Metallic materials characterization is a important part of phase transformation studies, and the sample preparation plays a crucial role in the measured properties. The methodologies available are not fully described and refer in its majority only to iron-based and aluminum-based alloys focusing in industrial analysis. In this paper the authors fully described the preparation methodology for metallic samples used by their research group. Controlled human infection model (CHIM) studies, which involve deliberate exposure of healthy human volunteers to an infectious agent, are recognised as important tools to advance vaccine development. These studies not only facilitate estimates of vaccine efficacy, but also offer an experimental approach to study disease pathogenesis and profile vaccine immunogenicity in a controlled environment, allowing correlation with clinical outcomes. Consequently, the data from CHIMs can be used to identify immunological correlates of protection (CoP), which can help accelerate vaccine development. In the case of invasive Salmonella infections, vaccination offers a potential instrument to prevent disease. Invasive Salmonella disease, caused by the enteric fever pathogens Salmonella enterica serovar Typhi (S. Typhi) and S. Paratyphi A, B and C, and nontyphoidal Salmonella (iNTS), remains a significant cause of mortality and morbidity in low- and middle-income countries, resulting in over 200,000 deaths and the loss of 15 million DALYs annually. CHIM studies have contributed to the understanding of S. Typhi infection and provided invaluable insight into the development of vaccines and CoP following vaccination against S. Typhi. However, CoP are less well understood for S. Paratyphi A and iNTS. This brief review focuses on the contribution of vaccine-CHIM trials to our understanding of the immune mechanisms associated with protection following vaccines against invasive Salmonella pathogens, particularly in relation to CoP.

Summary: Klebsiella pneumoniae is among the World Health Organization's list of priority pathogens, notorious for its role in causing healthcare-associated infections and neonatal sepsis globally. Containment of K. pneumoniae transmission depends on the continued effectiveness of antimicrobials and of biocides used for topical antisepsis and surface disinfection. Klebsiella pneumoniae is known to disseminate antimicrobial resistance (AMR) through a large auxiliary genome made up of plasmids, transposons and integrons, enabling it to evade antimicrobial killing through the use of efflux systems and biofilm development. Because AMR mechanisms are also known to impart tolerance to biocides, AMR is frequently linked with biocide resistance (BR). However, despite extensive research on AMR, there is a gap in knowledge about BR and the extent to which AMR and BR mechanisms overlap remains debatable. The aim of this paper is to review and summarise the current knowledge on the determinants of BR in K. pneumoniae and highlight content areas that require further inquiry.

Este trabalho tem como proposta verificar a temática Vida Extraterrestre e Astrobiologia em alguns documentos oficiais como os Parâmetros Curriculares Nacionais (PCN), Currículo Básico Comum de Minas Gerais (CBC Mineiro) e a Base Nacional Comum Curricular (BNCC), tendo como referência a Física do Ensino Médio. Pesquisou-se também o conhecimento dos estudantes sobre o tema Vida Extraterrestre e Astrobiologia, por meio de um questionário aplicado em quatro escolas estaduais de Minas Gerais. A pesquisa foi realizada no segundo ano do Ensino Médio, somando um total de 82 participantes. A análise das respostas fundamentou-se nas técnicas de análise de conteúdo. Verificou-se na análise documental que nos PCN e na BNCC, os temas Vida Extraterrestre e Astrobiologia são obrigatórios, porém no CBC Mineiro esses conteúdos não são abordados. Apesar de ser um tema recomendado pelos documentos oficiais, verificou-se a necessidade de mais pesquisas educacionais e publicações em congressos e periódicos explorando esse conteúdo.

In Botswana, Special Needs Education has been implemented for 25 years with some success but there is still a need for evidence-based methods like Frequency Building, behavioural fluency, and Precision Teaching to be used to measure and improve school performance and learning. We explored the impact of these behavioural technologies on reading performances of four children with learning disorders (ADHD, speech impairment and acquired brain disorder) in a special school in Gaborone. At the assessment, two children were unable to read letter sounds and two could not read sight words. Reading performances were measured with frequency and displayed on a standard celeration chart. During the intervention, the length of the tasks was reduced and then augmented. Findings revealed that after 3 months of intervention children significantly increased their score stimulating self-confidence and enthusiasm during activities. This work demonstrates that behavioural technologies can be applied in Africa without using expensive or time-consuming resources.

