1. Safety and immunogenicity of live intranasal pertussis vaccine GamLVP in the experimental infant hamadryas baboon model

Abstract

Introduction. The Gamaleya National Center of Epidemiology and Microbiology has developed a live intranasal pertussis vaccine, GamLVP, for protection against whooping cough. It is indicated for vaccination of infants and revaccination of adults of all age groups. Preclinical studies on suckling mice or rats and adult monkeys as well as clinical trials involving adult volunteers demonstrated safety and efficacy of the GamLVP vaccine. The expansion of the GamLVP vaccine to be used for vaccination of infants requires additional preclinical studies to assess its safety and immunogenicity in the most suitable experimental model of infant hamadryas baboons (Papio hamadryas).The aim of the study was to assess safety and immunogenicity of the GamLVP vaccine administered intranasally for a single dose, two-dose, and three-dose immunization of P. hamadryas infants.Materials and methods. The study was performed in three 1–2-month-old P. hamadryas infants kept, together with their mothers, in a separate cage. The results of the complete blood count and biochemical profile tests were measured before and after the immunization and experimental infection. The enzyme immunoassay (EIA) was used to detect any changes in the levels of specific IgG antibodies in sera from the mothers and infants; the agglutination test (AT) was used to measure titers of total anti-pertussis antibodies.Results. The intranasal immunization of P. hamadryas infants with the GamLVP vaccine triggered development of a specific humoral immune response mediated by IgG antibodies (pertussis toxin + filamentous hemagglutinin), increased titers of total agglutinating anti-pertussis antibodies, caused no local and systemic reactions, caused no changes in the complete blood count and biochemical profile. The experimental infection of the GamLVPimmunized P. hamadryas infants did not cause any changes in the laboratory blood test values and any clinical manifestations typical of the pertussis infection.

Abstract

The absence of an adequate experimental animal model makes difficult study of immunity against whooping cough and its pathogenesis. Experimental whooping cough reported by us earlier in pubescent non-human primates of the Old World was accompanied by specific clinical and laboratory marks in the absence of cough. The possibility of pertussis modelling while experimental whooping cough in impuberal hamadryad baboons was investigated. In the process of selection of monkeys for the further studies for perfecting of experimental model for pertussis research unexpectedly were detected specific pertussis antibodies in impuberal hamadryad baboons.The aim of the study: revealing of source of infection and transmission of pertussis to hamadryad baboons and investigation of response of antibody-positive impuberal hamadryad baboons to secondary contagion by B. pertussis bacteria while experimental infection.Results. 18 veterinary checked, somatically healthy hamadryad baboons of various gender managed in two neighboring cages. Specific pertussis IgM and IgG antibodies were found in blood serum of all the animals and one of the monkey keepers. By real-time PCR in nasopharyngeal swabs of the monkey keeper and three 7- and 9-month-old hamadryad baboons were registered single B. pertussis genom equivalents. Seropositive impuberal hamadryad baboons were experimentally challenged by virulent B. pertussis 475 strain. Quantity of B. pertussis genom equivalents and percentage of IgM and IgG antibodies in impuberal hamadryad baboons after experimental infection were detected. These results were comparable with such received after secondary experimental challenge of monkeys by B. pertussis. Humoral immuneresponse was characterized by booster effect and rapid B. pertussis elimination.Conclusion. The case of transmission of B.pertussis bacteria to hamadryad baboons by natural contagion and epizootic focus of pertussis in apery conditions were registered. In dynamics of immune response and level of bacterial load in experimentally infected impuberal and pubescent hamadryad baboons were not revealed significant differences. The possibility of asymptomatic B.pertussis transmission from man to monkey and from monkey to man without definitive spasmodic cough was reviewed. Pertussis research perspectives using experimental model of non-human primates of the Old World were marked.

