This paper presents modeling of the plasma load in vacuum chamber of a remote plasma source (RPS) and its generator is designed and implemented correspondingly. Traditionally, equivalent plasma load modeling usually acquired parameters measured by Langmuir probe which is hard to conduct and is basically intrusive. A simple method to model the plasma load into frequency and flow rate related equivalent RLC load without special device is proposed in this study. And two different resonant inverters which are operated around 400 kHz as an igniter and a sustainer of the plasma source generator are designed based on the equivalent load. The igniter generates high voltage 5 kV with argon gas as a trigger to start the plasma progress and the sustainer provides a maximum power of 2.5 kVA to keep plasma generating with nitrogen gas. To avoid interruption of plasma generation under gas flow rate varying from 100 to 600 sccm, a simple PI control with frequency modulation on the sustainer to regulate its output current is adopted. Both system design and equivalent load modeling are verified with simulated and experimental results in the paper.

This paper presents design and implementation of remote-plasma-source (RPS) generator for semiconductor manufacturing chamber cleaning. The generator consists of a full-bridge resonant inverter, an inductive coupling transformer, and a chamber with low pressure gases. To design the power converter for generating plasma sources, the equivalent load of a plasma chamber is required. A traditional approach to acquire equivalent plasma load is measuring plasma density through Langmuir probe. But the intrusive characteristic and power stress limitation make Langmuir probe not a convenient method to be applied in some plasma source design. This study proposes an engineering approach to deriving equivalent plasma load which takes switching frequency and gas flow rate into account without special devices needed. The transformer and the chamber are modeled with an RLC load which is used for designing the resonant tanks of the inverter and the igniter. The system configuration and modeling processes are presented in detail, from which the inverter and igniter designs are presented correspondingly. Simulated and experimental results have verified the analyses, design approaches and discussions of the RPS generator.

Vehicular emissions contribute a significant portion to fine particulate matter (PM2.5) air pollution in urban areas. Knowledge of the relative contribution of gasoline versus diesel powered vehicles is highly policy relevant and yet there lacks an effective observation-based method to determine this quantity, especially for its robust tracking over a period of years. In this work, we present an approach to track separate contributions by gasoline and diesel vehicles through positive matrix factorization (PMF) analysis of online monitoring data measurable by relatively inexpensive analytical instruments. They are PM2.5 organic and elemental carbon (OC and EC), C2–C9 volatile organic compounds (VOCs) (e.g., pentanes, benzene, xylenes, etc) and nitrogen oxides concentrations. The method was applied to monitoring data spanning over six years between 2011 and 2017 in a roadside environment in Hong Kong. We found that diesel vehicles accounted for ~ 70–90 % of the vehicular PM2.5 (PMvehicle) over the years and the remaining from gasoline vehicles. The diesel PMvehicle during a truck- and a bus-dominated periods showed declining trends, in coincidence with control efforts targeting at diesel commercial vehicles and franchised buses in the intervening period. The combined PMvehicle from diesel and gasoline vehicles by PMF agrees well with an independent estimate by the EC-tracer method, both confirming PMvehicle contributed significantly to the PM2.5 in this urban environment (~ 4–8 µg m−3, representing 30–60 % in summer and 10–20 % in winter). Our work shows that long-term monitoring of roadside VOCs and PM2.5 OC and EC is effective for tracking gaseous and PM pollutants from different vehicle categories. This work also demonstrates the value of evidence-based approach in support of effective control policy formulation.

Reliable markers for the rapid discrimination of severe renal damage remain a vital concern for lupus nephritis (LN). To determine a better tool for kidney damage detection, the present study compared the evaluation ability of novel urinary cytokines and chemokines (namely urinary monocyte chemoattractant protein 1 (uMCP-1), tumor necrosis factor-like weak inducer of apoptosis (uTWEAK)) with traditional serum or urinary markers (namely urinary alpha 1-microgrobulin (uα1-MG), beta 2-microglobulin (uβ2-MG) and serum complement C3 (C3), complement C4 (C4), creatinine (Cr), blood urea nitrogen (BUN) and cystatin C (Cys C)) in discriminating LN renal damage. Correlations between markers with Systemic Lupus Erythematosus Disease Activity Index (SLEDAI) renal SLEDAI scores, biopsy activity index (BAI) and biopsy chronicity index (BCI) scores were evaluated. Receiver operating characteristic (ROC) curves were generated to evaluate a single or combined model in discriminating active renal involvement (rSLEDAI scores 0) and patients with poor pathological outcome (BAI scores ≥ 7). uMCP-1 and uTWEAK possess higher correlation coefficients with renal damage and larger areas under ROC curves (AUCs) than other markers. A combined model of uMCP-1 and uTWEAK showed an AUC of 0.887, sensitivity of 86.67% and specificity of 80.00% to discriminate active LN, and an AUC of 0.778, sensitivity of 75.00% and specificity of 81.82% to discriminate LN with poor outcome, which are better than the utility of any markers individually.

This report details the newly identified antigens in existing blood group systems and presents three new blood group systems.

We found that the formation of single or double carbon vacancies in graphene are critical for the adsorption of Li atoms. While the N-doping facilitates the formation of vacancies, it introduces over binding issue and hinders the Li diffusion. The presence of B takes the excessive electrons from Li and N and reduces the energy barrier of Li diffusion on substrates. We perceive that these clear insights are crucial for the further development of graphene based anode materials for lithium-ion batteries.

