Abstract In this fourth session of our motivational interviewing workshop, we will go from theory to practice through simple and direct examples from clinical practice. The participants will be able to watch previously filmed clinical cases and participate in role plays, where they can practice using different motivational interviewing techniques that they had learned before. RESUMO O Brasil tem usado dívida externa pelo menos desde sua independência - e problemas em administrá-la vêm acontecendo desde então. Este artigo discute como parece ter sido de 1967-8 que a dinâmica mudou com a abertura financeira do país e como o país se tornou o país mais endividado do mundo em desenvolvimento.

Candida albicans is a human commensal fungus and the etiologic agent of nosocomial infections in immunocompromised individuals. Candida spp. is the most studied human fungal pathogen, and the mechanisms by which this fungus can evade the immune system affecting immunosuppressed individuals have been extensively studied. Most of these studies focus on different species of Candida, and there is much to be understood in virulence variability among lineages, specifically different C. albicans clinical isolates. To better understand the main mechanisms of its virulence variability modulated in C. albicans clinical isolates, we characterized L3881 lineage, which has been previously classified as hypovirulent, and SC5314 lineage, a virulent wild-type control, by using both in vitro and in vivo assays. Our findings demonstrated that L3881 presented higher capacity to avoid macrophage phagocytosis and higher resistance to oxidative stress than the wild type. These characteristics prevented higher mortality rates for L3881 in the animal model of candidiasis. Conversely, L3881 has been able to induce an upregulation of pro-inflammatory mediators both in vitro and in vivo. These results indicated that in vitro and in vivo functional characterizations are necessary for determination of virulence in different clinical isolates due to its modulation in the host–pathogen interactions.

Abstract Predator–prey interactions are important evolutionary drivers of defensive behaviors, but they are usually difficult to record. This lack of data on natural history and ecological interactions of species can be overcome through museum specimens, at least for some reptiles. When facing aggressive interactions, reptile species may exhibit the defensive behavior of autotomy by losing the tail, which is also known as “urotomy”. The inspection of preserved specimens for scars of tail breakage can reveal possible ecological and biological correlates of urotomy. Herein, we investigated how the probability of urotomy in the worm lizard Amphisbaena vermicularis is affected by sex, body size, temperature, and precipitation. We found higher chances of urotomy for specimens with larger body size and from localities with warmer temperatures or lower precipitation. There was no difference in urotomy frequency between sexes. Older specimens likely faced – and survived – more predation attempts through their lifetime than smaller ones. Specimens from warmer regions might be more active both below‐ and aboveground, increasing the odds to encounter predators and hence urotomy. Probability of urotomy decreased with increased precipitation. Possibly, in places with heavier rainfall worm lizards come more frequently to the surface when galleries are filled with rainwater, remaining more exposed to efficient predators, which could result in less survival rates and fewer tailless specimens. This interesting defensive behavior is widespread in squamates, but yet little understood among amphisbaenians. The novel data presented here improve our understanding on the correlates of tail breakage and help us to interpret more tales of lost tails.

BackgroundMental disorders are normally diagnosed exclusively on the basis of symptoms, which are identified from patients’ interviews and self-reported experiences. To make mental health diagnoses and monitoring more objective, different solutions have been proposed such as digital phenotyping of mental health (DPMH), which can expand the ability to identify and monitor health conditions based on the interactions of people with digital technologies. ObjectiveThis article aims to identify and characterize the sensing applications and public data sets for DPMH from a technical perspective. MethodsWe performed a systematic review of scientific literature and data sets. We searched 8 digital libraries and 20 data set repositories to find results that met the selection criteria. We conducted a data extraction process from the selected articles and data sets. For this purpose, a form was designed to extract relevant information, thus enabling us to answer the research questions and identify open issues and research trends. ResultsA total of 31 sensing apps and 8 data sets were identified and reviewed. Sensing apps explore different context data sources (eg, positioning, inertial, ambient) to support DPMH studies. These apps are designed to analyze and process collected data to classify (n=11) and predict (n=6) mental states/disorders, and also to investigate existing correlations between context data and mental states/disorders (n=6). Moreover, general-purpose sensing apps are developed to focus only on contextual data collection (n=9). The reviewed data sets contain context data that model different aspects of human behavior, such as sociability, mood, physical activity, sleep, with some also being multimodal. ConclusionsThis systematic review provides in-depth analysis regarding solutions for DPMH. Results show growth in proposals for DPMH sensing apps in recent years, as opposed to a scarcity of public data sets. The review shows that there are features that can be measured on smart devices that can act as proxies for mental status and well-being; however, it should be noted that the combined evidence for high-quality features for mental states remains limited. DPMH presents a great perspective for future research, mainly to reach the needed maturity for applications in clinical settings.

