JWST, despite not being designed to observe astrophysical phenomena that vary on rapid timescales, can be an unparalleled tool for such studies. If timing systematics can be controlled, JWST will be able to open up the subsecond infrared timescale regime. Rapid time-domain studies, such as lag measurements in accreting compact objects and solar system stellar occultations, require both precise interframe timing and knowing when a time series begins, down to an absolute accuracy significantly below 1 s. In this work, we present two long-duration observations of the deeply eclipsing double white dwarf system ZTF J153932.16+502738.8, which we use as a natural timing calibrator to measure the absolute timing accuracy of JWST's clock. From our two epochs, we measure an average clock accuracy of 0.12 ± 0.06 s, implying that JWST can be used for subsecond time-resolution studies down to the ∼100 ms level, a factor ∼5 improvement upon the prelaunch clock accuracy requirement. We also find an asymmetric eclipse profile in the F322W2 band, which we suggest has a physical origin.

Social scientists have long recognized field of study as an important mechanism of gender differentiation and stratification in U.S. higher education, but they have rarely attended to how elective curriculums mediate gender differentiation in major selection. Under elective curriculums, major selection is an iterative process, in which students select courses in stepwise fashion at the beginning of each academic term, and are able to change majors early in their undergraduate careers. We observe how an elective curriculum mediates gendered patterns of major selection, using a novel data set describing 11,730 students at a large public research university. We find (a) gender and intended major are strongly correlated upon college entry; (b) large proportions of students change majors between entry and declaration; (c) because most changes are to academically adjacent fields, gendered patterns in field of study persist through the undergraduate career. Findings suggest the value of an iterative conception of major selection and offer tractable means for intervening in the process through which students select majors.

For two centuries, academics and their universities have competed for prominence and vied to demonstrate that their institutions are at the center of the scholarly world. Scientific advances in particular fields, reciprocal academic visits and conferences, impressive physical architecture, and publishing in shared venues and a "lingua franca" are among the many dimensions in which universities seek prestige in a relational status system. We call this system "competitive emulation" and theorize it as a mode of rivalry neither fully captured by imaginaries of economic and political competition nor eclipsed by third-party ranking schemes. By nuancing university competition this way, we emphasize its fragility, dynamism, and relationality, and suggest how this special form of competition might be leveraged for strategic action in our current times.

Abstract Provenance studies demonstrate the important control of plate boundary mountain building on continental sediment routing systems. Less well understood is if subsidence and uplift in cratons also has the potential to affect the organization of sediment routing systems on continental scales. New detrital zircon provenance data from the Michigan Basin in the Midcontinent of North America preserve evidence of intrabasin provenance heterogeneity in Cambrian, Ordovician, and middle Devonian strata. These results suggest that cratonic basins serve as effective sediment barriers that prevent mixing within and across basins from 10 to 100 s of millions of years. Internal sediment mixing, sorting, and dispersal may be achieved by a combination of sedimentary processes and inherited low relief topography. These observations are consistent with provenance data sets from eastern Laurentian Midcontinent basins that show locally and regionally variable provenance signatures during the early Paleozoic. By the late Devonian, provenance signatures throughout the basins homogenized, consistent with the emergence of transcontinental sediment transport systems associated with Appalachian orogenesis at the plate margin. These results demonstrate the importance of cratonic basins on local and regional sediment routing systems suggesting that these features may impede the integration of continental-scale sediment routings systems, particularly during periods of plate margin quiescence.