The rapid conversion of the i to the I phenotype on postnatal red blood cells (RBCs) reflects the elaborate functions of cell surface branched-chain I antigenic structure. The adult i phenotype has been noted to be partially associated with congenital cataracts. These phenomena make the molecular genetics of the blood group I system and the regulation mechanism for I antigen expression in postnatal RBCs intriguing. It has been demonstrated that the human I locus expresses three IGnT forms, designated IGnTA, IGnTB, and IGnTC, which have different exon 1, but identical exons 2 and 3, coding regions. The uncommon molecular genetics of the I locus offers a new perspective of the formation and expression of the I antigen in different cells, and provides an insight into the questions derived from investigation of the adult i phenotype. The results obtained from molecular analysis of two adult i groups, with and without congenital cataracts, support the proposed molecular mechanism for the partial association of the two traits, namely that mutations that lead to the production of mutant IGnTC only, may result in an adult i phenotype but not congenital cataracts, whereas mutation events that occur in the common exon 2 or exon 3 regions, which result in the elimination of the activity of all three IGnT enzymes, would seem to lead to the development of both traits. This suggests that an I-gene defect may lead directly to the development of congenital cataracts. However, the result of the gene knock-out mouse model does not confirm this suggestion. Analysis of the regulation for I antigen expression shows that i-to-I phenotypic transition during erythroid differentiation is regulated by the transcription factor CCAAT/enhancer binding protein α (C/EBPα), which enhances transcription of the IGnTC gene, consequently leading to the formation of the cell surface I antigen. Subsequent investigation further showed that phosphorylation of the Ser-21 residue on C/EBPα blocks C/EBPα activity on IGnTC gene induction, while dephosphorylation of C/EBPα Ser-21 occurs in both the granulopoietic and erythropoietic processes, leading to stimulation of IGnTC gene expression and consequently I antigen formation. These results demonstrate that the formation of I antigen during the development of erythrocytes, and also granulocytes, is determined by a mechanism that is known to be crucial to determining granulopoiesis, namely involvement of the transcription factor C/EBPα and Ser-21 dephosphorylation of this protein.

We previously reported that vanilloid receptor type 1 (VR1, or TRPV1) was up-regulated in dorsal root ganglion (DRG) and the spinal dorsal horn after chronic inflammatory pain produced by complete Freund's adjuvant (CFA) injection into the plantar of rat hind paw. In the present study, we found that subcutaneous or intrathecal application of capsazepine (CPZ), a TRPV1 competitive antagonist, could inhibit thermal hyperalgesia on day 1 and on day 14 but not on day 28 after CFA injection. With extracellular electrophysiological recording, the effect of CPZ on noxious electrical or heat stimulation evoked responses of wide dynamic range (WDR) neurons in the deep layers of the spinal dorsal horn was evaluated. Under noxious electrical stimulation to sciatic nerve, CPZ applied to the spinal cord produced an inhibition on Adelta- and C-fiber evoked responses of WDR neurons on day 1 and 14, but not on day 28. Under radiant heat stimulation to the receptive field skin, subcutaneous application of CPZ significantly inhibited the background activity and extended the response latency of WDR neurons on day 14. These results provide new evidence for the functional significance of TRPV1 at the early stage, but not the late stage, in the rat model of CFA-induced inflammatory pain.

Background: The present study aims to investigate the role of transient receptor potential vanilloid 1 (TRPV1) in dorsal root ganglion (DRG) neurons in chronic pain including thermal hyperalgesia and mechanical allodynia. Chronic inflammatory nociception of rats was produced by intraplantar injection of complete Freund's adjuvant (CFA) and data was collected until day 28 following injection. Results: Thermal hyperalgesia was evident from day 1 to day 28 with peak at day 7, while mechanical allodynia persisted from day 1 to day 14 and was greatest at day 7. Intrathecal administration of AMG 9810 at day 7, a selective TRPV1 antagonist, significantly reduced thermal hyperalgesia and mechanical allodynia. TRPV1 expression in DRG detected by Western blotting was increased relative to baseline throughout the observation period. Double labeling of TRPV1 with neuronal marker neurofilament 200 (NF200), calcitonin gene-related peptide (CGRP) or isolectin B4 (IB4) was used to distinguish different subtypes of DRG neurons. TRPV1 expression was increased in the medium-sized myelinated A fiber (NF200 positive) neurons and in small non-peptidergic (IB4 positive) neurons from day 1 to day 14 and was increased in small peptidergic (CGRP positive) neurons from day 1 to day 28. Conclusion: TRPV1 expression increases in all three types of DRG neurons after CFA injection and plays a role in CFA-induced chronic inflammatory pain including thermal hyperalgesia and mechanical allodynia.

To investigate the functional brain pain center and default mode network response to electro acupuncture stimulate in weizhong acupoints(BL40) and dachangshu acupoints(BL25).During January to February 2015, volunteers were enrolled in this study from the staff and student interns of Gansu Province Traditional Chinese Medicine Hospital. A total of 20 healthy, right-handed subjects, male 9, female 11, age (23±3) years, participated in this study. Block design task functional magnetic resonance imaging(fMRI) 3.0 T was performed in all subjects by electro acupuncture stimulating at BL40 and BL25 from the same experienced acupuncturist.The needle connected electric acupuncture apparatus through tow long coaxial-cable. A block design with five 120 s blocks of rest time (OFF block, electric acupuncture turn off ) interspersed between five 60 s blocks of stimulation (ON block, electric acupuncture turn on) fMRI scan. Magnetic resonance data of brain function was collected and FSL(fMRI Software Library) software was used to analyze the data.All subjects' data were analyzed except 2 cases whose head movement were more than 2 mm. Activated brain function regions by electro acupuncture stimulate included temporal lobe lateral sulcus, lobus insularis, thalamus, supramarginal gyrus, prefrontal medial frontal gyrus. Negative activated brain regions included middle frontal gyrus, parahippocampal gyrus, cingulate cortex abdominal segment, parietal cortex.The functional pain central and default mode network were changed when electro acupuncture stimulate in(BL40) and(BL25).There are several brain activation regions and negative activated brain regions when administering electro acupuncture stimulation at BL40 and BL25.