RESUMO Este artigo objetivou comparar o senso de autoeficácia dos professores da Pré-Escola e Ensino Fundamental I para ensinar estudantes público-alvo da Educação Especial (PAEE) na sala de aula regular da rede pública municipal e relacionar o senso de autoeficácia com dados demográficos e de atuação profissional. Participaram da pesquisa 17 professores da Pré-Escola e 27 do Ensino Fundamental 1 de um município do interior paulista. Os dados foram obtidos mediante questionário de caracterização e a Escala de Eficácia Docente para Práticas Inclusivas (EEDPI) via tendência central, dispersão e correlacionais. Quando comparados com os professores do Fundamental 1, os da Pré-Escola possuem mais idade, menos alunos em sala e sentem-se mais preparados para incluir. Os valores de autoeficácia foram semelhantes entre os grupos e são descritos os dados de correlação entre os instrumentos. Corroborou-se na sinalização da potência e sensibilidade da EEDPI para avaliação da autoeficácia docente e da influência do constructo no contexto educativo.

Este artigo avalia como veículos jornalísticos de checagem de fatos podem, indiretamente, realizar crítica de mídia. Para isso, é proposta uma tipologia dos efeitos pedagógicos da verificação jornalística como uma crítica de mensagens (refutando cada informação incorreta), veículos (denunciando produtores que publicam informações falsas), meios (criticando plataformas que permitem circulação de desinformação), recepção (educando usuários sobre como identificar fake news e checar eles mesmos conteúdos on-line) e controle (discutindo propostas para alterar o sistema midiático, por meio de leis ou alterações no funcionamento de plataformas), além de, ao mesmo tempo, serem alvo de uma forma de metacrítica – uma crítica contra os checadores e suas checagens, que trata dos critérios para a crítica midiática publicada por jornalistas.

Cervical cancer remains a significant cause of morbidity and mortality in women worldwide and is the leading cause of cancer-related death in Botswana. It is well established that women with HIV have a higher risk of persistent HPV infection leading to cervical cancer. We assessed HPV prevalence and genotype distribution in 126 tissue specimens from confirmed invasive cervical cancer cases using Abbott real-time PCR assay. Overall, 88 (69.8%) women were HIV-infected. Fifty-seven (64.8%) of the HIV-infected women had a baseline CD4+ count ≥350 cells/μl, and 82 (93.2%) were on antiretroviral therapy at the time of cervical cancer diagnosis. Background: The epidemiology of extended-spectrum cephalosporin-resistant Enterobacterales (ESCrE) in hospitalized patients in low- and middle-income countries (LMICs) is poorly described. Although risk factors for ESCrE clinical infection have been studied, little is known of the epidemiology of ESCrE colonization. Identifying risk factors for ESCrE colonization, which can predispose to infection, is therefore critical to inform antibiotic resistance reduction strategies. Methods: This study was conducted in 3 hospitals located in 3 districts in Botswana. In each hospital, we conducted ongoing surveillance in sequential units hospitalwide. All participants had rectal swabs collected which were inoculated onto chromogenic media followed by confirmatory testing using MALDI-TOF MS and VITEK-2. Data were collected via interview and review of the inpatient medical record on demographics, comorbidities, antibiotic use, healthcare exposures, invasive procedures, travel, animal contact, and food consumption. Participants with ESCrE colonization (cases) were compared to noncolonized participants (controls) using bivariable and multivariable analyses to identify risk factors for ESCrE colonization. Results: Enrollment occurred from January 15, 2020, to September 4, 2020, and 469 participants were enrolled. The median age was 42 years (IQR, 31–58) and 320 (68.2%) were female. The median time from hospital admission to date of sampling was 5 days (IQR, 3–12). There were 179 cases and 290 controls (ie, 38.2% of participants were ESCrE colonized). Independent risk factors for ESCrE colonization were a greater number of days on antibiotic, recent healthcare exposure, and tending swine prior to hospitalization. (Table). Conclusions: ESCrE colonization among hospitalized patients was common and was associated with several exposures. Our results suggest prior healthcare exposure may be important in driving ESCrE. The strong link to recent antibiotic use highlights the potential role of antibiotic stewardship interventions for prevention. The association with tending swine suggests that animal husbandry practices may play a role in community exposures, resulting in colonization detected at the time of hospital admission. These findings will help to inform future studies assessing strategies to curb further emergence of hospital ESCrE in LMICs.

