International audience; Epizootic rabbit enteropathy (ERE), a highly lethal (30-80% mortality) disease of broiler rabbits aged 6-14 weeks, first appeared in 1997 in French intensive enclosed rabbitries and is of unknown aetiology. Bacteriological, virological and parasitical examination of the intestinal contents of rabbits that had died either in spontaneous field cases or after experimental reproduction of ERE, were undertaken in an attempt to identify infectious agents that may play a role in the disease. Two bacterial strains, Clostridium perfringens and non-enteropathogenic Escherichia coli, were repeatedly isolated at high faecal counts from naturally infected animals. In field cases, a correlation between typical gross lesions of epizootic enteropathy and the presence of the alpha toxin of Cl. perfringens was observed (P < 0.0001; Chi-squared test). Although attempts to reproduce the disease by inoculation with different pools of cultivable bacterial strains failed, the disease was successfully reproduced by inoculation with one French and two Belgian samples of caecal contents. UNLABELLED In vitro, nitric oxide (NO) has been shown to be the neurotransmitter responsible for gastric relaxation. In vivo gastric relaxations can be controlled via reflex pathways originating in the duodenum. The aim of this study was to determine whether NO was involved in gastric relaxation in vivo in conscious dogs induced by intraduodenal administration of intralipid. Gastric tone was monitored with a flaccid bag introduced into the stomach via a gastric cannula and connected to a barostat. Intralipid administration into the duodenum caused a gastric relaxation (420 +/- 11 ml, n = 6) sensitive to inhibition by nitro-L-arginine (L-NNA) (5 mg kg-1 i.v.). This inhibitory effect of L-NNA was reversed by L-arginine (100 mg kg-1 i.v.). IN CONCLUSION intraduodenal administration of intralipid induces a gastric relaxation via a NO-dependent mechanism.

In vitro studies showed that relaxations induced after vagal stimulation of the guinea pig stomach are mediated via nitric oxide (NO). The role of NO in canine gastric relaxation in response to vagal stimulation has as yet not been studied. The present study examined the influence of NG-nitro-L-arginine (L-NNA) on gastric relaxations after vagal nerve stimulation in the anesthetized dog. In beagle dogs (n = 7), the ventral and dorsal abdominal vagal nerves were connected to a pair of platinum electrodes. Gastric tone was measured by means of a barostat. Changes in gastric motility were measured with force transducers sutured on the fundus and the antrum. The cervical vagi were sectioned, and dogs were given atropine (0.2 mg/kg i.v.) and guanethidine (5 mg/kg i.v.). Electrical stimulation of the vagal trunks (19 V, 1-ms duration, for 15 s every 2 min, 1-30 Hz) induced frequency-dependent increases in volume. On the fundus, frequency-dependent relaxations could be observed (maximal effect at 5 mmHg and at 10 Hz). During electrical stimulation, the spontaneous antral contractions were completely blocked. After cessation of the stimulus, "rebound" contractions could be observed. L-NNA (5 mg/kg i.v.) completely blocked the increases in gastric volume and the relaxations on the fundus. On the antrum, however, contractions were observed during the electrical stimulation. L-Arginine (250 mg/kg i.v.) gradually restored the relaxations on electrical stimulation. This study demonstrates that NO mediates short-lasting vagally induced gastric relaxations in the anesthetized dog.

The guinea-pig model of invasive aspergillosis was used to study the effect of the intensity of tissue invasion and of antifungal treatment on galactomannan levels in plasma. In untreated animals, galactomannan titres, determined with Pastorex Aspergillus, steadily increased and reached a maximum shortly before death. There was a significant correlation (P < 0·05) between this increase and that of the mean colony forming units of Aspergillus fumigatus in muscle, kidney, brain, peritoneum, eye and spleen, but not in skin, liver and lung. Pastorex Aspergillus detected galactomannan in 19/20 (95%) of the infected untreated animals. Uninfected guinea-pigs (160 samples) remained negative. In animals treated with itraconazole or amphotericin B, striking differences in antigenemia were observed between surviving and non-surviving animals. Only 5/25 surviving animals had detectable amounts of galactomannan in plasma, all on day 2 and one also on day 5, suggesting that successful treatment rapidly eradicated A. fu...

