15. Investigation on biofilm composition and virulence traits of S. pseudintermediusisolated from infected and colonized dogs

Abstract

Staphylococcus pseudintermedius, which is part of the skin microbiome of dogs, causes a variety of opportunistic infections. These infections may become more difficult to treat due to the formation of biofilm. The capacity of S. pseudintermediusto form biofilm, as well as the associated genes, has not been elucidated. This study evaluated the production and composition of S. pseudintermediusbiofilm. Samples were collected from both infected dogs and asymptomatic dogs. Isolates were identified using mass spectrometry and Multiplex-PCR. Biofilm production and composition were assessed using a quantitative microtiter plate assay. The presence of icaoperon genes and spsgenes was investigated using conventional PCR. The investigation of Agr type and virulence genes was conducted in silico on 24 sequenced samples. All strains could produce strong biofilms, with most of the isolates presenting a polysaccharide biofilm. 63.6% of the isolates carried the complete icaoperon (ADBC). All samples showed the presence of the genes spsK, spsA, and spsL, while the distribution of other genes varied. Agr type III was the most prevalent (52.2%). All sequenced samples carried the cytotoxins hlb, luk-S, luk-F, as well as the exfoliative toxins sietand se_int. No isolate displayed other exfoliative toxins. Only LB1733 presented a set of different enterotoxins (sea, seb, sec_canine, seh, sek, sel, and seq). Our findings suggest that S. pseudintermediusis a strong producer of biofilm and carries virulence genes.

Abstract

Osteomyelitis often involves Staphylococcus spp. as the isolated genus in domestic animal cases. Implant-related infections, frequently associated with biofilm-forming microorganisms like staphylococci species, necessitate careful material selection. This study assessed biofilm formation by Staphylococcus pseudintermedius on titanium nuts used in veterinary orthopaedic surgery. Biofilm quantification employed safranin staining and spectrophotometric measurement, while bacterial counts were determined in colony-forming units (CFU). Scanning Electron Microscopy (SEM) evaluated the biofilm morphology on the surface of titanium nuts. All samples had CFU counts. Absorbance values that evidence biofilm formation were observed in seven of the eight samples tested. SEM images revealed robust bacterial colonization, and significant extracellular polymeric substance production, and the negative control displayed surface irregularities on the nut. Whole genome sequencing revealed accessory Gene Regulator (agr) type III in six samples, agr IV and agr II in two each. Genes encoding hlb, luk-S, luk-F, siet, se_int, and the icaADCB operon were identified in all sequenced samples. Other exfoliative toxins were absent. Biofilm formation by S. pseudintermedius was detected in all samples, indicating the susceptibility of orthopaedic titanium alloys to adhesion and biofilm formation by veterinary species. Abstract

Sporothrix brasiliensis is the most pathogenic species, responsible for the Brazilian cat-transmitted sporotrichosis hyperendemic. In this scenario, an investigation of the pathogen–host interaction can provide relevant information for future treatment strategies. To this end, the invertebrate Galleria mellonella has proven to be a suitable alternative for evaluating the virulence of pathogenic fungi, since the insect immune system is similar to the mammalian innate immune response. The aim of this work was to investigate phenotypic and molecular aspects of the immune response of G. mellonella throughout the S. brasiliensis infection. Hemocyte density and the evolution of the fungal load were evaluated. In parallel, RT-qPCR expression analysis of genes encoding antimicrobial peptides (Gallerimycin and Galiomycin) and stress management genes (C7 Contig 15362 and C8 Contig 19101) was conducted. The fungal load and hemocyte densities increased simultaneously and proportionally to the deleterious morphological events and larvae mortality. Gallerimycin, C7 Contig 15362 and C8 Contig 19101 genes were positively regulated (p < 0.05) at distinct moments of S. brasiliensis infection, characterizing a time-dependent and alternately modulated profile. Galiomycin gene expression remained unchanged. Our results contribute to the future proposal of potential alternative pathways for treating and consequently controlling S. brasiliensis zoonosis, a major public health issue in Latin America. Abstract