Como resposta à preocupação coletiva com a disseminação de notícias falsas on-line, o Congresso tem debatido o Projeto de Lei 2.630, que pretende instituir a Lei Brasileira de Liberdade, Responsabilidade e Transparência na Internet — apelidado de “PL das Fake News”. Esta pesquisa avalia como o PL 2.630 define e afeta práticas jornalísticas, considerando como a reação do público sobre o projeto trata do seu impacto sobre a circulação de informações jornalísticas. A metodologia adotada envolve a comparação dos projetos de lei e a análise de conteúdo dos comentários do público na plataforma da Câmara dos Deputados. A mudança no foco do PL — da proibição de disseminação de desinformação à remuneração de veículos da imprensa — ampliou as resistências do público, que considera a proposta restritiva de espaços digitais, criticando o fortalecimento de meios tradicionais.

The use of copper as an antimicrobial agent has a long history and has gained renewed interest in the context of the COVID-19 pandemic. In this study, the authors investigated the antimicrobial properties of an alloy composed of copper with a small percentage of silver (Cu-0.03% wt.Ag). The alloy was tested against various pathogens, including Escherichia coli, Staphylococcus aureus, Candida albicans, Pseudomonas aeruginosa, and the H1N1 virus, using contact exposure tests. Results showed that the alloy was capable of inactivating these pathogens in two hours or less, indicating its strong antimicrobial activity. Electrochemical measurements were also performed, revealing that the small addition of silver to copper promoted a higher resistance to corrosion and shifted the formation of copper ions to higher potentials. This shift led to a slow but continuous release of Cu2+ ions, which have high biocidal activity. These findings show that the addition of small amounts of silver to copper can enhance its biocidal properties and improve its effectiveness as an antimicrobial material.