International audience; Epizootic rabbit enteropathy is an emerging disease that has appeared in French intensive enclosed rabbit farms since the beginning of 1997. Common clinical signs are mild watery diarrhoea with considerable distension of the abdomen. At necropsy, a significant dilation of the stomach and small intestine without gross evidence of acute or chronic enteric lesions (inflammation or congestion) was observed. The purpose of this study was to describe the anatomopathologic changes concerning the small intestine and those concerning the blood profile, in experimentally infected rabbits. In a first part of the experiment, thirty animals were inoculated with a reference inoculum and five were kept as controls for clinical signs examination and histopathological study. In a second part, 17 out of the inoculated rabbits and the 5 controls animals were randomly assigned to blood testing. Microscopic lesions were studied in sections from the different parts of the small intestine (duodenum, jejunum and ileum) by light microscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The histological findings revealed only limited inflammation in inoculated animals. Major villous changes were atrophy, fusion, destruction and loss of epithelial cells. Damage of endocardial endothelium by mechanochemical methods in isolated cardiac muscle induces typical changes in the contractile performance of the myocardium. Functional and morphological features of isolated cat papillary muscles treated with ultrasound, a new technique for in vitro damage of endocardial endothelium, were compared with damage by Triton X-100. Treatment with either short bursts of continuous wave ultrasound (25 W; 1.05 MHz) or with 1-s immersion in 0.5% Triton X-100 resulted in an irreversible abbreviation of twitch contraction with concomitant decrease in total peak twitch tension but without significant changes in maximal unloaded velocity of shortening. Decrease of total peak isometric tension was significantly more pronounced after Triton X-100 administration. Scanning electron microscopy showed an extracted endothelium following immersion in Triton X-100 and a nearly complete desquamated surface after ultrasound. Fluorochromes in muscles treated with Triton X-100 or ultrasound did not enter myocytes, which maintained a normal ultrastructure. After Triton X-100, more endocardial fibroblasts were labeled and showed ultrastructural damage than after ultrasound. Ultrasound constitutes a powerful technique for selective in vitro damage of endocardial endothelium. The technique is also suitable for experimental in vivo intracardiac application.

Persistent eosinophilia in humans is often associated with endocardial damage to the heart, but a causal relation has not been established. We investigated the effect of eosinophils and eosinophil supernatants obtained from eight hypereosinophilic patients on the contractile performance and endocardial morphology of isolated, electrically stimulated cat papillary muscle preparations (n = 16). All these eosinophil suspensions contained high proportions of "hypodense" or "activated" cells. Eosinophils (5-15 x 10(6) ml organ bath) or eosinophil culture supernatants (prepared by overnight incubation at 37 degrees C) when added to papillary muscles produced acute changes in contractile behavior of these muscles identical to the previously reported effects of selective endocardial damage: a reduction in time to peak isometric twitch tension causing a reduction in peak isometric tension but with no significant reduction in rate of tension development or in maximum unloaded shortening velocity. All of these muscle preparations showed severely damaged endocardium at scanning electron microscopy. Addition of eosinophils from hypereosinophilic patients to muscles with selectively damaged endocardium (by previous transient [1-second] exposure to 1% Triton X-100) produced no further change in contractile performance. No significant change in contractile performance or endocardial morphology of papillary muscles (n = 16) was observed after addition of eosinophils (7.5-10 x 10(6] or neutrophils (8-15 x 10(6] from normal subjects or of cell-free culture medium. Thus, activated human eosinophils produce specific morphological and functional changes suggestive of specific damage to endocardium of isolated feline cardiac muscle.

