The inventory of the taxa collected in 2021 during the annual field trip of the Working Group for Floristics, Systematics, and Evolution of the Italian Botanical Society is reported. The field trip was held in the Mount Pennino massif, between Umbria and Marche administrative regions (central Italy). The flora documented for the study area amounts to 413 specific and subspecific taxa (including one hybrid), belonging to 234 genera and 64 families. Thirty-seven taxa are endemic to Italy; 42 are included in the IUCN Red List of the Italian Flora. Only three alien taxa were found. Ten taxa and one hybrid are considered floristic novelties because either new or confirmed for the regional flora of Umbria and/or Marche. Particularly, 6 taxa are new and 4 are confirmed for Umbria; one taxon and one hybrid are new for Marche.

Abstract. Marine diazotrophs convert dinitrogen (N2) gas intobioavailable nitrogen (N), supporting life in the global ocean. In 2012, thefirst version of the global oceanic diazotroph database (version 1) waspublished. Here, we present an updated version of the database (version 2),significantly increasing the number of in situ diazotrophic measurements from13 565 to 55 286. Data points for N2 fixation rates, diazotrophic cellabundance, and nifH gene copy abundance have increased by 184 %, 86 %, and809 %, respectively. Version 2 includes two new data sheets for the nifH genecopy abundance of non-cyanobacterial diazotrophs and cell-specific N2fixation rates. The measurements of N2 fixation rates approximatelyfollow a log-normal distribution in both version 1 and version 2. However,version 2 considerably extends both the left and right tails of thedistribution. Consequently, when estimating global oceanic N2 fixationrates using the geometric means of different ocean basins, version 1 andversion 2 yield similar rates (43–57 versus 45–63 Tg N yr−1; rangesbased on one geometric standard error). In contrast, when using arithmeticmeans, version 2 suggests a significantly higher rate of 223±30 Tg N yr−1 (mean ± standard error; same hereafter) compared to version 1(74±7 Tg N yr−1). Specifically, substantial rate increases areestimated for the South Pacific Ocean (88±23 versus 20±2 Tg N yr−1), primarily driven by measurements in the southwestern subtropics,and for the North Atlantic Ocean (40±9 versus 10±2 Tg N yr−1). Moreover, version 2 estimates the N2 fixation rate in theIndian Ocean to be 35±14 Tg N yr−1, which could not be estimatedusing version 1 due to limited data availability. Furthermore, a comparisonof N2 fixation rates obtained through different measurement methods atthe same months, locations, and depths reveals that the conventional15N2 bubble method yields lower rates in 69 % cases compared tothe new 15N2 dissolution method. This updated version of thedatabase can facilitate future studies in marine ecology andbiogeochemistry. Influenza remains a serious global health concern, causing significant morbidity and mortality each year. Vaccination is crucial to mitigate its impact, but requires rapid and efficient manufacturing strategies to handle timing and supply. Traditionally relying on egg-based production, the field has witnessed a paradigm shift toward cell culture-based methods offering enhanced flexibility, scalability, and process safety. This review provides a concise overview of available cell substrates and technological advancements. We summarize crucial steps toward process intensification – from roller bottle production to dynamic cultures on carriers and from suspension cultures in batch mode to high cell density perfusion using various cell retention devices. Moreover, we compare single-use and conventional systems and address challenges including defective interfering particles. Taken together, we describe the current state-of-the-art in cell culture-based influenza virus production to sustainably meet vaccine demands, guarantee a timely supply, and keep up with the challenges of seasonal epidemics and global pandemics.