This can also be attributed to African genomes that contain more variation than those from other parts of the world. The purpose of this review is to summarize published evidence on pharmacogenetic and pharmacokinetic aspects related to specific available treatments and the known genetic variabilities associated with metabolism and/or transport of breast cancer drugs, and treatment outcomes when possible. The emphasis is on the African genetic variation and focuses on the genes with the highest strength of evidence, with a close look on CYP2A6, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, CYP3A4/5, CYP19A1, UGT1A4, UGT2B7, UGT2B15, SLC22A16, SLC38A7, FcγR, DPYD, ABCB1, and SULT1A1, which are the genes known to play major roles in the metabolism and/or elimination of the respective anti-breast cancer drugs given to the patients. The genetic variability of their metabolism could be associated with different metabolic phenotypes that may cause reduced patients’ adherence because of toxicity or sub-therapeutic doses. Background: The combination of rivaroxaban plus aspirin compared with aspirin alone reduces the risk of major adverse cardiovascular and limb events for high-risk patients with peripheral artery disease. It is unknown whether rivaroxaban plus aspirin improves intermittent claudication for adults with lower-risk peripheral arterial disease. Methods: In this randomized, open-label, multicenter, 24-week clinical trial, we randomly assigned patients with peripheral artery disease and intermittent claudication to receive either 2.5 mg of rivaroxaban twice daily plus 100 mg of aspirin once daily or 100 mg of aspirin once daily. The primary outcome was a 24-week change in total walking distance, measured by the 6-minute walking test. The primary safety outcome was the incidence of major bleeding or clinically relevant nonmajor bleeding. Results: Eighty-eight patients were randomly assigned to either rivaroxaban plus aspirin (n=46) or aspirin alone (n=42). The mean age was 67 years, and 54% were female. The total walking distance measured by 6-minute walk test improved by 89 ± 18 m (mean±standard error) in the rivaroxaban-plus-aspirin group versus 21 ± 16 m in the aspirin-alone group. This corresponded to an absolute difference of 68 ± 24 m (95% confidence interval [CI], 19 to 116 m; P=0.007) and a relative improvement over the aspirin-alone group of 327% (95% CI, 94 to 560%). No major bleeding events were observed in either group. Conclusions: In patients with peripheral artery disease and intermittent claudication, 2.5 mg of rivaroxaban twice daily plus 100 mg of aspirin daily improved the total walking distance by a 6-minute walking test compared with 100 mg of aspirin daily alone. Abstract Background The variation of human papillomavirus (HPV) genotypes shapes the risks of cervical cancer and these variations are not well defined in Africa. Nucleotide changes within the L1 gene, nucleotide variability, and phylogeny were explored in relation to HIV in samples from Botswana and Kenya. Methods A total of 98 HPV-positive cervical samples were sequenced to identify different HPV variants. Phylogenetic inferences were used to determine HPV genotypes and investigate the clustering of sequences between women living with HIV (WLWHIV) and -women not living with HIV (WNLWHIV). Results Out of 98 generated sequences, 83.7% (82/98) participants had high-risk (HR) HPV genotypes while 16.3% (16/98) had low-risk (LR) HPV genotypes. Este artigo avalia a condenação do jornalista Rubens Valente, autor do livro Operação banqueiro, e sua editora por terem difamado o ministro Gilmar Mendes do Supremo Tribunal Federal (STF). A pesquisa avalia como o processo se encaixa na tradição do STF de tutela da liberdade de expressão em colisão com outros direitos da personalidade, como a honra, em particular nos casos envolvendo seus próprios ministros. Foca-se na decisão do ministro Alexandre de Moraes, confirmada pela primeira turma do STF, que determinou o pagamento de indenização e obrigou a publicação da petição de Mendes e das decisões judiciais condenando o jornalista em futuras edições da obra. O artigo discute, além disso, como a indenização e a publicação de documentos legais extensos podem inviabilizar a obra, caracterizando um cenário análogo ao de censura.

Abstract In this paper, a new methodology is applied to optimize the geometry of conductors of overhead transmission lines (TL) with high precision and low computational cost. This methodology is based on the sensitivity analysis of the electrical charge of the transmission lines using the adjoint method. This information is used with the gradient method and the golden section algorithm to minimize the electric field at the ground level of three-phase TL with two cables per phase. The approximation using central finite differences to obtain sensitivity is adopted for validation and comparisons. The TL's with high surge impedance loading is obtained after the optimization process.

Cervical cancer burden is still high in low- and middle-income countries, including Botswana. Persistent human papillomavirus (HPV) infection is the leading cause of cervical cancer. Accurate knowledge of HPV diversity associated to cervical cancer in sub-Saharan Africa may provide accurate understanding of the natural history of HPV infection in these contexts. The goal of this review was to consolidate existing evidence on cervical HPV infection and to conduct a pooled analysis of data from all eligible Botswana studies. After a successful review of twelve studies on cervical HPV genotypes that met the inclusion criteria, HPV-16 genotype was the most frequently discovered in women with pre-cancerous and cancer lesions, followed by HPV-18. HPV-16 in HIV-positive women with precancerous lesions to cancer is between 45% and 47.7%, and between 4.5% and 26.1% for HPV-18. With reference to other HPV genotypes, the proportion of HPV-35 and HPV-58 (13-16%) seems relatively consistent among the studies, however HPV-58 appears to be more common in HIV-positive subjects compared to HIV-negative women. Indeed, HPV-45 seems to be frequently detected in women with cervical cancer compared to women with precancerous lesions. Regarding the low-risk HPV genotypes, an appropriate breakdown has been provided. In conclusion, the current prophylactic vaccines against HPV-16 and HPV-18, which have demonstrated good immunogenicity in HIV-infected populations, may still prevent infection and ultimately cancer.

