Abstract

Life-threatening "breakthrough" cases of critical COVID-19 are attributed to poor or waning antibody (Ab) response to SARS-CoV-2 vaccines in individuals already at risk. Preexisting auto-Abs neutralizing type I IFNs underlie at least 15% of critical COVID-19 pneumonia cases in unvaccinated individuals; their contribution to hypoxemic breakthrough cases in vaccinated people is unknown. We studied a cohort of 48 individuals (aged 20 to 86 years) who received two doses of a messenger RNA (mRNA) vaccine and developed a breakthrough infection with hypoxemic COVID-19 pneumonia 2 weeks to 4 months later. Ab levels to the vaccine, neutralization of the virus, and auto-Abs to type I IFNs were measured in the plasma. Forty-two individuals had no known deficiency of B cell immunity and a normal Ab response to the vaccine. Among them, 10 (24%) had auto-Abs neutralizing type I IFNs (aged 43 to 86 years). Eight of these 10 patients had auto-Abs neutralizing both IFN-α2 and IFN-ω, whereas two neutralized IFN-ω only. No patient neutralized IFN-β. Seven neutralized type I IFNs at 10 ng/ml and three at 100 pg/ml only. Seven patients neutralized SARS-CoV-2 D614G and Delta efficiently, whereas one patient neutralized Delta slightly less efficiently. Two of the three patients neutralizing only type I IFNs at 100 pg/ml neutralized both D614G and Delta less efficiently. Despite two mRNA vaccine inoculations and the presence of circulating Abs capable of neutralizing SARS-CoV-2, auto-Abs neutralizing type I IFNs may underlie a notable proportion of hypoxemic COVID-19 pneumonia cases, highlighting the importance of this particularly vulnerable population.

13. Modelación y simulación de un secador de lecho vibrofluidizado para sólidos que contienen mezclas solventes

Abstract

Se estudió el secado de un sólido granular en un secador continuo de lecho vibrofluidizado por medio de simulaciones. Se desarrolló un modelo considerando el secado de una capa delgada de partículas humedecidas con una mezcla multicomponente. El modelo asume mezclado perfecto en la dirección del flujo de aire y considera solamente los cambios longitudinales del contenido líquido, composición y temperatura de la partícula. Los principales mecanismos para la transferencia de masa y calor dentro de las partículas son la difusión interactiva y conducción. Asumiendo una matriz constante de coeficientes efectivos de difusión para las partículas, se obtienen las soluciones analíticas de las ecuaciones de difusión y conducción. La variación de los coeficientes de difusión y de la conductividad térmica de las partículas a lo largo del secador es tomada en cuenta por una aplicación paso a paso de la solución analítica en intervalos de espacio con coeficientes promedios del paso anterior. La solución analítica representa un método apropiado para estudiar la selectividad y la retención de aroma durante el secado de alimentos. La solución es computacionalmente rápida; por lo tanto, la verificación experimental de este modelo aproximado sería muy beneficiosa ya que el tratamiento riguroso del secado multicomponente implica cálculos dilatados y tediosos.

Abstract

En este trabajo se estudió la cinética de secado de la levadura cervecera tomando en consideración el fenómeno de encogimiento presentado por la misma. Los datos experimentales se obtuvieron a tres temperaturas diferentes. Se evaluó el encogimiento volumétrico y de grosor de la levadura por medición directa determinada durante el secado. Se encontró que el encogimiento de la levadura es casi isotrópico e incide significativamente en la velocidad de secado. Además, la levadura durante el secado presenta solamente un periodo de velocidad decreciente y no exhibe curva característica de secado.

Abstract

BackgroundWe previously reported that impaired type I IFN activity, due to inborn errors of TLR3- and TLR7-dependent type I interferon (IFN) immunity or to autoantibodies against type I IFN, account for 15-20% of cases of life-threatening COVID-19 in unvaccinated patients. Therefore, the determinants of life-threatening COVID-19 remain to be identified in ~ 80% of cases.MethodsWe report here a genome-wide rare variant burden association analysis in 3269 unvaccinated patients with life-threatening COVID-19, and 1373 unvaccinated SARS-CoV-2-infected individuals without pneumonia. Among the 928 patients tested for autoantibodies against type I IFN, a quarter (234) were positive and were excluded.ResultsNo gene reached genome-wide significance. Under a recessive model, the most significant gene with at-risk variants was TLR7, with an OR of 27.68 (95%CI 1.5-528.7, P = 1.1 × 10-4) for biochemically loss-of-function (bLOF) variants. We replicated the enrichment in rare predicted LOF (pLOF) variants at 13 influenza susceptibility loci involved in TLR3-dependent type I IFN immunity (OR = 3.70[95%CI 1.3-8.2], P = 2.1 × 10-4). This enrichment was further strengthened by (1) adding the recently reported TYK2 and TLR7 COVID-19 loci, particularly under a recessive model (OR = 19.65[95%CI 2.1-2635.4], P = 3.4 × 10-3), and (2) considering as pLOF branchpoint variants with potentially strong impacts on splicing among the 15 loci (OR = 4.40[9%CI 2.3-8.4], P = 7.7 × 10-8). Finally, the patients with pLOF/bLOF variants at these 15 loci were significantly younger (mean age [SD] = 43.3 [20.3] years) than the other patients (56.0 [17.3] years; P = 1.68 × 10-5).ConclusionsRare variants of TLR3- and TLR7-dependent type I IFN immunity genes can underlie life-threatening COVID-19, particularly with recessive inheritance, in patients under 60 years old.