Abstract

The objective: to evaluate safety and efficacy of bedaquiline in the treatment of patients with multiple drug resistant tuberculosis (MDR-TB).Subjects and Methods. Clinical data of 523 patients above 18 years old registered for MDR TB treatment as per regimen 4 were retrospectively analyzed. HIV-negative patients were selected and divided into two groups by the use of bedaquiline in the chemotherapy regimen.Results. In MDR TB treatment regimen containing bedaquiline, adverse reactions were observed in 95.7% of cases, the majority of them (65.2%) were of mild or moderate degree of severity. In 30.4% of such cases, bedaquiline was temporarily discontinued (mostly due to hepatotoxic reactions). No cardiotoxicity was observed as a response to bedaquiline, the median corrected QT interval made 413 ms, the maximum value was 438 ms. Adding bedaquiline to the regimen resulted in faster (in the first 3 months of treatment) and more frequent (in 94.7%) sputum conversion by culture (in the regimens with no bedaquiline, it was achieved in 63.8% of cases, p = 0.019). Treatment with regimens containing bedaquiline was effective in 82.61% of patients, while regimens with no bedaquiline were effective in 65.2% of cases, p = 0.085.

6. CLASSIFICING CATEGORY OF GENDER IN UKRAINIAN AND OTHER LANGUAGES OF DIFFERENT SYSTEMS: TO THE PROBLEM OF LANGUAGE TEACHING FOR FOREIGN STUDENTS

Abstract

The article under review outlines the problems that a language teacher who works with a foreign language to foreign students may be faced. The grammar of each language is a system, but this system is, of course, specific. Different grammatical categories of language, non-identical are the general meanings by which grammatical forms are opposed to each other. It is known that with the same number of forms in different languages, their meanings can be correlated differently. In addition, homogeneous grammatical categories can be distributed unequally between different parts of speech. In some languages, affixes as morphemes, which express semantic and syntactic elements of meanings are not relationally connected to the word form and the whole system of its forms in inflectional languages – such as Russian, Ukrainian, Belarusian, etc., as well as derivational ones. Therefore, while analyzing certain languages, researchers do not describe the word change (in the broadest sense of the term), but they work with word formation, because it is the word-formation system that involve a nominal (noun, adjective, verb, numeral, pronoun) and verb paradigm, in which a "deep" morphological and syntactic system of the gender is formed and it outlines the structure, lexical and semantic components of animate and inanimate nature. Grammatical systems are different in not only grammatical meanings, but also in how these meanings are expressed. Taking into account all these features, it turns out that the grammatical system of each language is deeply unique. Despite all these features and characteristics of certain grammatical categories in the languages ​​of different systems, we can outline that you can compare and contrast any of the existing languages, because the grammatical system of languages ​​reveals common features that allow to develop principles for studying of universal categories. Such universals include the category of gender, which is qualified as a morphological-syntactic category of language, which realizes its potential in syntactic structures. The problem of classifying the category of gender in the languages ​​of different grammatical systems is directly related to the research in terms of comparative and contrastive linguistics, and to the problems of studying interfering factors in language.

Abstract

The article considers the problem of linguistic globalization of the modern world and the role of English as the first world language in the history of mankind, the reason for the government’s concern about the lack of English language proficiency in our country since in 2019 the analysts from the International Education Company assessed the level of language proficiency in our country as low. The leadership of Ukraine sees a direct connection between this fact and the low level of economic development of the country and its competitiveness in the international markets, which explains its desire to modernize the Ukrainian higher education system, to adapt it to modern globalization. The authors examined a number of official documents related to the use of English at higher education institutions: the Concept of English Language Development in Universities, Bill № 3717 of the Parliamentary Committee on Education and Science on the Opportunity to Studying a Foreign Language and the Ministry of Education and Science Order “On Approval of improvement in educational programs, which provide training for foreigners and stateless persons in higher education institutions of Ukraine, taking into account the peculiarities of their training». Special attention is paid to the issues of education at higher education institutions of foreign citizens, which is a special item of state income. The article raises a number of issues regarding the duration of Ukrainian language training for foreign students of the 1-4 years of study, the mandatory use of English during Ukrainian language training, the requirements from the Ministry of education and science and university ladministrations on English proficiency at B2 level for teachers who work with foreign students. Contemporary methods of teaching foreign languages, including Ukrainian as a foreign language, which provide for the use of an intermediary language during training or categorically deny such a need, are analyzed, and a number of proposals are made on the issues considered. Based on the studied official documents, the analysis of contemporary methods of teaching foreign languages and personal teaching experience, the authors concluded that the use of English as an intermediary language in teaching foreigners is justified at the initial stage, but in the dosed volume and on the condition of gradual reduction of recourse to it; Since the foreign language classes in student groups are attended by representatives of different countries and the level of English of the majority of foreign students is A2, it is not advisable to require teachers of philology who work with foreigners at the preparatory department and in junior courses of different faculties to be in command of English at B2 level. According to the authors, the problem of raising the status of English in the world and its more active use in higher education institutions in Ukraine is part of a more general problem of language globalization, which makes further study of this issue urgent.