Background. The continued spread of infectious diseases by mosquitoes remains a formidable obstacle to the well-being of the people all over the world. Arboviruses are spread from one vertebrate host to another by vectors through intricate transmission cycles that involve the virus, the vertebrate host, and the vector. It is essential to acquire a better understanding of the current abundance and distribution of major vectors in order to adequately prepare for the possibility of arbovirus outbreaks. This is because the abundance and distribution of these major vectors determines the human populations that are at risk for the diseases that they transmit. The effects of climate change on the amount of mosquitoes and their ability to survive the seasons have had a substantial impact on the spread of diseases that are transmitted by vectors in many different parts of Botswana. Methods. The purpose was to collect mosquito samples in Gaborone and the neighboring areas in southern Botswana, including border stations. We collected different stages of the mosquito from each place, raised them to maturity, and then identified them. Both morphological and genetic studies were utilized in order to successfully identify the organism. The species of Culex mosquitoes accounted for 88.3% of the 5177 mosquitoes that were collected and identified, whereas the species of Aedes aegypti and Anopheles mosquitoes accounted for 11.5% and 0.2% respectively. Conclusions. These findings give entomological baseline data that will aid in the study of vectorial patterns and the estimation of future arboviral hazards provided by mosquitoes. Additionally, these findings document the diversity and abundance of mosquito species.

The global malaria community has picked up the theme of malaria elimination in more than 90% of the world’s population in the next decade. Recent reports of Plasmodium vivax (P. vivax) in sub-Saharan Africa, including in Duffy-negative individuals, threaten the efforts aimed at achieving elimination. This is not only in view of strategies that are tailored only to P. falciparum elimination but also due to currently revealed biological characteristics of P. vivax concerning the relapse patterns of hypnozoites and conservation of large biomasses in cryptic sites in the bone marrow and spleen. A typical scenario was observed in Botswana between 2008 and 2018, which palpably projects how P. vivax could endanger malaria elimination efforts where the two parasites co-exist. The need for the global malaria community, national malaria programs (NMPs), funding agencies and relevant stakeholders to engage in a forum to discuss and recommend clear pathways for elimination of malaria, including P. vivax, in sub-Saharan Africa is warranted.

Objectives: Plasmodium vivax malaria was thought to be rare in Africans who lack the Duffy blood group antigen expression. However, recent studies indicate that P. vivax can infect Duffy-negative individuals and has spread into areas of high Duffy negativity across Africa. Our study compared epidemiological and genetic features of P. vivax between African regions. Methods: A standardized approach was used to identify and quantify P. vivax from Botswana, Ethiopia, and Sudan, where Duffy-positive and Duffy-negative individuals coexist. The study involved sequencing the Duffy binding protein (DBP) gene and inferring genetic relationships among P. vivax populations across Africa. Results: Among 1215 febrile patients, the proportions of Duffy negativity ranged from 20–36% in East Africa to 84% in southern Africa. Average P. vivax prevalence among Duffy-negative populations ranged from 9.2% in Sudan to 86% in Botswana. Parasite density in Duffy-negative infections was significantly lower than in Duffy-positive infections. P. vivax in Duffy-negative populations were not monophyletic, with P. vivax in Duffy-negative and Duffy-positive populations sharing similar DBP haplotypes and occurring in multiple, well-supported clades. Conclusions: Duffy-negative Africans are not resistant to P. vivax, and the public health significance of this should not be neglected. Our study highlights the need for a standardized approach and more resources/training directed towards the diagnosis of vivax malaria in Africa.

Abstract The meshless element-free Galerkin method (EFGM) is used to solve partial differential equations responsible for obtaining the electromagnetic fields generated by a transmission line. For this purpose, a 2D model based in a real transmission line is constructed and simulated. The results obtained by EFGM have good accuracy and they are compared against analytical solution and Finite Element Method in order to verify the effectiveness, advantages and disadvantages of the method.