Abstract

Extracellular vesicles (EVs) released by virus-infected cells have the potential to encapsulate viral peptides, a characteristic that could facilitate vaccine development. Furthermore, plasma-derived EVs may elucidate pathological changes occurring in distal tissues during viral infections. We hypothesized that molecular characterization of EVs isolated from COVID-19 patients would reveal peptides suitable for vaccine development. Blood samples were collected from three cohorts: severe COVID-19 patients (G1), mild/asymptomatic cases (G2), and SARS-CoV-2-negative healthcare workers (G3). Samples were obtained at two time points: during the initial phase of the pandemic in early 2020 (m0) and eight months later (m8). Clinical data analysis revealed elevated inflammatory markers in G1. Notably, non-vaccinated individuals in G1 exhibited increased levels of neutralizing antibodies at m8, suggesting prolonged exposure to viral antigens. Proteomic profiling of EVs was performed using three distinct methods: immunocapture (targeting CD9), ganglioside-capture (utilizing Siglec-1) and size-exclusion chromatography (SEC). Contrary to our hypothesis, this analysis failed to identify viral peptides. These findings were subsequently validated through Western blot analysis targeting the RBD of the SARS-CoV-2 Spike protein’s and comparative studies using samples from experimentally infected Syrian hamsters. Furthermore, analysis of the EV cargo revealed a diverse molecular profile, including components involved in the regulation of viral replication, systemic inflammation, antigen presentation, and stress responses. These findings underscore the potential significance of EVs in the pathogenesis and progression of COVID-19.

Light pollution is spreading around the world, causing a variety of problems, particularly for nocturnal animals such as bats. As urban areas expand, attic-dwelling bats face the problem of being exposed to more and more artificial light as they move to their dark foraging areas. Avoiding street lights could force bats to make energetically costly detours, which could eventually lead to the collapse of urban colonies. We therefore investigated the response of attic-dwelling bats to street lighting, using the brown long-eared bat (Plecotus auritus) as an example. Using radio-tracking, we followed 17 bats during their commutes through two urban areas: a small town and a village in rural Germany. After a conservative filtering process to ensure a high spatial accuracy of the tracking data, we analysed the data of nine bats in detail, using a use-versus-availability approach for the small town and a descriptive approach for the smaller dataset obtained from the village. The best-fitting model confirmed that the bats responded to artificial light at night. Close to the roost (∼10 m), bats preferred areas with low light intensities, but seemed to tolerate some light levels. We speculate that this pattern was caused by bats being forced to leave the roost into an illuminated area. At medium distances from roosts (∼400 m), most bats avoided street lights. In fact, all bats avoided being in close proximity to street lights, even when the lanterns produced only low levels of light. We conclude that brown long-eared bats avoid street lights when moving through urban areas. We recommend the establishment of dark corridors in urban areas to support high bat diversity, including all species along the continuum of light-opportunistic and light-sensitive species.

Industrialization [...], Circuit courts are divided as to whether Section 602 of Title VI ofthe 1962 Civil Rights Act creates a private right of action enforceable under 42 U.S.C. [section] 1983. In the past, many environmental justice groups have brought their claims under Section 602, since it is easier to prove disparate impact as opposed to direct discrimination. However, recent trends in Section 1983 jurisprudence seem to indicate that unless the statute expressly creates a private right of action, it is usually not enforceable under Section 1983. Moreover, courts have muddled substantive due process claims related to "state-created dangers" by adding subjective thresholds (e.g., private actor requirements, "shock the conscience"standards, and mens rea requirements). The Administrative Procedure Act ("APA") allows individuals to challenge agency compliance with the National Environmental Policy Act ("NEPA") and seek judicial review of agency decisions implicating environmental justice issues; therefore, NEPA challenges through the APA would be an appropriate alternative to Section 1983 actions. In addition to the procedural mechanisms available through the APA, the Inflation Reduction Act of 2022 ("IRA") includes several sources of funding dedicated to addressing environmental justice issues throughout the country. With a clear mandate from Congress, NEPA challenges will be the most effective strategy to address environmental justice concerns along the U.S.-Mexico border where most environmental justice issues are linked to major federal actions.

