As an epistemological, axiological, and methodological paradigm, Critical Race Theory (CRT; Crenshaw et al., 2000; Harris, 1993) is a scholarly tool to identify and disrupt inequities, possible via CRT's core tenets towards troubling systemic racism. We argue that political movements in North Carolina (NC) exhibit attempts to delegitimize critical race scholarship and curricula that accurately portray history and contemporary student populations' racialized experiences, a manifestation of the conservative agenda to whitewash the state's history that is predicated on racism and white supremacy. In alignment with radical theorizations and research that examine ideologies at the root of ill-informed hysteria, we present a Critical Discourse Analysis (CDA) of the effects of political power in foreclosing educational possibilities toward building equitable societies through our analysis of data from NC's Fairness and Accountability in the Classroom for Teachers and Students for North Carolinian "FACTS" submissions (Robinson, 2021). FACTS is a reporting tool targeting NC educators who employ critical lenses in their instruction that promotes unfounded antagonism toward CRT. The significance of this research is a localized example of CRT being targeted by conservative politicians toward the intent of delegitimizing critical scholarship and education and thus perpetuating ahistorical ideals rooted in racism and white supremacy.

SARS-CoV-2’s rapid global spread caused the declaration of COVID-19 as a pandemic in March 2020. Alongside humans, domestic dogs and cats are also susceptible to infection. However, limited reports on pet infections in Chile prompted a comprehensive study to address this knowledge gap. Between March 2021 and March 2023, the study assessed 65 pets (26 dogs and 39 cats) from 33 COVID-19+ households alongside 700 nasal swabs from animals in households with unknown COVID-19 status. Using RT-PCR, nasal, fecal, and environmental samples were analyzed for the virus. In COVID-19+ households, 6.06% tested positive for SARS-CoV-2, belonging to 3 dogs, indicating human-to-pet transmission. Pets from households with unknown COVID-19 status tested negative for the virus. We obtained 2 SARS-CoV-2 genomes from animals, that belonged to Omicron BA.4.1 variant, marking the first report of pets infected with this lineage globally. Phylogenetic analysis showed these sequences clustered with human sequences collected in Chile during the same period when the BA.4.1 variant was prevalent in the country. The prevalence of SARS-CoV-2 in Chilean pets was relatively low, likely due to the country’s high human vaccination rate. Our study highlights the importance of upholding and strengthening human vaccination strategies to mitigate the risk of interspecies transmission. It underscores the critical role of the One Health approach in addressing emerging zoonotic diseases, calling for further research on infection dynamics and risk factors for a comprehensive understanding.

Aligned with critical scholarship and upholding minoritized populations' agency and power, this survey research study addresses Latinx college students' resistance strategies in two Nuevo South states by examining the social issues that students are aware of, engage with, and the nature of their interactions with these issues. We apply Southern epistemology as a framework to center the unique Nuevo South sociopolitical context and examine modern issues Latinx communities face in this geographic region and how Latinx college students resist hostility. Findings empirically establish how the Southern epistemology remains to modernly construct and constrain the lives of Latinx college students in the Nuevo South.

Abstract Objective The MGDrivE (MGDrivE 1 and MGDrivE 2) modeling framework provides a flexible and expansive environment for testing the efficacy of novel gene-drive constructs for the control of mosquito-borne diseases. However, the existing model framework did not previously support several features necessary to simulate some types of intervention strategies. Namely, current MGDrivE versions do not permit modeling of small molecule inducible systems for controlling gene expression in gene drive designs or the inheritance patterns of self-eliminating gene drive mechanisms. Results Here, we demonstrate a new MGDrivE 2 module that permits the simulation of gene drive strategies incorporating small molecule-inducible systems and self-eliminating gene drive mechanisms. Additionally, we also implemented novel sparsity-aware sampling algorithms for improved computational efficiency in MGDrivE 2 and supplied an analysis and plotting function applicable to the outputs of MGDrivE 1 and MGDrivE 2.

Cryptococcus neoformans can invade the central nervous system by crossing the blood-brain barrier via a transcellular mechanism that relies on multiple host factors. In this narrative, we review the evidence that a direct interplay between C. neoformans and brain endothelial cells forms the basis for invasion and transmigration across the brain endothelium. Adherence and internalization of C. neoformans is dependent on transmembrane proteins, including a hyaluronic acid receptor and an ephrin receptor tyrosine kinase. We consider the role of EphA2 in facilitating the invasion of the central nervous system by C. neoformans and highlight experimental evidence supporting macropinocytosis as a potential mechanism of internalization and transcytosis. How macropinocytosis might be conclusively demonstrated in the context of C. neoformans is also discussed.