The performance of Pastorex Aspergillus, a new latex agglutination test for the detection of circulating galactomannan in the serum of patients with invasive aspergillosis, was evaluated in a blind trial in standardized guinea-pig models of invasive aspergillosis and other invasive mycoses. In these animal models, the invasive nature of the fungal infection was confirmed by re-isolation of the etiologic agent from the organs of every animal. Ninety-two plasma samples from 42 animals with invasive aspergillosis were submitted to the test. In 41 of these animals, at least one plasma sample was positive with the latex test (sensitivity 97.6%), titers ranging from 1/1 to 1/512. In general, antigen titers increased as a function of time, reaching the highest values shortly before death. Guinea-pigs infected with Penicillium marneffei also yielded positive agglutination reactions but antigen titers were lower (maximal titer 1/8). Plasma samples from animals with invasive candidosis (23), disseminated trichophytosis (11) and cryptococcosis (23) were all negative with the latex test. In 80 guinea-pigs without fungal infection, 3 false positive results (titers 1/1) were observed, which means a specificity of 96.2% in this control group.

Ultrasound has been widely used for the evaluation of the shoulder, mainly for rotator cuff pathology. Developments in technology, as well as better knowledge of the pathology and the anatomy make this examination one of the most useful in the exploration of the shoulder, especially in the hands of the experienced radiologist. Ultrasound is low-cost, readily available and should be considered with plain films as the first step examination of the shoulder. Ultrasound is not only useful for the evaluation of the rotator cuff pathology and impingement syndrome, but is also performing in the evaluation of non rotator cuff pathology such as biceps tendon pathology, shoulder instability, mass evaluation, infection, degenerative and inflammatory arthropathies and nerve entrapment syndromes. Making a complete evaluation of the shoulder helps to differentiate between rotator cuff pathology and others that can mimic rotator cuff disorders. This article will review shoulder anatomy and examination technique and the different pathologies that can be assessed by ultrasound.

1. We aimed to characterize 5-HT receptors mediating contraction and relaxation to 5-HT in dog proximal stomach longitudinal muscle (LM) strips. 2. Of the tryptamine analogues tested, 5-HT was the most potent contractile agent at basal length, while 5-CT was the most potent relaxant of PGF(2alpha)-induced contraction. Neither the contractions to 5-HT, nor the relaxations to 5-CT were influenced by tetrodotoxin, illustrating that action potential propagation is not involved. 3. The 5-HT-induced contraction was antagonized by mesulergine (0.03 to 0.3 microM) and ketanserin (2 - 20 nM), but the antagonism was not of a simple competitive nature, indicating multiple receptor involvement. Ketanserin (3 to 30 nM) and mesulergine (30 nM) competitively antagonized the alpha-Me-5-HT-induced contraction (pK(B): 8.83+/-0.09 and pA(2): 8.25+/-0.06 respectively). These affinity values are in line with literature affinities of ketanserin and mesulergine at 5-HT(2A) receptors in various bioassays. 4. The 5-CT-induced inhibition of PGF(2alpha)-induced contraction was competitively antagonized by mesulergine (pK(B) estimate: 8.52+/-0.12) and by the selective 5-HT(7) receptor antagonist SB-269970 (pK(B) estimate: 9.36+/-0.14). Both pK(B) estimates are in line with literature affinities of these compounds for 5-HT(7) receptors. Mesulergine (30 nM) and SB-269970 (10 nM) shifted the relaxant curve to 5-HT parallel to the right in the presence of ketanserin (0.3 microM) (pA(2) estimates of 8.08+/-0.10 and 8.75+/-0.14 respectively), indicative of 5-HT(7) receptor involvement. 5. It is concluded that 5-HT induces dog proximal stomach (LM) contraction via smooth muscle 5-HT(2A) receptors and relaxation via smooth muscle 5-HT(7) receptors.