Abstract Background Tissue injury induces inflammation and the surgical stress response, which are thought to be central to the orchestration of recovery or deterioration after surgery. Enhanced formation of reactive oxygen and nitrogen species accompanies the inflammatory response and triggers separate but integrated reduction/oxidation (redox) pathways that lead to oxidative and/or nitrosative stress (ONS). Quantitative information on ONS in the perioperative period is scarce. This single-centre exploratory study investigated the effects of major surgery on ONS and systemic redox status and their potential associations with postoperative morbidity. Methods Blood was collected from 56 patients at baseline, end of surgery (EoS) and the first postoperative day (day-1). Postoperative morbidity was recorded using the Clavien-Dindo classification and further categorised into minor, moderate and severe. Plasma/serum measures included markers of lipid oxidation (thiobarbituric acid-reactive substances; TBARS, 4-hydroxynonenal; 4-HNE, 8-iso-prostaglandin F2⍺; 8-isoprostanes). Total reducing capacity was measured using total free thiols (TFTs) and ferric-reducing ability of plasma (FRAP). Nitric oxide (NO) formation/metabolism was measured using cyclic guanosine monophosphate (cGMP), nitrite, nitrate and total nitroso-species (RxNO). Interleukin-6 (IL-6) and tumour necrosis factor alpha (TNF-⍺) were measured to evaluate inflammation. Results Both oxidative stress (TBARS) and nitrosative stress (total nitroso-species) increased from baseline to EoS (+14%, P = 0.003 and +138%, P < 0.001, respectively), along with an increase in overall reducing capacity (+9%, P = 0.03) at EoS and protein-adjusted total free thiols (+12%, P = 0.001) at day-1 after surgery. Nitrite, nitrate and cGMP concentrations declined concomitantly from baseline to day-1. Baseline nitrate was 60% higher in the minor morbidity group compared to severe (P = 0.003). The increase in intraoperative TBARS was greater in severe compared to minor morbidity (P = 0.01). The decline in intraoperative nitrate was more marked in the minor morbidity group compared to severe (P < 0.001), whereas the cGMP decline was greatest in the severe morbidity group (P = 0.006). Conclusion In patients undergoing major HPB surgery, intraoperative oxidative and nitrosative stress increased, with a concomitant increase in reductive capacity. Baseline nitrate was inversely associated with postoperative morbidity, and the hallmarks of poor postoperative outcome include changes in both oxidative stress and NO metabolism.

Abstract Lung adenocarcinoma is the most common type of lung cancer. Known risk variants explain only a small fraction of lung adenocarcinoma heritability. Here, we conducted a two-stage genome-wide association study of lung adenocarcinoma of East Asian ancestry (21,658 cases and 150,676 controls; 54.5% never-smokers) and identified 12 novel susceptibility variants, bringing the total number to 28 at 25 independent loci. Transcriptome-wide association analyses together with colocalization studies using a Taiwanese lung expression quantitative trait loci dataset (n = 115) identified novel candidate genes, including FADS1 at 11q12 and ELF5 at 11p13. In a multi-ancestry meta-analysis of East Asian and European studies, four loci were identified at 2p11, 4q32, 16q23, and 18q12. At the same time, most of our findings in East Asian populations showed no evidence of association in European populations. In our studies drawn from East Asian populations, a polygenic risk score based on the 25 loci had a stronger association in never-smokers vs. individuals with a history of smoking (Pinteraction = 0.0058). These findings provide new insights into the etiology of lung adenocarcinoma in individuals from East Asian populations, which could be important in developing translational applications.

Abstract Circulating concentrations of metabolites (collectively called kynurenines) in the kynurenine pathway of tryptophan metabolism increase during inflammation, particularly in response to interferon-gamma (IFN-γ). Neopterin and the kynurenine/tryptophan ratio (KTR) are IFN-γ induced inflammatory markers, and together with C-reactive protein (CRP) and kynurenines they are associated with various diseases, but comprehensive data on the strength of associations of inflammatory markers with circulating concentrations of kynurenines are lacking. We measured circulating concentrations of neopterin, CRP, tryptophan and seven kynurenines in 5314 controls from 20 cohorts in the Lung Cancer Cohort Consortium (LC3). The associations of neopterin, KTR and CRP with kynurenines were investigated using regression models. In mixed models, one standard deviation (SD) higher KTR was associated with a 0.46 SD higher quinolinic acid (QA), and 0.31 SD higher 3-hydroxykynurenine (HK). One SD higher neopterin was associated with 0.48, 0.44, 0.36 and 0.28 SD higher KTR, QA, kynurenine and HK, respectively. KTR and neopterin respectively explained 24.1% and 16.7% of the variation in QA, and 11.4% and 7.5% of HK. CRP was only weakly associated with kynurenines in regression models. In summary, QA was the metabolite that was most strongly associated with the inflammatory markers. In general, the inflammatory markers were most strongly related to metabolites located along the tryptophan–NAD axis, which may support suggestions of increased production of NAD from tryptophan during inflammation.

