1. Deciphering dose and time dependent effect of supplementing SNP and L-NAME in extender on progressive motility of Murrah bull spermatozoa

Abstract: Male germ cells have the potential to generate reactive oxygen species (ROS) during different stages of differentiation, laboratory manipulation and during their interaction with dissolved oxygen in extender. These ROS and its subset reactive nitrogen species (RNS) perform a concentration specific bimodal role, favouring oxidative eustress at physiological concentrations. The present study was aimed to investigate the dose and time dependent effect of the most potent RNS, i.e. nitric oxide (NO) compounds, specifically SNP (Sodium nitroprusside) and L-NAME (N-Nitro-L-Arginine Methyl Ester) supplementation in extender on progressive motility. The semen samples were divided into 8 aliquots comprising of various concentrations of SNP (Control, 0.1, 1 and 10 μmol/L) and L-NAME (Control, 1, 10 and 100 μmol/L) and refrigerated for 0, 6, 24 and 48 h. The progressive motility (%) improved significantly particularly with the addition of 1 Î¼M SNP and 10 μM L-NAME as compared to control during all time intervals. Subsequently semen sample was cryopreserved with optimum level of SNP (1μmol/L) and L-NAME (10 μmol/L) from these results by adding it in extender and assessed for progressive motility before cryopreservation and after 24 h, 1 month and 2 months of cryopreservation. It is concluded that the addition of SNP and L NAME in cryopreservation media at 1 μM and 10 μM respectively improved the sperm motility significantly. On further evaluation a significant increase in progressive motility was observed for L-NAME treated extender at 24 h and SNP treated extender at 1 and 2 month interval in comparison to control.

2. Angiotensin-II drives changes in microglia–vascular interactions in rats with heart failure

Abstract: Abstract Activation of microglia, the resident immune cells of the central nervous system, leading to the subsequent release of pro-inflammatory cytokines, has been linked to cardiac remodeling, autonomic disbalance, and cognitive deficits in heart failure (HF). While previous studies emphasized the role of hippocampal Angiotensin II (AngII) signaling in HF-induced microglial activation, unanswered mechanistic questions persist. Evidence suggests significant interactions between microglia and local microvasculature, potentially affecting blood–brain barrier integrity and cerebral blood flow regulation. Still, whether the microglial-vascular interface is affected in the brain during HF remains unknown. Using a well-established ischemic HF rat model, we demonstrate the increased abundance of vessel-associated microglia (VAM) in HF rat hippocampi, along with an increased expression of AngII AT1a receptors. Acute AngII administration to sham rats induced microglia recruitment to brain capillaries, along with increased expression of TNFα. Conversely, administering an AT1aR blocker to HF rats prevented the recruitment of microglia to blood vessels, normalizing their levels to those in healthy rats. These results highlight the critical importance of a rather understudied phenomenon (i.e., microglia-vascular interactions in the brain) in the context of the pathophysiology of a highly prevalent cardiovascular disease, and unveil novel potential therapeutic avenues aimed at mitigating neuroinflammation in cardiovascular diseases.

Abstract: Abstract Coronary artery calcium (CAC) scans contain valuable information beyond the Agatston Score which is currently reported for predicting coronary heart disease (CHD) only. We examined whether new artificial intelligence (AI) applied to CAC scans can predict non-CHD events, including heart failure, atrial fibrillation, and stroke. We applied AI-enabled automated cardiac chambers volumetry and calcified plaque characterization to CAC scans (AI-CAC) of 5830 asymptomatic individuals (52.2% women, age 61.7 ± 10.2 years) in the multi-ethnic study of atherosclerosis during 15 years of follow-up, 1773 CVD events accrued. 4. Unbiased Scene Graph Generation in Videos