Gastrin delays gastric emptying of liquids probably by reducing gastric tone. The mechanism responsible for the relaxatory effect induced by pentagastrin was unknown. The aim of this study was to investigate the effects of pentagastrin and the underlying mechanisms responsible for these effects. Could nitric oxide (NO) be involved as a mediator? Gastric tone was monitored with a flaccid bag introduced into the stomach via a gastric cannula and connected to a barostat. A series of pressure–volume curves with a 30-min interval were constructed by increasing intragastric pressure to a maximum of 14 mmHg (2-mmHg steps). Pentagastrin (4 μg kg−1 s.c.) facilitated the volume increases induced by isobaric gastric distension. This effect could be completely blocked by pretreatment with cimetidine (10 mg kg−1 s.c.) or by omeprazole (10 mg kg−1 p.o.). The effect induced by pentagastrin could be mimicked by histamine (0.16 mg kg−1 s.c.) and to a lesser extent by insulin (0.2 IU kg−1 i.v.). Cimetidine and omeprazole had no intrinsic effect on gastric tone. With an opened cannula, allowing the gastric secretions to leave the stomach, no increased gastric relaxation could be observed either in the presence of pentagastrin or in the presence of histamine or insulin. Nitro-l-arginine (L-NNA, 5 mg kg−1 i.v.) reduced the volume increases induced by distension. Unexpectedly, even in the presence of L-NNA, pentagastrin remained effective. In conclusion, pentagastrin induces a gastric relaxation via a mechanism which involves gastric secretion but not nitric oxide.

UD-CG-212 CL is a metabolite of pimobendan (UD-CG-115 BS), a non-glycosidic, non-adrenergic positive inotropic agent. In the present study we investigated the effect of UD-CG-212 CL on cat papillary muscles in the presence or absence of an intact endocardial endothelium. The endocardium was damaged by a very short detergent treatment. We demonstrated that, in muscles with an intact endocardial endothelium, UD-CG-212 CL induced a moderate, but significant inotropic effect resembling the changes induced by adrenoceptor agonists. Addition of an α- and or β-blocker reduced this positive inotropic effect. The effect induced by UD-CG-212 CL, was completely abolished after the endocardial endothelium was damaged. We conclude that the endocardium played a modulatory role in the action of UD-CG-212 CL through the release of various factors with inotropic activity.

The increasing number of patients with virus and drug-induced immunodeficiencies has also lead to a higher incidence of invasive candidosis and other opportunistic infections. Because of the bad prognosis of systemic Candida infections, early and accurate diagnosis is very important. Diagnosis is difficult, however, since traditional methods as blood culture and antibody determination are not very reliable. This has stimulated research on the presence of Candida antigens circulating in the blood. Although several recent reports have focussed on a 47 kD cytoplasmic protein 1, 2, the most extensively studied circulating antigen is mannan, a major cell wall polysaccharide 3, 5. Its detection in patient serum is a good indicator for deep-seated disease 6, 7.

We have studied the effects of vasopressin on isolated cat papillary muscle both before and after damaging the endocardium. The experiments were performed in physiological conditions of temperature (35 degrees C) and calcium concentration (1.25 mM Ca2+). Isometric and isotonic twitches as well as maximal unloaded velocity of shortening (Vmax) were measured. In muscles with an intact endocardium (n = 13), vasopressin (10(-12) to 10(-6) M) induced early twitch relaxation with a significant reduction of time to half isometric tension decline and with concomitant significant decrease in peak twitch performance. In a second group of muscles (n = 13) the endocardial endothelial surface was damaged by briefly (1 s) exposing the muscles to a 0.5% Triton X-100 followed by abundant wash with Krebs-Ringer solution, thereby irreversibly decreasing time to half twitch relaxation and peak twitch tension without significantly affecting Vmax. After this intervention, vasopressin had a positive inotropic effect with a significant increase in peak twitch tension and Vmax with no significant changes in twitch duration. Accordingly, in the presence of a functional endocardium, the direct myocardial positive inotropic effect of vasopressin was reversed, with early twitch relaxation and diminished peak twitch performance. At the highest concentrations of vasopressin, vasopressin-induced functional and morphological damage of the endocardium was observed.