After several years of public road testing, the commercial deployment of fully autonomous vehicles—or Automated Driving Systems (ADS)—is poised to scale substantially following significant technological advancements and recent regulatory approvals. However, the fundamental question of whether an ADS is safer than its human counterparts remain largely unsolved due to several challenges in establishing an appropriate real-world safety comparison method. As scaling ensues, the lack of an established method can contribute to misinterpretations or uncertainties regarding ADS safety and impede the continuous and consistent assessment of ADS performance. This study introduces three research developments to define a robust and replicable safety comparison method to address this critical methodological gap. First, we introduce the use of liability insurance claims data to measure the comparative safety between ADS and human drivers. Second, we use Swiss Re insurance claims data to establish the first zip code- and responsibility-calibrated human performance benchmark, composed of over 600,000 private passenger vehicle claims and 125 billion miles of driving exposure. Third, we perform a case study by applying the developed baseline to evaluate the safety impact of the Waymo Driver. We find that when benchmarked against zip code-calibrated human baselines, the Waymo Driver significantly improves safety towards other road users. The comparison method established in this study can be replicated for other regions or ADS deployments to aid the decision-making of ADS safety stakeholders such as regulators, and instill trust in the general public.

The inventory of the taxa collected in 2022 during the annual field trip of the Working Group for Floristics, Systematics and Evolution of the Italian Botanical Society is reported. The field trip was held from 19th to 22th April in the islands of Lipari and Panarea (Aeolian Islands, Sicilia). Overall, 1,664 herbarium specimens were deposited in public and private herbaria. The flora documented for the studied area amounts to 386 specific and subspecific taxa, belonging to 241 genera and 74 families. Centaurea aeolica, Helichrysum litoreum (Asteraceae), and Dianthus rupicola subsp. aeolicus (Caryophyllaceae) were the only three Italian endemics found in the study area, whereas 48 alien taxa were recorded. Dimorphotheca ecklonis (Asteraceae), Nassella tenuissima (Poaceae), Solanum torvum (Solanaceae), and Viola wittrockiana (Violaceae) are casual alien species new to Sicilia, whereas Oenothera odorata (Onagraceae) is a new naturalized alien species for the Italian vascular flora.

Abstract Innovations in viral vaccine manufacturing are crucial for pandemic preparedness and to meet ever‐rising global demands. For influenza, however, production still mainly relies on technologies established decades ago. Although modern production shifts from egg‐based towards cell culture technologies, the full potential has not yet been fully exploited. Here, we evaluate whether implementation of state‐of‐the‐art technologies for cell culture‐based recombinant protein production are capable to challenge outdated approaches in viral vaccine process development. For this, a fully automated single‐cell cloning strategy was established to generate monoclonal suspension Madin‐Darby canine kidney (MDCK) cells. Among selected cell clones, we could observe distinct metabolic and growth characteristics, with C59 reaching a maximum viable cell concentration of 17.3 × 106 cells/mL and low doubling times in batch mode. Screening for virus production using a panel of human vaccine‐relevant influenza A and B viruses in an ambr15 system revealed high titers with yields competing or even outperforming available MDCK cell lines. With C113, we achieved cell‐specific virus yields of up to 25,000 virions/cell, making this cell clone highly attractive for vaccine production. Finally, we confirmed process performance at a 50‐fold higher working volume. In summary, we present a scalable and powerful approach for accelerated development of high‐yield influenza virus production in chemically defined medium starting from a single cell.

Abstract Microbial secondary metabolites play important roles in biotic interactions in microbial communities and yet, we do not understand how these compounds impact the assembly and development of microbial communities. To address the implications of microbial secondary metabolite production on biotic interactions in the assembly of natural seawater microbiomes, we constructed a model system where the assembly of a natural seawater biofilm community was influenced by the addition of the marine biofilm forming Phaeobacter inhibens that can produce the antibiotic secondary metabolite tropodithietic acid (TDA), or a mutant incapable of TDA production. Because of the broad antibiotic activity of TDA, we hypothesized that the potential of P. inhibens to produce TDA would strongly affect both biofilm and planktonic community assembly patterns. We show that 1.9 % of the microbial composition variance across both environments could be attributed to the presence of WT P. inhibens, and especially genera of the Bacteriodetes were increased by the presence of the TDA producer. Moreover, network analysis with inferred putative microbial interactions revealed that P. inhibens mainly displayed strong positive associations with genera of the Flavobacteriaceae and Alteromonadaceae, and that P. inhibens acts as a keystone OTU in the biofilm exclusively due to its potential to produce TDA. Our results demonstrate the potential impact of microbial secondary metabolites on microbial interactions and assembly dynamics of complex microbial communities.