Metabolomic analysis is a technology which seeks to provide a comprehensive profile of all the metabolites present in a biological sample ( Fiehn 2002 ; Noguchi et al. 2003 ; Watkins et al. 2002 ). Metabolomics is not a new science, but emerging as an exciting application which can span the scope of biotechnology and medicine, providing metabolic profile and a complement to the genomic and proteomic data ( German et al. 2003 ). Studies have already begun to explore the effects of toxicological, pharmaceutical, nutritional, and environmental intervention and to build integrated databases of metabolite concentrations throughout biological systems, ranging from microbes to plants to human and research animal populations ( Bruskiewich et al. 2003 ; Bailey et al. 2003 ; Fiehn 2003 ; Novotna et al. 2003 ). This may provide a tool for discovering a novel pathway, or determining the relationship of these metabolite concentrations to disease, and the extent to which nutrition can modulate metabolite concentrations ( Weckwerth and Fiehn 2002 ). Metabolites are the products of enzymatic processes. Their levels can be regarded as the ultimate response of biological systems to genetic or environmental changes ( Fiehn 2002 ). While the amount of an enzyme protein in a biological system could be quantified by using specific antibodies or by measuring the mRNA responsible for their production, factors such as the availability of cofactors and coenzymes, feedback regulation, pH, compartmentalization, etc. could affect their activity, thus, in turn, affect the metabolite's (i.e., substrates and their enzymatic products) levels. There have been a number of different interpretations of "metabolites." In pharmacological and toxicological studies, metabolites could be considered as the enzymatic degradative products of drugs ( Nishikawa et al. 2003 ; Plumb et al. 2002 ; Reo 2002 ; Robosky et al. 2002 ; Slim et al. 2002 ). In other studies, metabolites could be referred to as amino acids ( Noguchi et al. 2003 ), keto acids ( Bailey et al. 2003 ), or lipids ( German et al. 2003 ), which are substrates and products of selected metabolic pathways. Noguchi et al. (2003) proposed using "subset" to indicate the type of metabolites to be studied. Thus, the amino acid subset, for example, would indicate the levels of all the amino acids in a sample and lipomic subset would indicate the levels of all metabolites involved in lipid metabolism. In biochemical, physiological, and toxicological studies, animal models are preferred. In fact, a large amount of information on metabolites balance has been accumulated for a number of pharmacological, toxicological, and pathological conditions ( Henry et al. 1985 ; Needleman et al. 1968 ; Tourtellotte et al. 1966 ; McDougal, Jr. et al. 1997 ; Yang et al. 1985 ; King et al. 1967 ; Lowry et al. 1983 ; Gupta et al. 2000 ; Gupta et al. 2001a ,b; Gupta and Dettbarn 2003). Only recently has attention been directed towards studying metabolites in cell culture. Cell culture, with intact membrane and cytoplasmic organelles that operate under well understood biochemical and molecular events, provides a simple model that is best used for studying the basic mechanisms of toxicological and pharmacological actions of xenobiotics. Other advantages of using cell culture are to lighten the heavy workload and complications involved in performing animal studies, as well as to reduce the use of animals, which is also advocated on the ground of good animal ethics. To correlate the results in vivo, the data need to be transformed taking into consideration the level of toxicant at the site of action.

The central nucleus of amygdala (CeA) plays an important role in pain regulation. Calcitonin gene-related peptide (CGRP)-like immunoreactive fibers and CGRP receptors are distributed densely in CeA. The present study was performed to elucidate the role of CGRP in nociceptive regulation in the CeA of rats. Intra-CeA injection of CGRP induced dose-dependent increases in the hind-paw withdrawal latency tested by hotplate test and Randall Selitto Test, indicating an antinociceptive effect of CGRP in CeA. Furthermore, the antinociceptive effect of CGRP was blocked by intra-CeA administration of the CGRP receptor antagonist CGRP8-37, suggesting that CGRP receptor1 is involved in the CGRP-induced antinociception. The CGRP-induced antinociception was attenuated by s.c. injection of the opioid antagonist naloxone, suggesting an involvement of endogenous opioid systems in CGRP-induced antinociception. Moreover, it was demonstrated that opioid receptors in the periaqueductal gray, but not in CeA, contributed to the CGRP-induced antinociception, indicating the importance of the pathway between CeA and the periaqueductal gray in CGRP-induced antinociception. Combining retrograde fluorescent tracing with immunohistochemistry, we found that met-enkephalinergic neurons were innervated by CGRP-containing terminals in CeA. Furthermore, most neurons in the CeA retrogradely traced from the periaqueductal gray were contacted by CGRP-containing terminals and some of them were surrounded by characteristic basket-like structures formed by the terminals, suggesting that CGRP innervates the neurons which project from CeA to the periaqueductal gray. The results indicate that CGRP activates the met-enkephalinergic neurons, which project from CeA to the periaqueductal gray, producing antinociceptive effect in rats.