Abstract Background In spite of the global effort to eliminate malaria, it remains the most significant vector-borne disease of humans. Plasmodium falciparum is the dominant malaria parasite in sub-Saharan Africa. However, Plasmodium vivax is becoming widely spread throughout Africa. The overuse of vector control methods has resulted in a remarkable change in the behaviour of mosquito that feeds on human as well as on vector composition. The aim of this study was to identify Anopheles mosquito species in vivax malaria endemic regions and to investigate their role in P. vivax circumsporozoite protein (Pvcsp) allele diversity. Methods Mosquito samples were collected from Central Sudan (Rural Khartoum and Sennar) and Eastern Sudan (New Halfa, Kassala state) using pyrethrum spray catch (PSC) and CDC light traps. Mosquitoes were identified using appropriate morphological identification keys and Anopheles gambiae complex were confirmed to species level using molecular analysis. A subset of blood-fed anopheline mosquitoes were dissected to determine the presence of natural infection of malaria parasites. In addition, the rest of the samples were investigated for the presence of Pvcsp gene using nested-PCR. Results A total of 1037 adult anopheline mosquitoes were collected from New Halfa (N = 467), Rural Khartoum (N = 132), and Sennar (N = 438). Morphological and molecular identification of the collected mosquitoes revealed the presence of Anopheles arabiensis (94.2%), Anopheles funestus (0.5%), and Anopheles pharoensis (5.4%). None of the dissected mosquitoes (N = 108) showed to be infected with malaria parasite. Overall P. vivax infectivity rate was 6.1% (63/1037) by Pvcsp nested PCR. Co-dominance of An. arabiensis and An. pharoensis is reported in Sennar state both being infected with P. vivax. Conclusion This study reported P. vivax infection among wild-caught anopheline mosquitoes in Central and Eastern Sudan. While An. arabiensis is the most abundant vector observed in all study areas, An. funestus was recorded for the first time in New Halfa, Eastern Sudan. The documented Anopheles species are implicated in Pvcsp allele diversity. Large-scale surveys are needed to identify the incriminated vectors of P. vivax malaria and determine their contribution in disease transmission dynamics.

Polymorphisms of the CYP2B6 gene influence drug levels in plasma and possibly virological outcomes. The aim of this study was to explore the potential impact of CYP2B6 genotype and haplotype variation on the risk of developing EFV/NVP drug resistance mutations (DRMs) in HIV-1 patients receiving EFV-/NVP-containing regimens in Botswana.Patients and Methods: Participants were a sub-sample of a larger study (Tshepo study) conducted in Gaborone, Botswana, among HIV-infected individuals taking EFV/NVP containing ART. Study samples were retrieved and assigned to cases (with DRMs) and controls (without DRMs). Four single-nucleotide polymorphisms (SNPs) in the CYP2B6 gene (− 82T>C; 516G>T; 785A>G; 983T>C) were genotyped, the haplotypes reconstructed, and the metabolic score assigned. The possible association between drug resistance and several independent factors (baseline characteristics and CYP2B6 genotypes) was assessed by Binary Logistic Regression (BLR) analysis. EFV/NVP resistance status and CYP2B6 haplotypes were also analyzed using Z-test, chi-square and Fisher’s exact test statistics.Results: Two hundred and twenty-seven samples were analysed (40 with DRMs, 187 without DRMs). BLR analysis showed an association between EFV/NVP resistance and CYP2B6 516G allele (OR: 2.26; 95% CI: 1.27– 4.01; P=0.005). Moreover, haplotype analysis revealed that the proportion of EFV/NVP-resistant infections was higher among CYP2B6 fast than extensive/slow metabolizers (30.8% vs 16.8%; P= 0.035), with the 516G allele more represented in the haplotypes of fast than extensive/slow metabolizers (100.0% vs 53.8%; P< 0.001).Conclusion: We demonstrated that the CYP2B6 516G allele, and even more when combined in fast metabolic haplotypes, is associated with the presence of EFV/NVP resistance, strengthening the need to assess the CYP2B6 genetic profiles in HIV-infected patients in order to improve the virologic outcomes of NNRTI containing ART.Keywords: ART, CYP2B6 gene, drug resistance selection, fast metabolizers, HIV

