Abstract

Cancer is a leading cause of death worldwide. Glioblastoma (GBM) is a major challenge in oncology due to its highly invasive nature and limited treatment options. GBM's aggressive migration beyond tumor margins and rapid tumor growth hinders success in patient treatment. Localized therapeutic delivery, such as the use of transition metals like copper, is highlighted as a novel therapeutic agent for many potential biomedical applications. Herein, it is aimed to study the effects of Cu release on the proliferation and invasiveness of cancer cells. To this end, novel copper‐based nanostructures with different release patterns are designed. Using a complex 3D cell culture model to mimic the tumor microenvironment, it is shown that different patterns of copper ion release have a strong impact on GBM progression and invasiveness. The findings highlight the importance of optimizing localized copper release patterns to tailor different tumor treatment strategies. They also show the potential and suitability of 3D microchips as instruments to study the behavior of tumor spheroids. In spite of their limitations, these 3D microdevices enable a controlled and close monitoring of the influence of environmental factors (such as the presence of Cu ions) on the proliferation and invasiveness of the cells, with a better approach to reality compared to 2D models and with a more controlled environment, compared to an in vivo model.

6. Gold-Platinum Nanoparticles with Core-Shell Configuration as Efficient Oxidase-like Nanosensors for Glutathione Detection

Abstract

Nanozymes, defined as nanomaterials that can mimic the catalytic activity of natural enzymes, have been widely used to develop analytical tools for biosensing. In this regard, the monitoring of glutathione (GSH), a key antioxidant biomolecule intervening in the regulation of the oxidative stress level of cells or related with Parkinson’s or mitochondrial diseases can be of great interest from the biomedical point of view. In this work, we have synthetized a gold-platinum Au@Pt nanoparticle with core-shell configuration exhibiting a remarkable oxidase-like mimicking activity towards the substrates 3,3′,5,5′-tetramethylbenzidine (TMB) and o-phenylenediamine (OPD). The presence of a thiol group (-SH) in the chemical structure of GSH can bind to the Au@Pt nanozyme surface to hamper the activation of O2 and reducing its oxidase-like activity as a function of the concentration of GSH. Herein, we exploit the loss of activity to develop an analytical methodology able to detect and quantify GSH up to µM levels. The system composed by Au@Pt and TMB demonstrates a good linear range between 0.1–1.0 µM to detect GSH levels with a limit of detection (LoD) of 34 nM.

Abstract

There are four principles in evolution: variation, inheritance, selection and time. These principles summarizes natural selection. In the same way, sexual selection can be defined as the evolution of certain conspicuous physical traits, such as pronounced coloration, increased size, or striking adornments that may grant the possessors of these traits greater success in obtaining mates. Both concepts join on time in the branch termed evolutionary biology (EB). Current EB since Darwin had a primer at the end of the past century in the Trivers-Wilard’s hypothesis based on the relationships of parents’ effort with the condition of offspring. This hypothesis has required new brands termed “Sex-allocation“, “Parental Investment”, “Delayed Maturation” and “Sexy-sons” to conform the framework of the current EB. Here, I try to summarize the key concepts of these theories trying to bring the theme towards birds.

10. Enzyme–iron oxide nanoassemblies: a review of immobilization and biocatalytic applications

Abstract

In the search for new biotechnological advances, increasing attention is currently being paid to the development of magnetic nanoplatforms loaded with enzymes, since, on the one hand, they can be recovered and reused, and on the other hand, they improve their catalytic activity and increase their stability, avoiding processes such as aggregation or autolysis. In this review, we evaluate a series of key parameters governing the enzyme–nanoparticle immobilization phenomena from a thermodynamic and kinetic point of view. We also focus on the use of magnetite nanoparticles (MNPs) as multifunctional vectors able to anchor enzymes, summarize the most relevant aspects of functionalization and immobilization and, finally, describe some recent and relevant applications of the enzyme–MNP hybrids as biocatalysts with especial emphasis on cancer therapy.

Abstract

Platinum nanoparticles (Pt NPs) have a well-established role as a classic heterogeneous catalyst. Also, Pt has traditionally been employed as a component of organometallic drug formulations for chemotherapy. However, a new role in cancer therapy is emerging thanks to its outstanding catalytic properties, enabling novel approaches that are surveyed in this review. Herein, we critically discuss results already obtained and attempt to ascertain future perspectives for Pt NPs as catalysts able to modify key processes taking place in the tumour microenvironment (TME). In addition, we explore relevant parameters affecting the cytotoxicity, biodistribution and clearance of Pt nanosystems. We also analyze pros and cons in terms of biocompatibility and potential synergies that emerge from combining the catalytic capabilities of Pt with other agents such as co-catalysts, external energy sources (near-infrared light, X-ray, electric currents) and conventional therapies., The authors gratefully acknowledge funding from the European Commission (ERC Advanced Grant-742684 CADENCE).