29. Drivers for the Development of Computational Thinking in Costa Rican Students

Abstract

This study provides evidence about factors that facilitate the development of computational thinking (CT) in Costa Rican elementary school students, including the description of the contribution of the LIE++ proposal that addresses CT knowledge and practices through programming and physical computing projects. A quasi-experimental design was used to compare a group of students from the LIE++ educational proposal with a group of students from another proposal called LIE-Guides, which emphasizes learning with digital technologies. The study sample comprised 14,795 voluntary students, who answered an online test that was constructed and validated to estimate the scores achieved in CT. The results showed that the students participating in the LIE++ proposal obtained better scores compared to the LIE-Guides group. A multilevel regression model demonstrated that students' personal and social variables, as well as the proposal's execution scheme, positively affected student learning in CT. This research is a first approach to the subject in this context. It refers to the importance of providing educational opportunities that focus on more advanced computing knowledge and skills, as well as the relevance of continuing to develop tools and methodologies that help generate evidence about CT in education in order to improve educational interventions.

Abstract

Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Abstract

BackgroundAntiretroviral therapy (ART) intensification and disruption of latency have been suggested as strategies to eradicate HIV. ABX464 is a novel antiviral that inhibits HIV RNA biogenesis. We investigated its effect on HIV transcription and total and intact HIV DNA in CD4+ T cells from ART-suppressed participants enrolled in the ABIVAX-005 clinical trial (NCT02990325).MethodsPeripheral CD4+ T cells were available for analysis from 9 ART-suppressed participants who were treated daily with 150 mg of ABX464 for 4 weeks. Total and intact HIV DNA and initiated, 5'elongated, unspliced, polyadenylated, and multiply-spliced HIV transcripts were quantified at weeks 0, 4, and 8 using ddPCR.ResultsWe observed a significant decrease in total HIV DNA (P = .008, median fold change (mfc) = 0.8) and a lower median level of intact HIV DNA (P = not significant [n.s.], mfc = 0.8) after ABX464 treatment. Moreover, we observed a decrease in initiated HIV RNA per million CD4+ T cells and per provirus (P = .05, mfc = 0.7; P = .004, mfc = 0.5, respectively), a trend toward a decrease in the 5'elongated HIV RNA per provirus (P = .07, mfc = 0.5), and a lower median level of unspliced HIV RNA (P = n.s., mfc = 0.6), but no decrease in polyadenylated or multiply-spliced HIV RNA.ConclusionsIn this substudy, ABX464 had a dual effect of decreasing total HIV DNA (and possibly intact proviruses) and HIV transcription per provirus. To further characterize its specific mechanism of action, long-term administration of ABX464 should be studied in a larger cohort.Clinical trials registrationNCT02990325.

36. Understanding How African Americans Develop Resilience in Overcoming the Social Paradox of Opportunity: A Qualitative Study

Abstract

The purpose of this descriptive phenomenological qualitative study was to understand the lived experiences of African Americans who have leveraged their resilience to overcome barriers to complete post-secondary education. The study used a descriptive phenomenological qualitative research design, while the population studied was African Americans, particularly those who have completed their college education. The preferred sampling technique was purposive sampling, and the sample size obtained was 10 participants. An in-depth interview was the instrument used for collecting the desired data for the study. The interviews occurred via Zoom. Thematic analysis was employed for analyzing the collected data. The findings showed that African American students experience prejudice or inequity in college, but they leverage resilience in a variety of ways that ultimately helps them triumph over their encounters with discrimination and inequality. The findings also showed that African American students usually experience a social paradox of opportunity whereby they struggle to access similar opportunities as their white counterparts. However, through resilience developed through their support systems, they manage to overcome those adversities and graduate from college. The findings give devoted support for prior research by integrating the findings on how resilience enables individuals to overcome obstacles and achieve their goals. In addition, the study seeks to resolve the social paradox of opportunity that African American students face in universities, thereby improving the prospects of African American and other minority students to complete their degrees and secure well-paying careers in which they can make positive contributions to society. Abstract

This study explores the matrices of serial style present in Aaron Sorkin's dramaturgical writing for television serials. The focus is on the way in which teams of directors associated with Sorkin’s productions employ solutions for the staging of these works, thereby establishing their recognizable styles. The analysis values the heuristic uses of the concept of “style”, in disciplines of art history and literature studies, aiming to understand the dynamics under which television serials results from creative encounters between dramaturgy and mise-en-scène. As proving grounds of this examination, we analyse the works of two directors of Aaron Sorkin’s show, The West Wing, evaluating the diversified manners in which these two propose solutions for the staging of Sorkin’s writing.

