We provide non-asymptotic, relative deviation bounds for the eigenvalues of empirical covariance and gram matrices in general settings. Unlike typical uniform bounds, which may fail to capture the behavior of smaller eigenvalues, our results provide sharper control across the spectrum. Our analysis is based on a general-purpose theorem that allows one to convert existing uniform bounds into relative ones. The theorems and techniques emphasize simplicity and should be applicable across various settings.

3. The AIR and Apt-AIR Frameworks of Epistemic Performance and Growth: Reflections on Educational Theory Development

- Abstract
The nurturing of learners' ways of knowing is vital for supporting their intellectual growth and their participation in democratic knowledge societies. This paper traces the development of two interrelated theoretical frameworks that describe the nature of learners' epistemic thinking and performance and how education can support epistemic growth: the AIR and Apt-AIR frameworks. After briefly reviewing these frameworks, we discuss seven reflections on educational theory development that stem from our experiences working on the frameworks. First, we describe how our frameworks were motivated by the goal of addressing meaningful educational challenges. Subsequently, we explain why and how we infused philosophical insights into our frameworks, and we also discuss the steps we took to increase the coherence of the frameworks with ideas from other educational psychology theories. Next, we reflect on the important role of the design of instruction and learning environments in testing and elaborating the frameworks. Equally important, we describe how our frameworks have been supported by empirical evidence and have provided an organizing structure for understanding epistemic performance exhibited in studies across diverse contexts. Finally, we discuss how the development of the frameworks has been spurred by dialogue within the research community and by the need to address emerging and pressing real-world challenges. To conclude, we highlight several important directions for future research. A common thread running through our work is the commitment to creating robust and dynamic theoretical frameworks that support the growth of learners' epistemic performance in diverse educational contexts.

6. Generalization in Kernel Regression Under Realistic Assumptions

It is by now well-established that modern over-parameterized models seem to elude the bias-variance tradeoff and generalize well despite overfitting noise. Many recent works attempt to analyze this phenomenon in the relatively tractable setting of kernel regression. However, as we argue in detail, most past works on this topic either make unrealistic assumptions, or focus on a narrow problem setup. This work aims to provide a unified theory to upper bound the excess risk of kernel regression for nearly all common and realistic settings. Specifically, we provide rigorous bounds that hold for common kernels and for any amount of regularization, noise, any input dimension, and any number of samples. Furthermore, we provide relative perturbation bounds for the eigenvalues of kernel matrices, which may be of independent interest. These reveal a self-regularization phenomenon, whereby a heavy tail in the eigendecomposition of the kernel provides it with an implicit form of regularization, enabling good generalization. When applied to common kernels, our results imply benign overfitting in high input dimensions, nearly tempered overfitting in fixed dimensions, and explicit convergence rates for regularized regression. As a by-product, we obtain time-dependent bounds for neural networks trained in the kernel regime.

- Abstract
This article discusses challenges posed to the design and enactment of twenty-first century school curricula by examining three core issues: pedagogical autonomy, the balance and integration of knowledge and thinking skills, and curricular flexibility. It focuses on how a committee of experts commissioned by the Israeli ministry of education to advise it on reforming the state curriculum responded to these challenges. The discussion highlights two fundamental questions in curriculum design: how to integrate global research finding with local conditions? and how to respond to a changing and unpredictable reality? To deal with the challenges, the article offers a model that aims to enhance the flexibility of the curriculum, and a list of criteria to assist educators on all levels in making informed curricular decisions. Although the analysis is situated within a particular cultural and national context the issues addressed have general and global implications.