Abstract Background The role of the lung for the initiation and progression of rheumatoid arthritis (RA) is still unclear. Up to 10% of RA patients develop interstitial lung disease which remains a clinical challenge. Understanding early disease mechanisms is of great importance. The objective of this study was to determine whether there is an association between peripheral neutrophil phenotypes and presence of pulmonary abnormalities (PA) on chest high-resolution computed tomography (HRCT) in untreated early RA (ueRA). Methods Clinical data and blood were collected, and HRCT performed at diagnosis on 30 consecutive anti-citrullinated protein antibody (ACPA) and/or rheumatoid factor (RF) positive ueRA patients. HRCTs were evaluated for the presence of RA-associated parenchymal, airway and/or pleural abnormalities. Expression of phenotype markers on neutrophils were determined by flow cytometry. Levels of calprotectin, ACPA and RF were measured using immunoassays. Results The frequency of having any PA was 60%. Airway abnormalities were present in 50%, parenchymal nodules in 43% and interstitial lung abnormalities (ILA) in 10%. Unsupervised multivariate data analysis showed clustering of any PA with neutrophil activation, parameters of inflammation and RF titres. In univariate analysis, the patients with PA displayed significantly increased CD11b and decreased CD62L expression on neutrophils (1.2-fold, p = 0.014; 0.8-fold, p = 0.012) indicating activation and significantly increased RF IgM titre and CRP (5.7-fold, p = 0.0025; 2.3-fold, p = 0.0035) as compared to no PA. Titres of RF, but not ACPA, correlated with expression of the neutrophil activation marker CD11b. A stratified analysis demonstrated that airway involvement was the PA subtype with the strongest association with neutrophil activation. Conclusion We report a strong association between radiographic airway findings and activation of circulating neutrophils in early RA supporting a role of innate immunity and the lung at disease onset. Our results also indicate different contributions of RF and ACPA in the RA pathogenesis.

Superconducting (SC) tokamak JT-60SA plays an essential role in fusion research and development by supporting and complementing the ITER project, providing directions to the DEMO design activity and fostering next generation scientists and engineers. Since the short circuit incident at the terminal joints of equilibrium field coil #1 during the integrated commissioning (IC) in March 2021, both EU and JA implementing agencies (IAs) have examined how to ensure safe operation of JT-60SA by mitigating the risk of possible discharge occurrence inside the cryostat. Based on the experience of the global Paschen tests, the IAs have established a strategy of risk mitigation measures, which is a combination of (i) reinforcement of insulation, (ii) avoiding unnecessary voltage application to the coil systems and (iii) immediate de-energization of the coils when deteriorated vacuum conditions are detected. Thanks to the considerable efforts of the Integrated Project Team members, the IC restarted in May 2023. After confirmation of the SC state of the coil systems (TF, EF and CS), the coil energization test and the plasma operation phase 1 (OP-1) started. The first plasma was successfully achieved on 23 October 2023 with a limited value of voltage and current applied to the coils. The plasma configuration control was also confirmed with low plasma current and low auxiliary heating power conditions. Based on the IO–F4E–QST collaboration, activities of JT-60SA have been shared with the IO and provided an important lesson for ITER assembly and commissioning, and will provide an outstanding contribution to fusion research at large. After OP-1, maintenance & enhancement phase 1 (M/E-1) starts from January 2024, in which in-vessel components are installed, and heating and diagnostic systems are extensively upgraded to allow a high power heating experiment planned in OP-2. In order to make the best use of JT-60SA, a newly organized JT-60SA experiment team will refine the research plan for the future high heating power operation phase.

