Abstract

Abstract Background Phytophthora capsici root rot (PRR) is a disastrous disease in peppers (Capsicum spp.) caused by soilborne oomycete with typical symptoms of necrosis and constriction at the basal stem and consequent plant wilting. Most studies on the QTL mapping of P. capsici resistance suggested a consensus broad-spectrum QTL on chromosome 5 named Pc.5.1 regardless of P. capsici isolates and resistant resources. In addition, all these reports proposed NBS-ARC domain genes as candidate genes controlling resistance. Results We screened out 10 PRR-resistant resources from 160 Capsicum germplasm and inspected the response of locus Pc.5.1 and NBS-ARC genes during P. capsici infection by comparing the root transcriptomes of resistant pepper 305R and susceptible pepper 372S. To dissect the structure of Pc.5.1, we anchored genetic markers onto pepper genomic sequence and made an extended Pc5.1 (Ext-Pc5.1) located at 8.35 Mb–38.13 Mb on chromosome 5 which covered all Pc5.1 reported in publications. A total of 571 NBS-ARC genes were mined from the genome of pepper CM334 and 34 genes were significantly affected by P. capsici infection in either 305R or 372S. Only 5 inducible NBS-ARC genes had LRR domains and none of them was positioned at Ext-Pc5.1. Ext-Pc5.1 did show strong response to P. capsici infection and there were a total of 44 differentially expressed genes (DEGs), but no candidate genes proposed by previous publications was included. Snakin-1 (SN1), a well-known antimicrobial peptide gene located at Pc5.1, was significantly decreased in 372S but not in 305R. Moreover, there was an impressive upregulation of sugar pathway genes in 305R, which was confirmed by metabolite analysis of roots. The biological processes of histone methylation, histone phosphorylation, DNA methylation, and nucleosome assembly were strongly activated in 305R but not in 372S, indicating an epigenetic-related defense mechanism. Conclusions Those NBS-ARC genes that were suggested to contribute to Pc5.1 in previous publications did not show any significant response in P. capsici infection and there were no significant differences of these genes in transcription levels between 305R and 372S. Other pathogen defense-related genes like SN1 might account for Pc5.1. Our study also proposed the important role of sugar and epigenetic regulation in the defense against P. capsici.

8. Transthoracic Echocardiography A Predictor of Complications in Patients with Severe Preeclampsia

Abstract

Background: Severe preeclampsia (PE) and eclampsia are a hypertensive disease of pregnancy associated with significant morbidity and mortality and require special monitoring. Lung ultrasonography and transthoracic echocardiography are a novel monitoring and diagnostic tools in intensive care and are widely used nowadays. Objective: To detect the role and value of transthoracic echocardiography (TTE) in postoperative monitoring in patients with severe PE. Patients and Methods: This was a cross sectional study of 55 patients with severe preeclampsia, 35 patients with eclampsia and 60 healthy controls. Transthoracic echocardiography was done to evaluate diastolic function using E/A ratio with continuous wave Doppler, ejection fraction using M mode in long axis parasternal view and inferior vena cava (IVC) diameter in subcostal view. All measurements were done postoperatively. Results: There was significant difference between the 3 groups regarding diastolic function (p value 0.0001). Abnormal diastolic function in severe PE, eclampsia and controls was 81.2%, 94.3% and 5% respectively. There was no difference between the 3 groups of the study as regard IVC diameter. There was significant difference between the 3 groups as regards EF (P=0.007), the mean of EF in severe PE, eclampsia and controls was 66.89 ± 5.54, 63.029 ± 8.45 and 66.43 ± 4.16 respectively. Conclusions: The use of transthoracic echocardiography is mandatory in postoperative monitoring in patients with severe PE and eclampsia as they can adjust management and detect complications. Restricted fluid therapy policy is indicated with the higher grades of diastolic dysfunction and lower ejection fraction (EF).

Abstract

A tissue culture experiment was conducted to attain the adventitious buds initiation and growth from Fragraia x ananassa cv. Festival in vitro leaves. The leaves were excited from shoot propagated in vitro and cultured on Murashige and Skoog (MS) medium supplemented with 4.50, 6.75, and 9.00 mmol l-1 thidiazuron (TDZ) for six weeks. Then explants were transferred to MS medium (salts and vitamins) free plant growth regulators for 8 weeks. This study included the histological and morphological responses of strawberry leaves with TDZ treatments in relation to some biochemical components. All TDZ treatments resulted in adventitious shoots development on leaf explants without callus formation. Growth in 6.75 mmol l-1 TDZ led to an increment in the most morphological parameters compared to the rest treatments. The results showed that 6.75 mmol l-1 of TDZ treatment significantly increases the number of shootlets per explant (5.0), total shoot (51.3), the number of leaves/explant (12) as compared with other treatments while the rest of parameters (the survival percentage, shoot length (cm), number of leaves/explant, root length (cm), and number of roots/explant) did not show any significant differences. Accumulation of chlorophylls, total sugars, and total soluble phenols was enhanced by 6.75 mmol l-1 of TDZ. The histological observations showed that adventitious shoots initiated from some parenchyma of ground tissue in the midvein zone of the explant, whereas the parenchyma mesophyll and the epidermal cells did not involve in this process. The ontogenesis of adventitious shoots was described by successive stages with the following distinguishable anatomical structures: meristemoids, bud primordium, shoot apex with leaf primordial, branching of adventitious shoots. After four weeks of culture, cell dedifferentiation was recorded in the midvein parenchyma and forming small groups of divided cells called meistemoid centres. These centres forming new meristematic masses embedded in the ground tissue of the main vein. After six weeks, further development of these masses resulted in the formation of bud primordial with normal organized shoot apical meristems and leaf primordial and arising from the explant as small protrusions. After two weeks from subculture on MS free TDZ, the adventitious shoots continue to elongate with forming new lateral branches and clearly observed on the adaxial side of the explant. This procedure provides a simple and rapid approach to regenerate strawberry plants via direct organogenesis.

