Technology development and the general availability of information have affected modern society in such a way that the educational system requires immediate and decisive changes. The situation escalated during the pandemic when distance learning became an integral part of life of every teacher and student. Modern researchers call the educational system built on the flipped classroom model a pedagogical breakthrough, so it is important to study its impact from all sides; this explains the relevance of this paper. The purpose of this research was to study the effectiveness of flipped classroom as a distance learning component for students. The study was conducted at St. Petersburg State University and enrolled 56 students in the control and experimental groups (n = 28 per group). The researchers used the questionnaire "Studying the motives of students' educational activities" by A.A. Rean and V.A. Yakunin and a cross-section of grades to obtain information about academic performance and a survey for feedback from students. The findings show that flipped classroom had a positive effect on academic performance and student motivation. The number of "excellent" students increased by 17.9% and the numbers of "good" and "satisfactory" students decreased by 3.6% and 14.3%, respectively. The overall motivation of the group increased from 4.8 to 5.0. At the same time, the number of students with low motivation decreased by 7.2%, with medium motivation increased by 10.7%, and with high motivation decreased by 3.4%. A feedback survey showed that the vast majority of students were satisfied with the flipped classroom. At that, 89.2% of students answered that this model is suitable for knowledge assimilation, 92.8% believe that flipped classroom arouses their research interest, and 82.1% call the flipped classroom model the most suitable for interesting learning. The respondents noted the following advantages of the flipped classroom: saving time (82.7%), the opportunity to discuss more interesting topics in class (64.2%), the absence of dependence on time and place (38.1%), and the possibility of a deeper study (53.5%). The disadvantages included the inability to independently study the material (10.7%), a large amount of material (17.8%), and technical problems (7.1%). These findings are of value for further study of the effectiveness of flipped classroom introduction into the educational system and may be used to compile statistics or as a basis for a similar experiment.

Tay-Sachs disease (TSD) is a rare genetic disorder with diverse clinical manifestations, often leading to underdiagnosis due to symptom similarities with other neurological conditions. In this study, we aimed to identify the genetic mutations underlying late-onset TSD in a 27-year-old patient with progressive neurological symptoms. Whole-exome sequencing revealed two hexA gene mutations associated with TSD: a previously known variant, c.805G > A (p.Gly269Ser), and a novel splice-site mutation, c.346 + 2dupT. Through clinical assessments, genetic analysis, and functional investigations—including RNA sequencing and enzymatic activity assays—we confirmed the pathogenicity of the novel mutation. Our findings highlight the efficacy of advanced genomic technologies in diagnosing intricate genetic disorders and emphasize the significance of functional validation to confirm the effects of mutations. Identifying compound heterozygous mutations in the hexA gene also provides insight into Mendelian inheritance patterns. This case highlights the diagnostic challenges posed by overlapping clinical phenotypes and emphasizes the need for increased genetic awareness among clinicians. Accurate diagnosis of TSD has significant implications for patients and their families, allowing for informed genetic counseling and guiding clinical management decisions. While current treatment options are limited, timely and accurate diagnosis holds promise for future research and therapeutic interventions. This study highlights the value of a multidisciplinary approach in exploring the molecular basis of complex genetic diseases and informing clinical decisions.

The displacement efficiency and residual oil saturation are the most important characteristics determining the recoverable volumes of oil and the waterflooding efficiency. The main data about these parameters are provided by laboratory core studies. Specialized field tests make it possible to evaluate the same characteristics in-situ, directly on a well, thus excluding the influence of scale effect and impact of core sampling and preparation.This paper presents the results of a field study to evaluate the displacement efficiency and residual oil saturation on a well draining terrigenous Devonian formations on one of the areas of the Romashkinskoye field. The results are compared with generalized dependencies based on core data. Significant influence of the scale effect (the effect of sweep multiplicity) on the estimated parameter values was noted, which indicates the need to supplement laboratory experiments with specialized field studies.