Abstract: The task of dynamic scene graph generation (SGG) from videos is complicated and challenging due to the inherent dynamics of a scene, temporal fluctuation of model predictions, and the long-tailed distribution of the visual relationships in addition to the already existing challenges in image-based SGG. Existing methods for dynamic SGG have primarily focused on capturing spatio-temporal context using complex architectures without addressing the challenges mentioned above, especially the long-tailed distribution of relationships. This often leads to the generation of biased scene graphs. To address these challenges, we introduce a new framework called TEMPURA: TEmporal consistency and Memory Prototype guided UnceRtainty Attenuation for unbiased dynamic SGG. TEMPURA employs object-level temporal consistencies via transformer-based sequence modeling, learns to synthesize unbiased relationship representations using memory-guided training, and attenuates the predictive uncertainty of visual relations using a Gaussian Mixture Model (GMM). Abstract: Summary: Background: People with diabetes are at increased risk of hospitalisation, morbidity, and mortality following SARS-CoV-2 infection. Long-term outcomes for people with diabetes previously hospitalised with COVID-19 are, however, unknown. This study aimed to determine the longer-term physical and mental health effects of COVID-19 in people with and without diabetes. Methods: The PHOSP-COVID study is a multicentre, long-term follow-up study of adults discharged from hospital between 1 February 2020 and 31 March 2021 in the UK following COVID-19, involving detailed assessment at 5 and 12 months after discharge. The association between diabetes status and outcomes were explored using multivariable linear and logistic regressions. Findings: People with diabetes who survived hospital admission with COVID-19 display worse physical outcomes compared to those without diabetes at 5- and 12-month follow-up. People with diabetes displayed higher fatigue (only at 5 months), frailty, lower physical performance, and health-related quality of life and poorer cognitive function. Differences in outcomes between diabetes status groups were largely consistent from 5 to 12-months. In regression models, differences at 5 and 12 months were attenuated after adjustment for BMI and presence of other long-term conditions. Interpretation: People with diabetes reported worse physical outcomes up to 12 months after hospital discharge with COVID-19 compared to those without diabetes. These data support the need to reduce inequalities in long-term physical and mental health effects of SARS-CoV-2 infection in people with diabetes. Abstract: Introduction: Postpartum depression (PPD) is a form of depression that affects new mothers within the first year after childbirth. PPD can negatively impact both the mother’s quality of life and the child’s development. Untreated PPD can lead to adverse outcomes, such as infant growth retardation, poor child survival, impaired development, and undernutrition. The burden of PPD is a significant public health concern, with a global prevalence increase of 18.4% from 2005 to 2015. Early detection and intervention of PPD can mitigate the severity and chronicity of symptoms. Aims: This study aimed to determine the prevalence of PPD among mothers in an urban area of Kolkata and identify any influencing factors. Materials and Methods: The study utilized a cross-sectional design, surveying 189 mothers within the postpartum period of 6 weeks to 1 year. Edinburg Postnatal Depression Scale was used to identify possible PPD. Results: About 29% of the women had probable PPD, 19% had possible depression, and 15% had no depression. Factors associated with depression included low birth weight and primiparity, while financial independence served as a protective factor. Multinomial regression analysis revealed that low-birth-weight babies, marital conflict, lack of support at home, and primiparity had higher odds of depression, whereas financial independence was a protective factor. Age was not statistically significant. Conclusion: A high prevalence of PPD and significant associations with factors emphasized the importance of screening all mothers for PPD, providing medical care and counselling, and ensuring acceptance and support from partners, family, and society.

Abstract: Background: Tuberculosis (TB) and malnutrition are epidemiologically interrelated two major public health problems in India. Food security also influences nutritional status. This aspect needs evaluation in diverse geographical areas. This study aimed to assess the level of food security, nutritional status and correlates among adult TB patients in a block of Murshidabad, West Bengal. Methods: This cross-sectional study was undertaken in the Nabagram block of Murshidabad district during April–July 2023. All adult (≥18 years) patients registered under the Tuberculosis Unit of the block during a reference period of 6 months (October 2022–March 2023) were considered study subjects. Eighty such eligible participants recruited randomly were interviewed with a pre-designed schedule for socio-demographic and programmatic characteristics. Household food security was measured using a validated U.S. Household Food Security Survey Module. Anthropometric measurements were taken, and nutritional status was determined based on body mass index. Statistical Package for the Social Sciences (SPSS) version 26 was used for analysis. Results: Among 80 TB patients, 51% were underweight and 20% were severely underweight. The majority (90%) of them had high or marginal food security, and 10% had low or very low food security. On multivariable logistic regression, Hindu religion (adjusted odds ratio (AOR): 6.74, 95% confidence interval (CI): 2.12–29.39), presence of any chronic morbidity (AOR: 11.61, 95%CI: 2.71–49.78) and receipt of dietary counselling by a health worker (AOR: 7.25, 95%CI: 1.22–43.13) appeared as predictors of underweight. Conclusions: Underweight among TB patients is quite prevalent in the area, and few programmatic interventions are influencing factors. This underscores the importance of nutritional counselling services and the universal provision of Poshan benefits.