ABSTRACT In the search for novel drug candidates, diverse environmental microbiomes have been surveyed for their secondary metabolite biosynthesis potential, yet little is known about the biosynthetic diversity encoded by divergent microbiomes from different ecosystems, and the environmental parameters driving this diversity. Here, we used targeted amplicon sequencing of adenylation (AD) and ketosynthase (KS) domains along with 16S sequencing to delineate the unique biosynthetic potential of microbiomes from three separate habitats (soil, water, and sediments) exhibiting unique small spatial scale physicochemical gradients. The estimated richness of AD domains was highest in marine sediments with 656 ± 58 operational biosynthetic units (OBUs), while the KS domain richness was highest in soil microbiomes with 388 ± 67 OBUs. Microbiomes with rich and diverse bacterial communities displayed the highest PK potential across all ecosystems, and on a small spatial scale, pH and salinity were significantly, positively correlated to KS domain richness in soil and aquatic systems, respectively. Integrating our findings, we were able to predict the KS domain richness with a RMSE of 31 OBUs and a R2 of 0.91, and by the use of publicly available information on bacterial richness and diversity, we identified grassland biomes as being particularly promising sites for the discovery of novel polyketides. Furthermore, a focus on acidobacterial taxa is likely to be fruitful, as these were responsible for most of the variation in biosynthetic diversity. Overall, our results highlight the importance of sampling diverse environments with high taxonomic diversity in the pursuit for novel secondary metabolites. IMPORTANCE To counteract the antibiotic resistance crisis, novel anti-infective agents need to be discovered and brought to market. Microbial secondary metabolites have been important sources of inspiration for small-molecule therapeutics. However, the isolation of novel antibiotics is difficult, and the risk of rediscovery is high. With the overarching purpose of identifying promising microbiomes for discovery of novel bioactivity, we mapped out the most significant drivers of biosynthetic diversity across divergent microbiomes. We found the biosynthetic potential to be unique to individual ecosystems, and to depend on bacterial taxonomic diversity. Within systems, and on small spatial scales, pH and salinity correlated positively to the biosynthetic richness of the microbiomes, Acidobacteria representing the taxa most highly associated with biosynthetic diversity. Ultimately, understanding the key drivers of the biosynthesis potential of environmental microbiomes will allow us to focus bioprospecting efforts and facilitate the discovery of novel therapeutics.

Rationale: We recently demonstrated that patients with coronary artery disease (CAD) and obstructive sleep apnea (OSA) carrying the tumor necrosis factor-alpha (TNF-α) A allele had increased circulating TNF-α levels compared with the ones carrying the TNF-α G allele. In the current study, we addressed the effect of TNF-α (-308G/A) gene polymorphism on circulating TNF-α levels following continuous positive airway pressure (CPAP) therapy. Methods: This study was a secondary analysis of the RICCADSA trial (NCT00519597) conducted in Sweden. CAD patients with OSA (apnea–hypopnea index) of ≥15 events/h and an Epworth Sleepiness Scale (ESS) score of TNF-α was measured by immunoassay (Luminex, R&D Systems). Genotyping of TNF-α-308G/A (single nucleotide polymorphism Rs1800629) was performed by polymerase chain reaction–restriction fragment length polymorphism. Results: In all, 239 participants (206 men and 33 women; mean age 64.9 (SD 7.7) years) with polymorphism data and circulating levels of TNF-α at baseline and 1-year follow-up visits were included. The median circulating TNF-α values fell in both groups between baseline and 12 months with no significant within- or between-group differences. In a multivariate linear regression model, a significant change in circulating TNF-α levels from baseline across the genotypes from GA to GA and GA to AA (standardized β-coefficient −0.129, 95% confidence interval (CI) −1.82; −0.12; p = 0.025) was observed in the entire cohort. The association was more pronounced among the individuals who were using the device for at least 4 h/night (n = 86; standardized β-coefficient −2.979 (95% CI −6.11; −1.21); p = 0.004)), whereas no significant association was found among the patients who were non-adherent or randomized to no-CPAP. The participants carrying the TNF-α A allele were less responsive to CPAP treatment regarding the decline in circulating TNF-α despite CPAP adherence (standardized β-coefficient −0.212, (95% CI −5.66; −1.01); p = 0.005). Conclusions: Our results suggest that TNF-α (-308G/A) gene polymorphism is associated with changes in circulating TNF-α levels in response to CPAP treatment in adults with CAD and OSA.

