For most CO2 and CH4 satellites, only a small percentage (∼10%) of observations yield successful retrievals, with the remaining ∼90% rejected, primarily due to the effects of clouds. Discarding this large fraction of data is an inefficient strategy worth reconsidering due to the costs involved in developing, launching and operating the satellites to make these observations. However, if real-time cloud data are available together with pointing capability, cloud data can guide the instrument pointing in an “intelligent pointing” strategy for cloud avoidance. In this work, multiple intelligent pointing simulations were conducted, demonstrating the significant advantages of this approach for satellites in a highly elliptical orbit (HEO), from which nearly the whole Earth disk can be observed. Multiple factors are shown to contribute to intelligent pointing efficiency such as the size and shape (or aspect ratio) of the field of view (FOV). For the current baseline orbit and Imaging Fourier Transform Spectrometer (IFTS) observing characteristics for the proposed Arctic Observing Mission (AOM), the monthly fraction of cloud-free observations is roughly a factor of 2 (ranging from ∼1.5–2.5) more than obtained with standard pointing (in which cloud information is not used). A similar efficiency is expected in a geostationary orbit (GEO) with an IFTS, however, for a dispersive instrument in HEO or GEO, the gain is more modest. This result is primarily attributed to the ∼1:1 aspect ratio of the IFTS FOV, since it is more efficient for cloud avoidance and scanning irregularly-shaped land masses than the long and narrow slit projection of a typical dispersive spectrometer. These results have implications for the design of future CO2 or CH4 monitoring satellites and constellation architectures, as well as other fields of satellite earth observation in which clouds significantly impact observations.

Abstract An idealized aquaplanet moist global atmospheric model with realistic radiative transfer but no clouds and no convective parameterization is found to possess multiple climate equilibria. When forced symmetrically about the equator, in some cases the Inter Tropical Convergence Zone (ITCZ) migrates to an off‐equatorial equilibrium position. Mechanism denial experiments prescribing relative humidity imply that radiation‐circulation coupling is essential to this instability. The cross‐equatorial asymmetry occurs only when the underlying slab ocean is sufficiently deep and the atmosphere's spectral dynamical core is sufficiently coarse (∼T170 or less with our control parameters). At higher resolutions, initializing with an asymmetric state indicates metastability with very slow (thousands of days) return to hemispheric symmetry. There is some sensitivity to the model timestep, which affects the time required to transition to the asymmetric state, with little effect on the equilibrium climate. The instability is enhanced when the planetary boundary layer scheme favors deeper layers or by a prescribed meridional heat transport away from the equator within the slab. The instability is not present when the model is run with a convective parameterization scheme commonly utilized in idealized moist models. We argue that the instability occurs when the asymmetric heating associated with a spontaneous ITCZ shift drives a circulation that rises poleward of the perturbed ITCZ. These results serve as a warning of the potential for instability and non‐uniqueness of climate that may complicate studies with idealized models of the tropical response to perturbations in forcing.

Using co-located space-based measurements of carbon dioxide (CO2) from the Orbiting Carbon Observatory-2 and Orbiting Carbon Observatory-3 (OCO-2/3) and carbon monoxide (CO) and nitrogen dioxide (NO2) from the TROPOspheric Monitoring Instrument (TROPOMI), we calculate total column enhancements for observations influenced by anthropogenic emissions from urban regions relative to clean background values. We apply this method to observations taken over or downwind of 27 large (population of >1 million) urban areas from around the world. Enhancement ratios between species are calculated and compared to emissions ratios derived from four globally gridded anthropogenic emissions inventories. We find that these global inventories underestimate CO emissions in many North American and European cities relative to our observed enhancement ratios, while smaller differences were found for NO2 emissions. We further demonstrate that the calculation and intercomparison of enhancement ratios of multiple tracers can help to identify the underlying biases leading to disagreement between observations and inventories. Additionally, we use high-resolution CO2 inventories for two cities (Los Angeles and Indianapolis) to estimate emissions of CO and NO2 using our calculated enhancement ratios and find good agreement with both a previous modelling study for the megacity of Los Angeles and California Air Resources Board (CARB) inventory estimates.