19. Impact of Small Anaerobic Digester on Household Economy of Bangladeshi Livestock Farmers

Abstract: An extensive survey was performed covering all the regions of the country to find out the overall impacts of bio-digester on the economy of livestock farmers. Five districts were selected; ten farmers with having bio-digester of 3.2 m3 on average and ten farmers who have no bio-digester were selected from each district. Through direct interviewing and farm monitoring, all farm characteristics, i.e., diurnal biogas production, power generation, cooking time, income and expenditures, farmer’s gross earnings, and manure management practices data were collected accordingly. Descriptive statistics and student t-test was made to express the comparison response of the farms by using XL and SPSS software. It was observed that the owners of anaerobic digesters earned significantly (p < 0.001) more than the traditional farmers by selling animals and biogas (1715 & 306; 1146 & 0.00 USD, respectively). Not only that, by selling milk and fresh manure, the owners of bio-digester harvested more (p < 0.05) annual income than non-bio-digester farmers (4162, 3408 & 60.91, 44.63 USD, respectively). Though the expenditure of farmers having digester was high, but in a single fiscal year, they earned more (p < 0.05) profit than the conventional farmers (USD 4329 & 2842, respectively). However, owners of bio-digester used 67.2 % of their produced manure for gas production. Regarding storing manure as biomass and using it for cooking purposes significant difference (p < 0.001) was observed that was also reflected in the total manure management system of a farm. The farmers having no bio-digester stored 71.95% of their total manure in solid form, whereas the farmers who had bio-digester only stored 20.4% of their manure, which made a significant (p < 0.001) difference. From the biogas chamber, in an average one farmer used a gas stove for 4-5 hours and a gas lamp for 6-8 hours, which saved at least the expenditure of 18 USD per month/household. The notable thing was that the bio-digester alone contributed 7% to those farmers’ gross economy by producing gas. It can be recommended that the rural householders could generate power by installing bio-digester and turn a small bio-digester as a beneficial avenue of their household economy.

Abstract: Both detailed and rapid population studies alike predict that binary black hole (BHBH) formation is orders of magnitude more efficient at low metallicity than high metallicity, while binary neutron star (NSNS) formation remains mostly flat with metallicity, and black hole–neutron star mergers show intermediate behavior. This finding is a key input to employ double compact objects as tracers of low-metallicity star formation, as spectral sirens, and for merger rate calculations. Yet the literature offers various (sometimes contradicting) explanations for these trends. We investigate the dominant cause for the metallicity dependence of double compact object formation. We find that the BHBH formation efficiency at low metallicity is set by initial condition distributions, and conventional simulations suggest that about one in eight interacting binary systems with sufficient mass to form black holes will lead to a merging BHBH. We further find that the significance of metallicities in double compact object formation is a question of formation channel. The stable mass transfer and chemically homogeneous evolution channels mainly diminish at high metallicities due to changes in stellar radii, while the common envelope channel is primarily impacted by the combined effects of stellar winds and mass-scaled natal kicks. Outdated giant wind prescriptions exacerbate the latter effect, suggesting that BHBH formation may be much less metallicity-dependent than previously assumed. NSNS formation efficiency remains metallicity-independent, as they form exclusively through the common envelope channel, with natal kicks that are assumed to be uncorrelated with mass. Forthcoming gravitational-wave observations will provide valuable constraints on these findings.

