1. High nutritional value instant flakes produced from various cereal grains

Abstract

The current study was carried out to prepare functional flakes from various cereals and to assess the nutritional value of breakfast cereal flakes and their sensory acceptability. Oat, soft wheat and durum wheat, barley (hull-less and hulled), triticale, millet and sorghum grains have been used. Physicochemical, functional, phytochemical properties and sensory evaluation were determined. The developed cereal flakes have high nutritional value and are high in dietary fiber. Concerning the overall acceptability of flakes, durum wheat, hull-less barley and triticale were more preferred than the other samples. Meanwhile, hulled barley and millet flakes showed the lowest scores compared with other flake samples due to the lowest score of their appearance and color. Triticale, durum wheat and hulled barley turned to be good alternatives for oat to prepare flakes of high-quality characteristics, as they have high protein (13.46, 11.92 and 11.67%, respectively) and ash contents along with low content of fat and low calories. In terms of nutritional quality, the results indicated that oat flakes were higher in Mg and P content, durum wheat flakes showed higher content of Mn, triticale had the highest content of K and Ca, and millet had the highest content of Zn and Fe. Triticale flakes showed significantly higher content of dietary fiber and water absorption index at room and hot temperatures when compared with other flakes. While comparing total phenolic content, the millet and barley flakes showed the highest values. Additionally, most flake samples had significantly higher values of antioxidant activity compared to oat flakes used as control samples.

Percutaneous active mechanical circulatory support (MCS) devices are being increasingly used in the treatment of acute myocardial infarction-related cardiogenic shock (AMICS) despite conflicting evidence regarding their effect on mortality. We aimed to ascertain the effect of early routine active percutaneous MCS versus control treatment on 6-month all-cause mortality in patients with AMICS.

Abstract

Abstract Diabetes mellitus is a worldwide problem characterized by hyperglycemia as well as the damage of the microscopic structure of the beta cells of Langerhans pancreatic islets. In the present study, the histological, immunohistochemical, morphometric, and biochemical alterations to pancreatic beta cells in streptozocin (STZ)-induced diabetes were assessed in rats treated with curcumin (CU) (100 mg/kg/day) or nano-curcumin (nCU) (100 mg/kg/day) for 1 month. Twenty-four adult male Wistar albino rats were distributed into four groups: the nondiabetic control group, the diabetic untreated group, and two diabetic groups treated with CU or nCUR, respectively. Blood glucose, serum insulin levels, and lipid profile were measured. The pancreatic tissues were collected and processed into paraffin sections for histological and immunohistochemical examination, oxidative stress markers, and real-time PCR expression for pancreatic and duodenal homeobox 1 (PDX1). The insulin expression in beta cells was assessed using immunohistochemistry. Morphometrically, the percentage area of anti-insulin antibody reaction and the percentage area of islet cells were determined. STZ-induced deteriorating alteration in beta cells led to declines in the number of functioning beta cells and insulin immunoreactivity. In STZ-treated rats, CU and nCUR significantly reduced blood glucose concentration while increasing blood insulin level. It also caused a significant increase in the number of immunoreactive beta cells to the insulin expression and significant reduction of the immunoreactive beta cells to the caspase-3 expression. In conclusion, CU and nCUR could have a therapeutic role in the biochemical and microscopic changes in pancreatic beta cells in diabetes-induced rats through STZ administration with more bio-efficacy of nCUR.