Background: Anthropophobia, a subtype of social phobia, is prevalent in Chinese and Japanese societies. This study investigated sociocultural influences on the course of this culturally specific mental disorder. Method: One hundred and fifty subjects, including 50 anthropophobic, 50 neurasthenic, and 50 community subjects, were interviewed in Beijing, China for the assessment of their early life experiences (child-parent relationships and sexual experiences), collectivism disposition, sexual attitudes, and communication behaviors. Logistic and linear regression analyses were performed to examine significant predictors of the occurrence and the symptom level of anthropophobia. Results: Regression models explained 69% of variance in the diagnosis and 57% of variance in the symptom level of anthropophobia among anthropophobic and community subjects. They also explained 48% and 47% of variance respectively in the diagnosis and the level of symptoms among anthropophobic and neurasthenic subjects. Anthropophobic subjects had more problematic relationships with parents than did community and neurasthenic subjects. They also exhibited significantly stronger characteristics of collectivism than did community subjects. Sexual contact with a non-family member prior to age 19 and a feeling of discomfort when interacting with the opposite sex were significantly associated with the diagnosis and symptom level of anthropophobia. Conclusions: It was concluded that anthropophobic subjects' early sexual experiences and need for parental approval shaped their conformity to social norms and negative sexual attitudes, which were reinforced by the collective-orientated cultural environment, and contributed to the development of anthropophobia.

The present study was performed in rats with experimentally induced mononeuropathy after left common sciatic nerve ligation. The hindpaw withdrawal latencies to thermal and mechanical stimulation increased significantly after intra-periaqueductal grey injection of 2 or 3 nmol, but not 1 nmol of galanin in rats with mononeuropathy. Intraperitoneal administration of 4.5 mg/kg morphine induced significant increases in hindpaw withdrawal latencies to both noxious stimulation, which were attenuated by following intra-periaqueductal grey injection of 2 nmol of the galanin antagonist galantide. Furthermore, the antinociceptive effect induced by intra-periaqueductal grey injection of 26.6 nmol of morphine was attenuated significantly by following intra-periaqueductal gray administration of 2 nmol of galantide. The results demonstrated that in periaqueductal grey galanin plays an antinociceptive role in rats with mononeuropathy and galanin is involved in the mechanisms of opioid-induced antinociception.

Due to the escalating interest in zero ODP (ozone depletion potential) blowing agents throughout the world, hydrocarbons are seriously being considered as potential candidates for rigid foam applications. However, many rigid foam applications require a certain degree of fire resistance, a characteristic which the flammable nature of hydrocarbons would seem to oppose. Last year we presented a paper which detailed our initial work with hydrocarbon-blown foams produced on a lab-scale high pressure machine [1]. In that work, several techniques were explored which improve the flame resistance of isocyanurate foams. These techniques included increased index, inclusion of phosphorous, and the use of inerting co-blowing agents. This paper will present further work concerning the flammability of rigid isocyanurate foams blown with hydrocarbons, with a particular emphasis on cyclopentane as the primary blowing agent. In this study we have investigated the use of various foaming techniques to suppress flammability. As in our previous paper, all foams were produced on a high pressure foam machine. Options which were explored include fire retardants, co-blowing agents, and increased foam indices. Results from small-scale fire tests are presented in order to compare the effectiveness of the different approaches. Mini-tunnel and hot plate test results are featured.

Objective: To compare the measurement of quantitative DNA in squamous cell carcinoma of the head and neck by flow cytometry and image cytometry. Design: Comparison of image cytometry to the more commonly used flow cytometry using paraffin-embedded tissues. Setting: University of California, Davis Medical Center, Sacramento. A 472-bed university teaching hospital. Patients: Records of 26 patients with squamous cell carcinoma of the tongue, base of tongue, and larynx were obtained from the case files of an otolaryngologist–head and neck surgeon. They were reviewed for staging and follow-up. Results: We demonstrated a 96% concordance rate between the methods. A solitary discrepant case was aneuploid by image cytometry and diploid by flow cytometry. The specimen involved tumor infiltrated by lymphocytes that may have masked the aneuploid population to measurement by flow cytometry. Quantitative DNA analysis correlated moderately well with tumor grade, tumor stage, and patient outcome with a minimum of 6 years of follow-up. All patients with diploid tumors were long-term survivors. Conclusions: Both methods provide accurate quantitative DNA analyses in squamous cell carcinoma of the head and neck. The methods are highly correlative and yield similar predictive data regarding tumor behavior and prognosis. (Arch Otolaryngol Head Neck Surg. 1994;120:294-297)

Aims/Objectives: The purpose of this study was to explore the molecular basis of the K0 phenotype of a Taiwanese blood donor found to have anti-Ku alloantibodies. Background: With respect to Kell blood group antigens, almost all Taiwanese have the (K−, k+) phenotype. Materials and Methods: Alloantibody identification and KEL antigen typing were performed. Enzymatic function assays were carried out to detect the Kell glycoprotein on RBCs. The KEL genes were sequenced to detect genetic variation. To determine the origin of this novel allele, family studies were conducted. Results: The alloantibody was identified as anti-Ku. The donor was typed K0. The KEL gene-sequencing data revealed that this K0 donor is a compound heterozygote with two different null alleles. He bears a novel 730delG mutation in one allele. Family studies suggested that the donor inherited the 730delG mutation from his father. The endothelin-converting activity assay indicated that his RBCs had no functional Kell glycoprotein. Other family members who had only one null allele with the 730delG mutation had the phenotype (K−, k+). Conclusion: For blood transfusion safety, it is important to establish an effective screening algorithm to identify rare phenotypes, such as the K0 phenotype, and to establish a database of rare blood groups.