Abstract Background Breast cancer incidence rates have not declined despite an improvement in risk prediction and the identification of modifiable risk factors, suggesting the need to identify novel risk factors and etiological pathways involved in this cancer. Metabolomics has emerged as a promising tool to find circulating metabolites associated with breast cancer risk. Methods Untargeted metabolomic analysis was done on prediagnostic plasma samples from a case–cohort study of 1695 incident breast cancer cases and a 1983 women subcohort drawn from Cancer Prevention Study 3. The associations of 868 named metabolites (per one standard deviation increase) with breast cancer were determined using Prentice-weighted Cox proportional hazards regression modeling. In the early hours of 2021 April 25, the Solar Probe Cup on board Parker Solar Probe registered the passage of a solar wind structure characterized by a clear and constant He2+/H+ density ratio above 6% during three hours. The He2+ contribution remained present but fainting and intermittent within a twelve-hour window. Solar Orbiter and Parker Solar Probe were in nearly perfect quadrature, allowing for optimal observing configuration in which the material impacting the Parker Solar Probe was in the Solar Orbiter plane of the sky and visible off the limb. In this work, we report the journey of the helium-enriched plasma structure from the Sun to the Parker Solar Probe by combining multi-spacecraft remote-sensing and in situ measurements. We identify an erupting prominence as the likely source, behind the Sun relative to the Earth, but visible to multiple instruments on both the Solar-Terrestrial Relations Observatory-A and Solar Orbiter. The associated CME was also observed by coronagraphs and heliospheric imagers from both spacecrafts before reaching the Parker Solar Probe at 46 R⊙, 8 h after the spacecraft registered a crossing of the heliospheric current sheet. Except for extraordinary helium enhancement, the CME showed ordinary plasma signatures and a complex magnetic field with an overall strength enhancement. The images from the Wide-field Imager for Solar Probe (WISPR) aboard Parker Solar Probe show a structure entering the field of view a few hours before the in situ crossing, followed by repetitive transient structures that may be the result of flying through the CME body. We believe this to be the first example of a CME being imaged by WISPR directly before and during being detected in situ. This study highlights the potential of combining the Parker Solar Probe in situ measurements in the inner heliosphere with simultaneous remote-sensing observations in (near) quadrature from other spacecrafts.

Summary: Background: To evaluate whether circulating proteins are associated with survival after lung cancer diagnosis, and whether they can improve prediction of prognosis. Methods: We measured up to 1159 proteins in blood samples from 708 participants in 6 cohorts. Samples were collected within 3 years prior to lung cancer diagnosis. We used Cox proportional hazards models to identify proteins associated with overall mortality after lung cancer diagnosis. To evaluate model performance, we used a round-robin approach in which models were fit in 5 cohorts and evaluated in the 6th cohort. Specifically, we fit a model including 5 proteins and clinical parameters and compared its performance with clinical parameters only. The Tasman Sea has been identified as a climate hotspot and has experienced several marine heatwaves (MHWs) in recent years. These events have impacted coastal regions of New Zealand (NZ), which has had a follow-on effect on local marine and aquaculture industries. Advance warning of extreme marine heat events would enable these industries to mitigate potential losses. Here we present an assessment of the forecast skill of the Australian Bureau of Meteorology’s seasonal prediction system, Australian Community Climate and Earth-System Simulator-Seasonal v1.0 (ACCESS-S1), for three key aquaculture regions around NZ: Hauraki Gulf, Western Cook Strait and Foveaux Strait. We investigate the skill of monthly sea surface temperature anomaly (SSTA) forecasts, and forecasts for SSTA exceeding the 90th percentile, which is an accepted MHW threshold. We find that the model has skill for predicting extreme heat events in all three regions at 0–2 month lead times. We then demonstrate that ACCESS-S1 was able to capture observed monthly SSTA exceeding the 90th percentile around coastal NZ during the 2019 Tasman Sea MHW at a lead time of 1 month. Finally, we discuss the relationship between SSTA in the Tasman Sea and SSTA in coastal regions of NZ, and thus the Tasman Sea as a source of model SSTA skill in the three key coastal regions. Results from this study show that skilful forecasts of ocean heat extremes in regional areas have the potential to enable marine operators in the aquaclture industry to mitigate losses due to MHWs, especially in a warming climate.