Abstract The development of craniofacial skeletal structures is fascinatingly complex and elucidation of the underlying mechanisms will not only provide novel scientific insights, but also help develop more effective clinical approaches to the treatment and/or prevention of the numerous congenital craniofacial malformations. To this end, we performed a genome-wide analysis of RNA transcription from non-coding regulatory elements by CAGE-sequencing of the facial mesenchyme of human embryos and cross-checked the active enhancers thus identified against genes, identified by GWAS for the normal range human facial appearance. Among the identified active cis-enhancers, several belonged to the components of the PI3/AKT/mTORC1/autophagy pathway. To assess the functional role of this pathway, we manipulated it both genetically and pharmacologically in mice and zebrafish. These experiments revealed that mTORC1 signaling modulates craniofacial shaping at the stage of skeletal mesenchymal condensations, with subsequent fine-tuning during clonal intercalation. This ability of mTORC1 pathway to modulate facial shaping, along with its evolutionary conservation and ability to sense external stimuli, in particular dietary amino acids, indicate that the mTORC1 pathway may play a role in facial phenotypic plasticity. Indeed, the level of protein in the diet of pregnant female mice influenced the activity of mTORC1 in fetal craniofacial structures and altered the size of skeletogenic clones, thus exerting an impact on the local geometry and craniofacial shaping. Overall, our findings indicate that the mTORC1 signaling pathway is involved in the effect of environmental conditions on the shaping of craniofacial structures.

Pathological processes in the skin of domestic animals are one of the most common diseases of domestic animals both in the Kyrgyz Republic and throughout the world. In connection with the complex differential diagnosis of oncological and non-neoplastic diseases in animals based only on clinical signs, the purpose of this work was to conduct a statistical and histomorphological analysis of cases of atheroma development in dogs. Clinical, histological and statistical methods were used during the research. The research was conducted on a controlled group of domestic dogs in the amount of 12 animals, which were followed for several years. When collecting the anamnesis, in addition to clinical signs, the breed, age and concomitant diseases of the dogs were recorded. During surgical removal of atheroma, histological preparations were prepared to clarify the diagnosis and study histomorphological changes in the tissues. Atheroma of the skin in dogs was most often registered in old animals, which were more than 9 years old at the time of diagnosis. At the same time, such pathology was most often localized in dogs in the area of the head and neck. Among the breeds, the Boxer turned out to be the most susceptible to the formation of sebaceous gland cysts. Also, the incidence among females was 75% of all cases of registered diseases. Histological studies revealed the prevalence of cases with the formation of multiple cysts – 67%, when a number of smaller formations are found near one huge atheroma, while cases of purulent inflammation were only 16%. Surgical removal of the atheroma was an effective method of treatment, while no cases of re-development of cysts were observed during the long postoperative follow-up. The obtained results make it possible to expand knowledge on the clinical manifestation, diagnosis and treatment of non-neoplastic skin diseases in domestic animals