Fluorescence in situ hybridization (FISH) was applied to detect the copy number of the retinoblastoma (RB1) tumor suppressor gene in metaphase chromosomes and interphase nuclei. We used 14 λ phage clones spanning the whole RB1 gene region as a probe and obtained a specific hybridization signal in normal metaphase chromosomes at 13q14. Normal interphase nuclei showed two RB1 signals in about 90% of cases, whereas two cell lines with cytogenetically defined deletions involving the RB1 gene showed only one hybridization signal in about 80% of the nuclei. Analogous changes were detected in metaphase chromosomes. Multicolor FISH with subsets of the phage clones allowed visualization of subregions within the 200-kb gene in interphase nuclei. Analysis of clinical breast cancer samples showed that most of the cells contained two copies of the RB1 gene, even when restriction fragment length polymorphism analysis showed loss of heterozygosity (LOH) at the RB1 locus. This indicates that LOH at the RB1 locus in breast cancer cells probably involves mechanisms other than physical deletion.

The committee met in Macao Special Administrative Region,China, during the 2008 International Society of Blood Trans-fusion (ISBT) Congress. Some changes to the classificationdocumented in Blood Group Terminology 2004 [1] and updatedin 2007 [2] were agreed and are described below. The fullupdated classification can be found on the blood groupterminology website at http://www.blood.co.uk/ibgrl. A newblood group system, the RHAG system, was established andnew antigens were added to the Rh, Kell, and Dombrocksystems (Table 1). A total of 308 antigens are now recognized,270 of which are clustered in 30 blood group systems.

When chemically skinned muscle fibres are activated by Ca2+ at an ionic strength of 170 mM, the spacing between the filaments has been shown to decrease with increasing force, suggesting that the cross-bridges can generate force not only in the axial but also in the radial direction. In the present study, radial force and radial stiffness of activated single skinned rabbit psoas fibres were studied by X-ray diffraction. The responses of the lattice spacing to changes in osmotic pressure by application of dextran T500, which is equivalent to force applied in the radial direction, was examined. The radial force generated by the attached cross-bridges was calculated, with the approximation that a negligible fraction of cross-bridges was attached in the relaxed muscle at the same ionic strength of 170 mM. 2. The active radial force was found to be a slightly non-linear function of lattice spacing, reaching zero at 34 nm. The radial force was compressive at lattice spacing greater than 34 nm and expansive at less than 34 nm. 3. The active axial force, on the other hand, was found to be much less affected by the application of dextran T500. Active axial force increased by 4% to a plateau at 4% dextran T500 and then decreased by 10% at 8% dextran T500. 4. While not under osmotic pressure, the radial force of the activated fibre was determined to be 400 pN (single thick filament)-1. This is of the same order of magnitude as the axial force. The radial stiffness was also comparable to the axial stiffness at 7 pN (thick filament)-1 (0.1 nm)-1. 5. The radial elasticity of the fully activated fibre differs significantly from that of the fibre in rigor. The radial stiffness exhibited by fibres in rigor was approximately five times higher, at 30 pN (thick filament)-1 (0.1 nm)-1 and the point where the radial force reached zero was 38 nm. 6. In the activated state, the point at which radial force reaches zero is independent of the level of Ca2+ activation, i.e. independent of the number of cross-bridges attached to actin in the force-generating state. We suggest that the zero-force point is equivalent to the equilibrium point of a spring and is an intrinsic property of the radial elasticity of the cross-bridge. 7. It is concluded that activated and rigor cross-bridges exhibit a spring-like property in the radial direction.(ABSTRACT TRUNCATED AT 400 WORDS)

Single crystals and powders of the layered, wide bandgap semiconductor, Cd 2 P 2 S 6 , have been intercalated with Ru(bipy) 2+ 3 by a cation exchange mechanism. Cation vacancies are created in the lattice to compensate for the charge of the intercalated ions. The photoluminescence spectrum of the intercalated material has been examined over the temperature range 1.8–77 K and compared with the photoluminescence properties of the host lattice and the free intercalate. Analysis of the spectra indicates that only weak interactions exist between the intercalate and the host lattice. The polarization of the photoluminescence suggests that the Ru(bipy) 2+ 3 ions are partially oriented with their C 3 symmetry axes perpendicular to the host lattice layers.

Changes in cellular energy and redox states were studied in the C6 glioma cells following exposure to chemicals that affect glutathione metabolism. It was demonstrated that treatment with sublethal concentrations (25, 50 and 100 microM) of buthionine sulfoxamine (BSO) did not affect cellular energy state as measured by total adenosine nucleotides (TAN=ATP+ADP+ AMP), ATP:ADP:AMP and energy charge potential (ECP=[ATP + 0.5 (ADP)]/TAN). However, there was a significantly decrease in cellular GSH/GSSG and total glutathione (TG=[GSH+GSSG]/ TAN). The change was due to a significant decrease in intracellular GSH level without significant change in [GSSG]. Cells exposed to BSO for 24 hr were much more sensitive to subsequent injuries caused by Cd (0.6 mM for 3 hr). The results indicated that while a significant reduction of intracellular redox state did not affect cell viability, it could increase the susceptibility of cells to subsequent chemical stress. N-acetylcysteine (NAC), on the other hand, caused a dose (1, 5 and 10 mM)-dependent increase in GSH/GSSG without significant changes in intracellular energy state. Improvement of intracellular GSH/GSSG offered no protection against subsequent Cd induced cell death unless NAC was present at the time Cd was added. The pattern of cell death also correlated with the increase in intracellular free radial generation as measured by the fluorescence labeling with 27- dichlorofluorescin. Results of the present study demonstrated that intracellular redox states could be manipulated by addition of chemicals that affect glutathione metabolism. While the redox state may not be the sufficient condition to cause cell death, it could modulate the response of cells to subsequent Cd treatment. Furthermore, the action of NAC against Cd cytotoxicity may not be related to intracellular redox status.