Animal studies show aging varies between individuals as well as between organs within an individual1-4, but whether this is true in humans and its effect on age-related diseases is unknown. We utilized levels of human blood plasma proteins originating from specific organs to measure organ-specific aging differences in living individuals. Using machine learning models, we analysed aging in 11 major organs and estimated organ age reproducibly in five independent cohorts encompassing 5,676 adults across the human lifespan. We discovered nearly 20% of the population show strongly accelerated age in one organ and 1.7% are multi-organ agers. Accelerated organ aging confers 20-50% higher mortality risk, and organ-specific diseases relate to faster aging of those organs. We find individuals with accelerated heart aging have a 250% increased heart failure risk and accelerated brain and vascular aging predict Alzheimers disease (AD) progression independently from and as strongly as plasma pTau-181 (ref. 5), the current best blood-based biomarker for AD. Our models link vascular calcification, extracellular matrix alterations and synaptic protein shedding to early cognitive decline. We introduce a simple and interpretable method to study organ aging using plasma proteomics data, predicting diseases and aging effects.

Abstract While previous studies identified common genetic variants associated with longevity in centenarians, the role of the rare loss-of-function (LOF) mutation burden remains largely unexplored. Here, we investigated the burden of rare LOF mutations in Ashkenazi Jewish individuals from the Longevity Genes Project and LonGenity study cohorts using whole-exome sequencing data. We found that centenarians had a significantly lower burden (11-22%) of LOF mutations compared to controls. Similar effects were also observed in their offspring. Gene-level burden analysis identified 35 genes with depleted LOF mutations in centenarians, with 14 of these validated in the UK Biobank. Mendelian randomization and multi-omic analyses on these genes identified RGP1, PCNX2, and ANO9 as longevity genes with consistent causal effects on multiple aging-related traits and altered expression during aging. Our findings suggest that a protective genetic background, characterized by a reduced burden of damaging variants, contributes to exceptional longevity, likely acting in concert with specific protective variants to promote healthy aging.

13. Cognitive Aspects in Problem Solving: The Case of a Data Structures Course for IS Students

- Abstract
Data Structures and Algorithms (DS) is a basic computer science course that is a prerequisite for taking advanced information systems (IS) curriculum courses. The course aims to teach students how to analyze a problem, design a solution, and implement it using pseudocode to construct knowledge and develop the necessary skills for algorithmic problem solving and abstract thinking. While the literature acknowledges the difficulty of this course, few references were found that examine the process students undergo while solving DS algorithmic problems. The study's objective is to explore and describe IS students' problem-solving processes and challenges requiring a high level of abstract thinking in a "black box" approach. During the study, 13 students were observed while solving a complex problem, using "think aloud" (TA) techniques. Each observation was recorded, transcribed, and iteratively analyzed using principles of provisional coding in qualitative data analysis. The findings suggest that the quality and correctness of the solutions depend on three main factors: abstract thinking, flexibility applied during the solution process, and an absence of misconceptions related to concepts and the basic understanding of the problem. The students' levels of abstract thinking also influenced the quality of visualization used while trying to solve the problem. This study's findings may raise the awareness of DS course designers and instructors regarding the importance of the role of abstract thinking, possible misconceptions, and strategies used in problem solving as factors influencing students' ability to solve complex problems.

14. Grasping Psychological Evidence: Integrating Evidentiary Practices in Psychology Instruction

The spread of misinformation has underscored the importance of cultivating citizens' competency to critically evaluate popular accounts of scientific evidence. Extending the prevailing emphasis on evidence in the natural sciences, we argue for fostering students' understanding of psychological evidence and its communication in the media. In this study, we illustrate how this goal can be advanced in undergraduate psychology instruction by actively engaging students in evidence evaluation and design. We employed the Grasp of Evidence framework to document students' evidence evaluation ideals and processes and how these changed over a course in which students engaged in a series of collaborative evidence evaluation and design tasks. Prior to instruction, students exhibited a mechanistic understanding of scientific methods, coupled with substantial reliance on personal experience. Following instruction, students demonstrated three key shifts in grasp of evidence: a shift in perception of the sources of psychological knowledge, a shift in views of scientific objectivity, and a shift in definitions of psychological phenomena. Analysis of students' collaborative discourse highlighted three design principles that supported increasingly complex understandings of psychological evidence: collaborative critique and redesign of flawed studies, engagement with diverse sources of popular evidence, and confronting elusive conceptual constructs.