While SHAP (SHapley Additive exPlanations) has proven to be a robust tool for explaining machine learning predictions, its high computational cost limits its practicality for real-time use. To address this, we introduce C-SHAP (Clustering-Boosted SHAP), a hybrid method that combines SHAP with K-means clustering to reduce execution times significantly while preserving interpretability. C-SHAP excels across various datasets and machine learning methods, matching SHAP’s accuracy in selected features while maintaining an accuracy of 0.73 for Random Forest with substantially faster performance. Notably, in the Diabetes dataset collected by the National Institute of Diabetes and Digestive and Kidney Diseases, C-SHAP reduces the execution time from nearly 2000 s to just 0.21 s, underscoring its potential for scalable, efficient interpretability in time-sensitive applications. Such advancements in interpretability and efficiency may hold value for enhancing decision-making within software-intensive systems, aligning with evolving engineering approaches.

Abstract: Metarrestin (ML246) is a first-in-class pyrrole–pyrimidine-derived small molecule that selectively targets the perinucleolar compartment (PNC). PNC is a distinct subnuclear structure predominantly found in solid tumor cells. The occurrence of PNC demonstrates a positive correlation with malignancy, serving as an indicator of tumor aggressiveness, progression, and metastasis. Various promising preclinical results have led to the clinical translation of metarrestin into a first-in-human trial. This review aims to summarize (i) the current understanding of the structure and function of PNC and its role in cancer progression and metastasis, (ii) key findings from studies examining the effect of metarrestin on various cancers across the translational spectrum, including in vitro, in vivo, and human clinical trial studies, and (iii) the pharmaceutical relevance of metarrestin as a promising anticancer candidate. Furthermore, our molecular docking and MD simulation studies show that metarrestin binds to eEF1A1 and eEF1A2 with a strong and stable affinity and inhibits eEF1A2 more efficiently compared to eEF1A1. The promising results from preclinical studies suggest that metarrestin has the potential to revolutionize the treatment of cancer, heralding a paradigm shift in its therapeutic management.

Abstract: Adolescence is a critical developmental period of biological and social change during which 1 in 3 youth experience significant anxiety symptoms. The social-emotional lives of the majority of adolescents are largely conducted via digital media use (DMU; e.g., social media, text messaging). Yet the past decade of research on DMU and anxiety has yielded mixed results (e.g., Keles et al., 2020 review), leaving the complex role that DMU might play in the emergence and maintenance of anxiety poorly understood. A key step forward is to leverage psychophysiology to identify individual differences in cognitive and emotional processes that confer vulnerability to potential negative effects of DMU. Further, given the ubiquity of DMU, a greater focus is needed on measurements that move beyond sheer frequency to capture DMU in comparison to face-to-face (FTF) social interactions. This study examined attention bias (AB), characterized by selective and exaggerated attention toward or away from threat, as a moderator of the link between DMU and anxiety in adolescents (N = 75; 42 female) aged 12–14 years (M = 13.28, SD = 0.87). AB was indexed during a dot probe task using reaction time metrics (i.e., trial-level bias) and via ERPs capturing attentional selection and discrimination (N170) and cognitive control (N2) to threat compared to neutral faces. AB moderated associations between DMU and anxiety. A greater preference to use DMU vs FTF predicted greater anxiety among those with a greater behavioral bias away from threat, blunted N170, and blunted N2 in the presence of threat. Future research should examine potential causal and bidirectional links between DMU and anxiety and explore whether preferences for technology-mediated interactions and individual differences in threat processing increase risk.

Abstract: Background The long-term outcomes of COVID-19 hospitalisation in individuals with pre-existing airway diseases are unknown. Methods Adult participants hospitalised for confirmed or clinically suspected COVID-19 and discharged between 5 March 2020 and 31 March 2021 were recruited to the Post-hospitalisation COVID-19 (PHOSP-COVID) study. Participants attended research visits at 5 months and 1 year post discharge. Clinical characteristics, perceived recovery, burden of symptoms and health-related quality of life (HRQoL) of individuals with pre-existing airway disease (i.e., asthma, COPD or bronchiectasis) were compared to the non-airways group. Results A total of 615 out of 2697 (22.8%) participants had a history of pre-existing airway diseases (72.0% diagnosed with asthma, 22.9% COPD and 5.1% bronchiectasis). 31. Energy-Accuracy Trade-Offs for Resistive In-Memory Computing Architectures