Abstract

Background: Valproic acid (VPA) is a potent anticancer and antiangiogenic agent. However, design and synthesis of chemical derivatives with improved antiangiogenic and anticancer activities are still necessary. In this study a library of novel derivatives of VPA was synthesized and tested. Methods: A human liver cancer cell line (HepG2) and a human normal embryonic kidney cell line (HEK 293) were exposed to various concentrations of VPA derivatives for 24 hours and cell viability was checked by MTT colorimetric assay. Anti-angiogenic properties were evaluated in transgenic zebrafish embryos. Results: N-valproylglycine derivatives suppressed survival almost 70% (p value 0.001) in HepG2 cells but only 10-12% in HEK 293 cells (p value 0.133). They also suppressed angiogenic blood vessel formation by 80% when used between 2-20 µM in zebrafish embryos. Valproic acid hydrazides showed moderate level of anticancer activity by affecting 30-50% (p value 0.001) of cell viability in HepG2 cells and 8-10% in HEK293 cells (p value 0.034). Conclusion: The majority of compounds in this study showed potent and stronger antiangiogenic and anticancer activity than VPA. They proved selectively toxic to cancer cells and safer for normal cells. Moreover, these compounds inhibited developmental angiogenesis in zebrafish embryos. Based on the fact that liver is a highly vascularized organ, in case of liver carcinoma these compounds have the potential to target the pathological angiogenesis and could be an effective strategy to treat hepatocellular carcinoma.

Abstract

Three molecular markers, including start codon targeted (SCoT) polymorphism, directed amplification of minisatellite-region DNA polymerase chain reaction (DAMD-PCR), and inter simple sequence repeat (ISSR) markers, were compared in terms of their informativeness and efficiency for analysis of genetic relationships among 38 accessions of eight annual Cicer species. The results were as follows: (1) the highest level of detected polymorphism was observed for all three marker types; (2) the rate of diversity for the three marker techniques was approximately equal, and the correlation coefficients of similarity were statistically significant for all three marker systems; (3) the three molecular markers showed relatively similar phylogenetic grouping for examined species. Diversity analysis showed that Cicer reticulatum is the closest wild species to the cultivated chickpea, and this finding supports the hypothesis that C. reticulatum is the most probable progenitor of the cultivated species. C. bijugum, C. judaicum, and C. pinnatifidum were clustered together, and in other clusters C. yamashitae and C. cuneatum were grouped close together. To our knowledge, this is the first detailed comparison of performance among two targeted DNA region molecular markers (SCoT and DAMD-PCR) and the ISSR technique on a set of samples of Cicer. The results provide guidance for future efficient use of these molecular methods in genetic analysis of Cicer.

Abstract

The exploration and production activities of the natural gas in Abu Madi region (in middle delta) progressed in the last decade. The condensate hydrocarbon and formation water are bi‐products produced during the natural gas industry. The accompanied water contains 10–15% condensate (gasoline). This water with gasoline was drained into the Shehab El‐Din drain. The farmers were used this water to irrigate their fields. Great areas of the fields were out of order of planting as the result of gasoline pollution. This work concentrates on washing the polluted soil using locally prepared surfactant (LABSA‐Na) in the washing process. The soil sample was picked up from the Abu Madi field and was divided into pots (four pots for each treatment). The experiment was designed on the basis of polluting the soil by gasoline (condensate) from 1 to 10%. The washing process was carried out in presence of 0.1, 0.3, and 0.5% surfactant solutions. Abstract

Global shortage of freshwater is expected to reduce aquaculture activities that depend upon freshwaters. Mining the wild genetic resources of fish species can aid much the selection and development of new strains capable of withstanding unusual culturing conditions, such as tolerance to unusual salinities. Nile tilapia is a supreme international aquaculture species, with a limited degree of tolerance to brackish waters. In the current study, DNA barcoding for the Nile Tilapia Oreochromis niloticus , using the mitochondrial cytochrome oxidase subunit I gene, was carried out. Samples were collected from two locations at Manzala Lake in the North of Egypt (El-Serow, salinity 1‰; and Ashtoom El-Gamil, Salinity 20‰); and from Lake Nasser (salinity 1‰;) in the South of Egypt. Numbers, locations and phylogenetic interrelationships among these haplotypes were determined, appending to the comparison the world haplotypes available in GenBank database. The results indicated the main haplotype lineages in the area of study and the world. Also, a newly identified haplotype with natural tolerance to different salinities could be identified from both studied lakes. This can contribute much to the genetic enhancement of tilapia aquaculture using new parents with more capability to tolerate salinity differences, producing then pure or hybrid offsprings with further ameliorated salinity tolerance.