Sporothrix brasiliensiswith low susceptibility isolates were described from the Brazilian zoonotic sporotrichosis hyperendemics. The aim of this work was to evaluate distinct fractions of Ocotea pulchella, Ocotea notata, Myrciaria floribunda, and Hypericum brasilienseplant extracts against itraconazole-sensitive and low susceptibility S. brasiliensisisolates. Crude extracts were tested against clinical isolates and the ATCC MYA4823 to determine the minimum inhibitory concentrations (MICs) and fungicidal or fungistatic activities (MFC). A high MICs and MFCs amplitude (1 – > 128 µg/mL) were obtained for seven extracts. The highest antimicrobial activities against sensitive S. brasiliensiswere displayed by the ethyl acetate extracts of O. notata(MIC = 2–128 μg/mL) and M. floribunda(MIC = 1–8 μg/mL). A fungicidal effect was observed for all fraction extracts. Ocoteaspp. and M. floribundaethyl acetate extracts provide promising profiles against itraconazole-sensitive or low susceptibility S. brasiliensis. Future studies will determine if these extracts can contribute as alternative therapies to this neglected zoonosis.

Abstract

Staphylococcus aureus is one of the most common opportunistic pathogens in humans and several animals. Some strains can form a biofilm, which is related to chronic diseases. Green Propolis are used frequently in alternative medicine. Several studies have demonstrated the chemical similarities between Green Propolis and its botanical source Baccharis dracunculifolia. This study evaluated the antibacterial profile of the hydroalcoholic extracts of Green Propolis and Baccharis dracunculifolia against S. aureus. The antibiofilm effect of the extracts was also evaluated against a reference strain of S. aureus. The Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) for each isolate were assessed. MIC and MBC values for both extracts were similar against all bacterial strains. The extracts demonstrated good performance against the biofilm of the S. aureus ATCC 25923. Green Propolis and Baccharis dracunculifolia extract antibiofilm activities were mainly on biofilm's newly formed and consolidated moments. [ABSTRACT FROM AUTHOR]

40. Detection of the international lineage ST71 of methicillin-resistant Staphylococcus pseudintermediusin two cities in Rio de Janeiro State

Abstract

Staphylococcuspseudintermediusis the main coagulase-positive staphylococci associated with canine skin/soft tissue infections (SSTI), otitis externa, and surgical site infections. The international spread of an epidemic and multiresistant lineage of methicillin-resistant Staphylococcus pseudintermedius(MRSP), the so-called European clone—displaying sequence type (ST) 71—requires attention. The first isolation of an MRSP ST71 isolate in South America was reported in Rio de Janeiro city, in 2010; however, a limited number of canine isolates were analyzed. Thus, to have a better panel of the MRSP spread in this city, we were stimulated to continue this study and search for the presence of MRSP in 282 colonized or infected dogs in the city of Rio de Janeiro. Among the MRSP isolates collected (N= 17; 6.1%), the pulsed-field gel electrophoresis (PFGE) patterns were similar to those of European clone. All 17 isolates were classified as ST71 by multilocus sequence typing (MLST). In order to assess whether isolates of MRSP ST71 may have also spread to the Rio de Janeiro state countryside, we collected samples from 124 infected dogs in the city of Campos dos Goytacazes (232 km away from Rio de Janeiro city). Our data showed the presence of ST71 lineage in one isolate among three MRSP detected. S. pseudintermediuswas isolated from 40.6% of the clinical samples (N= 165/406). A relatively high incidence of methicillin resistance, detected by a PCR-based method, was found in 12.1% of the S. pseudintermediusrecovered from animals (N= 20/165). The resistance profile of these isolates was similar to that described for the international ST71 strains whose genomes are publicly available in the GenBank. The prospect of ST71 isolates being resistant to virtually all antimicrobials used in veterinary medicine is alarming and should be considered a central issue considering that MRSP ST71 spreads over large geographic distances and its transmission from animals to humans.