Abstract Diverse geographic, environmental, and ecological factors affect gene flow and adaptive genomic variation within species. With recent advances in landscape ecological modelling and high‐throughput DNA sequencing, it is now possible to effectively quantify and partition their relative contributions. Here, we use landscape genomics to identify determinants of genomic differentiation in the forest tent caterpillar, Malacosoma disstria, a widespread and irruptive pest of numerous deciduous tree species in North America. We collected larvae from multiple populations across Eastern Canada, where the species experiences a diversity of environmental gradients and feeds on a number of different host tree species, including trembling aspen (Populus tremuloides), sugar maple (Acer saccharum), red oak (Quercus rubra), and white birch (Betula papyrifera). Using a combination of reciprocal causal modelling (RCM) and distance‐based redundancy analyses (dbRDA), we show that differentiation of thousands of genome‐wide single nucleotide polymorphisms (SNPs) among individuals is best explained by a combination of isolation by distance, isolation by environment (spatial variation in summer temperatures and length of the growing season), and differences in host association. Configuration of suitable habitat inferred from ecological niche models was not significantly related to genomic differentiation, suggesting that M. disstria dispersal is agnostic with respect to habitat quality. Although population structure was not discretely related to host association, our modelling framework provides the first molecular evidence of host‐associated differentiation in M. disstria, congruent with previous documentation of reduced growth and survival of larvae moved between natal host species. We conclude that ecologically mediated selection is contributing to variation within M. disstria, and that divergent adaptation related to both environmental conditions and host association should be considered in ongoing research and management of this important forest pest.

Abstract Temporal separation of reproductive timing can contribute to species diversification both through allochronic speciation and later reinforcement of species boundaries. Such phenological differences are an enigmatic component of evolutionary divergence between two major forest defoliator species of the spruce budworm complex: Choristoneura fumiferana and C. occidentalis. While these species interbreed freely in laboratory settings, natural hybridization rates have not been reliably quantified due to their indistinguishable morphology. To assess whether temporal isolation is contributing to reproductive isolation, we collected adult individuals throughout their expected zone of sympatry in western Canada at 10‐day intervals over two successive years, assigning taxonomic identities using thousands of single nucleotide polymorphisms. We found unexpectedly broad sympatry between C. fumiferana and C. occidentalis biennis and substantial overlap of regional flight periods. However, flight period divergence was much more apparent on a location‐by‐location basis, highlighting the importance of considering spatial scale in these analyses. Phenological comparisons were further complicated by the biennial life cycle of C. o. biennis, the main subspecies of C. occidentalis in the region, and the occasional occurrence of the annually breeding subspecies C. o. occidentalis. Nonetheless, we demonstrate that biennialism is not a likely contributor to reproductive isolation within the species complex. Overall, interspecific F1 hybrids comprised 2.9% of sequenced individuals, confirming the genomic distinctiveness of C. fumiferana and C. occidentalis, while also showing incomplete reproductive isolation of lineages. Finally, we used FST‐based outlier and genotype–environment association analyses to identify several genomic regions under putative divergent selection. These regions were disproportionately located on the Z linkage region of C. fumiferana, and contained genes, particularly antifreeze proteins, that are likely to be associated with overwintering success and diapause. In addition to temporal isolation, we conclude that other mechanisms, including ecologically mediated selection, are contributing to evolutionary divergence within the spruce budworm species complex.

The pedestrian-induced oscillation of the London Millennium Bridge is considered as an example of emerging synchronisation. Belykh et al. provide an alternative mechanism for emergence of coherent oscillatory bridge dynamics where synchrony is a consequence, not the cause, of the instability.

Although therapeutically efficacious, ipilimumab can exhibit dose-limiting toxicity that prevents maximal efficacious clinical outcomes and can lead to discontinuation of treatment. We hypothesized that an acidic pH-selective ipilimumab (pH Ipi), which preferentially and reversibly targets the acidic tumor microenvironment over the neutral periphery, may have a more favorable therapeutic index. While ipilimumab has pH-independent CTLA-4 affinity, pH Ipi variants have been engineered to have up to 50-fold enhanced affinity to CTLA-4 at pH 6.0 compared to pH 7.4. In hCTLA-4 knock-in mice, these variants have maintained anti-tumor activity and reduced peripheral activation, a surrogate marker for toxicity. pH-sensitive therapeutic antibodies may be a differentiating paradigm and a novel modality for enhanced tumor targeting and improved safety profiles.