This paper proposes a new methodology for sensitivity analysis evaluation, fast and with high precision of the electric potential distribution near the transmission lines (TL's). The TL is modeled by the finite element method and the sensitivity of the cable positions is obtained using the adjoint method. The sensitivity of the objective function during the optimization process by using methods based on gradient information is obtained by using the adjoint method. The exact sensitivity obtained by the adjoint method concerning the numeric model of the TL's results in new geometries of the bundles conductors with high surge impedance loading. These geometries are not possible to get using analytic models. The sensitivities of a large number of conductors by phase are obtained with high precision and very low computational cost using adjoint sensitivities, which are independent of the number of design variables. The central finite difference method is used to calculate the sensitivities and to validate the adjoint method. With this methodology, the FEM model of the TL can be used during the optimization process without compromising the required computational processing time. [ABSTRACT FROM AUTHOR]

A proliferação de informações falsas em redes sociais online criou um cenário de desconfiança em relação às fontes informativas. A preocupação em relação à ameaça de notícias falsas se tornou um terreno fértil para demandas coletivas por propostas para combater a desinformação. Esta pesquisa parte de estudos sobre respostas sociais às notícias falsas, avaliando a proliferação de sites de checagem de fatos, ameaças de controle legal e alterações técnicas em algoritmos online, contrastando essas estratégias com o potencial de iniciativas baseadas na educomunicação. Baseado na metodologia da pesquisa-ação, foi analisado um panorama de propostas de educação midiática existentes em diferentes plataformas (canais de vídeo, páginas multimídia, oficinas presenciais, curso online aberto e massivo sincrônico ou não), comparado possibilidades de interatividade, participação e engajamento do público. Como resultado, foi identificado a falta de uma proposta de curso online gratuito, assincrônico, massivo, multimídia, interativo, acessível, atraente e com potencial de viralização em redes sociais. Em resposta às lacunas nesse campo, foi desenvolvido um novo mecanismo de combate à desinformação usando dos princípios da educomunicação, mas ocupando o mesmo espaço de proliferação das notícias falsas – as redes sociais – e usando seus formatos específicos para lutar contra as fake news em seu próprio ambiente. Desenvolvido como resultado dessa pesquisa, o curso ‘Vaza, Falsiane!’ obteve em dois anos 200 mil visitantes, e seus conteúdos alcançaram mais de 10 milhões pelo Facebook.

Abstract. The two non-nucleoside reverse transcriptase inhibitors (NNRTIs), efavirenz (EFV) and nevirapine (NVP), are currently the core antiretroviral drugs for treatment of HIV in sub-Saharan Africa including Botswana. The drugs are metabolized by Cytochrome P450 2B6 (CYP2B6) liver enzyme. The CYP2B6 gene that encodes for metabolism of these drugs is known to be highly polymorphic. One of the polymorphism in the CYP2B6 gene, 516G>T, particularly the 516T allele, is known to confer poor metabolism of EFV and NVP. This may lead to high levels of plasma drug concentrations and development of treatment toxicities, like central nervous system toxicities, and cutaneous and hepatic toxicities, for EFV and NVP, respectively. The CYP2B6 516G allele on the other hand is associated with an extensive metabolism of the two NNRTIs drugs. We sought to establish association between possible developments of NNRTIs toxicities with CYP2B6 516G>T variation in Botswana. A total of 316 peripheral blood mononuclear cells samples were used in a retrospective view. All the samples were from participants on EFV/NVP-containing regimen with known toxicity output. TaqMan Real-Time PCR approach was applied for assessing CYP2B6 516 allele variation in cases with treatment toxicity and those without. Analysis was performed by chi-square statistics and logistic regression analysis. The rate of poor metabolizers among participants with toxicity and those without toxicity was 18.4% and 15.1%, respectively. The CYP2B6 516 genotype distribution comparisons between the participants with toxicity and those without were not statistically different (chi-square = .326; P = .568). CYP2B6 516 variation was not associated with NNRTI toxicity. No other factors were associated with toxicity when considering age, baseline body mass index, baseline CD4, baseline HIV viral load and adherence. The results were discussed in the context of all the studies done in Botswana to date.