Abstract

L’élevage des ruminants est caractérisé par une diversité importante de conduites et de pratiques ayant un impact sur le bien-être des animaux. En plein air, les animaux peuvent être exposés à des variations climatiques. En bâtiment, la liberté des mouvements et la qualité de l’aire de couchage sont des facteurs déterminants du confort ressenti par l’animal et de son état sanitaire. Une alimentation inadaptée aux besoins physiologiques, peut provoquer des perturbations métaboliques ou des déviations comportementales. Les modifications des groupes sociaux sont également potentiellement néfastes. Par exemple, des remaniements fréquents de la composition d’un groupe conduisent à un stress et une dégradation de l’état sanitaire et des performances zootechniques. Les interventions comme la castration, l’écornage ou la césarienne, semblent avoir un impact d’une durée limitée sur l’animal. Enfin, l’importance de la relation homme – animal sur le bien-être des ruminants semble accrue du fait de la tendance à une augmentation du nombre d’animaux par soigneur et une diminution du temps passé par animal. En conclusion, une multitude de facteurs, correspondant majoritairement aux conditions d’élevage et aux interventions zootechniques, peut influencer le bien-être des ruminants. Les recherches dans ce domaine doivent s’orienter sur l’impact des pratiques ainsi que les alternatives à développer.

Using online survey methods (n = 153), this study investigates whether exposure to different levels of customization in games will increase the experiences of enjoyment, both directly and indirectly, through the experience of avatar embodiment, as well as whether these relationships are influenced by gender identity. Results indicate a positive relationship between level of exposure to customization and enjoyment for women-identified participants. 22. Teenagers’ vocabulary knowledge and self-efficacy as elements in effective L2 online searches

Abstract

As English dominates the Internet, most education systems need to prepare pupils to perform L2 information searches. Generating efficient search terms and understanding the information retrieved requires good vocabulary knowledge. This study examined the relationship between vocabulary knowledge, L2 online searching, and attitudes towards online searching. A sample of Swedish pupils’ abilities in locating suitably difficult texts and learning words were tested by comparing their Vocabulary Size Test scores with the lexical density of their search results and subsequent text production. Long-term vocabulary recall and self-efficacy for searching online were also tested. The results indicate that pupils tended to read and write texts below their ability level. They were unaware that they could achieve more as their self-efficacy measures indicated high confidence levels despite rather modest skills. The challenge posed by the findings is to enable teenagers to recognize how much more effectively they could use the Internet, without damaging their confidence.

23. Searching in the wild:Swedish teenagers’ online searching during English lessons

Abstract

All school leavers should have efficient online searching skills, defined here as the ability to select suitable search terms, feed them into a search engine, and make active decisions about which options to open from the results page. An additional element in our study includes the ability to search in English. As less than 1% of the Internet is in Swedish and about 60% is in English, this study assumes that Swedish school leavers will need to be able to use both languages. The study provides a qualitative, naturalistic account of the searching activities of Swedish teenagers in English classes from Grade 8 through to English 6 (ages 14‐18 years). Screen recordings from normal English lessons were analysed and supplemented with stimulated recall interviews with 40% of the pupils. The study reveals low levels of ambition and limited digital competence combined with high levels of confidence. Most pupils were unaware that they could have achieved more. The same pattern emerged across four school years in eight different classes. We conclude that improvements in digital competence education require more modelling of skills such as searching, so that teenagers recognize the value of aiming high.

Abstract

Youth get their sexual health information from social media, often from social media influencers (SMIs) or microcelebrities with large followings. Previous research suggests that SMIs have powerful persuasive effects on attitudes and behaviors. Thus, it is important to examine the ways in which sexual health information, such as birth control, is conveyed by SMIs. Using framing theory as a theoretical framework, this study examines characteristics of SMIs and their shared experiences pertaining to birth control. A content analysis of YouTube vlogs (n = 50) posted from December 2019–2021 was conducted on SMIs who talk about their experiences using hormonal and non-hormonal birth control. SMI status was determined based on the number of people subscribed to the YouTube channels. Results suggest that SMI YouTube videos are primarily about the discontinuation of hormonal birth control and may provide inaccurate sexual health information. Reasons for discontinuation of hormonal birth control provided by the SMIs are discussed. Future research should explore the effects of influencer sexual health messaging on beliefs, attitudes, and behaviors. [ABSTRACT FROM AUTHOR]