Abstract Microstructure profiling was utilized to estimate vertical mixing (via vertical turbulent buoyancy flux) during a tidal pulse in the interior Merrimack River plume in calm winds. Multiple stratified shear mixing regimes appear and evolve with time. Initially the plume acts as a nearfield jet, with mixing in the plume (plume layer mixing) and over the plume‐ambient interface (nearfield interfacial mixing). As the plume grows, interfacial mixing is suppressed offshore of the nearfield as currents slow, diminishing turbulent exchange between plume and shelf. At the end of ebb, ambient tidal currents reverse direction below plume, initiating another mode of internal, interfacial mixing (coined here as tidal interfacial mixing), allowing exchange between plume and ambient waters offshore. This work highlights previously unreported tidally modulated mixing within the near and midfield of a river plume.

The study of orbital resonances allows for the constraint of planetary properties of compact systems. We can predict a system’s resonances by observing the orbital periods of the planets, as planets in or near mean motion resonance (MMR) have period ratios that reduce to a ratio of small numbers. However, a period ratio near commensurability does not guarantee a resonance; we must study the system’s dynamics and resonant angles to confirm resonance. Because resonances require in-depth study to confirm, and because two-body resonances require a measurement of the eccentricity vector which is quite challenging, very few resonant pairs or chains have been confirmed. We thus remain in the era of small-number statistics, not yet able to perform large population synthesis or informatics studies. To address this problem, we build a python package to find, confirm, and analyze MMRs, primarily through N -body simulations. We then analyze all near-resonant planets in the Kepler/K2 and TESS catalogs, confirming over 60 new resonant pairs and various new resonant chains. We additionally demonstrate the package’s functionality and potential by characterizing the mass–eccentricity degeneracy of Kepler-80g, exploring the likelihood of an exterior giant planet in Kepler-80, and constraining the masses of planets in Kepler-305. We find that our methods overestimate the libration amplitudes of the resonant angles and struggle to confirm resonances in systems with more than three planets. We identify various systems that are likely resonant chains but that we are unable to confirm, and highlight next steps for exoplanetary resonances.

Although resonant planets have orbital periods near commensurability, resonance is also dictated by other factors, such as the planets’ eccentricities and masses, and therefore must be confirmed through a study of the system’s dynamics. Here, we perform such a study for five multiplanet systems: Kepler-226, Kepler-254, Kepler-363, Kepler-1542, and K2-32. For each system, we run a suite of N -body simulations that span the full parameter space that is consistent with the constrained orbital and planetary properties. We study the stability of each system and look for resonances based on the libration of the critical resonant angles. We find strong evidence for a two-body resonance in each system; we confirm a 3:2 resonance between Kepler-226c and Kepler-226d, confirm a 3:2 resonance between Kepler-254c and Kepler-254d, and confirm a three-body 1:2:3 resonant chain between the three planets of Kepler-363. We explore the dynamical history of two of these systems and find that these resonances most likely formed without migration. Migration leads to the libration of the three-body resonant angle, but these angles circulate in both Kepler-254 and Kepler-363. Applying our methods to additional near-resonant systems could help us identify which systems are truly resonant or nonresonant and which systems require additional follow-up analysis.

Agrowing avenue for determining the prevalence of life beyond Earth is to search for “technosignatures” from extraterrestrial intelligences/agents. Technosignatures require significant energy to be visible across interstellar space and thus intentional signals might be concentrated in frequency, in time, or in space, to be found in mutually obvious places. Therefore, it could be advantageous to search for technosignatures in parts of parameter space that are mutually derivable to an observer on Earth and a distant transmitter. In this work, we used the L -band (1.1–1.9 GHz) receiver on the Robert C. Byrd Green Bank Telescope to perform the first technosignature search presynchronized with exoplanet transits, covering 12 Kepler systems. We used the Breakthrough Listen turboSETI pipeline to flag narrowband hits (∼3 Hz) using a maximum drift rate of ±614.4 Hz s ^−1 and a signal-to-noise threshold of 5—the pipeline returned ∼3.4 × 10 ^5 apparently-localized features. Visual inspection by a team of citizen scientists ruled out 99.6% of them. Further analysis found two signals of interest that warrant follow up, but no technosignatures. If the signals of interest are not redetected in future work, it will imply that the 12 targets in the search are not producing transit-aligned signals from 1.1 to 1.9 GHz with transmitter powers >60 times that of the former Arecibo radar. This search debuts a range of innovative technosignature techniques: citizen science vetting of potential signals of interest, a sensitivity-aware search out to extremely high drift rates, a more flexible method of analyzing on-off cadences, and an extremely low signal-to-noise threshold.