The present study examined correlates of life satisfaction among 350 Chinese elderly aged 65 or older. Subjects consisted of a clinic sample (n = 200) and a randomly selected community sample (n = 150) recruited from the same area in Beijing. Linear regression analysis was performed, using health, financial status, and family support as independent variables. Results showed that the regression model explained 38 percent of the variance in life satisfaction in the total sample, 34 percent and 43 percent of the variance respectively in the clinical and community samples. Life satisfaction was significantly predicted by health and financial status among community elderly and by health, financial status, as well as family support among outpatient elderly. Findings suggest that the socio-cultural context has exerted important impacts on the Chinese elderly's life satisfaction.

Studies have shown that 5-hydroxytryptamine (5-HT) plays an important role in the descending pathway of pain modulation from brainstem to the spinal cord. Using selective 5-HT receptor antagonists, the present study investigated which type of 5-HT receptor(s) in the spinal cord was involved in the morphine-induced anti-nociception in intact rats, in rats with nerve injury and in rats with inflammation. The hindpaw withdrawal latencies decreased significantly after sciatic nerve injury and hindpaw inflammation compared with intact rats. Intrathecal administration of 25 or 10 μg of the selective 5-HT 1A recepter antagonist spiroxatrine, but not 1 μg of spiroxatrine, significantly blocked the increased hindpaw withdrawal latencies to thermal and mechanical stimulation induced by intra-periaqueductal gray injection of 1 μg of morphine in intact rats. Intrathecal injection of the 5-HT 2 receptor antagonist RS 102221 and the 5-HT 3 receptor antagonist MDL 72222 had no significant effects on the increased hindpaw withdrawal latencies to both noxious stimulations induced by intra-periaqueductal gray injection of morphine. Furthermore, intrathecal administration of spiroxatrine, but not RS 102221 nor MDL 72222, significantly attenuated the increased hindpaw withdrawal latencies induced by intra-periaqueductal gray administration of morphine in rats with nerve injury and in rats with inflammation. The results demonstrate that the 5-HT 1A receptor, not 5-HT 2 nor 5-HT 3 receptor, plays an important role in the descending pathway of anti-nociception from the brainstem to the spinal cord in intact rats, in rats with nerve injury and in rats with inflammation.

The troponin complex in a muscle fiber can be replaced with exogenous troponin by using a gentle exchange procedure in which the actin-tropomyosin complex is never devoid of a full complement of troponin (Brenner et al. (1999) Biophys J 77: 2677-2691). The mechanism of this exchange process and the factors that influence this exchange are poorly understood. In this study, the exchange process has now been examined in myofibrils and in solution. In myofibrils under rigor conditions, troponin exchange occurred preferentially in the region of overlap between actin and myosin when the free Ca2+ concentration was low. At higher concentrations of Ca2+, the exchange occurred uniformly along the actin. Ca2+ also accelerated troponin exchange in solution but the effect of S1 could not be confirmed in solution experiments. The rate of exchange in solution was insensitive to moderate changes in pH or ionic strength. Increasing the temperature resulted in a two-fold increase in rate with each 10 degrees C increase in temperature. A sequential two step model of troponin binding to actin-tropomyosin could simulate the observed association and dissociation transients. In the absence of Ca2+ or rigor S1, the following rate constants could describe the binding process: k1 = 7.12 microM(-1) s(-1), k(-1) = 0.65 s(-1), k2 = 0.07 s(-1), k(-2) = 0.0014 s(-1). The slow rate of detachment of troponin from actin (k(-2)) limits the rate of exchange in solution and most likely contributes to the slow rate of exchange in fibers.

The role of nociceptin, the endogenous ligand for the opioid receptor-like (ORL1) receptor, in nociceptive processing is controversial. Most studies demonstrate hyperalgesia following supraspinal administration, analgesia following intrathecal and peripheral administration at higher doses, and hyperalgesia following intrathecal and peripheral application at lower doses. The present study investigates the effect of nociceptin on synovial plasma extravasation and its ability to modulate 5-hydroxytryptamine-induced synovial plasma extravasation using the rat knee joint model of inflammation. Nociceptin alone does not alter synovial plasma extravasation from baseline. Nociceptin at concentrations up to 1 nM enhances 5-hydroxytryptamine-induced synovial plasma extravasation (up to 50%) and nociceptin at concentrations above 100 nM inhibits 5-hydroxytryptamine-induced synovial plasma extravasation (down to 45%). The novel, selective ORL1 receptor antagonist J-113397 potently inhibits the pro-inflammatory effect of nociceptin, but only partly inhibits, at higher concentrations, the anti-inflammatory effects of nociceptin. These findings demonstrate a dose-dependent bi-directional effect of nociceptin on inflammatory processes and may indicate a target for novel therapeutics.

A single layer of epithelial cells lines the gastrointestinal tract, forming a critical barrier between the lunminal contents, which includes antigens and other noxious substances, and the body proper. It has become clear in recent years that the role of mast cells in the gastrointestinal mucosa is not only to react to antigens, but also to actively regulate the barrier and transport properties of the intestinal epithelium. Mucosal mast cells respond to both IgE/antigen-dependent and non-IgE-dependent stimulation, releasing bioactive mediators into adjacent tissues where they induce physiological responses. Studies in models of hypersensitivity and stress have provided evidence that changes in mucosal function are due to either direct action of mast cell mediators on epithelial receptors and/or indirect action via nerves/neurotransmitters.