Le vote en ligne est souvent évoqué ces dernières années comme une solution pour lutter contre la hausse de l’abstention. Emmanuel Macron avait par exemple prévu dans son programme de 2017 de « généraliser le vote électronique d’ici 2022 ». Si la promesse n’a pas été tenue, le contexte de crise sanitaire et la distanciation sociale généralisée qui s’en est suivie tout autant que l’abstention massive de ces dernières années (26 % d’abstention au premier tour de l’élection présidentielle de 2022), ont amené de nombreuses personnalités politiques à se prononcer en faveur du vote en ligne. Pour autant, en France, où se rendre au bureau de vote reste un rituel républicain,on peut penser que le vote en ligne serait accueilli plus difficilement par les citoyens que dans certains autres pays. Les premiers résultats de notre enquête post-électorale dans le cadre People 2022 nous permettent d’éclairer ce débat.

Abstract

International audience; Introduction: At the beginning of the COVID-19 pandemic, a lockdown was implemented in France, leading to profound changes in families’ life. Studies showed emotional and behavioral difficulties in children and adults during this period. However, few research has focused on the longitudinal interactions between children’s emotion regulation and parental emotional experience of the lockdown. Aim: This study aimed to examine the longitudinal bidirectional relationships between children’s emotion regulation and parental emotional experience of the lockdown. Method: One hundred and twenty parents of children aged 5 to 12 answered two online questionnaires, at the start of the first lockdown (Time 1), and one month later (Time 2). Emotional dysregulation in children and parental emotional experience of the lockdown were assessed. Results: The results of the autoregressive cross-lagged analyses revealed that children’s emotion regulation at Time 1 predicted lockdown-related parental emotional experience at Time 2, whereas lockdown-related parental emotional experience at Time 1 didn’t predict children’s emotion regulation at Time 2. Conclusion: This study suggested that parents of children with greater emotion regulation difficulties have had a worse emotional experience of the lockdown, without affecting the emotional regulation of children. Clinical implications and protective factors are discussed.; Introduction : Au début de la pandémie de COVID-19, un confinement a été mis en place en France, entraînant de profonds changements dans la vie des familles. Des études ont montré des difficultés émotionnelles et comportementales chez les enfants et les adultes pendant cette période. Cependant, peu de recherches se sont concentrées sur les interactions longitudinales entre la régulation émotionnelle des enfants et l'expérience émotionnelle parentale du confinement. Objectif : Cette étude visait à examiner les relations bidirectionnelles longitudinales entre la régulation émotionnelle des enfants et l'expérience émotionnelle parentale du confinement. Méthode : Cent vingt parents d'enfants âgés de 5 à 12 ans ont répondu à deux questionnaires en ligne, au début du premier confinement (Temps 1), et un mois plus tard (Temps 2). La dysrégulation émotionnelle chez les enfants et l'expérience émotionnelle parentale du confinement ont été évaluées. Résultats : Les résultats des analyses autorégressives croisées ont révélé que la régulation émotionnelle des enfants au temps 1 prédisait l'expérience émotionnelle parentale liée au confinement au temps 2, alors que l'expérience émotionnelle parentale liée au confinement au temps 1 ne prédisait pas la régulation émotionnelle des enfants au temps 2. Conclusion : Cette étude suggère que les parents d'enfants ayant de plus grandes difficultés de régulation émotionnelle ont vécu une expérience émotionnelle du confinement plus difficile, sans affecter la régulation émotionnelle des enfants. Les implications cliniques et les facteurs de protection sont discutés.