Wide neural networks are biased towards learning certain functions, influencing both the rate of convergence of gradient descent (GD) and the functions that are reachable with GD in finite training time. As such, there is a great need for methods that can modify this bias according to the task at hand. To that end, we introduce Modified Spectrum Kernels (MSKs), a novel family of constructed kernels that can be used to approximate kernels with desired eigenvalues for which no closed form is known. We leverage the duality between wide neural networks and Neural Tangent Kernels and propose a preconditioned gradient descent method, which alters the trajectory of GD. As a result, this allows for a polynomial and, in some cases, exponential training speedup without changing the final solution. Our method is both computationally efficient and simple to implement.

Abstract The highly polygenic nature of human longevity renders pleiotropy an indispensable feature of its genetic architecture. Leveraging the genetic correlation between aging-related traits (ARTs), we aimed to model the additive variance in lifespan as a function of the cumulative liability from pleiotropic segregating variants. We tracked allele frequency changes as a function of viability across different age bins and prioritized 34 variants with an immediate implication on lipid metabolism, body mass index (BMI), and cognitive performance, among other traits, revealed by PheWAS analysis in the UK Biobank. Given the highly complex and non-linear interactions between the genetic determinants of longevity, we reasoned that a composite polygenic score would approximate a substantial portion of the variance in lifespan and developed the integrated longevity genetic scores (iLGSs) for distinguishing exceptional survival. We showed that coefficients derived from our ensemble model could potentially reveal an interesting pattern of genomic pleiotropy specific to lifespan. We assessed the predictive performance of our model for distinguishing the enrichment of exceptional longevity among long-lived individuals in two replication cohorts (the Scripps Wellderly cohort and the Medical Genome Reference Bank (MRGB)) and showed that the median lifespan in the highest decile of our composite prognostic index is up to 4.8 years longer. Finally, using the proteomic correlates of iLGS, we identified protein markers associated with exceptional longevity irrespective of chronological age and prioritized drugs with repurposing potentials for gerotherapeutics. Together, our approach demonstrates a promising framework for polygenic modeling of additive liability conferred by ARTs in defining exceptional longevity and assisting the identification of individuals at a higher risk of mortality for targeted lifestyle modifications earlier in life. Furthermore, the proteomic signature associated with iLGS highlights the functional pathway upstream of the PI3K-Akt that can be effectively targeted to slow down aging and extend lifespan.

Abstract High-throughput microscopy is vital for screening applications, where three-dimensional (3D) cellular models play a key role. However, due to defocus susceptibility, current 3D high-throughput microscopes require axial scanning, which lowers throughput and increases photobleaching and photodamage. Point spread function (PSF) engineering is an optical method that enables various 3D imaging capabilities, yet it has not been implemented in high-throughput microscopy due to the cumbersome optical extension it typically requires. Here we demonstrate compact PSF engineering in the objective lens, which allows us to enhance the imaging depth of field and, combined with deep learning, recover 3D information using single snapshots. Beyond the applications shown here, this work showcases the usefulness of high-throughput microscopy in obtaining training data for deep learning-based algorithms, applicable to a variety of microscopy modalities.

IntroductionThere are limited data on prevalence of dementia in centenarians and near-centenarians (C/NC), its determinants, and whether the risk of dementia continues to rise beyond 100.MethodsParticipant-level data were obtained from 18 community-based studies (N = 4427) in 11 countries that included individuals ≥95 years. A harmonization protocol was applied to cognitive and functional impairments, and a meta-analysis was performed.ResultsThe mean age was 98.3 years (SD = 2.67); 79% were women. After adjusting for age, sex, and education, dementia prevalence was 53.2% in women and 45.5% in men, with risk continuing to increase with age. Education (OR 0.95;0.92-0.98) was protective, as was hypertension (odds ratio [OR] 0.51;0.35-0.74) in five studies. Dementia was not associated with diabetes, vision and hearing impairments, smoking, and body mass index (BMI).DiscussionAmong the exceptional old, dementia prevalence remains higher in the older participants. Education was protective against dementia, but other factors for dementia-free survival in C/NC remain to be understood.