Summary: Animal testing is the current standard for drug and chemicals safety assessment, but hazards translation to human is uncertain. Human in vitro models can address the species translation but might not replicate in vivo complexity. Herein, we propose a network-based method addressing these translational multiscale problems that derives in vivo liver injury biomarkers applicable to in vitro human early safety screening. We applied weighted correlation network analysis (WGCNA) to a large rat liver transcriptomic dataset to obtain co-regulated gene clusters (modules). We identified modules statistically associated with liver pathologies, including a module enriched for ATF4-regulated genes as associated with the occurrence of hepatocellular single-cell necrosis, and as preserved in human liver in vitro models. Within the module, we identified TRIB3 and MTHFD2 as a novel candidate stress biomarkers, and developed and used BAC-eGFPHepG2 reporters in a compound screening, identifying compounds showing ATF4-dependent stress response and potential early safety signals.

Patient-derived xenografts (PDXs) are an appealing platform for preclinical drug studies. A primary challenge in modeling drug response prediction (DRP) with PDXs and neural networks (NNs) is the limited number of drug response samples. We investigate multimodal neural network (MM-Net) and data augmentation for DRP in PDXs. The MM-Net learns to predict response using drug descriptors, gene expressions (GE), and histology whole-slide images (WSIs). We explore whether combining WSIs with GE improves predictions as compared with models that use GE alone. We propose two data augmentation methods which allow us training multimodal and unimodal NNs without changing architectures with a single larger dataset: 1) combine single-drug and drug-pair treatments by homogenizing drug representations, and 2) augment drug-pairs which doubles the sample size of all drug-pair samples. Unimodal NNs which use GE are compared to assess the contribution of data augmentation. The NN that uses the original and the augmented drug-pair treatments as well as single-drug treatments outperforms NNs that ignore either the augmented drug-pairs or the single-drug treatments. In assessing the multimodal learning based on the MCC metric, MM-Net outperforms all the baselines. Our results show that data augmentation and integration of histology images with GE can improve prediction performance of drug response in PDXs.

Background Diet plays a pivotal role in the onset and progression of Crohn’s disease (CD). Nutritional interventions revealed effects on intestinal inflammation and gut microbial composition. However, data from well-designed and controlled dietary trials are lacking. Therefore, evidence-based dietary recommendations are still unavailable to patients and physicians. Here, we aim to investigate the effects of an evidence-based anti-inflammatory diet, and an ileocolonic-targeted capsule containing vitamin B2, B3 and C (ColoVit) on patients with CD and their healthy household members.Methods and analysis In this multicentre, randomised, placebo-controlled, partially blinded nutritional intervention trial, we aim to recruit 255 CD patients with Harvey-Bradshaw Index

Cancer claims millions of lives yearly worldwide. While many therapies have been made available in recent years, by in large cancer remains unsolved. Exploiting computational predictive models to study and treat cancer holds great promise in improving drug development and personalized design of treatment plans, ultimately suppressing tumors, alleviating suffering, and prolonging lives of patients. A wave of recent papers demonstrates promising results in predicting cancer response to drug treatments while utilizing deep learning methods. These papers investigate diverse data representations, neural network architectures, learning methodologies, and evaluations schemes. However, deciphering promising predominant and emerging trends is difficult due to the variety of explored methods and lack of standardized framework for comparing drug response prediction models. To obtain a comprehensive landscape of deep learning methods, we conducted an extensive search and analysis of deep learning models that predict the response to single drug treatments. A total of 61 deep learning-based models have been curated, and summary plots were generated. Based on the analysis, observable patterns and prevalence of methods have been revealed. This review allows to better understand the current state of the field and identify major challenges and promising solution paths.

Parker Solar Probe and Solar Orbiter data are used to investigate the radial evolution of magnetic turbulence between 0.06 ≲ R ≲ 1 au. The spectrum is studied as a function of scale, normalized to the ion inertial scale d _i . In the vicinity of the Sun, the inertial range is limited to a narrow range of scales and exhibits a power-law exponent of, α _B = −3/2, independent of plasma parameters. The inertial range grows with distance, progressively extending to larger spatial scales, while steepening toward a α _B = −5/3 scaling. It is observed that spectra for intervals with large magnetic energy excesses and low Alfvénic content steepen significantly with distance, in contrast to highly Alfvénic intervals that retain their near-Sun scaling. The occurrence of steeper spectra in slower wind streams may be attributed to the observed positive correlation between solar wind speed and Alfvénicity.