31. Prevalence of 16S rRNA methylase genes among β-lactamase-producing Enterobacteriaceae clinical isolates in Saudi Arabia

Abstract

Background: Co production of 16S rRNA methylases gene and β-Lactamase gene among Enterobacteriaceae isolates conferring resistance to both therapeutic options has serious implications for clinicians worldwide. Methods: To study co existence of 16S rRNA methylases (armA, rmtA, rmtB, rmtC, rmtD, and npmA) and β-Lactamase (blaTEM-1, blaSHV-12, blaCTX-M-14) genes, we screened all phenotypic positive β-Lactamase producing enterobacteriaceae by polymerase chain reaction (PCR) targeting above genes. A total of 330 enterobacteriaceae strains were collected during study period out of that 218 isolates were identified phenotypically as β-Lactamase producers, which include 50 (22.9%) Escherichia coli; 92 (42.2%) Klebsiella pneumoniae, 44 (20.2%), Citrobactor freundii and 32 (14.7%) Enterobacter spp. Results: Among this 218, only 188 isolates harbored the resistant gene for β-Lactamase production. Major β-Lactamase producing isolates were blaTEM-1 type. 122 (56 %) isolates were found to produce any one of the 16S rRNA methylase genes. A total of 116 isolates co produced β-Lactamase and at least one 16S rRNA methylases gene Co production of armA gene was found in 26 isolates with rmtB and in 4 isolates with rmtC. The rmtA and rmtD genes were not detected in any of the tested isolates. Six isolates were positive for a 16S rRNA methylase gene alone. Conclusion: β-Lactamase producing isolates appears to coexist with 16S rRNA methylase predominantly armA and rmtB genes in the same isolate. We conclude the major β-Lactamase and 16S rRNA methylases co-producer was K. pneumoniae followed by E. coli. We suggest further work on evaluating other β-lactamases types and novel antibiotic resistance mechanisms among Enterobacteriaceae.

Abstract

Glycemic traits are used to diagnose and monitor type 2 diabetes and cardiometabolic health. To date, most genetic studies of glycemic traits have focused on individuals of European ancestry. Here we aggregated genome-wide association studies comprising up to 281,416 individuals without diabetes (30% non-European ancestry) for whom fasting glucose, 2-h glucose after an oral glucose challenge, glycated hemoglobin and fasting insulin data were available. Trans-ancestry and single-ancestry meta-analyses identified 242 loci (99 novel; P < 5 x 10-8), 80% of which had no significant evidence of between-ancestry heterogeneity. Analyses restricted to individuals of European ancestry with equivalent sample size would have led to 24 fewer new loci. Compared with single-ancestry analyses, equivalent-sized trans-ancestry fine-mapping reduced the number of estimated variants in 99% credible sets by a median of 37.5%. Genomic-feature, gene-expression and gene-set analyses revealed distinct biological signatures for each trait, highlighting different underlying biological pathways. Our results increase our understanding of diabetes pathophysiology by using trans-ancestry studies for improved power and resolution.

Abstract

Serological assays for SARS-CoV-2 are being utilized at an exponential rate for surveillance programs. This enterprise was designed to develop and validate a qualitative immunochromatographic test, via the Lateral Flow Assay (LFA), for detection of immunoglobulins M and G (IgM and IgG) against both nucleocapsid (N) and the receptor-binding domain (RBD) of the spike protein of SARS-CoV-2. Both targeted proteins were cloned and expressed in baculovirus expression system utilizing insect cells Sf9. The recombinant RBD and N proteins were purified and conjugated with gold nanoparticles (AuNPs) to set up the coating antigens pad. Both anti-human IgG and IgM were dispensed on nitrocellulose membrane to capture human antibodies in serum samples. A home-made dispensing system was developed to draw identical test and control lines. The validity of the developed LFA was verified by testing serum samples from 103 convalescent COVID-19 patients who were PCR positive for SARS-CoV-2 along with 28 control serum samples. The developed strips showed distinctive bands for IgM and IgG of both proteins (RBD and N) in positive samples. The sensitivity of RBD-based LFA was 70.9% and 39.8% for IgG and IgM, respectively, with a specificity of 100% for both. The N-based LFA exhibited a sensitivity of 73.8% and 35.9% for IgG and IgM, respectively, while its specificity was 75% and 100% for IgG and IgM, respectively. Our developed LFA could afford a tool for surveillance programs in low-resource countries. Moreover, it might be functional for rapid and inexpensive monitoring of the anti-SARS-CoV-2 antibodies in the sera of vaccinated individuals.