CRISPR-based gene-drives can carry the Cas9 and guide RNA (gRNA) components in a single-linked cassette or in separate elements inserted into different genomic loci. Here the authors genetically transform and compare full versus split drives, with the former performing less efficiently than predicted.

The objective of this preliminary observational study was to determine milking time behavior of cows in a free-flow automated (robotic) milking system (AMS) and identify potential factors that influenced the time waiting to be milked. Milking time behavior of 40 cows from 1 pen on a commercial dairy farm with a free-flow AMS was evaluated using video analysis over 2 d. For each study cow, data were assessed for waiting time to access the milking robot, the use of the fetch pen, robot refusals, and their lying behavior. On average, cows visited the robot to wait to be milked 6 times per day, for 15 min per visit, for a total daily waiting time of 88 min per cow (range 5 to 322 min). Daily waiting time was longer for primiparous cows and decreased with increasing days in milk, but this effect interacted with parity. Daily waiting time and number of visits to the robot were associated with voluntary use of the fetch pen. Furthermore, cows with long daily waiting times had shorter daily lying times compared with cows with short daily waiting times (9.5 vs. 11.1 h/d). It is possible that factors related to the design and layout of the AMS entry and fetch pen had an effect on waiting behavior. We inferred that adoption of grouping strategies intended to reduce competitive behavior, especially toward primiparous cows, could improve milking time behavior in a free-flow AMS. This preliminary observational data from a single herd highlights the need to confirm the findings across multiple AMS herds, both with free-flow and guided-flow systems.

Abstract Shark bites are of high public concern globally. Information on shark occurrence and behaviour, and of the effects of human behaviours, can help understand the drivers of shark‐human interactions. In Australia, a number of shark bite clusters occurred over the last decade. One of these took place in Cid Harbour the Whitsundays, Queensland, a region for which little was known about the shark community. Here, we describe and evaluate the research in response to that shark bite cluster. Fishing methods, acoustic and satellite tracking, and baited remote underwater video cameras (BRUVs) were used to identify the shark species using Cid Harbour, estimate relative abundance, and describe habitat use and residency. Side‐scan sonar and BRUVs were also used to assess prey availability. Recreational users were surveyed to understand human behaviour and their awareness and perceptions of ‘Shark Smart’ behaviours. This allowed shark occurrence and behaviour to be interpreted in the context of human behaviours in the Harbour. Eleven shark species were identified. Abstract Over the last century, many shark populations have declined, primarily due to overexploitation in commercial, artisanal and recreational fisheries. In addition, in some locations the use of shark control programs also has had an impact on shark numbers. Still, there is a general perception that populations of large ocean predators cover wide areas and therefore their diversity is less susceptible to local anthropogenic disturbance. Here we report on temporal genomic analyses of tiger shark (Galeocerdo cuvier) DNA samples that were collected from eastern Australia over the past century. Using Single Nucleotide Polymorphism (SNP) loci, we documented a significant change in genetic composition of tiger sharks born between ~1939 and 2015. The change was most likely due to a shift over time in the relative contribution of two well-differentiated, but hitherto cryptic populations. Our data strongly indicate a dramatic shift in the relative contribution of these two populations to the overall tiger shark abundance on the east coast of Australia, possibly associated with differences in direct or indirect exploitation rates.

Insecticide resistance (IR) poses a major global health challenge. Here, the authors generate common IR mutations in laboratory Drosophila strains and use a CRISPR-based allelic-drive to replace an IR allele with a susceptible wild-type counterpart, providing a potent new tool for vector control.

CRISPR-based engineering can be used to bias sex ratios. Here the authors develop a transgenic line of Drosophila melanogaster expressing Cas9 from the Y chromosome and functionally characterize the utility of this strain for both sex selection and gene drive.

