el registro documental de bases de datos científicas digitales, con criterios de búsqueda el período de las publicaciones (2018 a 2023) y las palabras clave. La información se clasificó en conceptos, prácticas y ejes, se sintetizó y se dedujeron patrones. Para profundizar el análisis, se integró una experiencia de NODESS de Sinaloa. Los resultados resaltan la importancia de las prácticas de la Economía Social y Solidaria en las comunidades y la aceptación de los NODESS en México, los ejes coincidentes fueron fortalecimiento de la gestión productiva, emprendimiento asociativo y profesionalización del personal. Se concluye que, las alianzas NODESS son una oportunidad para impulsar el sector primario en Sinaloa. Para futuras investigaciones se propone elaborar un compendio de buenas prácticas de los NODESS en Sinaloa, con la intención de divulgarlo e impulsar la creación de nuevas alianzas.

Abstract This study aims to evaluate the applicability of the high-resolution WaveFront Phase Imaging Sensor (WFPI) in eyes with Fuchs’ Endothelial Corneal Dystrophy (FECD) through qualitative and quantitative analysis using a custom-designed Automatic Guttae Detection Method (AGDM). The ocular phase was measured using the t $$\cdot$$ · eyede aberrometer and then was processed to obtain its High-Pass Filter Map (HPFM). The subjects were pathological and healthy patients from the Fundación Jiménez-Díaz Hospital (Madrid, Spain). The AGDM was developed and applied in pupils with 3 and 5 mm of diameter. A set of metrics were extracted and evaluated like the Root-Mean-Square error (RMS), Number of guttae, Guttae Area, and Area of Delaunay Triangulation (DT). Finally, a Support Vector Machine (SVM) model was trained to classify between pathological and healthy eyes. Quantitatively, the HPFM reveals a dark spots pattern according to the ophthalmologist’s description of the slit-lamp examination of guttae distribution. There were significant statistical differences in all the metrics when FECD and Healthy groups were compared using the same pupil size; but comparing both pupil sizes for the same group there were significant differences in most of the variables. This sensor is a value tool to objectively diagnose and monitor this pathology through wavefront phase changes.

Abstract: An investigation was performed to determine whether high concentrations of carbon dioxide (CO2) at 70-98% in atmospheric air are a suitable alternative for stunning rabbits compared to conventional approaches such as electronarcosis. Aversion to the gas and efficacy in causing prolonged unconsciousness and death were studied in a total of 480 rabbits by means of behavioural parameters, physiological indicators (presence of rhythmic breathing and corneal reflex) and electroencephalography (EEG, brain function). Objective: Given funding concerns and heightened work expectations at baccalaureate degree-granting community colleges, we set out to understand faculty advisors' emotional labor in such context. Methods: We conducted a supra analysis, which is a form of secondary analysis of qualitative data. Existing interview data were drawn from a mixed-methods study focused on changes in academic advising policies and practices at baccalaureate degree-granting community colleges. Results: We identified three interrelated themes: (a) putting students first; (b) overextending oneself selflessly; and (c) pressures of neoliberalism and bureaucratic checkpoints. Conclusion/Contributions: This work addresses an important gap in the literature as it pertains to the complexities of the work and professional lives of community college faculty at CCB-conferring colleges. In addition to highlighting the emotional labor of CCB faculty advisors, this work further illuminates how community colleges have "normalized emotional labor as part of the community college faculty role".