The positive inotropic response to angiotensin I and II in cardiac tissue of most mammalian species, as well as the exact site in the heart for conversion of local and systemic angiotensin I into angiotensin II, remains to be elucidated. In isolated cat papillary muscles, angiotensin I and angiotensin II (0.1 nM to 1 microM, 35 degrees C, 1.25 mM Ca2+) increased, in a dose-dependent manner, peak twitch tension with typical slight prolongation of twitch duration. This typical response did not necessitate the presence of an intact endocardial endothelium (EE), as a similar response was observed in muscles where the EE had been damaged by a 1-second exposure to 0.5% Triton X-100. After addition of captopril, an angiotensin converting enzyme inhibitor, the positive inotropic response to angiotensin I was completely abolished, both in the presence and the absence of an intact EE. Hence, the heart possesses angiotensin converting enzyme, which mediates the positive inotropic response to angiotensin I. An intact EE was not a prerequisite for this response; thus, myocytes as well as nonmyocytes may be possible locations (in addition to the EE) for angiotensin converting enzyme. In the presence of an intact EE, and after addition of captopril, the positive inotropic response to angiotensin II was significantly diminished (desensitization). By contrast, in the absence of an intact EE, but also after addition of captopril, the positive response to angiotensin II was potentiated (sensitization). Both desensitization and sensitization (in the presence or absence of an intact EE, respectively) of the response to angiotensin II induced by the addition of captopril were inhibited by indomethacin, a cyclooxygenase inhibitor, suggesting a role for prostaglandins.

Although the positive inotropic response to alpha 1-agonists has been well documented, the mechanism is incompletely understood. In isolated papillary muscle of cat and rabbit, low physiological concentrations of phenylephrine (PE) (10(-9)-10(-7) M; 35 degrees C) under beta-blockade (propranolol 10(-6) M) and in physiological [Ca2+]o (1.25 mM), significantly increased peak twitch tension (TT) with typical unchanged or slightly prolonged twitch contraction duration. This typical response necessitated the presence of an intact endocardial endothelium (EE) since no inotropic response was observed at or below 10(-7) M PE in muscles where the EE had been damaged by 1 s exposure to 1% Triton X-100 prior to the first PE addition. After Triton-induced EE damage prior to the first PE addition, a direct myocardial inotropic response could be demonstrated, but this response required higher concentrations of PE (10(-6)-10(-3) M) and was significantly less pronounced than in muscles with intact EE. At elevated (2.5 mM) or high (7.5 mM) Ca2+, a concentration dependent inotropic response to PE could be induced at the higher concentrations of PE (10(-6) M or higher), but the inotropic response to low concentrations of PE (10(-7) M or below) was absent even in the presence of an intact EE. In papillary muscles with intact EE at physiological [Ca2+]o of 1.25 mM, the higher concentrations of PE (10(-6) M or higher) destroyed the EE. Indeed, after washing with fresh bathing solution following prolonged exposure to these high concentrations of PE, the baseline twitches resembled twitches in muscles where the EE had been experimentally damaged. Selective destruction of the EE by PE in these conditions with no morphological damage to the subjacent myocardium was confirmed on electron microscopy. A second concentration-response curve to PE was significantly depressed, shifted to the right and with no response at or below 10(-7) M PE. Subsequent successive curves were not significantly different from the second one. At the higher Ca2+ (2.5 mM or 7.5 mM), neither functional nor morphological evidence of damage to the EE was observed regardless of the PE concentration and exposure time; in some muscles EE damage did occur at high Ca2+, however, but only after multiple exposures to high PE each followed by abundant wash. At all concentrations of PE, the alpha 1-antagonist prazosin (10(-5) M) blocked the inotropic response regardless of [Ca2+]o and prevented morphological endothelial damage at 1.25 mM Ca2+.

In contrast to vascular endothelium, the functional role of endocardial endothelium has been largely underestimated. It was now been demonstrated that selective removal of the endocardial endothelium from isolated heart muscle preparations resulted in reduction of twitch duration and peak twitch tension, but did not significantly alter maximal unloaded velocity of shortening (V max ). Alternatively, the presence of an intact endocardial endothelium increased peak twitch tension mainly by prolonging twitch duration.