Abstract

National audience; The site on which the Saint Thomas Chapel is located in Aizier was a medieval leprosarium, then a modern priory belonging to the Fécamp Abbaye. This establishment was the focus of a ten-year long excavation programme. The field work has now been completed. However, the research is still in progress. The living conditions of a small community of lepers, undoubtedly quite poor, who lived and died in Aizier between the 12th and 16th centuries, have already begun to emerge.; Le site de la « chapelle Saint-Thomas » à Aizier fut une léproserie médiévale, puis un prieuré moderne dépendant de l’abbaye de Fécamp. Cet établissement a fait l’objet d’une fouille programmée durant 10 ans. La phase terrain est à présent achevée, mais les études sont toujours en cours. Cependant commence déjà à se dessiner le cadre de vie d’une petite communauté de lépreux, sans doute fort pauvre, qui vécut et mourut à Aizier entre les XIIe et XVIe siècles.

46. Antibiotic resistance in Staphylococcus sp. isolated from the vaginal environment of squirrel monkeys ( Saimiri spp. ) bred ex situ.

Abstract

Background Squirrel monkeys ( Saimiri spp.) have been widely used as animal models; however, the occurrence of Staphylococcus sp in their vaginal microbiota remains to be described. Methods Samples were collected from 175 adult squirrel monkeys to isolate Staphylococcus sp and to test for susceptibility to a panel of nine antimicrobial agents. Results Isolates with characteristics of the genus Staphylococcus were detected in 95 of 175 samples. Coagulase-negative staphylococci (Co NS) were the most common (95.8%, 91/95) isolates. Resistance to antibiotics was observed in 47.3% (45/95) of isolates. Resistance to tetracycline was observed in 28.5% (26/91), chloramphenicol in 15.4% (14/91), and methicillin in 13.2% (12/91) of Co NS. Coagulase-positive staphylococci were resistant to tetracycline, erythromycin, and methicillin. Conclusions The presence of Staphylococcus sp in vaginal samples obtained from squirrel monkeys suggests that these animals were in a carrier state. Furthermore, isolating strains resistant to methicillin reinforces the biosafety care of a colony. [ABSTRACT FROM AUTHOR]

47. Use of ozonated sunflower oil in the treatment of infectious endometritis in mares.

Abstract

Endometritis can be classified as infectious or non-infectious, clinical or sub-clinical. It is one of the important causes of infertility and sub-fertility in mares. Taking into account the antimicrobial, immuno-stimulating capacity and the low cost of therapies with the combination oxygen-ozone (O2-O3), we aimed to evaluate the effectiveness of ozonized sunflower oil as a possible alternative treatment for endometritis in mares. Mares (n=20) with a known reproductive history of uterine infection and inflammation were selected and distributed into 2 groups: Control (n = 10), treated with non-ozonized sunflower oil and Treatment (n = 10), treated with oil of ozonated sunflower oil (Ozone & Life). Pre- and post-uterine changes were identified by means of transrectal ultrasound examination (B mode) and confirmed by cytological examination (cytological brush) and microbiological analysis (swab) through bacterial and fungalcultures. Collection of samples and treatment applications were performed during estrus. An insemination pipette coupled to a syringe filled with the respective oil (60ml) was inserted directly into the body of the uterus. The oil was left in the uterus for 24 hours, after which the uterus was washed with Ringer Lactate solution until a completely translucent flushing was obtained. Preliminary results demonstrate that ozonized sunflower oil has the potential to treat fungal and bacterial endometritis; one case of uterine infection caused by Aspergillus spp and 7of 9 cases of bacterial endometritis resolved. Bacteria involved in these infections were Staphylococcus delphini, Staphylococcus intermedius, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Enterobacter bugandensis, Staphylococcus chromogenes, Klebsiella pneumoniae and Staphylococcus sciuri.