Aims: The aim is to analyze the impact of macroeconomic factors on the dynamics of realizing the foreign trade potential of companies based on vector autoregression and the formation of strategic priorities for the companies’ economic activities. Methods: The research methodology is based on the application of methods of time series analysis and impulse response functions. The Granger causality test was used for the linear causal relationship between the analyzed indicators. The study used statistical data for the period 2001–2020 in the annual section of the World Bank Trade Map portal. Results: It is shown that the innovative development of the country contributes to an increase in non-raw material trade with countries geographically close to its borders. It was revealed that the level of agricultural development of the importing country influences the import of cereals with a lag effect of 3–4 years. Justified: The intensification of export-import operations contributes to the development of the production potential of countries as a whole, creating great potential for enhancing the economic activity of agricultural companies and creating incentives for the development of the economies of the country's regions. It has been determined that the foreign trade system is an equilibrium system that returns to a stable state 2-3 years after the shock impact of exogenous and endogenous factors. Novelty: The novelty of the study lies in the assessment of the dynamic impact of the shock impact of exogenous and endogenous factors on macroeconomic indicators, taking into account the specifics of the foreign trade turnover of countries. Conclusion: The results of the study can be used to develop a strategy for the development of foreign trade activities, including grain crops for both countries, to realize more reliable forecasting of foreign economic transactions with a lag of 3-5 years. IntroductionThe remarkable diversity observed in the structure and development of the molluscan nervous system raises intriguing questions regarding the molecular mechanisms underlying neurogenesis in Mollusca. The expression of SoxB family transcription factors plays a pivotal role in neuronal development, thereby offering valuable insights into the strategies of neurogenesis.MethodsIn this study, we conducted gene expression analysis focusing on SoxB-family transcription factors during early neurogenesis in the gastropod Lymnaea stagnalis. We employed a combination of hybridization chain reaction in situ hybridization (HCR-ISH), immunocytochemistry, confocal microscopy, and cell proliferation assays to investigate the spatial and temporal expression patterns of LsSoxB1 and LsSoxB2 from the gastrula stage to hatching, with particular attention to the formation of central ring ganglia.ResultsOur investigation reveals that LsSoxB1 demonstrates expanded ectodermal expression from the gastrula to the hatching stage, whereas expression of LsSoxB2 in the ectoderm ceases by the veliger stage. LsSoxB1 is expressed in the ectoderm of the head, foot, and visceral complex, as well as in forming ganglia and sensory cells. Conversely, LsSoxB2 is mostly restricted to the subepithelial layer and forming ganglia cells during metamorphosis. Proliferation assays indicate a uniform distribution of dividing cells in the ectoderm across all developmental stages, suggesting the absence of distinct neurogenic zones with increased proliferation in gastropods.DiscussionOur findings reveal a spatially and temporally extended pattern of SoxB1 expression in a gastropod representative compared to other lophotrochozoan species. This prolonged and widespread expression of SoxB genes may be interpreted as a form of transcriptional neoteny, representing a preadaptation to prolonged neurogenesis. Consequently, it could contribute to the diversification of nervous systems in gastropods and lead to an increase in the complexity of the central nervous system in Mollusca.

Purpose of the study: to evaluate the efficacy and feasibility of using brimonidine 0.2 % in the additive therapy of patients with advanced stages of glaucoma. Materials and methods. There were 59 patients (77 eyes) with advanced stages of primary open-angle, glaucoma (POAG) aged 68 to 83 years in the study. Patients were randomized into 2 groups depending on the received antihypertensive therapy. The first group (I) included 32 people (41 eyes) who used a fixed combination of a prostaglandin analogue (APG) or a carbonic anhydrase inhibitor (CAI) with a beta-blocker (BB) for the treatment of POAG. The second group (II) consisted of 27 patients (36 eyes) who had the α2-adrenergic agonist brimonidine — S3 added to the previously available combination of APG or ICA with BB. The observation period was 6 months. Results. Application of brimonidine — SZ in the additive therapy of advanced stages of glaucoma led to an additional hypotensive effect. By the end of the observation period in the group of patients treated with brimonidine, intraocular pressure decreased by an additional 10 % from the initial level. A good hypotensive effect contributed to an increase in functional parameters. Analysis of the results of static automatic perimetry showed a positive trend in the mean deviation (MD) and standard deviation pattern (PSD), which may indicate a neuroprotective effect of the drug. All patients noted good tolerability of treatment. Throughout the observation period, patients maintained a high level of adherence and performance.

