Abstract The prostate gland (PG) is a small organ in the male reproductive system that is currently the focus of biomedical research due to its leading position in morbidity and mortality from the tissue-specific cancer prostate cancer (PC). The PG epithelium, which undergoes a cancerous transformation, is formed and functions under the control of androgens. At the beginning of the disease, epithelial cells produce an androgen receptor (AR) and are sensitive to androgen-deprivation therapy. However, such therapy inevitably leads to the transition of the disease to the castration-resistant prostate cancer (CRPC), which manifests itself in metastasis and rapid mortality. In CRPC, the cells of the prostate epithelium change their phenotype, that may be associated with AR mutation and loss the sensitivity to specific therapy. The mechanism of PG phenotypic transformation may be hidden in the interaction and formation of the stromal and epithelial cells, which are evident during the establishment of the primary cultures. The aim of this study was to investigate the generation of human PG stromal cells in primary stromal and organoid cultures. We found that, in contrast to the rapid appearance and formation of a homogeneous population of mesenchymal cells in primary stromal cultures of most tissues, human PG cell cultures are formed initially from epithelial cells. They appear in the second week of cultivation and produce cytokeratins (CKs). A homogeneous population of mesenchymal cells producing vimentin is formed only at the end of the fourth week of cultivation. It is accompanied by the disappearance of epithelial cells. At the same time, some epithelial cells simultaneously produce CKs and vimentin. In PG organoid cultures, there is often a concomitant growth of epithelial, but not mesenchymal, cells on culture plastic. During the cultivation of epithelial cells arising from the organoid cultures, they, like the cells of the primary epithelium, exhibit the ability to spontaneous transformation into mesenchymal cells and simultaneously produce CKs and vimentin. Our data suggest that in primary and organoid PG cultures, stromal cells can be formed from epithelium due to the epithelial-to-mesenchymal transition (EMT). The tendency of PG epithelium toward spontaneous EMT may contribute to the mechanism of high sensitivity of prostate tissue to malignant transformation and metastasis. Understanding this mechanism may contribute to the development of effective antitumor therapy of prostate cancer.

Ischemic lesions of the heart, including myocardial infarction, are the most common pathologies of human cardiovascular system. Despite all the research and achievements of medicine in this field, the mortality from this disease remains heavy. Therefore, studying of processes occurring in the myocardium in the early and late postinfarction periods remains important. Rat left ventricular cardiomyocyte (CMC) ploidy, hypertrophy, hyperplasia, and ultrastructure were investigated in 2, 6, and 26 weeks after experimental myocardial infarction, caused by permanent ligation of left coronary artery. Cytofluorimetric study of CMC ploidy revealed no difference between normal, sham-operated, and infarcted animals for all the tested stages. However, interference microscopy indicated significant changes in cells size. CMC dry mass of infarcted rats in 2 weeks after surgery was 1.5 times lower than in control and sham operated groups. Electron microscopy analysis of CMC revealed disruption of sarcomere structure. However, in 6 weeks after surgery CMC dry mass was 1.6 times higher than in control. In 26 weeks after myocardial infarction CMC dry mass exceeded control only in peri-infarction zone. Cell counting showed that the number of left ventricular CMC, reduced as a result of myocardial infarction, was not restored during myocardial remodeling. © 2019 International Society for Advancement of Cytometry.

The reactions of perfluoro-3-isothiocyanato-2-methyl-2-pentene with PPh3 and P(NEt2)3 in the presence of NaBF4, KI, and NaBPh4 form phosphonium salts with the heterocyclic substituent (4E)-5,5-bis(trifluoromethyl)-4-(tetrafluoroethylidene)-4,5-dihydro-1,3-thiazol-2-yl, instead of involving desulfurization and formation of P-F-containing products. The reaction with tris(pentafluorophenyl)phosphine fails. The reactions with P(OEt)3 in the presence of ClSiMe3 or (CH3O)2POSiMe3 yield diethyl or dimethyl [(4E)-5,5-bis(trifluoromethyl)-4-(tetrafluoroethylidene)-4,5-dihydro-1,3-thiazol-2-yl]phosphonates and no intramolecular alkylation products. The 1H, 13C, 19F, and 31P spectra are presented, and the reaction pathways are discussed. Potential mechanisms of the biological and catalytic activity of the reaction products are considered.

The article assesses the filtration of periodic signals of different shapes with the use of optimized calculation of discrete low pass filtering system (LPFS) orders. The calculation is based on the minimization of the difference between approximated values and filtered values of a noisy signal. To assess the feasibility of the calculation of the optimized discrete filtering system order, a series of experiments has been carried out on generation and filtration of noisy periodic signals with different signal shapes and various signal-to-noise ratios. For all of the noisy signals, optimal (relative to the original noise-free signals) and optimized (relative to the approximated signals) LPFS orders based on a minimal mean squared error (MSE) have been calculated. After calculation of MSE between filtered and original signals, it was observed that the optimal and optimized filters allow us to get comparable noisy signal filtration quality. The article is completed by the conclusions about the possibility of using the discrete filtering system order calculation optimized by approximation.

Reactions of 2-(imidazol-1-yl)-perfluoro-5,5-dimethyl-4-ethylidene-2-thiazoline with a series of secondary amines in THF resulted in the replacement of the imidazole moiety by the amine moiety.

The reaction of benzimidazoline-2-thione with perfluoro-2-methylpent-2-ene in the presence of triethylamine afforded (2E)-2-(tetrafluoroethylidene)-3,3-bis(trifluoromethyl)-2,3-dihydrothiazolo[3,2-a]benzimidazole whose structure was confirmed by X-ray diffraction analysis. The reaction pathways are discussed.

Perfluoro-2-methylpent-2-ene reacts with pyrazole, imidazole, 1,2,4-triazole, and benzotriazole to give products of the replacement of the vinylic fluorine atom. In the case of imidazole, the product of allylic fluorine substitution, 1,3-bisimidazolylperfluoro-2-methylpent-2-ene, is also formed. The structures of the products were confirmed by spectral data.

The reactions of perfluoro-2-methylpent-2-en-3-yl isothiocyanate with ambident N,O- and N,S-nucleophiles (thiazole-2-thione, pyridine-2-thione, 2-hydroxypyridine, benzothiazole-2-thione, benzoxazole-2-thione, 3,4,5,6-tetrahydropyrimidine-2-thione) in the presence of triethylamine yield only 2-N-substituted 4,5-dihydrothiazole derivatives. The molecular structures of three products were determined by X-ray diffraction analysis. The reaction pathways are discussed.