Background Populations seem to respond differently to the global pandemic of severe acute respiratory syndrome coronavirus 2. Recent studies show individual variability in both susceptibility and clinical response to COVID-19 infection. People with chronic obstructive pulmonary disease (COPD) constitute one of COVID-19 risk groups, being already associated with a poor prognosis upon infection. This study aims contributing to unveil the underlying reasons for such prognosis in people with COPD and the variability in the response observed across worldwide populations, by looking at the genetic background as a possible answer to COVID-19 infection response heterogeneity. Methods SNPs already associated with susceptibility to COVID-19 infection (rs286914 and rs12329760) and severe COVID-19 with respiratory failure (rs657152 and rs11385942) were assessed and their allelic frequencies used to calculate the probability of having multiple risk alleles. This was performed on a Portuguese case-control COPD cohort, previously clinically characterized and genotyped from saliva samples, and also on worldwide populations (European, Spanish, Italian, African, American and Asian), using publicly available frequencies data. A polygenic risk analysis was also conducted on the Portuguese COPD cohort for the two mentioned phenotypes, and also for hospitalization and survival to COVID-19 infection. Findings No differences in genetic risk for COVID-19 susceptibility, hospitalization, severity or survival were found between people with COPD and the control group (all p-values > 0.01), either considering risk alleles individually, allelic combinations or polygenic risk scores. All populations, even those with European ancestry (Portuguese, Spanish and Italian), showed significant differences from the European population in genetic risk for both COVID-19 susceptibility and severity (all p-values < 0.0001). Conclusion Our results indicate a low genetic contribution for COVID-19 infection predisposition or worse outcomes observed in people with COPD. Also, our study unveiled a high genetic heterogeneity across major world populations for the same alleles, even within European sub-populations, demonstrating the need to build a higher resolution European genetic map, so that differences in the distribution of relevant alleles can be easily accessed and used to better manage diseases, ultimately, safeguarding populations with higher genetic predisposition to such diseases.

Mitochondrial Myopathy is a rare pathology caused by a defect in the mitochondrial DNA metabolism, leading to defects in the formation of adenosine triphosphate, in the Krebs citric acid cycle, fatty acid oxidation and oxidative phosphorylation. It is manifested by exercise intolerance, muscle fatigue on small efforts, muscle weakness, tachycardia, and difficulty breathing. There are few case reports on the operative management of adult patients suffering from mitochondrial myopathy. With this report, we intend to describe the anesthetic management of a patient with mitochondrial myopathy who underwent laparoscopic gastroplasty and outline some anesthetic considerations about this pathology.

Abstract There is a growing recognition that spatial scale is important for understanding ecological processes shaping community membership, but empirical evidence on this topic is still scarce. Ecological processes such as environmental filtering can decrease functional differences among species and promote functional clustering of species assemblages, whereas interspecific competition can do the opposite. These different ecological processes are expected to take place at different spatial scales, with competition being more likely at finer scales and environmental filtering most likely at coarser scales. We used a comprehensive dataset on species assemblages of a dominant ant genus, Pheidole, in the Cerrado (savanna) biodiversity hotspot to ask how functional richness relates to species richness gradients and whether such relationships vary across spatial scales. Functional richness of Pheidole assemblages decreased with increasing species richness, but such relationship did not vary across different spatial scales. Species were more functionally dissimilar at finer spatial scales, and functional richness increased less than expected with increasing species richness. Our results indicate a tighter packing of the functional volume as richness increases and point out to a primary role for environmental filtering in shaping membership of Pheidole assemblages in Neotropical savannas. Worldwide, there is great concern with the issue of bioaccumulation of organochlorine compounds in human milk and their potential impact on health. Breastfeeding is recognized as essential for the proper development and protection of children’s health. In this study, we collected human milk samples from 40 donors in Piracicaba - SP, and analyzed to determine the levels of residues of organochlorine pesticides: hexachlorobenzene (HCB), lindane, heptachlor, aldrin, heptachlor epoxide, o,p’ DDE, a- endosulfan, p,p’ DDE, dieldrin, endrin, b-endosulfan, endosulfan sulfate, and methoxychlor. The analysis was performed by solid phase extraction (SPE) and quantification by gas chromatography using electron capture detector (ECD). Residues of organochlorine pesticides were detected in 65% of the donors evaluated.