OBJECTIVES:. Anemia has been associated with an increased risk of both cardiac arrest and stroke, frequent complications of COVID-19. The effect of hemoglobin level at ICU admission on a composite outcome of cardiac arrest or stroke in an international cohort of COVID-19 patients was investigated. DESIGN:. Retrospective analysis of prospectively collected database. SETTING:. A registry of COVID-19 patients admitted to ICUs at over 370 international sites was reviewed for patients diagnosed with cardiac arrest or stroke up to 30 days after ICU admission. Anemia was defined as: normal (hemoglobin ≥ 12.0 g/dL for women, ≥ 13.5 g/dL for men), mild (hemoglobin 10.0–11.9 g/dL for women, 10.0–13.4 g/dL for men), moderate (hemoglobin ≥ 8.0 and < 10.0 g/dL for women and men), and severe (hemoglobin < 8.0 g/dL for women and men). PATIENTS:. Patients older than 18 years with acute COVID-19 infection in the ICU. INTERVENTIONS:. None. MEASUREMENTS AND MAIN RESULTS:. Of 6926 patients (median age = 59 yr, male = 65%), 760 patients (11.0%) experienced stroke (2.0%) and/or cardiac arrest (9.4%). Cardiac arrest or stroke was more common in patients with low hemoglobin, occurring in 12.8% of patients with normal hemoglobin, 13.3% of patients with mild anemia, and 16.7% of patients with moderate/severe anemia. Time to stroke or cardiac arrest by anemia status was analyzed using Cox proportional hazards regression with death as a competing risk. Covariates selected through clinical knowledge were age, sex, comorbidities (diabetes, hypertension, obesity, and cardiac or neurologic conditions), pandemic era, country income, mechanical ventilation, and extracorporeal membrane oxygenation. Moderate/severe anemia was associated with a higher risk of cardiac arrest or stroke (hazard ratio, 1.32; 95% CI, 1.05–1.67). CONCLUSIONS:. In an international registry of ICU patients with COVID-19, moderate/severe anemia was associated with increased hazard of cardiac arrest or stroke.

28. A Kernel Perspective of Skip Connections in Convolutional Networks

Over-parameterized residual networks (ResNets) are amongst the most successful convolutional neural architectures for image processing. Here we study their properties through their Gaussian Process and Neural Tangent kernels. We derive explicit formulas for these kernels, analyze their spectra, and provide bounds on their implied condition numbers. Our results indicate that (1) with ReLU activation, the eigenvalues of these residual kernels decay polynomially at a similar rate compared to the same kernels when skip connections are not used, thus maintaining a similar frequency bias; (2) however, residual kernels are more locally biased. Our analysis further shows that the matrices obtained by these residual kernels yield favorable condition numbers at finite depths than those obtained without the skip connections, enabling therefore faster convergence of training with gradient descent.

Background: Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease. Most patients with moderate-to-severe disease require long-term antibiotic treatment, or biologic treatments to control their disease. Despite these interventions, relapses are common. This study evaluated the effectiveness of piperacillin/tazobactam treatment in patients with Hurley stage II and III HS who experienced disease flares and did not respond to conventional antibiotic and biologic treatment. Methods: Patients with HS hospitalized at the Department of Dermatology, Sheba Medical Center between August 2021 and January 2023 were retrospectively analyzed. Results: A cohort of ten such patients were treated with piperacillin/tazobactam for 6–21 days. Eight (80%) and two (20%) patients respectively demonstrated 2- and 1-grade improvements, from their baseline HS-Physician Global Assessment score. During the follow-up period, nine patients were monitored. HS Clinical Response (HiSCR) was achieved in six (66.7%) and five (55.6%) patients at the 3- and 6-month follow-up visits, respectively. Conclusions: In conclusion, Piperacillin/tazobactam emerges as a promising therapeutic option for disease flare-up in patients with Hurley stage II and III HS who do not respond to conventional treatment. Thus, piperacillin/tazobactam should be considered as crisis therapy for this patient subset.