Abstract

The relevance of the research undertaken is determined by the insufficient description of the cognitive approach in relation to word formation units, the unjustifiably rare use of word formation data by the conceptology that is actively developing in linguistics, and the need to form semantic-cognitive analysis procedures relevant to the research object to determine the boundaries of complex units of the word-formation level. The article focuses on the philosophical issues of linguistics, in particular word formation. The peculiarity of linguistics at the end of the 20th century, the essence of which is the recognition of the cognitive conditioning of the main linguistic units and structures, is noted. It is noted that the materialization of certain concepts with the help of linguistic means always consists of a certain ordering, systematization, distribution by headings, categorization. The main function of the analysis and classification of derived words is determined not as groups of derived words or a system of derivational types, but the principles of construction and deployment of complex derivational fragments (blocks, rows, fields), taking into account derivational units larger than a word, analytical analogs of derived words, constructions in the derivation function, only way to represent the derivative and its semantics.

10. METHODS AND STRATEGIES OF STUDYING THE UKRAINIAN LANGUAGE AS A FOREIGN LANGUAGE ON THE BACKGROUND OF MODERN PROBLEMS IN THE WORLD

Abstract

The current situation in the world involves changes and corrections to the existing rules for studying. The transition to online lessons has formed a complex configuration: on the one hand – the technical side with many opportunities, on the other hand – tactics and learning strategies that need to be adjusted and synchronized according to the requirements and capabilities of online learning systems, platforms and programs. The objectives of the article do not include the description and comparison of such virtual learning platforms as, for example, Zoom and Microsoft Teams, but it is impossible not to mention them, because such platforms are modern tools for online learning and need to be used. The article highlights various nuances of teaching methods. In the real audience, the learning process is somewhat different, other systems are involved in communication, in the understanding between students and teachers, in control. I would like to emphasize that we are talking about teaching Ukrainian to foreign students of the preparatory department, about groups where there are students of different nationalities, religions, ages, basic training, as well as different temperaments, interests, life circumstances, and often such groups have no common intermediary languages. Therefore, it is necessary to rely on psychological, psycholinguistic, neurophysiological factors and taking its into account, it is possible to improve the speech, communicative, cognitive competence of students, increase the quality of learning. As for online learning, competencies develop very heterogeneously, depending on the physiological characteristics of the mind and memory, as well as on how honestly and responsibly students perform an independent part of the work. Understanding, working out and mastering different blocks of material takes more time, so it is important to rely on the conclusions of psychologists. Here the teacher's observation of the peculiarities of students' perception and timely influence on various factors, for example, on the motivation to study the material, curiosity, plays an important role. To improve the results in the development of the educational complex, the use of mnemonic techniques and technologies is suggested.

Abstract

Introduction. An increasing incidence of pertussis among different groups of population and shortcomings of the existing preventive solutions pinpoint urgency of development of new safe vaccines suitable for immunization of infants and for booster immunization of adolescents and adults.The purpose of this study is evaluation of safety, immunogenicity and protective activity of the new constructed attenuated Bordetella pertussis bacteria 4MKS by infecting immunized Macaca mulatta monkeys intranasally with virulent bacteria of the pertussis pathogen.Materials and methods. Five adult, clinically healthy Macaca mulatta monkeys aged 3–4 years were used for immunization and experimental infection. The re-immunization was performed in 6 months. Three non-immunized animals of the same age were used as controls.Results. The intranasal single-dose inoculation and re-inoculation of attenuated B. pertussis bacteria did not cause any nasopharyngeal inflammation in the rhesus monkeys and any changes in the blood lab test values after the nonhuman primates had been infected with virulent bacteria. No elevation of total IgE was detected in blood serum of the Macaca mulatta monkeys after the single-dose and double-dose immunization. When the monkeys were intranasally immunized with attenuated and virulent B. pertussis bacteria, they developed a defensive reaction to re-infection, namely suppression of the bacterial growth, increased rates of elimination of bacteria from the animals’ nasopharynxes and development of a humoral immune response to the infection. The development of immunity against pertussis re-infection is accompanied by a pronounced booster effect.Discussion. The obtained results suggest common mechanisms of development both of post-vaccination immunity after intranasal vaccination of animals and infection-acquired immunity against pertussis. Both of them provide protection against re-infection with B. pertussis bacteria and prevent development of clinical symptoms of pertussis.