Close-kin mark-recapture (CKMR) methods have recently been used to infer demographic parameters such as census population size and survival for fish of interest to fisheries and conservation. These methods have advantages over traditional mark-recapture methods as the mark is genetic, removing the need for physical marking and recapturing that may interfere with parameter estimation. For mosquitoes, the spatial distribution of close-kin pairs has been used to estimate mean dispersal distance, of relevance to vector-borne disease transmission and novel biocontrol strategies. Here, we extend CKMR methods to the life history of mosquitoes and comparable insects. We derive kinship probabilities for mother-offspring, father-offspring, full-sibling and half-sibling pairs, where an individual in each pair may be a larva, pupa or adult. A pseudo-likelihood approach is used to combine the marginal probabilities of all kinship pairs. To test the effectiveness of this approach at estimating mosquito demographic parameters, we develop an individual-based model of mosquito life history incorporating egg, larva, pupa and adult life stages. The simulation labels each individual with a unique identification number, enabling close-kin relationships to be inferred for sampled individuals. Using the dengue vector Aedes aegypti as a case study, we find the CKMR approach provides unbiased estimates of adult census population size, adult and larval mortality rates, and larval life stage duration for logistically feasible sampling schemes. Considering a simulated population of 3,000 adult mosquitoes, estimation of adult parameters is accurate when ca. 40 adult females are sampled biweekly over a three month period. Estimation of larval parameters is accurate when adult sampling is supplemented with ca. 120 larvae sampled biweekly over the same period. The methods are also effective at detecting intervention-induced increases in adult mortality and decreases in population size. As the cost of genome sequencing declines, CKMR holds great promise for characterizing the demography of mosquitoes and comparable insects of epidemiological and agricultural significance.

A. aegypti is the principal vector for arboviruses that impact on human health and wellbeing. Here the authors use precision guided sterile insect technique—pgSIT—to suppress or eliminate mosquito populations in multigeneration cage experiments.

Abstract Background Delimiting cryptic species in elasmobranchs is a major challenge in modern taxonomy due the lack of available phenotypic features. Employing stand-alone genetics in splitting a cryptic species may prove problematic for further studies and for implementing conservation management. In this study, we examined mitochondrial DNA and genome-wide nuclear single nucleotide polymorphisms (SNPs) in the brown-banded bambooshark, Chiloscyllium punctatum to evaluate potential cryptic species and the species-population boundary in the group. Results Both mtDNA and SNP analyses showed potential delimitation within C. punctatum from the Indo-Australian region and consisted of four operational taxonomic units (OTUs), i.e. those from Indo-Malay region, the west coast of Sumatra, Lesser Sunda region, and the Australian region. Each OTU can be interpreted differently depending on available supporting information, either based on biological, ecological or geographical data. We found that SNP data provided more robust results than mtDNA data in determining the boundary between population and cryptic species. Conclusion To split a cryptic species complex and erect new species based purely on the results of genetic analyses is not recommended. The designation of new species needs supportive diagnostic morphological characters that allow for species recognition, as an inability to recognise individuals in the field creates difficulties for future research, management for conservation and fisheries purposes. Moreover, we recommend that future studies use a comprehensive sampling regime that encompasses the full range of a species complex. This approach would increase the likelihood of identification of operational taxonomic units rather than resulting in an incorrect designation of new species.

Abstract Mutualism is a form of symbiosis whereby both parties benefit from the relationship. An example is cleaning symbiosis, which has been observed in terrestrial and marine environments. The most recognized form of marine cleaning symbiosis is that of cleaner fishes and their clients. Cleaner species set up cleaning stations on the reef, and other species seek out their services. However, it is not well understood how the presence of cleaning stations influence movements of large highly mobile species. We examined the role of cleaning stations as a driver of movement and habitat use in a mobile client species. Here, we used a combination of passive acoustic telemetry and in‐water surveys to investigate cleaning station attendance by the reef manta ray Mobula alfredi. We employed a novel approach in the form of a fine‐scale acoustic receiver array set up around a known cleaning area and tagged 42 rays. Within the array, we mapped structural features, surveyed the distribution of cleaner wrasse, and observed the habitat use of the rays. We found manta ray space use was significantly associated with blue‐streak cleaner wrasse Labroides dimidiatus distribution and hard coral substrate. Cleaning interactions dominated their habitat use at this site, taking precedence over other life history traits such as feeding and courtship. This study has demonstrated that cleaning symbiosis is a driver for highly mobile, and otherwise pelagic, species to visit inshore reef environments. We suggest that targeted and long‐term use of specific cleaning stations reflects manta rays having a long‐term memory and cognitive map of some shallow reef environments where quality cleaning is provided. We hypothesize that animals prefer cleaning sites in proximity to productive foraging regions.

NHEJ alleles and Cas9 remnants after a gene drive introduction are scientific and public concerns. Here, the authors use split drives with recoded rescue elements to target essential genes and minimize the appearance of NHEJ alleles while also leaving no trace of Cas9.

Gene drives may be impeded by the generation of resistant alleles following NHEJ. Here the authors develop a recoded gene-drive rescue system for the malaria mosquito, Anopheles stephensi, that targets the drive to the kynurenine hydroxylase gene for negative selection against mutated alleles.