With the goal of identifying metabolites that significantly correlate with the protective e2 allele of the apolipoprotein E (APOE) gene, we established a consortium of five studies of healthy aging and extreme human longevity with 3545 participants. This consortium includes the New England Centenarian Study, the Baltimore Longitudinal Study of Aging, the Arivale study, the Longevity Genes Project/LonGenity studies, and the Long Life Family Study. We analyzed the association between APOE genotype groups E2 (e2e2 and e2e3 genotypes, N = 544), E3 (e3e3 genotypes, N = 2299), and E4 (e3e4 and e4e4 genotypes, N = 702) with metabolite profiles in the five studies and used fixed effect meta-analysis to aggregate the results. Our meta-analysis identified a signature of 19 metabolites that are significantly associated with the E2 genotype group at FDR

- Abstract
Background: YouTube is widely used for learning about scientific issues in and out of school. However, much of the scientific information on YouTube is inaccurate. Prior studies have mostly focused on how students evaluate textual online information sources and have not yet systematically examined how they evaluate authentic scientific YouTube videos. Objectives: The purpose of this study was to examine how students evaluate authentic scientific YouTube videos in comparison to scientific webpages. Methods: Eighth grade students ranked six YouTube videos on the topic of climate change, justified their rankings, and responded to metacognitive prompts designed to elicit their evaluation criteria and strategies. Students also responded to a parallel webpage evaluation measure. We analysed and compared students' evaluation strategies and criteria. We also examined which evaluation criteria predict students' judgments of YouTube video quality. Results and Conclusions: Students predominantly relied on evaluation of video content, focusing mainly on criteria of communicative quality and explanation quality. Students tended to neglect source expertise and information validity criteria. Students evaluated videos mostly similarly to how they evaluated webpages. However, affective experiences played a greater role in video evaluation; whereas task relevance and verbal quality played a smaller role. Students' evaluation criteria predicted their judgments of video quality. Implications: The findings suggest that students are unprepared for critically evaluating scientific YouTube videos and that digital information literacy instruction should address this gap. The study also identifies some unique challenges of video evaluation that educators should attend to.

33. Mapping Multiple Documents: From Constructing Multiple Document Models to Argumentative Writing

Visualizations may help students understand the network of connections among multiple documents. This study explored how ninth-grade students used a novel digital document mapping scaffold to construct models of multiple documents. We examined students' maps and mapping processes in order to understand how they used the scaffold to visually represent multiple scientific documents on a controversial issue. We also investigated the relations between document mapping and argumentative writing in order to clarify how mapping might support written argumentation. Lastly, we explored how students perceived the purposes of document mapping. We found that students used the scaffold to construct several types of models that varied in their degree of source vs. content separation and integration. About half of the students constructed full documents models. Interleaving reading and mapping led to construction of more elaborate and well-integrated maps. Students who created more elaborate and two-sided maps also wrote essays that better integrated multiple documents in order to synthesize and weigh competing arguments. Students who revisited the maps and the documents while writing wrote essays that included and cited more documents. Students described document mapping as a tool for selecting and connecting sources and claims in order to support understanding and writing. These findings reveal how document mapping can support comprehension and integration of multiple documents. The findings also shed new light on how students construct documents models and on the relations between representations of multiple documents and argumentative writing. These results have implications for both theory and instruction.

Antler is one of the primary animal raw materials exploited for technical purposes by the hunter-gatherer groups of the Eurasian Upper Palaeolithic (UP) all over the ecological range of deers, and beyond. It was exhaustively employed to produce one of the most critical tools for the survival of the UP societies: hunting weapons. However, antler implements can be made from diverse deer taxa, with different ecological requirements and ethological behaviours. Identifying the antler's origin at a taxonomic level is thus essential in improving our knowledge of humans' functional, practical and symbolic choices, as well as the human-animal interface during Prehistoric times. Nevertheless, palaeogenetics analyses have focused mainly on bone and teeth, with genetic studies of antler generally focused on modern deer conservation. Here we present the results of the first whole mitochondrial genome ancient DNA (aDNA) analysis by means of in-solution hybridisation capture of antlers from pre-Holocene archaeological contexts. We analysed a set of 50 Palaeolithic and Neolithic (c. 34-8ka) antler and osseous objects from South-Western Europe, Central Europe, South-Western Asia and the Caucasus. We successfully obtained aDNA, allowing us to identify the exploited taxa and demonstrate the archaeological relevance of those finds. Moreover, as most of the antlers were sampled using a minimally-invasive method, further analyses (morphometric, technical, genetic, radiometric and more) remain possible on these objects.