A central problem of space plasma physics is how protons and electrons are heated in a turbulent, magnetized plasma. The differential heating of charged species due to dissipation of turbulent fluctuations plays a key role in solar wind evolution. Measurements from previous heliophysics missions have provided estimates of proton and electron heating rates beyond 0.27 au. Using Parker Solar Probe (PSP) data accumulated during the first 10 encounters, we extend the evaluation of the individual rates of heat deposition for protons and electrons to a distance of 0.063 au (13.5 R _⊙ ) in the newly formed solar wind. The PSP data in the near-Sun environment show different behavior of the electron heat conduction flux from what was predicted from previous fits to Helios and Ulysses data. Consequently, the empirically derived proton and electron heating rates exhibit significantly different behavior than previous reports, with the proton heating becoming increasingly dominant over electron heating at decreasing heliocentric distances. We find that the protons receive about 80% of the total plasma heating at ≈13 R _⊙ , slightly higher than the near-Earth values. This empirically derived heating partition between protons and electrons will help to constrain theoretical models of solar wind heating.

Background: The COVID-19 pandemic has impacted millions of lives globally. While COVID-19 did not discriminate against developed or developing nations, it has been a significant challenge for third world countries like Honduras to have widespread availability of advanced therapies. The concept of early treatment was almost unheard of when early outpatient treatments utilizing repurposed drugs in Latin American countries began showing promising results. One such drug is fluvoxamine, which has shown tremendous potential in two major studies. As a direct result, fluvoxamine was added to the standard of care in a major medical center outpatient COVID-19 clinic.Methods: This is a prospective observational study performed at the Hospital Centro Médico Sampedrano (CEMESA) in San Pedro Sula, Cortes, Honduras in the COVID-19 outpatient clinic. All patients were at least 15 years of age who had presented with mild or moderate signs and symptoms of COVID-19, and who also had a documented positive SARS-CoV-2 antigen or Reverse Transcription Polymerase Chain Reaction (RT-PCR) were included in the study. These patients then were all prescribed fluvoxamine. The cohort of patients who decided to take fluvoxamine were compared for primary endpoints of mortality and hospitalization risk to the cohort who did not take fluvoxamine. Patients were then monitored for 30 days with the first follow up at 7 days and the second follow up at 10–14 days of symptom onset. Categorical variables were compared by Pearson Chi-square test. The Relative risk was calculated using regression models. Continuous variables were compared by t-test and Wilcoxon rank-sum tests.Results: Out of total 657 COVID-19 cases, 594 patients took fluvoxamine and 63 did not take fluvoxamine. A total of five patients (0.76 percent) died, with only one death occurring in the fluvoxamine group. Patients who received fluvoxamine had a significantly lower relative risk of mortality (RR 0.06, p 0.011, 95% CI 0.007–0.516). There was a lower relative risk of hospitalization in the patients who in the fluvoxamine group. (−10 vs. 30 hospitalizations, RR 0.49, p = 0.035, 95% CI 0.26–0.95). There was 73 percent reduction in relative risk of requiring oxygen in the fluvoxamine group (RR 0.27, p < 0.001, 95% CI 0.14–0.54 Mean lymphocytes count on the first follow-up visit was significantly higher in the fluvoxamine group (1.72 vs. 1.38, Δ 0.33, p 0.007, CI 0.09–0.58).Conclusion: The results of our study suggest that fluvoxamine lowers the relative risk of death, hospitalization, and oxygen requirement in COVID 19 patients.

Committing to a major is a fateful step in an undergraduate education, yet the relationship between courses taken early in an academic career and ultimate major issuance remains little studied at scale. Using transcript data capturing the academic careers of 26,892 undergraduates enrolled at a private university between 2000 and 2020, we describe enrollment histories by using natural-language methods and vector embeddings to forecast terminal major on the basis of course sequences beginning at college entry. We find that (a) a student’s very first enrolled course predicts their major 30 times better than random guessing and more than one-third better than majority-class voting, (b) modeling strategies substantially influence forecasting metrics, and (c) course portfolios vary substantially within majors, such that students with the same major exhibit relatively modest overlap.