The mechanical properties of mammalian ventricular cardiac muscle have been studied in the presence and in the absence of an intact endocardial surface. Isotonic and isometric twitch contractions were obtained from papillary muscles of the right ventricle of cat at 29 degrees and 37 degrees C, at different extracellular calcium concentrations ([Ca2+]o), and at different initial muscle lengths. The endocardial surface was damaged by gentle abrasion of the muscle surface with a plastic blade or by brief immersion for 1 second with 1% Triton X-100. Although there was no evidence of damage to myocardial cells, damaging the endocardial surface resulted in an immediate and irreversible abbreviation of the twitch contractions with, except at the highest ([Ca2+]o, a decrease in peak isometric twitch tension. These changes induced 1) an asymmetrical shift of the tension-[Ca2+]o relation towards increasing [Ca2+]o but with no effect at the highest [Ca2+]o, and 2) a rightward and downward shift of the length-tension relation. Both shifts were significantly more pronounced at 37 degrees C than at 29 degrees C; they were not accompanied by significant changes in Vmax. The asymmetrical shift of the tension-[Ca2+]o relation suggests that the endocardium-mediated chain of events may be mediated by changes in the sensitivity of the contractile proteins to Ca2+. This hypothesis is also supported by the similar pattern of changes (i.e., modulation of the onset of early tension decline) induced by decreasing length at each [Ca2+]o and by the removal of a functional endocardium. Accordingly, the endocardium may help to control the performance of the heart by modulating peak contractile performance and relaxation of the underlying myocardium.

Ventricular mural thrombi complicate many cardiac diseases. The endocardial endothelium can modulate the mechanical performance of subjacent myocardium and mediate responses to certain physiopharmacologic agents. We studied the effects of aggregating platelets on the contractile performance of isolated cardiac muscle. The role of the endocardium was investigated by selectively damaging it by very brief (1 second) exposure to 1% Triton X-100 in some muscle preparations before experiments. Cat papillary muscles (n = 54) were attached to an electromagnetic length-tension transducer in organ baths containing Krebs-Ringer solution (1.25 mM Ca2+, 35 degrees C), and stimulated electrically at 0.2 Hz. Homologous washed platelets (final concentration 3 x 10(11)/l) aggregated spontaneously on addition to baths. Mechanical performance increased significantly more in muscles with damaged endocardium than in intact muscles (p less than 0.05); total peak isometric twitch tension increased by 31.8 +/- 7.8% (with damaged endocardium) and 11.8 +/- 2.6% (with intact endocardium), and peak isotonic twitch shortening increased by 36.7 +/- 7.8% (with damaged endocardium) and 9.6 +/- 2.0% (with intact endocardium). Increases in maximum velocity of unloaded shortening were similar in both muscle groups. Time to half isometric twitch tension decline decreased in intact muscles (3.6 +/- 1.0%) but increased in Triton-treated muscles (2.5 +/- 1.3%, p = 0.003 for difference between groups). The inotropic response to platelets in muscles with intact endocardium was unaltered by pretreatment of muscles with indomethacin (10 microM) or by stimulation of platelet aggregation with thrombin (0.1 unit/ml).(ABSTRACT TRUNCATED AT 250 WORDS)

Impairment of the endocardial surface has a profound influence on the mechanical performance of the underlying undamaged myocardium. It immediately and irreversibly shortens the duration of twitch tension development, particularly at a physiological extracellular [Ca++] and temperature, thereby affecting the relationship of peak isometric twitch tension development to both [Ca++]0 and length. Accordingly, the endocardium as the most primitive structure of the heart, may help to control the performance of the underlying myocardium by modulating the onset of early tension decline. These effects will result in important variations of peak contractile performance and of relaxation of the underlying myocardium.

Atrial natriuretic peptide, released by mammalian atria in response to volume overload, induces vasodilation and natriuresis. In this study, a direct effect on cardiac mechanical performance was demonstrated. Atriopeptin III (10(-9)-10(-7) M) induced early relaxation and decreased peak twitch of isometric and isotonic twitches of isolated papillary muscles of cat and rat, without affecting maximal unloaded velocity of shortening. This effect resembled the effects of dibutyryl cyclic GMP and of sodium nitroprusside on cardiac muscle. The action of atriopeptin III, but not of dibutyryl cyclic GMP or sodium nitroprusside, was abolished by mechanically or chemically damaging the endocardial endothelial surface. Thus, the early relaxation of cardiac muscle induced by atrial natriuretic peptide may be mediated through receptors on the endocardial endothelium.