Abstract

Introduction: Flaxseed extract promotes bone health and possibly functioned in inducing bone regeneration process. SHED (stem cell from exfoliated deciduous teeth) has been considered as an alternative source of MSC in tissue engineering because of their potential to differentiate into multiple cell-lineages including osteoblast. Although flaxseed extract has been shown to affect bone formation process, its effect in inducing stem cell into osteoblast lineage has not been reported. Objectives: To determine the effects of flaxseed extract on osteoblast differentiation potential of SHED. Materials and Methods: SHED cultured in osteoblast induction media (OIM) was treated with flaxseed extract at 4mg/ml. The RNA was collected and extracted with Total RNA Mini Kit (Geneaid) from cells cultured at day 1, 3, 7, 14 and 21 and subjected to reversetranscriptase PCR for osteoblast markers (OSX, OCN, DMP1). Alkaline phosphatase activity was determined by ALP assay and Alizarin Red S staining was performed to evaluate calcium deposits in SHED. Results: All osteoblast markers were expressed in all samples analysed. Interestingly, the expression of OSX appeared to be reduced in SHED treated with flaxseed extract. In addition, SHED treated with flaxseed extract resulted in low ALP activity compared to control (p<0.05) (t-test). Calcium deposition was positive in SHED cultured in OIM only. Conclusion: Flaxseed appears to reduce the expression of osteoblast markers, ALP activity and calcium deposition in SHED. Abstract

Brucellosis is a popular, zoonotic disease that has detrimental implications on human, animal, and economic health. This study compared the efficiency and effectiveness of various serological tests for the seroprevalence of brucellosis in cattle in four Egyptian governorates, including the buffered acidified plate antigen (BAPA) assay, Rose Bengal Plate Test (RBPT), indirect enzyme-linked immunosorbent assay (i-ELISA), and competitive enzyme-linked immunosorbent assay (c-ELISA). A total of 240 sera samples were collected from asymptomatic cattle from Cairo (n = 58), Ismailia (n = 62), Beni Suif (n = 60), and Fayom (n = 60) in Egypt. According to PABA, RBT, i-ELISA, and c-ELISA assay results, the overall prevalence of brucellosis was 141 (58.75%), 141 (58.75%), 169 (70.4%), and 160 (66.66%), respectively. i-ELISA as the gold standard: the sensitivity, specificity, and accuracy of RBT were evaluated as 99, 70 and 59%, respectively with a 95% confidence interval (CI) of 95 to 100%, 60 to 79% and 52 to 65%. While considering c-ELISA as the gold standard: the sensitivity, specificity, and accuracy of RBT were evaluated as 100, 81 and 59%, respectively with a 95% CI of 97 to 100%, 72 to 88% and 52 to 65%, respectively. In conclusion, both ELISA tests were proven to be superior serological tests than PABA and RBPT, and they may be suggested for use in Egypt in screening cattle for brucellosis. A national campaign to control and prevent brucellosis should also get underway to lower its incidence. Abstract

Background: Severe preeclampsia (PE) and eclampsia are a hypertensive disease of pregnancy associated with significant morbidity and mortality and require special monitoring. Lung ultrasonography and transthoracic echocardiography are a novel monitoring and diagnostic tools in intensive care and are widely used nowadays. Objective: To detect the role and value of transthoracic echocardiography (TTE) in postoperative monitoring in patients with severe PE. Patients and Methods: This was a cross sectional study of 55 patients with severe preeclampsia, 35 patients with eclampsia and 60 healthy controls. Transthoracic echocardiography was done to evaluate diastolic function using E/A ratio with continuous wave Doppler, ejection fraction using M mode in long axis parasternal view and inferior vena cava (IVC) diameter in subcostal view. All measurements were done postoperatively. Results: There was significant difference between the 3 groups regarding diastolic function (p value 0.0001). Abnormal diastolic function in severe PE, eclampsia and controls was 81.2%, 94.3% and 5% respectively. There was no difference between the 3 groups of the study as regard IVC diameter. There was significant difference between the 3 groups as regards EF (P=0.007), the mean of EF in severe PE, eclampsia and controls was 66.89 ± 5.54, 63.029 ± 8.45 and 66.43 ± 4.16 respectively. Conclusions: The use of transthoracic echocardiography is mandatory in postoperative monitoring in patients with severe PE and eclampsia as they can adjust management and detect complications. Restricted fluid therapy policy is indicated with the higher grades of diastolic dysfunction and lower ejection fraction (EF). [ABSTRACT FROM AUTHOR]