As we stand on the brink of the post-antibiotic era, we are in dire need of novel antimicrobial compounds. Microorganisms produce a wealth of so-called secondary metabolites and have been our most prolific source of antibiotics so far. However, rediscovery of known antibiotics from well-studied cultured microorganisms, and the fact that the majority of microorganisms in the environment are out of reach by means of conventional cultivation techniques, have led to the exploration of the biosynthetic potential in natural microbial communities by novel approaches. In this mini review we discuss how sequence-based analyses have exposed an unprecedented wealth of potential for secondary metabolite production in soil, marine, and host-associated microbiomes, with a focus on the biosynthesis of non-ribosomal peptides and polyketides. Furthermore, we discuss how the complexity of natural microbiomes and the lack of standardized methodology has complicated comparisons across biomes. Yet, as even the most commonly sampled microbiomes hold promise of providing novel classes of natural products, we lastly discuss the development of approaches applied in the translation of the immense biosynthetic diversity of natural microbiomes to the procurement of novel antibiotics.

This paper aims to contribute to the design of a new conceptual model for coping with obtrusive interpersonal tensions in the context of a systemic organizational development process. The model integrates notions of the Value and Development Square (VDS) into a structured process, with ideas from the hypnosystemic work and solution-focused questions. This results in an approach that can be taught to and applied by those seeking solutions for interpersonal tensions within an organization. For the model's educational applicability, we developed a conceptual bridge: we split the attentiveness process—which occurs during the search for solutions for an unpleasant tension—into interrelated dimensions. In doing so, the process using the VDS can be observed in "small steps," and, if necessary, suitable interventions can be developed. This paper provides ideas for using this augmented version of the square, including specific questions for those involved in adverse interpersonal tensions that induce reflection and/or inspire a perspective change. With this adaption of the original VDS, we aim to offer a valuable tool for leaders, coaches, mediators, and employees. It aids in rapidly structuring complex situations where stressful tensions persist and assists those engaged in interpersonal tensions to transform those tensions into useful levers for collective goals.

Herbaria are an important source of data and material useful in many fields, including plant conservation. Seeds preserved in herbarium specimens may have the potential to germinate, although few studies focused on this topic. Here, the first systematic assessment of six techniques, including priming techniques and melatonin application, aimed at improving the germination of seeds from herbarium specimens is presented. Seed germination of 26 species common in Europe, some of which congeneric to extinct species, collected in herbaria and in the wild (20,549 seeds in total, including 19,509 from 297 herbarium specimens from 8 different herbaria) was tested with the following treatments: exogenous melatonin addition to the germination medium, priming with melatonin, osmopriming, hydropriming for 24 and 48 hours, standard soil, heat sterilization and gibberellins addition. More than 85% of the fresh seeds and 1% of the seeds collected in herbaria germinated, including seeds older than 50 years. Data show that treatment with exogenous melatonin had a positive effect on the germination of fresh seeds, but a negative effect on the germination of herbarium‐derived seeds. Furthermore, osmopriming treatment had a slightly positive effect on the germination of herbarium‐derived seeds. Osmopriming and exogenous melatonin addition seem to be promising techniques that need further investigation and improvement and might be useful for the development of an optimal germination protocol for old and herbarium‐derived seeds. The germination of seeds from herbaria could be an important tool in plant conservation, with the aim of reversing the extinction trend of many species through de‐extinction, safeguarding biodiversity, and genetic variability. This study provides preliminary data for the development of germination protocols, especially for old seeds of species of conservation interest, to maximise the chance of recovering lost genetic diversity and leading to the first de‐extinction ever. The field trip was held in 2019 along the Sillaro, Santerno, and Senio high valleys located in Toscana (central Italy). The flora documented for the studied area amounts to 492 specific and subspecific taxa (including five hybrids), belonging to 254 genera and 77 families. Bromopsis caprina, Ophrys appennina, O. classica, Polygala flavescens subsp. flavescens, and Pulmonaria vallarsae subsp. apennina were the only five Italian endemics found in the study area, whereas 28 alien taxa were detected. Finally, nine taxa (seven native and two alien) have to be considered as new records for the regional flora of Toscana.