A locale (frame) L has a largest spatial sublocale generated by the primes (spectrum points), the spatial part SpL. In this paper we discuss some of the properties of the embeddings SpL ⊆ L. First we analyze the behaviour of the spatial parts in the assembly: the points of L and of S(L)^op (∼=the congruence frame) are in a natural one-one correspondence while the topologies of SpL and Sp(S(L)^op) differ. Then we concentrate on some special types of embeddings of SpL into L, namely in the questions when SpL is complemented, closed, or open. Background: Blood OX40-expressing CD4 T-cells from antiretroviral (ART)-treated people living with HIV (PWH) were found to be enriched for clonally-expanded HIV sequences, hence contributing to the HIV reservoir. OX40-OX40L is also a checkpoint regulator of inflammation in multiple diseases. We explored gut mucosal OX40+CD4+ T-cells and their potential significance in HIV disease. Methods: Biopsies of caecum and terminal-ileum of ART-treated PWH (n = 32) were obtained and mucosal damage and HIV reservoir were assessed. Mucosal OX40+ and Ki67+ CD4 T-cell subsets, as well as several tissue T-cell subsets modulating mucosal integrity and homeostasis (Th17, Th22, Treg, Tc17, Tc22, IL17+TCRγδ, IL22+TCRγδ) were quantified. Inflammatory-related markers, T-cell activation and thymic output were also determined in blood samples. Correlations were explored using Spearman rank test and corrected for multiple comparisons by Benjamini-Hochberg. BackgroundThe BCN02-trial combined therapeutic vaccination with a viral latency reversing agent (romidepsin, RMD) in HIV-1-infected individuals and included a monitored antiretroviral pause (MAP) as an efficacy read-out identifying individuals with an early or late (< or > 4weeks) viral-rebound. Integrated -omics analyses were applied prior treatment interruption to identify markers of virus control during MAP.MethodsPBMC, whole-genome DNA methylation and transcriptomics were assessed in 14 BCN02 participants, including 8 Early and 4 Late viral-rebound individuals. Chromatin state, histone marks and integration analysis (histone-3 acetylation (H3Ac), viral load, proviral levels and HIV-specific T cells responses) were included. REDUC-trial samples (n = 5) were included as a control group for RMD administration alone.FindingsDNA methylation imprints after receiving the complete intervention discriminated Early versus Late viral-rebound individuals before MAP. Also, differential chromatin accessibility and histone marks at DNA methylation level were detected. Importantly, the differential DNA methylation positions (DMPs) between Early and Late rebounders before MAP were strongly associated with viral load, proviral levels as well as the HIV-specific T-cell responses. Abstract

Coastal lagoons hold significant ecological value due to their rich biodiversity and essential roles in supporting ecosystems. However, they are increasingly threatened by accelerated climate change, and it is crucial to assess these environments’ ecological status for present and future conditions resulting from the impacts of climate change. In this context, the present work aims to evaluate the present and future ecological status of Ria de Aveiro through the application of the numerical model Delft3D. The model was validated, and the results demonstrate that it effectively captures the main characteristics of the lagoon dynamics, although achieving accurate water quality representation poses challenges due to interdependencies in solutions and the inherent complexity of associated processes. The model was explored to characterize the environmental factors of the lagoon and evaluate its ecological status through the computation of several indexes. According to the model results, the main environmental factors present seasonal variations consistent with temperate climates. Regarding the ecological status of Ria de Aveiro, the central channels of the lagoon mostly hold a Good/Moderate status, while regions near river inflows tend to exhibit Moderate to Poor conditions. In future conditions, water quality is expected to improve in winter and autumn due to reductions in river-borne pollutants resulting from the projected decrease in river flow. For spring and summer, a decline in water quality is projected mainly due to the increase in phosphate concentrations in the lagoon. This study provides valuable insights into the ecological dynamics of coastal lagoons under changing climatic conditions, contributing to improved management and mitigation strategies. The findings can guide future conservation efforts and help mitigate the adverse effects of climate change on these vital ecosystems.