Abstract Background The discovery of CRISPR-based gene editing and its application to homing-based gene drive systems has been greeted with excitement, for its potential to control mosquito-borne diseases on a wide scale, and concern, for the invasiveness and potential irreversibility of a release. Gene drive systems that display threshold-dependent behavior could potentially be used during the trial phase of this technology, or when localized control is otherwise desired, as simple models predict them to spread into partially isolated populations in a confineable manner, and to be reversible through releases of wild-type organisms. Here, we model hypothetical releases of two recently engineered threshold-dependent gene drive systems—reciprocal chromosomal translocations and a form of toxin-antidote-based underdominance known as UDMEL—to explore their ability to be confined and remediated. Results We simulate releases of Aedes aegypti, the mosquito vector of dengue, Zika, and other arboviruses, in Yorkeys Knob, a suburb of Cairns, Australia, where previous biological control interventions have been undertaken on this species. We monitor spread to the neighboring suburb of Trinity Park to assess confinement. Results suggest that translocations could be introduced on a suburban scale, and remediated through releases of non-disease-transmitting male mosquitoes with release sizes on the scale of what has been previously implemented. UDMEL requires fewer releases to introduce, but more releases to remediate, including of females capable of disease transmission. Both systems are expected to be confineable to the release site; however, spillover of translocations into neighboring populations is less likely. Conclusions Our analysis supports the use of translocations as a threshold-dependent drive system capable of spreading disease-refractory genes into Ae. aegypti populations in a confineable and reversible manner. It also highlights increased release requirements when incorporating life history and population structure into models. As the technology nears implementation, further ecological work will be essential to enhance model predictions in preparation for field trials.

Objective – This study uses quantitative methods to determine if the metadata requirements of institutional repositories (IRs) promote data discovery. This question is addressed through an exploration of an international sample of university IRs, including an analysis of the required metadata elements for data deposit, with a particular focus on how these metadata support discovery of research data objects. Methods – The researchers worked with an international universe of 243 IRs. A codebook of 10 variables was developed to enable analysis of the eventual randomly derived sample of 40 institutions. Results – The analysis of our sample IRs revealed that most had metadata standards that offered weak support for data discovery—an unsurprising revelation in view of the fact that university IRs are meant to accommodate deposit and storage of all types of scholarly outputs, only a small percentage of which are research data objects. Most IRs seem to have adopted metadata standards based on the Dublin Core schema, while none of the IRs in our sample used the Data Documentation Initiative metadata that is better suited for deposit and discovery of research datasets. Conclusion – The study demonstrates that while data deposit can be accommodated by the existing metadata requirements of multi-purpose IRs, their metadata practices do little to prioritize data deposit or to promote data discovery. Evidence indicates that data discovery will benefit from additional metadata elements.

Gene drives raise safety concerns around unintended propagation. Here the authors present a trans-complementing split-gene drive that requires inheritance of separate transgenes to assemble a fully functional drive.

Stochastic rainfall modelling is a commonly used technique for evaluating the impact of flooding, drought, or climate change in a catchment. While considerable attention has been given to the development of stochastic rainfall models (SRMs), significantly less attention has been paid to developing methods to evaluate their performance. Typical evaluation methods employ a wide range of rainfall statistics. However, they give limited understanding about which rainfall statistical characteristics are most important for reliable streamflow prediction. To address this issue a formal evaluation framework is introduced, with three key features: (i) streamflow-based, to give a direct evaluation of modelled streamflow performance, (ii) virtual, to avoid the issue of confounding errors in hydrological models or data, and (iii) targeted, to isolate the source of errors according to specific sites and seasons. The virtual hydrological evaluation framework uses two types of tests, integrated tests and unit tests, to attribute deficiencies that impact on streamflow to their original source in the SRM according to site and season. The framework is applied to a case study of 22 sites in South Australia with a strong seasonal cycle. In this case study, the framework demonstrated the surprising result that apparently “good” modelled rainfall can produce “poor” streamflow predictions, whilst “poor” modelled rainfall may lead to “good” streamflow predictions. This is due to the representation of highly seasonal catchment processes within the hydrological model that can dampen or amplify rainfall errors when converted to streamflow. The framework identified the importance of rainfall in the “wetting-up” months (months where the rainfall is high but streamflow low) of the annual hydrologic cycle (May and June in this case study) for providing reliable predictions of streamflow over the entire year despite their low monthly flow volume. This insight would not have been found using existing methods and highlights the importance of the virtual hydrological evaluation framework for SRM evaluation.