If you have ever tried to follow a discussion on a controversial topic on any social media platform such as Facebook or Twitter, you may have noticed that even the smallest deviation from the majority opinion can lead to the exclusion of the person from the ongoing discussion. Terms like cancel culture, online bashing, Twitter storm, etc., also describe this kind of disassociating communication. However, every ostracism decreases the size of the remaining in-group, to the point where society could end up fragmented into multitudes of small social systems. On one hand, a democratic society in which a dialog is only possible in smaller units tends to be far more complex and thus far less capable of acting than a society that favors a broader discourse. On the other hand, social interaction that allows and incorporates many different opinions, views, propositions, and conclusions seems to require a large effort. For an open-minded discourse to succeed, our communication shall transcend both the content dialog (first-order) and the meta-dialog (second-order) so as to set the dialog in relation to its context. In this paper we spotlight the differences between associating and disassociating communication. We also use the viewpoint of social systems theory to explore not only answers to questions about the consequences of avoiding responsibility for the quality of our dialogs, but also the solutions a distinction-based approach offers to communication challenges.

In our daily practice as management consultants we observe disorientation, misconceptions, and open questions about the suitability, limitations, and/or benefits of novel management approaches. Certainly, there is a strong demand for up-to-date management practices, though at the same time there exist the dangers of misuse and misleading expectations, not necessarily from malice but rather, according to our experience, from lack of self-observation. In this context, second-order concepts are revealed to be useful and solution-oriented. Even though in literature we can find approaches to distinguish first-order cybernetics (FOC) from second-order cybernetics (SOC), none of those focus on organizations as social living systems or the organization's basic operation: decision making. Consequently, in this paper we discuss the essential ideas of SOC-based management methods and tools, focusing on the dissimilarities of posture and potential performance of these concepts. To contrast them, we compare Design Thinking with Comparative Systemic (CS) Management, two concepts that use SOC ideas, with two well-known FOC management approaches: the Plan-Do-Check-Act-Cycle (PDCA Cycle) and Systems Dynamics. Finally, we present the fundamental differences between FOC and SOC based decision making in management. Basically, we differentiate between concepts based on FOC or SOC by means of three modes of action: how they propose to coordinate (temporal dimension), structure (factual dimension), or legitimate (social dimension) decisions.

Background: Oocyte in vitro maturation (IVM) is a reproductive technology that generates mature oocytes from patients who have received minimal or no ovarian stimulation (OS). The minimal-stimulation feature of IVM makes it a valuable option for cancer patients requiring fertility preservation (FP), before chemo- or radiotherapy. When ovarian tissue is collected for cryopreservation (OTC), only the ovarian cortex is preserved whereas the ovarian medulla containing growing follicles is discarded. To maximise the future fertility prospects for cancer patients, OTC should be coupled with a modified form of IVM, termed ex-vivo IVM, in which immature cumulus-oocyte complexes (COCs) are collected from antral follicles. Aim: To examine the utility of emergency ex-vivo IVM for FP. Method: A 31-year-old woman diagnosed with locally invasive appendiceal cancer required urgent FP as there was no time for conventional OS egg freeze before surgery, and OTC was contra-indicated due to the possibility of later regrafting of cancer cells. She underwent peritonectomy with bilateral oophorectomy with no prior ovarian stimulation or cycle management. Visible antral follicles were aspirated ex-vivo, the ovaries were then manually dissected, antral follicles punctured and intact COCs collected. COCs were washed and matured for 30-46h in G-2 PLUS medium supplemented with 0.1IU/mL rec-FSH, 0.5IU/mL rec-hCG, and 10% (v/v) autologous heat-inactivated patient serum. Results: 40 COCs were collected and 18 (45%) reached metaphase II, 1 was cryopreserved and 17 were inseminated by ICSI. 13/17 oocytes fertilised (2PN; 76%) and 7/13 (54%) blastocysts were generated (D5: 1× HB-1-1, 3× B-1-1, 1× B-2-1. D6: 1× HB-1-2, 1× B-2-2). Seven blastocysts and one oocyte were vitrified for future use by the patient. Conclusion: Ex-vivo IVM is a viable FP option for patients diagnosed with cancer. It can readily be coupled with OTC and/or a regular IVM oocyte pickup to maximise a cancer patient’s future fertility prospects.