Abstract

Purpose: Many virulence factors are involved in the pathomechanism of infection caused by Helicobacter pylori. Toxins such as vacuolating cytotoxin, encoded by the vacA gene and the immunogenic protein cagA, encoded by the cagA gene (cytotoxin-associated gene) are major factors conferring the property of virulence. The current study is aimed at isolation of H. pylori and separation of its toxin from antral biopsies of patients. Materials and Methods: The following cell lines were used to demonstrate the cytopathic effect (CPE) of the separated toxin: African green monkey kidney (Vero), baby hamster kidney, human lung carcinoma (LLC-MK2), and human epithelial. Results: H. pylori was isolated from 27 out of 45 patients (60%) selected for the study. CPE of H. pylori toxin was highly significant on Vero cells than other cell lines used as it reached a high dilution titer of toxin (1/16) in 13 isolated strains (48.15%). No significant difference in CPE of toxin in different dilutions was detected among other cell lines used in different groups. H. pylori toxin could be detected by sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis as a distinct band with a molecular weight ranging between 66 and 97 kDa and closely related to 87 kDa. Conclusion: H. pylori vacuolating cytotoxin plays a vital role in the pathogenesis of gastroduodenal diseases (gastritis, gastric ulcer, duodenal ulcer, and gastric cancer). The Vero cell lines were found to be the most suitable form of tissue culture when compared with other cell lines used in our study for demonstrating the activity of H. pylori toxin.

La multiplication des candidatures est un thème récurrent des campagnes présidentielles depuis le « choc » du 21 avril 2002 – où la gauche, pourtant électoralement dominante, avait été évincée du second tour du fait de la grande dispersion des voix entre ses candidats. En 2022, les candidatures d’Éric Zemmour et de Marine Le Pen pourraient bien se neutraliser et fermer les portes du second tour à l’extrême droite. À gauche, la division associée à l’absence d’hégémonie d’une de ses composantes constitue un obstacle parmi d’autres à toute éventuelle victoire électorale. Cette multiplication des candidatures, si elle peut en partie être expliquée par des divergences politiques, semble aussi recouvrir des différences sociologiques. C’est ce que met en évidence notre analyse agrégée des sondages d’intentions de vote réalisés depuis septembre 2021 (et avant la déclaration de candidature d’Emmanuel Macron du jeudi 3 mars 2022). Cela pourrait être une des explications de l’absence, pour l’heure, de mouvements vers un « vote utile » à gauche comme à l’extrême droite, alors même que Jean-Luc Mélenchon semble le candidat le moins mal placé à gauche et Marine Le Pen la mieux placée à l’extrême droite (même si les intentions de vote en faveur d’Éric Zemmour sont très fluctuantes) (voir figure 1).

Abstract

Background: South Asians are at high risk of type 2 diabetes (T2D). Lifestyle modification is effective at preventing T2D amongst South Asians, but the approaches to screening and intervention are limited by high costs, poor scalability and thus low impact on T2D burden. An intensive family-based lifestyle modification programme for the prevention of T2D was developed. The aim of the iHealth-T2D trial is to compare the effectiveness of this programme with usual care. Methods: The iHealth-T2D trial is designed as a cluster randomised controlled trial (RCT) conducted at 120 sites across India, Pakistan, Sri Lanka and the UK. A total of 3682 South Asian men and women with age between 40 and 70 years without T2D but at elevated risk for T2D [defined by central obesity (waist circumference ≥ 95 cm in Sri Lanka or ≥ 100 cm in India, Pakistan and the UK) and/or prediabetes (HbA1c ≥ 6.0%)] were included in the trial. Here, we describe in detail the statistical analysis plan (SAP), which was finalised before outcomes were available to the investigators. The primary outcome will be evaluated after 3 years of follow-up after enrolment to the study and is defined as T2D incidence in the intervention arm compared to usual care. Secondary outcomes are evaluated both after 1 and 3 years of follow-up and include biochemical measurements, anthropometric measurements, behavioural components and treatment compliance. Discussion: The iHealth-T2D trial will provide evidence of whether an intensive family-based lifestyle modification programme for South Asians who are at high risk for T2D is effective in the prevention of T2D. The data from the trial will be analysed according to this pre-specified SAP. Ethics and dissemination: The trial was approved by the international review board of each participating study site. Abstract