BACKGROUND: The use of telemedical technologies (TMT) is widespread in providing care to patients of various profiles. However, TMTs are hardly ever used in gastroenterology, despite the extensive digitalization of healthcare. Potential sites for the use of TMTs among gastroenterological patients were assessed. AIM: The study aimed to assess the effectiveness and benefits of TMTs in patients with digestive diseases. Patients with colorectal cancer (CRC): Screening and postoperative management. The TMT intervention increased the number of patients screened for CRC compared to standard face-to-face counseling. In addition, the quality of preparation for colonoscopy was higher in patients who interacted with the physician via TMTs compared to the control group. Most studies reported positive effects of TMTs on quality of life and physical activity in patients with CRC. However, the study by K. Beaver et al. showed that the incidence of CRC recurrence did not differ in the TMT and face-to-face groups. Patients with nonalcoholic fatty liver disease (NAFLD): Correction of lifestyle. When assessing the use of TMTs among NAFLD patients, a significant decrease in alanine aminotransferase and aspartate aminotransferase activity was found compared to that in face-to-face management. The results of the body mass index reduction are contradictory. The most pronounced tendency towards weight loss was observed with the telemedical follow-up of patients over a 6-month period. Patients with irritable bowel syndrome (IBS): Treatment and monitoring of symptoms. Two directions are distinguished in the treatment of IBS by ТМТs. The first is cognitive behavioral therapy (CBT). H. Everitt et al. showed clinically significant improvement in 72.8% of cases after 12 months of web-based CBT. The advantages of using telemedicine in the delivery of CBT include the geographical independence of the physician and the patient, personalization of treatment, and the possibility of ongoing support for the patient and his or her family. The second direction is the use of special diets, such as low-fermentable nutrient diets (FODMAP). Patients following the FODMAP diet supervised by the mobile app reduced symptom severity (odds ratio 55; 95% confidence interval 1198, p=0.01). Special mobile apps (IBS Constant Care) were used to monitor the condition of patients with IBS, which analyze input data and assess severity, providing information to both the patient and the physician. Patients with inflammatory bowel disease (IBD): Patient training and telemonitoring. The study by L. Pang et al. proved that the patients quality of life in the telemedicine intervention group was significantly higher (p=0.03). Another advantage of using TMTs is the provision of access to educational materials about IBD and the possibility of remote contact with the treating physician. The patients ability to obtain necessary information reduces the need for unplanned visits to medical facilities, thus reducing the cost of organizing medical care for patients with IBD. The degree of IBD activity and risk of recurrence and adherence to therapy did not differ significantly between the telemedicine intervention group and the standard face-to-face follow-up group. METHODS: A literature search relevant to the review was conducted in the PubMed database. The studies taken for assessment were those on the use of TMTs among patients with IBD, IBS, NAFLD, and CRC. TMTs were used to access educational information and dietary recommendations, provide information on lifestyle adjustments and physical activity, remind patients to take medicines, consult with physicians, and assess disease activity by completing online questionnaires. The various studies used text messaging, e-mail, mobile apps, websites, and videoconferencing apps as a means of communication. RESULTS: The use of telemedicine in the management of patients with digestive diseases showed high efficiency. TMTs contribute to: Improved quality of life in patients with digestive diseases. Increasing physical activity of patients with CRC and NAFLD. Improved quality of CRC screening. Maintaining remission of IBD and CRC as effectively as face-to-face monitoring. Moreover, the use of TMTs allows personalized treatment, ensures the ongoing support of the patient and his or her family by the medical staff, and provides patients with access to quality educational materials about the disease. CONCLUSIONS: Thus, the use of TMTs is promising in the treatment of digestive diseases and requires wider introduction into practice.

The relevance of the research consists in the fact that the folk wisdom of any nation contains morals for all occasions. Scientific researches usually consider only positive examples of educational value, despite the fact that folk wisdom contains thoughts that warn of danger. The problem is that it is necessary to uncover the thoughts of the Tatar people about ensuring life safety in everyday conditions, in particular in relations with a neighbor. The Tatar people are versatile and diverse in revealing and describing relations and interactions with their neighbors. Tatar folk proverbs and sayings contain more thoughts about a positive attitude towards a neighbor than careful handling of them. The purpose of this study is to reveal folk wisdom from the point of view of ensuring life safety at the household level. Empirical methods such as observation, comparing proverbs and sayings with the experience of people in real life, and theoretical methods such as abstraction, analysis and synthesis, idealization, induction and deduction, mental modeling, ascent from the abstract to the concrete, etc. have been used in the research. As a result, the authors have revealed that Tatar Folk Pedagogy, in particular, proverbs and sayings contain many examples of how to properly organize relations with neighbors, to keep distance, to be compliant, to live in harmony and peace, to know the recommendations and adhere to the instructions of Folk Pedagogy, so that one doesn’t run into trouble. All this in the end will preserve the life and health of people, it’s something everyone should know about, because everyone has beloved neighbors. The literature analysis confirms the interrelationship of thyroid gland pathology and glaucoma. Patients with diffusetoxic goiter (DTG) and endemic goiter have an especially high risk of developing glaucoma, while those with autoimmune thyroiditis face a moderate risk. The prevalence of primary open angle glaucoma (POAG) in patients over 40 with endocrine ophthalmopathy is reliably associated with the male gender and the duration of the disease longer than 60 months. An increased risk of POAG is noted in men with hypothyroidism averagely aged 69. In most cases, ophthalmic hypertension that accompanies endocrine pathology does not need any topical hypotensive treatment. The main pathogenetically validated therapy of thyroid disorders results in a reduction of IOP level and an improvement of fluid outflow from the eye.