14. Water reservoirs dynamics do not affect the abundance and productivity of the bearded tit (Panurus biarmicus) in 'El Hondo Natural Park' (SE Iberia)

Abstract

Long-term fluctuations of the abundance and productivity of marshland passerines are affected by multiple factors. Water levels fluctuations in man-made wetlands can affect severely the breeding and wintering attempts of highly dependent reed passerines and decrease their abundances simply by dispersal towards better sites. In this work I study if water outputs and inputs in a western Mediterranean coastal semi-arid reservoir “El Hondo Natural Park” do not affect, apparently, the long-term abundances and productivity of the small Spanish endangered population of Bearded Tit Panurusbiarmicus ubicated in it. The watershed parameters analyzed during sixteen years (1992-2007) by a Principal Components Analysis (PCA) show that only two main axes, water inputs (PC1) and outputs (PC2) explained most total variance and were the drivers of the water in the reservoirs. None of the bird’s dynamics indexes were significantly correlated with the axes, so that water dynamics seem have not taken in to account for the fluctuations of this species, but the flooding of thereservoir with output capacity “Embalse de Poniente” could likely influence the annual productivity of this species, due to the negative but unsignificant correlation with PC2. The maintenance of ponds of enough water quantity and quality (e.g. “La Raja”, “Charca Sur”) in the “El Hondo Natural Park” during the nesting periods is fundamental for the preservation of this endangered population. The results indicate that further evaluations of the key biotic and abiotic factors who drive this threatened Bearded Tit’s population in this isolated and endangered man-made wetland are needed.

Abstract

This article belongs to the Special Issue 10th Anniversary of Catalysts—Feature Papers in Photocatalysis., The present review aims at highlighting recent advances in the development of photocatalysts devoted to cancer therapy applications. We pay especial attention to the engineering aspects of different nanomaterials including inorganic semiconductors, organic-based nanostructures, noble metal-based systems or synergistic hybrid heterostructures. Furthermore, we also explore and correlate structural and optical properties with their photocatalytic capability to successfully performing in cancer-related therapies. We have made an especial emphasis to introduce current alternatives to organic photosensitizers (PSs) in photodynamic therapy (PDT), where the effective generation of reactive oxidative species (ROS) is pivotal to boost the efficacy of the treatment. We also overview current efforts in other photocatalytic strategies to tackle cancer based on photothermal treatment, starvation therapy, oxidative stress unbalance via glutathione (GSH) depletion, biorthogonal catalysis or local relief of hypoxic conditions in tumor microenvironments (TME)., This research was funded by the European Research Council (ERC) through an Advanced Research Grant (CADENCE, grant number 742684). The strong size and morphology dependence of metallic nanostructures to tune their visible to near-infrared (vis-NIR) light harvesting capabilities has been combined with the design of a wide variety of architectures for the semiconductor supports to promote the selective activity of specific crystallographic facets. The search for efficient heterojunctions has been subjected to numerous studies, especially those involving gold nanostructures and titania semiconductors. In the present review, we paid special attention to the most recent advances in the design of gold-semiconductor hetero-nanostructures including emerging metal oxides such as cerium oxide or copper oxide (CeO2 or Cu2O) or metal chalcogenides such as copper sulfide or cadmium sulfides (CuS or CdS). These alternative hybrid materials were thoroughly built in past years to target research fields of strong impact, such as solar energy conversion, water splitting, environmental chemistry, or nanomedicine. Herein, we evaluate the influence of tuning the morphologies of the plasmonic gold nanostructures or the semiconductor interacting structures, and how these variations in geometry, either individual or combined, have a significant influence on the final photocatalytic performance.

24. Sperm DNA integrity and meiotic behavior assessment in an infertile male carrier of a 9qh+++ polymorphism

Abstract

1. Introduction In the last few years, some papers have reported a high incidence of heterochromatin variants in infertile men [1-3]. Paracentric heterochromatin variants usually occur on the long arms [...], Although several reports on male infertility suggest a relationship between chromosome 9 polymorphisms and infertility, the effects on the phenotype have not been extensively reported. In this study, an infertile patient was found to carry a 9qh+++ chromosome. The flow cytometric TUNEL assay and SCD test have been applied to characterize sperm DNA integrity. In order to assess its meiotic behaviour, synapsis, recombination, and aneuploidy, analyses have been also performed. Sperm DNA fragmentation (SDF) was 77.81% and 87% for the TUNEL and SCD tests, respectively. Ninety-two percent of pachytene cells analyzed showed meiotic abnormalities. The mean number of MLH1 foci per pachytene in the control group was higher (49) than the mean found in the 9qh+++ patient (38) (P < .0001). In spermatozoa, significant increases of disomy rates were observed for chromosome 18 and for the sex chromosomes (P < .0001). These disturbances could be present in other male carriers of a less marked 9qh+.

Abstract

This article summarizes the situation of the Bearded Tit (Panurus biarmicus) in Iberia on the basis of the literature found in some Spanish Journals, books and own investigations. This is a species few studied in the centre of Spain, on the bulk of populations are, so furthermore investigations are needed from these areas.

Abstract

Doing this, researchers have developed complex sophisticated materials with properties, in some cases, similar to those found in nature. Of special interest is the development of scaffolds able to guide bone regeneration. The bone tissue comprises an organic matrix composed of aligned collagen fibers containing nanoapatite crystals oriented along the fiber direction. During bone mineralization, both processes, the self-assembly of collagen fibrils and mineralization occur simultaneously. Collagen fibers are able to control calcium phosphate nucleation and subsequent apatite crystal growth at a very limited range of collagen density and ionic concentration. In this study, we reproduced the simultaneity of both processes using an artificial peptide fluorenylmethoxycarbonyl-diphenylalanine (Fmoc-FF) that has the ability to self-assemble in water after the addition of Ca2+ ions. Therefore, the peptide self-assembly process and the mineralization of apatite are Ca-demanding processes and occur simultaneously. The role of peptide and ionic concentrations has been investigated affording organic/inorganic hybrid hydrogels with different degrees of homogeneity and mineralization.