The Connecticut River plume interacts with the strong tidal currents of the ambient receiving waters in eastern Long Island Sound. The plume formed during ambient flood tides is studied as an example of tidal river plumes entering into energetic ambient tidal environments in estuaries or continental shelves. Conservative passive freshwater tracers within a high-resolution nested hydrodynamic model are applied to determine how source waters from different parts of the tidal cycle contribute to plume composition and interact with bounding plume fronts. The connection to source waters can be cut off only under low-discharge conditions, when tides reverse surface flow through the mouth after max ambient flood. Upstream plume extent is limited because ambient tidal currents arrest the opposing plume propagation, as the tidal internal Froude number exceeds one. The downstream extent of the tidal plume always is within 20 km from the mouth, which is less than twice the ambient tidal excursion. Freshwaters in the river during the preceding ambient ebb are the oldest found in the new flood plume. Connectivity with source waters and plume fronts exhibits a strong upstream-to-downstream asymmetry. The arrested upstream front has high connectivity, as all freshwaters exiting the mouth immediately interact with this boundary. The downstream plume front has the lowest overall connectivity, as interaction is limited to the oldest waters since younger interior waters do not overtake this front. The offshore front and inshore boundary exhibit a downstream progression from younger to older waters and decreasing overall connectivity with source waters. Plume-averaged freshwater tracer concentrations and variances both exhibit an initial growth period followed by a longer decay period for the remainder of the tidal period. The plume-averaged tracer variance is increased by mouth inputs, decreased by entrainment, and destroyed by internal mixing. Peak entrainment velocities for younger waters are higher than values for older waters, indicating stronger entrainment closer to the mouth. Entrainment and mixing time scales (1–4 h at max ambient flood) are both shorter than half a tidal period, indicating entrainment and mixing are vigorous enough to rapidly diminish tracer variance within the plume.

AIM-North is a proposed satellite mission that would provide observations of unprecedented frequency and density for monitoring northern greenhouse gases (GHGs), air quality (AQ) and vegetation. AIM-North would consist of two satellites in a highly elliptical orbit formation, observing over land from ∼40°N to 80°N multiple times per day. Each satellite would carry a near-infrared to shortwave infrared imaging spectrometer for CO2, CH4, and CO, and an ultraviolet-visible imaging spectrometer for air quality. Both instruments would measure solar-induced fluorescence from vegetation. A cloud imager would make near-real-time observations, which could inform the pointing of the other instruments to focus only on the clearest regions. Multiple geostationary (GEO) AQ and GHG satellites are planned for the 2020s, but they will lack coverage of northern regions like the Arctic. AIM-North would address this gap with quasi-geostationary observations of the North and overlap with GEO coverage to facilitate intercomparison and fusion of these datasets. The resulting data would improve our ability to forecast northern air quality and quantify fluxes of GHG and AQ species from forests, permafrost, biomass burning and anthropogenic activity, furthering our scientific understanding of these processes and supporting environmental policy.

All measurements of XCO2 from space have systematic errors. To reduce a large fraction of these errors, a bias correction is applied to XCO2 retrieved from GOSAT and OCO-2 spectra using the ACOS retrieval algorithm. The bias correction uses, among other parameters, the surface pressure difference between the retrieval and the meteorological reanalysis. Relative errors in the surface pressure estimates, however, propagate nearly 1:1 into relative errors in bias-corrected XCO2. For OCO-2, small errors in the knowledge of the pointing of the observatory (up to ∼ 130 arcsec) introduce a bias in XCO2 in regions with rough topography. Erroneous surface pressure estimates are also caused by a coding error in ACOS version 8, sampling meteorological analyses at wrong times (up to 3 h after the overpass time). Here, we derive new geolocations for OCO-2's eight footprints and show how using improved knowledge of surface pressure estimates in the bias correction reduces errors in OCO-2's v9 XCO2 data.