Abstract Objective Limited sample sizes are often a problem for species of conservation concern when using genetic tools to make population assessments. Lack of analytical power from small sample sizes can be compensated for by use of a large marker set. Here we report on development and characterization of 17 novel microsatellite markers for the reef manta ray (Mobula alfredi). Results Loci were screened on 60 reef manta rays (M. alfredi) sampled from the east coast of Australia. The number of alleles per locus varied from 2 to 13 with observed heterozygosities ranging between 0.300 and 0.917. The development of these 17 additional markers increases the total number of microsatellite markers available for this species to 27.

Manta rays forage for zooplankton in tropical and subtropical marine environments, which are generally nutrient-poor. Feeding often occurs at predictable locations where these large, mobile cartilaginous fishes congregate to exploit ephemeral productivity hotspots. Investigating the zooplankton dynamics that lead to such feeding aggregations remains a key question for understanding their movement ecology. The aim of this study is to investigate the feeding environment at the largest known aggregation for reef manta rays Mobula alfredi in the world. We sampled zooplankton throughout the tidal cycle, and recorded M. alfredi activity and behaviour, alongside environmental variables at Hanifaru Bay, Maldives. We constructed generalised linear models to investigate possible relationships between zooplankton dynamics, environmental parameters, and how they influenced M. alfredi abundance, behaviour, and foraging strategies. Zooplankton biomass changed rapidly throughout the tidal cycle, and M. alfredi feeding events were significantly related to high zooplankton biomass. Mobula alfredi switched from non-feeding to feeding behaviour at a prey density threshold of 53.7 mg dry mass m−3; more than double the calculated density estimates needed to theoretically meet their metabolic requirements. The highest numbers of M. alfredi observed in Hanifaru Bay corresponded to when they were engaged in feeding behaviour. The community composition of zooplankton was different when M. alfredi was feeding (dominated by copepods and crustaceans) compared to when present but not feeding (more gelatinous species present than in feeding samples). The dominant zooplankton species recorded was Undinula vulgaris. This is a large-bodied calanoid copepod species that blooms in oceanic waters, suggesting offshore influences at the site. Here, we have characterised aspects of the feeding environment for M. alfredi in Hanifaru Bay and identified some of the conditions that may result in large aggregations of this threatened planktivore, and this information can help inform management of this economically important marine protected area.

Homing-based gene drives, engineered using CRISPR/Cas9, have been proposed to spread desirable genes throughout populations. However, invasion of such drives can be hindered by the accumulation of resistant alleles. To limit this obstacle, we engineer a confinable population modification home-and-rescue (HomeR) drive in Drosophila targeting an essential gene. In our experiments, resistant alleles that disrupt the target gene function were recessive lethal and therefore disadvantaged. We demonstrate that HomeR can achieve an increase in frequency in population cage experiments, but that fitness costs due to the Cas9 insertion limit drive efficacy. Finally, we conduct mathematical modeling comparing HomeR to contemporary gene drive architectures for population modification over wide ranges of fitness costs, transmission rates, and release regimens. HomeR could potentially be adapted to other species, as a means for safe, confinable, modification of wild populations.

Reef manta rays (Mobula alfredi) are capable of long-distance dispersal when habitat is continuous. In the Ningaloo Reef World Heritage Area located on Australia’s mid-west coast, M. alfredi is sighted year-round and is a focal species for ecotourism in the region. Despite value to local tourism and demographic vulnerability to exploitation, basic information regarding M. alfredi movements and area use in this region is lacking. Here we used satellite tags and a long-term photographic-identification database collected by researchers and citizen scientists to examine the species’ visitation patterns and regional space use. Tagged M. alfredi remained in coastal shelf waters, preferentially occupying shallow depths < 20 m (76% of depth observations). Comparison of real tracks against those produced by correlated random walk simulations revealed directional dispersal and connectivity between the Ningaloo and Shark Bay World Heritage Areas. The greatest linear distance an individual moved after tagging was ∼700 km. This movement range is comparable to seasonal M. alfredi migration observed at similar latitudes on Australia’s east coast. Photographic-identification of 1121 individual M. alfredi, from 5146 sighting events, revealed that some individuals exhibited long-term site affinity to locations within Ningaloo Marine Park, with the longest spanning 15.16 years (9.8% individuals > 10 years). Lagged Identification Rate analysis suggested site visitation was best described by a pattern of emigration and reimmigration. While our observations of movement and residency establishes basic information previously unknown for the species in this region, the overlap of core area use with existing UNESCO World Heritage Areas demonstrates the potential for well-positioned marine parks to provide protection of critical habitat for M. alfredi.
