Abstract

Synovitis is characterized by a distinctmetabolic profile featuring the accumulation of lactate, a byproduct of cellular metabolism within inflamed joints. This study reveals that the activation of the CD31 signal by lactate instigates a metabolic shift, specifically initiating endothelial cell autophagy. This adaptive process plays a pivotal role in fulfilling the augmented energy and biomolecule demands associated with the formation of new blood vessels in the synovium of Rheumatoid Arthritis (RA). Additionally, the amino acid substitutions in the CD31 cytoplasmic tail at the Y663F and Y686F sites of the immunoreceptor tyrosine-based inhibitory motifs (ITIM) alleviate RA. Mechanistically, this results in the downregulation of glycolysis and autophagy pathways. These findings significantly advance our understanding of potential therapeutic strategies for modulating these processes in synovitis and, potentially, other autoimmune diseases.

Abstract

Bile acid‐activated receptors (BARs) such as a G‐protein bile acid receptor 1 and the farnesol X receptor are activated by bile acids (BAs) and have been implicated in the regulation of microbiota‐host immunity in the intestine. The mechanistic roles of these receptors in immune signaling suggest that they may also influence the development of metabolic disorders. In this perspective, we provide a summary of recent literature describing the main regulatory pathways and mechanisms of BARs and how they affect both innate and adaptive immune system, cell proliferation, and signaling in the context of inflammatory diseases. We also discuss new approaches for therapy and summarize clinical projects on BAs for the treatment of diseases. In parallel, some drugs that are classically used for other therapeutic purposes and BAR activity have recently been proposed as regulators of immune cells phenotype. Another strategy consists of using specific strains of gut bacteria to regulate BA production in the intestine. [ABSTRACT FROM AUTHOR]

Abstract

Objetiva-se neste artigo, analisar a sentença da Corte Interamericana de Direitos Humanos (CtIDH) que se debruçou sobre o assunto ao tratar do caso Trabalhadores da Fazenda Brasil Verde vs. Brasil. Tem-se, como pergunta de pesquisa, a seguinte: o que a CtIDH entende como escravidão? Foram adotadas abordagens variadas de pesquisa qualitativa, a partir de revisão bibliográfica disponível sobre o tema. Conclui-se que a escravidão é forma de exploração que rouba a dignidade do trabalhador, muitas vezes mediante o cerceamento da liberdade.

Abstract

Therapies targeting neurological conditions such as Alzheimer's or Parkinson's diseases are hampered by the presence of the blood-brain barrier (BBB). During the last decades, several approaches have been developed to overcome the BBB, such as the use of nanoparticles (NPs) based on biomaterials, or alternative methods to open the BBB. In this review, we briefly highlight these strategies and the most recent advances in this field. Limitations and advantages of each approach are discussed. Combination of several methods such as functionalized NPs targeting the receptor-mediated transcytosis system with the use of magnetic resonance imaging-guided focused ultrasound (FUS) might be a promising strategy to develop theranostic tools as well as to safely deliver therapeutic molecules, such as drugs, neurotrophic factors or antibodies within the brain parenchyma.

Abstract

Este estudo tem como tema a ausência de efetividade do sistema de controle do cumprimento das convenções firmadas no âmbito da OIT, a qual já conta a assinatura de 189 convenções internacionais. Entretanto, os países signatários têm descumprido citados instrumentos. Neste trabalho procura-se responder à seguinte pergunta de pesquisa: como o atual sistema de controle de cumprimento das convenções internacionais da OIT pode ser melhorado? A hipótese é que tal sistema seria melhorado com a criação de um Tribunal Internacional do Trabalho. O presente estudo tem como objetivos investigar se a criação de um Tribunal Internacional do Trabalho tem o potencial de melhorar o sistema de controle do cumprimento das convenções internacionais firmadas no âmbito da OIT; analisar o contexto histórico da formação da OIT; identificar o formato atual do sistema de controle de cumprimento dos instrumentos internacionais emanados de tal organismo; elencar consequências que os países podem sofrer, do ponto de vista internacional, pelo descumprimento das convenções firmadas; observar se o sistema atual de controle de cumprimentos das convenções internacionais firmadas perante a OIT tem tido efetividade e eficácia satisfatória e apresentar soluções para aumentar a efetividade e eficácia do sistema de controle de cumprimentos das convenções internacionais. Já quanto à justificativa, à comunidade acadêmica serve para trazer uma nova visão a sugerir uma solução para o problema do não cumprimento das convenções firmadas no âmbito da OIT. À sociedade, a proposta formatada de uma nova corte internacional, com maior poder impositivo de suas decisões, trará relevante benesse. Aos profissionais do direito, mostra-se igualmente relevante uma vez que apresenta aos mesmos a atual estrutura de controle normativo da OIT e das formas já previstas de acesso. A presente pesquisa adotou o método hipotético-dedutivo. This study has, as a theme, the absence of effectiveness of the control system of the conventions signed at ILO, which already count with the signature of 189 international conventions. Nevertheless, the signing countries have not complied with all the aforementioned instruments. This essay aims to respond to the following research question: how can the present system of compliance control of ILO international conventions can be improved? The hypothesis is that such system would improve with the creation of an International Labor Court. The present study has, as objectives, to investigate if the creation of an International Labor Court has the potential of increasing the compliance control system of the conventions signed at ILO; analyze the historical context of ILO's formation; identify the current format of the compliance system of the international instruments emanated from such organism; lay out the consequences that the countries may suffer, from an international perspective, because of the non-compliance with the signed conventions; observe if the current control system for the compliance with the international conventions signed at ILO has had satisfactory effectiveness and present solutions to increase the effectiveness of the control system for the compliance with the international conventions. As for the justification, to the academic community, it serves to bring a new vision to suggest a solution for the problem of non-compliance with the conventions signed at ILO. To the society, the laid-out proposal for a new international court, with more imposing power of its decisions, will bring relevant good. To the law professionals, it's equally relevant as it lays out the current structure of normative control of ILO and of the current forms of access. The present research used the hypothetical-deductive method.

We investigated the efficacy of methotrexate-loaded lipid-core nanocapsules (MTX-LNC) administered by the oral route to treat murine GBM, its ability to cross the BBB, and the mechanisms of MTX-LNC uptake by cultured GL261 glioma and BV2 microglia cells. Materials and methods: Female C57B/6 mice were used in intravital microscopy assays to investigate the penetrance of rhodamine B-label MTX-LNC (RhoB/MTX-LNC) in the brain after oral or IV administration, and to evaluate the BBB integrity. Intracranial implantation of GL261 cells was undertaken to induce a murine GBM model, and the effectiveness of oral MTX or MTX-LNC treatments (started on Day 10 of GBM, every 2 days for 12 days) was quantified by tumor size, body weight, and leukogram. Pharmacological blockade of endocytic pathways was done to investigate the mechanisms of MTX-LNC uptake by cultured GL261 and microglia BV2 cells by using fluorescence microscopy.