The Kell blood group system is polymorphic, and 23 antigens have been defined to date. The Kell antigens are located on a single red cell transmembrane glycoprotein, encoded by the 19 exons of the KEL gene. The different Kell phenotypes result from point mutations leading to amino acid changes in the Kell glycoprotein. An unusual phenotype, which is defined as the complete lack of all of the Kell antigens, has been identified and designated as the Kell-null or Ko phenotype. The coding region of the KEL gene of the Ko individual showed a normal KEL2/KEL4/KEL7 gene sequence; nevertheless, a G to C mutation at the splice donor site (5' splice site) of intron 3 was found to be present as a homozygote in the individual. The mutation destroys the conserved GT sequence of the splice donor site. Reverse transcription-polymerase chain reaction analysis showed the absence of the complete KEL mRNA. Instead, a major transcript with the exon 3 region skipped was found. The exon 3 of the KEL gene encodes the transmembrane domain of the Kell glycoprotein, and a transcript without exon 3 is predicted to have a premature stop codon that abolishes the translation of C-terminal segment. The segment contains all of the known positions responsible for characterizing different Kell antigens, and this explains the lack of all Kell antigens in Ko red cells.

Intact caldesmon and particularly the actin-binding C-terminal fragment (20-kDa) of caldesmon have been shown in skeletal muscle fibers to selectively displace low affinity, weakly bound cross-bridges from actin without significantly altering the actin attachment of force producing, strong binding cross-bridges (Brenner et al., 1991; Kraft et al., 1995a). However, the sarcomeric distribution and the specific binding of externally added caldesmon to the myofilaments of skeletal muscle fibers was not known. It was e.g., unclear whether caldesmon binds along actin in a manner similar to tropomyosin or whether it also binds to myosin. In this study, we determined the binding pattern of exogenously added intact caldesmon and its C-terminal 20-kDa fragment, respectively, in MgATP-relaxed rabbit skeletal muscle fibers using electron (EM) and confocal fluorescence microscopy (CFM). EM showed that similar to what has been demonstrated earlier for smooth muscle thin filaments (Lehman et al., 1989), intact caldesmon binds periodically every 38 nm along the thin filaments. CFM revealed that rhodamine-labeled intact caldesmon and the 20-kDa caldesmon fragment bind along nearly the entire length of the thin filaments. A portion of the I-band near the Z-line appears unlabeled, both when equilibrated at normal and long sarcomere lengths. The width of the unlabeled region seems to depend on ionic strength. The 20-kDa C-terminal caldesmon fragment binds in essentially the same pattern as intact caldesmon. This indicates that the high fluorescence intensity in the overlap region seen with intact caldesmon does not depend on caldesmon binding to myosin. X-ray diffraction was used to monitor the effects of filament lattice. Intact caldesmon at0.3 mg/ml induced disorder in the myofilament lattice. No such disordering was observed, however, when fibers were equilibrated with up to 0.8 mg/ml of the 20-kDa caldesmon fragment.

The human Secretor alpha(1, 2)fucosyltransferase gene determines the ABH secretor status and influences the Lewis phenotype of an individual. Studies were carried out on the Lewis (a+b-) nonsecretors of different groups indigenous to Taiwan to demonstrate their se genotypes.The Lewis phenotype of the blood samples was determined by a microplate method. The se genotypes of the individuals with the Lewis (a+b-) phenotype were analyzed by a polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method designed for the se alleles reported previously. PCR and cloning techniques were used to determine the coding sequence of the novel se gene.A new se allele, se685, with a three-nucleotide deletion of GTGGT to GT in the coding region of nucleotides 685 through 689 was identified in a Le (a+b-) nonsecretor from the Ami tribe indigenous to Taiwan. The deletion predicts the loss of the amino acid Val230 in the corresponding secretor enzyme's C-terminal segment. The distribution of the se685 allele in the Ami tribe was further verified by PCR-RFLP analysis.The Se gene exhibits heterogeneity with some Se alleles being common but others displaying a unique distribution in different ethnic populations. The newly identified se685 allele seems to exist only in the Ami tribe indigenous to Taiwan.

Two dimensional x-ray diffraction was obtained from skinned rabbit psoas muscle fibers. The goal is to correlate structures of the cross-bridge population with various intermediate states in the cross-bridge cycle by using nucleotides and their analogs. It was found that in a relaxed muscle in ATP containing solutions, cross-bridges are distributed in three populations in equilibrium: those detached from actin and ordered on the myosin helix, those that are detached and disordered, and those weakly attached to actin in random orientations. The distribution among the three populations is highly dependent on temperature. Those that are detached and yet ordered in a helical structure surrounding the myosin backbone are very likely in the M.ADP.Pi state, supporting an earlier suggestion by Wray (1987). It was also found that the attached cross-bridges with bound MgADP are structurally distinct from those without nucleotide, in agreement with one of our earlier findings by osmotic compression (Xu et al., 1993). Another finding of interest is that the analog AMP-PNP was found to be an ATP analog, rather than an ADP analog as it has been reported previously by many research groups.