Miltefosine, an orally administered drug, is an important component of the therapeutic arsenal against visceral and mucosal forms of leishmaniasis. However, data regarding the safety and tolerability of miltefosine treatment for cutaneous leishmaniasis (CL) are relatively limited. The aim of this study was to evaluate the tolerability, safety, and adverse events (AEs) of miltefosine treatment in patients with CL. In this cohort study, we reviewed the medical records of all miltefosine-treated patients between 1 January 2016 and 31 December 2022, at Israel Defense Forces military dermatology clinics and the dermatology and Tropical Medicine Clinics at Chaim Sheba Medical Center, Ramat-Gan, Israel. A total of 68 patients (54 males, 79%) with a median age of 30.3 ± 15.6 years (range: 18–88) were included in this study. Leishmania species were identified as L. major (n = 37, 54.4%), L. tropica (n = 12, 17.6%), L. braziliensis (n = 18, 26.5%), and L. infantum (n = 1, 1.5%) using polymerase chain reaction (PCR). Miltefosine tablets were administered orally at a dose of 50 mg, three times daily, for 28 days. Overall, 44 patients (65%) completed the 28-day treatment, and the remaining patients required dose reduction or early discontinuation of treatment. AEs (of any degree) were common, reported in 91% of patients. Both previously reported and previously unreported AEs were documented. Gastrointestinal symptoms (66.1%) and malaise (23.5%) typically occurred during the first two weeks of treatment and tended to subside. Other AEs, including acute renal failure (20.6%), sudden and severe pleuritic chest pain (7.6%), acne exacerbation (11.8%), suppuration of CL lesions (17.8%), and AEs related to the male genitourinary system (39.6% of males), typically occurred towards the end of treatment. The latter included testicular pain, epididymitis, diminution or complete absence of ejaculate, inability to orgasm, and impotence. Severe AEs necessitated treatment discontinuation (29.4%) or hospitalization (10.3%). URTI-like symptoms, arthritis, cutaneous eruption, pruritus, and laboratory abnormalities were also observed. Overall, the cure rate (for all patients combined) evaluated 3 months after the completion of treatment was 60%. The tolerability of miltefosine treatment for CL is low. Close clinical and laboratory monitoring is required during treatment, as severe AEs are not uncommon. As new insights regarding its toxicities emerge, further studies are required to define the role of miltefosine in the treatment of CL.

Israel is endemic for Old-World cutaneous leishmaniasis. The most common species is Leishmania major. However, the available treatment options are limited. This study’s objective was to compare the authors’ experience with different antimony intralesional treatments of Leishmania major cutaneous leishmaniasis. A retrospective evaluation was undertaken for cases of Leishmania major cutaneous leishmaniasis treated by pentavalent antimony in a university-affiliated medical centre in Israel. The previous treatment of intralesional sodium stibogluconate (Pentostam®) was compared with the current treatment of meglumine antimoniate (Glucantime®). One hundred cases of cutaneous leishmaniasis were treated during the study period, of whom 33 were treated with intralesional sodium stibogluconate and 67 were treated with intralesional meglumine antimoniate. The patients were 78 males and 22 females, mean age 24 (range 10–67) and there was a total of 354 skin lesions. Within 3 months from treatment, 91% (30/33) of the intralesional sodium stibogluconate group and 88% (59/67) of the intralesional meglumine antimoniate group had complete healing of the cutaneous lesions after an average of 3 treatment cycles (non-statistically significant). In conclusion, the 2 different medications have the same efficacy and safety for treating cutaneous leishmaniasis. Pentavalent antimoniate intralesional infiltration treatment is safe, effective, and well tolerated with minimal side effects for Old-World cutaneous leishmaniasis.