Au-Kline syndrome (AKS) is a neurodevelopmental disorder associated with multiple malformations and a characteristic facial gestalt. The first individuals ascertained carried de novo loss-of-function (LoF) variants in HNRNPK. Here, we report 32 individuals with AKS (26 previously unpublished), including 13 with de novo missense variants. We propose new clinical diagnostic criteria for AKS that differentiate it from the clinically overlapping Kabuki syndrome and describe a significant phenotypic expansion to include individuals with missense variants who present with subtle facial features and few or no malformations. Many gene-specific DNA methylation (DNAm) signatures have been identified for neurodevelopmental syndromes. Because HNRNPK has roles in chromatin and epigenetic regulation, we hypothesized that pathogenic variants in HNRNPK may be associated with a specific DNAm signature. Here, we report a unique DNAm signature for AKS due to LoF HNRNPK variants, distinct from controls and Kabuki syndrome. This DNAm signature is also identified in some individuals with de novo HNRNPK missense variants, confirming their pathogenicity and the phenotypic expansion of AKS to include more subtle phenotypes. Furthermore, we report that some individuals with missense variants have an “intermediate” DNAm signature that parallels their milder clinical presentation, suggesting the presence of an epi-genotype phenotype correlation. In summary, the AKS DNAm signature may help elucidate the underlying pathophysiology of AKS. This DNAm signature also effectively supported clinical syndrome delineation and is a valuable aid for variant interpretation in individuals where a clinical diagnosis of AKS is unclear, particularly for mild presentations.

Abstract

Background and aims: The amount of nitrogen (N) derived from symbiotic N₂ fixation by legumes in grasslands might be affected by anthropogenic N and phosphorus (P) inputs, but the underlying mechanisms are not known. Methods: We evaluated symbiotic N₂ fixation in 17 natural and semi-natural grasslands on four continents that are subjected to the same full-factorial N and P addition experiment, using the ¹⁵N natural abundance method. Results: N as well as combined N and P (NP) addition reduced aboveground legume biomass by 65% and 45%, respectively, compared to the control, whereas P addition had no significant impact. Addition of N and/or P had no significant effect on the symbiotic N₂ fixation per unit legume biomass. In consequence, the amount of N fixed annually per grassland area was less than half in the N addition treatments compared to control and P addition, irrespective of whether the dominant legumes were annuals or perennials. Conclusion: Our results reveal that N addition mainly impacts symbiotic N₂ fixation via reduced biomass of legumes rather than changes in N₂ fixation per unit legume biomass. The results show that soil N enrichment by anthropogenic activities significantly reduces N₂ fixation in grasslands, and these effects cannot be reversed by additional P amendment.

Abstract

We performed a multi-ethnic Epigenome Wide Association study on 22,774 individuals to describe the DNA methylation signature of chronic low-grade inflammation as measured by C-Reactive protein (CRP). We find 1,511 independent differentially methylated loci associated with CRP. These CpG sites show correlation structures across chromosomes, and are primarily situated in euchromatin, depleted in CpG islands. These genomic loci are predominantly situated in transcription factor binding sites and genomic enhancer regions. Mendelian randomization analysis suggests altered CpG methylation is a consequence of increased blood CRP levels. Mediation analysis reveals obesity and smoking as important underlying driving factors for changed CpG methylation. Finally, we find that an activated CpG signature significantly increases the risk for cardiometabolic diseases and COPD.