Abstract Background More than half of human protein-coding genes have an alternative transcription start site (TSS). We aimed to investigate the contribution of alternative TSSs to the acute-stress-induced transcriptome response in human tissue (skeletal muscle) using the cap analysis of gene expression approach. TSSs were examined at baseline and during recovery after acute stress (a cycling exercise). Results We identified 44,680 CAGE TSS clusters (including 3764 first defined) belonging to 12,268 genes and annotated for the first time 290 TSSs belonging to 163 genes. The transcriptome dynamically changes during the first hours after acute stress; the change in the expression of 10% of genes was associated with the activation of alternative TSSs, indicating differential TSSs usage. The majority of the alternative TSSs do not increase proteome complexity suggesting that the function of thousands of alternative TSSs is associated with the fine regulation of mRNA isoform expression from a gene due to the transcription factor-specific activation of various alternative TSSs. We identified individual muscle promoter regions for each TSS using muscle open chromatin data (ATAC-seq and DNase-seq). Then, using the positional weight matrix approach we predicted time course activation of “classic” transcription factors involved in response of skeletal muscle to contractile activity, as well as diversity of less/un-investigated factors. Conclusions Transcriptome response induced by acute stress related to activation of the alternative TSSs indicates that differential TSSs usage is an essential mechanism of fine regulation of gene response to stress stimulus. A comprehensive resource of accurate TSSs and individual promoter regions for each TSS in muscle was created. This resource together with the positional weight matrix approach can be used to accurate prediction of TFs in any gene(s) of interest involved in the response to various stimuli, interventions or pathological conditions in human skeletal muscle.

Th e article presents an algorithm for diff erential search for the cause of gestational arterial hypertension, clinical and laboratory data and therapeutic measures provided to the patient, on the basis of which endocrinologists together with obstetricians and gynecologists can determine the tactics of pregnancy and delivery of women with hormone-active adrenal tumors.

The Siberian wood frog Rana amurensis Boulenger, 1886 is the most hypoxia-tolerant amphibian. It can survive for several months in an almost complete absence of oxygen. Little is known about the mechanisms of this remarkable resilience, in part because studies of amphibian genomes are impeded by their large size. To make the Siberian wood frog more amenable for genetic analysis, we performed transcriptome sequencing and de novo assembly for the R. amurensis brain under hypoxia and normoxia, as well as for the normoxic heart. In order to build a de novo transcriptome assembly of R. amurensis, we utilized 125-bp paired-end reads obtained from the brain under normoxia and hypoxia conditions, and from the heart under normoxia. In the transcriptome assembled from about 100,000,000 reads, 81.5 % of transcripts were annotated as complete, 5.3 % as fragmented, and 13.2 % as missing. We detected 59,078 known transcripts that clustered into 22,251 genes; 11,482 of them were assigned to specific GO categories. Among them, we found 6696 genes involved in protein binding, 3531 genes involved in catalytic activity, and 576 genes associated with transporter activity. A search for genes encoding receptors of the most important neurotransmitters, which may participate in the response to hypoxia, resulted in a set of expressed receptors of dopamine, serotonin, GABA, glutamate, acetylcholine, and norepinephrine. Unexpectedly, no transcripts for histamine receptors were found. The data obtained in this study create a valuable resource for studying the mechanisms of hypoxia tolerance in the Siberian wood frog, as well as for amphibian studies in general.