Phytophagous insects may choose host plants based on conditions that enhance offspring performance. However, some insect species may also select plants based on attributes that enhance their own performance regardless of the consequences for offspring survival. An approach evaluating both hypotheses could provide a more comprehensive understanding of the host plant selection by phytophagous insects. In this study, we described the life stages of a Neotropical stink bug, Edessa contermina, co-occurring on Byrsonima verbascifolia plants in a conservation area of the Brazilian Savannah. We also empirically evaluated how food supply, shelter availability and competitors’ density on the host plants affected the densities of nymphs, adults and mating pairs. We identified and described five life stages of E. contermina. The amount of plant resources did not explain the nymph, adult and mating pairs’ density. However, adults and mating pairs chose plants with a low density of nymphs, probably because egg laying on the host plants with a high density of competitors may negatively affect offspring performance.

Purpose: This study aimed to analyze the degree to which the meaning of work, specifically the work centrality dimension, and the types of achievement goal orientations at work, predict professional fulfillment. Originality/value: The main contribution is to provide background for the identification of antecedents of a concept yet narrowly studied, which is professional fulfillment, signaling how counselors and organizations may facilitate the greater achievement of what is most valued in a career. We evidence the need to consider work an important sphere of life and to invest efforts in self-development to attain professional fulfillment. Design/methodology/approach: This is quantitative, survey-type research involving 140 people who have been working for at least 6 months. An online questionnaire was answered containing absolute and relative work centrality, achievement goal orientation at work and professional fulfillment scales, as well as socio-demographic questions. Data were analyzed using bivariate correlations and multiple regression analysis. Findings: Significant relationships of prediction were found between work centrality and professional fulfillment, as well as between one of the types of goal orientation and professional fulfillment. The results support two of the three predicted hypotheses, in addition to converging with previous research on the positive impacts of work centrality and the mastery approach orientation. Objetivo: O artigo é conduzido pela seguinte questão: como as narrativas de grupos de interesses contribuem para o diálogo entre a perspectiva da estratégia como prática e a teoria dos stakeholders?. O objetivo é analisar as mudanças, permanências e tensões nas narrativas dos stakeholders, vistas como práticas estratégicas, no contexto da mineração. Originalidade/valor: A estratégia como uma prática social diz respeito às relações entre organizações, indivíduos e sociedade em busca de coexistência. Este estudo estabelece um diálogo entre a estratégia como prática social e uma perspectiva política dos stakeholders, considerando que interagem em redes, buscando a criação de valor a partir da mutualidade. Design/metodologia/abordagem: A pesquisa de campo foi conduzida numa abordagem qualitativa e longitudinal. A coleta de dados utilizou técnicas de pesquisa documental em atas e vídeos de audiências públicas, pesquisa em jornal e realização de entrevistas. Resultados: Os resultados mostram interações narrativas de seis grupos de interesse: organizações empreendedoras, poder público, educação, meio ambiente, moradores da área de implantação e setor produtivo. O resultado sugere um papel de relevância das tensões relacionais como elementos que restringem e possibilitam as mudanças e permanências, e evidenciam a natureza dinâmica das interações intra e intergrupos de stakeholders. O diálogo teórico proposto amplia o campo dos estudos da estratégia, em uma vertente pós-moderna, ao estabelecer um diálogo com uma vertente política da ação dos stakeholders, como alternativa a uma perspectiva funcionalista tradicional no campo.

ABSTRACT The increasing demand for electricity and the scarcity of resources, require renewable energy sources and efficient equipment that reduce the consumption of electricity. The Green Office (GO) of the Universidade Tecnológica Federal do Paraná (UTFPR) is a sustainable building that uses strategies to reduce impacts to the environment, one of them being the use of the grid connected photovoltaic system (on-grid). The on-grid was installed in 2011 and since then has been feeding the GO and another building (block V) belonging to UTFPR. This article presents a comparison between an estimate of energy consumption and the generation of energy through the on-grid. By means of the estimated consumption, a survey in loco of the scenario of expenditures of the electrical equipment used in the GO was made, estimating hours of use and power, so the consumption scenario was 145 kWh/month. The power generation of the EV is lower in the months May - July, period in which the solar irradiation is smaller, but according to the measurements the on-grid produces more energy than it consumes.