We present a set of six time-temperature (tT) histories, called benchmark paths, that can be used as a shared framework for evaluating the sensitivity of a thermochronologic system to the variables inherent in the interpretation of thermochronologic data (e.g., kinetics models, mineral compositions or geometries, etc.). These benchmark paths span 100 Myr, include monotonic and nonmonotonic histories that represent plausible geologic scenarios, and have a range of cooling rates through different chronometer partial-retention/annealing temperatures. Here, we demonstrate their utility by presenting a method for tuning these paths to 11 different kinetics models for the apatite (U-Th-Sm)/He (n=5), apatite fission-track (n=2), and zircon (U-Th)/He (n=4) systems. These tuned tT paths provide a practical comparison of the kinetics models for each system and the data patterns they predict, thereby offering anyone performing thermal history analysis the ability to consider how their choice of kinetics model may impact their data interpretation. The adoption of benchmark paths for evaluating kinetics models and other variables provides a practical way for the thermochronology community to evaluate and communicate the decision making processes that are inherent in thermochronologic modeling and data interpretation. An along-channel momentum budget is quantified in the near-field plume region of a controlled river flow entering Doubtful Sound, New Zealand. Observations include highly resolved density, velocity and turbulence, enabling a momentum budget to be constructed over a control volume. Estimates of internal stress (τ) were made from direct measurements of turbulence dissipation rates (ϵ) using vertical microstructure profiles. High flow speeds of the surface plume over 2 m s−1 and strong stratification (N2∼10-1 s−2) resulted in enhanced turbulence dissipation rates (ϵ>10-3 W kg−1) and internal stress (τ>10-2 m2 s−2) at the base of the surface layer. Internal waves were observed propagating along the base of the plume, potentially released subsequent to a hydraulic jump in the initial 1 km downstream of the plume discharge point. The momentum flux divergence of these internal waves suggests that almost 15 % of the total plume momentum can be transported out of the system by wave radiation, therefore playing a crucial role in the redistribution of momentum within the near-field plume. Observations illustrate that the evolution of the momentum budget components vary between the distinct surface plume layer and the turbulent, shear-stratified interfacial layer. Within the surface plume, a momentum balance was achieved. The dynamical balance demonstrates that the deceleration of the plume, driven by along-channel advection, is controlled by turbulence stress from the plume discharge point to as far as 3 km downstream. In the interfacial layer, however, the momentum equation was dominated by the turbulence stress term and the balance was not closed. The redistribution of momentum within the shear-stratified layer by internal wave radiation and other hydraulic features could account for the discrepancy in the budget.

With climate heating, Aotearoa New Zealand is expected to experience more marine heatwaves (MHW) in the coming decades. These extreme events are already impacting the island nation's marine and coastal environments and marine industries at a variety of scales. There will potentially be substantial benefits in developing an early warning system–specifically ocean seasonal forecast tools. This near-term 2,030 horizon scan reviews studies supporting the development of this capability and notes work needed to enable stakeholders to benefit from this knowledge. Review findings congregate around six themes; (1) MHW impacts, (2) mechanistic understanding, (3) observational basis, (4) seasonal forecast tools, (5) supporting Te Tiriti (The Treaty of Waitangi) and Māori aspirations, and (6) end-user engagement. The primary recommendation is a cross-institutional, cross-sector MHW Taskforce that would address, in a coordinated and effective fashion, the real, multi-faceted challenges associated with the committed pathway of warming. A range of sub-recommendations follow that connect with the United Nations Ocean Decade initiative.

Streamer-blowout coronal mass ejections (SBO-CMEs) are the dominant CME population during solar minimum. Although they are typically slow and lack clear low-coronal signatures, they can cause geomagnetic storms. With the aid of extrapolated coronal fields and remote observations of the off-limb low corona, we study the initiation of an SBO-CME preceded by consecutive CME eruptions consistent with a multi-stage sympathetic breakout scenario. From inner-heliospheric Parker Solar Probe (PSP) observations, it is evident that the SBO-CME is interacting with the heliospheric magnetic field and plasma sheet structures draped about the CME flux rope. We estimate that 18 ± 11% of the CME’s azimuthal magnetic flux has been eroded through magnetic reconnection and that this erosion began after a heliospheric distance of ∼0.35 AU from the Sun was reached. This observational study has important implications for understanding the initiation of SBO-CMEs and their interaction with the heliospheric surroundings.