The objective of this study was to determine whether rabbits fed in a restricted regimen (75%) showed increased competition for feeding, drinking and use of specific areas of the cages as compared with those provided feed ad libitum. This evaluation was carried out by measuring their space utilisation in the cage, the incidence of agonistic behaviour and rates of mortality. In total, 504 rabbits between 31 and 66 days of age were used in this study. A total of 200 heavy-weight rabbits and 56 light-weight rabbits were randomly housed in 32 cages, each cage containing eight rabbits: 25 cages housing heavy rabbits and seven cages housing the light-weight ones. They were all fed ad libitum (AD). In addition, a total of 208 heavy-weight rabbits and 40 light-weight rabbits were randomly housed in 31 cages, each of them containing eight rabbits: 26 cages housing heavy weight rabbits and five cages housing light-weight ones. They were all fed a restricted diet (R) regimen. The restriction was calculated to be 75% of the feed consumed by the AD group. The total space available in the cage was 3252 cm2, with a stocking density of 24.6 animals/m2. Animals between 32 and 60 days of age from 20 different cages were observed nine times per week (morning or afternoon) by means of scan and focal sampling by one observer. During each period, cages were assessed for 5 min, registering every minute the position of all the animals in relation to Area A (feeder), Area B (central part) or Area C (back and drinker area). The incidence of agonistic behaviour such as displacement, biting and jumping on each other was also assessed. Performance variables such as daily gain and feed conversion ratio, in addition to general health status and mortality rates, were recorded for all rabbits. When the rabbits were under restricted feeding, the competition for feed and drink increased with clear signs of agonistic behaviour such as biting, displacement and animals jumping on top of each other. Although this competition was maintained during the entire growing period, the BW homogeneity between animals in the same cage was similar in both cases, suggesting that all animals could consume similar quantities of feed. The possible advantages of a restricted diet, such as better feed conversion ratio, were observed in this study only in the last few weeks of the growing period.

The objective of this work was to study the postmortem evolution of potential biomarkers of autophagy (Beclin 1, LC3-II/LC3-I ratio) and oxidative stress (total antioxidant activity, TAA; superoxide dismutase activity, SOD and catalase activity, CAT) in the Longissimus dorsi muscle of entire male ((Large White×Landrace)×Duroc) pigs subjected to different management treatments that may promote stress, such as mixing unfamiliar animals at the farm and/or during transport and lairage before slaughter. During the rearing period at the farm, five animals were never mixed after the initial formation of the experimental groups (unmixed group at the farm, UF), whereas 10 animals were subjected to a common routine of being mixed with unfamiliar animals (mixed group at the farm, MF). Furthermore, two different treatments were used during the transport and lairage before slaughter: 10 pigs were not mixed (unmixed group during transport and lairage, UTL), whereas five pigs were mixed with unfamiliar animals on the lorry and during lairage (mixed group during transport and lairage, MTL). These mixing treatments were then combined into three pre-slaughter treatments – namely, UF-UTL, MF-UTL and MF-MTL. Anxiety is a critical component of the development and maintenance of drug addiction; however, anti-anxiety medications such as benzodiazepines and beta-blockers (β-adrenergic receptor antagonists) are not used for the treatment of substance use disorder, except for the management of acute withdrawal syndrome. Preclinical studies have shown that beta-blockers may reduce stress-induced relapse; however, the effect of beta blockers on the escalation and maintenance of drug intake has not been tested. To address this issue, we chronically administered the β-adrenergic receptor antagonist propranolol during the escalation or maintenance of cocaine intake in a model of extended access (6 h) to cocaine self-administration (0.5 mg/kg). The behavioural specificity of propranolol was tested using a non-drug reward (saccharin). Daily administration of propranolol (15 mg/kg) prevented the development of escalation of cocaine self-administration and partially reversed self-administration after the establishment of escalation of intake. Moreover, propranolol dose-dependently decreased the motivation for cocaine tested under a progressive ratio schedule of reinforcement during the development of escalation and after maintenance. Finally, propranolol administration had no effect on the escalation and maintenance of saccharin self-administration. These results demonstrate that chronic treatment with propranolol provides therapeutic efficacy in reducing cocaine self-administration during the development and after the establishment of escalation of cocaine self-administration in an animal model relevant to cocaine use disorder. These results suggest that beta blockers should be further investigated as a target for medication development for the treatment of cocaine use disorder.