Resumo Introdução: O perfil clínico de pacientes brasileiros adultos com síndrome nefrótica por doença de lesões mínimas (LM) e glomeruloesclerose segmentar e focal (GESF) é pouco conhecido. Objetivo: Avaliamos as características clínico-laboratoriais e resposta a tratamento em pacientes adultos com síndrome nefrótica e diagnósticos histológicos de LM ou GESF. Métodos: Fez-se a análise retrospectiva de 50 pacientes adultos com LM e 120 com GESF. Todos os pacientes foram inicialmente tratados com corticosteroide. Os desfechos do estudo foram: resposta a corticosteroide, prevalência de remissão total, progressão para doença renal crônica estágio 5 (DRC5) e necessidade de terapia de substituição renal por DRC5. Resultados: Níveis iniciais de creatinina sérica foram 24% mais elevados entre pacientes com GESF (p = 0,02) e os de proteinúria foram 36% mais altos em LM (p < 0,001). Pacientes com LM foram córtico-sensíveis em 80% dos casos, com remissão total em 74%, e os pacientes com GESF em 58% (p = 0,01), com remissão total em 30% (p = 0,002). A prevalência de insuficiência renal aguda em pacientes com GESF foi de 39% (vs. 12%, p = 0,013) e DRC5 de 10% (vs. 0%, p < 0,001). Remissão completa ou parcial com o uso de corticosteroide reduziu em 83% o risco de DRC5 (p < 0,001) e remissão total associou-se a redução no risco de DRC5 de 89% (p < 0,001). Conclusão: A resposta positiva à corticoterapia foi o fator mais importante relacionado à preservação da função renal ao longo de mais de uma década de seguimento, e GESF relacionou-se a menor índice de resposta a corticosteroide.

RESUMOO objetivo deste trabalho foi avaliar possíveis efeitos do extrato das vargens de bordão-de-velho (Samanea tubulosa benth) sobre a reprodução em ratas Wistar. O extrato foi preparado a partir da secagem e trituração das vagens, seguida por maceração com etanol e, posteriormente, concentrado em rotavapor. Ratas Wistar gestantes foram tratadas com 75 mg.100 g-1 de peso corporal de extrato de S. tubulosa e o grupo controle recebeu 1ml/100 g-1 de peso corporal de solução salina por gavagem durante 19 dias. Após eutanásia no 20º dia, as seguintes variáveis foram analisadas: peso corporal fetal, peso dos fetos e placentas, número de corpos lúteos e de reabsorções, implantação, fetos vivos e mortos. O extrato de vagens de S. tubulosa apresenta toxicidade para o feto causando malformações, reabsorções fetais e diminuição pronunciada do peso ao nascer. Contudo, o extrato de vagens de S. tubulosanão influencia no número de corpos lúteos e número de implantações.

Theoretical molecular structures of the complexes [PdCl2(HmPz)2] (1) and [PdCl2(HIPz)2] (2) (HmPz = 4-methylpyrazole; HIPz = 4-iodopyrazole) were studied using B3LYP/DFT method. The new complex (2) and the complex (1) were synthesized and characterized by elemental analysis and IR spectroscopy. The calculated bond distances and angles showed that both compounds exhibited a square planar coordination environment around the palladium center. The theoretical IR spectra of Cs symmetry (electronic state 1A’) of the complexes agree well with the experimental data. A description of the molecular orbital (HOMO and LUMO) for compounds (1) and (2) was also given in this work.

The life cycle of Plasmodium falciparum, the agent responsible for malaria, depends on both cytosolic and apicoplast translation fidelity. Apicoplast aminoacyl-tRNA synthetases (aaRS) are bacterial-like enzymes devoted to organellar tRNA aminoacylation. They are all encoded by the nuclear genome and are translocated into the apicoplast only after cytosolic biosynthesis. Apicoplast aaRSs contain numerous idiosyncratic sequence insertions: An understanding of the roles of these insertions has remained elusive and they hinder efforts to heterologously overexpress these proteins. Moreover, the A/T rich content of the Plasmodium genome leads to A/U rich apicoplast tRNA substrates that display structural plasticity. Here, we focus on the P. falciparum apicoplast tyrosyl-tRNA synthetase (Pf-apiTyrRS) and its cognate tRNATyr substrate (Pf-apitRNATyr). Cloning and expression strategies used to obtain an active and functional recombinant Pf-apiTyrRS are reported. Functional analyses established that only three weak identity elements in the apitRNATyr promote specific recognition by the cognate Pf-apiTyrRS and that positive identity elements usually found in the tRNATyr acceptor stem are excluded from this set. This finding brings to light an unusual behavior for a tRNATyr aminoacylation system and suggests that Pf-apiTyrRS uses primarily negative recognition elements to direct tyrosylation specificity.