Abstract: Resistive in-memory computing (IMC) architectures currently lag behind SRAM IMCs and digital accelerators in both energy efficiency and compute density due to their low compute accuracy. This article proposes the use of signal-to-noise-plus-distortion ratio (SNDR) to quantify the compute accuracy of IMCs and identify the device, circuit, and architectural parameters that affect it. We further analyze the fundamental limits on the SNDR of magnetoresistive random access memory (MRAM-), resistive random access memory (ReRAM-), and ferroelectric field effect transistor (FeFET)-based IMCs employing parameter variation and noise models that were validated against measured results from a recent MRAM-based IMC prototype in a 22 nm process. At high-output signal magnitude, we can find that the maximum achievable SNDR is limited by the pre-analog-to-digital-converter (ADC) array nonidealities, such as the conductance variations (CVs), parasitic resistances, and current mirror mismatch (MM), whereas the ADC thermal (AT) noise limits the SNDR at small signal magnitudes. Furthermore, for large dot-product (DP) dimensions ( $N > 50$ ), the maximum achievable SNDR is highest for FeFET, followed by ReRAM and then MRAM. Finally, the increase in conductance contrast ( ${g_ {\text {ON}} }/ {g_ {\text {OFF}} }$ ) enhances the maximum achievable SNDR only until it reaches a value of approximately 12. ReRAMs and FeFETs demonstrate high energy efficiencies while achieving high SNDR, as their low conductance values lead to lower currents and lower noise due to wire parasitics. In all cases, across all three device types, DP dimension, ADC precision, and conductance contrast, the maximum achievable SNDR is found to be in the range of 18–22 dB, barely meeting the minimum needed for achieving an inference accuracy close to an equivalent fixed-point digital architecture. Finally, we demonstrate a network-level accuracy of 84.5% when mapping an ResNet-20 (CIFAR-10) by ReRAM-based architecture at a SNDR of 22 dB, in which MRAM- and FeFET-based architectures cannot realize. This result clearly implies the need for other approaches, e.g., algorithmic- and learning-based methods, to improve the inference accuracy of resistive IMC architectures.

32. Effect of oxygen vacancy and Si doping on the electrical properties of Ta2O5 in memristor characteristics

Abstract: Abstract The resistive switching behavior in Ta2O5 based memristors is largely controlled by the formation and annihilation of conductive filaments (CFs) that are generated by the migration of oxygen vacancies (OVs). To gain a fundamental insight on the switching characteristics, we have systematically investigated the electrical transport properties of two different Ta2O5 polymorphs ( $$\epsilon$$ ϵ -Ta2O5 and λ-Ta2O5), using density functional theory calculations, and associated vacancy induced electrical conductivity using Boltzmann transport theory. The projected band structure and DOS in a few types of OVs, (two-fold (O2fV), three-fold (O3fV), interlayer (OILV), and distorted octahedral coordinated vacancies (OεV)) reveal that the presence of OILV would cause Ta2O5 to transition from a semiconductor to a metal, leading to improved electrical conductivity, whereas the other OV types only create localized mid-gap defect states within the bandgap. On studying the combined effect of OVs and Si-doping, a reduction of the formation energy and creation of defect states near the conduction band edge, is observed in Si-doped Ta2O5, and lower energy is found for the OVs near Si atoms, which would be advantageous to the uniformity of CFs produced by OVs. These findings can serve as guidance for further experimental work aimed at enhancing the uniformity and switching properties of resistance switching for Ta2O5-based memristors.

Abstract: Background Pre-existing cardiovascular disease (CVD) or cardiovascular risk factors have been associated with an increased risk of complications following hospitalisation with COVID-19, but their impact on the rate of recovery following discharge is not known.Objectives To determine whether the rate of patient-perceived recovery following hospitalisation with COVID-19 was affected by the presence of CVD or cardiovascular risk factors.Methods In a multicentre prospective cohort study, patients were recruited following discharge from the hospital with COVID-19 undertaking two comprehensive assessments at 5 months and 12 months. Patients were stratified by the presence of either CVD or cardiovascular risk factors prior to hospitalisation with COVID-19 and compared with controls with neither. Full recovery was determined by the response to a patient-perceived evaluation of full recovery from COVID-19 in the context of physical, physiological and cognitive determinants of health.Results From a total population of 2545 patients (38.8% women), 472 (18.5%) and 1355 (53.2%) had CVD or cardiovascular risk factors, respectively. Compared with controls (n=718), patients with CVD and cardiovascular risk factors were older and more likely to have had severe COVID-19. Full recovery was significantly lower at 12 months in patients with CVD (adjusted OR (aOR) 0.62, 95% CI 0.43 to 0.89) and cardiovascular risk factors (aOR 0.66, 95% CI 0.50 to 0.86).Conclusion Patients with CVD or cardiovascular risk factors had a delayed recovery at 12 months following hospitalisation with COVID-19. Targeted interventions to reduce the impact of COVID-19 in patients with cardiovascular disease remain an unmet need.Trail registration number ISRCTN10980107.