The climate regime shift during the 1980s had a substantial impact on the terrestrial ecosystems and vegetation at different scales. However, the mechanisms driving vegetation changes, before and after the shift, remain unclear. In this study, we used a biophysical dynamic vegetation model to estimate large-scale trends in terms of carbon fixation, vegetation growth, and expansion during the period 1958–2007, and to attribute these changes to environmental drivers including elevated atmospheric CO2 concentration (hereafter eCO2), global warming, and climate variability (hereafter CV). Simulated leaf area index (LAI) and gross primary production (GPP) were evaluated against observation-based data. Significant spatial correlations are found (correlations > 0.87), along with regionally varying temporal correlations of 0.34–0.80 for LAI and 0.45–0.83 for GPP. More than 40 % of the global land area shows significant positive (increase) or negative (decrease) trends in LAI and GPP during 1958–2007. Regions over the globe show different characteristics in terms of ecosystem trends before and after the 1980s. While 11.7 % and 19.3 % of land have had consistently positive LAI and GPP trends, respectively, since 1958, 17.1 % and 20.1 % of land saw LAI and GPP trends, respectively, reverse during the 1980s. Vegetation fraction cover (FRAC) trends, representing vegetation expansion and/or shrinking, are found at the edges of semi-arid areas and polar areas. Environmental drivers affect the change in ecosystem trend over different regions. Overall, eCO2 consistently contributes to positive LAI and GPP trends in the tropics. Global warming mostly affects LAI, with positive effects in high latitudes and negative effects in subtropical semi-arid areas. CV is found to dominate the variability of FRAC, LAI, and GPP in the semi-humid and semi-arid areas. The eCO2 and global warming effects increased after the 1980s, while the CV effect reversed during the 1980s. In addition, plant competition is shown to have played an important role in determining which driver dominated the regional trends. This paper presents new insight into ecosystem variability and changes in the varying climate since the 1950s.

Abstract Background Researchers are required to seek consent from Indigenous communities prior to conducting research but there is inadequate information about how Indigenous people understand and become fully engaged with this consent process. Few studies evaluate the preference or understanding of the consent process for research with Indigenous populations. Lack of informed consent can impact on research findings. Methods The Picture Talk Project was initiated with senior Aboriginal leaders of the Fitzroy Valley community situated in the far north of Western Australia. Aboriginal people were interviewed about their understanding and experiences of research and consent processes. Transcripts were analysed using NVivo10 software with an integrated method of inductive and deductive coding and based in grounded theory. Local Aboriginal interpreters validated coding. Major themes were defined and supporting quotes sourced. Results Interviews with Aboriginal leaders (n = 20) were facilitated by a local Aboriginal Community Navigator who could interpret if necessary and provide cultural guidance. Participants were from all four major local language groups of the Fitzroy Valley; aged 31 years and above; and half were male. Themes emerging from these discussions included Research—finding knowledge; Being respectful of Aboriginal people, Working on country, and Being flexible with time; Working together with good communication; Reciprocity—two-way learning; and Reaching consent. Conclusion The project revealed how much more there is to be learned about how research with remote Aboriginal communities should be conducted such that it is both culturally respectful and, importantly, meaningful for participants. We identify important elements in community consultation about research and seeking consent.

Objective: Kinase Suppressor of Ras 2 (KSR2) is a molecular scaffold coordinating Raf/MEK/ERK signaling that is expressed at high levels in the brain. KSR2 disruption in humans and mice causes obesity and insulin resistance. Understanding the anatomical location and mechanism of KSR2 function should lead to a better understanding of physiological regulation over energy balance. Methods: Mice bearing floxed alleles of KSR2 (KSR2fl/fl) were crossed with mice expressing the Cre recombinase expressed by the Nestin promoter (Nes-Cre) to produce Nes-CreKSR2fl/fl mice. Growth, body composition, food consumption, cold tolerance, insulin and free fatty acid levels, glucose, and AICAR tolerance were measured in gender and age matched KSR2−/− mice Results: Nes-CreKSR2fl/fl mice lack detectable levels of KSR2 in the brain. The growth and onset of obesity of Nes-CreKSR2fl/fl mice parallel those observed in KSR2−/− mice. As in KSR2−/− mice, Nes-CreKSR2fl/fl are glucose intolerant with elevated fasting and cold intolerance. Male Nes-CreKSR2fl/fl mice are hyperphagic, but female Nes-CreKSR2fl/fl mice are not. Unlike KSR2−/− mice, Nes-CreKSR2fl/fl mice respond normally to leptin and AICAR, which may explain why the degree of obesity of adult Nes-CreKSR2fl/fl mice is not as severe as that observed in KSR2−/− animals. Conclusions: These observations suggest that, in the brain, KSR2 regulates energy balance via control of feeding behavior and adaptive thermogenesis, while a second KSR2-dependent mechanism, functioning through one or more other tissues, modulates sensitivity to leptin and activators of the energy sensor AMPK.