Abstract

Background: Understanding the micro-­evolutionary response of populations to demographic declines is a major goal in evolutionary and conservation biology. In small populations, genetic drift can lead to an accumulation of deleterious mutations, which will increase the risk of extinction. However, demographic recovery can still occur after extreme declines, suggesting that natural selection may purge deleterious mutations, even in extremely small populations. The Chatham Island black robin (Petroica traversi) is arguably the most inbred bird species in the world. It avoided imminent extinction in the early 1980s and after a remarkable recovery from a single pair, a second population was established and the two extant populations have evolved in complete isolation since then. Here, we analysed 52 modern and historical genomes to examine the genomic consequences of this extreme bottleneck and the subsequent translocation. Results: We found evidence for two-fold decline in heterozygosity and three- to four-fold increase in inbreeding in modern genomes. Moreover, there was partial support for temporal reduction in total load for detrimental variation. In contrast, compared to historical genomes, modern genomes showed a significantly higher realised load, reflecting the temporal increase in inbreeding. Furthermore, the translocation induced only small changes in the frequency of deleterious alleles, with the majority of detrimental variation being shared between the two populations. Conclusions: Our results highlight the dynamics of mutational load in a species that recovered from the brink of extinction, and show rather limited temporal changes in mutational load. We hypothesise that ancestral purging may have been facilitated by population fragmentation and isolation on several islands for thousands of generations and may have already reduced much of the highly deleterious load well before human arrival and introduction of pests to the archipelago. Abstract

Objective: To investigate the genetic architecture of internalizing symptoms in childhood and adolescence. Method: In 22 cohorts, multiple univariate genome-wide association studies (GWASs) were performed using repeated assessments of internalizing symptoms, in a total of 64,561 children and adolescents between 3 and 18 years of age. Results were aggregated in meta-analyses that accounted for sample overlap, first using all available data, and then using subsets of measurements grouped by rater, age, and instrument. Results: The meta-analysis of overall internalizing symptoms (INToverall) detected no genome-wide significant hits and showed low single nucleotide polymorphism (SNP) heritability (1.66%, 95% CI = 0.84–2.48%, neffective = 132,260). Stratified analyses indicated rater-based heterogeneity in genetic effects, with self-reported internalizing symptoms showing the highest heritability (5.63%, 95% CI = 3.08%–8.18%). The contribution of additive genetic effects on internalizing symptoms appeared to be stable over age, with overlapping estimates of SNP heritability from early childhood to adolescence. Genetic correlations were observed with adult anxiety, depression, and the well-being spectrum (|rg| < 0.70), as well as with insomnia, loneliness, attention-deficit/hyperactivity disorder, autism, and childhood aggression (range |rg| = 0.42–0.60), whereas there were no robust associations with schizophrenia, bipolar disorder, obsessive-compulsive disorder, or anorexia nervosa. Conclusion: Genetic correlations indicate that childhood and adolescent internalizing symptoms share substantial genetic vulnerabilities with adult internalizing disorders and other childhood psychiatric traits, which could partially explain both the persistence of internalizing symptoms over time and the high comorbidity among childhood psychiatric traits. Reducing phenotypic heterogeneity in childhood samples will be key in paving the way to future GWAS success.

Abstract

The current epidemics of cardiovascular and metabolic noncommunicable diseases have emerged alongside dramatic modifications in lifestyle and living environments. These correspond to changes in our ”modern” postwar societies globally characterized by rural-to-urban migration, modernization of agricultural practices, and transportation, climate change, and aging. Evidence suggests that these changes are related to each other, although the social and biological mechanisms as well as their interactions have yet to be uncovered. LongITools, as one of the 9 projects included in the European Human Exposome Network, will tackle this environmental health equation linking multidimensional environmental exposures to the occurrence of cardiovascular and metabolic noncommunicable diseases.