Autophagy is a highly conserved catabolic process in eukaryotic cells. Reactive nitrogen species play roles as inductors and signaling molecules of autophagy. A key mechanism of NO-mediated signaling is S-nitrosylation, a post-translational modification (PTM) of proteins at cysteine residues. In the present work, we analyzed the patterns of protein S-nitrosylation during the induction of autophagy in Triticum aestivum roots. The accumulation of S-nitrosylated proteins in the cells during autophagy induced with KNO2 and antimycin A was visualized using monoclonal antibodies with a Western blot analysis, and proteins were identified using a standard bottom-up proteomics approach. Protein S-nitrosylation is a labile and reversible PTM, and therefore the SNO group can be lost during experimental procedures. A subsequent bioinformatic analysis using predictive algorithms and protein-ligand docking showed that identified proteins possess hypothetical S-nitrosylation sites. Analyzing protein–protein interaction networks enabled us to discover the targets that can directly interact with autophagic proteins, and those that can interact with them indirectly via key multifunctional regulatory proteins. In this study, we show that S-nitrosylation is a key mechanism of NO-mediated regulation of autophagy in wheat roots. A combination of in silico predictive algorithms with a mass spectrometry analysis provides a targeted approach for the identification of S-nitrosylated proteins.

Autophagy is a highly conserved process that degrades damaged macromolecules and organelles. Unlike animals, only scant information is available regarding nitric oxide (NO)-induced autophagy in plants. Such lack of information prompted us to study the roles of the NO donors’ nitrate, nitrite, and sodium nitroprusside in this catabolic process in wheat roots. Furthermore, spermine, a polyamine that is found in all eukaryotic cells, was also tested as a physiological NO donor. Here, we show that in wheat roots, NO donors and spermine can trigger autophagy, with NO and reactive oxygen species (ROS) playing signaling roles based on the visualization of autophagosomes, analyses of the levels of NO, ROS, mitochondrial activity, and the expression of autophagic (ATG) genes. Treatment with nitrite and nitroprusside causes an energy deficit, a typical prerequisite of autophagy, which is indicated by a fall in mitochondrial potential, and the activity of mitochondrial complexes. On the contrary, spermine sustains energy metabolism by upregulating the activity of appropriate genes, including those that encode glyceraldehyde 3-phosphate dehydrogenase GAPDH and SNF1-related protein kinase 1 SnRK1. Taken together, our data suggest that one of the key roles for NO in plants may be to trigger autophagy via diverse mechanisms, thus facilitating the removal of oxidized and damaged cellular constituencies.

Family and preschool institutions are the most influential environments for education and development. Therefore, the aim of this research is to study the prospects and drawbacks of early childhood development in the interaction of preschool and family education in Russia. The participants were 79 preschool teachers and 327 parents of children who attend kindergartens. Findings show that the most convenient forms of family-preschool interaction are group lessons, tours and group celebrations. Employees of preschool institutions are the least interested in the utilisation of information and commutation technologies. Commonly, the staff may have children aged 3-5 listening to music, playing board games, reading books, modelling, making applique, drawing and performing small routine tasks (collecting toys and dishes, watering the flowers). In contrast to the institute of family education, preschool education draws more attention to the child development. Findings demonstrate that unlike educators, parents endorse the use of digital technologies but 74.7% of them use technologies for leisure applications, rather than for educational and developmental purposes. The mentioned development methods are applied to reach child development by 28% of parents and 43.65% of educators. The findings may be applied to facilitate communication between parents and preschool teachers.

In recent years, however, due to the development of digital scanning and data processing technologies and the emergence of non-invasive highly informative diagnostic methods, it has become possible to reveal structural and functional changes of the optic disc in vivo, in addition to the traditional detection of histological changes in cadaveric eyes.Glaucomatous process modeling is one of the challenges in ophthalmology.