Comparative genomic hybridization (CGH) allows entire genomes to be scanned for whole and segmental aneuploidy and thus may be an appropriate tool for the detection of clinically important abnormalities during fetal and neonatal screening. Criteria to distinguish between significant aberrations and experimental artifacts are essential for these applications. This report describes the use of a t-statistic to detect changes in CGH profiles that differ significantly from variations that occur in CGH profiles of normal samples. Eleven cell lines derived from fetal or neonatal patients were analyzed in this study. Aneuploidies in these lines included trisomies for chromosomes 13, 16, 18, and 21 and monosomy for distal 5p and tetrasomy 18p. Aneuploidy was detected in all samples by using the t-statistic, although the extent of the aneuploid region was not correctly estimated in some cases. A detailed description of the t-statistic fused for making these CGH comparisons is described in a companion paper (Moore et al., Cytometry 28:183-190, 1997.

This study examines self-perceived physical and mental health among 213 Chinese elderly who visited the Geriatric Outpatient Clinic of Beijing Hospital, the People's Republic of China. The study hypothesizes that cultural factors, specified by family relations, along with demographic factors, number of diseases, economic well-being, and living conditions have a significant impact on subjects self-perceived health status. Pearson correlation, linear and logistic regression analyses are performed. Results indicate that age, number of diseases, perceived family respect, neighborhood relations, and percentage of income spent on rent are significant predictors of self-perceived physical health. These same factors plus preference to live with a son and personal monthly income are significant predictors of self-perceived mental health. Socio-cultural implications of these findings are examined.

Massage-like stroking induces acute antinociceptive effects that can be reversed by an oxytocin antagonist, indicating activation of oxytocin on endogenous pain controlling systems. We now demonstrate an increase in hindpaw withdrawal latencies (HWLs), in response to thermal and mechanical stimuli, which was present after six treatments of massage-like stroking every other day and which continued to increase through the remaining seven treatments. Repeated massage-like stroking also resulted in increased oxytocin-like immunoreactivity (oxytocin-LI) levels in plasma and periaquaductal grey matter (PAG). Furthermore, increases in HWLs were also present after injections of oxytocin into the PAG (0.1, 0.5 and 1.0 nmol). Intra-PAG oxytocin injection of 1 nmol followed by 1 or 20 nmol of naloxone attenuated the increments in HWL. Also, there was a dose-dependent attenuation of the oxytocin-induced antinociceptive effects following intra-PAG injection of the µ-opioid antagonist β-funaltrexamine (β-FNA) and the κ-opioid antagonist nor-binaltorphimine (nor-BNI) but not the δ-antagonist naltrindole. The long-term antinociceptive effects of massage-like stroking may be attributed, at least partly, to the oxytocinergic system and its interaction with the opioid system, especially the µ- and the κ-receptors in the PAG.

The present study was performed in rats with experimental mononeuropathy after left common sciatic nerve constriction. A bilateral decrease in hindpaw withdrawal latency to thermal and mechanical stimulation was observed after unilateral ligation of the left common sciatic nerve; however, it was more pronounced on the lesioned side. Compared with sham-operated rats, the content of calcitonin gene-related peptide-like immunoreactivity was significantly decreased in the left dorsal horn of the spinal cord and left dorsal root ganglia in rats with mononeuropathy. Blocking the receptor of calcitonin gene-related peptide, by intrathecal injection of 5 or 10 nmol of calcitonin gene-related peptide (8–37), induced a significant bilateral increase in hindpaw withdrawal latency to both thermal and mechanical stimulation which, however, was significantly less pronounced in mononeuropathic rats than in intact rats. The effect of calcitonin gene-related peptide (8–37) was reversed by intrathecal administration of the opioid antagonist naloxone. The contribution of calcitonin gene-related peptide and its receptors to transmission of presumed nociceptive information appears to be reduced in the sciatic nerve constriction model. The decrease in reflex responsiveness induced by calcitonin gene-related peptide (8–37) was counteracted by naloxone, indicating that opioids control the net effect of excitation in the spinal cord circuitry induced by calcitonin gene-related peptide and possibly other co-released neurotransmitters.

A missense mutation (A385 to T), predicting an Ile129 to Phe substitution, in the human Secretor alpha 1,2-fucosyltransferase gene was present in double dose in Lewis(a+b+) individuals, but not in Lewis(a-b+) individuals. Co-segregation of the Lewis(a+b+) phenotype with homozygosity for the mutation was also verified. These results yield a potential molecular basis for the weak Secretor allele (Sew) accounting for the Lewis(a+b+) phenotype.

Bone marrow transplantation has become an important therapeutic strategy in the treatment of pediatric malignancies and certain congenital disorders. We initiated a bone marrow transplant program at Children's Hospital in New Orleans at the start of January 1989. Both allogeneic and autologous bone marrow transplantations have been performed. To date, a total of 20 children have received transplants for a variety of diseases. Their ages ranged from 17 months to 20 years. The overall 2-year survival is 40%. For the good-risk patients, the projected 2-year disease-free survival is 85%, compared to 30% for the poor-risk patients. Regimen-related mortality has been extremely low in our settings primarily due to improvement of available supportive care. We conclude that bone marrow transplantation offers a potential cure for these otherwise fatal diseases.

1. Drugs were administered to a suprachiasmatic nucleus through a chronically implanted cannula at the second day of a torpor bout of hibernating ground squirrels. After naltrexone injection, the body temperature of the hibernating animals increased and they aroused from hibernation within 20 hr after the injection. 2. Further experiments show that intra-suprachiasmatic nucleus perfusion of 1 nmol of ICI 174864 or nor-BNI, not beta-FNA, were able to increase the body temperature of hibernating ground squirrels and aroused them from hibernation within 20 hr after the injection.