In this article, the mental construct "meander" is described from the standpoint of cognitive science and the theory of sign systems, it is considered as a universal prototypical symbol in language and culture. "Meander" is qualified as a deep sign, the mental fields of which are actualized by universal metaphorically structured images. Being an ambivalent "universal" Word, the "meander" in its deep structuring is objectified by mental fields of both positive and negative connotations. Based on the well-known position of Yu. S. Stepanov, according to which language as a construct and it consists of a core, social and mental shells, we were able to not only analyze and describe, but also to show the deep nature and potential capabilities of the prototypical symbol of the "meander". No less important for this work is the thesis that language is "a system capable of generating an infinite number of texts". "Meander" is characterized by a complex system of symbols, in which each element carries deep meanings and senses. Understanding the complexity of the prototypical symbol system, it should be noted that the study of such a scientific object as a "meander" should not be limited only from the position of linguistic disciplines. At the same time, "Meander" is attributed to both a stereotype and a prototypical symbol, since both concepts are associated with the content side of language and culture, that is, they are understood as mental constructs that correlate with the picture of the world. The purpose of this article is to describe the multi-component mental construct "meander" from the point of view of its deep structuring. “Meander” as a “stereotype” and a prototypical symbol incorporates the meanings of language and culture, and therefore is understood as a mental construct that adequately reflects the linguistic image of the world. "Stereotypes" and "prototypical symbols", to which we also refer the mental construct "meander", are transmitted by cognitive mechanisms, and, accordingly, are verbalized by one or another sense and meaning. Since the "meander" is a deep abstract prototypical symbol, it is able not only to combine many similar forms of the same object, but also to structure abstract connotative images that are categorized at a deep level. And in this sense, "Meander" is considered by us both as a prototypical symbol, and as a stereotype, and as a pattern, since it is structured according to a model that is often repeated. The results of the analysis showed that the universal, "common human" symbol "meander" is a deeply structured mental construction objectified by meanings and meanings in languages and cultures.

Abstract

In this article, the labour market is considered from the perspective of its development goals in the digital economy. The analysis of scientific literature is carried out taking into account the main directions of labour market research, their classification is proposed. Among the areas of research, the formation of a joint economy, the creation of digital platforms and a transparent information environment, labour market mobility, advanced training and retraining of workers, the development of the educational system, and the importance of human potential are highlighted. A concept has been developed to study the characteristics of the labour market development in the digital economy, which is based on aspects of the development and realization of human potential. The experience of foreign countries was studied, the state of the Russian and Abkhaz labour markets was investigated. The results of a study of the prospects for the educational system development in the context of the digitalization of the labour market are presented, the directions of education modernization and advanced training of workers are established. The features are identified and the risks of the labour market development of Russia and Abkhazia are specified.

Abstract

Avirulent B. pertussis bacteria containing IS elements in the bvgAS operon were detected during the study of whooping cough patients and bacilli carriers. The present work is devoted to the study of the accumulation dynamics and the mechanisms of generation of persistent forms of the B. pertussis bacteria in lower monkeys as the most adequate model for extrapolation ofthe experiment results to humans. By means of the real-time PCR method, it was established that the B. pertussis bacteria lived more than three months in the upper respiratory tract after a single intranasal monkey infection; the period was reduced to 14-28 days during repeated infection. An increase in the portion of B. pertussis Bvg mutants in the population to tens of percent from the total number of registered bacteria was registered. The experimental confirmation ofthe development and accumulation of avirulent B. pertussis Bvg mutants during the development of the infectious process was obtained. Further study of the composition of the B. pertussis persistent bacteria population at different stages of the disease will make it possible to formulate new approaches to the whooping cough diagnostics and prevention and creation of fundamentally new drugs.