Abstract: Pseudoaneurysm is a rare but fatal complication of myocardial infarction (MI). With the advances in cardiovascular disease detection and treatments, fatal structural complications post-MI are now rare. When they occur, advanced diagnostic modalities can be used for early diagnosis, aiding surgical planning, and improving prognosis. In our case, post-MI left ventricle pseudoaneurysm complicated by hemopericardium was diagnosed using cardiac computed tomography angiography (CCTA). Use of attenuation measurement on CCTA helped diagnose active extravasation into the hemopericardium. This case highlights the high index of suspicion needed for rare but fatal complications post-MI and the utility of CCTA in their management.

36. MRI tractographic validation of drug-enhanced hepatic clearance of amyloid-beta and the therapeutic potential for Alzheimer's Disease: A pilot study

Abstract: Alzheimer's disease (AD) may require alternative therapeutic perspectives as current interventions may sometimes be sub-optimal. Amyloid-beta 42 (Aβ42) is mainly eliminated by hepatic clearance which diminishes in AD, hence hepatomodulatory drugs enhancing this clearance may have potentiality for therapeutic implication. Here, we clinically substantiate our systems-biology investigation on repurposed hepatomodulating drugs (metformin, cilostazol and rifampicin) which enhance brain insoluble Aβ42 clearance through liver-bile-faeces route. Through MRI-tractographic analysis, we now formulate a three-segmental basis of brain Aβ42 spread: fronto-thalamic region (segment-1), temporo-occipital region (segment-2), and dorso-cingulate region (segment-3). This segmental pattern is corroborated histopathologically by Braak's stages A, B and C. We further observed that the aforesaid three pharmaceuticals respectively acted on those three segmental regions differentially. We analysed MRI and DTI images of 15 healthy controls (CDR: 0; MMSE: 24–20), and 15 AD patients (CDR: 0.5–1.0; MMSE: 20–26). We found that, tractographically, there is a significant reduction in neuronal integrity in the three aforesaid regions in untreated AD compared to controls. Nevertheless, the three drugs increased neural activation of AD patients in the three corresponding areas. These three drugs act by regulating respectively the genes ABCB11, ABCA1 and MDR1. These genes were correspondingly downregulated in the above-mentioned anatomical segments 1, 2 and 3 of Alzheimer's disease. Thus personalized patient-specific hepatomodulative drugs for AD intervention may be explored, as corroborated by the neuroanatomical involvement in AD.

Abstract: Purpose: Teaching and training of glaucoma fellows on the technique of Ahmed glaucoma valve (AGV) implantation surgery is very important. The purpose of this study was to describe the clinical outcomes and complications of AGV surgery performed by glaucoma fellows of a tertiary eye center from eastern India. Methods: This was a retrospective study based on electronic medical records. Thirty-five eyes operated with AGV by five glaucoma fellows from January 2016 to November 2020 were included. Best-corrected visual acuity, intraocular pressure (IOP), and number of antiglaucoma medications (AGMs) were the study outcome measures. Results: The mean (±standard deviation) age of the patients was 40.5 (±19.7) years. The follow-up period was 24.37 (±13.01) months. Both complete success (IOP 21 mmHg with AGM beyond 3 months) in seven eyes. Despite a high rate of failure of the primary surgery, with the consultants’ intervention, there was statistically significant improvement in vision and IOP in all patients (P < 0.01) at the last follow-up. Conclusion: AGV implantation is a challenging surgical skill to be acquired by the glaucoma fellows.
